AFLP分子标记鉴别大白菜品种   总被引:11,自引:0,他引:11  
本试验采用AFLP技术,研究了90份来自7个不同栽培地区的大白菜品种材料。共筛选了20对引物,不同引物组合检测多态性谱带的能力有很大的差异,多态性谱带的数量从9条到32条不等。其中E—ACA/M—CTG是大白菜品种十分高效的引物组合,共产生7l条清晰的扩增带,其中有32条多态性谱带,多态性谱带的百分率为45.7%。通过该引物组合,能将90个品种全部区分开来。同时应用该引物组合检测2个大白菜杂交品种(北京新2号,京夏王)各10株,其中有1株北京新2号的谱带异常,其余同一品种不同单株的带型完全一致。表明AFLP标记用于研究品种指纹图谱,并鉴别品种是完全可行的。  相似文献

本试验采用AFLP技术分析了来自全国9个栽培地区的44个甘蓝主栽品种,共筛选了40对E 3/M 3引物组合,多态性条带的数量从0条到15条不等。其中引物组合E-AAC/M-CTA是甘蓝品种中多态性最高的引物,有15条多态性条带,多态性条带的百分率为30%,但该引物不足以区分44个供试的甘蓝品种。同时筛选了11组E 2/M 3对引物组合,其中引物组合E-AG/M-CTC产生了13条清晰的多态性条带。以E-AAC/M-CTA和E-AG/M-CTC这两对引物组合的多态性条带构建了44份甘蓝品种材料的指纹图谱,此指纹图谱可以将供试的44份材料一一区分。  相似文献

Amplified fragment length polymorphisms (AFLPs) are now widely used in DNA fingerprinting and genetic diversity studies, the construction of dense genetic maps and in fine mapping of agronomically important traits. The AFLP markers have been chosen as a source to extend and saturate a linkage map of rye, which has previously been generated by means of restriction fragment length polymorphism, random amplified polymorphic DNA, simple sequence repeat and isozyme markers. Gaps between linkage groups, which were known to be part of chromosome 2R, have been closed, thus allowing the determination of their correct order. Eighteen EcoRI‐MseI primer combinations were screened for polymorphism and yielded 148 polymorphic bands out of a total of 1180. The level of polymorphism among the different primer combinations varied from 5.7% to 33.3%. Eight primer combinations, which revealed most polymorphisms, were further analysed in all individuals of the F2 mapping population. Seventy‐one out of 80 polymorphic loci could be integrated into the linkage map, thereby increasing the total number of markers to 182. However, 46% of the mapped AFLP markers constituted four major clusters located on chromosomes 2R, 5R and 7R, predominantly in proximity to the centromere. The integration of AFLP markers caused an increase of 215 cM, which resulted in a total map length of almost 1100 cM.  相似文献

福建省致病疫霉菌SRAP遗传多样性分析   总被引:1,自引:0,他引:1  
为了解福建省致病疫霉菌的群体遗传结构,为该病原菌的遗传进化提供理论依据,笔者应用SRAP分子标记技术对福建省致病疫霉菌的群体遗传多样性,及不同地区菌株间的关系进行比较分析。利用10个菌株从110对引物组合中筛选出多态性引物10对,对分离自福建省10个不同市(县)的62个致病疫霉菌菌株DNA进行PCR扩增,共产生92条谱带,其中多态性标记90条,多态检测率为97.8%。利用NTSYpc Version2.1软件对供试菌株间的遗传距离进行聚类分析并构建系统树状图。以遗传距离0.57为阈值,可将供试62个菌株划分为4个遗传聚类组,SRAP分组与菌株的地理来源、寄主均无明显相关性。聚类分析结果表明,福建省不同地区的致病疫霉菌整体亲缘关系相近,但各菌株间存在遗传差异。  相似文献

Forty-one apple (Malus × domestica Borkh.) cultivars were screened for RAPD (Random Amplified Polymorphic DNA) and AFLP(Amplified Fragment Length Polymorphism) markers. RAPD analysis was performed with 35 arbitrary 10-mer primers, selected from 60 primers tested (kits A, C and E, Operon Technologies, Inc.). Of a total of 362bands observed, 208 (57.5%) were polymorphic. Three-hundred-and-eighty-one AFLP fragments were obtained with 8primer combinations, of which 218 (57.2%) were polymorphic. Cultivars differentiated through mutation were included in this study and showed identical patterns when analysed with both RAPD and AFLP analysis. The estimated genetic relationships were correlated (r = 73.7%) between the analysis with the two different markers. UPGMA analysis was performed and dendrograms were constructed using either the data apart from each(RAPD and AFLP) method or combined in a single joint matrix. The relationships among the forty-one studied cultivars were basically consistent with the known lineage and geographic origins of the cultivars. The four Portuguese cultivars included in this study clustered together and diverged from the other cultivars. Apparently they constitute an independent genetic pool, which could be of interest for apple plant breeders. 花生属分子标记领域的研究远落后于其他物种,而栽培种花生因其遗传基础狭窄,用大多数分子标记技术都难以检测到丰富的分子标记,因此限制了花生属野生种在改良花生栽培种方面的利用以及建立花生分子标记辅助育种技术体系。本文分别对花生属4个区组的16份种质资源和8份花生栽培种资源采用与功能基因相关的SCoT分子标记技术研究了花生属种间和栽培种内遗传多样性和亲缘关系。23条SCoT引物在花生属试材基因组中的扩增位点共194个,其中多态性位点130个,多态性达67.01%,通过聚类分析研究了它们之间的亲缘关系;在栽培种内筛选出19条多态性引物,在8份试材基因组中扩增位点198个,其中多态性位点67个,多态性为33.84%,表明SCoT分子标记技术能在花生栽培种内检测出一定程度的DNA多态性。  相似文献

新疆彩色棉23个品种指纹图谱的构建及遗传多样性分析   总被引:6,自引:0,他引:6  
以新疆截止2012年审定的23份新彩棉品种为材料,利用SSR标记进行DNA指纹图谱的构建和遗传多样性分析。从5000对SSR引物中,挑选出多态性高、稳定性好、均匀分布在棉花26条染色体上的52对引物,在23份新彩棉品种中筛选出核心引物47对,SSR扩增检测到多态性基因型位点数共计162个,每个标记检测到的基因型位点数在2~7之间,平均为3.45个;引物多态信息量(PIC)值介于0.4537~0.8686之间,平均值为0.7096。结果显示:在23份新彩棉品种中,14份品种采用特异或特征引物可以一次性区分开,其余9份品种需要采用引物组合来实现区别该品种与其他品种。最少选用18对特异引物及组合引物就可以完全区分开新彩棉1~23号品种。利用18对SSR标记构建了新彩棉1号至23号品种的指纹图谱。利用NTSYS-pcV2.10软件聚类分析表明:23个新彩棉品种遗传相似系数变化范围是0.3781~0.9298,平均为0.5511,表明新彩棉品种之间存在着丰富的遗传多样性。  相似文献

以白菜高抗TuMV白心株系91 112和高感TuMV桔红心株系T12 91为亲本建立的小孢子DH系作为图谱构建群体,以AFLP标记来构建白菜连锁图谱。通过对64对AFLP引物组合的筛选,利用20对引物得到了263个AFLP多态性位点。经Mapmaker/EXP3.0软件处理,构建了1张含255个标记位点,10个连锁群,覆盖长度为883.7cM的连锁图。255个AFLP位点中偏离孟德尔遗传的比率(P<0.05)为15 0%和27 5%(P<0.01)。  相似文献

旨在阐明不同柱花草品种的遗传差异。利用AFLP、SSR两种分子标记技术对7个柱花草品种进行遗传多样性分析。12对AFLP引物组合共扩出966个条带,其中多态性条带有850条,平均每对引物的多态性条带为70.8条,多态性比率为87.99%。10对SSR引物共扩出26个条带,其中多态性条带有23个,多态性比率为88.46%;10对引物的观察杂合度(Ho)的范围是0~0.7143,平均为0.1857,Shannon’s遗传多样性指数(I)平均值为0.6545,平均多态性含量(PIC)为0.727。根据非加权成对平均数法(UPGMA)进行聚类分析,获得品种聚类树形图,AFLP与SSR分析的聚类图相似性显著,7个品种明显分成两类。‘矮柱花草’与‘西卡柱花草’的遗传关系最为疏远,‘奥克雷柱花草’与‘爱德华柱花草’、‘格拉姆柱花草’与‘热研5号柱花草’关系最为密切。  相似文献

Estimates of genetic diversity can be based on different types of data. The aim of this research were to study genetic diversity among Croatian wheat cultivars by random amplified polymorphic DNA (RAPD) markers, morphological traits and pedigree records; to analyse differences between wheat cultivars from two breeding centres; and to evaluate usability of RAPD markers for estimation of genetic diversity among wheat cultivars in comparison with morphological traits and pedigree record data. Studies were conducted on 14 wheat cultivars and breeding lines from two breeding centres in Croatia. For the RAPD analysis 36 primers were screened and the 14 most polymorphic ones yielded 341 polymorphic bands. Twelve morphological traits were used for morphological analysis. Pedigrees were composed of seven generations of ancestors. RAPD markers showed a high level of polymorphism among the cultivars examined and the breeding lines. No significant correlations were observed among the methods tested.  相似文献

DNA amplification fingerprinting (DAF) techniques were used to evaluate genetic relationships among 24 randomly selected white lupin (Lupinus albus L.) accessions originating from four endemic regions. Parameters affecting DAF were optimized to obtain high resolution and reproducible banding patterns. DNA amplification was tolerant to a wide range of both template and primer concentrations but sensitive to Mg2+ concentration, and annealing temperature and time. Pure template DNA was found to be essential for obtaining reproducible banding patterns. Primer annealing occurred at multiple sites, permitting detection of polymorphism using only a single primer. Of 56 octamer primers tested, 22 yielded highly polymorphic DAF bands that could be used to distinguish closely related accessions. Genetic similarity coefficients were calculated and a dendrogram, using the unweighted pair group method with arithmetic average (UPGMA), was constructed. A high level of genome diversity was found among the accessions, but several from the Middle East and West Africa tended to cluster together. The results support the future use of DAF markers for the characterization and identification of white lupin germplasm.  相似文献

利用感蚜高粱品种千三与抗蚜品种河农16杂交获得的100个F2单株作为图谱构建群体,以AFLP标记构建高粱连锁图谱。通过对192对AFLP引物组合进行筛选,共得到75对多态性引物,利用筛选出的75对引物进行选择性扩增,得到了154个AFLP多态性位点。经Map maker/EXP 3.0软件处理,构建了包含12个连锁群,93个遗传标记的连锁图谱,该图谱覆盖686 cM,平均图距为7.38 cM。  相似文献

The AFLP (amplified fragment length polymorphism) technique has been applied in establishing an extended linkage map of sugar beet. A total of 120 AFLPs were integrated into an existing linkage map based on RFLP markers. Four primer combinations yielded between 19 and 40 polymorphic bands in an F2 population consisting of 94 plants. The AFLP loci were evenly distributed over the nine linkage groups, with the exception of linkage group V where the number of AFLPs was significantly low. The AFLPs were found to be reproducible even against the background of different combinations of Taq DNA polymerases and buffers. However, the quantity of higher molecular weight fragments (>400 bp) was reduced when using plant DNA of poor quality as a template. The results of these experiments are discussed, together with possible applications of AFLPs in sugar beet breeding.  相似文献

One‐hundred and twenty‐four amplified fragment length polymorphism (AFLP) and 49 random amplified polymorphic DNA (RAPD) markers have been used to distinguish between 20 and 23 commercial chicory cultivars, respectively. These were all Cichorium intybus var. foliosum F1 hybrids, currently used in hydroponic forcing. Five‐hundred and twenty RAPD primers (OPERON) were tested, of which 156 resulted in reproducible patterns and 26 yielded polymorphisms. Two‐hundred and fifty‐six AFLP primer‐combinations were tested and six combinations were selected for identification purposes. Similarity indices were measured and clustering has been done using pairwise comparison. Both types of marker provide similar conclusions. Two major clusters are formed, representing late and early cultivars. All cultivars were identified using 10 informative RAPD primers or three AFLP primer combinations. A low degree of polymorphism was detected between some early cultivars, suggesting a narrow genetic base in their breeding strategy.  相似文献

Summary Morphological, isozyme and random amplified polymorphic DNA (RAPD) markers were used to estimate genetic variation within and between cultivars of red clover (Trifolium pratense L.), an important temperate forage legume. Two cultivars of red clover, Essi from Europe and Ottawa from Canada, were evaluated. Six monogenic morphological characters were observed for 80 plants from each of these two cultivars. All six morphological loci were polymorphic in the cultivar Essi whereas only four loci were polymorphic in the cultivar Ottawa. Forty plants from each cultivar were assayed for isozyme markers. A total of 21 enzyme-coding loci with 43 alleles was detected using twelve enzyme systems. Thirteen and nine of these loci were polymorphic in Essi and Ottawa, respectively. The mean number of alleles per locus was 1.81 in Essi and 1.67 in Ottawa. Seventeen random 10-mer primers were screened for RAPD markers. Nine primers which gave clear and consistent amplified products were used to assay 20 individuals from each cultivar. Each primer gave from 7 to 20 amplified bands with an average of 14.8 bands per primer. One hundred and eight of 116 putative loci were polymorphic in Essi and 90 of 98 loci were polymorphic in Ottawa. High within-cultivar variation was observed in both cultivars using both isozyme and RAPD markers. This high polymorphism makes these markers useful for germplasm characterization and genetic studies in red clover.  相似文献

为了从分子水平上揭示花椰菜种质资源间的亲缘关系,为其种质搜集、鉴定、利用和遗传改良提供一定的理论基础。本研究采用SRAP分子标记技术对24份花椰菜种质资源进行遗传多样性研究。从30个引物组合中筛选得到23对多态性引物组合,共检测到257条扩增条带,平均每对引物扩增等位基因数从5到20不等。其中多态性片段为185条,多态性比例为71.98%,表明花椰菜种质间具有相对丰富的遗传多样性。相似系数分析表明种质间遗传相似系数为0.5491~0.9626,平均为0.7490。SRAP分子标记聚类分析结果显示来源和地域可作为花椰菜种质聚类的农性状之一。  相似文献

我国棉花抗枯、黄萎病骨干品种(系)基于AFLP的遗传多样性   总被引:10,自引:2,他引:8  
利用AFLP分子标记技术,对我国20世纪50年代开展抗枯、黄萎病育种以来培育的105份骨干品种(系)的遗传多样性进行了研究。结果表明,筛选的20个AFLP引物组合在该品种群体中扩增了1498个AFLP标记,其中多态性标记232个,占总标记数的15.5%。单一引物组合扩增的DNA标记数变化在49111之间,平均每个引物组合扩增的总标记和多态性标记分别为74.9和11.6。在该抗病品种群体中,有46个品种(系)具有特异标记,占品种总数的43.81%,其中引物组合E41/M50可使10个品种产生特异标记。品种(系)之间的成对欧氏距离总平均值为4.353,变幅为1.7326.708,单一品种欧氏距离平均值变幅为3.5315.705,高于总平均值的品种数不足50%,表明该陆地棉抗病品种群体的遗传多样性较低。基于AFLPs多态性数据的聚类分析,105个品种(系)被划分为5个陆地棉品种类群(Uplandcottongroups,UCGs),每个UCG包含的品种数目不同。  相似文献
