为了研究肌细胞生成素(myogenin,MyoG)基因与鸭屠宰性状的关系,试验采用基因克隆以及PCR产物直接测序的方法,将MyoG基因作为鸭屠宰性状的候选基因,首次对鸭的5个外显子进行研究,对兴义鸭的MyoG基因外显子进行多态性检测。结果表明:共发现7处单核苷酸多态性(SNPs)位点,即第1外显子SNP1(G238C)、SNP2(C397G),第3外显子SNP3(C8T),第5外显子SNP4(G23A)、SNP5(G47A)、SNP6(C74T)、SNP7(G104C)。最小二乘分析结果显示,SNP1与胸肌率有显著相关(P0.05),SNP3和SNP7与全净膛率有显著相关(P0.05),SNP7与全净膛重有显著相关(P0.05)。  相似文献

为研究MEF2C基因启动子区SNPs对屠宰性状的影响,本实验运用直接测序法筛选兴义鸭MEF2C基因启动子区的SNP位点,并分析其与屠宰性状的相关性。结果表明:在兴义鸭MEF2C基因启动子上存在8个SNPs位点,分别为T-190A、A-232G、A-285G、A-617T、G-1420A、A-1437C、T-1504G、A-1524G;生物信息学分析发现T-190A、A-232G、G-1420A突变位点周围HOXA4、GCN4、Pit-1a、IRF-1等重要转录因子结合位点消失,继而生成Oct-1、ICSBP 2个新的转录因子结合位点;关联分析结果表明,T-190A不同基因型个体的兴义鸭活重存在显著差异,G-1420A不同基因型个体的兴义鸭腿肌率具有显著差异。由此推断,T-190A、G-1420A位点可能对调控兴义鸭MEF2C基因启动子区功能元件有重要影响,且2个多态性位点可能为影响兴义鸭屠宰性状的重要功能性SNP。本研究结果将为进一步确定MEF2C基因功能提供试验基础。  相似文献

本研究采用PCR-SSCP和DNA直接测序技术相结合来检测兴义鸭CYP7A1基因外显子3的SNP位点,分析其与血清生化指标的关联性。结果表明,检测到3种基因型AA、AB和BB,2个等位基因A和B,AA和A分别为优势基因型和等位基因,频率分别为0.882和0.921,序列比对发现5个SNP位点G744A、G783A、T858C、C951G和G960A,均为同义突变。关联分析显示,BB基因型个体的血清白蛋白、球蛋白和总蛋白显著高于AA基因型(P0.05),AA基因型白球比值显著高于AB型(P0.05),推测CYP7A1基因外显子3的多态可能对兴义鸭血清蛋白有一定影响,BB基因型可能是血清蛋白组成的有利基因型,等位基因B可能是有利等位基因。  相似文献

为了解三穗鸭肌细胞增强因子(myocyte enhancer factor 2A,MEF2A)SNPs与屠宰性状的相关性,本研究以60只三穗鸭为研究对象,采用PCR-SSCP方法结合PCR产物直接测序技术对三穗鸭MEF2A基因多态性进行检测,并进行SNPs与屠宰性状各指标的相关分析。结果表明,在MEF2A基因中共发现了2个SNPs:第11外显子的g.47915G>A位点和g.47918G>A位点,g.47915G>A位点发生的G/A突变使密码子由GAA变为AAA,翻译出的氨基酸由谷氨酸变成赖氨酸,g.47918G>A位点的G/A突变引起的密码子由GAT变成AAT,翻译出的氨基酸由天冬氨酸变成天冬酰胺。SNPs与屠宰性状的关联性分析表明,g.47915G>A和g.47918G>A位点影响全净膛率。这一结果揭示了MEF2A基因的多态性对三穗鸭屠体性状具有重要的影响。  相似文献

早胜牛钙蛋白酶Ⅰ基因(CAPN1)多态性分析   总被引:1,自引:0,他引:1  
试验旨在研究CAPN1基因在早胜牛群体中的遗传变异,为进一步寻找与早胜牛肉质性状相关的分子标记奠定基础。采用DNA混合池测序法对早胜牛320个样本CAPN1基因所有外显子的SNPs位点进行筛选,同时进行序列分析,并利用测序图中SNP位点等位基因峰高的比值估算各等位基因的频率。结果表明:在早胜牛CAPN1基因外显子区共筛选到7个多态性位点G3717A、A3854G、C5709G、C6004T、G11058A、G15299A和G15682A,其中G3717A和G15299A为同义突变,其余5个SNPs位点为错义突变。该研究结果将为进一步研究早胜牛肉质性状相关候选基因提供理论依据。  相似文献

【目的】分析叉头框家族转录因子A2(forkhead box A2,FOXA2)基因第3外显子多态位点在地方猪种中的遗传多样性及其与松辽黑猪繁殖性状的相关性。【方法】采集90头健康经产松辽黑猪母猪耳组织,采用直接测序法检测FOXA2基因第3外显子SNP位点,利用SPSS 26.0软件分析FOXA2基因第3外显子SNP位点多态性(基因型频率、基因频率、遗传纯合度、遗传杂合度、有效等位基因数、多态信息含量)及其与松辽黑猪母猪繁殖性状(总产仔数、产活仔数、乳头数以及仔猪初生重、3周龄重、断奶重、断奶仔猪数)的相关性。【结果】松辽黑猪FOXA2基因第3外显子共检测出3个SNPs位点,1 337 bp处SNP位点(C1337A)上发现CC、CA和AA 3种基因型;1 343 bp处SNP位点(C1343G)上发现CC、CG和GG 3种基因型;1 626 bp处SNP位点(C1626T)上发现CC、CT和TT 3种基因型。卡方适合性检验表明,松辽黑猪FOXA2基因第3外显子的C1337A、C1626T位点符合哈迪-温伯格平衡状态(P>0.05),C1343G位点偏离哈迪-温伯格平衡状态(P&...  相似文献

为研究CD36基因在早胜牛群体中的遗传变异及其与肉质性状的关联性,本试验采用DNA混合池测序技术检测了320个样本的SNPs位点,并估算了SNPs位点的等位基因频率。结果表明,外显子区域有2个多态位点,分别是外显子8的79201bp处A缺失和外显子12的86351bp处T→A突变。内含子区域有3个多态位点,分别是内含子8的79309bp处C→A突变和内含子12的86446bp处C→A突变、86447bp处G→A突变,其余区域未检测到碱基突变。外显子8的A缺失可能引起氨基酸序列发生改变。外显子12的T→A突变属同义突变,未引起氨基酸改变。后续我们将对CD36基因多态性与早胜牛肉质性能相关性进行分析,以期寻找到与肉质性能关联的SNPs位点,为研究提高早胜牛肉质性能的工作提供参考。  相似文献

试验旨在研究白细胞表面抗原DRB1基因外显子3多态性与哈萨克羊布鲁氏菌病易感性的相关性。运用混合DNA池结合PCR产物直接测序方法,对哈萨克羊DRB1基因外显子3进行多态性分析,经卡方检验分析每个SNP位点的等位基因频率、基因型频率及其多态性与布鲁氏菌病易感性的相关性,利用生物信息学分析软件对PCR扩增所获序列进行RNA二级结构及蛋白质的二级结构和抗原表位分析。结果表明,在282bp的外显子3序列中共检测到7个SNPs,分别为:T10C、C119T(Trp→Arg)、G215C(Gln→Glu)、A238G、T245G(Ser→Ala)、G256A、C259T,这些位点在病例组和对照组之间的等位基因频率及各基因型间不存在显著性差异(P0.05);进一步分析发现,各突变位点均引起RNA二级结构和最小自由能的改变,各错义突变位点均未引起蛋白质二级结构和抗原表位的改变。由此得出,DRB1基因外显子3的7个SNPs位点(T10C、C119T、G215C、A238G、T245G、G256A和C259T)与哈萨克羊布鲁氏菌病易感性无相关性。  相似文献

为探索ADAMTS1基因在绵羊繁殖中的作用,本研究以贵州半细毛羊为试验对象构建DNA池,分别设计引物扩增第1、2、5-7和9外显子,研究ADAMTS1基因的遗传多态性.结果显示,ADAMTS1基因第6外显子有1个SNP (A255C),第9外显子有4个SNPs (A127G、G243T、G303T、A401G).生物信息学分析表明,SNPs位点对ADAMTS1基因RNA二级结构和蛋白结构均有一定影响.ADAMTS1基因可能影响贵州半细毛羊的繁殖性状.  相似文献

为探究寡腺苷酸合成酶1(oligoadenylate synthase 1,OAS1)基因多态性与松辽黑猪繁殖性状的关联性,试验选取130头松辽黑猪母猪为研究对象,利用Sanger直接测序法测序查找OAS1基因外显子1~8的SNP位点,使用SPSS 19.0软件分析OAS1基因SNP位点与松辽黑猪繁殖性状的关联性。结果显示,在松辽黑猪OAS1基因外显子2、3和6上共检测到33个突变位点;其中在外显子2的110 bp处存在1个SNP位点(G110C),存在3种基因型:GG、GC和CC;在外显子3的176 bp处存在1个SNP位点(C176T),存在3种基因型:CC、CT和TT;在外显子6的145 bp处存在1个SNP位点(C145T),存在3种基因型:CC、CA和AA;在166 bp处存在1个SNP位点(G166A),存在3种基因型:GG、GA和AA;在206 bp处存在1个SNP位点(A206G),存在3种基因型:AA、AG和GG。卡方适合性检验结果显示,松辽黑猪OAS1基因G110C突变位点符合Hardy-Weinberg平衡状态,C176T、C145A、G166A和G206A位点均偏离Hardy-Weinberg平衡状态。群体遗传参数分析结果显示,各SNPs位点遗传杂合度均位于中等水平,为中度多态(0.25<PIC<0.5)。关联分析结果发现,G110C位点GC基因型个体总产仔数、产活仔数和断奶仔猪数均显著高于GG基因型个体(P<0.05);C176T位点CT基因型个体断奶仔猪数显著高于CC基因型个体(P<0.05);C145T位点CC基因型个体总产仔数和产活仔数均显著高于AA基因型个体(P<0.05);G166A位点GA基因型个体断奶仔猪数显著高于GG基因型个体(P<0.05);A206G位点GG基因型个体总产仔数和产活仔数显著高于AA基因型个体(P<0.05)。结果表明,OAS1基因外显子区存在突变位点,对松辽黑猪部分繁殖性状有显著性影响。  相似文献

To understand the relationship between single nucleotide polymorphism sites (SNPs) of myocyte enhancer factor 2A (MEF2A) gene and slaughter traits in Sansui duck, a total of 60 individuals of Sansui ducks were selected to investigate in this study, direct sequencing of PCR and PCR-SSCP methods were used on single nucleotide polymorphisms of MEF2A gene, and genetic effects of its on slaughter traits were analyzed.The results showed that two SNPs which include g.47915G>A and g.47918G>A of exon 11 were found in MEF2A gene, and the G/A mutation in the g.47915G>A SNP resulted in the change of codon from GAA to AAA, and the coding amino acid from Glu to Lys, and the G/A mutation in the g.47918G>A SNP resulted in the change of codon from GAT to AAT, and the coding amino acid from Asp to Asn.The result of association of SNPs with slaughter traits showed various results were as follows:g.47915G>A and g.47918G>A had affected the eviscerated percentage.This result revealed that the polymorphism of MEF2A gene had basilic influence on slaughter traits.  相似文献

Our previous cDNA microarray study showed that the growth hormone (GH) gene may involve in the duck egg formation process. The purpose of this study was to investigate the relationship between GH genotypes of single nucleotide polymorphisms (SNPs) and reproductive traits of Tsaiya ducks. Primer pairs for the coding region in the GH were designed based on the duck genomic sequence. Polymorphisms were detected by polymerase chain reaction (PCR)-single strand polymorphism (SSCP) and were verified by DNA sequencing. Nineteen SNPs were identified in the duck GH gene, of which three coding SNPs (C3169T, C3700T and C5058G) were genotyped to investigate the associations with reproductive traits. The results showed that each SNP was associated with at least one duck fertility-related trait (p < 0.05). Haplotypes constructed on these three SNPs were associated with fertility rate (FR) and maximum duration of fertility (MDF) (p < 0.05). In particular, diplotype H1H1 was dominant for FR and MDF. This suggested that GH gene polymorphisms are associated with duck fertility-related traits. The SNPs in this gene may be used as potential markers for marker-assisted selection.  相似文献

This study was aimed to analyze the association of myostatin (MSTN) gene polymorphisms with meat quality traits in Chishui Black-bone chicken. The DNA pooling was constructed and the direct sequencing technology of PCR product was used to detect the single nucleotide polymorphism (SNP) of MSTN gene. The results showed that C324T were detected in exon 1 of MSTN gene in Chishui Black-bone chicken. However, there was not found SNP site in exon 2 and exon 3 of MSTN gene. The C324T site was devised into three genotypes of CC, CT and TT. The association relationships between the genotype of MSTN gene and meat quality traits revealed that chest muscle pH and leg muscle pH value of Chishui Black-bone chicken with CT genotype were significantly higher than that of CC and TT genotypes (P<0.05), the shear force of CT genotype was significantly higher than that of TT genotype (P<0.05), there was no significant difference in other meat quality traits (P>0.05). This results suggested that the pH value and shear force of MSTN gene had a larger genetic effect in Black-bone chicken,MSTN gene was one of genes which could control the preliminary inference, might affect the pH value and shear force or with the main gene chain in Black-bone chicken, and could be used as pH value and shear force of molecular markers of Black-bone chicken.  相似文献

To explore the influence of Gaopo pig myostatin (MSTN) gene polymorphism on meat quanlity traits (moisture,crude fat,crude protein,crude ash, loin muscle area, marbling score, pH value, color of meat,tenderness,drip loss, water loss rate), MSTN gene was chosen as a candidate gene for research meat quality traits in this study. 50 Gaopo pigs at 10 months old were selected,the single nucleotide polymorphism (SNP) of three exons of MSTN gene in Gaopo pig were detected by direct sequencing of PCR amplification products, and the correlation between MSTN gene SNP and meat quality traits was analyzed using SPSS 20.0 software. The results showed that a C/T mutation had been found at 63 bp of MSTN gene exon 3,the site was silent mutation, and did not caused changes of encoding amino acid. Genotype analysis showed only three samples had mutations in the site, CC genotype was the dominant type and allele C was the dominant gene, but there was no TT homozygous genotype. The test of Hardy-Weinberg equilibrium showed the research group obeyed the genetic equilibrium (P>0.05). Population genetic parameter analysis showed the heterozygosity (He) of C63T was lower, and had a low variation in Gaopo pig population. Polymorphism information content (PIC) analysis showed that it was a low polymorphic loci (PIC<0.25), and the site could provide a little genetic imformation. There was no significant difference between the different genotypes and meat quality traits (P>0.05).Given the above, the polymorphism of MSTNgene exon was found in Gaopo pig, with less variation and relatively conservative.It's needed to expand the number of sample for further explore whether the site could be considered as genetic markers for meat quality traits of Gaopo pig or not.  相似文献

为了探讨肌肉生长抑制素(myostatin,MSTN)基因的多态性对高坡猪肉质性状(水分、粗脂肪、粗蛋白质、粗灰分、眼肌面积、大理石纹、pH、肉色、嫩度、滴水损失、失水率)的影响,试验以MSTN基因作为肉质性状候选基因,以10月龄的50头高坡猪为研究对象,采用PCR扩增产物直接测序的方法对高坡猪MSTN基因3个外显子的单核苷酸多态性(SNP)进行研究,运用SPSS 20.0软件分析MSTN基因SNP与肉质性状的关联性。结果表明,仅在高坡猪MSTN基因第3外显子63 bp处检测到1个C/T突变位点,该突变为同义突变,未引起编码氨基酸的改变。基因型分析发现,仅有3个个体在该位点发生突变,CC基因型为优势基因型,C为优势等位基因,无TT纯合基因型。经χ2适合性检验分析,该SNP在研究群体中处于Hardy-Weinberg平衡状态(P>0.05)。群体遗传参数分析发现,C63T标记位点的杂合度(He)相对较低,表明其在高坡猪群体中的变异较小;就多态信息含量(PIC)而言,该位点属于低度多态(PIC<0.25),说明该遗传标记能够提供少量的遗传信息。将该位点的不同基因型与肉质性状指标进行关联分析表明,所有肉质性状指标在不同基因型间差异均不显著(P>0.05)。综上所述,MSTN基因外显子在高坡猪中存在多态性,但变异较少,相对保守,能否作为高坡猪肉质性状的遗传标记有待于扩大样本数量进一步研究。  相似文献

本研究旨在探讨肌肉生长抑制素(myostatin,MSTN)基因多态性及其与赤水乌骨鸡肉质的关联性。采用DNA池、PCR产物直接测序方法对赤水乌骨鸡MSTN基因的外显子进行多态性检测,结果发现,在赤水乌骨鸡MSTN基因的外显子1中检测到1个SNP位点C324T,表现为3种基因型,分别命名为CC、CT和TT,而在MSTN基因外显子2、3上没有发现多态位点。对基因型与肉质性状关联分析显示,CT基因型胸肌、腿肌pH均显著高于CC和TT基因型(P<0.05),CT基因型剪切力显著高于TT基因型(P<0.05),其他肉质性状在不同基因型间差异均不显著(P>0.05)。此结果提示,MSTN基因对赤水乌骨鸡pH和剪切力具有较大的遗传效应,初步推断 MSTN是控制这一性状的众多基因之一,且可能是影响赤水乌骨鸡pH和剪切力的一个主效基因或与主效基因相连锁,可作为选育赤水乌骨鸡pH和剪切力的分子标记,用于标记辅助选择意义重大。  相似文献

利用PCR-SSCP技术对萨福克、陶赛特、得克塞尔及滩羊4个绵羊品种358个个体Leptin基因等2、3外显子进行多态性分析,共检测到7个SNPs,其中新发现5个SNPs。测序结果表明,在外显子2上无突变。内含子2上存在A99G、G115A、C150T、C171T位点。外显子3上,存在G271A;C316A;G387T位点。外显子3上的SNPs使编码的氨基酸发生变化。统计分析表明A99G、C150T和A99G+C150T位点与生长发育性状存在相关性。在A99G位点,Aa基因型初生质量、日增质量、体高、胸围和尻宽指标上均高于AA基因型,初生质量、日增质量和体高指标差异显著(P〈0.05),胸围和尻宽指标差异极显著(P〈0.01)。C150T和A99G+C150T位点结果一致,突变基因型日增重、体高、体长、胸围和尻宽指标均高于野生基因型,差异显著(P〈0.05)。  相似文献

【目的】 研究中国草原红牛丙酮酸脱氢酶激酶4(pyruvate dehydrogenase kinase 4,PDK4)基因多态性与肉质性状的关系。【方法】 挑选120头中国草原红牛为研究对象,采用Sanger测序法检测PDK4基因第1~11外显子的单核苷酸多态性(SNP)位点,分析SNP位点的基因型频率、基因频率及群体遗传参数等。利用SPSS 21.0软件对中国草原红牛PDK4基因SNP位点多态性与肉质性状(熟肉率、肉嫩度、失水率、pH、滴水损失、肌内脂肪含量、初水分含量、蛋白质含量)进行关联分析。【结果】 在中国草原红牛PDK4基因外显子8和外显子11上共检测到3个SNPs;PDK4基因第8外显子57 bp处存在1个SNP位点(G57C),且引起编码氨基酸的改变,存在GG、GC和CC 3种基因型;PDK4基因第11外显子在330和389 bp处存在2个SNPs位点(G330T和C389T),在G330T位点上存在GG、GT和TT 3种基因型;在C389T位点上存在CC、CT和TT 3种基因型。卡方适合性检验结果显示,中国草原红牛第8外显子G57C位点偏离Hardy-Weinberg平衡状态(P<0.05),第11外显子的G330T和C398T符合Hardy-Weinberg平衡状态(P>0.05);群体遗传参数分析发现,第8外显子G57C突变位点属于低度多态性位点(PIC<0.25),等位基因数为1.1429,表明其在中国草原红牛中的变异较小;第11外显子G330T和C398T属于中度多态性位点(0.25<PIC<0.5),等位基因数分别为1.6431和1.6447,说明该遗传标记能够提供遗传信息。关联分析结果表明,PDK4基因第8外显子中G57C位点GG和CC基因型个体肌内脂肪含量显著高于GC基因型(P<0.05);第11外显子G330T位点GG基因型滴水损失和初水分量显著高于GT基因型(P<0.05);GG基因型肉嫩度显著高于TT基因型(P<0.05);GT基因型肌内脂肪含量显著高于TT基因型(P<0.05),C389T处CT基因型失水率显著低于TT基因型(P<0.05)。【结论】 PDK4基因多态性与中国草原红牛肉质性状相关,可作为肉质性状的候选基因。  相似文献
