The potential for gene flow between Camelina sativa, a promising edible and industrial oil crop in Canada, and its wild North American relatives C. alyssum, C. microcarpa and C. rumelica subsp. rumelica, was investigated. Sequence analysis of the nuclear ribosomal internal transcribed spacer region (ITS) was used to differentiate Camelina species and/or accession‐specific nucleotide markers and identify interspecific F1 hybrids. ITS analysis identified hybrids in progeny of C. alyssum × C. sativa, C. microcarpa accession 36010 × C. sativa and C. sativa × C. rumelica subsp. rumelica. Seed set on C. alyssum × C. sativa F1 and F2 progeny was similar to the parents; few seeds were produced on hybrid progeny of C. microcarpa accession 36010 × C. sativa and C. sativa × C. rumelica subsp. rumelica. The study provided evidence that should the species have sympatric distributions and overlapping flowering periods, gene flow between C. sativa and its wild North American relatives is possible and that it would most likely occur with C. alyssum given the high fertility of the F1 hybrids recovered.  相似文献

In order to introduce the Tomato Spotted Wilt Virus (TSWV) resistance from Nicotiana alata into Nicotiana tabacum, a cytoplasmic male sterility (CMS) line of N. tabacum (N. tabacum L. cv. (gla.) S ‘K326’), was successfully crossed with N. alata. Despite a high DNA content variability, F1 hybrids could be classified in two subgroups, a major one encompassing fertile hybrids morphologically similar to their tobacco maternal parent but TSWV sensitive, and a minor one displaying sterile hybrids showing an intermediate phenotype and TSWV resistant. In order to elucidate the unexpected fertility recovery of the fertile F1 plants, some N. alata fertility restoration ppr genes were cloned and were shown to be differentially expressed between parental lineages as well as between both F1 subgroups, suggesting that N. alata contains fertility restoring allele able to overcome the CMS of N. tabacum.  相似文献

It is crucial to develop a rapid technique for identifying sexuality in the seedling stage of persimmon (Diospyros kaki Thunb.), and the elimination of male progeny has been regarded as an important strategy for enhancing breeding efficiency. In this study, phenotype characterization and genotyping of the male‐linked OGI marker were carried out using 205 accessions, including persimmon cultivars, F1 progeny and nine related Diospyros species. All persimmon cultivars displayed consistent results regarding OGI amplification and sex phenotype. A total of 143 F1 progeny were derived from 11 crosses, among which 95 individuals had flowered. In the flowering full‐sib families, the amplification of the OGI marker in agreement with the sex phenotype was obtained in 85 plants (89.5%). The segregation of OGI in ‘Huashi 1’ × ‘Luotian Tianshi’ and ‘Huashi 1’ × Male 3 F1 populations fit a 1 : 1 ratio. Furthermore, high OGI transferability was observed in nine related species. Overall, the results indicated that the OGI locus could be used to distinguish male from female persimmon plants at an early stage.  相似文献

Cultivation of the same varieties of mungbean and blackgram across different seasons and locations is constrained by their photo‐ and thermo‐sensitive behaviour. Developing insensitive genotypes, which can fit well across all seasons, requires robust donors which would provide genes imparting this trait. This study was undertaken to identify such donors in the Vigna species. Forty‐eight accessions belonging to 13 Vigna species and eight released cultivars were evaluated under natural field conditions. Among these, two accessions, viz. V. umbellata (IC251442) and V. glabrescens (IC251372) were found photo‐ and thermo‐insensitive as these were able to flower and set pods at temperatures as high as 43.9°C and as low as 2.7°C. Pollen viability studies indicated viable pollen (>75% at 2.7°C and >85% at 41.9°C) and normal pollen tube growth at both the extremes of temperature. The identified V. glabrescens accession has long, constricted pods and dark green, mottled seeds while V. umbellata has smooth, curved pods and shining, oval, large seeds. Both these accessions can be utilized in developing photo–thermo insensitive genotypes in cultivated Vigna species.  相似文献

The Fragaria genus has become relevant due to worldwide economic importance of the octoploid hybrid F. × ananassa. Interspecific hybridizations are widely employed in breeding programmes to introduce useful characteristics from wild species to cultivated varieties, although many reproductive barriers prevent gene flow between species. Hybrid genotypes that can act as a bridge between related wild species and cultivated strawberry could be useful to overcome those barriers. The aim of this work was to obtain interspecific hybrids of Fragaria and to analyse possible reproductive barriers. In the Banco de Germoplasma de Frutilla‐UNT, crossings were carried out among wild species of Fragaria with different ploidy levels and cultivated strawberry. To confirm hybrid condition of genotypes obtained, histological techniques and microsatellite markers were used. When wild species were crossed with cultivated strawberry in both directions, there was low production of achenes, while in crosses between wild species of Fragaria genus, achene production was very high. The percentage of germination of achenes was high when crossed species had the same ploidy level, and very low or null when they presented different ploidy levels. Although pre‐zygotic incompatibility associated to the degree of domestication and postzygotic barriers related to different ploidy levels of the progenitors were detected, new hybrid genotypes of Fragaria were obtained. These new hybrids could be used as “bridge species” in breeding programmes, since preliminary results showed no incompatibility barriers when they were crossed with Duchesnea indica.  相似文献

We developed a new disomic addition line M11028‐1‐1‐5 (2n = 44 = 21” + 1”) from a cross between wheat cv. ‘7182’ and octoploid Tritileymus M47 (2= 8x = 56, AABBDDN sNs ). Cytological observations demonstrated that M11028‐1‐1‐5 contained 44 chromosomes and formed 22 bivalents during meiotic metaphase I. The genomic in situ hybridization (GISH) investigations showed this line contained 42 wheat chromosomes and a pair of L. mollis chromosomes. SSR, EST and PCR‐based landmark unique gene (PLUG) markers were screened to determine the homoeologous relationships of the introduced L. mollis chromosomes in wheat background. Nine markers, i.e. Xwmc256, Xgpw312, Swes123, CD452568, BF483643, BQ169205, TNAC1748, TNAC1751 and TNAC1752, all of which were located on the homoeologous group 6 chromosomes of common wheat, amplified bands unique to L. mollis in M11028‐1‐1‐5. Gliadin analysis also confirmed that the added chromosomes in M11028‐1‐1‐5 were correlated with the sixth group chromosome. This indicated that M11028‐1‐1‐5 contained a pair of introduced L. mollis chromosome belonging to homoeologous group 6, which we designated it as Lm#6 Ns disomic addition line. This is the first report of a common wheat–L. mollis disomic addition line.  相似文献

Interspecific hybridization is an important approach to broaden the genetic base and create novel plant forms in breeding programs. However, interspecific hybridization in Ipomoea is very difficult due to the cross incompatibility. Here we report two novel interspecific F1 hybrids between I. batatas (L.) Lam. (2n = 6x = 90) and two wild species, I. grandifolia (2n = 2x = 30) and I. purpurea (2n = 2x = 30). Hybridization was stimulated by applying plant growth hormones. Morphological, molecular and cytological tests were conducted to confirm their hybridity. We found that the two hybrids were quite distinctive in leaf color and morphology, and yielded intermediate sizes of storage roots compared to their respective parents. Inter-simple sequence repeat analysis showed that the unique DNA bands from the wild parents could be detected in these two hybrids. The cluster analysis also showed that the two F1 hybrids were closer to I. batatas in phylogeny relationship. The number of chromosomes of the two hybrids was both 60, indicating that the hybrids were tetraploid. The meiotic configuration analysis of the H1 of I. batatas × I. grandifolia revealed the occurrence of 17.58I + 14.28II + 1.36III + 2.48IV at metaphase I in average chromosome association per pollen mother cells (PMCs), 4.26I + 18.32II + 2.56III + 3.12IV was average meiotic configuration in the H2 of I. batatas × I. purpurea. Both hybrids appeared to be polyads and multi-microcytes at tetrad phase and differed in their pollen fertility.  相似文献

To transfer new traits into the gene pool of garden dahlias (Dahlia variabilis), crosses between garden dahlias (2n = 64) and the epiphytic species Dahlia macdougallii (2n = 32) from the section Epiphytum were conducted. Six hybrid plants were obtained. The hybrid status was verified using three SSR markers. In addition, flow cytometry was performed to determine the genome size of the hybrids and to show that the hybrids were hexaploid as expected from crosses between tetraploids and octoploids. The open pollinated progeny of the hybrids produced four progeny with octoploid genomes. The hybrids exhibited indeterminate vegetative growth and the formation of flowers from axillary buds, similar to the father D. macdougallii. This result is of interest for breeding new varieties of dahlia with traits that are not present in the current gene pool.  相似文献

Wild Lens taxa are invaluable sources of useful traits for broadening genetic base of cultivated lentil. Nine inter‐sub‐specific and interspecific crosses were made successfully between cultivated (Lens culinaris ssp. culinaris) and wild lentils (L. culinaris ssp. orientalis, odemensis, lamottei and ervoides). The effect of species groups, day length and temperature on crossability in lentils was evident under normal winter sowing in New Delhi and in summer Himalayan nursery at Sangla in Himachal Pradesh, India, although pollen fertility assessed in all the cross‐combinations showed no significant variation. True hybridity of nine inter‐sub‐specific and interspecific crosses was confirmed through morphological and molecular (ISSR) markers, in which three of 120 primers could confirm the hybridity of all the crosses. All cross‐combinations were also studied for important quantitative traits related to yield. The range, mean and coefficient of variation were estimated in parental lines, F1 and F2 generations to determine the extent of variability generated in cultivated lentils through the introgression of genes from wild L. taxa. A high level of heterosis was observed in F1 crosses for important traits studied. Substantially higher variations for seed yield and its attributing traits were exhibited in F2 generations indicating transgressive segregation. The results of the present investigation revealed that wild L. taxa can be successfully exploited for lentil improvement programmes, and the variations generated could be easily utilized for broadening the genetic base of cultivated lentil gene pool for improving the yield as well as wider adaptation.  相似文献

To select superior seed parents for vegetable hybrid seed production, we conducted interspecific crosses between male sterile Brassica juncea (2n = 36, AABB) and eight inbred lines of Brassica rapa (2n = 20, AA). Alloplasmic lines of B. rapa with the cytoplasm of B. juncea were developed from B. juncea × B. rapa hybrids by repeated backcrossing using B. rapa as the recurrent male parent until the BC3 generation. Seed fertility, male sterility and chlorophyll content were investigated in these plants cultivated under four different temperature conditions (5, 10, 12 and 20°C). At 10°C, the alloplasmic lines of B. rapa with the cytoplasm of B. juncea were male sterile with partly chlorotic leaves. The alloplasmic B. rapa had lower chlorophyll a, chlorophyll b and carotenoid contents than those of the original B. rapa. The leaves recovered from chlorosis when the plants were cultivated at 20°C. An alloplasmic line of B. rapa (A6) is available as a seed parent for vegetable hybrid seed production and contributes seed fertility, slight chlorosis and stable male sterility.  相似文献

Development of wheat–alien translocation lines has facilitated practical utilization of alien species in wheat improvement. The production of a compensating Triticum aestivumThinopyrum bessarabicum whole‐arm Robertsonian translocation (RobT) involving chromosomes 6D of wheat and 6Eb of Th. bessarabicum (2n = 2x = 14, EbEb) through the mechanism of centric breakage–fusion is reported here. An F2 population was derived from plants double‐monosomic for chromosome 6D and 6Eb from crosses between a DS6Eb(6D) substitution line and bread wheat cultivar ‘Roushan’ (2n = 6x = 42, AABBDD) as female parent. Eighty F2 genotypes (L1–L80) were screened for chromosome composition. Three PCR‐based Landmark Unique Gene (PLUG) markers specific to chromosomes 6D and 6Eb were used for screening the F2 plants. One plant with a T6EbS.6DL centric fusion (RobT) was identified. A homozygous translocation line with full fertility was recovered among F3 families and verified with genomic in situ hybridization (GISH). Grain micronutrient analysis showed that the DS6Eb(6D) substitution line and T6EbS.6DL stock have higher Fe and Zn contents than the recipient wheat cultivar ‘Roushan’.  相似文献

Trifolium occidentale is a diploid wild relative of T. repens (white clover) with adaptation to dry, saline coastal habitats. Transfer of drought and salt‐tolerant adaptive traits to white clover could be potentially valuable if interspecific hybridization can be achieved efficiently and leads to fertile hybrid populations. To achieve hybridization, 4x plants of T. occidentale were generated. Efficient techniques for generation of 4x plants and their identification using dry pollen shape are described. Interspecific 4x F1 plants were achieved without embryo rescue. F2 populations and first backcross hybrids to white clover were also efficiently achieved. Although male and female fertility were lower than in white clover, they were adequate to produce large amounts of seed from small numbers of inflorescences. Thus, early generation interspecific hybrid prebreeding populations can be readily developed, opening the way for transfer of traits from T. occidentale to white clover.  相似文献

Brassica napus is an important oil species with short history and narrow genetic background. Interspecific hybrids from crosses between B. oleracea and different B. rapa were obtained. We found the hybrids with white petal resembling B. oleracea, the flavonoid and phenolic content decreased in hybrids, agreeing with the expressional changes of flavonoid biosynthesis genes. Seed coat of hybrids resembled diploid parents, or partly resembled to each parent with a clear outline. The palisade layer in hybrids was thicker than parents, with similar pigment accumulation as B. oleracea but more than B. rapa. Differentially sized protein bodies (PBs) were found in hybrids. The radical and inner cotyledon of all hybrids were identified with larger but less PBs than parents. The average size of PBs in outer cotyledon of resynthesized B. napus was also larger than parents, but the number of PBs was not significantly reduced. The phenotypic and seed structural variations after polyploidization of B. napus would be interesting for genetic broadening and breeding of rapeseed.  相似文献

Sexual hybridization is an important generator of biodiversity and a powerful breeding tool. Hybridization can also overcome ploidy barriers when it involves 2n gametes, as in the case of unilateral sexual polyploidization (USP) that has been utilized in several crops, among which alfalfa. This research was aimed at gaining insights into the effects of USP on genome methylation and on phenotypic traits in alfalfa, an important forage species. The Methylation-Sensitive Amplified Polymorphism technique was used to estimate the cytosine methylation changes occurring in a tetraploid (2n = 4x = 32) USP progeny from crosses between a diploid Medicago sativa subsp. falcata genotype that produces 2n eggs and a cultivated tetraploid Medicago sativa subsp. sativa variety. De novo methylation or demethylation in the USP progeny were observed for 13% of the detected genomic sites, indicating that methylation changes can be relevant. USP plants showed larger surface area of the leaf epidermis cells than both parents, but this did not result in larger leaf size or higher plant biomass. They displayed significant higher ovule sterility than the tetraploid parent, but normal fertility was observed in crosses with unrelated male testers. We conclude that hybridization and sexual polyploidization resulted in novel variation in terms of remodeling of the methylation landscape as well as changes in phenotypic traits in alfalfa.  相似文献

Portugal has a diverse common bean germplasm, which is still grown in farmers' fields. In this work, we searched for resistance to rust (Uromyces appendiculatus) and powdery mildew (Erysiphe diffusa) in a representative collection of the Portuguese common bean germplasm. Despite many accessions depicting intermediate levels of resistance when compared to the susceptible check, 24 and 13 accessions showed low levels of infection, in spite of a compatible interaction (disease severity (DS) values lower than 5% and infection type (IT) of 4), to rust and powdery mildew, respectively, indicative of partial resistance. Moreover, a resistant reaction was observed in 11 accessions when inoculated with powdery mildew (IT = 0–1) and in additional 11 accessions (one in common) when inoculated with rust (IT = 0–2). The levels of resistance found in this report anticipate great potential of the Portuguese national germplasm, recently reported as genetically closer to the Andean common bean gene pool, for disease resistance breeding of this important crop.  相似文献

Brachiaria humidicola is a tropical grass that grows in seasonally swampy grasslands in Africa. In Brazil, two apomictic cultivars (2n = 54) of this species are widely used as pastures in poorly drained soils. The recent discovery of a sexual polyploid accession (2n = 36) in the germplasm collection at the Embrapa Beef Cattle Research Center allowed intraspecific hybridization with the objective of broadening the genetic variability and selection of superior genotypes in this species. Hybridization, however, depends on accessions with the same ploidy level. Cytological analyses of 55 accessions revealed that 19 apomictic accessions also presented 2n = 36 chromosomes. Chromosome pairing in hexavalent association at diakinesis and metaphase I suggested that the basic chromosome number for this species is x = 6. Cytological analysis revealed abnormalities in variable frequencies in the meiosis of these hexaploid (2n = 6x = 36) accessions. The most common were those related to irregular chromosome segregation which led to unbalanced gamete formation, but chromosome stickiness was also recorded. These results clearly demonstrate the value of cytogenetics in the choice of genitors and for superior hybrids to be obtained in the breeding of this species. For that both the ploidy level and the frequency of abnormalities need to be considered, besides other favorable agronomic characteristics.  相似文献

Most of the hybrid seed in chilli are produced manually, but the use of male sterility (MS) can reduce the cost of hybrid seed production. MS‐12, a nuclear male‐sterile (NMS) line developed at Punjab Agricultural University, Ludhiana (India), has been utilized to develop commercial F1 hybrids. A recessive gene, designated as ms10, governs MS in MS‐12. Due to recessive gene control, development of new NMS lines incorporating ms10 gene is tedious and time‐consuming. We identified SSR markers AVRDC‐PP12 and AVRDC_MD997* linked to the ms10 gene. A total of 558 primer pairs were screened following bulked segregant analysis (BSA). Linkage analysis in 210 F2 plants indicated that the two SSR markers were linked to the ms10 gene and the marker AVRDC‐PP12 was closest to the gene at 7.2 cM distance. The marker was mapped to chromosome 1 at genome position 175 694 513 to 175 694 644. Until more closely linked markers are developed, the marker AVRDC‐PP12 would facilitate transfer of ms10 gene through marker‐assisted selection (MAS). Fine mapping would lead to cloning of the ms10 gene.  相似文献

Phenylpropenes play an important role in plant defense against animals and microorganisms, and in attracting pollinators and insects. We report the genetic inheritance of methyl chavicol and eugenol following a cross between the sweet basil varieties ‘Perrie’ and ‘Cardinal,’ eugenol and methyl chavicol chemotypes, respectively. Methyl chavicol was detected only in ‘Cardinal,’ accounting for more than 95% of the total phenylpropenes. Eugenol was most abundant in ‘Perrie,’ accounting for more than 99% of the total phenylpropenes. Eugenol, chavicol and methyl chavicol were detected in F1 hybrids at intermediate levels (10%–52%) without statistical differences (> .05) for any compound among the F1 progeny arising from the different crossed pairs. The F2 progeny segregated into three groups, 23%–25% to a eugenol chemotype, 23%–25% to a methyl chavicol chemotype, and the remaining (~50%) into an intermediate mixture of the two compounds. This distribution fitted a segregation ratio of 1:2:1 (χ2 = 1.71; = .4249), suggesting that the phenylpropene phenotype is regulated by a single bi‐allelic gene with incomplete dominance. A putative association with biosynthesis enzymes is discussed.  相似文献

Soybean breeders have not exploited the diversity of the 26 wild perennial species of the subgenus Glycine Willd. that are distantly related to soybean [Glycine max (L.) Merr.] and harbour useful genes. The objective of this study was to develop a methodology for introgressing cytoplasmic and genetic diversity from Glycine tomentella PI 441001 (2= 78) into cultivated soybean using ‘Dwight’ (2= 40) as the male parent. Immature seeds (19–21 days post‐pollination) were cultured in vitro to produce F1 plants (2= 59). Amphidiploid (2= 118) plants, induced by colchicine treatment, were vigorous and produced mature pods and seeds after backcrossing with ‘Dwight’. The BC1 plants (2= 79) produced mature seeds in crosses with ‘Dwight’. Chromosome numbers in BC2F1 plants ranged from 2= 41–50. From BC2F2 to BC3F1, the number of plants in parentheses with 2= 40 (275), 2= 41 (208), 2= 42 (80), 2= 43 (27), 2= 44 (12) and 2= 45 (3) were identified. Fertile lines were grown in the field during 2012 and 2013. This is the first report of the successful development of new alloplasmic soybean lines with cytoplasm from G. tomentella.  相似文献

An intergeneric hybrid between Brassica napus (AC, n = 19) and Raphanus raphanistrum (Rr, n = 9) was developed using the sequential ovary–ovule culture. Morphological, molecular, cytological studies and fluorescent genomic in situ hybridization were conducted to establish the hybridity and genomic relatedness. F1 hybrid plants were male sterile and morphologically intermediate between the two parents. Cytological analysis of the hybrid plants revealed 10II + 8I as the predominant meiotic configuration with the mean bivalent frequency of 8.83. A maximum of 13II were observed in 8.24% of the pollen mother cells (PMCs). GISH studies facilitated the identification of two allopairs, confirming homoeologous pairing between A/C and Rr genomes. This may have significant implications in terms of transgene flow and possible introgression into Rraphanistrum, which is a common canola weed in Canada and Australia. The intergeneric hybrid Bnapus × Rraphanistrum so produced also has the potential to be used as a bridging species for the transfer of desirable genes to amphiploid crop Brassica species carrying A/C genomes.  相似文献
