ABSTRACT

Aims To compare wound healing 7 days after cautery disbudding of dairy calves treated immediately after disbudding with a traditional antimicrobial spray (OXY) or with a topical anaesthetic gel containing cetrimide, adrenaline and two local anaesthetics, lignocaine and bupivacaine (TA).

Method Eighty-one female dairy calves between 6–8 weeks of age were disbudded using a standard cautery disbudding protocol (sedation, cornual block, and analgesia), with complete removal of the horn bud. After disbudding, the wound on the right and left horn buds within each animal were randomly allocated to receive either OXY or TA. One week after disbudding, wounds were visually assessed for the presence of exudate, necrotic tissue, crust, or granulation tissue which were each assigned a grade from 1–3, where 1?=?no evidence, 2?=?moderate presence; and 3?=?marked presence.

Results At 7 days after disbudding, the prevalence of wounds with exudate or necrotic tissue was very low, and independent of treatment. The odds of a disbudding wound treated with TA being scored as having granulation tissue and was 5.2 (95% CI?=?1.72–15.7) times that of a wound treated with OXY. Conversely, the odds of wounds treated with TA being scored as having crusts was 0.18 (95% CI?=?0.06–0.57) that of a wound treated with OXY. No sign of infection was seen in any calves.

Conclusion and clinical relevance The greater prevalence of granulation tissue and reduction in crusting in disbudding wounds treated with topical anaesthetic gel compared to those treated with the antimicrobial spray suggests that use of the topical anaesthetic gel may speed wound healing compared to using antimicrobial spray. This should be further tested in a larger study undertaken over a longer period.  相似文献

AIM: To assess the effect of sedation and local anaesthesia (LA) at disbudding, and the addition of meloxicam or ketoprofen treatment, on weight gain in dairy calves following disbudding.

METHODS: Friesian-Jersey cross calves, from four dairy farms, were enrolled when 3–6 weeks old. All calves (n=271) were disbudded by veterinary personnel and randomly assigned to six groups: 136 were disbudded without sedation or LA, of which 31 received 20 mg meloxicam S/C and 75 received 150 mg ketoprofen I/M. A further 135 were disbudded with sedation (0.25 mg/kg xylazine I/M) and LA, of which 30 also received meloxicam and 75 received ketoprofen. Calves were weighed 3 days before, and 15 and 30 days after, disbudding (Day 0). Daily weight gain was analysed using mixed models and ANOVA.

RESULTS: Complete results were obtained from 263 calves. From Day ?3 to Day 15, the growth rate of calves disbudded without pain relief (0.53 (95% CI=0.47–0.60) kg/day) was less that of calves disbudded with some form of pain relief (0.65 (95% CI=0.62–0.68) kg/d; p=0.004). There was no difference between the effect of meloxicam or ketoprofen (p=1.00). An interaction between use of sedation and LA and additional non-steroidal anti-inflammatory drugs (NSAID) meant that NSAID treatment did not increase growth rates in calves disbudded with sedation and LA but did increase growth rates for calves disbudded without pain relief (p<0.05). From Day 16 to Day 30 there was no effect of NSAID treatment on growth rate, but calves receiving LA and sedation grew faster (0.74 (95% CI=0.69–0.80) kg/day) than calves disbudded without LA and sedation (0.66 (95% CI=0.61–0.71) kg/day; p=0.018). From Day ?3 to Day 30, calves disbudded with sedation and LA grew faster (0.71 (95%CI=0.64–0.77) kg/day) than calves disbudded without sedation and LA (0.60 (95% CI=0.55–0.65) kg/day; p=0.011). However, addition of NSAID to sedation and LA made no further difference to growth rates (p=0.69).

CONCLUSIONS: Dairy calves disbudded with no pain relief had slower growth rates than calves receiving pain relief. From Day 15 to 30 calves given no pain relief, or NSAID alone, grew more slowly than those receiving sedation and LA at disbudding. The addition of NSAID treatment to sedation and LA did not further increase growth rates.

CLINICAL RELEVANCE: This study adds to the evidence that pain management when disbudding is beneficial for calf productivity as well as calf welfare.  相似文献

ABSTRACT

Aims: To compare the effects of intrathecal anaesthesia using procaine and xylazine, with and without sedation with I/V xylazine and butorphanol, on sedation and cardiorespiratory measures in calves undergoing umbilical surgery.

Methods: Male dairy calves, aged <3 months, were recruited that had enlargement of the umbilical stalk which was abnormal when palpated. They were assigned to receive either intrathecal injection between the sixth lumbar and first sacral vertebrae of 4?mg/kg of 2% procaine and 0.2?mg/kg 2% xylazine, with I/V injection of 0.02?mg/kg xylazine and 0.1?mg/kg of butorphanol (IT?+?SED group; n?=?6), or the same intrathecal injection and I/V injection of 0.9% saline (IT group; n?=?7). Surgery to correct abnormalities was carried out with calves positioned in dorsal recumbency. Rescue analgesia with injections of 2% procaine around the surgical wound was administered when movements triggered by surgery were observed. Post-operative analgesia was provided using I/V 0.5?mg/kg meloxicam. Duration of surgery was recorded, as well as degree of sedation, heart rate, systolic (SAP), diastolic (DAP) and mean (MAP) arterial blood pressure during surgery.

Results: All anaesthetic and surgical procedures were successfully performed. Mean total duration of surgery was similar for the IT?+?SED and the IT groups (30.33 (SD 10.09) and 31.00 (SD 10.21) minutes, respectively) (p?=?0.92). All calves were at least mildly sedated from 5 minutes after injections to the end of the surgery. One calf in the IT?+?SED group and three calves in the IT group required rescue analgesia when the umbilicus was manipulated. Between 0 and 10–15 minutes after injection, decreases in mean heart rate, SAP, MAP and DAP were observed in both groups. Mean SAP was lower in the IT?+?SED than the IT group. Hypotension (MAP<60?mm Hg) was present in four calves from the IT?+?SED group and in one from the IT group.

Conclusions and clinical relevance: Intrathecal administration of 2% procaine and 2% xylazine allowed the successful completion of umbilical surgery, but 30% of calves needed rescue analgesia during surgery. Clinically, the addition of I/V sedation seemed to provide better analgesia than intrathecal block alone but resulted in greater hypotension.  相似文献

ObjectiveTo study the effects of oromucosal detomidine gel administered sublingually to calves prior to disbudding, and to compare its efficacy with intravenously (IV) administered detomidine.Study designRandomised, prospective clinical study.AnimalsTwenty dairy calves aged 12.4 ± 4.4days (mean ± SD), weight 50.5 ± 9.0 kg.MethodsDetomidine at 80 μg kg?1 was administered to ten calves sublingually (GEL) and at 30 μg kg?1 to ten control calves IV (V. jugularis). Meloxicam (0.5 mg kg?1) and local anaesthetic (lidocaine 3 mg kg?1) were administered before heat cauterization of horn buds. Heart rate (HR), body temperature and clinical sedation were monitored over 240 minutes. Blood was collected from the V. cephalica during the same period for drug concentration analysis. Pharmacokinetic variables were calculated from the plasma detomidine concentration-time data using non-compartmental methods. Statistical analyses compared routes of administration by Student’s t-test and linear mixed models as relevant.ResultsThe maximum plasma detomidine concentration after GEL was 2.1 ± 1.2 ng mL?1 (mean ±SD) and the time of maximum concentration was 66.0 ± 36.9 minutes. The bioavailability of detomidine was approximately 34% with GEL. Similar sedation scores were reached in both groups after administration of detomidine, but maximal sedation was reached earlier in the IV group (10 minutes) than in the GEL group (40 minutes). HR was lower after IV than GEL from 5 to 10 minutes after administration. All animals were adequately sedated, and we were able to administer local anaesthetic without resistance to all of the calves before disbudding.Conclusions and clinical relevanceOromucosally administered detomidine is an effective sedative agent for calves prior to disbudding.  相似文献

ObjectiveTo compare the sedative effects of intramuscular xylazine alone or combined with levomethadone or ketamine in calves before cautery disbudding.Study designRandomized, blinded, clinical trial.AnimalsA total of 28 dairy calves, aged 21 ± 5 days and weighing 61.0 ± 9.3 kg (mean ± standard deviation).MethodsCalves were randomly allocated to three groups: xylazine (0.1 mg kg–1) and levomethadone (0.05 mg kg–1; group XL), xylazine (0.1 mg kg–1) and ketamine (1 mg kg–1; group XK) and xylazine alone (0.2 mg kg–1; group X). Local anaesthesia (procaine hydrochloride) and meloxicam were administered subcutaneously 15 minutes after sedation and 15 minutes before disbudding. The calves’ responses to the administration of local anaesthesia and disbudding were recorded. Sedation was assessed at baseline and at intervals up to 240 minutes postsedation. Times of recumbency, first head lift and first standing were recorded. Drug plasma concentrations were measured.ResultsData were obtained from 27 animals. All protocols resulted in sedation sufficient to administer local anaesthesia and to perform disbudding. Sedation scores significantly correlated with drug plasma concentrations (p ≤ 0.002). Times to recumbency did not differ among protocols (2.8 ± 0.3, 3.1 ± 1.1 and 2.1 ± 0.8 minutes for groups XL, XK and X, respectively), whereas interval from drug(s) administration until first head lift was significantly shorter in group XK than X (47.3 ± 14.1, 34.4 ± 5.3 and 62.6 ± 31.9 minutes for groups XL, XK and X, respectively). The area under the time-sedation curve was significantly greater in group X than XK or XL (754 ± 215, 665 ± 118 and 1005 ± 258 minutes for groups XL, XK and X, respectively).Conclusions and clinical relevanceLevomethadone or ketamine with a low dose of xylazine produced short but sufficient sedation for local anaesthesia and disbudding with minimum resistance.  相似文献

The objective of this study was to assess cortisol and behaviour changes in calves hot-iron disbudded after different analgesic protocols. We assessed the response of 27 calves at 1, 3, 6 and 24 h after disbudding with regional anaesthesia (DA), regional anaesthesia plus carprofen (DAC), disbudded only (D) or sham-disbudded (ND). At 1h cortisol was higher in D compared with all other groups. At 3h DA showed higher cortisol than ND but did not differ from baseline. At 15 min and 1h pain-related behaviours were more frequent in the D group than in all other groups. Group D (3 h) and DA (3 and 6 h) showed more ear flicks and head rubs compared with DAC and ND. We concluded that head rubbing, head shaking and ear flicking are useful behaviours for evaluating pain after hot-iron disbudding. Disbudding causes severe pain in calves and only the association of regional anaesthesia with carprofen assures good welfare for 24 h.  相似文献

Aims. To evaluate the use of local anaesthesia and systemic analgesia for alleviating the distress associated with dehorning by monitoring stimulated behaviours in calves.

Methods. Eight behaviours were monitored in 3-4 month-old calves dehorned with and without local anaesthesia and/or a non-steroidal anti-inflammatory drug, from 2 hours after treatment.

Results. Four of the behaviours, lying, grazing or ruminating, tail shaking and ear flicking, were significantly different during the first 4-6 hours of observation in control calves, which were not dehorned, compared to calves dehorned without anaesthesia or analgesia, which suggests that these behaviours are useful indices of the acute pain experienced by calves after dehorning. The behaviour of calves dehorned after both a local anaesthetic and a non-steroidal anti- inflammatory agent had been administered was similar to control calves. The behaviour of calves given only local anaesthesia or systemic analgesia prior to dehorning was similar to that of calves dehorned without any pain relief.

Conclusion. These results suggest that calves should be given both a local anaesthetic and a systemic analgesic before dehorning to alleviate the pain associated with this procedure.  相似文献

ABSTRACT

Aims: The main goal of the current study was to evaluate, on a commercial beef cattle farm, the impact of infection with gastrointestinal nematodes resistant to both ivermectin (IVM) and moxidectin (MXD) on the productivity of calves.

Methods: Male Aberdeen Angus calves, aged 9–11 months, with faecal nematode egg counts (FEC) ≥200 epg and body weight ≥190?kg, were allocated to two herds. Herd A (n?=?90) grazed a maize-winter forage crop rotation and Herd B (n?=?90) grazed a 2-year-old Agropyrum pasture. On Day 0 in each herd, calves were randomly allocated into five groups (n?=?18), which were treated with 0.2?mg/kg IVM; 0.2?mg/kg MXD; 3.75?mg/kg ricobendazole (RBZ), both IVM and RBZ, or remained untreated. Faecal samples collected on Days ?1 and 19 were used to determine the percentage reduction in FEC, and genera of the nematodes were determined by the identification of the third-stage larvae recovered from faecal cultures. Total weight gain was determined from body weights recorded on Days ?1 and 91.

Results: Overall mean reduction in FEC was 42% for IVM, 67% for MXD, 97% for RBZ and 99% for IVM?+?RBZ. The reduction in FEC for Cooperia spp. was ≤78% for IVM and MXD, and for Haemonchus spp. was 0 and 36% for IVM and MXD, respectively, confirming the presence of parasites resistant to both anthelmintics. Only IVM?+?RBZ treatment resulted in 100% efficacy against Haemonchus spp. The overall estimated mean total weight gain for calves treated with IVM was 15.7 (95% CI?=?11.9–19.7) kg and for calves treated with IVM?+?RBZ was 28.8 (95% CI?=?25–32.5) kg (p?<?0.001). Mean total weight gain for calves treated with MXD was 23.5 (95% CI?=?19.7–27.2) kg.

Conclusions and clinical relevance: In calves naturally infected with resistant nematodes, under the production system assessed here, weight gains were lower in calves treated with anthelmintics that were moderately or highly ineffective compared to those treated with highly effective anthelmintics. These results demonstrate to farmers and veterinarians the importance of a sustainable and effective nematode control under field conditions.  相似文献

Cattle are disbudded or dehorned using a variety of methods. In this study, plasma cortisol concentrations were used to monitor distress in 6-week-old Friesian calves during the 9 hours following disbudding. Disbudding was carried out with a cautery iron or a dehorning scoop, with or without local anaesthetic. Cautery caused a transient rise in cortisol concentrations which returned to control values within 3 hours. The cortisol response to the scoop was more prolonged, as the plasma cortisol concentrations did not return to control levels until 7.5 hours after disbudding. The administration of a local anaesthetic reduced the cortisol response during the first 2 hours after scoop dehorning. This reduced response was followed by a delayed rise in cortisol concentrations between 2 and 7.5 hours. Cautery caused less distress than the scoop. The administration of local anaesthetic had little effect in alleviating distress in calves disbudded using the cautery iron.  相似文献

Approved analgesic compounds in cattle are not currently available in the United States due to the lack of validated pain assessment methods and marker residue depletion studies. In this study, we compared the pharmacokinetic parameters and effect of preemptive analgesics administered to calves subjected to dehorning with local anesthesia. Holstein steers were randomly assigned to receive one of the following treatments per os (PO) or intravenously (IV) (n = 8/group): meloxicam (1 mg/kg PO), gabapentin (15 mg/kg PO), meloxicam (1 mg/kg), and gabapentin (15 mg/kg) PO, flunixin (2.2 mg/kg IV), or a placebo. Plasma drug, haptoglobin, substance P (SP) concentrations, serum cortisol concentrations, ocular thermography, mechanical nociceptive threshold (MNT), and average daily gain (ADG) were evaluated. Data were analyzed using mixed‐effects models and noncompartmental pharmacokinetic analysis. Meloxicam, gabapentin, and meloxicam with gabapentin at the present doses did not reduce cortisol concentrations. Analgesic‐treated calves had significantly lower plasma SP concentrations and improved ADG compared with controls. Flunixin calves had reduced circulating cortisol compared with controls. Meloxicam‐treated calves showed an increase in MNT at two horn bud sites compared with the other treatments. Analgesics improved ADG and reduced biomarkers of pain, but effects differed by compound and route of administration.  相似文献

This study assessed the efficacy of ketoprofen for mitigating pain following dehorning with an electric cautery iron. Forty Holstein heifer calves, 4 to 8 wk of age, were randomized to receive a lidocaine cornual nerve block and either an injection of ketoprofen (3 mg/kg IM) or physiological saline, 10 min prior to dehorning. Cortisol was measured from serum obtained 10 min prior to dehorning and at 3 and 6 h post-dehorning. Calf behavior was video-recorded between 0 to 2, 3 to 5, and 6 to 8 h post-dehorning, and frequency of ear flicks, head shakes, head rubs, lying, standing, feeding, and grooming were recorded. Finally, 24-h intake of calf starter was recorded. There was no effect of treatment on cortisol (P > 0.1); however, ketoprofen-treated calves displayed less ear-flicks and total head behavior (P < 0.05), and tended to consume more starter (P = 0.09) than control calves. Ketoprofen is effective for mitigating behavioral effects of postsurgical pain following dehorning in 4- to 8-week-old calves.  相似文献

ABSTRACT

Aims: To investigate associations between lamb growth to weaning and dam udder and teat scores measured at pre-mating, pre-lambing, docking and weaning.

Methods: Mature Romney ewes (n?=?1,009) were enrolled from a commercial sheep flock located near Masterton, in the Wellington region of New Zealand in 2017. A range of udder and teat traits were scored in all ewes, using visual assessment and palpation, at pre-mating, pre-lambing, docking and weaning. During the lambing period, each newborn lamb was matched to its dam and lamb sex, birthweight and birth-rank were recorded. A rearing rank was allocated to each live-lamb at weaning, when all lambs were weighed (n?=?1,570), allowing calculation of daily growth rates (g/day). Associations between udder and teat scores and lamb growth rates to weaning were examined using multivariable models for each udder-scoring time.

Results: Growth rates of lambs whose dams had udder palpation scores of hard, or both teats recorded as abnormal, pre-mating were lower than lambs whose dams had normal scores (229.9 (95% CI?=?213.2–246.6) vs. 254.5 (95% CI?=?245.6–263.5) g/day; p?=?0.011) and (227.4 (95% CI?=?208.3–246.6) vs. 247.9 (95% CI?=?235.7–260.2) g/day; p?=?0.024), respectively. Growth rates of lambs whose dams had clinical mastitis at docking or weaning were lower than those without mastitis (215.8 (95% CI?=?199.9–231.7) vs. 235.4 (95% CI?=?225.4–255.0) g/day; p?=?0.007) and (220.0 (95% CI?=?205.2–234.8) vs. 254.7 (95% CI?=?248.9–260.5) g/day; p?<?0.001), respectively. Growth rates of lambs whose dams had asymmetrical udders at docking or weaning were lower than lambs whose dams had symmetrical udders (204.6 (95% CI?=?189.7–219.5) vs. 240.2 (95% CI?=?225.4–255.0) g/day; p?<?0.001) and (223.3 (95% CI?=?213.9–232.7) vs. 242.2 (95% CI?=?229.4–255.0) g/day; p?=?0.014), respectively.

Conclusions and Clinical Relevance: Pre-mating udder palpation and teat palpation scores can be used to identify ewes whose lambs are predicted to have lower growth to weaning. Assuming a mean lamb age at weaning of 84.4 days, lambs born to ewes with a pre-mating score of hard would be expected to have a mean weaning weight that was 2.1?kg less than those whose dams had normal scores. Udder palpation, udder symmetry and clinical mastitis scores during lactation were also associated with lamb growth rates.

Abstract

Two selenium (Se) supplementation trials were conducted in successive years involving a total of 70 red deer calves 3–15 months of age grazing pasture containing 30–57 ppb of Se on a dry matter basis. The trials compared growth rate, whole blood Se and glutathione peroxidase (GSHpx) concentrations of calves which received periodic doses of oral Se or a single injection of barium selenate (equivalent to 50 mg Se) or no Se supplementation. There were no significant weight gain differences between treated and untreated groups in either trial. Whole blood GSHpx levels were strongly correlated with blood Se levels (r = 0.9278) and produced the following regression equation: GSHpx 0.0155 Se ? 2.292.

In both years the 3 month old calves had GSHpx levels of 6–9 kU/l which probably derived from maternal transfer of Se. The GSHpx levels in unsupplemented calves declined from these levels to a minimum in winter (group means ? 2.6) and then progressively rose the following spring and summer.

Periodical oral dosing with Se or a single injection of barium selenate significantly elevated blood Se and GSHpx levels throughout the trials.  相似文献

ObjectiveTo investigate the pharmacokinetics, efficacy and convulsive dose of articaine hydrochloride in goat kids.Study designExperimental prospective study.AnimalsA total of 18 (n = 6 animals per experiment) male Saanen goat kids (2–4 weeks old).MethodsThe study consisted of three experiments. The first determined the pharmacokinetics of articaine following intravenous administration of articaine hydrochloride (8 mg kg–1). The second experiment investigated the anaesthetic efficacy and pharmacokinetics following cornual nerve block using 1.5% articaine hydrochloride. Anaesthesia of horn buds was evaluated using the response to pinprick test. Non-compartmental analysis was used. The final experiment determined the convulsive dose of articaine and its corresponding plasma concentration following intravenous infusion of articaine hydrochloride (4 mg kg–1 minute–1). Data are shown as mean ± standard deviation.ResultsThe mean terminal half-life (t1/2λz), mean volume of distribution at steady state (Vdss) and mean plasma clearance (CL) of articaine following intravenous administration were 0.66 hour, 3.81 L kg–1 and 5.33 L hour–1 kg–1, respectively. After cornual nerve block, the mean maximum plasma concentration of articaine was 587 ng mL–1 at 0.22 hour and its mean t1/2λz was 1.26 hours. Anaesthesia of horn buds was observed within 4 minutes following cornual nerve block. The mean dose required to produce convulsions was 16.24 mg kg–1 and mean convulsive plasma concentrations of articaine and articainic acid were 9905 and 1517 ng mL–1, respectively.ConclusionsIntravenous administration of 8 mg kg–1 of articaine hydrochloride did not cause any adverse effects. Pharmacokinetic data suggest that articaine was rapidly eliminated and cleared. Cornual nerve block using 1.5% articaine hydrochloride alleviated the response to the acute nociceptive stimulus during disbudding.Clinical relevanceArticaine hydrochloride appears to be a safe and effective local anaesthetic for disbudding in goat kids.  相似文献

ObjectiveTo compare the postoperative analgesic effects of intravenous (IV) lidocaine, meloxicam, and their combination in dogs undergoing ovariohysterectomy.Study designProspective, randomized, double‐blind, controlled clinical trial.AnimalsTwenty‐seven dogs aged (mean ± SD) 16.1 ± 7.5 months and weighing 22.4 ± 17.9 kg scheduled for ovariohysterectomy.MethodsAnaesthesia was induced with propofol and maintained with isoflurane. Dogs (n = 9 in each group) were allocated to receive just prior to and during surgery one of the following regimens: M group, 0.2 mg kg?1 IV meloxicam then a continuous rate infusion (CRI) of lactated Ringer's at 10 mL kg?1 hour?1; L group, a bolus of lidocaine (1 mg kg?1 IV) then a CRI of lidocaine at 0.025 mg kg?1 minute?1; and M + L group, both the above meloxicam and lidocaine treatments. Pain and sedation were scored, and venous samples taken for serum cortisol and glucose measurement before and at intervals for 12 hours after anaesthesia. Pain scores were assessed using a multi‐parameter subjective scoring scale (cumulative scale 0–21) by three observers. The protocol stated that dogs with a total score exceeding 9 or a sub‐score above 3 in any one category would receive rescue analgesia. Sedation was scored on a scale of 0–4.ResultsThere were no significant differences in subjective pain scores, serum cortisol, and glucose concentrations between the three groups. The highest pain score at any time was 5, and no dog required rescue analgesia. None of the three regimens caused any observable side effects during or after anaesthesia. At 1 and 2 hours after extubation dogs in group L were significantly more sedated than in the other two groups.Conclusions and Clinical relevanceThis study suggests that, with the scoring system used, IV lidocaine and meloxicam provide similar and adequate post‐operative analgesia in healthy dogs undergoing ovariohysterectomy.  相似文献

CASE HISTORY: Twelve of 150 goat kids, 4–10 days old, died 3 days after disbudding with a hot iron. Another 18 kids had been ill the previous day but survived following antibiotic therapy. Five of the dead kids were necropsied.

PATHOLOGICAL FINDINGS: There was necrosis and haemorrhage of the skin, subcutaneous tissues and frontal bone at disbudding sites in all five kids examined post mortem. Beneath disbudding sites in 4/5 kids there were bilateral, dark red, often cavitated areas of necrosis extending deep into the frontal cortex of the brain. Histologically, these areas consisted of coagulation necrosis, haemorrhage, vascular thrombosis and suppurative inflammation. Numerous bacteria, predominantly large Grampositive rods, were present in the necrotic brain tissue. In the remaining kid, bilateral areas of yellow discolouration and flattening of gyri in frontal lobes corresponded histologically to extensive polioencephalomalacia. A mixed growth of aerobes and anaerobes was cultured from the brain of one kid with suppurative lesions.

CLINICAL RELEVANCE: Thermal disbudding of neonatal kids is widely practised in dairy goat herds and is considered the method of choice for disbudding in New Zealand. However, the skull of goat kids is much thinner than that of calves and the safety margin for thermal injury to the brain is markedly reduced. This report highlights the risks associated with the technique and its potential as a welfare issue.  相似文献

AIMS: To determine the plasma disposition of meloxicam in goats following S/C, oral or I/V administration at a single dose of 0.5?mg/kg bodyweight.

METHODS: Five healthy Saanen goats, aged 12–14 months and weighing 35–40?kg, were used for a three phase cross-over design with a 10-day washout period, with meloxicam administered I/V, then orally and S/C. Heparinised blood samples (5?mL) were collected from all animals prior to drug administration (0 hours) and subsequently up to 96 hours. Concentrations of meloxicam in plasma were measured using high performance liquid chromatography. Concentration-time curves were fitted and pharmacokinetic parameters were estimated for each administration group.

RESULTS: Subcutaneous administration of meloxicam exhibited unique plasma distribution characteristics that differed from oral and I/V administration. Mean peak plasma concentrations were greater (1.91 (SD 0.39) vs. 0.71 (SD 0.17) µg/mL) and the time to reach them shorter (3.20 (SD 1.64) vs. 14.33 (SD 2.19) hours) following S/C compared with oral administration (p<0.05). The terminal half-life was longer (15.16 (SD 4.74) vs. 10.69 (SD 1.49) hours) and the MRT was shorter (15.67 (SD 2.37) vs. 24.33 (SD 3.12) hours) following S/C than oral administration (p<0.05), but bioavailability was similar (98.24 (SD 9.62) vs. 96.49 (SD 10.71)%).

CONCLUSION AND CLINICAL RELEVANCE: Subcutaneous administration of meloxicam resulted in long-term presence of drug at high concentration in goat plasma. This unique plasma disposition characteristic may offer an advantage in some clinical cases towards potentially improving the treatment efficacy in goats.  相似文献

ABSTRACT

Aims: To compare detection by real-time PCR of DNA from Mycoplasma bovis on mucosal swabs taken from the palatine tonsillar crypt and the mainstem bronchi of clinically asymptomatic calves after slaughter.

Methods: We compared the sensitivity of mucosal swabs taken from two sites: the palatine tonsillar crypt and the mainstem bronchi. Paired samples were taken post-mortem at slaughter from 55 clinically well calves from an infected herd and were tested by real-time PCR for the presence of M. bovis-specific DNA.

Results: Mycoplasma bovis DNA was detected in 51 palatine tonsillar crypt swabs (92.7 (95% CI?=?82.4–98.0)%) and seven mainstem bronchial swabs (12.7 (95% CI?=?5.3–24.5)%). All seven calves with positive mainstem bronchial swabs also had positive palatine tonsillar crypt swabs.

Conclusions: When compared to mucosal swabs of the mainstem bronchi, mucosal swabs of the palatine tonsillar crypt were seven times more sensitive for the post-mortem detection of M. bovis DNA. The viability of detected M. bovis was not assessed, because any cattle carrying viable or non-viable M. bovis DNA were determined to be a potential risk to eradication. Palatine tonsillar crypt mucosa may be a useful anatomical site for real-time PCR detection of M. bovis DNA in naturally infected calves. More work is needed to define the persistence and viability of M. bovis at this anatomical site.

Clinical relevance: The results of this study helped form the basis of surveillance tools used in M. bovis control and eradication efforts. Familiarity with these results may help veterinarians better communicate with their clients about the science behind the eradication efforts.  相似文献

Ingvast‐Larsson, C., Högberg, M., Mengistu, U., Olsén, L., Bondesson, U., Olsson, K. Pharmacokinetics of meloxicam in adult goats and its analgesic effect in disbudded kids. J. vet. Pharmacol. Therap. 34 , 64–69. The pharmacokinetics and analgesic effect of the nonsteroidal anti‐inflammatory drug meloxicam (0.5 mg/kg) in goats were investigated. In a randomized, cross‐over design the pharmacokinetic parameters were investigated in adult goats (n = 8) after single intravenous and oral administration. The analgesic effect was evaluated in kids using a randomized, placebo controlled and blinded protocol. Kids received meloxicam (n = 6) once daily and their siblings (n = 5) got isotonic NaCl intramuscularly while still anaesthetized after cautery disbudding and injections were repeated on three consecutive days. In the adult goats after intravenous administration the terminal half‐life was 10.9 ± 1.7 h, steady‐state volume of distribution was 0.245 ± 0.06 L/kg, and total body clearance was 17.9 ± 4.3 mL/h/kg. After oral administration bioavailability was 79 ± 19%, Cmax was 736 ± 184 ng/mL, Tmax was 15 ±5 h, although the terminal half‐life was similar to the intravenous value, 11.8 ± 1.7 h. Signs of pain using a visual analogue scale were smaller in kids treated with meloxicam compared with kids treated with placebo on the first day after disbudding, but subsequently no difference in pain was noticeable. Plasma cortisol and glucose concentrations did not differ between the two groups.  相似文献

Caustic paste is frequently used for disbudding young female dairy calves. Nerve blocking may not be completely effective after such chemical tissue damage. Regional anaesthesia, together with a non-steroidal-anti-inflammatory drug (NSAID), was shown to reduce plasma cortisol in calves disbudded using caustic paste. To find out whether pre-emptive NSAID alone could control pain or whether NSAID reduces cortisol response by a mechanism other than by pain control, we compared cortisol levels and behaviour of 10 chemically disbudded calves treated with IV flunixin-meglumine, five of which were injected at 5 min (F0) and five injected at 60 min before dehorning (F1), with 5 sham-dehorned (ND) and 5 non-treated chemically disbudded animals (CD). There was a higher (p < 0.001) cortisol level in both NSAID-treated groups compared with ND at 1 h after disbudding, but no differences from control animals (CD). Non-treated disbudded animals showed higher cortisol at + 3 h compared with ND. A higher incidence of pain-related behaviours was shown in disbudded animals up to 3 h post-disbudding.We conclude that pre-emptive analgesia treatment by itself is not effective in controlling pain and does not prevent blood cortisol increase after disbudding of calves with caustic paste.  相似文献
