[Objective] To research the effects of medicated serum of HERBA POLYGONI PERFOLIATI on HBsAg and HBeAg secretion in2215 cells. [Methods]Destruction rate of medicated serum of HERBA POLYGONI PERFOLIATI to 2215 cells and HL-7702 were detected by MTT method; HBsAg and HBeAg secretion in cell supernatant was detected by ELISA method when four different concentrations of medicated serums were applied in 2215 cells for 72 h. [Results]20% and 40% medicated serums showed inhibitory effects on HBsAg and HBeAg secretion,showing certain dose-effect relationship. At the same time,medicated serum of HERBA POLYGONI PERFOLIATI had less significant inhibitory effects on HL-7702 than that on 2215 cells. [Conclusions] Medicated serum of HERBA POLYGONI PERFOLIATI had certain toxic and anti-HBV effects in vitro.  相似文献

喹乙醇诱导小鼠睾丸生精细胞凋亡作用研究   总被引:2,自引:0,他引:2  
按毒理学方法给小鼠灌喂不同剂量喹乙醇,采用原位末端标记法检测该药物饲料添加剂对生精细胞凋亡作用的影响。结果表明:各试验组小鼠生精细胞凋亡指数较正常组和阴性对照组均有所增加,在不同时间,高剂量组均表现差异显著(p<0.05)或极显著(p<0.01),且连续给药8 d和12 d时,喹乙醇各试验组呈现出明显的剂量-效应关系,说明喹乙醇可以诱导小鼠睾丸生精细胞发生凋亡。  相似文献

[Objective]To research the apoptosis of hepatoma cells HepG-2 induced by shikimic acid from FRUCTUS ANISI STELLATI and its mechanism. [Methods]Hepatoma cells HepG-2 at logarithmic phase were randomly divided into drug group and control group. Shikimic acids from FRUCTUS ANISI STELLATI at the concentrations of 0. 125,0. 5 and 1 g /L were used. Inhibitory effects of shikimic acids on HepG-2 cell proliferation were detected by MTT method. Effects of shikimic acids at the concentrations of 0. 125,0. 5 and 1 g /L on the cell cycle of hepatoma cells HepG were researched by Flow Cytometer. Expression of Bcl-2 and Bax proteins were detected by immunocytochemistry detection. [Results]Results of MTT test showed that shikimic acid(0. 125,0. 5 and 1 g /L) had significant inhibitory effects on the proliferation of in vitro human liver cancer HepG-2 in a time-and dose-dependent relationship(P 0. 05). Shikimic acid blocked the hepatoma cells HepG-2 at G1 phase,so that the cells could not enter into S phase for DNA synthesis,which restricted the cell proliferation. Results of immunocytochemistry detection showed that protein expression of Bcl-2 decreased significantly,while Bax expression enhanced significantly,so that the proportion of Bcl-2 to Bax reduced. [Conclusions]shikimic acid blocked the hepatoma cells HepG-2 at G1 phase,and had significant inhibitory effects on the proliferation of human hepatoma cell through reducing the proportion of Bcl-2 to Bax.  相似文献

本试验旨在探究奶牛乳腺上皮细胞(bovine mammary epithelial cells,BMECs)最佳的冻存液以改善乳腺上皮细胞的冻存质量。BMECs传至第5代后分别加入以下10种不同配方的冻存液。A组:85%DMEM+10%胎牛血清+5%DMSO;B组:80%DMEM+10%胎牛血清+10%DMSO;C组:75%DMEM+10%胎牛血清+15%DMSO;D组:70%DMEM+10%胎牛血清+20%DMSO;E组:85%DMEM+5%胎牛血清+10%DMSO;F组:75%DMEM+15%胎牛血清+10%DMSO;G组:70%DMEM+20%胎牛血清+10%DMSO;H组:80%DMEM+10%胎牛血清+10%甘油;I组:70%DMEM+10%胎牛血清+20%甘油;J组:60%DMEM+10%胎牛血清+30%甘油,冻存前统一调整细胞密度到1×106 /mL冻存。分别对复苏后的细胞进行台盼蓝染色计算存活率和PI/Hoechst33258双染计算凋亡率。结果表明:BMECs经不同冻存剂冻存复苏后,细胞活力、形态学及凋亡率表现有所不同,其中B组和G组的活力和24 h贴壁率较其他组高,二者的凋亡率较低,二者之间差异无显著性(P>0.05);传代后B组细胞的生长状况最好。  相似文献

[Objective] To investigate the safety of compound Zhuqin extract.[Methods]In acute toxicity test,mice in test group were given compound Zhuqin extract( i. g.) by gavage,the dosage of which was 0. 8,0. 6,0. 45,0. 35 g /20 g,to determine the median lethal dose of compound Zhuqin extract( LD50). In maximum tolerance experiment,mice in test group were given compound Zhuqin extract( i. g.) by gavage,the dosage of which was 0. 5 g /20 g,to determine the maximum tolerance dose( MTD); In the sub- chronic toxicity test,rats in test group were given compound Zhuqin extract( i. g.) by gavage for 30 d,the dosage of which was 2. 0,1. 5,1. 0 g /200 g,and normal saline negative control group was set. Clinic condition of rat was observed everyday,hematology,blood biochemistry,anatomy,organ index and organization pathological observation were conducted after test.[Results]The acute toxicity test failed to detect median lethal dose of compound Zhuqin extract; the weight differences were not significant( P 0. 05); some blood biochemical criterion and blood routine index had significant differences( 0. 01 P 0. 05),and some indexes had extremely differences( P 0. 01); organ index,necropsy and histopathology had abnormalities.[Conclusions] Compound Zhuqin extract was safe and reliable.  相似文献

[Objective]To discuss the cutting propagation and cultivation technology of Plumbago zeylanica L. [Methods] The cutting,breeding,soil,fertilizer application and field management were researched. [Results]The branch age had little relationship with the survival rate. During the process of cutting,ABT rooting powder treatment was not needed for the branches. Survival rate had no significant correlation with the length of branches. The survival rate on loamy soil was 95%. Cutting period should be from June to August. The high soil maturation degree and fully decomposed organic fertilizer were helpful to the growth of P. zeylanica. Soil moisture should be paid special attention after cutting. Based on observation,the seedling stage of P. zeylanica was 20- 30 d after cutting; the optimal transplanting period was 20- 30 d after seed sowing. [Conclusions]The cultivated seedlings of P. zeylanica had significant and strong principal roots; the principal roots were10 cm in length with many fibrous roots(more than 15); the seedlings grew well with dark green leaves and strong stems.  相似文献

[Objective] To extract and isolate the effective components from Sorbus tianschanica,and to research the hypoglycemic effects of n-Butanol part. [Methods]S. tianchanica was extracted by reflux extraction. After gradient elution,n-Butanol part was obtained. With diabetic mice induced by STZ as the model,blood glucose test was carried out. [Results]n-Butanol part in administration group had significant differences in hypoglycemic effects with that in model group(P 0. 05),showing that n-Butanol part had certain antidiabetic functions on diabetic mice induced by STZ. And the hypoglycemic effects showed a reducing trend as the administration dosage decreased,indicating that administration dosage had positive correlation with hypoglycemic effects. n-Butanol in different parts could reduce the blood glucose of diabetic mice in different degrees. And the blood glucose value in high-dosage group(8 g /kg) reduced greatly at 2 h after administration,having great differences with that in model group(P 0. 01). This showed that high dosage was helpful to the glucose tolerance of diseased mice,and could effectively protect against the rapid increase of blood glucose after meal. n-Butanol had no significant effects on the blood glucose of normal mice. However,the morality rates in high-and middle-dosage groups were both higher than that in low-dosage group; and the morality rate in administration group was lower than that in model group,showing that n-Butanol could promote the survival rate of diabetic mice. [Conclusions]S. tianchanica had certain effects of anti-diabetes.  相似文献

[目的]观察不同浓度复方杜仲对肉鸡免疫器官发育及新城疫血凝抑制(ND-HI)效价的影响。[方法]1日龄AA肉鸡400只随机均分4组。Ⅰ、Ⅱ、Ⅲ、Ⅳ组分别在饮水添加0 g/L、1.25 g/L、2.5 g/L和5 g/L的复方杜仲。每周末各组随机抽样8只称重、心脏采血、屠宰取胸腺、脾脏和法氏囊称重。[结果] 1至6周胸腺指数、法氏囊指数各组间无差异。14日龄脾指数Ⅱ组高于Ⅰ组(P<0.05);21日龄Ⅱ组高于其他组(P<0.05),其中Ⅱ、Ⅲ组间差异极显著(P﹤0.01);42日龄复方杜仲Ⅱ、Ⅲ组脾指数均低于Ⅰ组(P<0.05),Ⅳ组极显著低于Ⅰ组(P<0.01)。21日龄Ⅲ、Ⅳ组ND-HI抗体水平高于Ⅰ组(P<0.05);28日龄达到最高水平,Ⅱ、Ⅲ组高于Ⅰ组(P﹤0.05),Ⅳ组极显著高于Ⅰ组(P﹤0.01)。[结论] 复方杜仲水煎液对前3周肉鸡脾指数有一定的影响,且以低剂量组效果最佳。对ND-HI效价有增强作用,呈现量效递增关系。  相似文献

[Objective]To determine the contents of inorganic elements in the stems of wild Plumbago zeylanica Linn. in different harvest periods( from January to December),and to discuss the effects of harvest period on the content of inorganic elements. [Methods]Microwave digestion and Inductively Coupled Plasma Optical Emission Spectrometer( ICP-OES) were both used to determine the inorganic elements in the stems of wild P. zeylanica. The detection results were researched by correlation analysis,principal component analysis and cluster analysis. [Results]The contents of inorganic elements in the stems of P. zeylanica varied in different harvest periods. Al,Ca,Fe,Sn,Ti,As,V, Zn,Mn,Mg and Ni were the eigenelements of P. zeylanica stems. Cluster analysis showed that the 12 samples of P. zeylanica stems could be divided into two categories: September was the first major category,and the others were the second category. [Conclusions]The detection results offered useful data for the synergic relationship among elements,and provided theoretical basis for the better utilization and development of P. zeylanica resources.  相似文献

将试验小鼠分为对照、模型和剂量(低、中、高)组,建立免疫抑制模型。对照组、模型组饲喂基础饲料,剂量组分别饲喂含不同板蓝根粗多糖的饲料。6周后测小鼠血常规指标、脏/体比、血清白介素-2(IL-2)及肿瘤坏死因子-α(TNF-α)的含量。结果表明,免疫抑制小鼠的白/红细胞数、血小板数和血红蛋白值均随粗多糖剂量的增加而增大;体增质量对照组最大,低、中、高剂量组和模型组依次递减;各剂量组脏/体比均明显小于对照组;各剂量组小鼠血清中IL-2和TNF-α水平均显著上升。说明板蓝根粗多糖能提升小鼠免疫功能,但对小鼠的发育有一定影响。  相似文献

为研究日粮中添加千里光超微粉对仔猪生长性能和部分血清生化指标的影响,选取体重为10 kg左右的杜×长×大三元杂交仔猪90头,按随机区组设计、公母各半的原则,将仔猪分成5个处理组,每个处理3个重复,每个重复6头仔猪。Ⅰ组饲喂基础日粮,Ⅱ组饲喂添加75 mg/kg金霉素和20 mg/kg硫酸粘杆菌素的日粮,Ⅲ、Ⅳ、Ⅴ组分别饲喂添加0.4%、0.8%、1.2%千里光超微粉的日粮。试验期为30天。结果发现,添加千里光超微粉能提高仔猪平均日增重和平均日采食量,降低料肉比,其中添加0.8%的千里光超微粉能显著提高仔猪的平均日增重(P<0.05)、显著降低料肉比(P<0.05),且与Ⅱ组的效果相当(P>0.05);添加千里光超微粉能在一定程度上提高血清GLU、TG、T3、T4、IGF-I含量,降低血清BUN含量,其中添加0.8%的千里光超微粉能显著提高血清T3、IGF-I的含量(P<0.05),显著降低BUN含量(P<0.05)。表明仔猪日粮中添加0.8%的千里光超微粉能显著提高仔猪平均日增重和血清T3、IGF-I含量,显著降低料肉比和血清BUN含量,改善仔猪的生长性能。  相似文献

为了探索一种简便易行、具有良好应用性并适用于中医药领域免疫功能低下的动物模型,以及为福建黄兔提供一种新型实验动物的实验依据。实验采用氢化可的松为细胞免疫抑制剂,6个实验组,其中1个对照组,5个剂量组(分别每天20、25、30、35、40 mg/kg),采用流式细胞检测法测CD4、CD8细胞含量,并通过摘取脾脏的方式计算脾脏指数。实验结果显示:B、C剂量组(分别每天25、30 mg/kg)的福建黄兔在注射药物前后CD4、CD8细胞含量呈现明显的下降趋势,证实上述2个剂量组是福建黄兔免疫功能低下模型造模的较好剂量;同时,A~E剂量组的黄兔脾脏相较于对照组均呈现不同程度的缩小,脾脏指数降低,B、C、D组与对照组差异显著,反映其免疫功能受损,其中尤以C剂量组(30 mg/kg)下降最为明显。由此可见,C剂量组是本实验中免疫功能低下动物模型造模的最适宜剂量。  相似文献

以叶片诱导的愈伤组织为材料,用秋水仙素溶液浸泡法和秋水仙素加入培养基法分别进行诱导。实验结果表明,秋水仙素加入培养基法要好于秋水仙素溶液浸泡法,具体表现为:用秋水仙素加入培养基法诱导出的愈伤组织褐化死亡率低,分化率高,四倍体细胞平均比例高。而在处理浓度与时间的选择中,以高浓度的秋水仙素与短时间处理组合较好。秋水仙素的浓度为600 mg/L处理3 d效果最佳,四倍体细胞比例达41.5%。  相似文献

目的:通过流式细胞术观察拯救CIAV M突变株对宿主细胞MSB1的生长周期及凋亡的影响。方法:拯救CIAV M突变株接于MDCC-MSB1细胞系(试验组),并相应设立阴性组(正常SPF鸡肝脏细胞液),阳性组(CUX-1标准毒株),进行培养24hr-48hr。用PI及FITC Annexin V染色方法,用流式细胞仪测定宿主细胞生长周期及凋亡,并用SPSS13.0进行结果统计学分析。结果:接种拯救CIAV M突变株的宿主细胞G0/G1期的累积细胞百分率为59.33%±4.04%,S期为32.92%±3.65%,G2/M期为7.75%±3.11%,与阴性组相比,G0/G1期细胞累积百分率有所上升;与CUX组相比,G0/G1期有所下降,但结果经统计学分析,差异均不显著(P>0.05)。试验组宿主细胞凋亡率为37.00%±4.65%,与阴性组相比,凋亡率上升,统计学差异显著(P<0.05);与阳性组相比,凋亡率下降,统计学差异显著(P<0.05)。结论:本次试验结果显示,经过VP3基因突变的CIAV M突变株,经过病毒拯救并回归本动物后,对宿主细胞MSB1的生长周期没有显著的抑制,但能引起宿主细胞的显著性凋亡,且凋亡程度与CUX-1引起的宿主细胞凋亡率相比有所降低。可以表明,通过VP3密码子移位的基因突变,能够将CIAV的致凋亡作用降低,从而相应降低病毒的致病性,为以后构建CIAV减毒疫苗株奠定理论基础。  相似文献

摘 要 杜×长×大三元杂交肥育猪48头被随机分成三组,即对照组、试验1组和试验2组。三组猪试验处理如下:在基础饲粮中加黄霉素20mg/kg后,喂对照组猪;在基础饲粮中加0.15﹪金银花-黄芪复合提取物(LJAMCE)后,喂试验1组猪;在基础饲粮中加0.3﹪LJAMCE后,喂试验2组猪。饲养试验期为40d。饲养试验结果如下: ①试验1、2组猪的日增重均在不同程度上快于对照组猪,分别提高了20.4﹪(P<0.05)和10.0﹪(P>0.05);②LJAMCE可提高饲料转化率,分别比对照组提高9.8﹪(P>0.05)和5.2﹪(P>0.05)。③饲粮中加0.15﹪LJAMCE对GPT活性、IgG、TG、vLDL等血液生化指标都有不同程度的积极作用;④LJAMCE在饲粮中适宜添加量为0.15﹪。 关键词 LJAMCE;生长性能;生化指标;育肥猪  相似文献

3种中药复方提取液对肉鸡生理、免疫指标的影响   总被引:1,自引:0,他引:1  
为探讨复方中药提取液对肉鸡生理机能的影响,验证中药改善肉鸡生产性能的功效,研究以党参、黄芪、六神曲、麦芽、山楂等组方,设计3个中药复方(方Ⅰ、方Ⅱ和方Ⅲ),水提后浓缩,1日龄开始混饮健康肉鸡(0.5%),并进行免疫器官指数、血清蛋白、血清酶、新城疫病毒(Newcastle disease virus,NDV)血清抗体滴度、白介素2(Interleukin-2,IL-2)、白介素3(Interleukin-3,IL-3)及平均日增重、料肉比、死亡率测定。结果显示,3个复方中药提取液对肉鸡免疫器官发育、血清蛋白及天冬氨酸转移酶(AST)、丙氨酸转移酶(ALT)、乳酸脱氢酶(LDH)含量没有影响(P0.05),方Ⅱ组肉鸡21、28日龄血清碱性磷酸酶(ALP)含量显著高于对照(P0.05);试验鸡21日龄时,3个复方中药组NDV抗体滴度显著高于对照(P0.05),28日龄时方Ⅰ组抗体滴度极显著高于对照(P0.01),方Ⅲ组抗体滴度显著高于对照(P0.05);方Ⅰ组IL-2活性在21日龄、28日龄时显著高于对照(P0.05),方Ⅲ组IL-2活性在21日龄时显著高于对照(P0.05),方Ⅰ组IL-3活性在28日龄时显著高于对照(P0.05);试验鸡28日龄时,方Ⅰ、方Ⅱ2组平均日增重高于对照,最高为方Ⅱ组72.72g/天,方Ⅰ、方Ⅱ2组料肉比低于对照,最低为方Ⅱ组2.08;死亡率最低为方Ⅰ组1%。上述结果表明,3个复方中药提取液安全性较高,对肉鸡生理机能均有一定改善作用,其中方Ⅰ在延缓血清抗体滴度降低速度,促进细胞因子分泌方面效果明显,而方Ⅱ在提高肉鸡生产性能方面效果较好。  相似文献

为了探讨蟾酥注射液对新城疫疫苗免疫效果的影响。选取1日龄健康雏鸡240只,随机分成4组,每组60只,设试验组(低剂量组、中剂量组和高剂量组)和对照组,选择不同浓度的蟾酥注射液与新城疫疫苗联合使用作为试验组。分别在21、28、35、42、49和56日龄,通过β-微量法,检测血清中ND-HI抗体;通过MTT法,检测外周血T细胞吸光度值(OD值);通过称量体重和免疫器官重量的方法,检测免疫器官指数。结果显示:蟾酥注射液各组与对照组比较,ND-HI抗体水平、外周血T细胞转化、胸腺指数、法氏囊指数和脾指数均有不同程度的提高。说明蟾酥注射液能够提高机体的体液免疫和细胞免疫水平,从而增强新城疫疫苗的免疫效果。  相似文献

[Objective] To discuss the effects of Yuezhitang decoction on promoting lactating function of maternal mice,enhancing the quality of newborn mice,and improving the survival ratio of newborn mice by using the overload lactation model of mice.[Methods]Overload lactation model of maternal mice was established.Maternal mice were given different dosages of samples by gavage.The lactation volume,accumulative lactation volume,body weight change of newborn mice at lactation period were researched.The mammary gland index was detected; the changes of breast tissue morphology and physiology were observed.[Results] Compared with model group,11 and 16.5 g/kg dosage groups significantly improved the lactation volume of maternal mice(P0.05).Mammary gland index of 11 g/kg dosage group increased significantly.In 11 and 16.5 g/kg dosage groups,mammary gland acinus had dense structure; the acinus was arranged regularly with complete structure and significant increase in quantity,showing typical characters of secretory mammary gland.[Conclusions]Yuezhitang decoction promoted the lactation of maternal mice.  相似文献

为了探讨中草药对哲罗鲑增重率、死亡率以及血液生化指标的影响,并为相关中草药药剂的研制提供相关的参考数据,笔者选用黄芪、当归、甘草、茯苓及贯众5种中草药,按不同比例添加到饲料当中鉰喂1+龄哲罗鲑。试验设加药组及对照组,每组30尾试验鱼,按照鱼体重的2.0%分别投喂含不同中草药的饲料,对照组投喂基础饲料,均设平行。试验结束后,对各组的增重率和死亡率进行统计,并采集血液,进行生化指标的测定。测定结果显示,甘草组对鱼体增重效果较好,而茯苓组呈现抑制效果。死亡率方面,除贯众和茯苓,其他试验组均无死亡。生化指标方面,总胆红素在贯众高剂量组有明显升高;甘草低剂量组及黄芪高剂量组尿素氮在血清中的含量明显升高;其余几项生化指标与对照组相比没有明显变化。试验结果表明,虽然选用的5种中草药皆具有一定免疫促进功能,但作为饲料添加剂对哲罗鲑的生长及血液生化指标的改变还是有一定区别的,与剂量也密切相关。  相似文献
