Abstract. The possibility of oral transmission of the pleistophorid microsporidian parasite Vavraia parastacida Langdon from marron, Cherax tenuimanus (Smith), was investigated by feeding infected marron and yabby, Cherax albidus Clark, muscle to juvenile marron and yabbies. Infections were established in a proportion of marron and yabbies with tissue from both crayfish, and both mounted a vigorous haemocytic response which failed to arrest progress of the disease. The disease appeared to progress more rapidly in the yabbies, in which no natural cases have been found. It would appear inadvisable to mix marron and yabbies on crayfish farms unless the marron are first demonstrated to be free of the parasite. As an intermediate host is not essential to transmission, which can occur through cannibalism, the exclusion of possible vectors will not control vavraiasis on crayfish farms, It remains possible that an alternate life cycle involving intermediate or transport hosts also takes place.  相似文献

The effects of salinity and temperature on plasma protein concentration and total haemocytic prophenoloxidase (proPO) were determined. Groups of 10 juvenile yellowleg shrimp, Penaeus californiensis Holmes, were acclimated for 20 days at different salinities (28%o, 32%o, 36%o, 40%o and 44%o at 25 °C) or temperatures (18, 22, 25, 28 and 32 °C at 36%o). While total protein levels were not affected, the quantities of proPO increased as salinity was elevated. Temperature affected both haemolymph parameters, showing a significant decrease in proPO at 32 °C, and an increase of protein at 28 and 32 °C. These results may help to explain the elevated disease susceptibility of shrimp at higher salinities and/or temperatures.  相似文献

Vibrio parahaemolyticus, an important pathogen of freshwater prawn, Macrobrachium rosenbergii causes high mortality. Four strains of V. parahaemolyticus were isolated from the gill, haepatopancreas, hemolymph and carapace of diseased prawn. The species were characterized based on biochemical and serological tests. On performing an antibiotic assay, these strains were found to be highly susceptible to cephalaxin, tetracycline and erythomycin. During the in vitro pathogenicity test, all the strains were found to be positive to the Congo red binding assay and to be haemolytic in nature, whereas an in vivo pathogenicity test revealed that 2 × 109 CFUs mL?1of bacteria induced disease symptoms such as black colouration on the carapace, red discolouration of the exoskeleton and loss of appendages within 6 days and 80% mortality. In histopathological studies, a prominent necrosis was seen in gill lamellae, and branchial arches were thickened at places due to hyperplasia and haemocytic infiltration. Hepatopancreatic tissue showed dilation of tubules, vacuolation of hepatocytes and marked necrosis in acinar cells. Development of an immunological technique for the detection and screening of V. parahaemolyticus infection and its treatment is highly important in freshwater prawn aquaculture.  相似文献

A histopathological survey of commercial farms in Ecuador culturing redclaw crayfish, Cherax quadricarinatus (Von Martens), revealed the presence of different viral, bacterial and non‐infectious pathologies. A total of 536 animals from 27 different farms were analysed during a 3‐year period; 25, 154 and 357 specimens in 1995, 1996 and 1997, respectively. From the total sampled, a virus referred to as Cherax quadricarinatus bacilliform virus (CqBV) had a prevalence of 44% in animals examined during 1996 and 33% in 1997. A rickettsia‐like organism (RLO) was detected in five specimens sampled from three different farms in 1995. This RLO was detected in 36 and 12% of samples analysed in 1996 and 1997, respectively, and was the infectious agent most frequently associated with mortalities above 60%. Co‐infection by CqBV and the RLO was observed in 18 and 3% of the crayfish examined in 1997 and 1998, respectively. Five specimens out of 25 examined presented no sign of an infectious agent during 1995. The proportion of specimens testing negative by histopathology to known or suspected infectious agents was 25 and 32% in 1996 and 1997, respectively. Although different infectious agents were present in cultured crayfish, farms in general had overall survivals higher than 50% in most ponds. Other conditions observed included an intracellular prokaryotic organism, iron granules in the R and F cells of the hepatopancreas, different species of epibionts on the surface of the gills and body, and two cases of haemocytic enteritis. Two cases of serious mortality in different farms were associated with the presence of Cowdry type A inclusions, pyknosis, karyorrhexis and haemocytic infiltration in the stomach hypodermis and the antennal gland.  相似文献

In this study, black disease infecting fairy shrimps, Streptocephalus sirindhornae Sanoamuang, Murugan, Weekers & Dumont, and Branchinella thailandensis Sanoamuang, Saengphan & Murugan, in Thailand, was investigated. The typical signs of the disease are the appearance of black spots on the cuticle, located mainly on the dorsal side and thoracopods. A number of rod-shaped bacteria aggregated in the black spots and were visualized by scanning electron microscopy. The histopathological results showed that a haemocytic response to the infection resulted in a dense melanized core of bacteria. In addition, generalized septicaemia by rod-shaped bacteria was also observed in the infected tissue. Of the 31 isolates, Aeromonas spp. were predominantly isolated and six strains were selected for the experimental infections. The most pathogenic strain was identified molecularly as A. hydrophila. When fairy shrimp were infected at bacterial concentrations of 10(4) and 10(6) cfu mL(-1) , the overall infection levels were 73.33 ± 6.67% and 93.33 ± 6.67%, respectively. The experimentally infected fairy shrimp showed abnormal swimming and died within 24-48 h after the appearance of the dark pigment.  相似文献

An epidemic of ‘milky disease’ in the mud crab (Scylla paramamosain) generally breaks out in the fall when the crab is near maturity, resulting in large economic losses in crab farming. Vibrio parahaemolyticus has been proven to be one of the major pathogens. In this study, the mud crabs were challenged with V. parahaemolyticus, and their innate immune responses were investigated in terms of total haemocyte counts (THCs), haemocytic enzyme activities and gene expression levels during a 114‐h period. The THCs of the mud crabs decreased significantly after 42 h of exposure. The activities of the haemocytic enzymes, including acid phosphatase‐alkaline phosphatase, phenoloxidase, superoxide dismutase (SOD), peroxidase and nitric oxidase synthethase, were significantly enhanced during the challenge course. The gene expression levels also significantly increased for all tested genes (proPO, Cu/Zn‐SOD, Prx, LYS, CRU and ALF) with the exception of CAT down‐regulated expression. The results may imply that the immune responses of the mud crab could be activated by the pathogens, and the data here will provide many clues for further systematic investigation of ‘milky disease’ caused by V. parahaemolyticus and the disease prevention in mud crab S. paramamosain.  相似文献

The influence of rearing conditions on Flavobacterium columnare infection of rainbow trout, Oncorhynchus mykiss (Walbaum), was studied experimentally in the laboratory and at a fish farm. In experiment I, the effect of parasitic infection on columnaris disease was studied using F. columnare carrier fish. The fish were exposed to Diplostomum spathaceum cercariae and a set of other stressors in order to induce clinical columnaris infection. Parasitic infection and other stressors failed to induce the disease. Disease occurred when the fish were challenged with F. columnare, but D. spathaceum infection did not enhance the severity of the infection. In experiment II, the influence of rearing density and water temperature was studied. Overall mortality was highest in fish at normal rearing density with high temperature (+23 degrees C). At low temperature (+18 degrees C) mortality was not affected by rearing density, but the transmission of columnaris disease was faster at normal rearing density at both temperatures. This supports the view that reduction of fish density could be used in prevention of columnaris disease especially if water temperature is high. Because the lower rearing density can also decrease the transmission of ectoparasites and penetrating endoparasites, it could be an efficient tool in ecological disease management.  相似文献

用纯化栉孔扇贝(Chlamys farrei)急性病毒性坏死症病毒(AVNV)免疫兔子,以兔抗血清为一抗,荧光标记的羊抗兔抗体为二抗,采用冰冻切片技术,建立了栉孔扇贝AVNV的间接免疫荧光检测方法。用该方法分析栉孔扇贝和海湾扇贝(Argopecten irradians)体内AVNV感染强度,并对感染率进行统计。结果表明,栉孔扇贝的肾脏、肝胰腺中,AVNV阳性信号最强,呈现中度到重度感染,其AVNV感染率100%。鳃丝、性腺及闭壳肌中未检测到阳性信号。在海湾扇贝的肝胰腺及肾中AVNV的感染率也较高。提示AVNV感染扇贝的靶器官主要是肾脏、肝胰腺。对与栉孔扇贝同一海区养殖的海湾扇贝在栉孔扇贝发病期间存活率较高的原因进行了讨论。  相似文献

In this study experimental challenges with Moritella viscosa, the causative agent of winter ulcers in salmonids, were performed on juvenile Atlantic cod and Atlantic halibut. The challenges involved both intramuscular (i.m.) and intraperitoneal (i.p.) injections and bath with a strain originally isolated from Atlantic salmon. Cod was found to be significantly more sensitive than halibut to the infection. Both fish species were found to be more sensitive to i.m. than i.p. challenges. Both challenges induced a systemic disease in cod and halibut, but only cod was infected with an experimental bath challenge. Pathognomonic signs were found to be comparable with those described in M. viscosa-infected salmon and turbot. The main distinguishing pathological sign was that the cod showed host response to M. viscosa infection resulting in granuloma formation in infected tissues, which is a known response of cod to a infection with another Gram-negative bacterium, Aeromonas salmonicida. Re-isolation of M. viscosa from kidneys of cod and halibut with clear disease signs was problematic and optimization of isolation measures is needed. The results of this study indicate that M. viscosa infection may be a risk factor in cod farming, but that halibut is more resistant.  相似文献

Amoebic gill disease (AGD) is a pathogenic disease in salmonids caused by Neoparamoeba perurans. Treatment of AGD infection has been through freshwater bathing of the fish. However, as the availability of fresh water is often limited, hydrogen peroxide has been introduced as an alternative treatment. This study investigated the effect of hydrogen peroxide as treatment for AGD‐infected salmon (Salmo salar L.,) at different seawater temperatures and hydrogen peroxide dosages. In total, 600 fish were challenged with N. perurans and the severity of the AGD infection was measured using a gill score scale. After challenge and disease development, the fish were distributed into 12 tanks. The treatment was performed at different seawater temperatures (8°C, 12°C, 17°C) using different hydrogen peroxide doses. Each temperature included an untreated control group. Linear models were used to analyse gill score. A significant effect of treatment was found (?0.68 ± 0.05) regardless of dose and temperature, suggesting that hydrogen peroxide was effective in treating AGD. When the model included dose, a negative linear relationship between dose and gill score was found. The study proved that treatment of AGD with hydrogen peroxide was successful, as gills partially recovered following treatment and further disease development was delayed.  相似文献

Mortalities of puerulus and juvenile rock lobsters, Jasus edwardsii (Hutton, 1875), held in shore-based ongrowing facilities at water temperatures between 10 and 18 °C were associated with infection by an invasive oomycete identified as Haliphthoros sp. Gross signs of disease included loss of appetite, lethargy and 1–3 brown/black focal necrotic lesions in the gills near insertion of the walking legs. Hyphae were observed in wet preparations of gill filaments excised from lesions. Histology of gill lesions showed hyphae inside the gill cuticle, invasion and lysis of the skeletal muscle and massive haemocyte infiltration and melanization at the base of walking legs adjacent to infected gill filaments. Lobsters over approximately 30 mm carapace length appeared resistant to infection. Death of affected lobsters usually occurred prior to or during the moult and in some cases may have been associated with secondary bacterial infection. Haliphthoros sp. was isolated in pure culture and marine agar 2216 was the medium that produced best growth in vitro . Two isolates were exposed in vitro to five chemicals to determine if an effective treatment could be found. Chemicals that interrupted the life cycle by killing zoospores or preventing sporulation included malachite green, trifuralin, formalin and copper sulphate. The appearance of the disease was associated with poor hygiene and its elimination may be achievable by improving husbandry practices.  相似文献

Modifications of great scallop (Pecten moximus) shells have been noticed in many sites of scallop fisheries in Brittany, especially in shallow waters. These calcification abnormalities are linked to the appearance of a brown colouration of the internal calcified shell layer, due to the presence of a eumelanin associated with the insoluble organic matrix of the biocrystals. The appearance of this pigment generates many disturbances of the calcified foliated microstructure of the scallop internal shell layer. The mantle structure is not modified in brown shells as compared with white ones. No pathogenic signs such as hyperplasia or haemocytic infiltration have been observed. According to this observation, we hypothesize that the brown colour phenomenon is more a result of environmental disturbance rather than a symptom of a pathogenic disease.The colour abnormalities of the internal shell layer can be detected by a spectral analysis of its reflectance before it can be detected with the naked eye. This method, correlated to microstructural observations, gives a rapid and precise analysis of the appearance of the pigmentation on adult or juvenile scallops. It may be a useful method for the evaluation of the influence of environmental parameters, for example, on calcification and its abnormalities.  相似文献

Abstract. Myxobolus insidiosus from the striated musculature of chinook salmon, Oncorhynchus tshawytscha (Walbaum), was studied in disease free and naturally contaminated waters. Infection could not be induced in susceptible fish in a disease free water supply by any means triad. Experiments in naturally contaminated waters provided data on minimum and other exposure times. Studies were also carried out on the effect of water temperature, different substrates and flow rates, time of year and size of fish on infection. The rainbow trout, Salmo gairdneri Richardson, was also checked for infection and not found with this disease in naturally contaminated waters. Transovarian infection was ruled out.  相似文献

Under normal farming conditions, shrimp can experience starvation periods attributable to disease outbreaks or adverse environmental conditions. Starvation leads to significant morphological changes in the hepatopancreas (HP), being the main organ for absorption and storage of nutrients. In the literature, limited research has described the effect on the HP of periods of starvation followed by refeeding and none in whiteleg shrimp (Penaeus vannamei) using computer‐assisted image analysis (CAIA). This study describes the effect of starvation and starvation followed by refeeding on the HP of whiteleg shrimp using CAIA. Visiopharm® software was used to quantify the following morphological parameters, measured as ratio to the total tissue area (TLA): total lumen area (TLA:TTA), haemocytic infiltration area in the intertubular spaces (HIA:TTA), B‐cell vacuole area (VBA:TTA), lipid droplet area within R cells (LDA:TTA) and F‐cell area (FCA:TTA). Significant changes were measured for HIA:TTA and LDA:TTA during starvation (increase in HIA:TTA associated with decrease in LDA:TTA) and starvation followed by refeeding (decrease in HIA:TTA associated with increase in LDA:TTA). In the future, HIA:TTA and LDA:TTA have the potential to be used in a pre‐emptive manner to monitor the health of the HP, facilitate early diagnosis of diseases and study the pathophysiology of the organ.  相似文献

Infectious salmon anaemia (ISA) is a viral disease of farmed Atlantic salmon, Salmo salar L., in Norway. However, in laboratory experiments, the virus has also been found to propagate in trout, Salmo trutta L., where it is a persistent infection. It is crucial for the management of ISA in Norway, and for the prevention of spread to fish in other countries, that possible carriers of the virus are found. Another possible salmonid reservoir species is the rainbow trout, Oncorhynchus mykiss (Walbaum). After experimental infection, the ISA virus was found to be present in O. mykiss as late 28 days after infection, with a peak around day 20. The infection resulted in a significant drop in the haematocrit and haemorrhages on the liver in some specimens. The ISA virus was seen budding from endothelial cells lining blood vessels in the heart ventricle of challenged rainbow trout. It is concluded that the ISA virus is able to propagate in O. mykiss and that this species may function as a reservoir when cultured in areas where the virus is found. The virus does not seem to cause any significant mortalities of infected rainbow trout.  相似文献

Channel catfish virus (CCV) disease is an acute haemorrhagic disease in juvenile channel catfish ( Ictalurus punctatus ). To date channel catfish is the only species affected by natural outbreaks of the CCV but juvenile large mouth bass ( Micropterus salmoides ) and silurus ( Silurus meriaionalis ) have suffered high mortalities in recent years in China. Histopathological phenomenon of sick fish is similar to CCV disease, and the identified virus was CCV. In this report, the pathogenicity of infectious CCV was examined by infection trials on the first known host species, the channel catfish and other teleosts. Our results indicated that there were higher detection rates of CCV from large mouth bass and silurus fish. Channel catfish virus did not induce mortality in other cypriniformes, but histopathological studies revealed that carp might be infected by both bathing and intraperitoneal infection. No deaths, clinical or histopathological signs, were found in the six other species exposed by immersion or injection. Experimental infection studies confirm that CCV infect not only channel catfish but also other species (large mouth bass, silutus and carp). The outbreaks of CCV disease only occurred when the cultured temperature was above 25 °C.  相似文献

Tick-borne encephalitis (central european encephalitis) and Lyme disease (erythema chronicum migrans) are diseases mainly transmitted by ticks of the ixodus ricinus species. The tick-borne encephalitis, a virus infection, can produce severe encephalitic symptoms. There is no specific treatment but the infection can be prevented by active or passive immunization. Lyme disease is caused by a spirochete (borrelia burgdorferi). Due to the variety of often uncharacteristic symptoms this infection is not easily recognized. However, early diagnosis and antibiotic treatment are necessary to prevent chronic damage to skin, joints, heart or nervous system.  相似文献

Age‐dependent susceptibility to nervous necrosis virus (NNV) was demonstrated for barramundi (Lates calcarifer). The experiment used juvenile barramundi produced from a single spawning that were challenged consecutively by immersion with a redspotted grouper nervous necrosis virus (RGNNV) isolate. The dose and environmental conditions (35 ppt salinity and 30 °C) were constant. Fish and water were sampled longitudinally for histopathology and RT‐qPCR analysis to examine the evolution of the disease, virus replication, immune response and release of virus into water. Viral nervous necrosis (VNN) disease occurred in barramundi challenged at 3 and 4 weeks of age while fish challenged at 5, 7 and 9 weeks of age developed subclinical infection. Replication of NNV occurred faster and the concentration of virus reached higher concentrations in the younger fish with clinical disease. Virus isolation and qPCR tests indicated that infectious NNV was released from carcasses into water when fish were affected with clinical disease but not when NNV infection was subclinical. Based on these observations, we consider that carcasses from clinically infected fish have a potentially important role in the horizontal transmission of NNV, and barramundi juveniles should be protected from exposure to NNV until they are 5 weeks of age and reach the disease resistance threshold.  相似文献
