Germination of sclerotia ofSclerotium rolfsii on agar nutrient medium was delayed or slightly inhibited by concentrations of propiconazole between 0.4 and 4.0 μg ml?1, but was strongly inhibited by 8 μg ml?1 and completely inhibited by 16 μg ml?1. On the other hand, growth of hyphae from the germinated sclerotia was strongly inhibited by propiconazole at 1 μg ml?1 or greater. Hyphal growth from agar discs on agar medium was about 8 times less sensitive than hyphal growth from the sclerotia or from hyphal inoculum in liquid media. Propiconazole at 0.25 and 1.0 μg ml?1 strongly inhibited ergosterol biosynthesis, but this was not associated with large accumulations of C-14 methyl sterols. The ratio of eburicol to ergosterol in hyphae grown in the presence of 0.25 μg ml?1 propiconazole for 16, 30 or 45 h was 0.11, 0.13 and 0.04, respectively, for the three intervals while for hyphae grown in the presence of 1 μg ml?1, the ratios were 0.29, 0.36 and 0.30, respectively, for the same intervals. In view of a ratio of 23.5 for14C-acetate incorporation into the two sterols during the initial 6 h growth period in the presence of propiconazole, it is believed that the lack of large accumulation of C-14 methyl sterols is due to the feedback inhibition by eburicol or to cell lysis when the content of ergosterol becomes too low in the actively growing cells.  相似文献

A technique is described for testing isolates of Septoria tritici from winter wheat for resistance to benzimidazole fungicides. Secondary spores from 23 isolates were tested on Czapek Dox V-8 agar amended with benomyl at 1–10 μg ml. Twenty-one isolates were recovered from eight crops in England in 1984 and two (PBI isolates) were obtained in 1973. Thirteen isolates, including both PBI isolates, were sensitive to benomyl at 1 μg ml?1 and nine were resistant at 10 μg ml?1. The remaining isolate had a low proportion (1:3.7 x 104) of resistant spores. The minimum inhibitory concentration for sensitive isolates was 0.2–0.4 μg ml 1 benomyl and for resistant isolates was more than 1000 μg ml 1. Benomyl-resistant isolates were cross-resistant to carbendazim, thiabendazole and thiophanate-methyl, but not to 12 other fungicides with different modes of action. The implications of these findings are discussed.  相似文献

The uptake and metabolism of DDT, fenitrothion and chlorpyrifos were studied in cultures of the ciliate protozoan Tetrahymena pyriformis. When cultures were treated with DDT in concentrations varying from 0.01 to 0.5 μg ml−1, concentrations found in T. pyriformis were 3.8 to 335 μg g−1 dry weight. The accumulation of fenitrothion ranged from 28.7 μg g−1 in cultures treated with 1 μg ml−1 to 2260 μg g−1 in cultures treated with 10 μg ml−1. Under similar experimental conditions chlorpyrifos was accumulated from 24.7 to 15400 μg g−1. The patterns of uptake were dependent on the growth cycle, the ability of the organism to metabolise insecticide and the type of the insecticide used. Maximum accumulation of DDT, fenitrothion and chlorpyrifos occurred in 2, 4 and 6 h respectively. Tetrahymena metabolised DDT to DDD and DDE but failed to metabolise fenitrothion and chlorpyrifos. The effects on growth and morphology of T. pyriformis were studied over a period of 5 days. Higher concentrations (10, 50 and 100 μg ml−1) of DDT inhibited only the growth of the organisms and did not change cell morphology. Fenitrothion was extremely toxic to the organisms and at 5 and 10 μg ml−1 cells became more or less spherical and died after 48 h. However, concentrations of 0.5, 1 and 2.5 μg ml−1 fenitrothion caused growth inhibition, but only at 2.5 μg ml−1 was this permanent. Chlorpyrifos inhibited the growth of the organisms at 1, 5 and 10 μg ml−1 but the morphology was affected only at 5 and 10 μg ml−1.  相似文献

The antibiotic nucleoside tubercidin produced by Streptomyces viola-ceoniger was evaluated for in-vivo efficacy and in-vitro activity against Phytophthora capsici, Magnaporthe grisea and Colletotrichum gloeosporioides. Tubercidin was more effective against P. capsici and M. grisea than against C. gloeosporioides in inhibiting mycelial growth. The bioassay on TLC plates was the most sensitive method and allowed the evaluation of antifungal activity of tubercidin even at a low concentration of 0.1 μgml?1. As compared to the systemic fungicide metalaxyl, tubercidin was similar or somewhat higher in inhibition of mycelial growth of P. capsici. When applied to pepper stems, tubercidin was equally as effective as metalaxyl in the control of phytophthora blight in pepper plants, irrespective of application time and concentration. The treatment with 1000 μg ml?1 tubercidin induced phytotoxicity in pepper plants. No control efficacy of phytophtora blight was observed in pepper plants supplied with a soil drench of tubercidin. Treatment with tubercidin at 500 μg ml?1 completely protected pepper plants at first branch stage from phytophthora blight until four days after application. The control efficacy of tubercidin drastically declined seven days after application.  相似文献

Cette adaptation s'accompagne d'une diminution de la croissance sur milieu sans fongicide et du pouvoir pathogéne. La fréquence de zoospores capables de former spontanément une colonie sur 100 μg ml?1 de métalaxyl est de l'ordre de 10?4. Un traitement des zoospores à la N-méthyl-N′-nitro-N-nitrosoguanidine (30 μg ml?1 pendant 10 min) augmente cette fréquence par un facteur de 10–16 fois. Les souches résistantes ainsi obtenues ont la même croissance sur milieu sans fongicides et sont aussi pathogènes sur feuilles d'agrumes que les souches mères, alors que les souches sélectionnées sans traitement mutagène présentent les mêmes caractéristiques que celles obtenues par adaptation.  相似文献

The effect of a granular formulation of sodium azide, applied annually to pine nursery beds at rates of 0, 67.2, and 134.5 kg a. i. ha?1 under water seal or plastic seal, on soil fungal populations was determined over a 3-year period. Populations of fungi in the soil decreased following application of the sodium azide each year; the greatest decrease occurred at the highest rate of application. Populations of fungi in soil treated with the azide generally remained lower than in the controls throughout each of the 3 years; however, the population disparity between treated and control plots decreased in magnitude with each succeeding year. Populations of Trichoderma spp., in plots treated with 134.5 kg sodium azide ha?1, increased 2 weeks after treatment each year, and the population peaks increased in magnitude each year. In addition the effect of sodium azide (technical grade > 99%) at concentrations of 0, 2, 5, 10, 20 and 50 μg ml?1 in potato dextrose agar and blackstrap molasses agar media was determined in vitro for 14 isolates of Trichoderma harzianum. Growth and sporulation differed among the isolates and between the two media tested. Generally, the azide temporarily inhibited growth of the fungi, but the majority of the isolates were able to grow on either medium containing 50 μg sodium azide ml?1, although sporulation was more profuse on the molasses than on the potato dextrose agar medium.  相似文献

Field isolates of Botrytis cinerea with moderate levels of resistance to dicarboximide fungicides (ED50 1.0–4.9 μg ml?1) and to dicloran were obtained from glasshouses where vinclozolin and iprodione failed to control grey mould. From sensitive and moderatcly-resistant cultures, laboratory isolates were selected on dicarboximide-amended medium, which were highly resistant to these fungicides (ED50 125->3000 μg ml?1). Conidia of all the resistant isolates germinated well on media amended with 100 μg ml?1 of the dicarboximides vinclozolin, iprodione, procymidone and myclozolin and with 5 μg ml?1 of metomeclan. However, the spores of the moderately resistant isolates did not germinate on 100 μg ml?1 metomeclan while the spores of the highly resistant isolates germinated well. Using media with 100 μg ml?1 of metomeclan to distinguish between the two phenotypes, no highly resistant strain was detected among 312 resistant samples from five cucumber glasshouses with a high frequency of moderately resistant strains. From air-borne inoculum of five glasshouses with 100% resistant populations, 1604 colonies were recovered on vinclozolin-amended (100 μg ml?1) medium and none on metomeclan-amended (100 μg ml?1) medium. It is concluded that strains of B. cinerea highly resistant to dicarboximides are absent from field populations.  相似文献

The effects of atrazine were studied on growth, production of aflatoxin, and fatty acid and sterol biosynthesis by four isolates of Aspergillus in vitro. There was little effect of atrazine on Aspergillus spp. at concentrations up to 20 μg ml?1 but at 40 μg ml?1 or above, growth, production of aflatoxin, and fatty acid and sterol biosynthesis were remarkably reduced. Palmitic, stearic and linoleic acid synthesis were inhibited in three of the isolates tested at 60 μg ml?1. At 100 μg ml?1, except ergosterol, the cholesterol and 5, 7-ergostadienol synthesis was totally inhibited in all isolates. The effects of tebuconazole, a systemic fungicide, on the morphology, structure, cell wall components and toxin production of Fusarium culmorum were investigated in vitro. Treatment was by application of four filter paper strips (0.75 cm × 5.0 cm) soaked in 20 µg ml ?1 fungicide placed around a point inoculum in Petri dishes. Mycelial growth was strongly inhibited by fungicide treatment. Scanning electron microscopic observations showed that the fungicide caused irregular swelling and excessive branching of hyphae. The morphological changes induced by the fungicide at the ultrastructural level included considerable thickening of the hyphal cell walls, excessive septation, the formation of the incomplete septa, extensive vacuolisation, accumulation of lipid bodies and progressing necrosis or degeneration of the hyphal cytoplasm. Non‐membrane inclusion bodies were often detected in the hyphal cytoplasm. Furthermore, the formation of new hyphae (daughter hyphae) inside collapsed hyphal cells was common following treatment. The daughter hyphae also displayed severe alterations such as irregular thickening of the cell walls and necrosis of the cytoplasm. Using cytochemical techniques, the labelling densities of chitin and β‐1,3‐glucan in the cell walls of the fungicide‐treated hyphae were more pronounced than in those of the control hyphae. Moreover, immunogold labelling with antiserum against deoxynivalenol (DON) revealed that Fusarium toxin DON was localized in the cell walls, cytoplasm, mitochondria and vacuoles of the hyphae from the control and the fungicide treatment, but the labelling density in the fungicide‐treated hyphae decreased dramatically compared with the control hyphae, indicating that tebuconazole reduced Fusarium toxin production of the fungus. © 2001 Society of Chemical Industry  相似文献

The effects of several herbicides used in pome fruit orchards on the germination of spores and growth of mycelia of Botrytis cinerea, Mucor piriformis and Penicillium expansum in vitro and the survival of propagules of these fungi in soil were studied. Diuron in agar at 4–128 μg ml?1 reduced germination of spores of B. cinerea and M. piriformis, and 2,4-D and paraquat at 32 μg ml?1 similarly affected B. cinerea and P. expansum. Several herbicides at 128 μg ml?1 in agar reduced growth of B. cinerea and M. piriformis but were ineffective against P. expansum. Propagule survival levels of the three fungi generally were lower in both autoclaved and non-autoclaved soil amended with herbicides than in non-amended soil. This effect was greatest in non-autoclaved soil, suggesting involvement of microbial antagonists. The most effective herbicides for reduction of fungal propagules in soil were 2,4-D, diuron, and paraquat.  相似文献

Eleven sterol biosynthesis-inhibiting fungicides were compared in experiments to determine the physico-chemical properties required for most effective control of take-all by soil treatment. All were active in agar culture against an isolate of the pathogen which causes take-all, Gaeumannomyces grammis var. tritici, with prochloraz being the most toxic (EC50 0.02 μg ml?1) and PP 969 the least (EC50 0.44 μg ml?1). Penconazole and PP 969 had vapour activity against the fungus in further bioassays on agar. In soil in pots, the most strongly lipophilic compound, buthiobate, was ineffective against take-all in wheat; triadimenol was most effective and, like flutriafol, nuarimol and PP 969, retained some effectiveness after 12 weeks in soil. PP 969, unlike penconazole or nuarimol, was effective in soil treated unevenly by mixing the fungicides in layers. PP 969 is relatively polar, and it is suggested that this property, allowing redistribution in soil water rather than as vapour, outweighed its poor intrinsic toxicity. The ideal soil-treatment fungicide should therefore be polar and also have good intrinsic activity and moderate persistence. None of the compounds tested had all these properties.  相似文献

Fenpropimorph was found to be highly active against Penicillium italicum (EC50 0.01/μg ml?1). Conidia of P. italicum, treated with low concentrations of fenpropimorph, swelled in size and showed distorted germ tubes. During the initial stages of mycelial growth, fenpropimorph had little or no effect on the dry weight increase, which became strongly inhibited within 24 h after addition of the toxicant (0.05, 0.1 and 0.2 μg ml?1). Irregular deposition of β–1,3 and β–1, 4 polysaccharides, probably chitin, was observed after treatment with fenpropimorph or imazalil. Fenpropimorph (0.05 and 0.2 μ ml?1) caused the accumulation of a major demethyl-sterol that was different from ergosterol. It was identified as ergosta-8, 14, 24(28)-trien-3β-ol by mass, infrared, ultraviolet and proton nuclear magnetic resonance, spectrometric procedures. At both concentrations, the accumulation was already detected after incubation for 2 h. In contrast, imazalil (0.1 μg ml?1) caused the accumulation of several methyl- and dimethyl-sterols which were tentatively identified as eburicol (24-methylene-24, 25-dihydrolanosterol), 4, 14α-dimethylergosta-8, 24(28)-dien-3-one, 14α-methylergosta-8, 24(28)-dien-3-one and obtusifoliol (4, 14α-dimethylergosta-8, 24(28)-dien-3α-ol). The accumulation of ergosta-8, 14,24(28)-trien-3β-ol indicates inhibition of the Δ14-reductase in P. italicum in a similar manner to that found previously in Ustilago maydis.  相似文献

Disposition kinetics of cypermethrin and fenvalerate were investigated in lactating black Bengal goats following single dose intravenous administration at 57 and 45 mg kg?1 respectively. The maximum and minimum blood concentrations of cypermethrin were 18.49 (±3.17) and 0.06 (±0.002) μg ml?1, while the corresponding values for fenvalerate were 14.58 (±2.37) and 0.04 (±0.005) μg ml?1 respectively. Both cypermethrin and fenvalerate remained present in blood for 36 h. The mean t1/2β) and Vdarea values were 5.56 (±0.28) h and 10.38 (±2.20) litre kg?1 for cypermethrin and 5.66 (±0.35) h and 11.31 (±2.20) litre kg?1 respectively for fenvalerate. Both cypermethrin and fenvalerate persisted in goat milk for 36 h. The t1/2β) and AUC values of fenvalerate were 7.37 (±1.84) h and 122.38 (±11.65) μg h ml?1 whilst the corresponding values for cypermethrin were 6.66 (±1.54) h and 99.48 (±7.81) μg h ml?1 in milk respectively.  相似文献

As one of the most widely applied agricultural chemicals in the world, glyphosate has many effects on the environment. The present study quantified plant responses to exposure by glyphosate through the root zone for a range of concentrations (0, 10, 100 and 1000 μg L?1). Ludwigia peploides and Polygonum hydropiperoides were grown in a greenhouse and given a single exposure to glyphosate via the root zone. The growth and physiological parameters were measured before exposure and for 18 days postexposure. The growth variables that were measured included the relative growth rate, stem length increase, biomass and root‐to‐shoot‐ratios. The physiological variables that were measured were the chlorophyll content index and chlorophyll fluorescence. The data analyses revealed that the root‐zone glyphosate affected some of the measured variables in P. hydropiperoides more than for L. peploides. Polygonum hydropiperoides showed a significant decrease in the root‐to‐shoot ratios for the 100 μg L?1 treatment, compared to the 10 μg L?1 treatment. The chlorophyll content index of the treated plants was significantly reduced in P. hydropiperoides, compared to the untreated plants on Days 7 and 18. Ludwigia peploides was affected only on the day after exposure, with the chlorophyll fluorescence parameters being significantly less for the 1000 μg L?1 treatment, compared to the 10 μg L?1 treatment. Glyphosate‐treated P. hydropiperoides showed a decreased chlorophyll content and reduced chlorophyll fluorescence parameters. In contrast, L. peploides showed a decrease in the chlorophyll fluorescence parameters but no reduction in its chlorophyll content. In addition to demonstrating the adverse effects of root exposure to glyphosate for the study species, these data help to partially explain the highly invasive and persistent nature of L. peploides in marginal aquatic environments, such as agricultural ditches.  相似文献

Two radiolabelled forms of racemic [14C]cypermethrin (14C at the benzylic carbon or at C-1 of the cyclopropane ring) were separately administered twice daily to lactating cows in portions of the feed. The amounts dosed were equivalent to 0.2, 5 and 10 μg of cypermethrin per g of feed. The radioactivity eliminated in the milk indicated that the ingestion and elimination of radioactivity were in balance at about day 4 after the start of dosing. Urine and faeces were equally the major routes of elimination, and only a fraction of a percent of the dose appeared in the milk. The residue in the milk was unchanged cypermethrin and was found at a concentration that was proportional to the dose. At the high cypermethrin intake of 10 μg g?1 of diet, the residue in the milk was 0.03 μg g?1. Concentrations of residues in the tissues, measured after 7, 20 or 21 days of treatment, were low and in the order: liver>kidney>renal fat>subcutaneous fat>blood>muscle>brain. The major residue in the liver and kidney of a cow that received 10 μg of cypermethrin per g of diet was N-(3-phenoxybenzoyl)glutamic acid. Other conjugates of 3-phenoxybenzoic acid and of 3-(4-hydroxyphenoxy)benzoic acid (unidentified, with the exception of the glycine conjugate) were also present. The residue in fat (about 0.1 μg g?1 from an intake of 10 μg g?1 of feed) consisted mainly of cypermethrin.  相似文献

Seventeen field isolates of Botrytis cinerea were compared by determining their radial growth on synthetic media containing various amounts of 21 antifungal compounds. Twelve of these compounds were fungicides that are recommended for the control of Botrytis infections. There were marked differences between the isolates in their sensitivity to the compounds. Individual isolates displayed high levels of resistance to some of the fungicides, including benomyl, carbendazim, iprodione, thiabendazole, thiophanate-methyl, vinclozolin and zineb. The most potent growth inhibitors were benomyl and carbendazim (ED95 values for most isolates <0.1 μg fungicide ml?1 media), dichlofluanid, iprodione, nystatin, thiabendazole, thiophanatemethyl and vinclozolin (ED95 values for most isolates < 1.0 μg ml?1), and captan, chlorothalonil, dicloran and thiram (ED95 values for most isolates < 6.0 μg ml?1). Zineb was much less potent than the other recommended anti-Botrytis fungicides; it was no more effective than carboxin, dinocap and mancozeb (ED95 values for most isolates > 25 μg ml?1).  相似文献

The cyclic depsipeptide PF1022A, given orally to mice, showed very good anthelmintic activity against Heligmosomoides polygyrus and Heterakis spumosa at 50 mg kg−1. In vitro, PF1022A was very active against Trichinella spiralis and had good activity against Nippostrongylus brasiliensis at 1 μg ml−1. An 18-membered enniatin analogue, JES 1798, showed good activity only against N. brasiliensis at 10 μg ml−1. The optical antipode of PF1022A had poor activity even at 100 μg ml−1. The effects of PF1022A on the membrane potential and input conductance of somatic muscle of Ascaris suum were examined using a two-microelectrode current-clamp technique. PF1022A did not antagonize the effects of the selective nicotinic agonist levamisole. PF1022A and an analogue, JES 1798, but not the PF1022A antipode, produced a small time-dependent increase in input conductance associated with no potential change. The increase in input conductance did not occur in the Cl-free bathing solution, suggesting that the increase in input conductance was mediated by Cl ions. The addition of high concentrations of Ca2+ to the preparation after the addition of PF1022A did not lead to production of Ca2+-activated Cl channels, suggesting that its mode of action was not that of a Ca2+ ionophore. The mechanism by which the cyclic depsipeptide might increase the Cl conductance is discussed.  相似文献

Two bioassay methods are described which use detached tobacco leaves to measure the sensitivity of Peronospora tabacina to systemic fungicides. Tobacco leaves (13–15 cm2), treated with fungicides before or after detachment from the plant, were inoculated with sporangia in water drops and, after incubation in beakers and Petri plates, the disease severity and/or production of sporangia was determined 4–7 days after treatment with the fungicides. Of 15 systemic fungicides applied to detached leaves, eight N-phenylamides at 0.066?1.0 μg ml?1 controlled blue mould; metalaxyl was the most effective fungicide. Isolates of P. tabacina, collected in the field from tobacco plants grown in soil treated with metalaxyl, were not resistant to the fungicide applied to detached leaves prior to inoculation. The fungicide, applied to leaves before detachment, was used to measure the efficacy of five systemic N-phenylamide fungicides sprayed on the basal and unsprayed distal portions of the leaves. Blue mould was controlled on the basal portion of the leaf by all the fungicides at 0.66?1.0 μg ml?1, but it required the application of 3–30 times more chemical on the basal portion to achieve comparable blue mould control on the distal part of the leaf.  相似文献

Methods are described for the extraction and analysis by gas-liquid and high-pressure liquid chromatography of the fungicide imazalil, 1-(β-allyloxy-2, 4-dishlorophenethyl) imidazole, on potatoes. Before storage, over 80% was recovered from potatoes treated with 0.01–3.0 mg imazalil kg?1, with a detection limit of 2 μg kg?1. Imazalil applied to potatoes at 10 g t?1 before storage decreased the incidence of gangrene (Phoma exigua), silver scurf (Helminthosporium solani), skin spot (Polyscytalum pustulans) and black scurf (Rhizoctonia solani), and was at least as effective as thiabendazole applied at 40 g t?1. At 1 g t?1 it also decreased skin spot and silver scurf. Incidence of black dot (Colletotrichum coccodes) was unaffected by these fungicide treatments.  相似文献

The activity of two neem extracts, AZT and NEEM-AZAL (containing 30 and 3 mg azadirachtin ml?1 respectively) and synthetic azadirachtin (AZ) against second-instar larvae (L2) of Plutella xylostella L. was examined using leafdip bioassays. On Chinese cabbage, AZ was significantly (P <0.05) less toxic (3 to 4-fold; LC50 0.54 μg AZ ml?1) than either neem extract against a laboratory strain of P. xylostella (FS). The LC50 values for AZT against the FS and another laboratory strain (Wellcome) were not significantly different on Chinese cabbage. The activity of AZT against the FS and Wellcome strains was similar on Chinese cabbage and Brussels sprout. AZT was significantly less toxic (3-fold) on Brussels sprout against an acylurea-resistant field strain (Sawi) when compared with the FS strain on Chinese cabbage. Larval mortality (at day 13) was found to increase with increasing exposure time of P. xylostella (FS) larvae to AZT-treated Chinese cabbage, although there was little difference in mortality between 48 and 120 h exposure. When AZT, NEEM-AZAL and AZ were applied at a dose (1 μg AZ ml?1) which gave end-point mortalities between 50 and 90% (at day 13), all treatments delayed the development of a proportion of surviving larvae but no morphogenetic abnormalities were observed in larvae which reached pupation. Evidence for antifeedant (reduced weight gain) and repellant effects (choicechamber) for AZT were observed with L2 P. xylostella (Wellcome) on Chinese cabbage. AZT was also shown to have ovicidal activity against P. xylostella (Wellcome) at relatively high dose ranges (10-1000 μg AZ ml?1) as well as some contact activity (FS strain) in topical bioassays. In residual bioassays on glass with adults of the hymenopteran endo-larval parasitoid of P. xylostella, Diadegma semiclausum (Ichneumonidae), AZT showed little or no activity at rates up to 1000 μg AZ ml?1. In medium-volume (MV, 200 litre ha?1) and ultra-low-volume (c. 1 litre ha?1) spray bioassays on Brussels sprout, AZT gave 16-92% and 88-100% mortality respectively (Wellcome strain) at rates approximating to 1-20 g AZ ha?1. The residual activity of AZT and NEEM-AZAL against P. xylostella (FS) on Brussels sprout (MV spray) was observed to decrease appreciably after three days, the decline in activity being particularly marked for NEEM-AZAL.  相似文献
