Four microsatellite markers and a mitochondrial DNA (mtDNA) fragment were used to investigate two possible explanations for a reported decline in productivity of genetically improved farmed tilapia (GIFT) in Fiji: (i) a decline in genetic diversity (GD) and (ii) genetic introgression from feral tilapia populations. Genetic diversity was estimated using θ and allelic richness, while Bayesian clustering was used to assign individuals to genetic groups (K=2 or 3) to test for introgression. Differentiation among groups was estimated using FST analysis. Results indicate that genetic diversity had declined compared with a GIFT reference stock from WorldFish Centre, while there was little evidence for introgression from feral tilapia populations. Loss of genetic diversity most probably resulted from practices that have not actively managed genetic resources in the hatchery. While GIFT is considered to be an improved line of Nile tilapia (Oreochromis niloticus), mtDNA analysis here revealed haplotypes assigned previously to three discrete Oreochromis species (O. niloticus, Oreochromis mossambicus and Oreochromis aureus) in both the Fijian strain and the WorldFish Centre strain. Possible sources for the three divergent lineages are discussed. Results have implications for the management and future expansion of the tilapia culture industry in Fiji as well as in other Pacific island nations.  相似文献

Feral Australian Oreochromis mossambicus (Peters) (Pisces: Cichlidae) and an interspecific hybrid population (most probably originally derived from crosses of O. mossambicus and O. niloticus stocks) were used as model organisms to study the inheritance patterns of 24 allozyme loci and 31 random amplified polymorphic DNA (RAPD) loci in tilapia. Single‐paired matings of parents of known genotype were used to generate families, and 10–15 full‐sib offspring from each mating were used to test for mode of inheritance. The majority of allozyme and RAPD loci tested segregated in a Mendelian fashion. Allozyme markers in general showed co‐dominant inheritance patterns, while RAPD markers conformed to expectations for band presence/absence under a dominant allele model. Although only a small number of families and offspring were used, the results highlight the suitability of allozymes and RAPDs as genetic markers for population analysis in tilapia.  相似文献

A stripspawning methodology was evaluated for tilapia (oreochromid) species. This technique achieved an average hatching success of 68.6 ± 3.6% (N= 31). Female fecundity and spawning frequency were dependant on both genetical and husbandry factors. Egg yields for Oreochromis niloticus, O. mossambicus, and O. niloticus±O. mossambicus hybrids averaged 4.54, 10.86 and 10.36 eggs/g female/spawn, respectively. Female broodstock that were adapted to an intensive spawning regime exhibited a significant increase in fecundity. Additionally, egg survival was not affected by hydration for up to 15 minutes prior to fertilization. Results suggest that the strip spawning of tilapia species may be an efficient method of providing viable gametes for hatchery purposes.  相似文献

Among three tilapiine species, Oreochromis aureus (Steindachner), O mossambicus (Peters) and O. niloticus (L.), fifteen banding positions of haemoglobin were resolved using citric acid-aminopropylmorpholine pH 7.0 buffer. O. mossambicus and O. aureus displayed 7 and 8 fixed band locations, respectively, whereas eight strains of O. niloticus from Asian cultured and African wild populations showed a total of 14. Variability in the O. niloticus strains, manifested as presence or absence of haemoglobin bands, was observed in banding positions 3, 4, 6, 7, 8, 9, 10, 13 and 14. Six banding positions in O. niloticus were shared with O. mossambicus and 8 with O. aureus. A dendrogram constructed from distance coefficients between popu-lation pairs using the presence or absence data showed the close affinity among O. niloticus strains belonging to the O. niloticus niloticus subspecies. A slight separation of Kenya O. niloticus from the O. niloticus niloticus strains is consistent with its subspecific distinction as O. niloticus vulcani. Among three species, O. niloticus had a closer affinity to O. aureus than to O. mossambicus. The phylogenetic relation-ships using differences in haemoglobin patterns and the higher genetic variability of O. niloticus reflect similar results obtained from existing studies using allozymes. In general, the presence of multiple fish haemoglobins is attributed to their adaptability to haeterogeneous environments. This attribute is typical of tilapias.  相似文献

This study was aimed at detecting differences among tilapia species in immunological and biochemical parameters that are indicators for fish health. In addition, six blood parameters that are indicators of stress were measured, glucose concentration, ceruloplasmin activity, lysozyme activity, respiratory burst activity, haematocrit and leucocrit, and their levels were compared among groups. A calibration experiment was conducted with commercial stock of tilapia to optimize a protocol for measuring stress response in tilapia. An air exposure stress was induced to six groups of fish and blood samples were taken at six different times: 15 min, 1, 2.5, 4.5, 6 and 24 h, after inducing stress. The highest responses to stress were observed after 2.5–4.5 h. A second experiment was conducted with four tilapia species: Oreochromis aureus (Steindachner), O. mossambicus (Peters), a red strain and the wild type of O. niloticus L. Levels of serum biochemical components and parameters of the innate immunity response to stress were compared in ten fish from each species. Significant differences were observed. Oreochromis aureus differed from the other three species (notably from O. mossambicus) in most of the measured immune response traits (glucose concentration, lysozyme activity, haematocrit and levels of total protein and IgM after stress) and serum biochemical components (protein, albumin, triglyceride, cholesterol, calcium, magnesium, phosphorus, bilirubin and β‐hydroxybotiric acid). The identified differences suggest that hybrid families from O. aureus and O. mossambicus may be used to construct a segregating population for genetic analysis of the innate immune response to stress. Thus, these two species were bred for segregating F2 population, suitable for quantitative trait loci studies for the innate immune response to stress.  相似文献

A comparison was made of some productive traits of Stirling Nile tilapia (wild type) (Oreochromis niloticus, Linnaeus) and red hybrid tilapia (Florida red tilapia× Stirling red O. niloticus) males during a 98‐day grow‐out period. Twenty‐two males from each genetic group with initial weights of 139.0 g for O. niloticus and 207.3 g for the red hybrid were placed in triplicate tanks. The fish were fed with a feed containing 36.8% crude protein. Survival was 97.0% for the red hybrid and 83.3% for O. niloticus. Daily individual weight gains were 2.95 and 2.50 g and final body weights were 473.0 and 348.8 g for the red hybrid and O. niloticus respectively. Fillet yield was similar for both the species, with 33.4% for the red hybrid and 32.0% for O. niloticus. Fresh fillet lipid content was perceptibly less in the red hybrid (0.33%) than in O. niloticus (2.07%). Some benefits of a red low‐fat tilapia genotype are discussed.  相似文献

A comparison of composition and sensory evaluation were performed on fillets of Oreochromis niloticus (wild type) and red hybrid tilapia (RHT) (Florida red tilapia ♂× red O. niloticus♀). Fifty male tilapia, 25 O. niloticus (initial weight 159.3 g) and 25 red hybrid (initial weight 132.4 g), were placed in each of three 2.0 m3 volume tanks. The fish were fed a commercial feed containing 35.9% crude protein during the 120‐day experimental period and then killed in cold water (3°C). All the fish were filleted. Twelve fillets from each treatment were used for proximate analyses, five fillets from each treatment were used for fatty acids analyses and the remaining fillets were used for sensory evaluation. The compositional analyses showed similar moisture, true protein and ash content in both genetic groups, but a lower crude fat content was measured in the red hybrid fillets (0.70%) compared with that of O. niloticus fillets (0.97%). No differences between O. niloticus and the red hybrid were observed in their fillet profile of fatty acids. In the sensory evaluation test, a difference in fillet flavour between both genetic groups was perceived. Of the 112 consumers in the preference test, 81.2% perceived a difference in fillet flavour between the two tilapias, with a general preference for the red hybrid over the wild‐type O. niloticus. The benefits of cultivating a RHT with a low lipid content are discussed.  相似文献

The influence of salinity and dietary composition on blood parameter values (haematocrit, leucocrit, immature lymphocytes, mature lymphocytes, granulocytes, plasma osmolarity and total plasma protein) of red hybrid tilapia, Oreochromis niloticus (Linnaeus) x O. mossambicus (Peters), was studied. Two groups of tilapia were fed a high-protein diet while kept in fresh or brackish water, respectively, and compared with two groups fed a low-protein diet under similar environmental conditions. Treatments were executed in duplo. Results show that salinity influenced all cellular blood parameters except the haematocrit. Dietary composition influenced the total plasma protein and haematocrit, while all parameters changed as time progressed during the experiment. It was concluded that the environmental parameters investigated in the present study should be considered when estimating fish health based on blood parameter values.  相似文献

Four red tilapia hybrids were evaluated for growth in earthen ponds and for colour distribution: ‘red’O. niloticus (L.) x O. niloticus (L.) or O. aureus (Steindachner). and O. niloticus or O. aureus x ‘white’ segregate of a Philippine red tilapia (Oreochromis sp.). The best growth was obtained in the ‘red’O. niloticus x O. niloticus and O. aureusx‘white’ hybrids, although they constituted only a little over 50% males. Of these two hybrids, the former is all-red, while the latter segregates into 35% red and 65% less attractive bronze-coloured individuals. Ways for establishing all-male populations of these two hybrids, for possible improvement of their performance, are discussed. Incompatibility between ‘red’O. niloticus females and O. aureus males, limiting fry production of this hybrid, was observed. The few fish of this hybrid obtained and tested proved to be all-male and all-red.  相似文献

Genetic diversity of the Nile tilapia Oreochromis niloticus collected from the river Nile (Cairo, Assuit and Qena) and two Delta lakes (Burullus and Manzalla) in Egypt was examined by the analysis of randomly amplified polymorphic DNA (RAPD). Of 25 primers examined,21 primers produced 230 RAPD bands. The percentage of polymorphic bands in Manzalla (29.4%) and Burullus (24%) populations was low compared with Assuit (30.54%), Cairo (33.5%) and Qena (44.84%) populations. The highest percentage of polymorphic bands was observed in the Qena population,suggesting a greater potential for use in breeding programs. The molecular phylogenetic tree constructed by unweighted pair‐group method of analysis shows Manzalla and Burullus populations strongly linked and separate from the Assuit and Cairo populations, with Qena population as outgroup. The data serve as a baseline analysis of the current genetic diversity found among O. niloticus populations in Egypt.  相似文献

Despite a worldwide boom in tilapia aquaculture, South Africa has failed to follow suit, resulting in a small, very much undeveloped local industry. Much of the justification for this reality lies on the inadequacy of the species used and the stringent legislation preventing the use of the Nile tilapia, Oreochromis niloticus. With momentum gained in recent years, the local industry lobbying for the legalization of the O. niloticus has made great progress and its use in certain systems has recently been facilitated. This study was conducted to compare the growth performance between the indigenous Mozambique tilapia, Oreochromis mossambicus, another African alternative gaining momentum in Zambia, the Three Spot tilapia, Oreochromis andersonii, and the globally reputable O. niloticus. The study was conducted in a biofloc technology (BFT) system, showing great potential as an environmentally friendly technology. The trial was run for 10 weeks with 60 fish/tank stocked into four repeats tanks per species. Fish were sampled on six occasions with a regression fitted to the mass data. Oreochromis niloticus showed a significantly higher growth rate with an average daily gain of 0.693 ± 0.018 g/day and the lowest feed conversion ratio (FCR) of 1.01 ± 0.05, followed by O. mossambicus with average daily gain of 0.405 ± 0.025 g/day and FCR of 2.24 ± 0.16, and then O. andersonii with an average daily gain of 0.185 ± 0.025 g/day and an FCR of 2.53 ± 0.28, respectively. From this study, O. niloticus therefore appears to be the most adequate species for use in BFT systems in South Africa.  相似文献

To assess the genetic diversity of Nile tilapia Oreochromis niloticus, a total of 250 fish from five Egyptian populations were genotyped using six microsatellite markers. Heterozygosity and Wright's F‐statistics (FIS, FST, and FIT) were calculated to determine the genetic variation within and between these populations. Observed heterozygosities were in the range of 0.4 (Burullus) to 0.96 (Qena), with FIS values ranging from 0.082 to 0.282. The mean FST showed that approximately 96.5% of the genetic variation was within‐population and 3.5% was among populations. Standard genetic distances were used to classify the five populations into two major groups. The deeper lotic river Nile populations of Assuit and Cairo formed one group and the shallow less lotic Delta lakes populations of Manzalla and Burullus formed the second group, with the upstream Nile Qena population being an outgroup. The findings from the current study help understanding of the broad‐scale population structuring of the Nile tilapia (O. niloticus) allowing the population groupings identified to act as potential sources of genetic variation. These populations could be included in future Marker‐Assisted‐Selection programs for economically desired production traits.  相似文献

Fertilization of ultraviolet (UV) irradiated oocytes of Nile tilapia, Oreochromis niloticus (L.), with sperm from O, niloticus or blue tilapia, O, aureus (Steindachner), and subsequent suppression of the first cleavage of fertilized eggs successfully induced androgenesis in Nile and blue tilapia. The optimal doses of UV irradiation to denucleate a female genome of Nile tilapia prior to androgenesis ranged from 5940 to 6930 erg mm?2 for 54-63 s at a fixed intensity of 110 erg mm?2 s?1. Putative androgenetic fish were created from eggs which were irradiated at various times and several durations of heat-shock. Eggs which were treated for 5 min at 41.6 oC at 2 7.5 min after fertilization were the most successful at suppressing the first cleavage and producing viable androgenetic diploids in Nile or hybrid Nile X blue tilapia. The maximal survivals of putative androgenetic diploids in relation to the control were 1.60% and 0.90% in Nile and hybrid Nile X blue tilapia, respectively. The androgenetic offspring established exhibited active feeding and normal growth.  相似文献

Mass selection was trialed on Fijian hybrid tilapia (Oreochromis niloticus × Oreochromis mossambicus) to reduce the amount of black spots on otherwise red phenotypes. The responses of two selection intensities (low‐selection line, top 50% – L) and (high‐selection line, top 30% – H) were compared with that of a control line (no selection – C) across three generations. The relative growth performance of treatments (C, L and H) was examined in parallel in each generation to assess whether mass selection had a correlated negative effect on growth performance. The results show clearly that red phenotype can be improved significantly by applying mass selection, without affecting growth performance. We propose that black spots on an otherwise red phenotype could represent the allelic products of a second genetic locus influencing skin colour, which can be expressed in red individuals (genotype Rr) but which may be masked in black individuals (genotype rr).  相似文献

A study was carried out to investigate the genetic diversity during domestication of Oreochromis shiranus (Trewavas) and to see if it could be associated with events in the known history of aquaculture development in Malawi. Five polymorphic microsatellite loci were scored in 14 populations of O. shiranus and one population of O. mossambicus (Peters). The mean number of alleles per locus ranged from 4.4 ± 1.03 to 13.2 ± 3.31 and was higher in the wild populations than in the domesticated populations. Other measures of genetic diversity were also lower in the domesticated compared with the wild populations, and the decline in diversity was correlated with the time elapsed since the founding of the farm stocks. Ordination analysis grouped domesticated populations into three: (1) those that trace their genealogy from Lakes Chiuta and Chilwa populations and are now spread all over the country; (2) those that come from Lakes Malawi and Malombe; and (3) hybrids between O. shiranus and O. mossambicus. Genetic differentiation among farms was strongly influenced by the pattern of known exchanges among the farmers and introgressive hybridization that had occurred between O. shiranus and O. mossambicus in the farmers’ ponds. Thus, the process of genetic changes in the species subsequent to domestication are best explained and predicted by socio-economic factors that influence the behaviour of farmers, rather than by the time-and-distance models of standard population genetics.  相似文献

Abstract. Conical and round-bottomed incubating containers were evaluated for efficiency in producing Oreochromis fry. For Orcochromis niloticus (L.) and Oreochromis mossambicus (Peters): 0-, 12-, 24-, 48- and 72-h-old egg clutches were artificially incubated at 28°C in conical and round-bottomed containers. Hatching occurred within 72–84h and 90–102h after fertilization respectively compared to 96–120h for naturally incubated egg clutches. No significant differences (P > 0·05) were found between the hatch and survival rates of O. niloticus and O. mossambicus fry or between fry from the various ages of eggs. The type of containers, however, significantly (P < 0·05) influenced the hatchability and survival rates, the overall survival rate from fertilization being 60% and 85% for conical and round-bottomed containers respectively. Possible reasons for these differences are discussed. The feasibility of using the superior round-bottomed containers for artificial egg and fry incubation and the advantages for broodstock management to mass produce Oreochromis fry are discussed.  相似文献

First-feeding stage tilapia fry were reared for 40 or 50 days in 20-l capacity aquaria at six stocking densities (2, 4, 6, 8, 10, 12 l?1) and three feeding levels (6, 12, 24% of body weight daily). Fry survival and growth were improved at the higher feeding levels, but the percentage of fry surviving showed no consistent relationship to stocking density. Cannibalism accounted for a mortality rate of 10–35% in each experimental group and was inversely related to the level of feeding. Under equivalent conditions, O. niloticus female × O. aureus male fry had a higher rate of survival, better food conversion and were more uniform in size than O. mossambicus fry.  相似文献

脆肉罗非鱼是近年快速发展起来的特色淡水养殖产品,具有肉质爽脆、富有弹性、久煮不烂等优点,受到消费者的广泛欢迎。本研究主要是通过对脆肉尼罗罗非鱼与普通尼罗罗非鱼肌肉在质构特性、感官评价、基本化学成分、脂肪酸分布和游离氨基酸组成等进行比较研究,从而揭示脆肉尼罗罗非鱼与普通尼罗罗非鱼的肌肉理化与营养特性的差异。研究结果显示:脆肉尼罗罗非鱼肉的硬度、弹性和咀嚼性与普通尼罗罗非鱼差异显著(p<0.05);脆肉尼罗罗非鱼肌肉中的粗蛋白和灰分与普通尼罗罗非鱼无明显差异,但水分含量显著降低,总脂含量显著升高(p<0.05);脂肪酸组成分析显示脆肉罗非鱼肌肉中所含的多不饱和脂肪酸显著高于普通尼罗罗非鱼(p<0.05),其中亚油酸、二十碳五烯酸和二十二碳六烯酸分别是对比组的2.12、3.36和3.71倍,脆肉尼罗罗非鱼肌肉的游离必需氨基酸总量,显著高于普通尼罗罗非鱼(p<0.05),但鲜味氨基酸甘氨酸略低于普通尼罗罗非。研究结果表明,脆肉尼罗罗非鱼和普通尼罗罗非鱼肌肉在质构特性、含水量、脂肪酸含量和游离氨基酸分布上都存在有明显的差异。  相似文献

Several tilapia species in Mexican reservoirs have been used as food source for more than 40 years but little effort has been devoted to the conservation and genetic selection of those species. Our objective was to evaluate genetic variability in feral (n = 59) and hatchery-produced (n = 58) Oreochromis niloticus stocked in the Benito Juarez dam. Allozyme analyses for 13 enzymatic systems and general proteins yielded 27 genetic loci. Seventeen loci were polymorphic in at least one sample. The observed heterozygosity was lower than expected and the imbalance in the Hardy–Weinberg equilibrium in almost all loci was caused by heterozygote deficiency. The inbreeding coefficient was 84%. Although the evidence suggests moderate genetic differentiation, it is probable that a great proportion of the source of hatchery organisms is from feral organisms. According to our results, management of the hatchery strain is inadequate, therefore other breeding strategies are needed to increase the genetic variability of O. niloticus from Oaxaca.  相似文献
