Chawali and Lekali are two common farmer’s barley varieties or landraces in Jumla, Nepal (2,240–3,000 m) with small to bold grains and wide adaptation from irrigated low lands to high hills. This study was undertaken to test whether features of the traditional seed system can significantly influence the diversity of a crop and its conservation on-farm. In Jumla (high-hill), the barley seed system is completely informal and is mainly from farmer to farmer. In the present study, the seed flows and the pattern of genetic diversity in barley were investigated to detect differences between the two varieties and test the divergence among populations of each variety These data suggested that Chawali, the more common variety, was less subject to homogenising gene flow between farms than was Lekali. A total of 128 farming households were surveyed for seed supply information and 128 populations for each landrace from two villages: Kartikswami and Talium were collected for SSR diversity analysis. Some 92 SSRs were screened in an initial sample of 20 barley populations of both landraces and 2 improved varieties (LG-51 and Soluwa). Of the 81 SSRs that consistently amplified, only 15 SSRs (19%) were polymorphic with gene diversity values ranging from 0.09 to 0.71. A medium to low diversity was detected among the landrace populations of barley varieties. Chawali populations were less polymorphic within ecological groups, and more divergent between than were Lekail populations. This result accords with Chawali having a more conservative local seed system.

Farming communities in traditional agroecosystems have been playing an important role in conserving agricultural diversity. They are not only the custodians but also managers of the crop diversity and maintain the dynamic processes of crop evolution and adaptation, the key elements of sustainable agricultural productions. The Himalayan highlands are important centres of crop plant diversity due to high ecological heterogeneity and high local socio-cultural integrations. The crop genetic diversity of Kumaon Himalaya in Uttarachal State of India has been documented in the present study. Existing crop genetic diversity at inter- and intra-species level was assessed and factors for changes in crop compositions and farming systems during the recent past were studied. Farmer management of rice landrace populations were studied in greater detail. Various benefit enhancing options for farmers from local crop diversity were scrutinized based on farmer perceptions and priorities for efficient management of local crop diversity on-farm and its sustainable utilization for agricultural production.

Local patterns of seed regeneration and trade that occur outside the formal breeding sector (seed exchange networks) can have a strong influence on the genetic diversity and evolution of traditional crop varieties. Despite this, little is known about the extent to which seed exchange networks influence gene flow and genetic structure in traditional crop varieties. Here we study barley (Hordeum vulgare subsp. vulgare) in rural communes of Northern Morocco in 2008 and 2009. We quantified seed regeneration and exchange by farmers within the seed exchange network using structured interviews. Using SSR markers, we also quantified the neutral genetic diversity and structure of a complex of traditional varieties referred to as Beldi that is managed in this exchange network. The majority of farmers (>88 %) report cultivating Beldi. Most seeds of Beldi (70–90 %) are maintained on-farm, while the remainder of seeds are obtained from local markets within the commune. Beldi has high genetic diversity and there is weak but significant genetic structure between communes (FST = 0.031). From SSR marker data there is evidence of a high level of gene flow between communes not reported in interviews. Seeds purchased in local markets likely represent seeds from a larger geographic region, leading to lower genetic structure among communes than expected based on the reported level of on-farm seed regeneration and local sourcing of seed. We discuss the implications of this seed exchange network for the conservation of traditional barley varieties in the study region.

Cowpea cultivation in many countries around the Mediterranean Basin depends on a number of locally adapted populations conserved on-farm at a small scale, rather than on the use of modern varieties. Documentation, characterization and exploitation of traditional local populations could contribute to their conservation and utilization as sources of desirable characteristics. Therefore, a study was conducted to (a) characterize, (b) assess diversity and (c) classify 23 on-farm conserved local cowpea populations based on 32 agro-morphological traits. Investigations on diversity of characteristics related to seed yield, mineral and seed crude protein content as well as on correlations among them were carried out. A relatively high phenotypic diversity was observed. In particular, a high level of within population diversity was found (\( \bar{H}s \) = 0.34) exceeding that among populations’ diversity (Gst = 0.27). Principal component analysis classified the majority of local populations into two groups (mainly according to populations’ seed coat color and eye color), further divided into six subgroups regardless of the populations’ geographical origin. Significant differences were also observed among the populations studied for potassium and calcium, as well as for their seed crude protein content which ranged from 22.14 to 28.37 %. The results show appreciable levels of intra- and inter-phenotypic diversity in on-farm conserved cowpea populations, which indicates the existence of a valuable gene pool for future exploitation in breeding programs.

Brassica oleracea L. is an important species that originated in the eastern Mediterranean area. From there, B. oleracea L. subsp. capitata (L.) DC. convar. botrytis (L.) Alef. var. italica Plenck (broccoli) was introduced into Italy where considerable diversification took place. Several landraces of broccoli, which are highly appreciated by local people for their organoleptic traits, are presently cultivated in both large fields and home gardens. In this study, 10 landraces, 4 synthetics derived from landraces and 4 of the most used F1 hybrids were characterised for morpho-physiological and molecular traits with the principal aim of feeding information into the landrace protection schemes being developed in Italy and into breeding programs. The landraces and derived synthetics are well differentiated from each other and from the F1 hybrids for important agronomic and genetic traits. The principal component analysis based on analysis of morpho-physiological traits showed that overall the 2 main components accounted for 67.74 % of the total variation. The neighbour joining tree, based on 23 microsatellite markers, grouped accessions into 2 main clusters. One includes one synthetic variety and 2 F1 hybrids, while the other cluster includes all the other accessions. There were also several sub clusters. Most of the materials examined showed within-accession diversity. The information gathered in this study can be used to protect landraces through the schemes presently being developed in Italy and to register them in the European common catalogue of “conservation varieties”. In situ (on-farm) conservation of landrace diversity is discussed with reference to landrace use.

Failure to take into account the ecological complexity of landrace populations of crop plants limits our ability to conserve their genetic resources in situ. Soil seed banks are a central feature of the ecology of landrace populations of cassava; their existence has consequences for conservation. Seedlings recruited from seed banks are incorporated by farmers into their stocks of clones of this vegetatively propagated crop, transforming pure clonality into a mixed clonal/sexual reproductive system. Soil seed banks, and farmers’ responses to them, play an important role in maintaining diversity in populations of cassava landraces. In a study combining genetic and ethnobiological approaches, we showed the following: (i) Recruitment from soil seed banks increased diversity of populations at the local scale. At the level of a field, the presence of plants issued from seeds resulted in significantly greater diversity of genotypes and phenotypes than if only individuals planted by farmers had been present. (ii) Farmers’ use of seed banks has enabled indirect ‘exchange’ of locally adapted cassava germplasm between cultural groups, without requiring that groups actually encountered one another and engaged in social exchange of cultivars. (iii) Farmers have responded to catastrophic crop failure by using seed banks to regenerate stocks of clones. This use of seed banks should enable cassava populations to respond to disasters by an increase of genetic diversity, rather than by a narrowing of the genetic base, often feared in such situations.

In Kenya, potato (Solanum tuberosum L.) is the second most important staple food crop after maize and plays a major role in national food and nutritional security. However, there is an inadequate supply of certified seeds, with informal sources (farm-saved, local markets, or neighbors) accounting for 99% of the seeds available to farmers. Most farmers conserve potato seeds on-farm as tubers, using simple methods such as spreading them on the floor, storing them in pits lined with dry leaves and covered with straw, or placing them in gunny bags or wooden crates. Use of diffused light stores (DLS) is the recommended method of seed potato storage, as it lengthens the storage period and reduces storage losses. In the National Potato Research Program in KARI, potato germplasm is conserved in ex situ field genebanks in the form of tubers, which are planted out regularly or as in vitro plantlets.

This study describes the genetic differentiation of a collection of 134 watermelon landrace accessions from Mali, representing red fleshed dessert and white fleshed seed and cooking type watermelons from five regions, plus three commercial dessert type cultivars with red flesh. The material was represented by in total 397 plants and was analysed using 24 microsatellite primer sets, which differentiated 129 alleles across all loci. Analyses of molecular variance ascribed 51 % of the variation among the landrace accessions, and 14 % of the variation could be ascribed to the regions. Partitioning the accessions into use groups (dessert, cooking, seed processing) explained 25 % of the variation. When categorising the accessions further into 10 landrace types, differentiated on the basis of use groups, local accession name, flesh colour and seed phenotype, these landrace types explained 26 % of the variation. Analysis with the software Structure revealed that the accessions with confidence could be separated into two major genetic groups, related to flesh colour (red and white) of the watermelon fruits. The same analysis further indicated that the material may be differentiated into eight genetic sub-groups. One group included again the red fleshed dessert types with local and commercial origin, while the remaining seven genetic sub-groups comprised the white fleshed landrace types used for seed processing and cooking, as well as white fleshed types of one dessert type. Some of the seed and cooking types were to a large extent different and assigned to one genetic sub-group each. A Mantel test, based on geographical and genetic distance matrices showed a positive correlation, indicating that seed exchange has not overcome local adaptation.

On-farm conservation of plant genetic resources for food and agriculture has received strong support worldwide in recent years. It has been justified on appealing assumptions: it complements ex situ conservation, allows co-evolutionary interaction of host–pathogens and crop–weed complexes, and involves local knowledge systems. This article illustrates how on-farm conservation being set for its sake is extremely difficult under farmers’ dynamic management of plant genetic resources based on sorghum. The dynamics of their management could be explained by continued introduction, displacement, loss and maintenance of aboriginal landraces that have distinct functional attributes, patch-occupancy and relative abundance profiles. Such management and hence the dynamic landrace demography has largely been triggered by co-evolving biophysical stresses, spatial and seasonal variations. The best viable alternative to support farmers’ management of genetic resources is to link conservation to crop improvement both to enhance on-farm genetic diversity and make the biophysical environment a comfortable home for the plant genotypes.

Genetic diversity in 12 landraces of bambara groundnut (Vigna subterranea), an indigenous African legume, was evaluated using Random Amplified Polymorphic DNA (RAPD) markers. DNA from individuals of each landrace was also analysed to determine the level of heterogeneity within landraces. RAPDs revealed high levels of polymorphism among landraces. The percentage polymorphism ranged from 63.2% to 88.2% with an average of 73.1% for the 16 RAPD primers evaluated. The construction of genetic relationships using cluster analysis groups the 12 landraces in two clusters. RAPDs are useful for the genetic diversity studies in V. subterranea and can identify variation within landraces.

Ethiopia is a centre of diversity and hosts rich genetic resources of tetraploid wheats. Through time, the wheat materials were subject to genetic erosion. Closer investigation was made to assess the status of loss, and identify the possible causes by studying two districts from East Shewa. Information from primary and secondary sources was reviewed and analysed. Farmers identified 26 tetraploid wheat landraces (21 from Akaki and 17 from Ejere), which were once widely grown in the area. Of these, only six were currently available. Compared to the formerly available number of landraces, the loss of diversity in the study area was estimated to be 77%. In the Ejere locality, the loss was 100% before the launching of the on farm landrace conservation programme, and for Akaki it was 95%. Major factors that contributed to the loss include: (1) introduction and expansion of bread wheat varieties; (2) expansion of tef; (3) lack of a mechanism to re-supply seeds of tetraploid wheat landraces; (4) decline in size of landholdings; (5) changes in land use and cropping patterns; (6) lack of policy support; and (7) expansion of improved tetraploid wheat varieties. The lessons from this study underscore the importance of strengthening the local seed supply system as a prerequisite for sustaining on farm conservation of landraces. Moreover, it is necessary to initiate diversity studies focusing on the distribution and status of tetraploid wheat landraces across the country. Complementing these by molecular analyses is essential in order to assess the genetic distinctness of the landraces.

Genetic variability within and among eight landrace populations of tea (Camellia sinensis (L.) O. Kuntze) located in southern Kyoto, Japan, was surveyed with six microsatellite markers. The average number of alleles per locus was 3.83 to 4.67 for landrace populations, whereas the corresponding value among modern cultivars and breeding lines was 6.63. Expected heterozygosity values averaged over loci within landrace populations ranged from 0.498 to 0.723. A similar level of variation, 0.682, was observed for cultivars and breeding lines. High fixation index values (0.177–0.417) for each population are consistent with biparental inbreeding within the population. Genetic differentiation among local populations was extremely low with F ST = 0.062, although AMOVA revealed significant differentiation among landrace populations. We propose that these populations share a common ancestral gene pool and that some degree of artificial selection within each population has been performed by local farmers. Neighbor-joining analysis revealed that genetic relationships among populations reflect geographical location of populations. This might result from more frequent genetic exchange by nearby farmers.

Adaptability of traditional agricultural systems is suggested by their success over time, but documentation of how this happens is rare. This paper shows how genetic diversity in a rice landrace enables rice farming system of northern Thailand to adapt to a constraint of an insect pest, microenvironments of mountainous landscape and people’s different tastes in rice. Resistance to laboratory-reared gall midge varied among accessions the rice landrace Muey Nawng and gall midge populations. Higher rice yield in farmers’ fields reflected adaptation to local environment as well as resistance to gall midge. Microsatellite variation of the accessions correlated negatively with their gall midge resistance, but there was also variation in heading time and endosperm starch. Presence of non-waxy endosperm in glutinous rice provides opportunity to select for rice that is cooked into non-glutinous rice preferred by minority groups who live at higher elevations, where the gall midge is emerging as a new threat, possibly because of climate change. These data show how genetic diversity of a rice landrace coupled with seed management by farmers enabled a rice farming system to adapt to the varied microenvironment of a mountainous landscape under the constraint of an insect pest and people’s different tastes in rice.

In Nepal, in traditional rice farming systems many diverse landraces are grown in all of the rice agro-ecosystems from low to high altitude. Three case study sites were selected to represent the major rice agro-ecozones: Bara (100–150 m) for the low-altitude terai (plain); Kaski (700–1,206 m) for the mid-hill zone; and Jumla (2,200–3,000 m) for the high-hill zone. The diversity in rice varieties was compared in these three sites and nine survey villages in a series of surveys conducted in 1998, 1999 and 2006. The level and distribution of diversity on farm varied with the physical and socio-economic settings of the farming communities. The mid-hill site (Kaski) had the highest rice landrace diversity. This was adapted to the diverse agro-ecosystems found there and there was equal diversity in Kule khet (irrigated lands by seasonal canals) and Sim khet (marshy wet land). The next most diverse system was Nicha khet (irrigated lowlands) in Bara, the low-altitude site. The high-hill site (Jumla) had the lowest rice diversity. Across all sites many of the landraces were rarely grown and then only in small areas, reflecting the specialized uses to which they were put. At all sites the most common single landrace occupied less than half of the rice area. Resource-rich farmers were the more important custodians of on-farm rice varietal diversity across the sites. There was more rice diversity in favourable environments than in less favourable ones. This was true whether diversity was measured across sites or across rice domains within sites.

Carbon footprint (CF) quantification of major rice production systems (RPSs) is a prerequisite for developing strategies for climate change mitigation in agriculture. Total life cycle greenhouse gas emissions (LC-GHGs) from rice production to consumption might provide precise CFs for RPSs. Therefore, we assessed three segments (pre-farm, on-farm, and post-farm) of LC-GHGs under five major contrasting RPSs, i.e., aerobic rice (AR), shallow lowland rice(SLR), system of rice intensification (SRI), ...

Farmers who still cultivate bambara groundnut (Vigna subterranea) rely on landraces and seed retained from previous harvests. Given that the crop is typically cultivated in semi-arid regions, seed quality of farmers' retained seed might be compromised due to water stress experienced by maternal plants during production. The aim of this study was to determine the effect of water stress on maternal plants on subsequent seed quality of bambara groundnut. A single bambara groundnut landrace was characterised into four distinct selections based on seed coat and speckling colour. Initial seed quality (viability and vigour) was evaluated prior to planting seed in a field trial under irrigated and rainfed conditions. Final yield and yield components were determined at harvest. Thereafter, seed quality (viability and vigour) of progeny of different landrace selections was evaluated. Yield was lower under rainfed than irrigated conditions. Overall, subsequent seed quality showed improvement from initial seed quality of the original seedlot. Seed viability was higher in seeds produced under irrigated than rainfed conditions. Seed quality of bambara groundnut may be reduced underwater-limited production conditions. Seed enhancement practices such as priming may assist farmers to achieve better emergence. In the long term, seed production should be done under optimum conditions in order to achieve high-seed quality.

Germplasm conserved as seeds in genebanks requires regular regeneration. In this process, selection and genetic drift may cause loss of genetic diversity from accessions. In the case of selfing crops, separation of distinct lines into different accessions may be an efficient strategy to avoid these negative effects. In order to evaluate the applicability of this method for collection management, knowledge about the level of intra-accession genetic diversity is required. By means of AFLP analysis intra-accession variation was investigated in two cultivars, two landraces and two wild populations of ex situ conserved barley germplasm. In the total sample of 216 individuals analysed (36 per accession), 22 genotypes were observed based on 104 polymorphic loci. The number of genotypes detected ranged from 1 to 3 per accession, except for a Nepalese landrace that revealed 12 genotypes. An UPGMA cluster analysis grouped the genotypes unambiguously into the accession they belonged to and genotypes within accessions were generally found to be closely related. In order to determine the repeatability of the results obtained, 11 individuals belonging to 4 genotypes from the Nepalese landrace were scored for a second set of AFLP markers. Matrices of genetic distances calculated for the two AFLP datasets were found to be highly correlated (r = 0.9346, P < 0.001). Separation of genotypes into different accessions was considered a relevant option only for the Nepalese landrace. Analysis of molecular variance indicated that this accession could be well divided into 8 distinct lines. Further implications of the results for genebank practices are discussed.

Fonio (Digitaria exilis (Kipp.) Stapf, white fonio; Digitaria iburua Stapf, black fonio), is one of the oldest native, but neglected, cereal crops of cultural, nutritional and economic importance of West Africa. To gather key information that will help to preserve its diversity in Benin and develop modern and well-adapted varieties that could meet both farmers’ and consumers’ needs, a participatory research appraisal survey was conducted in 15 villages randomly selected in the fonio production zone of Benin. Altogether 15 farmer-named landraces were recorded and further grouped into five morphotypes of which four belong to D. exilis and one to D. iburua. The number of landraces inventoried varied from one to five per village (three on average) and from one to three per household. Their distribution and extent are presented and their importance in terms of genetic resources conservation is discussed. Farmers’ preferences for fonio varieties were assessed and eight preference criteria (earliness, culinary characteristics, ease of processing, productivity, facility of harvesting, grains size, storability, and drought tolerance) were identified and prioritised according to the gender. The traditional seed supply system and on-farm seed maintenance were investigated. Two plant species (Digitaria horizontalis Willd. and Digitaria longiflora (Retz.) Pers.) were reported by farmers as putative parents of cultivated fonio. Of these, only Digitaria longiflora has interesting agronomic traits (erect habit; stem long, big, strong and resistant to lodging; panicle long and rich in grains; large sized seeds) that are useful for the improvement of cultivated fonio. In the entire study area, the crop is under threat. Therefore, concerted efforts should be deployed to preserve its diversity and increase its use to combat food insecurity and malnutrition in Benin.
