2003年3月31日,兰州市动物园1对斑马突然暴亡,经尸体剖检和实验室检验,从采取的脏器及组织分离出大量高致病力的大肠杆菌,经中国兽医药品监察所对2株大肠杆菌血型O、K抗原鉴定,结果一株为O2和O74混合型,另一株为O6;但K抗原血清鉴定未获结果。初步确诊该对斑马死亡是高致病力的大肠杆菌所致。  相似文献

Tick salivary proteins are promising targets for the development of anti-tick vaccines. Recently, we described two paralogous anti-complement proteins, called Ixodes ricinus anti-complement (IRAC) proteins I and II, that are co-expressed in tick I. ricinus salivary glands. However, our previous attempts to immunize rabbits against IRAC via infection with recombinant Bovine herpesvirus 4 (BoHV-4) vectors invariably failed although both recombinants expressed high levels of functional IRAC proteins in vitro. As IRAC are soluble monovalent antigens, one of the possible explanations is that monovalent ligation of the B-cell receptor induces receptor activation but fails to promote antigen presentation, a phenomenon that is thought to induce a state of B-cell tolerance. In the present study, we tried to increase IRAC immunogenicity by expressing them as oligovalent antigens. To this end, IRAC were fused to membrane anchors and BoHV-4 vectors expressing these recombinant forms were produced. The immunization potentials of recombinant viruses expressing either secreted or transmembrane IRAC proteins were then compared. While the former did not induce a detectable immune response against IRAC, the latter led to high titres of anti-IRAC antibodies that only marginally affected tick blood feeding. All together, the data presented in this study demonstrate that the immunogenicity of a soluble antigen can be greatly improved by anchoring it in membrane.  相似文献

The aim of this work was to determine levels of carcinoembryonic antigen (CEA) and cancer antigen (CA 15‐3) in the blood serum of 45 bitches. A modified procedure was used to determine the CEA and CA 15‐3 markers with the human kits using the radioimmunoassay method. Samples collected from extirpated tumour of mammary glands were histologically processed and classified as per WHO guidelines. The average age of animals with tumour was 10.00 ± 2.2 years; for healthy bitches average age was 4.2 ± 3.2 years. Values of CEA and CA 15‐3 were considered positive, if they exceeded 0.23 ng mL?1 and 7 IU mL?1, respectively. Average levels of CEA in the tumour group were 0.25 ± 0.06 versus 0.20 ± 0.03 in healthy bitches (P = 0.0001). The average CA 15‐3 value in bitches with tumour was 8.58 ± 1.27 versus 5.14 ± 1.34 in healthy animals (P < 0.0001).  相似文献

本文综述了棘球蚴病疫苗的研究进展。棘球蚴病基因工程重组抗原疫苗研究已取得突破性进展,实验室研究试验羊减囊率达到90%以上,田间试验减囊率在80%以上,在我国即将商品化应用。核酸疫苗由于安全有效、操作简单、价廉易得、便于贮存与运输、省时省力,具有极大的潜在应用价值。多肽疫苗由于不存在毒力返祖或灭活不完全的问题,是棘球蚴病疫苗未来发展的方向之一。  相似文献

禽巴氏杆菌荚膜多糖-蛋白载体抗原的研究   总被引:4,自引:1,他引:3  
用EDC法和CNBr法分别交联禽巴氏杆菌荚膜多糖(CPS)与破伤风类毒素(TT),经Sephadex-G150柱层析和薄层层析鉴定表明;两种方法均获得一种大分子的CPS-TT结合物,该结合物免疫鸡的血清经二巯基乙醇(2-ME)处理后用间接血凝试验(IHA)检测出较高滴度的IgG抗体和一定量的IgM抗体,抗体消长情况监测结果;IgG抗体的峰值(4.88)在免疫后第21天,其后缓慢下降,持续至第24周左右。IgM抗体的峰值(4.26)在第14天,其后陡降,至第8周降至阴性血清水平,与对照组差异极显著(P<0.01)。第8周和第24周时测出较强的二次反应和回忆反应。重复试验测出的IgG、IgM抗体滴度与本试验结果相近,第4周时攻毒保护率分别为100%和75%,且IgG抗体滴度与攻毒保护率的相关性较好,1:16以上可获得全保护。试验结果表明交联方法稳定,重复性较好,制备的CPS-TT载体抗原实现了CPS由Ti抗原向Td抗原的转换。为研究一种新型的禽霍乱载体菌苗提供了理论依据和实验手段。  相似文献

Serologically coronavirus free kittens were placed in 2 catteries with a history of feline infectious peritonitis (FIP), each cattery representing 1 of the 2 different predominant clinical characteristics of FIP - effusive and granulomatous. The kittens were clinically observed for 100 days. A 100% morbidity and a 90% mortality was observed. The first signs were observed after 14 and 27 days respectively. The clinical pattern of the disease was similar in all kittens and showed a pattern of recurrent periods of conjunctivitis, upper respiratory and gastrointestinal signs. Once developed, wasting and signs of CNS disturbances were consistent. The “effusive strain” had a 2 weeks earlier onset of signs and death, and a 40% outcome of effusive FIP. Mean survival times during the observation period were 57 ±26 and 57 ±16 (mean ±SD in days), respectively. The death rates were similar in both groups. Feline coronavirus (FCoV) antigen was immunohistochemically detected using indirect immunofluorescence and was present in all kittens and in 93% of the 5 investigated organs (lung, liver, spleen, kidney, and mesenteric lymph node).  相似文献

以原头节可溶性粗抗原经Sephadex G-200层析纯化抗原为包被抗原,兔抗羊IgG-HRP结合物为显色剂,建立检测羊脑多头蚴病血清抗体的Dot-ELISA,并以ELISA作平行对照。结果,粗抗原和层析纯化抗原检测86头份羊脑多头蚴病阳性血清,其敏感性分别为94.18%和93.02%,两种抗原的敏感性无显著差异;检测122头份绦虫蚴病阴性血清,18头份棘球蚴病阳性血清,35头份细颈囊尾蚴病阳性血清,其特异性分别为90.29%和95.43%。两种抗原的特异性差异显著。Dot-ELISA和ELISA两种方法的符合率为100%。层析纯化抗原比粗抗原的特异性有了明显提高,而敏感性没有降低。层析纯化抗原和操作术式都具有良好的重复性,Dot-ELISA和ELISA对比试验结果相近,且具简便、快速及不需要复杂设备等优点,是一种检测羊脑多头蚴病血清抗体的理想方法。  相似文献

A 19-year-old Quarter Horse gelding was evaluated for respiratory distress and a rapidly enlarging retropharyngeal mass. Initial evaluation revealed severe respiratory distress, and a large, firm mass, visibly appreciable as 12 × 12 cm, in the left retropharyngeal and perilaryngeal region, with surrounding left and right retropharyngeal swelling. No significant abnormalities were present on complete blood count and serum biochemistry analyses. Endoscopy revealed severe pharyngeal collapse restricting airflow without gross abnormalities of the pharyngeal mucosa other than inflammation and irritation. A multilobular retropharyngeal mass, diffusely heterogeneous in echogenicity, was present adjacent to, but not occluding, the carotid artery as assessed by ultrasonography. Initial needle aspirate suggested lymphoma. Tissue biopsy and histopathology confirmed a round cell tumour. A temporary tracheotomy was performed to provide respiratory relief, and the horse was managed on oral antibiotics and anti-inflammatory medications while awaiting histopathological results. The decision was made to humanely euthanise the horse after biopsy results indicated lymphoma. Definitive diagnosis of T cell rich, large B cell lymphoma was made by combination of cytology, immunohistochemistry and molecular clonality PCR (PARR) testing. Lymphoma should be considered in horses with focal masses of the retropharyngeal region. Although treatment was not pursued, PARR testing was successful in this case and may be helpful for accurate characterisation of lymphoma in horses to more precisely determine prognosis and the most effective treatment plans, as it has been in human patients and small animals.  相似文献

利用抗伊氏锥虫表膜抗原的McAb双抗体夹心法,检测了48匹健康马(骡)、5匹人工接种伊氏锥虫马(骡),9匹先用锥虫弱毒株免疫后用强毒株攻毒马(骡)共278份血清的循环抗原(CA),同时用常规间接ELISA检测了相应的循环抗体(CAb)。结果: 278份血清的CA、CAb阳性率无显著差异,但其阳性检出时机不完全吻合;人工接种马(骡)的CA阳性检出率高于CAb,于接虫后经1-2个CA高峰而死,濒死前4/5动物的CA呈现阳性,免疫马(骡)在攻虫前CA即可呈现阳性,多经1-3个CA高峰后逐渐转阴;人工接种马(骡)的CA水玉高于免疫马(骡),二次攻虫后的CA水平高于首次。  相似文献

本实验应用生化萃取技术和体外人工培养技术,制备了肌旋毛虫匀浆(HO)抗原和排泄—分泌物(ES)抗原,并对两种不同来源抗原的部分生化特性和免疫学特性进行了比较。HO抗原通过SephadexG—200又分为第一峰(FP)和第二峰(SP)。在5~20%的聚丙烯酰胺梯度凝胶电泳和薄层聚丙烯酰胺凝胶等电聚焦电泳上,ES和FP抗原出现许多条电泳区带。ES、FP和HO抗原对小鼠的保护力(肌幼虫减少率)分别为78%,76%和46%。  相似文献

四环素类抗生素属小分子半抗原物质 ,必须与大分子物质结合才能产生免疫原性。本试验采用重氮化法合成土霉素 牛血清白蛋白 (BSA OTC)及土霉素 鼠血清白蛋白 (RSA OTC)人工抗原 ,用紫外扫描对其进行鉴定。以BSA OTC免疫家兔制备抗血清 ,以RSA OTC为包被抗原 ,采用双向免疫扩散法和ELISA方法对抗血清进行鉴定。结果表明用重氮化法合成的人工抗原对四环素类药物有特异的抗原抗体反应 ,抗血清效价达 1 2 8× 1 0 5,完全能够满足四环素类药物残留的ELISA检测要求  相似文献

探讨热修复中不同pH缓冲液与不同修复温度对鸡睾丸组织免疫荧光结果的影响。选取可被抗体标记的阳性组织,采取不修复、胰蛋白酶修复、不同条件的热修复进行比较。结果表明,抗原修复是必要的,本试验中以缓冲液pH 7.0、97℃下的热修复效果最好。  相似文献
