Milk and serum samples from 35 dairy herds in 17 states were evaluated for cow- and herd-level Mycobacterium avium subspecies paratuberculosis (MAP) antibody test agreement. Evaluation of 6,349 samples suggested moderate agreement between milk and serum enzyme-linked immunosorbent assay (ELISA) results, with a kappa value of 0.50. Cow-level sensitivity (Se) for 18 dairy operations with 1,921 animals was evaluated relative to fecal culture results. At the cow level, the milk ELISA relative Se was not significantly different from that of the serum ELISA (21.2 and 23.5%, respectively). Logistic regression models revealed a positive association between lactation number and milk ELISA status. Non-Holstein cows were more likely to test milk ELISA positive than Holstein cows. Cows in the first 2 weeks of lactation and after week 45 of lactation were more likely to test milk ELISA positive than cows between 3 and 12 weeks of lactation. Milk production > 80% of herd average was negatively associated with testing milk ELISA positive. Animals in the West and Midwest regions were less likely than animals in the Southeast region to test ELISA positive by either test. Estimates for herd-level sensitivity for the milk and serum ELISA, relative to fecal culture results, ranged from 56 to 83%. At the cow and herd levels, milk ELISA performed equivalent to serum ELISA using fecal culture as a reference for MAP infection and has the advantage of decreased labor costs on farms that use Dairy Herd Improvement Association testing.  相似文献

Isotype-capture ELISAs for BCV-specific IgA and IgM were developed and tested on milk and serum samples from Swedish cattle. The capture ELISAs showed higher sensitivity than indirect ELISAs for detection of BCV-specific IgA and IgM. In the capture ELISAs the agreement between detection in milk and serum samples was 94% for IgA and 86% for IgM. The correlation between log(10) titres in milk and serum was r=0.82 (P<0.001) for IgA and 0.84 (P<0.001) for IgM. Milk seemed a better target than serum for diagnosing specific IgA at low levels. There was no variation in the isotype-specific BCV antibody titres between healthy quarters of the same udder, but subclinical mastitis was associated with higher levels of IgA antibodies and weak false IgM positive reactions in undiluted milk. Bovine IgA and IgM antibodies in milk and serum showed high stability towards freezing and thawing and storage at room temperature.The antibody responses to BCV were followed in milk and serum from six dairy cows and in serum from four calves for a period of 1 year after an outbreak of winter dysentery (WD). In this outbreak some animals became reinfected with BCV. The IgA and IgM capture ELISAs differentiated between primarily BCV infected and reinfected animals. In the primarily infected cattle, IgM antibodies were first detected in milk and serum four to nine days after the first WD symptoms observed, and were subsequently detected for at least 2-3 weeks. IgM was also detected in the reinfected cows, but mostly at lower levels and for a shorter period of time than in the primarily infected animals. In milk, however, the IgM response of the reinfected cows was detected for a longer period of time than in serum. Six months after the outbreak, IgA was still detected in both serum and milk of all six cows and also in serum of one calf. The reinfected cows showed higher and more long-lasting peak levels of IgA in milk and serum than the primarily infected cows, indicating boosting of the IgA response.  相似文献

Bulk and individual milk samples from 117 herds located in Brittany (west France) were used to assess: (i) the performance characteristics of an indirect enzyme‐linked immunosorbent assay (ELISA) applied to individual milk for the detection of antibodies to bovine viral diarrhoea virus (BVDV); and (ii) the relationship between the bulk milk result obtained from this test and the within‐herd prevalence of antibody‐positive lactating cows. This ELISA test was based on a monoclonal antibody directed against non‐structural protein NS2‐3 of pestiviruses. At the individual level, based on 1113 matched milk/serum samples, the sensitivity and specificity of this test applied to milk, compared with the virus neutralization test on serum, were 95.0 and 97.7%, respectively. At the herd level, the relationship between the optical density percentage (OD%) of bulk milk and the within‐herd prevalence of antibody‐positive lactating cows was assessed using the receiver operating characteristics (ROC) analysis. Classes of OD% of bulk milk were determined so that they were associated with minimum intraclass and maximum between‐class variances of within‐herd prevalence of antibody‐positive cows. The ROC analysis resulted in two classes of bulk milk results corresponding to different expected levels of within‐herd prevalence. Herds with an OD% of bulk milk <75% and ≥75% had a mean observed prevalence of antibody‐positive cows of 8.9 and 60.6%, respectively. Herds with a bulk milk result <75% were expected to be BVDV free, whereas large variations in prevalence of antibody‐positive cows existed in the herds with OD% ≥75%. The test described in this study is suitable to identify herds likely to have a low prevalence of BVDV antibody‐positive cows.  相似文献

A retrospective longitudinal study was conducted to examine whether the exposure of dairy herds to bovine virus diarrhoea virus (BVDV) affected udder health. All Norwegian dairy herds that had experienced a marked increase in the BVDV antibody titres in bulk milk (from a level corresponding to an optical density (OD) <0.25 to >0.55, as determined by an indirect enzyme-linked immunosorbent assay) between two nation-wide herd screening examinations carried out late in 1992 and 1993, respectively, were considered to have been exposed to BVDV during the period between the examinations. The reference group included all dairy herds in which the bulk milk was BVDV antibody-negative or had only very low titres of BVDV antibodies (OD <0.25) each year from 1992 to 1995. The annual incidence rate of clinical mastitis, the bulk-milk somatic-cell count, and the annual rate of culling because of mastitis in the herds, were compared in the year of BVDV exposure (1993) as well as in a period prior to exposure (from 1988 to 1992) and two years following the year of exposure. In herds exposed to BVDV, there was a 7% increase in the incidence rate of clinical mastitis in the year of exposure, as compared with the nonexposed herds. No significant changes attributable to BVDV exposure were observed in the bulk-milk somatic-cell count or in the rate of culling because of mastitis.  相似文献

Antibodies against non-structural protein 3 (NS3, p80) of bovine viral diarrhoea virus (BVDV) were determined in milk from cows vaccinated with an inactivated BVDV vaccine and compared to serum antibody levels. Animals in one herd were vaccinated with an inactivated BVDV vaccine according to the standard protocol and animals from a second herd with an intensive schedule. Serum and milk samples were tested for BVDV NS3 antibodies using five commercial ELISAs. With a few exceptions, vaccination according to the standard schedule did not induce BVDV NS3-specific antibodies in serum or milk. However, after vaccination according to the intensive schedule, anti-NS3 antibodies were detected for a short time in serum and, to a lesser extent, in milk. Bulk milk was a suitable substrate for BVDV monitoring of herds vaccinated with the inactivated BVD vaccine.  相似文献

The performance characteristics of a blocking ELISA test applied to serum and individual milk for the detection of antibodies to bovine viral diarrhoea virus (BVDV) were assessed using 1189 matched milk/serum samples collected from cows of 42 dairy herds located in Brittany (west of France). This test was based on a monoclonal antibody directed against non-structural protein NS2-3 of pestiviruses. All tests were performed blind. For each type of sample, negative/positive cut-off values were determined using receiver operating characteristic (ROC) analysis. Sensitivity and specificity were estimated using the virus neutralisation test as a reference. For sera, the ROC analysis provided a negative/positive inhibition percentage cut-off value of 50% giving a sensitivity and a specificity of 96.9 and 97.8%. For individual milk samples, the cut-off was fixed at 30%, leading to a sensitivity and a specificity of 96.9 and 97.3%. Using this test, a good overall agreement was found between results obtained on matched milk/serum samples (Kappavalue=0.95). The present results indicate that this blocking ELISA test is reliable enough for use in a mass screening and control scheme on BVDV.  相似文献

On 81 farms blood samples were taken from adult dairy cattle, on pasture in October 1986 and after stabling in December 1986, to measure antibody titres against the nematodes Dictyocaulus viviparus, Cooperia spp. and Ostertagia spp., and the trematode Fasciola hepatica, and serum pepsinogen values. Faecal samples, collected in October, were examined to confirm the presence of parasites by means of egg counts and larval identifications. From December until the end of the stabling period, dry cows were either treated with albendazole or left untreated in alternate sequence of calving date. Treated cows produced 132.9 kg milk per cow per lactation more than untreated cows (P less than 0.01). Fat and protein percentage were not significantly influenced by anthelmintic treatment. The mean herd milk yield response to treatment varied from -889 to +1231 kg milk per cow per lactation. There was a significant between-herd variation in antibody titres against nematodes and in pepsinogen values. However no significant correlations between these parameters and the mean herd milk yield response to treatment were found.  相似文献

A longitudinal study was performed to investigate the variations of Neospora caninum antibody levels in individual milk during lactation as well as the association between antibody levels in serum and milk. Serum and milk samples of 15 milking cows were collected between February 2003 and September 2004 in three smallholder dairy farms in Khon Kaen province in northeast Thailand. All samples were analyzed for presence of antibodies by an N. caninum iscom ELISA test kit and the results were given as percent positivity (PP). The effects of time between calving and sampling, lactation number, and season on milk and serum PP were studied using Generalized Estimation Equations methods. All cows were antibody positive in either milk or serum at the first two consecutive samplings. Although serum and milk PP varied considerably, milk PP was consistently positive throughout the study. Cows of all lactation groups had a higher adjusted mean of milk PP at calving compared to later months after calving although the only significant difference was in first lactation. Serum and milk PP were always lower in first lactation than in second and later lactations. An adjusted mean of milk PP for cows classified as having serum PP> or =55 was significantly (P<0.05) higher than that of cows classified as having lower serum PP. Our results indicate that individual milk can be an alternative material to demonstrate presence of N. caninum antibodies in lactating cows.  相似文献

An enzyme linked immunosorbent assay (ELISA) was developed to detect antibody to bovine viral diarrhoea virus (BVDV) in bovine serum. The ELISA results were compared with those of the serum neutralisation test (SNT) using serums from 6 experimentally infected calves bled at intervals from 0 to 154 days postinfection and 886 field samples. The optical density (OD) produced by a single dilution of test serum was compared with a standard curve and the result expressed in ELISA units. Despite wide variation between absolute ELISA and SNT results, an agreement of 97% was obtained when reciprocal SNT titres greater than or equal to 8 and ELISA units greater than or equal to 10 were taken as indicative of a specific reaction. The ELISA was shown to be an efficient method of measuring antibody in bovine serum samples and would assist in any large scale screening of cattle herds for BVDV antibody.  相似文献

OBJECTIVE: To determine whether passively acquired antibodies prevent development of a protective immune response to live virus in calves. ANIMALS: 18 calves. PROCEDURES: Calves were caught immediately after birth and tested free of bovine viral diarrhea virus (BVDV) and serum antibodies against BVDV. Within 48 hours, 12 calves were fed colostrum that contained antibodies against BVDV and 6 calves received BVDV antibody free milk replacer. Three milk replacer fed and 6 colostrum fed calves were exposed to virulent BVDV2-1373 at 2 to 5 weeks of life when passively acquired serum antibody titers were high. After serum antibody titers against BVDV had decayed to undetectable concentrations (at 7 to 9 months of age), the 3 remaining milk replacer fed calves, 6 colostrum fed calves previously exposed to BVDV2-1373, and 6 colostrum fed calves that had not been exposed to the virus were inoculated with BVDV2-1373. RESULTS: Passively acquired antibodies prevented clinical disease in inoculated colostrum fed calves at 2 to 5 weeks of life. Serum antibody titers did not increase in these calves following virus inoculation, and serum antibody titers decayed at the same rate as in noninoculated colostrum fed calves. Inoculated colostrum fed calves were still protected from clinical disease after serum antibody titers had decayed to nondetectable concentrations. Same age colostrum fed calves that had not been previously exposed to the virus were not protected. CONCLUSIONS AND CLINICAL RELEVANCE: A protective immune response was mounted in calves with passive immunity, but was not reflected by serum antibodies titers. This finding has implications for evaluating vaccine efficacy and immune status.  相似文献

The purpose of this study was to determine the effects of bovine leukemia virus (BLV) infection on production, reproduction and longevity in dairy cattle. The study population was a commercial Holstein dairy herd of approximately 400 milking cows. Cattle were tested for antibodies to BLV at least annually for three years and when culled. Four groups of culled cows were compared: seronegative cows (n = 79), seropositive cows without lymphocytosis (n = 176), seropositive cows with lymphocytosis (> or = 9,000 lymphocytes/microliter) (n = 74), and seropositive cows with lymphosarcoma (n = 29). Seropositive groups of cows were bred more times and had longer calving intervals than seronegative cows. The seropositive groups had greater 305-day ME (mature equivalent) FCM (3.5% fat-corrected milk) per lactation and were older when culled than seronegative cows. However, the percent fat per lactation was greater in seronegative cows. In the last complete lactation, differences in 305-day ME FCM, days open and cull age between groups were reduced and none were significant (p > 0.05). In the cull lactation, only cows with lymphocytosis had reduced milk production relative to seronegative cows, although this difference was not significant. After adjustment for initial production and reproductive values, only seropositive nonlymphocytotic cows were culled at a significantly older age than seronegative cattle. Lymphocytotic cows were culled four months younger on average than nonlymphocytotic seropositive cows. Hence, BLV infected cows had greater milk production on average than uninfected cows. Adverse effects of BLV infection were primarily limited to lymphocytotic cows which were culled earlier and had reduced milk production in the cull lactation.  相似文献

Neutralising serum antibodies against bovine virus diarrhoea virus (BVDV) were monitored for three years in 35 cattle that were infected with the virus as calves; 24 of the calves were inoculated intramuscularly or intranasally, and 11 contracted the infection naturally. All the experimentally infected calves seroconverted within 14 to 28 days after inoculation, and all the animals still had high serum levels of antibodies to BVDV three years after infection. Determinations of antibody levels in milk and blood samples excluded the possibility that the calves had been reinfected with BVDV during the study.  相似文献

Bulk tank milk samples, collected twice with a 1 year interval, from 213 Swedish dairy herds with no vaccination programme against bovine virus diarrhoea virus (BVDV), were tested for antibodies to BVDV using an indirect enzyme-linked immunosorbent assay. The herds were classified into four different BVDV groupings based on changes in the estimated prevalence of BVDV antibody-positive cows in the herds. The estimated mean prevalences of BVDV antibody-positive cows were maintained as > 80% in 58 (27.2%) and as < 10% in 84 (39.4%) of the herds. A recent introduction of the infection was deemed to have occurred in seven (3.3%) of the herds studied. The BVDV groups were compared with regard to parameters related to disease and fertility at herd level. Relationships were assessed using logistic and ordinary linear regression analyses. The risks for clinical mastitis, retained placenta and oestrus-stimulating treatments were higher and the calving intervals were longer in BVDV infected herds, i.e. those herds with an increasing or maintained high prevalence of BVDV antibody-positive cows.  相似文献

The aim of the study was the assessment of rise and persistence of neutralizing antibodies (nAb) to bovine viral diarrhea virus (BVDV) and border disease virus (BDV) after a two step vaccination using an inactivated BVDV/BDV (Mucobovin) and a modified live BVDV vaccine (Vacoviron). In a first experiment eight heifers were kept in isolation and were serologically surveyed regularly over a three year period after vaccination. The same experiment was done with 80 vaccinated cattle kept under field conditions. Neutralizing antibody titres were monitored using homologous as well as heterologous BVDV and one BDV strain, respectively. Maximum titres were obtained two to three months after vaccination. During the three years of monitoring the antibody titres decreased but never fell below the detection limit. This slow antibody regression demonstrates that a single two step vaccination elicited high nAb titres which persist over at least three years. These results might serve as a decision tool when considering the necessity and time of revaccination of cattle, which have been vaccinated using the two step method.  相似文献

A dynamic, stochastic model simulating the technical and economic consequences of bovine virus diarrhoea virus (BVDV) infections for a dairy cattle herd for use on a personal computer was developed. The production and state changes of the herd were simulated by state changes of the individual cows and heifers. All discrete events at the cow level were triggered stochastically. Each cow and heifer was characterized by state variables such as stage of lactation, parity, oestrous status, decision for culling, milk production potential, and immune status for BVDV. The model was controlled by 170 decision variables describing biologic and management variables including 21 decision variables describing the effect of BVDV infection on the production of the individual animal. Two markedly different scenarios were simulated to demonstrate the behaviour of the developed model and the potentials of the applied modelling principle. The validation problem in relation to the model was discussed. A comparison between real and simulated data using data from a published case report was shown to illustrate how user acceptance can be obtained.  相似文献

Levels of passively transferred maternal antibodies to three canine viruses, rabies virus (RV), canine distemper virus (CDV) and infectious canine hepatitis (ICH) virus, in serum specimens from 14 fetal pups and in serial serum specimens collected up to 45 days after whelping from 14 neonate pups were compared with levels of antibodies to these viruses in milk and sera collected concurrently from their respective dams. Radioimmunoassays using RV-, CDV- and ICH virus-specific antigens showed that sera from all fetal pups had detectable levels of antibodies to all three canine viruses and ICH neutralising antibodies were detected in sera from 10 of the 14 fetal pups. As the time after whelping increased, titres of RV-, CDV- and ICH virus antibodies measured by radioimmunoassay and ICH virus neutralising antibody tests in serially collected specimens of milk from dams rapidly decreased, while titres of the antibodies in serum specimens from newborn pups in their litters steadily decreased. Individual fetal and newborn pups with a high titre of antibody to one virus also had high titres to the other two viruses, although a wide range of titres was observed among pups in each of the litters studied. Markedly higher titres of antibody to all three viruses were observed in serially collected specimens of sera from dams than in sera from fetal and newborn pups in their litters. Results show that maternal RV, CDV and ICH virus antibodies are transferred from dams to pups in utero and by nursing. Levels of these maternal virus-specific antibodies in newborn pup sera decreased at similar rates as time after whelping increased.  相似文献

AIMS: To assess the sensitivity and specificity of a bulk tank milk (BTM) antibody enzyme-linked immunosorbent assay (ELISA) to detect likely infection of a dairy herd with bovine viral diarrhoea virus (BVDV). The ELISA was subsequently used to estimate the prevalence of likely infected herds in parts of the North Island of New Zealand. METHODS: BTM samples from 724 randomly selected dairy herds in the Waikato, Bay of Plenty and Northland regions of New Zealand were tested for BVDV antibodies. From this group, 20 herds were again randomly selected from each of the quartiles of the ELISA percentage inhibition (%INH) result. From each participant herd, serum from 15 randomly selected calves aged 6-18 months and 15 cows was collected and tested using an indirect blocking ELISA for BVDV antibodies. RESULTS: Among serum results from calves from 50 herds available for analysis, 34 (68%) herds were classified as likely non-infected (0-3 seropositive among 15 calves) and 16 (32%) as likely infected (5-15 seropositive among 15 calves). Receiver-operator characteristic (ROC) analysis identified an optimal cut-off for BTM of 80%INH associated with 81% sensitivity and 91% specificity for likely herd infection. The prevalence of BVDV antibodies in cows within herds and %INH for BVDV in bulk milk were positively correlated (p<0.01). The association between bulk milk %INH and the prevalence of BVDV antibodies in calves was stronger than the same association in cows. Based on the threshold of 80%INH, the 95% confidence interval (CI) for prevalence of likely infection in the 724 herds in the Waikato, Bay of Plenty and Northland regions of New Zealand was 12-17%. Vaccination against BVDV was not significantly associated with the likely infection status of the herd based on prevalence of BVDV antibodies among calves. CONCLUSION: An ELISA test result for BVDV antibodies in BTM >/=80%INH can be used as a threshold to indicate the presence of likely infection with BVDV in dairy herds in New Zealand, with 81% sensitivity and 91% specificity.  相似文献
