BACKGROUND: The brown planthopper, Nilaparvata lugens (Stål), is a serious pest that causes enormous losses to the rice crop in Asia. The genetic basis of imidacloprid resistance was investigated in N. lugens. RESULTS: The resistant strain, selected for imidacloprid resistance from a field population of N. lugens collected from Nanjing, Jiangsu Province, China, showed a 964‐fold resistance compared with the laboratory strain. Progenies of reciprocal crosses (F1 and F1′) showed similar dose–mortality responses (LC50) to imidacloprid, and also exhibited a similar degree of dominance (D), 0.58 for F1 and 0.63 for F1′. Chi‐square analyses of self‐bred and backcross progenies (F2, F2′ and BC respectively) rejected the hypothesis for a single gene control of the resistance. The estimated realized heritability (h2) of imidacloprid resistance was 0.1141 in the resistant strain of N. lugens. CONCLUSION: The results showed that imidacloprid resistance in N. lugens was autosomal and was expressed as an incompletely dominant trait, probably controlled by multiple genes. Susceptibilities of UK and mainland European samples of Trialeurodes vaporariorum (Westwood) to the neonicotinoid insecticide imidacloprid were investigated over a 7 year period. All 24 strains collected between 1997 and 2003 showed similar baseline levels of susceptibility to that of a known susceptible laboratory strain when exposed to a diagnostic concentration (128 mg L(-1)) of formulated imidacloprid. Two samples collected during 2004, one from the UK and one from The Netherlands, demonstrated reduced susceptibility at this concentration. Using dose-response assays, the presence of resistant individuals was disclosed in both these strains; some individuals were unaffected at doses high enough to induce phytotoxic effects. This report represents the first confirmed cases of neonicotinoid resistance inducing control failures in T. vaporariorum, and highlights a need for careful vigilance to sustain the effectiveness of imidacloprid and related neonicotinoid insecticides.  相似文献

2006-2009年,用稻茎浸渍法连续监测了广西南宁市、广东阳江市、湖南东安县、福建福清市、江西上高县、湖北孝感市、浙江金华市、江苏通州市和安徽和县共9个地区褐飞虱种群对吡虫啉、噻嗪酮和氟虫腈的抗性变化。结果表明:褐飞虱种群对吡虫啉的抗性仍处于高水平至极高水平抗性阶段(105.5~459.7倍),但2009年监测到东安、孝感、上高种群对吡虫啉的抗性已有下降趋势;褐飞虱种群对氟虫腈的抗性有增长趋势,2006到2009年褐飞虱种群对氟虫腈由敏感至低水平抗性(<6.9倍)发展到了中水平至高水平抗性(13.5~43.3倍);由于2005年吡虫啉在高抗地区的禁用,褐飞虱种群对噻嗪酮的抗性上升速度加快,2009年已处于低水平至中水平抗性阶段(7.0~14.4倍)。这表明在吡虫啉、氟虫腈被禁用后,大面积单一使用噻嗪酮进行防治,褐飞虱对噻嗪酮的抗性有可能加速发展。  相似文献

BACKGROUND: Imidacloprid has been a major neonicotinoid insecticide for controlling Aphis gossypii (Glover) (Homoptera: Aphididae) and other piercing–sucking pests. However, the resistance to imidacloprid has been recorded in many target insects. At the same time, cross‐resistance of imidacloprid and other insecticides, especially neonicotinoid insecticides, has been detected. RESULTS: Results showed that the level of cross‐resistance was different between imidacloprid and tested neonicotinoid insecticides (no cross‐resistance: dinotefuran, thiamethoxam and clothianidin; a 3.68–5.79‐fold cross‐resistance: acetamiprid, nitenpyram and thiacloprid). In the study of sublethal effects, imidacloprid at LC20 doses could suppress weight gain and honeydew excretion, but showed no significant effects on longevity and fecundity of the imidacloprid‐resistant cotton aphid, A. gossypii. However, other neonicotinoid insecticides showed significant adverse effects on biological characteristics (body weight, honeydew excretion, longevity and fecundity) in the order of dinotefuran > thiamethoxam and clothianidin > nitenpyram > thiacloprid and acetamiprid. CONCLUSION: The results indicated that dinotefuran is the most effective insecticide for use against imidacloprid‐resistant A. gossypii. To avoid further resistance development, the use of nitenpyram, acetamiprid and thiacloprid should be avoided on imidacloprid‐resistant populations of A. gossypii. BACKGROUND: In 2003 the development of insecticide resistance against neonicotinoids in the brown planthopper (BPH), Nilaparvata lugens (Stål) (Homoptera: Delphacidae), was first observed in Thailand and has since been found in other Asian countries such as Vietnam, China and Japan. However, the LD50 values of BPH and the whitebacked planthopper (WBPH), Sogatella furcifera (Horváth), against both neonicotinoid and phenylpyrazole insecticides have been poorly reported in many Asian countries. RESULTS: The topical LD50 values for imidacloprid in the BPH populations collected from East Asia (Japan, China, Taiwan) and Vietnam in 2006 were 4.3–24.2 µg g?1 and were significantly higher than those collected from the Philippines (0.18–0.35 µg g?1). The BPH populations indicated a positive cross‐resistance between imidacloprid and thiamethoxam. Almost all the WBPH populations from Japan, Taiwan, China, Vietnam and the Philippines had extremely large LD50 values (19.7–239 µg g?1 or more) for fipronil, except for several populations from the Philippines and China. CONCLUSION: Species‐specific changes in insecticide susceptibility were found in Asian rice planthoppers (i.e. BPH for imidacloprid and WBPH for fipronil). Insecticide resistance in BPH against imidacloprid occurred in East Asia and Indochina, but not in the Philippines. Effective control of the brown planthopper, Nilaparvata lugens Stål, across rice-growing regions of Asia has been seriously compromised over the last 2 years by the appearance of widespread resistance to the neonicotinoid insecticide, imidacloprid. Sequence analysis of the ligand-binding domain of the nicotinic acetylcholine receptor α1 subunit from two field-collected resistant strains (CHN-2 and IND-11) did not reveal the Y151S point mutation previously implicated in conferring target-site resistance in this species. This result was supported by ligand-binding studies with [3H]-imidacloprid that showed no significant change in insecticide binding to isolated membranes from susceptible and resistant strains. In contrast, there was an approximate 5-fold increase in the mixed function oxidase activity for the two resistant strains suggesting that imidacloprid metabolism by increased cytochrome P450 monooxygenase activity is the major mechanism of resistance in these strains.  相似文献

Nine different strains of the two-spotted spider miteTetranychus urticae Koch (Acari: Tetranychidae) were collected on cotton from Adana, Antalya, Izmir, Manisa and Urfa in Turkey. Their responses to bifenthrin were investigated using conventional bioassay and biochemical assays. LC50 and LC90 values of bifenthrin were determined for all strains by using a residual bioassay with a petri dish-spray tower. Resistance ratios were determined by comparing the samples with a standard susceptible strain, GSS. The resistance ratios of the strains ranged from <1 to 669-fold (at LC50). Of the investigated field strains, only three (two from Adana and one from Urfa) were resistant to bifenthrin. There was a correlation between esterase enzyme activity and bifenthrin resistance according to polyacrylamide gel electrophoresis and microtiter plate assays in the three resistant strains. http://www.phytoparasitica.org posting May 17, 2005.  相似文献

BACKGROUND: Imidacloprid is the primary insecticide for controlling the tobacco‐adapted form of the green peach aphid (TGPA), Myzus persicae (Sulzer), a major pest of tobacco worldwide. This study used leaf‐dip bioassays to assess TGPA resistance to imidacloprid in the eastern United States from 2004 through 2007. RESULTS: When combined over the 4 year study, 18, 14 and 3% of the TGPA had imidacloprid resistance ratios (RRs) of 10–20‐fold, 20–30‐fold and 30–90‐fold, respectively, compared with the most susceptible colony tested. This indicates that some colonies have developed moderate levels of resistance to imidacloprid. A colony collected near Clayton, North Carolina, had the highest RR of 91 (LC50 value = 31 mg L?1). This resistance declined for six tests over a 3 year period in the laboratory culture from >130‐fold RR (LC50 = 48 mg L?1) to 40‐fold RR (LC50 = 15 mg L?1). Over the same period, the most susceptible colony and a standard colony not exposed to imidacloprid for over 7 years had consistently low LC50 values. CONCLUSION: Moderate levels of resistance to imidacloprid are noticed among TGPA colonies from the eastern United States. The variation in resistance indicates that the factors responsible are present in the populations at low frequencies and are just not enough to cause field failures yet. 采用稻苗浸渍法测定了长、短翅型褐飞虱对烯啶虫胺、环氧虫啶、呋虫胺、噻虫嗪、噻虫胺、吡虫啉、毒死蜱、敌敌畏、噻嗪酮、异丙威、吡蚜酮和醚菊酯的敏感性,并对其体内解毒酶活力进行了比较分析。结果表明:长翅型与短翅型褐飞虱若虫对新烟碱类杀虫剂呋虫胺、噻虫嗪、噻虫胺和吡虫啉的敏感性存在显著差异,长翅型比短翅型更敏感;相反,对于有机磷类杀虫剂毒死蜱,短翅型褐飞虱则更敏感;2种生物型对烯啶虫胺、环氧虫啶、敌敌畏、噻嗪酮、异丙威、吡蚜酮和醚菊酯的敏感性无显著差异。解毒酶相对比活力测定结果表明,长翅型褐飞虱若虫酯酶比活力显著高于短翅型,细胞色素P450单加氧酶比活力显著低于短翅型,而谷胱甘肽S-转移酶比活力无显著性差异。本研究结果可为褐飞虱的有效防控提供科学参考。  相似文献

采用玻璃管药膜法,建立了以吡虫啉、啶虫脒、噻虫嗪、噻虫胺、烯啶虫胺5种新烟碱类杀虫剂及氟啶虫胺腈对桃蚜室内敏感品系(SN)的LC90值作为区分剂量,测定桃蚜对6种杀虫剂敏感性变化的方法,并与浸叶法测得的抗性水平进行了相关性分析,验证了利用区分剂量快速测定桃蚜田间种群对6种杀虫剂抗性水平的准确性。结果表明:6种杀虫剂对桃蚜室内敏感品系的LC90值分别为150.01、1 170.81、54.19、951.34、245.98及133.60 ng/cm2。在此区分剂量下,河南省驻马店地区桃蚜种群(ZM)的死亡率在82%~96%之间;河北省玉田地区甘蓝桃蚜种群(GL)的死亡率在35%~82%之间,桃树桃蚜种群(TS)死亡率在3%~30%之间。分析表明,在选定的区分剂量下,桃蚜田间种群的死亡率与其对杀虫剂的抗性水平呈负相关,相关系数在0.818 8~0.999 9之间。同时,通过相关性方程计算得到的江苏省南京地区桃蚜种群(NJ)对6种杀虫剂的理论抗性水平与实际检测所得抗性水平结果接近。因此,以玻璃管药膜法确定的吡虫啉、啶虫脒、噻虫嗪、噻虫胺、烯啶虫胺及氟啶虫胺腈对桃蚜室内敏感品系的LC90值作为区分剂量,通过测定桃蚜田间种群的死亡率,可以快速表征田间种群对6种杀虫剂的敏感性变化,从而对其抗性水平进行初步评估。  相似文献

BACKGROUND: Development of pyriproxyfen and neonicotinoid resistance in the B-biotype whitefly and recent introduction of the Q biotype have the potential to threaten current whitefly management programs in Arizona. The possibility of integrating the novel anthranilic diamides chlorantraniliprole and cyantraniliprole into the current program to tackle these threats largely depends on whether these compounds have cross-resistance with pyriproxyfen and neonicotinoids in whiteflies. To address this question, the authors bioassayed a susceptible B-biotype strain, a pyriproxyfen-resistant B-biotype strain, four multiply resistant Q-biotype strains and 16 B-biotype field populations from Arizona with a systemic uptake bioassay developed in the present study. RESULTS: The magnitude of variations in LC50 and LC99 among the B-biotype populations or the Q-biotype strains was less than fivefold and tenfold, respectively, for both chlorantraniliprole and cyantraniliprole. The Q-biotype strains were relatively more tolerant than the B-biotype populations. No correlations were observed between the LC50 (or LC99) values of the two diamides against the B- and Q-biotype populations tested and their survival rates at a discriminating dose of pyriproxyfen or imidacloprid. CONCLUSION: These results indicate the absence of cross-resistance between the two anthranilic diamides and the currently used neonicotinoids and pyriproxyfen. Future variation in susceptibility of field populations to chlorantraniliprole and cyantraniliprole could be documented according to the baseline susceptibility range of the populations tested in this study. BACKGROUND: Biotype B of the sweetpotato whitefly, Bemisia tabaci (Genn.), is a worldwide pest that has developed resistance to many insecticides, including the neonicotinoid class. Florida field populations were monitored for susceptibility to the neonicotinoids imidacloprid and thiamethoxam using a cut leaf petiole bioassay method. RESULTS: Average RR50 values for imidacloprid increased from 3.7 in 2000 to 12.0 in 2003; decreased to 5.0 and 2.5 in 2004 and 2005, respectively; and then increased to 26.3 and 23.9 in 2006 and 2007, respectively. Populations with RR50 values of about 50 to 60 during generation one reverted to RR50 values of ?4 in six generations, when reared without further exposure to imidacloprid. Average RR50 values for thiamethoxam increased from 2.0 in 2003 to 24.7 in 2006 and decreased to 10.4 in 2007. Populations with RR50 values of about 22, 32 and 53 during generation one declined to 8, 5 and 6, respectively, after being reared for five generations without exposure to thiamethoxam. The correlation coefficient from the 26 populations that were bioassayed both with imidacloprid and thiamethoxam showed a significant positive correlation (R2 = 0.58) between these populations. CONCLUSION: The high level of RR50 values to imidacloprid and thiamethoxam suggest an unstable decline in the susceptibility of B. tabaci to imidacloprid and thiamethoxam, with possible cross‐resistance or predisposition for dual resistance selection. BACKGROUND: Neonicotinoid action as well as resistance involves interaction with nicotinic acetylcholine receptors (nAChRs). In the housefly, neonicotinoid resistance also involves cytochrome P450, as indicated by bioassay with synergist as well as altered expression. In bioassay, synergism was only partial and indicated possible target‐site resistance. The nAChR α2 subunit is important in neonicotinoid toxicity to insects, and gene expression of the Mdα2 subunit was investigated in field populations and laboratory strains of neonicotinoid‐resistant and insecticide‐susceptible houseflies, Musca domestica L. The genomic sequence covering exon III–VII of Mdα2 was analysed for mutations. RESULTS: Gene expression profiling of Mdα2 revealed notable differences between neonicotinoid‐resistant and insecticide‐susceptible houseflies. On average, the neonicotinoid‐resistant field population 766b and the imidacloprid selected strain 791imi had 60% lower copy numbers of Mdα2 compared with the susceptible reference strain. Sequencing of exon III–VII of the Mdα2, encoding acetylcholine binding‐site regions and three out of four transmembrane domains, did not reveal any mutations explaining the increased neonicotinoid tolerance in the strains examined. CONCLUSION: Previous discoveries and the results of this study suggest that the neonicotinoid resistance mechanism in Danish houseflies involves both cytochrome P450 monooxygenase‐mediated detoxification and reduced expression of the nAChR subunit α2. It is known from laboratory studies that tobacco-associated forms of Myzus persicae (Sulzer) and the closely related tobacco aphid Myzus nicotianae (Blackman) are often somewhat less susceptible to imidacloprid than non-tobacco strains of M. persicae. Choice tests (floating leaf technique) showed that tobacco aphids were also less susceptible to the antifeedant potential of imidacloprid in contact bioassays. Synergists like piperonyl butoxide or DEF did not enhance the susceptibility of tobacco-associated morphs of Myzus ssp. to imidacloprid, thus providing evidence that neither oxidative detoxication nor hydrolytic metabolization took place. However, in an attempt to study the influence of endosymbiotic bacteria on the efficacy of imidacloprid, we allowed small populations of tobacco aphids to feed on diets containing the antibiotic chlortetracycline prior to imidacloprid treatment. While the effectiveness of imidacloprid, i.e. lower LC50 values, could be improved in all strains, including the susceptible reference strain, there was no change in overall tolerance factors. In order to investigate any possible alteration of the target site, the affinity of imidacloprid and nicotine to nicotinic acetylcholine receptors in whole-aphid homogenates was measured. All strains (and clones) showed the same high-affinity binding sites and no detectable difference. Studies using the FAO dip method revealed that the lower susceptibility of M. nicotianae is not restricted to chloronicotinyls like imidacloprid or acetamiprid, because other insecticides with different modes of action such as pymetrozine and fipronil were also affected in laboratory studies. It is considered that the observed tolerance to chloronicotinyls in certain strains of Myzus ssp. is a natural variation in response, probably not coupled with any known mechanism of resistance in this species complex. © 1998 SCI  相似文献

BACKGROUND: The housefly, Musca domestica L., continues to be a major pest of confined livestock operations. Houseflies have developed resistance to most chemical classes, and new chemistries for use in animal agriculture are increasingly slow to emerge. Five adult housefly strains from four Florida dairy farms were evaluated for resistance to four insecticides (beta‐cyfluthrin, permethrin, imidacloprid and nithiazine). RESULTS: Significant levels of tolerance were found in most field strains to all insecticides, and in some cases substantial resistance was apparent (as deduced from comparison with prior published results). At the LC90 level, greater than 20‐fold resistance was found in two of the fly strains for permethrin and one fly strain for imidacloprid. Beta‐cyfluthrin LC90 resistance ratios exceeded tenfold resistance in three fly strains. The relatively underutilized insecticide nithiazine had the lowest resistance ratios; however, fourfold LC90 resistance was observed in one southern Florida fly strain. Farm insecticide use and its impact on resistance selection in Florida housefly populations are discussed. CONCLUSION: Housefly resistance to pyrethroids is widespread in Florida. Imidacloprid resistance is emerging, and tolerance was observed to both imidacloprid and nithiazine. If these insecticides are to retain efficacy, producer use must be restrained. Imidacloprid has been used as a key insecticide to control the brown planthopper, Nilaparvata lugens Stål, for several years, but no obvious resistance has been identified in field populations as yet. To evaluate the risk, a field population was collected and selected with imidacloprid in the laboratory. After 37‐generation selection a strain with 250‐fold resistance had been successfully achieved. Fitness analysis by constructing life tables demonstrated that resistant hoppers had obvious disadvantages in their reproduction. The fitness of highly resistant hoppers had decreased dramatically (0.169 and 0.104) to only one‐fifth to one‐tenth of that of the susceptible strain. Hence it was concluded that the brown planthopper had the potential to develop high resistance to imidacloprid but that the lower fitness of resistant hoppers could result in a quick recovery of sensitivity when the population did not come into contact with imidacloprid. This means that a reasonable resistance management programme with less imidacloprid use may efficiently delay or even stop resistance development. BACKGROUND: The cabbage whitefly, Aleyrodes proletella L., is emerging as a significant pest of field brassica crops in certain regions of the United Kingdom. In order to investigate the contribution of pesticide resistance to this phenomenon, A. proletella populations were sampled from five different areas in England in 2008 and 2009. Adult residual leaf‐dip bioassays were carried out using pyrethroid and neonicotinoid insecticides. RESULTS: Significant resistance to pyrethroids was found in multiple samples collected from two areas. No evidence of cross‐resistance to neonicotinoids was found in a subset of the pyrethroid‐resistant populations. While the patterns of resistance to different pyrethroids were broadly correlated, the magnitude of resistance factors differed substantially. Survival of strains at a putative diagnostic concentration of lambda‐cyhalothrin was found to provide a guide to their LC50. Significant differences in LC50 were found when different brassica crops were used in the bioassay, although the resistance patterns between strains were maintained. CONCLUSION: Reduced susceptibility to multiple pyrethroid insecticides exists in populations of A. proletella in the United Kingdom, corresponding to recent major outbreaks. The mechanism(s) of resistance are yet to be determined, but molecular structural differences in pyrethroids probably influence the magnitude of cross‐resistance within this group of insecticides. BACKGROUND: Citrus red mite, Panonychus citri (McGregor), is one of the most important pesticide‐resistant pests in China. In order better to understand its resistance status, six populations of the mite were collected from Chinese citrus orchards for monitoring of resistance to spirodiclofen and another five acaricides. RESULTS: All the samples collected in the field in 2006 were susceptible to spirodiclofen. However, the LC50 values in populations sampled in 2009 ranged from 3.29 to 418.24 mg L?1 spirodiclofen, a 127‐fold difference between the least and most sensitive populations. Compared with a susceptible strain, 50‐fold and 90.8‐fold resistance to spirodiclofen was detected in populations sampled from Pinghe and Fuzhou in 2009, as well as cross‐resistance to spirotetramat. The LC50 values for abamectin, fenpropathrin, hexythiazox and pyridaben in the collected samples ranged from 0.041 to 3.52 mg L?1, from 23.91 to 696.16 mg L?1, from 13.94 to 334.19 mg L?1 and from 48.90 to 609.91 mg L?1 respectively. CONCLUSION: Great variations in resistance to the tested acaricides were observed among the sampled populations. The Pinghe population developed resistance to all the acaricides tested. The Jianning population was susceptible to most acaricides tested, except pyridaben. Resistance management strategies were conducted on the basis of these observations. A field-collected population of the silverleaf whitefly, Bemisia argentifolii, was selected with the nicotinyl compound, imidacloprid, over 32 generations to determine if resistance would develop when maintained under continuous selection pressure in a greenhouse. Resistance was slow to increase at first with low to moderate levels of resistance (RR from 6- to 17-fold) in the first 15 generations of selection. Further selection steadily led to higher levels of resistance, with the greatest resistance ratio at 82-fold, the gradual rise suggesting the involvement of a polygenic system. At the end of the selection, slopes of probit regressions were substantially steeper than earlier, indicating increased homogeneity of imidacloprid resistance in this strain. A hydroponic bioassay featuring systemic uptake of imidacloprid through roots was developed to monitor the changes in resistance to imidacloprid in the selected whitefly strain and in seven field-collected strains from Imperial Valley, California. Six out of seven field-collected strains exhibited low LC50 values (0·002 to 0·512 mg ml-1) compared to the selected resistant strain, with one exception where the LC50 was 0·926 mg ml-1 (RR=15·0). Variation in responses to imidacloprid in the field strains suggest that this technique is sufficiently sensitive to detect differences in susceptibilities of whitefly populations. The imidacloprid-resistant strain showed no cross-resistance to endosulfan, chlorpyrifos or methomyl (RR ranging from 0·4- to 1·5-fold). A low level of cross-resistance was observed to bifenthrin in the IM-R strain at 7-fold. The success of selection for resistance to imidacloprid has serious implications for whitefly control programs that rely heavily on imidacloprid. ©1997 SCI  相似文献

BACKGROUND: In recent years, outbreaks of the brown planthopper, Nilaparvata lugens (Stål), have occurred more frequently in China. The objective of this study was to determine the susceptibility of N. lugens to neonicotinoids and other insecticides in major rice production areas in China. RESULTS: Results indicated that substantial variations in the susceptibility to different insecticides existed in N. lugens. Field populations had developed variable resistance levels to neonicotinoids, with a high resistance level to imidacloprid (RR: 135.3–301.3‐fold), a medium resistance level to imidaclothiz (RR: 35–41.2‐fold), a low resistance level to thiamethoxam (up to 9.9‐fold) and no resistance to dinotefuran, nitenpyram and thiacloprid (RR < 3‐fold). Further examinations indicated that a field population had developed medium resistance level to fipronil (up to 10.5‐fold), and some field populations had evolved a low resistance level to buprofezin. In addition, N. lugens had been able to develop 1424‐fold resistance to imidacloprid in the laboratory after the insect was selected with imidacloprid for 26 generations. CONCLUSION: Long‐term use of imidacloprid in a wide range of rice‐growing areas might be associated with high levels of resistance in N. lugens.
