Improvement of pulmonary arterial lesions after heartworm removal using flexible alligator forceps was investigated by measuring pulmonary arterial pressure (PAP), and by angiographic and histopathological examinations in 11 dogs. PAP obtained immediately after worm removal corresponded well with angiographic abnormalities. In 2 dogs, high PAP immediately after worm removal fell gradually by 12 weeks, and obstructions on angiogram were resolved at 4 to 12 weeks. In 4 dogs, slightly high PAP fell to the normal range at 4 weeks, and angiographic abnormalities were considerably reduced at 4 to 12 weeks. In 5 dogs, PAP returned to normal range immediately after worm removal, and angiographic changes almost disappeared at 4 to 8 weeks. On biopsy immediately after worm removal, samples of the main pulmonary arteries showed severe intimal proliferations with villous or papillary protrusion into the lumen. Autopsy at 12 to 20 weeks indicated that the intimal protrusions were remarkably reduced as compared with the biopsy samples in all cases. However, villous intimal protrusions were seen in the caudal lobar arteries in cases with remaining alive worms. New vessels seemed to develop into thromboemboli with time. From these findings, it is clear that the pulmonary arterial lesions improved after heartworm removal, and the clinical signs disappeared following the improvement in hemodynamics. Aspirin therapy (5 mg/kg/day) for 4 weeks after worm removal in 5 dogs did not improve the intimal lesions as compared to 3 control dogs.  相似文献

Carval syndrome is a severe heartworm infection where the worms have migrated to the right atrium and vena cava; this condition is associated with a myriad of clinical signs. Several non-surgical and interventional methods are currently used for mechanical worm removal. However, the success rate and complications related to these methods are heavily dependent on methodology and retrieval devices used. In this study, we developed a catheter-guided heartworm removal method using a retrieval basket that can easily access pulmonary arteries and increase the number of worms removed per procedure. With this technique, we successfully treated four dogs with caval syndrome.  相似文献

The performance of a method of estimating the worm burdens in young sheep flocks, by applying egg count data to a simple "formula" was evaluated on four farms in the Manawatu. On all farms there was a good fit between the estimated flock worm burden using this method and those obtained from worm counts. It is suggested that this procedure is likely to be successful in providing a reasonable approximation of the flock worm profile when 10 to 15 egg counts on randomly selected members are performed.  相似文献

The influence that decreased functional hepatic mass had on blood arsenic concentrations in dogs after they were treated with thiacetarsamide, on the clearance of indocyanine green (ICG), on arsenic concentrations in the heartworm (Dirofilaria immitis), and on drug efficacy was studied. Dogs which were partially hepatectomized and treated with thiacetarsamide (1.76 mg/kg, 2 times a day for 2 days) had a significantly (P less than 0.01) reduced ICG clearance, significantly (P less than 0.01) higher arsenic levels in heartworms, and a significantly (P less than 0.01) higher proportion of heartworms killed than did dogs that were sham operated and treated with thiacetarsamide or sham operated and untreated. There were no significant differences in blood arsenic (thiacetarsamide) concentrations 2 minutes after injection between hepatectomized and nonhepatectomized groups. More male heartworms were killed than were female worms in the thiacetarsamide-treated groups. Indocyanine green half-life was longer (12.43 minutes) in the hepatectomized group than it was in the nonhepatectomized sham-operated groups (5.09 and 4.94 minutes). Indocyanine green clearance rate was lower in the hepatectomized group (0.54 ml/min/kg) than that in the nonhepatectomized groups (1.36 and 1.56 ml/min/kg). A parallel seemed to exist between ICG and thiacetarsamide removal from the blood by the liver. This parallel also was suggested in the higher worm arsenic (thiacetarsamide) concentrations for the hepatectomized group vs that for nonhepatectomized groups. Apparently, the slower the removal of thiacetarsamide from the blood by the liver, the higher the worm arsenic level and, consequently, the higher the worm kill.  相似文献

Canine heart worm disease is often life-threatening due to its various complications, including right side heart failure, caval syndrome and pulmonary eosinophilic granulomatosis. Several preventive medications and melarsomine have been developed and they are very effective to control heartworm infestation. However, in a case of severe infestation, melarsomine therapy often results in an unfavorable outcome because of the severe immune reaction caused by rapid killing of the adult worm. Surgical removal and an interventional method using flexible alligator forceps have been well described in the literature. Despite the usefulness of mechanical removal using flexible alligator forceps, the methodology still needs to be upgraded for increasing the applicability for treating dogs with severe infestation. We describe herein a newly developed percutaneous removal method for heartworms and this was successfully applied to 4 dogs with severe heartworm infestation. The follow-up studies also showed favorable outcomes with no complications.  相似文献

A new approach to the interpretation of faecal egg counts using a simple "formula", is described. It is suggested that adoption of this procedure may enable the likely level and frequency distribution of strongyle worm burdens (Nematodirus excluded) in young sheep flocks to be estimated. The potential relevance of this information to the diagnosis of gastrointestinal parasitism is discussed.  相似文献

An excretory-secretory (ES) antigen was used in a serodiagnostic enzyme-linke immunosorbent assay (ELISA) for swine trichinosis. ELISA procedures included a double- antibody test, using either an anti-swine IgG or a protein A enzyme conjugate, and a triple-antibody test using a pig IgG heavy-chain specific second antibody with a conjugated third antibody. The ES antigen was effective in eliminating all false-positive reactivity in sera from farm-raised hogs. The triple-antibody procedure was more sensitive and demonstrated a greater efficiency in detecting positive animals and early seroconversions. Naturally-infected pigs with worm burdens as low as 0.01 larvae per gram (LPG) of diaphragm were seropositive using these procedures. Seroconversion in experimentally-infected animals receiving low doses of muscle larvae (500) occurred considerably later than in animals receiving high doses (10000). This might account for false-negative reactions in naturally-infected animals with very low (less than 0.1 LPG) worm burdens.  相似文献

Four groups of nine Saanen goat does with a naturally acquired mixed trichostrongylid infection were grazed on four paddocks. Two groups received a daily dose of Duddingtonia flagrans at the rate of 5 x 10(7) chlamydospores per animal per day for the 26-day grazing period. After a 19-day pasture resting period, 20 worm free 12-week-old tracer kids were introduced to the paddocks for 14 days prior to removal for worm burden analysis. Four groups of five does and four kids were drenched then turned out onto the paddocks and faecal egg count (FEC) monitored. The FEC between groups was comparable throughout the initial grazing period. There were significant reductions in number of Teladorsagia circumcincta (54.8%, P=0.004) and Haemonchus contortus (85.0%, P=0.02) worms recovered from tracer animals. FEC of animals subsequently grazing pasture were significantly reduced (P=0.036) with reductions of 44% observed 4 weeks post-turnout. No significant difference was observed after 6 weeks grazing. This trial has demonstrated the potential of D. flagrans to reduce larval numbers on pasture grazed by goats under New Zealand conditions.  相似文献

Several necropsy techniques are available for estimating the abundance of gastro-intestinal nematodes in abomasum of ruminants. Standardization of techniques is needed to allow accurate comparisons between laboratories. Here we propose a standardized technique for estimating the abundance of worms. We intend to compare the worms' number estimations in lambs and ewes based on contents and washings, to determine the uniformity of worm counts in aliquots, and to estimate the total worm number from washings. The digesta (or "contents") and the washings of the abomasum are treated separately. The worms of each subsample are diluted with water and the total number of worms is estimated on a small volume (aliquots) of these subsamples. The use of aliquots assumes that the worms are uniformly distributed in the whole volume of each subsample. We first confirmed that the use of aliquots is appropriate in most cases. We then show that the use of the washings alone allows a faster and a suitable estimation of the total worm burden for all strongyle species of the abomasum in both ewes and lambs. The evaluation of our necropsy procedure is a first step to a standardized technique which should be improved by validation in other laboratories.  相似文献

SUMMARY: Resistance to a range of benzimidazole anthelmintics was investigated in 2 strains of Ostertagia spp. One strain (SRBO) had been exposed to fenbendazole, oxfendazole and thiabendazole, the other strain (KR79) only to thiabendazole. Both strains showed a high degree of resistance to albendazole, fenbendazole, oxfendazole and thiabendazole, which had efficiencies of 11–38% and 0–60% against all developmental stages of SR80 and KR79, respectively. There was no significant reduction in the KR79 worm count by thiabendazole at 132 mg kg-1 given either as a single dose or divided into 12 equal portions, one administered every 6h. Nematodirus spp were also found to be resistant to all benzimidazoles tested. Levamisole at 7 mg kg-1 and naphthalophos at 30 mg kg-1 had efficiences of 89 and 66%, respectively, against SR80 Ostertagia and 99 and 19% against Nematodirus, all but the last of these being significant reductions in worm burdens. The resistant SR80 Ostertagia occurred on a research station, but possibly originated from a property which 3 years earlier had supplied ewes to the station. The use of benzimidazole anthelmintics and subsequent grazing on worm-free pasture may have enhanced the level of resistance. A relationship was established between egg counts and adult worm counts 10 days post-treatment, which suggested that for Ostertagia the worm count could be predicted from the geometric mean egg count from about 10 animals. Thus, where an Ostertagia population is suspected of being resistant, an anthelmintic efficiency assay using pre-and post-treatment faecal egg counts should provide a satisfactory diagnostic procedure.  相似文献

Two of the most important questions for immunologists studying nematode infections are what effect does the host response have on the parasite and which components of the host response are responsible for these effects. The number of nematodes and the mean length of adult female Ostertagia circumcincta was measured in over 500 6-7 month old lambs of the Scottish Blackface breed. Quantitative genetic analyses indicated that there was substantial genetic variation among lambs in faecal egg counts and in worm length but the analyses provided no evidence for genetic variation in worm numbers. Separate analyses have shown a strong relationship between decreased worm length and decreased worm fecundity. Therefore, genetic resistance in lambs appears to be due to control of worm growth and not to control of worm numbers. The only immune response that is consistently associated with reduced worm length is the IgA response to fourth-stage larvae. The association is remarkably strong. After allowing for the influence of worm number on worm length (density-dependence) essentially all of the variation among deliberately infected lambs in worm length can be accounted for, in a statistical sense, by variation in the strength and specificity of the local IgA response. Therefore, the IgA mediated suppression of worm growth and fecundity appears to be the major mechanism of resistance to O. circumcincta in lambs.  相似文献

The efficacy of fenbendazole (Panacur, Hoechst-Roussel) against inhibited early fourth-stage larvae of Ostertagia ostertagi and other nematodes of the abomasum and intestinal tract was investigated in naturally infected, yearling cattle in April 1978. The time when peak levels of inhibited larvae occurred was determined by epizootiologic study which began in November 1977. All animals were removed from pasture and maintained free from further helminth infection until slaughter (19 to 21 days). The fenbendazole liquid suspension was administered as an oral drench at dose level of 10 mg/kg to 10 animals and then at dose level of 15 mg/kg to an additional 10 animals at 10 days after removal from pasture. Eleven animals were maintained as untreated controls. In cattle given the dose of 10 mg/kg, the following reductions were observed: O ostertagi adults--100%, developing stages--80%, and inhibited larvae--97%; other worm genera in the abomasum and nematodes of the intestinal tract--100%. In the cattle given the larger dose, the following reductions were observed: O ostertagi adults--100%, developing stages--98%, and inhibited larvae--99%; other worm genera in the abomasum and nematodes of the intestinal tract--100%.  相似文献

A previously developed multiple regression algorithm was used as the basis of a stochastic model to simulate worm burdens in sheep naturally infected with Haemonchus contortus over five consecutive Haemonchus seasons (November to January/February) on a farm in the summer rainfall region in South Africa, although only one season is discussed. The algorithm associates haemoglobin levels with worm counts in individual animals. Variables were represented by distributions based on FAMACHA(?) scores and body weights of sheep, and Monte Carlo sampling was used to simulate worm burdens. Under conditions of high disease risk, defined as the sampling event during the worm season with the lowest relative mean haemoglobin level for a class of sheep, the model provided a distribution function for mean class H. contortus burdens and the probability of these occurring. A mean H. contortus burden for ewes (n=130 per sample) of approximately 1000 (range 51-28,768) and 2933 (range 78-44,175) for rams (n=120 per sample) was predicted under these conditions. At the beginning of the worm season when the risk of disease was lowest (i.e. when both classes had their highest estimated mean haemoglobin levels), a mean worm burden of 525 (range 39-4910) for ewes and 651 (range 37-17,260) for rams was predicted. Model indications were that despite being selectively drenched according to FAMACHA(?) evaluation, 72% of the ewes would maintain their mean worm burden below an arbitrarily selected threshold of 1000 even when risk of disease was at its highest. In contrast, far fewer rams (27%) remained below this threshold, especially towards the end of the worm season. The model was most sensitive to changes in haemoglobin value, and thus by extrapolation, the haematocrit, which is used as the gold standard for validating the FAMACHA(?) system. The mean class haemoglobin level at which there was a 50% probability of worm burdens being ≤ 1000 worms was 7.05 g/dl in ewes and 7.92 g/dl in rams.  相似文献

This study reports on the predictive relationship between serological, immunological and pathological responses following experimental inoculation with incremental doses of Fasciola gigantica in sheep. Fifty, 6-month-old, naive Merino wethers were allocated to one of 5 experimental groups, four of which received 50, 125, 225 and 400 metacercariae, respectively, whilst a 5th group acted as non-inoculated control. Strong individual correlations were observed between liver score, GLDH (glutamate dehydrogenase), GGT (gamma glutamyl transferase), CatL5 (cathepsin L5) antibody titre (IgG1, IgA), eosinophilia, and the total worm count or worm biomass. A combination of multiple indicator traits performed significantly better than any single indicator trait alone. The best predictive index accounted for up to 88% of observed worm burden (Wb) if information on inoculation dose was available. Without knowledge of inoculation dose, such as under field conditions, up to 67% of variation in worm burden could be predicted. In contrast, the best single predictor variable (liver damage score) accounted for up to 50% of worm burden, and in the absence of post-slaughter information, serum levels of anti-cathepsin IgA antibody titres accounted for 35% of predicted variation in worm burden. The utility of a predictive index under both field and experimental inoculation conditions is discussed.  相似文献

Adult feral swine, naturally infected with kidney worms (Stephanurus dentatus) and gastrointestinal nematodes, were divided into two groups of 10 pigs each. One group was treated with fenbendazole (Panacur, Hoechst AG, Frankfurt am. Main) mixed in feed at the rate of 3 mg kg-1 body weight for 3 days. The second group received feed only and was designated as non-treated controls. The animals in both groups were necropsied 3 weeks post-treatment and examined for the presence of live and dead adult kidney worms in the perirenal and ureteral area, ureteral penetration, the presence of kidney worm larvae in the liver, hepatic scars due to kidney worm larval migration, and for liver fibrosis. No live adult kidney worms were found in the perirenal and ureteral areas of treated pigs, and the non-treated pigs harbored an average of 42.8 live worms. No liver kidney worm larvae were found in the livers of treated pigs, and the non-treated pigs averaged 6.7 live larvae. At necropsy, urine samples from 8 of the 10 treated pigs contained no kidney worm eggs, and only 2 eggs were found in samples from each of the remaining 2 pigs in this group. In contrast, urine samples from 8 of the non-treated pigs contained numerous kidney worm eggs. Reductions in ascarid (Ascaris suum) and nodular worm (Oesophagostomum dentatum) egg counts were also observed in treated pigs.  相似文献

Groups of White Leghorn and New Hampshire male and female chicks were reared under helminth-free conditions and divided into two groups at an age of 8 weeks. One group was artificially infected with 100 embryonated Heterakis gallinarum eggs, while the other group was kept as uninfected control. After 8 weeks all animals were harvested and worm burden was established. Maximum of 60 worms per bird randomly selected were measured for length. Body weights were recorded at 8 and 16 weeks of age. Total worm burden was significantly (P<0.0001) higher in infected males than in infected females. Furthermore, White Leghorns showed a significantly (P=0.011) higher number of worms when compared with New Hampshire animals. Average worm lengths were not significantly different between the breeds (P=0.155) and sexes (P=0.062). Correlations between worm burden and worm length was 0.08 (P=0.140). The daily weight gains between 8 and 16 weeks of age of the infected animals were significantly (P=0.038) lower when compared with the control group. There was no significant correlation between daily weight gain and worm burden. The estimated heritabilities for logarithm (ln) worm burden were 0.41 (S.E.+/-0.086) in White Leghorn and 0.31 (S.E.+/-0.126) in New Hampshire, respectively. Based on the estimated heritabilities it is possible to select for H. gallinarum resistance in both genetically distinct chicken breeds. This approach may be of importance for chickens kept in floor husbandry systems.  相似文献

Helminth-free dogs and cats were experimentally infected with protoscoleces of Echinococcus multilocularis and used in controlled trials for efficacy evaluation of the cestodicide epsiprantel. In two separate trials each 4 dogs were treated at day 20 post infection (p.i.) with average oral dosages of 5.1 (4.9-5.3) and 5.4 (5.2-5.8) mg/kg body weight (b.w.) epsiprantel, respectively, and necropsied at day 24 p.i. Among each 4 dogs of the two untreated control groups all animals were infected and had high intestinal worm burdens with averages of 33.575 and 100.725 E. multilocularis specimens per animal (individual worm burdens in group Ib 59,500-149,800, group IIb 20,500-43,200); in the two groups of treated dogs the average worm burdens were reduced by 99.6 and 99.9%. Among 8 treated dogs 4 were helminth-free, the other 4 had residual worm burdens (10-70 in 3 dogs, 1480 in 1 dog). In each 5 cats single oral treatments with average doses of 2.7 (2.7-2.8) and 5.5 (5.5-5.5) mg/kg b.w. epsiprantel were 100% effective against E. multilocularis 20 days p.i. and eliminated the worm burdens from all 10 animals. In the untreated group of 5 cats the average worm burden was 2864 per animal (individual worm burdens 20-6830). Side effects of the drug treatment were not observed. The results of the study show that in single therapeutic dosages recommended by the producer (dogs 5.5 mg, cats 2.75 mg/kg b.w.) epsiprantel eliminates E. multilocularis to over 99% or completely, but residual worm burdens may persist in some animals.  相似文献

Over a period of 1 year, from May 1982 to April 1983, the gastrointestinal tracts of 240 camels were examined for the presence of parasitic helminths. The study quantifies the prevalence of gastrointestinal helminths and the seasonal fluctuations in intestinal worm burdens and faecal worm egg counts. Among the three species of cestodes and eight species of nematodes which were recorded for the first time from Kuwait, Trichostrongylus probolurus (93.8%), T. colubriformis (34.2%) and Stilesia vittata (30.0%) were the most prevalent in the small intestine and Camelostrongylus mentulatus (59.6%) in the abomasum. Estimation of the intestinal worm burdens and faecal worm egg counts showed that Trichostrongylus infections were by far the most predominant. The highest worm and egg counts were recorded in June and August, during the hot dry season. This rise is attributed to infections acquired from February to April, during the cool wet season. Possibly the most effective control can be achieved by a critical treatment at the end of the wet season coinciding with the first rise in nematode population.  相似文献

Two experiments were conducted to elucidate the timing and nature of the sheep immune response to Haemonchus contortus (Barber's pole worm). The first experiment examined the establishment of H. contortus populations and the immune response by comparing a bolus infection of third-stage larvae in na?ve sheep with a group previously primed by a trickle infection. The second experiment used staggered doses of ivermectin-resistant larvae to compare the development of adult worms during different durations of trickle infection with ivermectin-sensitive larvae. Infections successfully generated pathological signs of haemonchosis such as anaemia. Image analysis software was used to measure the area and perimeter of worms collected at post-mortem, and the number of eggs present in individual adult females (fecundity) was significantly correlated with worm size. A significant inverse correlation was found between blood lymphocyte counts and worm fecundity. The absence of correlation between worm fecundity and other leukocyte and erythrocyte counts highlighted the specificity of the lymphocyte response. This is the first report of a link between haematology profiles and worm fecundity in haemonchosis. The correlation observed between adult worm size and egg content leads to the hypothesis that egg production in H. contortus is limited by immune regulation of worm size and presumably growth. Mean worm size and fecundity declined as sheep received more prolonged trickle infections before necropsy, confirming previous reports that immune responses to adult worms are enhanced by ongoing larval challenge. Immunohistochemical results showed trends consistent with a Th2 (humoral) immune response which has been implicated in reducing nematode burdens in several species.  相似文献

The aim of the study was to determine the effect of alternative management interventions on levels of nematodes and the condition of working donkeys in South Africa. Twenty-four adult donkeys (Equus asinus) within an area of 200km radius were randomly allocated to eight paddocks. Two replicates each of three management interventions together with a control group were tested in a 16-month study. The interventions included monthly removal of feces from paddocks where the donkeys grazed, a pre-winter moxidectin treatment, and a combination of a pre-winter moxidectin treatment and monthly fecal removal. The influence of the different interventions on the nematode fecal egg counts, animal live weights, body condition scores and general blood chemistry were compared. In addition, herbage samples were collected from the pastures in each paddock to determine the number of third-stage larvae (L(3)) per kg dry matter. At the end of the study worm recoveries and counts were performed on eight of the animals following euthanasia. The cyathostomes represented the largest portion of the helminth species composition in both the fecal egg counts and larval cultures. Monthly fecal removal alone did not significantly reduce the L(3) on pasture and consideration of more frequent removal is discussed. Pre-winter moxidectin treatment resulted in a 100% reduction in fecal egg counts, an average egg reappearance period of 42-55 days, a reduced average egg count for up to 8 months, and reduced total helminth burdens in all the treated donkeys. It also resulted in improved live weights, hemoglobin concentration, packed cell volumes and to some extent body condition score of the donkeys.  相似文献
