The new taeniacidal antibiotic S15-1 (SQ 21,704) was evaluated against naturally occuring infections of Taenia pisiformis in 53 dogs, Dipylidium caninum in 35 dogs, T taeniaformis in 18 cats, and D caninum in 33 cats. It all instances, the compound was administered in gelatine capsules in a single oral dose. The doses tested were between and 200 mg/kg of body weight in dogs and between 15 and 45 mg/kg in cats. In dogs, doses of 25 mg/kg and greater were 100% effective against T pisiformis, whereas a dose of 50 mg/kg was necessary to clear D caninum. In cats, a single oral dose of 22.5 mg/kg was 100% efficacious against T taeniaeformis, and a single dose of 45 mg/kg (the largest dose tested) clearly seven of eight cats of D caninum. The efficacy was limited to tapeworms only; there was no efficacy against nematodes. The antibiotic was well tolerated by both species with no drug-related vomiting or other side-effects observed.  相似文献

A single dose of a new broad-spectrum anthelmintic, uredofos, was more effective than single doses of either bunamidine hydrochloride (HCI) or niclosamide in removing natural infections of Dipylidium coninum from pound dogs, as determined by necropsy examination for scolices. The efficacy of uredofos was equal to that of bunamidine HCI and exceeded that of niclosamide in removing Taenia spp from his host. All of 12 dogs harboring D caninum and all of 5 dogs harboring Taenia spp were completely cleared of tapeworm infection following treatment with uredofos at 50 mg/kg of body weight. Bunamidine HCl cleared 9 of 11 dogs (82%) of D caninum and 5 of 5 dogs of Taenia spp. Niclosamide had the least anticestodal activity of the 3 drugs; 2 or more scolices of D caninum were found at necropsy in each of 12 dogs treated with this drug. Four of 5 dogs were cleared of Taenia spp following niclosamide therapy. Untreated control dogs did not shed tapeworms of either species during a 3-day period of posttreatment fecal collections, but did have tapeworms at the time of necropsy.  相似文献

The efficacy of bunamidine hydrochloride (given at dose levels of 25 and 50 mg/kg of body wt) against immature and mature Echinococcus granulosus tapeworms was studied in experimentally infected dogs. The compound had average efficacies of 85.9 to 98.8 percent against the immature stages and was completely efficacious (100 percent clearance) against the mature worms. These data indicate that bunamidine HCl at the given doses can be used as a control measure against hydatid disease in the United States.  相似文献

A new broad-spectrum anthelmintic, uredofos, was tested in 146 dogs by single and multiple oral dosing. Single doses of 100 and 50 (but not 25) mg/kg were totally effective in removing Dipylidium caninum and Taenia spp from 46 dogs with infections of tapeworms. Among groups of 15 to 20 dogs, the average percentage efficacies against Toxocara canis for single soese of 100, 50, and 25 mg/kg were 98, 96, and 81%, respectively. The average percentage of efficacies against hookworm (Ancylostoma caninum) were greater than 96% in dogs treated with single doses of 100, 50, or 25 mg/kg and were 100% in the 35 dogs given 2 or 3 treatments (24-hour intervals) at dose levels of either 25 or 50 mg/kg. The whipworm, Trichuris vulpis, was not efficaciously eliminated by single doses of 25, 50, and 100 mg/kg (av percentage of efficacies of 30, 35, and 71%, respectively). Efficacy against T vulpis markedly improved when 2 doses were given at a 24-hour interval (av percentage of efficacies were 89% at dose level of 25 mg/kg and 99% at dose level of 50 mg/kg). At either dose (25 or 50 mg/kg), 3 daily treatments were no more efficacious against whipworms than were 2 doses. There was no evidence of drug toxicosis in any dogs tested. It was concluded that uredofos is highly effective against canine tapeworms, ascarids, and hookworms when given as a single dose of 50 mg/kg and against whipworm when given at dose level of 50 mg/kg/day for 2 days.  相似文献

The anthelmintic potential of epsiprantel, 2-(cyclohexylcarbonyl)-4-oxo-1,2,3,4,6,7,8,12b-octahydropyrazin [2,1-a] [2]benzapine, was revealed using the tapeworms Dipylidium caninum and Taenia taeniaeformis in the cat, and Taenia pisiformis and T. hydatigena in the dog. Subsequent controlled tests in cats demonstrated oral efficacy of 100% against D. caninum with a single dose of 2.5 mg/kg. Although consistently 100% effective against T. taeniaeformis at 5 mg/kg, a single worm was found in one cat treated at 7.5 mg/kg. In experimental infections of Taenia pisiformis in dogs, 100% activity was achieved from a single oral dose of 1 mg/kg. No adverse reaction or drug-associated toxicity were observed at dose levels used.  相似文献

Dogs and cats become infected with tapeworms by ingesting intermediate hosts that contain encysted juvenile tapeworms called larvae. The dog or cat is said to be the definitive host because it shelters the sexually reproductive, egg-producing stage of the tapeworm. The intermediate hosts, which are vertebrates in the case of Taenia and Mesocestoides and insects in the case of Dipylidium and Hymenolepis, become infected by ingesting unhatched but infective tapeworm eggs discharged in the feces of the dog or cat. The relatively less common Diphyllobothrium and Spirometra tapeworms discharge eggs that are undeveloped when passed in the feces and must fall into water to undergo development to the coracidium stage. Diphyllobothrium and Spirometra may have two or three aquatic intermediate hosts in series. The first of these, a copepod, ingests the free-swimming coracidium or ciliated oncosphere that has hatched from the egg. The final intermediate host containing the larva (plerocercoid) infective for the dog or cat is an aquatic vertebrate (fish, frog, water snake). Thus, dogs and cats become infected with tapeworms by eating uncooked meat or fish or by ingesting certain insects. These intermediate hosts are infected with juvenile tapeworms called larvae, which are the infective form for the dog or cat. The intermediate hosts, in turn, become infected by ingesting tapeworm eggs discharged in the feces of the dog or cat or, in the case of Diphyllobothrium and Spirometra, by ingesting coracidia that have subsequently developed in and hatched from such eggs. By far the most common tapeworms of dogs and cats in North America are D. caninum, T. pisiformis, and T. hydatigena. Therefore, the most common sources of tapeworm infection are, respectively, fleas, wild rabbits, and the uncooked meat and offal of ruminants and swine. Whenever a dose of tapeworm remedy is administered or dispensed, the client should be informed of these potential sources of reinfection. There is considerable overlap in the spectra of activity of currently available cestocides. Mebendazole, fenbendazole, niclosamide, bunamidine, and praziquantel are all effective against Taenia spp. Bunamidine is the drug of choice against Spirometra, Diphyllobothrium, and Mesocestoides. Praziquantel is the drug of choice against Echinococcus and Dipylidium.  相似文献

Fifty-five dogs, naturally infected with Taenia sp or Dipylidium caninum or both, were assigned to the following treatment groups for dose titration studies with epsiprantel: nonmedicated control dogs (n = 14), medicated dogs given a dosage of 2.75 mg/kg of body weight (n = 15), medicated dogs given a dosage of 5.5 mg/kg (n = 16), and medicated dogs given a dosage of 8.25 mg/kg (n = 10). Medication was given orally in a tablet formulation. Feces were examined for cestodes passed and the gastrointestinal tract was examined at necropsy for retained cestodes. Efficacy of epsiprantel was 92.9% against Taenia and 44.8% against Dipylidium for a dosage of 2.75 mg/kg, 100% against Taenia and 99.8% against Dipylidium for a dosage of 5.5 mg/kg, and 94.6% against Taenia and 100% against Dipylidium for a dosage of 8.25 mg/kg. For dose confirmation, 36 dogs naturally infected with Taenia sp or D caninum or both were allotted to 2 treatment groups: nonmedicated control dogs (n = 16) and dogs medicated with epsiprantel at a dosage of 5.5 mg/kg (n = 20). Eighteen dogs with naturally acquired helminth infections were used to evaluate the efficacy of nitroscanate against Ancylostoma caninum, Dipylidium caninum, and Trichuris vulpis. Approximately 15 minutes before treatment, the dogs were given 100 to 200 g of canned dog food. Ten dogs were treated with nitroscanate (50 mg/kg of body weight, PO), and 8 dogs were given placebo tablets PO. The dogs were euthanatized and necropsied 10 days after treatment and helminths were recovered from the small intestine and cecum. On the basis of the number of worms recovered from treated dogs vs the number recovered from control dogs, we determined the efficacy of nitroscanate to be 99.6% against A caninum, 99.8% against D caninum, and 0% against T vulpis.  相似文献

Nemadectin, a new broad-spectrum endectocide, was highly efficacious against natural infections of all the major canine gastrointestinal helminths. At single oral dosages of 0.2-0.4 mg kg-1 body weight (BW), a liquid formulation administered in gelatin capsules was 100% effective in eliminating natural infections of Toxocara canis, Toxascaris leonina, Ancylostoma caninum and Uncinaria stenocephala. Tablets (267 mg) containing 4.6% nemadectin given at a rate of 1/3 tablet per 20 kg BW (0.2 mg nemadectin kg-1) were 100% active against T. canis, A. caninum and U. stenocephala. With both formulations, an increase in the dose rate to 0.6-0.8 mg kg-1 BW resulted in 99-100% elimination of Trichuris vulpis infections. No adverse reactions were observed in any of the treated dogs.  相似文献

SUMMARY: The efficiency of the broad spectrum anthelmintic nitroscanate against tapeworm and nematode infections of dogs was tested in artificially or naturally infected dogs. The safety of the compound was also evaluated in acute, subacute and chronic toxicity tests. At the recommended single dose rate of 50 mg/kg given to the dogs with food, nitroscanate was 98% efficient against Taenia hydatigena, T. ovis and T. pisiformis. The drug was highly efficient against Echinococcus granulosus only at the dose rate of 200 mg/kg given twice but total elimination of worms was not achieved. Natural infections of Dipylidium caninum, Ancylostoma spp and Uncinaria stenocephala were totally eliminated from all dogs at the dose rate of 25 mg/kg or higher. Nitroscanate was 97% efficient against adult Toxocara canis at a single dose of 50 mg/kg. When the dose was repeated 24 hours later total elimination of both mature worms and immature worms in puppies aged 2 weeks was achieved. The repeated dose of 100 mg/kg removed 98% of early immature worms from puppies aged 3 days. The drug was 100% efficient against adult Toxascaris leonina at a single dose of 50 mg/kg and 90.5% efficiency was achieved against early 4th stage larvae by two doses of 50 mg/kg. Nitroscanate was not efficient against Trichuris vulpis. The drug was efficient for the removal of Toxocara cati and Ancylostoma tubaeforme from cats. Nitroscanate caused no serious symptoms of toxicity at dose rates up to 10,000 mg/kg in single or repeated doses in young or older adult dogs. The drug was safely given to dogs during pregnancy, to young puppies and to cats. The regular use of nitroscanate as a broad spectrum anthelmintic for the prevention and control of parasitic infections of dogs is discussed.  相似文献

Ninety-four dogs were randomly assigned to 1 of 6 groups (5 treated and 1 untreated control) for evaluation of anthelmintic efficacy. Three anthelmintic formulations were studied: a febantel paste, a praziquantel paste, and a combination paste containing both active ingredients. Treated dogs were dosed once daily for 3 days. The minimal effective dosages in combination were established as 10 mg of febantel and 1 mg of praziquantel/kg of body weight. At those dosages for the combination, 99.5% to 100% of the major nematodes and cestodes identified (ie, Ancylostoma caninum, Trichuris vulpis, Toxocara canis, Dipylidium caninum, and Taenia pisiformis) were eliminated. Febantel paste (10 mg/kg) alone was ineffective against cestodes (14.2% elimination of Dipylidium caninum) and praziquantel paste (1 mg/kg) alone was ineffective (0% to 2.4% elimination) against nematodes.  相似文献

A controlled trial involving 122 dogs experimentally infected with Echinococcus granulosus was carried out to compare the efficiency of arecoline hydrobromide and bunamidine hydrochloride. A single treatment with arecoline hydrobromide at a dose rate of 4 mg/kg or with bunamidine hydrochloride at 50 mg/kg gave a similar degree of efficiency against young worms. Serum level studies in cats and dogs dosed with 30 mg per kg per day of either oral or injectable formulations of a 1:5 trimethoprim/sulphadiazine combination showed good absorption by both routes of administration. Toxicity tests with the products demonstrated a high margin of safety in dogs given this dose level; the margin of safety in cats was lower but still adequate. The high success rate in clinical trials against a wide range of bacterial infections confirmed the predictions from the serum level results that a dose rate of 30 mg per kg per day orally or parentally, would be therapeutically efficacious in both species.  相似文献

Two separate controlled and blinded studies were conducted to confirm the dose and non-interference of spinosad and milbemycin oxime (MO) administered orally in combination or alone to dogs for the treatment and control of experimentally induced flea infestations (Ctenocephalides felis) and adult hookworm infections (Ancylostoma caninum). For each study, dogs were allocated randomly based on pre-treatment adult flea and hookworm egg counts to one of four treatment groups of 10 animals each. In each study, spinosad and MO in combination, using the lower half (30-45 mg/kg spinosad; 0.5-0.75 mg/kg MO) of the US commercial dose band (30-60 mg/kg spinosad; 0.5-1.0mg/kg MO) of each active ingredient, or individually alone using the full dose range, were given orally to dogs on Day 0 using a tablet formulation. A placebo control was treated similarly. In one study, on Days -1, 5, 12, 19, 28 and 35 each dog was infested with approximately 100 unfed adult C. felis obtained from the investigator's established flea colony. All dogs were infested via the same method. Forty-eight hour post-infestation flea comb counts were conducted on Days 1, 7, 14, 21, 30 and 37 and were used to determine the knockdown and residual flea activity. In the second study, on Day -27 each of 48 dogs were experimentally inoculated with 100 third-stage infective larvae of the hookworm, A. caninum. Dogs were treated on Day 0 and necropsied on Day 7 or Day 8. All nematodes in the intestinal tract were collected on Day 7 or Day 8, identified and counted by species and stage. Post-treatment, the geometric mean live flea counts were significantly different (p-value<0.0001) between the spinosad/MO combination and the spinosad only treatment groups as compared to the vehicle control group. The flea counts in the MO only group and the control group were not statistically different. The spinosad and MO combination group and the spinosad only treatment group demonstrated significantly different knockdown (100%) and post-treatment residual flea efficacy at Day 30 was 100% for both groups as compared to the vehicle control. The presence of MO in combination with spinosad did not interfere with the flea efficacy of spinosad as compared to the spinosad only group. MO alone did not demonstrate any flea efficacy. Post-treatment, the geometric mean A. caninum worm counts were significantly different (p-value<0.0001) between the spinosad and MO combination group as compared to the vehicle control group. The worm counts in the MO only group and the combination group were not statistically different. The spinosad and MO combination group (99.8% reduction) and the MO only treatment group (99.5% reduction) both demonstrated significantly different hookworm efficacy as compared to the vehicle control group. The presence of spinosad in combination with MO did not interfere with the hookworm efficacy of MO as compared to the MO only group. Spinosad alone did not demonstrate any hookworm efficacy. In summary, flavored spinosad and MO combination tablets administered orally to dogs at the lower end (30-45 mg/kg spinosad; 0.5-0.75 mg/kg MO) of the US commercial tablet unit dose range (30-60 mg/kg spinosad; 0.5-1.0mg/kg MO) were both safe and highly efficacious delivering 100% knockdown and 30 days of residual adult flea control on experimentally infested dogs as well as >99% adult hookworm efficacy evaluated under laboratory conditions. Interference between either drugs was not demonstrated for both of these dose limiting parasites.  相似文献

Praziquantel is a new type of acylated isoquinoline-pyrazine. A single, low oral or subcutaneous dose of the compound is reliably effective against all tested juvenile and adult cestodes in cats, dogs and sheep. Praziquantel is the first cestodicide which is also effective on bile duct cestodes. In cats and dogs, 5 mg praziquantel per kg is completely effective on all stages of Taenia hydatigena, T pisiformis, T ovis, T taeniaeformis, Dipylidium caninum, Mesocestoides corti, Echinococcus multilocularis and E granulosus. Because of its very wide therapeutic index praziquantel is thus particularly suited for eradication programmes, eg, echinococcosis.  相似文献

A study was initiated to determine the prevalence of tapeworms in horses in Southern Ontario and to investigate the efficacy of pyrantel pamoate, niclosamide and mebendazole. Fecal samples were taken from 580 horses of various breeds, ages and sexes in 24 locations and Anoplocephala perfoliata was found in 13.6%. This was regarded as a minimum, the true rate being probably significantly higher and the reasons for this are discussed. A brief review of the life cycle and effects of tapeworms in horses and a comparison of two flotation techniques for the diagnosis of A. perfoliata eggs in feces is given.Pyrantel pamoate, niclosamide and mebendazole were compared for efficacy in field and critical trials. In field trials, pyrantel base and niclosamide at 6.6 and 50 mg/kg respectively were found to be effective, but in critical trials their efficacy was poor, 15 and 5.6% respectively. These anthelmintics at these dose rates caused only an elimination of the terminal egg bearing segments and were without significant effect on the entire tapeworm. When pyrantel base was administered at 13.2 and 19.8 mg/kg (twice and three times the therapeutic dose rate for nematodes respectively) the efficacy was 97.8 and 100%. It would appear that pyrantel pamoate administered at 13.2 mg pyrantel base/kg is an effective therapeutic dose for tapeworms in horses. Further dose titration studies are needed for niclosamide. Mebendazole was without effect at up to four (35.2 mg/kg) times the therapeutic dose for nematodes.  相似文献

Dogs were allocated randomly based on pre-treatment quantitative nematode egg counts of each species of interest to one of two treatment groups of 10 or 11 animals each. In each study, spinosad and MO in combination, was given orally to dogs using the lower half (30-45 mg/kg spinosad; 0.5-0.75 mg/kg MO) of the US commercial dose band (30-60 mg/kg spinosad; 0.5-1.0mg/kg MO) of each active ingredient on Day 0 using a tablet formulation. A corresponding vehicle control group was treated similarly in each individual study. Dogs were necropsied post-treatment on Day 7/8. All nematodes in the intestinal tract collected at necropsy were identified and counted by species and stage. The spinosad and MO combination group demonstrated significantly different adult intestinal nematode efficacy in each individual study as compared to the vehicle control group. Efficacy values for whipworm, hookworm, T. canis and T. leonina were 100%, 99.8%, 100%, 93.3%, respectively. Minor non-serious adverse events were observed in a small number of control and treated dogs that were attributed primarily to the natural nematode infections. In summary, flavored spinosad and MO combination tablets administered orally to dogs were both safe and highly efficacious delivering >93% up to 100% adult intestinal nematode control in naturally infected dogs.  相似文献
