The efficacy of 22-oxacalcitriol (OCT), calcitriol, cholecalciferol, all-trans retinoic acid (ATRA) and 9-cis retinoic acid (9-cis RA) to differentiate in vitro four clonal cells of the canine osteosarcoma cell line POS into cells having properties of a functionally mature osteoblast bone cell were investigated. The induction of intracellular alkaline phosphatase (ALP) activity, osteocalcin (GLA-OC) and type I collagen (PIP) production after 72 h treatment were used as markers of differentiation. At a concentration of 10(-8)M, OCT and calcitriol significantly induced all markers, and ATRA only the ALP of osteoblast, chondroblast and undifferentiated clonal cells. At the same concentration, 9-cis RA and cholecalciferol induced ALP of chondroblast and osteoblast cells, respectively; ATRA, 9-cis RA and cholecalciferol induced PIP of chondroblast and undifferentiated cells. None of the drugs significantly differentiated fibroblast cells. The ability of these agents to differentiate osteosarcoma cells into cells that exhibit properties of functionally mature osteoblastic bone cells may promote normal osteogenesis and reverse the loss of control of their differentiation.  相似文献

Although canine osteosarcoma is one of the most malignant, aggressive and lethal neoplasms originating from undifferentiated bone cells, it may retain some capacity for normal differentiation. The purpose of this study was to ascertain if the residual capacity for differentiation could be used to suppress its malignant properties. We tested the efficacy of vitamin D and retinoids in inducing differentiation and inhibiting growth of the POS canine osteosarcoma and four of its clonal cell lines, POS 14A (fibroblast type), POS 53B (chondroblast type), POS 53C (undifferentiated type) and POS 53D (osteoblastic type). Treatment with 10(-10)to 10(-8)M concentrations of calcitriol, OCT, cholecalciferol, all-trans retinoic acid (ATRA) and 9-cis retinoic acid for 48-120 hours changed the morphology of POS, POS 53B, POS 53C and POS 53D cells to cells that were elongated and spindle-shaped. Increased number of cytoplasmic organelles and pronounced nuclear activities were induced by concentrations of 10(-8)M and 10(-7)M for 120 hours. All drugs at concentrations of 10(-10)to 10(-8)M for 72 hours inhibited POS growth dose-dependently. OCT significantly reduced the cell number in all cell lines when used at concentrations between 10(-9)and 10(-8)M for 72 hours and exerted significant anti-proliferative effects for eight days culture. This study demonstrated that changed morphology and inhibition of growth was induced by treatment of the cells with these vitamins, that the loss of control of differentiation in the neoplasia was not irreversible and that these drugs may be useful in the clinic.  相似文献

Pulmonary metastasis is a major cause of death and a major obstacle to the successful treatment of canine osteosarcoma. However, the residual capacity of the neoplasia for differentiation and its susceptibility to undergo apoptosis may be used to suppress its growth and metastatic properties. The highly metastasizing POS (HMPOS) canine osteosarcoma cell line which preferentially metastasize to the lungs was used to test the possible efficacy of 22-oxa-calcitriol (OCT) and all- trans retinoic acid (ATRA) to inhibit growth and pulmonary metastasis of the subcutaneously grown osteosarcoma in nude mice. Treatments in vitro, morphologically elongated and increased alkaline phosphatase activity and staining of cells. Tumour growth in vivo was inhibited significantly and the combination treatment of OCT and ATRA (OCT + ATRA) exerted a synergistic and stronger suppression at concentration of 1.0 microg kg(-1)body weight when given subcutaneously three times a week for 5 weeks. The subcutaneous tumours of the control mice consisted of osteoblast-like cells and isolated chondroblast-like cells, but formed several areas of osteoid and increased amount of collagen tissue in all treated mice. Pinpoint macrometastatic nodules developed only in all control mice. Micrometastatic nodule developed only in two of six mice treated with ATRA. However, nodule size and number, and lung wet weight were all reduced significantly. Metastasis were not seen in the mice treated with OCT or OCT + ATRA. This study demonstrated that inhibition of growth and pulmonary metastasis was induced by subcutaneous treatment with these drugs and suggest that both its differentiating and apoptotic inducing activities may be responsible for the antitumour effects. These drugs may be useful in the clinic as an adjunct for the treatment of canine osteosarcoma.  相似文献

Five ruminally, duodenally, and ileally cannulated steers (376 +/- 8.1 kg of initial BW) were used in a 5 x 5 Latin square to evaluate effects of cooked molasses block supplementation and inclusion of fermentation extract (Aspergillus oryzae) or brown seaweed meal (Ascophyllum nodosum) on intake, site of digestion, and microbial efficiency. Diets consisted of switchgrass hay (6.0% CP; DM basis) offered ad libitum, free access to water, and one of three molasses blocks (0.341 kg of DM/d; one-half at 0600 and one-half at 1800). Treatments were no block (control), block with no additive (40.5% CP; POS), block plus fermentation extract bolused directly into the rumen via gelatin capsules (2.0 g/d; FS), fermentation extract included in the block (2.0 g/d; FB), and seaweed meal included in the block (10 g/d; SB). Steers were adapted to diets for 14 d followed by a 7-d collection period. Overall treatment effect on hay OM intake tended (8.1 vs. 7.6 +/- 0.5 kg/d; P = 0.14) to increase with block supplementation. Total OM intake (8.4 vs. 7.6 +/- 0.5 kg/d; P = 0.01) increased in steers consuming block compared with control. Apparent and true ruminal OM digestibility increased (P = 0.05) with block consumption. Steers fed SB had greater (P = 0.10) true ruminal OM digestibility compared with steers fed POS (61.0 vs. 57.9 +/- 1.6%). True ruminal CP digestibility increased (P = 0.01) with block supplementation compared with control (37.5 vs. 23.6 +/- 3.7%). Addition of fermentation extract did not affect intake or digestion. Treatments did not alter ruminal pH, total VFA, or individual VFA proportions; however, ruminal ammonia increased (P = 0.01) with block supplementation. In situ disappearance rates of hay DM (3.14 +/- 0.44 %/h), NDF (3.18 +/- 0.47 %/h), and ADF (3.02 +/- 0.57 %/h) were not altered by treatment. Seaweed block increased (P = 0.01) slowly degraded CP fraction compared with POS (39.5 vs. 34.0 +/- 2.07%). Similarly, SB increased (P = 0.01) the extent of CP degradability (74.2 vs. 68.9 +/- 1.81%). No treatment effects (P = 0.24) were observed for microbial efficiency. Block supplementation increased intake, and use of brown seaweed meal seemed to have beneficial effects on forage digestibility in low-quality forage diets.  相似文献

Two experiments evaluated digestive and performance effects of field pea-based creep feed in nursing calf diets. In Exp.1, eight nursing steer calves (145 +/- 27 kg initial BW) with ruminal cannulas were used to evaluate effects of supplementation and advancing season on dietary composition, intake, digestion, and ruminal fermentation characteristics. Treatments were unsupplemented control (CON) and field pea-based creep (SUP; 19.1% CP, DM basis) fed at 0.45% BW (DM basis) daily. Calves grazed native range with their dams from early July through early November. Periods were 24 d long and occurred in July (JUL), August (AUG), September (SEP), and October (OCT). Experiment 2 used 80 crossbred nursing calves, 48 calves in yr 1 and 32 calves in yr 2 (yr 1 = 144 +/- 24 kg; yr 2 = 121 +/- 20 kg initial BW), to evaluate effects of field pea-based creep on calf performance. Treatments included unsupplemented control (CON); field pea-based creep feeds containing either 8% (LS); or 16% (HS) salt; and soybean meal/field pea-based creep containing (as-fed basis) 16% salt (HIPRO). Masticate samples from SUP calves in Exp.1 had greater CP (P = 0.05) than those from CON calves. Forage CP and ADIN decreased linearly with advancing season (P = 0.01 and 0.03, respectively). In vitro OM digestibility of diet masticate decreased from JUL to OCT (P < 0.01; 58.5 to 41.3%). Forage intake did not differ (P = 0.33) between treatments but increased linearly with advancing season (1.67, 1.90, 3.12, 3.38 kg/d for JUL, AUG, SEP, and OCT, respectively; P < 0.01). Milk intake (percentage of BW) did not differ (P = 0.56) between CON and SUP calves but decreased linearly (P < 0.01) with advancing season. Supplemented calves had greater (P = 0.03) total intake (g/kg of BW; forage + milk + creep) compared with CON calves. Treatment did not affect (P < 0.30) rate of in situ disappearance of forage or creep. Forage DM, CP, and creep DM disappearance rate decreased linearly (P < or = 0.02) with advancing season. Supplementation decreased (P = 0.05) ruminal pH, whereas ruminal ammonia and VFA concentrations were greater (P < or = 0.02) in SUP calves. In Exp. 2, creep-fed calves had greater ADG and final BW than CON calves (P < 0.01). Calves offered HS tended (P = 0.07) to have increased gain efficiency above CON than LS calves. Field peas can be used as an ingredient in creep feed to increase calf weight gain without negatively affecting ruminal fermentation and digestion.  相似文献

Differentiation induction of canine osteosarcoma cell lines by retinoids   总被引:2,自引:0,他引:2  
The effect of two retinoids, all- trans and 9- cis retinoic acid, on the differentiation of three canine osteosarcoma cells (OOS, HOS, and POS) was examined using markers specifically expressed by phenotypic osteoblasts. Both retinoids induced morphologic differentiation in all the canine osteosarcoma cells. Retinoids enhanced cell flattening and spreading, as well as reduction in cell overlapping. Alkaline phosphatase (ALP) activity and ALP staining was enhanced in OOS, and HOS cells, but decreased in POS cells. These results may suggest that OOS and HOS cells have immature osteoblastic properties and POS cells have mature osteoblastic properties. Retinoids decreased osteocalcin production in all the osteosarcoma cells. They induced an increase in production of type I collagen in HOS and POS cells, but a decrease in OOS cells. These results indicate that retinoids induce differentiation of canine osteosarcoma cells, resulting in an altered expression of their malignant phenotype.  相似文献

Three experiments were conducted to evaluate supplementation of dried distillers grains with solubles (DGS) to spring-calving beef cows (n = 120; 541 kg of initial BW; 5.1 initial BCS) consuming low-quality forage during late gestation and early lactation. Supplemental treatments included (DM basis) 1) 0.77 kg/d DGS (DGSL); 2) 1.54 kg/d DGS (DGSI); 3) 2.31 kg/d DGS (DGSH); 4) 1.54 kg/d of a blend of 49% wheat middlings and 51% cottonseed meal (POS); and 5) 0.23 kg/d of a cottonseed hull-based pellet (NEG). Feeding rate and CP intake were similar for DGSI and POS. In Exp. 1, cows were individually fed 3 d/wk until calving and 4 d/wk during lactation; total supplementation period was 119 d, encompassing 106 d of gestation and 13 d of lactation. Tall-grass prairie hay (5.6% CP, 50% TDN, 73% NDF; DM basis) was fed for ad libitum intake throughout the supplementation period. Change in cow BW and BCS during gestation was similar for DGSI and POS (-5.0 kg, P = 0.61 and -0.13, P = 0.25, respectively) and linearly increased with increasing DGS level (P < 0.01). Likewise, during the 119-d supplementation period, BW and BCS change were similar for DGSI and POS (-72 kg, P = 0.22 and -0.60, P = 0.10) and increased linearly with respect to increasing DGS (P < 0.01). The percentage of cows exhibiting luteal activity at the beginning of breeding season (56%, P = 0.31), AI conception rate (57%, P = 0.62), or pregnancy rate at weaning (88%, P = 0.74) were not influenced by supplementation. In Exp. 2, 30 cows from a separate herd were used to evaluate the effect of DGS on hay intake and digestion. Supplementation improved all digestibility measures compared with NEG. Hay intake was not influenced by DGS (P > 0.10); digestibility of NDF, ADF, CP, and fat linearly increased with increasing DGS. In Exp. 3, milk production and composition were determined for cows (n = 16/treatment) of similar days postpartum from Exp. 1. Daily milk production was not influenced by supplementation (6.3 kg/d, P = 0.25). Milk fat (2.1%) and lactose (5.0%) were not different (P > 0.10). Milk protein linearly increased as DGS increased (P < 0.05) and was greater for DGSI compared with POS. Similar cow performance was achieved when cows were fed DGS at the same rate and level of CP as a traditional cottonseed meal-based supplement. Increasing amounts of DGS did not negatively influence forage intake or diet digestibility.  相似文献

Right ventricular (RV) Tei index (index of myocardial performance) has been demonstrated to be clinically useful in estimating RV function in various human cardiac diseases. The purposes of this study were to validate the correlation between RV Tei index and RV function obtained by cardiac catheterization in healthy dogs, and to evaluate the RV Tei index in dogs with tricuspid regurgitation (TR). In healthy dogs, the RV Tei index significantly correlated with the RV peak +dP/dt (r=-0.80, p<0.0001) and -dP/dt (r=0.69, p=0.0001). In normal dogs, the RV Tei index was not significantly correlated with heart rate, body weight, and age. The RV Tei index significantly increased in dogs with moderate to severe TR (0.39 +/- 0.35, p=0.0015), filariasis (0.46 +/- 0.16, p=0.0131), and trivial to mild TR and severe mitral regurgitation (MR; 0.61 +/- 0.14, p=0.0017) when compared with the normal dogs (0.17 +/- 0.10). In addition, the RV Tei index in dogs with TR significantly increased in association with pulmonary hypertension [PH(-), 0.19 +/- 0.09; PH(+), 0.65 +/- 0.14; respectively p<0.0001]. Our study has demonstrated that RV Tei index is a feasible approach to estimate RV function in dogs and is not influenced by heart rate, body weight, and aging. Further investigations are required to clarify the clinical significance of RV Tei index in dogs with right-sided cardiac diseases.  相似文献

Pregnant St. Croix White and Barbados Blackbelly hair sheep ewes were used to evaluate the effect of supplemental nutrition around the time of lambing on ewe and lamb performance during the dry and wet seasons on St. Croix. Beginning 14 d before expected day of lambing (d 0) and for 21 d postpartum, one group of ewes was fed a pelleted supplement in addition to grazing guinea grass pasture (FEED). Other ewes in the flock grazed pasture only (CONTROL). This study was conducted during the dry season (June through September; FEED n = 14 and CONTROL n = 15) and during the wet season the next year (October through January; FEED n = 11 and CONTROL n = 12). The 24-h milk production of each ewe was measured on d 7, 21, 35, 49, and 63. Ewes were exposed to sterile rams equipped with marking harnesses to detect estrus during the postpartum period. The FEED ewes lost less weight postpartum during both seasons (P < 0.0001) and had higher milk production (P < 0.009) than CONTROL ewes during the dry season. During the dry season, the time to the first postpartum estrus did not differ (P > 0.10) between FEED and CONTROL ewes (46.9 +/- 2.7 vs 52.9 +/- 2.6 d, respectively). During the wet season, the time to first postpartum estrus was less (P < 0.07) in FEED than in CONTROL ewes (33.0 +/- 3.1 vs 41.1 +/- 2.9 d, respectively). The FEED ewes had higher lamb birth weight (P < 0.04) and weaning weight (P < 0.05) than CONTROL ewes (3.2 +/- 0.1 and 12.2 +/- 0.5 vs 2.9 +/- 0.1 and 10.9 +/- 0.5 kg, respectively) during the dry season. In the wet season, lamb birth weight and weaning weight were similar (P > 0.10) between FEED and CONTROL (3.2 +/- 0.1 and 15.5 +/- 0.7 vs 3.1 +/- 0.1 and 15.3 +/- 0.6 kg, respectively). Lambs born during the wet season had higher (P < 0.0001) ADG than lambs born during the dry season (194.4 +/- 5.9 vs 127.7 +/- 4.7 g/d, respectively). Strategic nutritional supplementation of hair sheep ewes may provide a method for increasing the weight of lambs produced during the dry season in the tropics, but it does not seem to be beneficial during the wet season.  相似文献

Residual feed intake (RFI) is the difference between the actual and expected feed intake of an animal based on its BW and growth rate over a specified period. The biological mechanisms underlying the variation in feed efficiency in animals with similar BW and growth rate are not well understood. This study determined the relationship of feedlot feed efficiency, performance, and feeding behavior with digestion and energy partitioning of 27 steers. The steers were selected from a total of 306 animals based on their RFI following feedlot tests at the University of Alberta Kinsella Research Station. Selected steers were ranked into high RFI (RFI > 0.5 SD above the mean, n = 11), medium RFI (RFI +/- 0.5 SD above and below the mean, n = 8), and low RFI (RFI < -0.5 SD below the mean, n = 8). The respective BW +/- SD for the RFI groups were 495.6 +/- 12.7, 529.1 +/- 18.6, and 501.2 +/- 15.5 kg. Digestibility and calorimetry trials were performed on a corn-or barley-based concentrate diet in yr 1 and 2, respectively, at 2.5 x maintenance requirements. Mean DMI (g/kg of BW(0.75)) during the measurements for high-, medium-, and low-RFI groups, respectively, were 82.7 +/- 2.0, 78.8 +/- 2.6, and 81.8 +/- 2.5 and did not differ (P > 0.10). Residual feed intake was correlated with daily methane production and energy lost as methane (r = 0.44; P < 0.05). Methane production was 28 and 24% less in low-RFI animals compared with high- and medium-RFI animals, respectively. Residual feed intake tended to be associated (P < 0.10) with apparent digestibilities of DM (r = -0.33) and CP (r = -0.34). The RFI of steers was correlated with DE (r = -0.41; P < 0.05), ME (r = -0.44; P < 0.05), heat production (HP; r = 0.68; P < 0.001), and retained energy (RE; r = -0.67; P < 0.001; energy values are expressed in kcal/kg of BW(0.75)). Feedlot partial efficiency of growth was correlated (P < 0.01) with methane production (r = -0.55), DE (r = 0.46), ME (r = 0.49), HP (r = -0.50), and RE (r = 0.62). With the exception of HP (r = 0.37; P < 0.05), feed conversion ratio was unrelated to the traits considered in the study. Feeding duration was correlated (P < 0.01) with apparent digestibility of DM (r = -0.55), CP (r = -0.47), methane production (r = 0.51), DE (r = -0.52), ME (r = -0.55), and RE (r = -0.60). These results have practical implications for the selection of animals that eat less at a similar BW and growth rate and for the environmental sustainability of beef production.  相似文献

Effects of ovulation rate and of fetal number and distribution within the uterus on pregnancy rate and fetal survival were evaluated in nulliparous (n = 1,331) and parous (n = 3,517) cattle selected for twinning. Cattle were divided into a spring (70 d) and fall (60 d) breeding season and bred by a combination of AI and natural service. Ovulation rate, pregnancy status, and fetal number and distribution were determined by transrectal, real-time ultrasonography of the uterus and both ovaries at the end of the breeding season. Pregnancy was reconfirmed by rectal palpation at 75 to 135 d of gestation. For heifers and cows combined, ovulation rate increased (P < 0.01) from 1.46 +/- 0.4 in 1994 to 1.89 +/- 0.4 in 2004; number of calves per parturition increased (P < 0.01) from 1.34 +/- 0.3 to 1.56 +/- 0.3, respectively, which included an increase in triplet and quadruplet ovulations and triplet births. Bilateral twin ovulations yielded proportionately more (P < 0.01) twin births than unilateral twin ovulations. Ovulation rate was greater (P < 0.01) in the fall than spring breeding season. Pregnancy rate at ultrasound diagnosis did not differ among females with 1, 2, or 3 ovulations (89.1 +/- 0.7, 91.2 +/- 0.7, or 91.5 +/- 2.8%, respectively), but rates at calving decreased (P < 0.01) with increasing ovulation rate (85.1 +/- 0.6, 82.7 +/- 0.6, or 64.2 +/- 2.7%, respectively). Pregnancy rate was less (P < 0.01) after twin or triplet births than single births. For dams birthing twins or triplets, pregnancy rate was less in the fall vs. spring, but rates were similar between seasons for dams with a single birth (type of birth x season, P < 0.05). Cows 60 d, regardless of type of birth. Maintenance of pregnancy to term differed (P < 0.01) among females diagnosed with 1, 2, or 3 fetuses (95.7 +/- 0.6, 87.8 +/- 0.8, and 54.9 +/- 2.3%, respectively). The reduced survival of twin and triplet fetuses in heifers had occurred (P < 0.01) by d 75 to 135 of gestation, and fetal losses were greater (P < 0.01) for unilateral than bilateral twins or triplets, whereas loss of twin or triplet fetuses in cows occurred later in gestation, and losses were not affected by uterine location. Thus, increased calf production from selecting for increased ovulation rate in beef cattle is tempered by increased fetal mortality, partially associated with the crowding of 2 or 3 fetuses within 1 uterine horn, especially in heifers.  相似文献

The aim of this study was to estimate the genetic correlations between 2 purebred Duroc pig populations (P1 and P2) and their terminal crossbreds [C1 = P1 x (Landrace x Large White) and C2 = P2 x (Landrace x Large White)] raised in different production environments. The traits analyzed were backfat (BF), muscle depth (MD), BW at slaughter (WGT), and weight per day of age (WDA). Data sets from P1, P2, C1, and C2 included 26,674, 8,266, 16,806, and 12,350 animals, respectively. Two-trait models (nucleus and commercial crossbreds) for each group included fixed (contemporary group, sex, weight, and age), random additive (animal for P1 and P2 and sire for C1 and C2), random litter, and random dam (C1 and C2 only) effects. Heritability estimates (+/-SE) for BF were 0.46 +/- 0.04, 0.38 +/- 0.02, 0.32 +/- 0.02, and 0.33 +/- 0.02 for P1, P2, C1, and C2, respectively. Heritability estimates for MD were 0.31 +/- 0.01, 0.23 +/- 0.02, 0.19 +/- 0.01, and 0.12 +/- 0.01 for P1, P2, C1, and C2, respectively. The estimates for WGT and WDA were 0.31 +/- 0.01, 0.21 +/- 0.02, 0.16 +/- 0.01, and 0.18 +/- 0.01 and 0.32 +/- 0.01, 0.22 +/- 0.02, 0.16 +/- 0.01, and 0.19 +/- 0.01, respectively. Genetic correlations between purebreds and crossbreds for BF were 0.83 +/- 0.09 (P1 x C1) and 0.89 +/- 0.05 (P2 x C2), for MD 0.78 +/- 0.05 (P1 x C1) and 0.80 +/- 0.08 (P2 x C2). For WGT and WDA, the correlations were 0.53 +/- 0.08 (P1 x C1), 0.80 +/- 0.10 (P2 x C2), and 0.60 +/- 0.07 (P1 x C1) and 0.79 +/- 0.09 (P2 x C2), respectively. (Co)variances in crossbreds were adjusted to a live BW scale. Compared with purebreds, the genetic variances in crossbreds were lower, and the residual variances were greater. Sire variances in crossbreds were approximately 20 to 30% of the animal variances in purebreds for BF and MD but were 13 to 25% for WGT and WDA. The efficiency of purebred selection on crossbreds, assessed by EBV prediction weights, ranged from 0.43 to 0.91 for line 1 and 0.70 to 0.92 for line 2. When nucleus and commercial environments differ substantially, the efficiency of selection varies by line and traits, and selection strategies that include crossbred data from typical production environments may therefore be desirable.  相似文献

Thermoregulation may limit exercise performance under hot and humid conditions. This study compared heart rate (HR), respiratory rate (RR), rectal temperature (Tr), packed cell volume (PCV) and total plasma protein concentration (TPP) during a submaximal incremental field exercise test under high vs. low ambient temperature and relative humidity. Ten horses were tested 3 times in summer (July) and 3 times in autumn (September). Heart rate was measured continuously, the other variables at rest and immediately after 4 min at 3.5, 4.5 and 7.0 m/s, separated by 3 min rest intervals, and after 5 and 10 min recovery. Data for all variables were significantly greater during exercise and recovery in the hot vs. cool conditions, respectively: after 4 min at 7.0 m/s, HR was 135+/-1 and 123+/-1/min (P<0.0001), Tr was 39.0+/-0.06 and 38.0+/-0.05 degrees C (P<0.0001), RR was 99+/-3 and 50+/-3/min (P<0.0001), PCV was 48.8+/-0.06 and 42.1+/-0.3% (P<0.0001) and TPP was 7.7+/-0.14 and 7.6+/-0.12 g/l (P = 0.026). These data reflect the thermal burden during submaximal exercise under hot conditions in the field. The greater relative PCV increase in the heat probably conferred a thermoregulatory advantage and reflected a greater circulating red cell volume increase rather than a decrease of plasma volume. Six experiments were conducted to evaluate dietary cation-anion difference (DCAD) in concentrate diets on urinary pH, feedlot performance, and N mass balance. In Exp. 1, 15 wether lambs (33.5 ± 3.0 kg) in five 3 × 3 Latin squares were fed a basal diet of 82.5% dry-rolled corn (DRC), 7.5% alfalfa hay, 5% molasses, and 5% supplement with different proportions of anionic and cationic salts. The DCAD was -45, -24, -16, -8, 0, +8, +16, +24, +32, and +40 mEq per 100 g of DM with the control basal diet (DCAD = +8) included in each square. Urinary pH increased (cubic, P < 0.01) as DCAD increased and DMI increased linearly (P < 0.01) with increasing DCAD. In Exp. 2 and 3, 8 Holstein steers (312 ± 24 kg) were used in 2 consecutive 4 × 4 Latin squares. Steers were fed either the same basal diet as Exp. 1 or a basal diet with 20% wet distillers grains (WDGS) replacing DRC. In Exp. 2, DCAD was adjusted to -2, -12, and -22 mEq per 100 g of DM from the basal diet (DCAD = +8) and DCAD was adjusted in Exp. 3 to -12, -22, and -32 mEq per 100 g of DM from the basal WDGS diet (DCAD = -2). Urinary pH decreased linearly as DCAD decreased (P < 0.01) in both experiments, whereas DMI decreased linearly in Exp. 2 (P = 0.02) but not Exp. 3 (P = 0.96). In Exp. 4, 6 crossbred steers (373 ± 37 kg) were used in a 2-period crossover design. Steers were fed the same basal diet as Exp. 3 with DCAD of -16 (NEG) and +20 (POS) mEq per 100 g of DM. Urinary pH and DMI (P < 0.05) were less for cattle fed the NEG diet compared with POS. In 2 experiments, steers (n = 96 each) were fed NEG or POS as calves (260 ± 22 kg of BW) for 196 d from November to May (Exp. 5) or as yearlings (339 ± 32 kg of BW) for 145 d from June to October (Exp. 6). Final BW, DMI, ADG, and HCW were not different (P > 0.11) among treatments in either experiment. Efficiency of BW gain was increased (P = 0.05) for steers fed NEG compared with POS in Exp. 5 but was not different (P = 0.11) in Exp. 6. Amount of N intake, retention, excretion, and manure N (kg/steer) were not different (P > 0.11) among treatments in either experiment. Manure pH (soil, feces, and urine) was decreased (P < 0.01) in pens fed NEG compared with POS in both experiments. Amount of N lost (kg/steer) was not different (P = 0.44) in Exp. 5, but tended (P = 0.09) to be 10.6% greater for POS compared with NEG in Exp. 6. Urinary pH was decreased by reducing DCAD, but this had minimal effect on N losses in open feedlot pens in these experiments.  相似文献

The objective was to determine the effects of repeated injections of PGF2alpha (Lutalyse; Pfizer) on semen and libido characteristics in terminal-line boars. Semen was collected once weekly from wk 0 to 15 and on four consecutive days during wk 16. Boars received an i.m. injection of 10 mg of PGF2alpha (n = 11) or 2 mL of vehicle (n = 11) immediately before entering the collection room. For the weekly collections, semen volume (220.3 +/- 3.2 mL; mean +/- SE), gel weight (38.7 +/- 0.7 g), total sperm cells (65.4 +/- 1.2 billion), motile sperm cells (67.4 +/- 0.6 %), and sperm velocity (125.9 +/- 1.2 microm/s), were affected by time (P < 0.01) but not by treatment (P > 0.10). Sperm concentration (0.31 +/- 0.01 billion/mL) was not affected (P > 0.10) by time or treatment. The percentage of morphologically normal sperm cells, assessed at wk 16, did not differ (P = 0.39) between groups (80.8 +/- 1.0). Libido was evaluated from wk 0 to 16. There were no effects of treatment or time (P > 0.10) on the period from injection to the start of ejaculation (225.6 +/- 9.1 s). Duration of ejaculation was affected by treatment (P < 0.01; 472.0 +/- 43.1 s and 280.4 +/- 43.1 s, for PGF2alpha-treated and control boars, respectively) and time (P < 0.01). During the intensive collection period (wk 16), semen volume (200.1 +/- 7.1 mL), gel weight (39.2 +/- 1.5 g), sperm concentration (0.19 +/- 0.01 billion/ mL), total sperm cells (39.5 +/- 3.6 billion), motile sperm cells (65.6 +/- 2.2%), and sperm velocity (117.8 +/- 3.7 microm/ s) were affected by time (P < 0.10) but not by treatment (P > 0.10). The period from injection to the start of ejaculation tended to decrease (by 44%) during the intensive collection period in PGF2alpha-treated boars, but not in controls (treatment x time, P = 0.07). Regardless, the period from injection to the start of ejaculation did not differ (P = 0.63) between groups on d 4 of the intensive collection period. Duration of ejaculation was affected by treatment (P < 0.01; 459.1 +/- 24.1 s for PGF22alpha-treated boars vs. 303.1 +/- 24.1 s for controls) but not by time (P > 0.10). Overall, there were no exceptional positive or negative effects of long-term treatment with PGF2alpha on semen characteristics and libido in boars.  相似文献

A 2-yr study using primiparous and multiparous, spring-calving, crossbred beef cows was conducted to evaluate the effects of supplemental whole corn germ on reproductive performance, calf performance, and serum leptin concentrations. Each year, cows were blocked by age and BCS and assigned randomly to one of three treatments: PRE (n = 115) cows received 1.14 kg/d (DM basis) of whole corn germ for approximately 45 d before calving; POST (n = 109) cows were fed 1.14 kg/d of whole corn germ for approximately 45 d after calving; and control cows (n = 118) were fed similar energy and protein from dry-rolled corn (1.82 kg of DM/d) for 45 d before and after calving. Additionally, PRE cows were grouped with controls after calving, and POST cows were grouped with control cows before calving, so that corn germ-supplemented cows received the control supplement in the alternate feeding period. Cow BW (538 +/- 13 kg) and BCS (5.4 +/- 0.13) did not differ among treatments at any time during the experiment. Calf birth weight (39 +/- 2 kg), weaning weight (225 +/- 7 kg), and age-adjusted weaning weight (234 +/- 8 kg) did not differ because of dam supplementation regimen. Treatment did not affect the proportion of cows exhibiting ovarian luteal activity before the start of the breeding season (67%) or pregnancy rate (91%). The interval from exposure to bulls until subsequent calving did not differ (P = 0.16) among PRE (298 +/- 2.3 d), POST (303 +/- 2.6 d), and control (304 +/- 2.3 d) cows. Leptin concentrations did not differ among treatments and were 2.15 +/- 0.75, 1.88 +/- 0.76, and 1.91 +/- 0.75 ng/mL for control, POST, and PRE cows, respectively. Age and week relative to calving influenced leptin concentration. Primiparous cows had similar leptin concentrations to 3-yr-old and mature cows for wk -7 and -6 relative to calving, but lower (P < 0.10) concentrations than mature cows for wk -5, and lower (P < 0.05) concentrations than either 3-yr-old or mature cows for wk -4 to +7 relative to calving. Serum leptin was correlated with BCS (P < 0.0001; r = 0.35) at initiation of the feeding period and was correlated with BCS (P = 0.02; r = 0.12) and weight (P < 0.01; r = 0.14) at the completion of the supplement period, but it was not correlated with initial BW or interim BCS. Calving interval was not correlated (P > 0.12) with weekly measures of serum leptin concentration. Supplementing beef cows with whole corn germ had no effect on cow performance, calf performance, or serum leptin concentrations of cows.  相似文献

To evaluate production and economic effects of feeding management strategy and age on intensively managed culled beef cows, a study was conducted using 125 cows of British breeding blocked by age (Young = 3 and 4 yr olds; LowMid = 5 and 6 yr olds; HighMid = 7 and 8 yr olds; and Aged = 9 yr and older) and assigned to one of three steam-flaked corn based feeding strategies. Treatments were as follows: Conservative (CSV), 30% roughage throughout; Standard (STD), decrease roughage from 30 to 10% over 20 d; and Aggressive (AGR), decrease roughage from 30 to 10% over 10 d. There were four pens per treatment in a randomized complete block design. Cows were fed for a total of 54 d, and BW was measured on d 0, 14, 28, 42, and 54. Half the cows from each pen were randomly selected and slaughtered at a commercial abattoir, and carcass data were collected. Average daily gain, daily DMI, and G:F during each weigh period and across the entire feeding period were calculated. Over the 54-d feeding period, strategies that employed more energy-dense diets numerically increased ADG (1.28, 1.63, and 1.55 +/- 0.14 kg/d for CSV, STD, and AGR; P = 0.26) and decreased DMI (11.91, 10.74, and 10.89 +/- 0.27 kg/d for CSV, STD, and AGR; P = 0.05), such that G:F was lower for CSV than for STD or AGR (0.105, 0.150, and 0.141 +/- 0.010; P = 0.05). Carcass weight was least for the CSV strategy (298 kg) and greatest for STD (328 kg); AGR resulted in intermediate carcass weight (317 +/- 6 kg; P = 0.04). Total cost of gain was over 30% greater for CSV strategy than for STD or AGR strategies (P < 0.01). In many cases, block effects (age) had a greater effect on responses than treatments. Average daily gain, DMI, and G:F decreased linearly with age (P < 0.01). Hot carcass weight, dressing percent, and fat thickness decreased linearly with age (P < 0.03); yield grade decreased and carcass maturity attributes increased linearly with age (P < 0.02). Performance and intake differences resulted in linear increases in total cost of gain (P < 0.01) and breakeven price (P = 0.03) with increasing age. These data indicate advantages to more aggressive feeding management strategies for culled beef cows, although maximal intake may be achieved with higher-roughage diets. Despite management effects, an increase in market price above purchase price may be required for intensive feeding of culled beef cows to be a profitable enterprise.  相似文献

This study determined whether the interval from estrus to ovulation was altered by giving P.G. 600 to sows at weaning. Mixed-parity sows received P.G. 600 i.m. (n = 72) or no treatment (n = 65) at weaning (d 0). Beginning on d 0, sows were observed for estrus twice daily. At the onset of estrus and thereafter, ultrasound was performed twice daily to determine the average size of the largest follicles and time of ovulation. Weaning age (20.1+/-0.4 d) did not differ (P > 0.10) between treatments. More P.G. 600 sows expressed estrus within 8 d (P < 0.01) than controls (94.4% vs 78.4%, respectively). Parity was associated with expression of estrus (P < 0.02), with 78% of first-parity and 93% of later-parity sows exhibiting estrus. However, no treatment x parity effect was observed (P > 0.10). The interval from weaning to estrus was reduced (P < 0.0001) by P.G. 600 compared with controls (3.8+/-0.1 d vs 4.9+/-0.1 d). Follicle size at estrus was not affected by treatment (P > 0.10). The percentage of sows that ovulated did not differ (P > 0.10) for P.G. 600 and control sows (90.3% vs 81.5%, respectively). Time of ovulation after estrus was not affected by treatment and averaged 44.8 h. However, univariate analysis indicated that the interval from weaning to estrus influenced the interval from estrus to ovulation (r = 0.43, P < 0.0001). Further, multivariate analysis showed an effect of treatment on the intervals from weaning to estrus, weaning to ovulation (P < 0.0001), and estrus to ovulation (P < 0.04). Within 4 d after weaning, 81% of the P.G. 600 sows had expressed estrus compared with 33% of controls. However, this trend reversed for ovulation, with only 35% of P.G. 600 sows ovulating by 36 h after estrus compared with 40% of controls. The estrus-to-ovulation interval was also longer for control and P.G. 600 sows expressing estrus < or = 3 d of weaning (45 h and 58 h, respectively) than for sows expressing estrus after 5 d (39 h and 32 h, respectively). Farrowing rate and litter size were not influenced by treatment. However, the interval from last insemination to ovulation (P < 0.02) indicated that more sows farrowed (80%) when the last insemination occurred at < or = 23 to > or = 0 h before ovulation compared with insemination > or = 24 h before ovulation (55%). In summary, P.G. 600 enhanced the expression of estrus and ovulation in weaned sows but, breeding protocols may need to be optimized for time of ovulation based on the interval from weaning to estrus.  相似文献

A cooperative study involving six experiment stations and 236 crossbred litters was conducted to determine the effect of nominal nipple drinker water flows of 700 mL/min and 70 mL/min (actual = 701 and 76 mL/min, respectively) during winter (November through February; 124 litters) and summer (June through August; 112 litters) seasons on performance of lactating sows and their litters. Within a season, sows were paired according to expected farrowing date and assigned at random to crates. Water flow rate treatments were assigned at random to sows within pairs. Sows were housed in farrowing crates from d 109 of gestation until either d 21 (two stations) or d 28 of lactation (four stations). Within 24 h after farrowing, litters were adjusted to contain 8 to 12 piglets. Sow feed intake (SFI) and litter weight (LW) were recorded weekly. Sow weights were recorded at d 109 of gestation, d 0, and d 21 of lactation. Sows lactating beyond 21 d were also weighed on d 28. Analysis of covariance was applied to sow weight change, average daily SFI, and LW data where litter size after crossfostering was the covariate. Average ambient temperature 30 cm above the floor at 0830 and 1600 was 24.6 +/- 0.15 degrees C and 29.4 +/- 0.14 degrees C, respectively, during summer and 20.7 +/-0.13 degrees C and 21.8 +/- 0.11 degrees C during winter trials. Restricted drinker water flow rate decreased SFI (P < 0.01; 4.59 vs. 3.94 kg/d, respectively, for 700 and 70 mL/min) and increased BW loss (P < 0.01; 0.56 vs 0.89 kg/d, respectively for 700 and 70 mL/min) but did not affect litter size (P > 0.87) or LW (P > 0.89) during the first 21 d of lactation. During d 22 to 28, the 70 mL/min flow decreased SFI (P < 0.01; 5.02 vs. 4.47 kg/d respectively, for 700 and 70 mL/min). Over the 21-d lactation period, the 70 mL/min treatment depressed (P < 0.01) SFI more during the winter (5.12 vs. 4.24 kg/d for 700 and 70 mL/ min, respectively) than during the summer (4.05 vs 3.65 kg/d for 700 and 70 mL/min, respectively). Season affected SFI (P < 0.01; 4.68 vs. 3.85 kg/d, respectively, for winter and summer), sow weight loss (P < 0.001; 0.46 vs 0.83 kg/d, respectively, for winter and summer), and LW at 21 d (P < 0.05; 52.8 vs. 49.6 kg, respectively, for winter and summer) but not (P > 0.96) the number of pigs per litter. Results of this study suggest that ample access to drinking water and controlling ambient temperature during summer months are essential for sow and litter performance.  相似文献
