Continuous low-dose-rate irradiation of animal tumors has become feasible through the implantation of iridium-192 by an afterloading technique. 192Ir is an ideal radioisotope for interstitial brachytherapy in veterinary medicine because of its 74.2-day half-life, medium-energy gamma radiation (0.35 MeV), moderate shielding requirements, low cost, and adaptability to most tumor configurations. The afterloading technique can easily be applied to veterinary patients. The effectiveness of interstitial brachytherapy may be enhanced by combination with surgery and/or hyperthermia. Computer-assisted dose calculations make afterloaded 192Ir implantation an accurate, precise method of interstitial brachytherapy.  相似文献

Seventeen horses with a total of 18 ocular or periorbital squamous cell carcinomas were irradiated with Strontium-90 surface therapy, or Radon-222, Iodine-125, or Iridium-192 interstitial implants. Tumors were surgically cytoreduced prior to irradiation in all but two horses. Follow-up data was obtained for 2 years in each horse. Two-year-non-recurrence rates (no grossly visible evidence of tumor) were 87.5% for 90Sr beta therapy, and 60% (70% when corrected for non-tumor-related deaths) for interstitial implant therapy. Data regarding frequency of occurrence, location of lesions, age of onset, and non-recurrence rates were similar to previous literature reports utilizing variable or shorter follow-up periods. The effects of cytoreductive surgery of the tumors was not studied.  相似文献

Equine sarcoid is the most common tumour of horses and accounts for over half of all equine skin tumours. Six types of sarcoid based on gross appearance and clinical behaviour have been described including occult, verrucous, nodular, fibroblastic, mixed and malevolent. Common locations for sarcoid development include the periocular region, ear pinnae, lips, neck, extremities and ventrum (including groin region). Bovine papillomavirus (BPV) is causally associated with equine sarcoid with genetic haplotype, fly vectors and skin trauma identified as potential risk factors for development of the disease. Histopathology is required for definitive diagnosis of equine sarcoid but incomplete excision is thought to activate latent BPV and stimulate growth. Although there are no uniformly effective treatment options, several modalities have been successful in eliminating or managing equine sarcoid. Surgical excision, intratumoural chemotherapy, cryotherapy, hyperthermia, radiotherapy, immunotherapy and immune modulators are used with degrees of success relative to the accessibility and invasiveness of the tumour. Prevention of equine sarcoid may be facilitated by future development of vaccines against bovine papillomavirus.  相似文献

An electronic form of interstitial brachytherapy was used to treat tumours on 3 different horses. One horse was treated for an ocular lymphoma, the second for a sarcoid, and the third for melanoma. This form of brachytherapy contains inherent advantages over previously used forms of brachytherapy and provides another treatment option for equine tumours that cannot be treated with, or do not respond to, conventional tumour therapies.  相似文献

Treatment of the equine sarcoid has posed a significant challenge to clinicians for years and many different methods have been tried with varying success, including ionising radiation. The aim of this study was to review the efficacy of iridium-192 interstitial brachytherapy for the treatment of eight periocular sarcoids and 15 non-ocular sarcoids on 18 horses. All the periocular sarcoids and 13 of the 15 non-ocular sarcoids were treated successfully.  相似文献

The leukotrienes (LT) LTD4 and LTB4 have been shown to cause bronchoconstriction and neutrophil accumulation, respectively, in horse lungs. Such changes are characteristic of the equine allergic respiratory disease, chronic obstructive pulmonary disease (COPD). To further investigate the role of these putative mediators in the pathogenesis of equine COPD the effect of a 5-lipoxygenase inhibitor, fenleuton, on antigen-induced changes in horses with this condition has been examined. Six horses with COPD underwent a series of four antigen challenges, one month apart, with placebo pre-treatment on three occasions and fenleuton (4 days oral dosing 5 mg/kg) pre-treatment on one occasion. Three horses received fenleuton prior to the second challenge and three horses received the drug prior to the fourth antigen challenge. Changes in radiolabelled neutrophil distribution, lung function and peripheral leucocyte counts were monitored on each occasion for 7 h following the start of antigen challenge. Antigen challenge caused an increase in radioactive counts over the lungs and a decrease in peripheral leucocyte count. Neither response was affected by fenleuton pre-treatment. Mean maximal changes in pleural pressure (ΔPplmax) and respiratory rate were also unaffected by fenleuton pre-treatment. However, in the two horses which responded to antigen-challenge with a particularly marked increase in ΔPplmax (>15 cm H2O), prior administration of fenleuton reduced the response by 64 and 63%. These results suggest that 5-lipoxygenase inhibitors warrant further investigation as bronchodilators in equine COPD.  相似文献

A retrospective study examined data regarding equine cutaneous and mucocutaneous neoplasms submitted to the Veterinary Diagnostic Laboratory at Oregon State University in a 3.5-year period. A total of 536 neoplasms were identified, accounting for 30% of the total equine pathology submissions. Sarcoid, squamous cell carcinoma, melanocytic tumors, papillomas, and mast cell tumors were the most common neoplasms, constituting 87.5% of all cutaneous neoplasms. Sarcoids represented 51.4% of all neoplasms and 15.18% of total equine accessions. Sarcoid was most common in paints, quarter horses, and Arabians, and was the only common tumor in donkeys and mules. Mean age at diagnosis of equine sarcoid was 9 years. Squamous cell carcinoma constituted 18.3% of all neoplasms and 5.41% of total equine accessions. Ocular squamous cell carcinoma was most common in paints and quarter horses, and penile/preputial squamous cell carcinoma was most common in appaloosas and quarter horses. The mean age of horses with ocular squamous cell carcinoma (13 years) and squamous cell carcinoma of the skin (15 years) was significantly less (P < 0.5) than that of horses with squamous cell carcinoma of the penis and prepuce (21 years) or vulva, anal, and perianal skin (19 years). Findings suggest that equine sarcoid and squamous cell carcinoma occur more frequently in the Pacific Northwest than in the northeastern United States.  相似文献

The osteoinductive capability of four particle sizes of equine demineralized bone matrix (DBM) was evaluated. Matrix particles were implanted in brachiocephalicus muscle pouches in six horses and were harvested 8 weeks later. Matrix particle sizes of 2.0 mm3 to 4.0 mm3 and 5.0 mm3 to 10.0 mm3 were associated with osteoinductive activity and minimal signs of local inflammation. The two smaller particle sizes (0.425 mm3 to 0.850 mm3 and 0.850 mm3 to 2.0 mm3) were minimally osteoinductive and were associated with a greater local inflammatory response. Microscopic events associated with new bone production in the two largest particle sizes included the presence of new osteoprogenitor cells in matrix vascular spaces, multinucleated osteoclast-like giant cells, and the formation of cartilage, osteoid, and new mineralized bone. The two smaller particle sizes were associated with increased fibrous tissue ingrowth and the presence of increased numbers of inflammatory cells. The temporal events of osteoinduction in response to heterotopic implantation of allogeneic DBM were delayed in horses when compared with other species. Although the demineralization technique used was effective in previous investigations on other species, incomplete demineralization occurred in the two smaller particle sizes that was not detected until after DBM implant harvest. The demineralization process may need to be altered for equine bone and precise monitoring for complete demineralization before implantation is required. Equine DBM is osteoinductive in muscle pouch sites in horses but matrix particle size does affect osteoinductive capability.  相似文献

In horses, osteosarcoma is a rare tumor, with the majority of reported cases occurring in the head, and, more specifically, in the mandible of young horses. The following report documents 8 cases of equine osteosarcoma, the majority occurring in male horses aged 7 years or older with a lack of metastasis identified in any horse. Six arose in the maxilla or mandible and one in the proximal tibia. The predominant subtype was fibroblastic osteosarcoma with fewer osteoblastic type tumors. All had osteoid and most had a chondromucinous matrix. Surgical excision was attempted in the majority of cases. An inability to completely excise the tumor and progressive disease typically resulted in euthanasia. To the authors' knowledge, this case series also documents the first report of an equine extraosseous osteosarcoma within the subcutaneous tissue caudal to the shoulder. Surgical excision appears successful with no recurrence of disease 14 months later. Further investigations of equine osteosarcoma and various chemotherapeutic agents are warranted to present additional treatment options.  相似文献

Beta2-adrenoceptor agonists are used as bronchodilators in both humans and horses. Of these drugs, clenbuterol is the one most frequently used when treating chronic obstructive pulmonary disease in the horse, while salbutamol and terbutaline are used in the treatment of human asthma. Little is known of the properties of the latter two drugs in equine medicine.
  We have compared salbutamol and terbutaline with clenbuterol in relation to their ability to relax muscle strips from equine tracheal muscle, pre- contracted with 40 n m carbachol, in tissue chambers. The affinities of these drugs to the β2-adrenoceptors in homogenates of the same muscle tissue were also examined. These experiments were performed with radioligand binding studies using the very potent β-adrenoceptor antagonist 125I-cyanopindolol.
  The three drugs were almost equipotent in relaxing the muscle strips. The EC50-values for salbutamol, terbutaline and clenbuterol were 5.6, 13.8 and 2.1 n m , respectively, and all three drugs relaxed the preparations completely. In the competitive binding study, however, the Kd-value of clenbuterol was much lower (24 n m ) than that of salbutamol and terbutaline (1100 n m and 3900 n m , respectively). The amount of receptors bound at the EC50-value of clenbuterol was 8% compared to less than 1% for salbutamol and terbutaline. This indicates a lower intrinsic efficacy of clenbuterol than of the other two drugs. The β-adrenoceptor density was 45 ± 14.3 fmol/mg protein (mean ± SD) and the Kd-value of 125I-cyanopindolol was 11.4 ± 3.3 p m .  相似文献

SUMMARY: Confinement of horses with their heads elevated for periods up to 24 hours was used to evaluate the extent and the effects of bacterial contamination of the equine lower respiratory tract. Significant (P < 0.05) increases in bacterial numbers (up to 109 colony forming units/mL in transtracheal aspirate derived samples) occurred within 6 or 12 hours in most horses. Pasteurella/Actinobacillus spp and Streptococcus spp were most commonly isolated. Lowering of the head for 30 minutes every 6 hours to facilitate postural drainage did not prevent multiplication of organisms to levels equivalent to those achieved by horses where the head was elevated for 24 hours. When horses were released from confinement and heads were no longer maintained in an elevated position, clearance of accumulated secretions and bacteria occurred within 8 to 12 hours. Thus, confinement with the head elevated resulted in significant bacterial contamination and multiplication within the lower respiratory tract during a period often encountered in routine management procedures, such as transportation. The clearance of accumulated secretions occurred over a prolonged period after release from such confinement.  相似文献

The distribution of equine leucocyte antigens (ELA) in Swedish Halfbreds affected by sarcoid tumors was determined and compared with that of control horses of the same breed. ELA-haplotype A3W13 appeared more frequently in affected horses, resulting in a chi 2 value of 4.45 (P = 0.034) for A3 and 9.05 (P = 0.0026) for W13, respectively. The relative risk factor (RR) could be estimated to 2.13 and 3.00 for A3 and W13, respectively. The etiology fraction (EF) was calculated to 28% and 37% for A3 and W13, respectively. Thus, in the population of Swedish Halfbreds approximately 40% (at least) of the disease appeared to be associated with the genetic background of the affected horse. Family studies established that ELA are codominantly expressed and inherited as simple Mendelian traits and that sarcoids among offspring are significantly associated with one of the parental haplotypes (P = 0.00942). This parental haplotype does not always include A3W13. These results confirm and extend previous results from other breeds and strongly suggest the existence of a predisposition for sarcoids among horses, that is due to an autosomal, dominant, ELA-linked gene with incomplete penetrance. In extension, this indicates a multifactorial etiology of equine sarcoids (additional non-MHC gene(s) and/or environmental factors).  相似文献

Introduction:  Greater than 50% of dogs with thyroid tumors present with surgically unresectable disease for which external beam radiotherapy has been reported to prolong survival. The success of 131I for control of thyroid tumors in cats and in humans suggests such therapy may also play a role in the management of canine thyroid cancer.
Methods:  Thirty‐nine dogs with WHO stage II/III (invasive or ectopic; n = 32) or IV (metastatic; n = 7) thyroid tumors were treated with 131I alone. Changes in thyroid function, 99MTc‐pertechnetate (99MTc) scintigraphic changes, and tumor response were recorded. Dogs with ventral cervical tumors were evaluated for feasibility of surgical resection following 131I.
Results:  Median overall survival was 839 days and 366 days for dogs with stage II/III and stage IV tumors, respectively. Thyroid hormone status, site and surgical resection were not associated with outcome in dogs with stage II/III tumors. Three dogs developed severe bone marrow suppression.
Conclusions:  These findings suggest 131I should be investigated more thoroughly in dogs with thyroid tumors not considered surgical candidates to more clearly characterize the indications for therapy and followup recommendations. 131I dosimetry in dogs with thyroid tumors remains problematic. Administration of 131I is currently based on empiric recommendations and, in general, the treatment is well tolerated although additional studies are indicated to optimize response and minimize toxicity.  相似文献

The purpose of this retrospective study was to evaluate six cases of equine sarcoidosis for initial presenting symptoms, response to therapy and actual outcome. Dermatologists and dermatopathologists from Europe, the United States, Australia and Canada were contacted to obtain these six cases, as this is a rare disease. Signalment, clinical signs, histological findings, clinical management and outcome were determined via a questionnaire and compared to former reports. There was no age or breed predilection, and four of six horses were geldings. Age of onset ranged from 3 months to 17 years. Onset of the disease was insidious or rapid. Interestingly, in five of six cases, scaling began on the trunk (girth and shoulder). Scaling, crusting and alopecia were seen in all six horses. In one horse, clinical signs of systemic disease were reported and included intermittent fever, prescapular lymphadenopathy, depression, poor body condition and nasal discharge. Treatment included phenylbutazone, deworming agents, antibiotics, short-term low-dose corticosteroids, and 1–1.5 mg/kg of prednisolone. One horse showed a partial response to trimethoprim and sulfonamide, and five of six went into clinical remission with corticosteroid treatment. Five of six horses were still alive 1 year after diagnosis; one horse was diagnosed <12 months ago. Two horses are in complete remission 4 and 8 years after diagnosis. In both horses, clinical signs recurred after cessation of therapy and went into remission again with reintroduction of treatment. Both of these horses have been in remission for several years without therapy.

This report presents the results of 12 adult horses with 16 sarcoids which received radiotherapy from iridium-192 interstitial implants. Fourteen of the 16 tumors (87.5%) were in remission for at least 12 months, and 13 (81.2%) were in remission for at least 24 months. Generally, normal tissue reactions were within acceptable limits. The results of this study further substantiate the beneficial nature of radioactive brachytherapy for equine sarcold.  相似文献

Pulmonary fibrosis and interstitial lung disease are poorly understood in horses; the causes of such conditions are rarely identified. Equine herpesvirus 5 (EHV-5) is a gamma-herpesvirus of horses that has not been associated with disease in horses. Pathologic and virologic findings from 24 horses with progressive nodular fibrotic lung disease associated with EHV-5 infection are described and compared with 23 age-matched control animals. Gross lesions consisted of multiple nodules of fibrosis throughout the lungs. Histologically, there was marked interstitial fibrosis, often with preservation of an "alveolar-like" architecture, lined by cuboidal epithelial cells. The airways contained primarily neutrophils and macrophages. Rare macrophages contained large eosinophilic intranuclear viral inclusion bodies; similar inclusion bodies were also found cytologically. The inclusions were identified as herpesviral-like particles by transmission electron microscopy in a single horse. In situ hybridization was used to detect EHV-5 nucleic acids within occasional macrophage nuclei. With polymerase chain reaction (PCR), the herpesviral DNA polymerase gene was detected in 19/24 (79.2%) of affected horses and 2/23 (8.7%) of the control horses. Virus genera-specific PCR was used to detect EHV-5 in all of the affected horses and none of the control horses. EHV-2 was detected in 8/24 (33.3%) of affected horses and 1/9 (11.1%) of the control horses. This disease has not been reported before, and the authors propose that based upon the characteristic gross and histologic findings, the disease be known as equine multinodular pulmonary fibrosis. Further, we propose that this newly described disease develops in association with infection by the equine gamma-herpesvirus, EHV-5.  相似文献

Intramuscular desmoid tumors (musculoaponeurotic fibromatosis) were discovered in two young adult horses. The tumor in one horse was in the lateral cervical musculature, and that in the second horse occurred in the pectoral musculature. Histopathologic features were similar in both horses and included proliferation of fibroblasts and cells expressing muscle actin (myofibroblasts), with extensive dissecting fibrosis within muscle. These features are similar to those of desmoid tumors in humans, particularly those also known as musculoaponeurotic fibromatosis. Dissection of these lesions revealed a single central (horse No. 1) or multiple central (horse No. 2) fluid-filled cavities with associated sterile inflammation. The presence of these cavities supports the hypothesis that equine desmoid tumors are traumatic in origin, possibly occurring at sites of injections or bursal rupture. Surgical excision of the tumor in horse No. 1 was apparently curative, but the extent of the tumor in horse No. 2 precluded surgical excision.  相似文献

Clinical observations on the use of BCG cell wall fraction in oil for treating seven horses with periocular sarcoids and five horses with sarcoids in other regions are described. Therapy was successful when used for periocular or solitary and smaller sarcoids. For sarcoids previously treated with cryosurgery, therapy appeared to be less efficient. A horse with a sarcoid on the stifle developed a septic gonitis due to necrosis of the sarcoid tissue adjacent to the joint. Sarcoids of the axilla appeared to be more aggressive than sarcoids at other locations and did not respond favourably to this form of therapy.  相似文献

The objective of this study was to investigate clinical signs indicating diseases with known or suspected hereditary components like equine sarcoid, insect bite hypersensitivity, osteochondrosis, allergic airway diseases, podotrochleosis, prognatism and wind-sucking in the franches-montagnes horse.We performed a clinical examination on 702 three-year-old, privately owned franches-montagnes horses, which were shown at the Swiss-Field-Tests in 2004.With the help of the owners a questionnaire on health, environment and feeding habits of the animals was completed. In 11.9% of the horses, sarcoids were detected, 6.2% carried one tumor and 5.7% had several. The prevalence was higher in chestnuts (16.6%) than in bays (10.1%). The prevalence of sarcoids in offspring from sires with known sarcoids (data from Station-Tests 1994-2005) was not significantly higher than in descendants from stallions without a known history of sarcoids. Clinical signs of insect bite hypersensitivity were only found in six horses (0.9%). In 12.0% hoof abnormalities like brittle horn, ring hoofs or hoof cracks of different degrees were recorded. The angle between hoof base and hoof wall was 56.7 +/- 0.1 degrees, the average hoof width was 13.7 +/- 0.3 cm in the front feet. For both of these measures we found no significant difference between left and right feet. With the exception of a high sarcoid prevalence, our results indicate that the franches-montagnes horse is a healthy breed overall.  相似文献
