The primary objective was to evaluate the role of non-ovarian oxytocin in the initiation of pulses of PGF2α, as measured by peripheral concentrations of 13,14-dihydro-15-keto-prostaglandin F2α (PGFM). A 2 × 2 factorial arrangement of estradiol and progesterone treatments was administered to groups of five ewes after ovariectomy on Day 12. Progesterone (10 mg) was administered at 0700 and 1900 hr on Day 12, and then either progesterone or its vehicle was administered on Days 13 and 14. Silastic implants, either empty or containing estradiol, was administered at ovariectomy. Oxytocin and PGFM were measured in jugular blood samples withdrawn from an indwelling catheter at 5-min intervals for 8 hr on Day 15. Statistically significant pulses of oxytocin, presumably of posterior pituitary origin, were detected in all ewes. Approximately one-half of the oxytocin pulses preceded a pulse in PGFM concentrations by 10 min or less. These pulses tended (P = 0.09) to have a longer duration than those not linked to pulses of PGFM. The number of PGFM pulses that followed or did not follow an oxytocin pulse by 10 min or less was similar (P > 0.2). The amplitude and duration of oxytocin-linked PGFM pulses were greater (P = 0.05) than non-linked pulses. Although several explanations for the lower than anticipated temporal relationship between oxytocin and PGFM pulses are possible, the finding that oxytocin-related PGFM pulses are distinguishable from other pulses is consistent with the concept that oxytocin initiates robust pulses in PGF2α secretion. 相似文献

Retrobulbar disease, while rare in horses, can pose a clinical challenge due to the advanced diagnostic procedures necessary to determine the underlying aetiology and extent of disease. The goal of this study was to explore definitive causes of exophthalmos within a hospital population and investigate prognoses for the various aetiologies. Retrospective medical record review was used to identify 15 horses diagnosed with a definitive cause of exophthalmos. Patient signalment, disease duration, eye affected, diagnostic procedures, therapies, and histopathological diagnosis were evaluated. Geldings (12/15, or 80%) were over-represented (P = 0.014). Mean age at presentation (14 ± 6.7 years) was not significantly different for horses with (14 ± 8.5 years) and without (15 ± 4.4 years) a diagnosis of neoplasia. Diagnosis of a neoplastic process was made in 8/15 horses (53.3%) with neuroendocrine tumour being most common. Eight of the 15 cases were classified as primary orbital disease, and seven represented extension from paranasal sinus disease. Two novel diagnoses were identified: cystic hamartoma arising from the lacrimal gland and orbital aspergillosis secondary to sinonasal disease. In total, eight of 12 horses for which follow-up was available were euthanised due to orbital disease, two immediately following computed tomography of the skull and six due to disease progression. One horse was euthanised following a peri-anaesthetic complication. Three horses were lost to follow-up. Of the 12 horses for which follow-up was available, only four (33.3%) were alive 21 months post-diagnosis. Considering only patients for whom a definitive diagnosis was obtained, retrobulbar disease was associated with a poor prognosis for life. 相似文献

本试验旨在研究干扰素刺激基因15(interferon-stimulated gene 15,ISG15)敲除对猪伪狂犬病病毒(PRV)复制的影响。通过CRISPR/Cas9技术构建猪ISG15基因敲除猪肾上皮(PK-15)细胞系,利用CCK-8试剂盒检测PK-15敲除ISG15基因对细胞活力的影响,采用间接免疫荧光技术检测PK-15以及PK15-ISG15-/-细胞感染PRV的增殖差异,通过RT-qPCR检测PRV-EP0、PRV-gE、PRV-VP16和IFN-β的转录水平,Western blot检测PRV-gE和ISG15的蛋白表达水平,以及通过病毒噬斑检测对子代病毒感染力的影响。结果表明,sgRNA1和sgRNA2均成功敲除ISG15基因;CCK-8试剂盒检测细胞活力结果表明,敲除ISG15基因对PK-15细胞活力无影响;间接免疫荧光检测结果表明,PRV感染后,PK15-ISG15-/-细胞中的荧光强度明显高于PK-15细胞;RT-qPCR和Western blot结果表明,敲除ISG15可以促进PRV的转录和蛋白表达;病毒噬斑试验进一步显示,敲除ISG15可以促进PRV的复制。另外,RT-qPCR结果显示,敲除ISG15可以抑制PRV感染引起的IFN-β转录上调。本研究成功构建了PK15-ISG15-/-细胞系,并通过PRV感染试验证实ISG15基因可以抑制PRV在PK-15细胞中的增殖,并推测这种抑制作用可能与IFN通路有关。 相似文献

The aim of this study was to compare the plasma profiles of 15-ketodihydro-PGF(2α) (PGM) in healthy neonates of three different species from birth until the third week of life. Twenty-four horse foals, 12 donkey foals, and 9 calves were studied. Blood samples were collected at 10, 20 and 30 min after birth, at 3, 24 and 72 h after birth, and at 7, 10, 17 and 21 days of life. All mothers experienced normal gestation lengths and normal, spontaneous deliveries. All newborns were judged mature and viable. Hormone concentrations were higher (p < 0.05) in horse foals 20 and 30 min after birth compared to later samples, and at 10 min in donkey foals compared to later samples (p < 0.05). In calves, higher (p < 0.05) concentrations of PGM were observed 10, 20, 30 min and 3 hours from parturition compared to later samples. These findings may be related to increased fetal placental unit production during parturition, while the relatively high PGF(2α) levels in the days after parturition may be connected with their role in completing organ maturation. Despite the existing differences between these species, the statistical analysis did not discover significant differences in PGM profiles during the first 3 weeks of life in donkey, horse and cattle newborns. The low levels observed 10 days after birth are possibly due to a fast completion of maturational development in these species. 相似文献
