Characterization of a New Barley Mild Mosaic Virus Pathotype in France   总被引:2,自引:1,他引:1  
In March 2002 in a French field, severe mosaic symptoms appeared on plants of the barley cultivar Tokyo with the rym5 locus controlling resistance to all European strains of barley yellow mosaic virus (BaYMV) and barley mild mosaic virus (BaMMV). Electron microscopic examination revealed that the disease symptoms were associated with the presence of flexuous particles which resemble bymoviruses. From these observations and after enzyme-linked immunosorbent assay analysis it was first determined that the plants could be infected by BaMMV and BaYMV. Mechanical transmission of these viruses to the barley cultivar Magie susceptible to both viruses was only possible for BaMMV. This new pathotype (BaMMV-Sil) from Sillery (Marne Department, 51, France), in contrast to another mechanically transmitted French BaMMV isolate (BaMMV-MF), could be transmitted mechanically to two barley cultivars (Tokyo, Misato Golden), Arachis hypogaea, Datura stramonium and Lactuca sativa. BaMMV-Sil was indistinguishable from three BaMMV isolates from Germany (G), Japan (Ka1) and France (PF) by monoclonal antibodies in ELISA while the Japanese isolate (Na1) and BaMMV-MF were distinguishable from all. The sequence of the 3-terminal region of BaMMV-Sil RNA1 was determined. Comparison with previously published sequence data of capsid proteins indicated that BaMMV-Sil was closely related to BaMMV-Ka1, BaMMV-G and another German isolate (BaMMV-ASL1). Resistance-breaking BaMMV strains able to infect cultivars carrying the rym5 locus have also been described in Japan (BaMMV-Na1) and Korea (BaMMV-Kor). No specific amino acid differences were detected between the capsid proteins of BaMMV-Sil, BaMMV-Na1, BaMMV-Kor and those BaMMV isolates that do not overcome the rym5 resistance gene. These results indicate that BaMMV-Sil is a new pathotype of BaMMV in France and suggests that the capsid protein is not the determining factor of the pathogenicity towards the resistance gene rym5.  相似文献

Improved diagnosis of barley yellow mosaic (BaYMV) and barley mild mosaic (BaMMV) viruses was obtained by adjusting the buffers used in immunospecific electron microscopy (ISEM) to ensure a pH ≥ 7-0 and in ELISA by replacing ovalbumin with 10 g/l full cream milk powder.
Over 70% of samples of winter barley with symptoms of mosaic received from different sites in the UK during 1987-90 had BaYMV and 37% had BaMMV, with 11% containing both viruses. BaMMV was much more common on malting cultivars than on those grown for feed and this resulted in an easterly bias to the geographical distribution of the virus. Both viruses were, however, widely distributed in areas where winter barley is grown intensively. A small number of BaYMV records were from cultivars previously regarded as resistant and these are probably a distinct strain of the virus.  相似文献

 采用抗原直接包被和双抗体夹心酶联免疫吸附测定法(ELISA)对采自云南、福建、湖南烟区烟草花叶病样品进行了病毒种类检测,利用三抗体夹心ELISA对黄瓜花叶病毒(Cucumber mosaic virus,CMV)的亚组类型进行了鉴定。在云南采集的520个花叶病样品中,烟草花叶病毒(Tobacco mosaic virus,TMV)、CMV和马铃薯Y病毒(Potato virus Y,PVY)总检出率分别为71.74%、55.01%和6.35%;在福建采集的150个花叶病样品中,TMV、CMV和PVY的总检出率分别为94%、24.66%和8.00%;在湖南采集的74个花叶病样品中,TMV、CMV和PVY的总检出率分别为58.11%、51.35%和2.70%。部分样品为2种以上病毒复合侵染。云南、福建和湖南采集的64个CMV阳性样品中,属亚组Ⅰ的样品为57个,占89.1%;属亚组Ⅱ的样品为10个,占15.6%;其中3个样品为亚组Ⅰ和亚组Ⅱ的复合侵染。  相似文献

The dynamics of wheat spindle streak mosaic bymovirus in winter wheat were studied during two crop cycles in a field site with a history of high virus incidence. Individual plants of two susceptible cultivars were sampled from autumn to spring and the presence of virus antigen in roots and leaves was determined by ELISA. Virus incidence was higher in cv. Frankenmuth than in cv. Augusta. During year one, incidence of viral antigen in roots remained very low for four months after sowing, and did not reach maximum levels until the following spring. During year two, incidence of viral antigen in roots rose to maximum levels in autumn, only three months after sowing. These results strongly suggested that root infection occurred in spring as well as in autumn. In both cultivars and in both years, we detected the virus in roots one month prior to its detection in leaves, suggesting that virus moves slowly from roots into leaves. Maximum incidence of virus in leaves occurred in spring of both years, coinciding with the period of symptom development. Typical symptoms (yellow streaks, spindles, and mosaic) were observed in year two, whereas only mild mosaic was observed in year one. Virus antigen was detected in nonsymptomatic leaves from two months after sowing through crop senescence. Because antigen could be detected in roots throughout the crop cycle, and zoosporangia and cystosori of the fungal vector could be detected one and two months, respectively, after sowing, it is possible that wheat spindle streak mosaic bymovirus is acquired and/or spread by the vector during the majority of the crop cycle.  相似文献

In April 2001, stunted barley plants bearing mosaic symptoms were observed in a field in France (Marne Department, 51). Rod-shaped and flexuous particles were visualized by electron microscopy and positive serological reactions were detected by ELISA with Barley yellow mosaic virus (BaYMV) and Soil-borne cereal mosaic virus (SBCMV) polyclonal antisera. The tubular virus which was soil transmissible to barley cv. Esterel was separated from BaYMV by serial mechanical inoculations to barley cv. Esterel. This furo-like virus, in contrast to a French isolate of SBCMV, could be transmitted to Hordeum vulgare, Avena sativa, Beta vulgaris and Datura stramonium. RT-PCR was used to amplify the 3′-terminal 1500 nucleotides of RNA1 and the almost complete sequence of RNA2. Nucleotide and amino acid sequence analyses revealed that the French virus infecting barley is closely related to a Japanese isolate of Soil-borne wheat mosaic virus (SBWMV-JT) which was originally isolated from barley. This French isolate was named SBWMV-Mar. The 3′ UTRs of both RNAs can be folded into tRNA-like structures which are preceded by a predicted upstream pseudoknot domain with seven and four pseudoknots for RNA1 and RNA2, respectively. The four pseudoknots strongly conserved in RNAs 1 and 2 of SBWMV-Mar show strong similarities to those described earlier in SBWMV RNA2 and were also found in the 3′ UTR of Oat golden stripe virus RNAs 1 and 2 and Chinese wheat mosaic virus RNA2. Sequence analyses revealed that the RNAs 2 of SBWMV-Mar and -JT are likely to be the product of a recombination event between the 3′ UTRs of the RNAs 2 of SBWMV and SBCMV. This is the first report of the occurrence of an isolate closely related to SBWMV-JT outside of Japan.  相似文献

Surveys were conducted in 2016 and 2017 across the main cassava-growing regions of Burkina Faso to assess the status of cassava mosaic disease (CMD) and to determine the virus strains causing the disease, using field observation and phylogenetic analysis. CMD incidence varied between regions and across years but was lowest in Hauts-Bassins (6.0%, 2016 and 5.4%, 2017) and highest in Centre-Sud (18.5%, 2016) and in Boucle du Mouhoun (51.7%, 2017). The lowest CMD severity was found in Est region (2.0) for both years and the highest in Sud-Ouest region (3.3, 2016) and Centre-Sud region (2.8, 2017). The CMD infection was primarily associated with contaminated cuttings in all regions except in Hauts-Bassins, where whitefly-borne infection was higher than cuttings-borne infection in 2016. PCR screening of 687 samples coupled with sequence analysis revealed the presence of African cassava mosaic-like (ACMV-like) viruses and East African cassava mosaic-like (EACMV-like) viruses as single infections at 79.5% and 1.1%, respectively. Co-infections of ACMV-like and EACMV-like viruses were detected in 19.4% of the tested samples. In addition, 86.7% of the samples positive for EACMV-like virus were found to be positive for East African cassava mosaic Cameroon virus (EACMCMV). Phylogenetic analysis revealed the segregation of cassava mosaic geminiviruses (CMGs) from Burkina Faso into three clades specific to ACMV, African cassava mosaic Burkina Faso virus (ACMBFV), and EACMCMV, confirming the presence of these viruses. The results of this study show that EACMCMV occurrence may be more prevalent in Burkina Faso than previously thought.  相似文献

During the last ten years yellow mosaic of barley has been spreading more and more in European countries and in some infested areas yield loss can only be prevented by growing resistant cultivars. Breeding of such cultivars is one of the most important goals of small-grain breeders. To select resistant cultivars, several methods are available such as mechanical inoculation, growing of plants in infested soil in the field or in growth chambers as well as using root powder of infected plants. The situation has recently been complicated by the discovery that yellow mosaic is caused by at least two distinct but closely related viruses (barley yellow mosaic virus and barley mild mosaic virus). Inoculation methods are not equally effective for these two viruses. Although it appeared initially that resistance to the two viruses was linked, there are now indications that some barley cultivars respond differently, and there may even be a third virus involved. Programmes for selection of resistant cultivars now have to take account of this.  相似文献

Soilborne wheat mosaic disease (SBWMD), originally attributed to infections by Soilborne wheat mosaic virus (SBWMV) and Wheat spindle streak mosaic virus (WSSMV), is one of the most frequent virus diseases and causes economic losses in wheat in southern Brazil. This study aimed to characterize molecularly the viral species associated with wheat plants showing mosaic symptoms in Brazil. Wheat leaves and stems displaying mosaic symptoms were collected from different wheat cultivars in Passo Fundo municipality, Rio Grande do Sul State, southern Brazil. Double-stranded RNA was extracted and submitted to cDNA library synthesis and next-generation sequencing. No sequences of SBWMV and WSSMV were detected but the complete genome sequence of a putative new member of the family Benyviridae was determined, for which the name wheat stripe mosaic virus (WhSMV) is proposed. WhSMV has a bipartite genome with RNA 1 and RNA 2 organization similar to that of viruses belonging to Benyviridae. WhSMV RNA 1 has a single open reading frame (ORF) encoding a polyprotein with putative viral replicase function. WhSMV RNA 2 has six ORFs encoding the coat protein, the major protein (read-through), triple gene block movement proteins (TGB 1, 2 and 3) and ORF 6 (hypothetical protein). In addition to the genomic organization and nucleotide and amino acid sequence identities, phylogenetic analyses also corroborated that WhSMV is a virus species of the Benyviridae. However, isolates of WhSMV formed a clade distinct from members of the genus Benyvirus. It was also demonstrated that the plasmodiophorid Polymyxa graminis is associated with wheat roots showing SBWMD symptoms and infected by WhSMV.  相似文献

Infection by barley yellow mosaic virus (BaYMV) was greater in a crop sown in October than in November and in cv. Maris Otter than in cv. Sonja. However, a greater proportion of plants of Sonja than of Maris Otter were infected, and more severely, with the soil-borne fungus vector of BaYMV. Polymyxagraminis. Indifferent years the yield of individual plants of Maris Otter was decreased by 15% and of Sonja by 40%. The spring barley cultivars Athos. Porthos. Georgie and Ark Royal were all infected with BaYMV when sown in autumn in infected soil. Infected winter barley cv. Igri lost up to 50% of yield when this was measured across a naturally infected patch of crop.  相似文献

辣椒是我国重要的蔬菜和经济作物,受多种病毒危害。2014年在北京市顺义区调查时发现部分种植的辣椒植株上叶片大面积黄化,边缘症状明显,个别植株叶片轻微上卷。提取典型症状样品的总RNA,反转录得到cDNA,分别用黄瓜花叶病毒(Cucumber mosaic virus,CMV)特异引物和马铃薯卷叶病毒属(Polerovirus)通用引物进行PCR检测,CMV特异引物和马铃薯卷叶病毒属通用引物分别扩增得到约650bp和1 400bp的特异条带。测序和核苷酸序列比对表明,其分别与CMV和甜菜西方黄化病毒(Beet western yellows virus,BWYV)序列同源性最高为99%和96%。这是对我国种植的辣椒上发生的CMV和BWYV复合侵染的首次报道。  相似文献

Two viruses that frequently occur in many Lilium species are Lily mottle virus (LMoV) and Cucumber mosaic virus (CMV), which usually co-infect lilies causing severe disease symptoms. Recent reports have revealed that the viral coat protein (CP) affects chloroplast ultrastructure and symptom development. This study used western blot analysis to confirm that in leaves infected by mixed virus infections of LMoV and CMV, CPs of both viruses were accumulated in lily chloroplasts. Immunogold labelling further demonstrated that both the LMoV CP and CMV CP were localized in the stroma and the thylakoid membranes of the chloroplasts. In addition, it was found that CPs of both viruses were rapidly transported into isolated, intact chloroplasts (in vitro), and their transport efficiencies were positively related to CP concentrations. The lowest transmembrane concentration of CMV CP decreased from 38 μg mL−1 recorded in the single CMV CP import system to 10 μg mL−1 in the mixed import system of LMoV CP and CMV CP. CPs of both viruses exhibited species selection in their transmembrane transport into chloroplasts. This is the first report that the CPs from two viruses (LMoV and CMV) are simultaneously present in lily chloroplasts. Accumulation of high levels of LMoV CP and CMV CP inside the chloroplast appears to contribute to a synergistic interaction inducing the development of mosaic symptoms.  相似文献

 2010-2011年采用大田人工接种鉴定的方法,对生产中大面积推广使用的24份烟草品种进行了烟草花叶病毒(TMV)和黄瓜花叶病毒(CMV)的抗性鉴定。结果表明,供试品种对TMV和CMV的抗病性存在较大差异,对TMV表现抗病的有Coker86、吉烟5号、Coker176、CV87、辽烟8号、CV91、中烟90等7份材料;表现中抗的有秦烟98、双抗70、C151等3份材料;表现中感的有秦烟96、G80、金星6007、龙江981、K326、秦烟201、NC89等7份材料;表现感病的有G28、云烟97、净叶黄、红花大金元、RG11、云烟85、云烟87等7份材料。对CMV表现抗病的有Coker86、龙江981、C151、秦烟201、云烟87等5份材料;表现中抗的材料是金星6007;表现中感的有CV91、RG11、Coker176、中烟90、K326、红花大金元、净叶黄、G80、G28等9份材料;表现感病的有秦烟98、云烟85、秦烟96、NC89、双抗70、云烟97、CV87、辽烟8号、吉烟5号等9份材料。其中兼抗TMV和CMV两种病毒病的材料有2份,分别是Coker86和C151。同时研究还发现,抗病性不同的烟草品种在受到病毒危害以后,对烟叶的产量和品质的影响也不同。明确了中国24个烟草品种的抗病性水平,为抗病品种的利用与品种合理布局提供科学依据,同时为烟草抗病毒病育种的亲本选择提供抗源信息。  相似文献

The concentration of African cassava mosaic virus (ACMV) was assessed by enzyme-linked immunosorbent assay in relation to symptom severity among resistant, moderately resistant and susceptible cassava genotypes. Resistant genotype NR 8083 had significantly lower symptom severity scores ( P  < 0·05) than the susceptible genotype TMS 91934, but the two genotypes contained similar levels of virus concentration. The moderately resistant genotypes TMS 30572 and NR 8082 expressed significantly lower symptom severities ( P  < 0·05) than the susceptible genotypes TMS 91934 and TME 117, but they contained significantly higher virus concentrations ( P  < 0·05) than TMS 91934 and similar virus concentration as in TME 117. However, two other resistant genotypes, TME 1 and TME 8, had low symptom severity scores and virus concentrations. There was significant interaction ( P  ≤ 0·05) between cropping season and virus concentration in all the genotypes except TMS 30572. The resistant and moderately resistant genotypes that had high virus concentrations sustained storage root yield losses. The severity of symptoms expressed was not necessarily a reflection of the virus concentration in some of the genotypes. In addition to the use of symptom severity scores to group genotypes into resistant classes, it is recommended that virus concentration should also be considered. Genotypes displaying mild symptoms, but with high levels of virus accumulation, could be an important source of inoculum in the spread of ACMV by the whitefly vectors. This suggests that each genotype should be tested for virus accumulation prior to its release to the farmers.  相似文献

The reaction of thirty-four barley cultivars from European and Asiatic origin was analysed in six soils infected with barley yellow mosaic virus complex (BaYMV, BaMMV). These soils were selected from 16 sites for their differences in cultivar response. Amongst the six cultivars carrying the ym4 gene (Esterel, Express, Labéa, Majestic, Réjane, Vanoise), only cv Majestic was infected at one site with BaYMV and BaMMV. Concerning BaYMV, three cultivars were infected on all the soils and 19 on none of them. Twelve cultivars were differentially infected depending on the soil. In the case of BaMMV, four cultivars were infected on all the soils and 19 on none of them. Eleven cultivars were differentially infected depending on the soil. ELISA tests revealed the presence, in these soils, of variants of BaYMV and BaMMV that were able to overcome at least seven of the 12 known resistance genes (ym3, ym4, ym6, ym8, ym9, ym10, ym11) and the resistance of three varieties (Tosan Kawa 73, OU1 and Taihoku A) in which the genetic basis is unknown. Amplification by RT-PCR of the N-terminal region for three of BaYMV variants was performed. Nucleotide and amino acid sequences were determined and compared with the corresponding sequence of a common strain of BaYMV-G. A few nucleotide differences were detected between all the French isolates, but there were no strain specific amino acid differences.  相似文献

 研究结果表明:陕西玉米病毒病毒原主要有玉米矮花叶病毒B株系(MDMV-B)、玉米粗缩病毒(MRDV)和大麦黄矮病毒(BYDV)。用血清鉴定225份病株样品中,MDMV-B、MRDV和BYDV侵染分别占40%、23%、4.5%。MDMV-B和MRDV复合侵染占23.5%。在田间自然条件下,MDMV-B主要通过玉米蚜和禾谷缢管蚜以非持久性传播,MRDV则由灰飞虱以持久传播。MDMV-B、MRDV和BYDV的粒子大小分别为735~755 nm×17 nm、70~75 nm和23~30 nm。随着玉米生育成熟,侵染逐渐降低,为害亦趋减轻。研究认为,玉米品种、播种期、播量、田间传毒介体数量及发生早晚和地膜覆盖等是影响玉米病毒病流行的主要因素。  相似文献

Two potyvirus isolates, one from germplasm of yard-long bean (Vigna unguiculata ssp.sesquipedalis) introduced into the Netherlands, and another one from soybean plants (Glycine max) in Indonesia, were compared with two virus isolates of blackeye cowpea mosaic virus (BICMV) from the USA and a Moroccan isolate of cowpea aphid-borne mosaic virus (CAMV). It is proposed that all five isolates be now considered BICMV on the basis of host ranges, symptoms and serology. From our results, and a reassessment of the literature it is suggested to drop the name CAMV in favour of BICMV.Samenvatting Twee potyvirussen, de een in Nederland ingevoerd met genenmateriaal vanVigna unguiculata ssp.sesquipedalis en de ander uit planten van sojaboon (Glycine max) in Indonesië, werden vergeleken met twee isolaten van blackeye cowpea mosiac virus (BICMV) en een Marokkaans isolaat van cowpea aphid-borne mosaic virus (CAMV). Op grond van waardplantenreeksen, symptomen en serologie stellen de auteurs voor om alle vijf isolaten te beschouwen als BICMV. Gebaseerd op de verkregen resultaten en een kritische beschouwing van de literatuur wordt de aanbeveling gedaan om de naam CAMV te laten vallen ten gunste van BICMV.  相似文献
