With the bioguided fractionation of the ethanol extracts from the leaves of Momordica charantia, we obtained two most active compounds against the feeding of the diamondback moth, Plutella xylostella larvae. The antifeedant activity of momordicine Ⅰ and momordicine Ⅱ against the second and the third instar larvae of Plutella xylostella were tested using leaf discs of cabbage in the laboratory. The results showed that momordicin Ⅰ and momordiein Ⅱ had significant antifeedant activity on the larvae of P. Xylostella, and momordicin Ⅱ was more active than momordicin Ⅰ. The concentrations for 50% antifeedant effects (AFC50) of momordicin Ⅱ against the second and the third instar larvae of P. Xylostella were 76.69 and 116.24 μg mL-1, whereas that of momordicin Ⅰ was 144.08 and 168.42 μg mL-1, respectively. In addition, momordicin Ⅰ and momordicin Ⅱ had significant inhibitive effect on the rate of weight gain and survival of P. Xylostella larvae.  相似文献

Active oxygen quantum yield, insecticidal activities and stability of diphenylthiophene were studied for the first time. The results showed that maximum diphenylthiophene absorbency variety was 0.438 after irradiation for 280 min and that of α-terthienyl (α-T) was 0.480 after irradiation for 200 min with UV. LC50 values ofdiphenylthiophene and α-T against 3rd instar larvae of Aedes albopictus were 9.18 × 10-3 and 9.69 × 10-4 μg mL-1 when treated for 24 h, respectively. LC50 values of the two chemicals against the 3rd instar larvae of Plutella xylostella were 267.87 and 222.22 μg mL-1 when treated for 24 h, respectively. The half lives of diphenylthiophene and α-T in methanol were 113.62 and 10.65 h. Difference between quantum yield of diphenylthiophene and α-T was not significant and they all possessed high toxicity to Aedes albopictus and Plutella xylostella, but diphenylthiophene was more stable than α-T. It could be concluded that diphenylthiophene has overcome the deficiency of photoactivated insecticides which degraded quickly in the environment and could not be applied on the field. Diphenylthiophene could kill the insects and accelerate the degradation rate of triazophos. The benefits of diphenylthiophene can be further exploited and applied on the field.  相似文献

【目的】研究光叶铁仔Myrsine stolonifera提取物对小菜蛾Plutella xylostella生长发育的影响。【方法】采用叶片载毒法,测定了光叶铁仔根、茎、叶甲醇提取物和大孔吸附树脂乙醇馏分对小菜蛾的生物活性;测定了大孔吸附树脂乙醇馏分对小菜蛾生长发育的影响。【结果】光叶铁仔根、茎、叶甲醇提取物和大孔吸附树脂乙醇馏分对小菜蛾具有胃毒作用,以叶部50%(φ)乙醇馏分的效果较佳,72 h后校正死亡率达到75.56%。根茎部95%(φ)乙醇馏分与叶部50%(φ)乙醇馏分,对小菜蛾的幼虫具有一定的拒食作用与体质量抑制效果,其拒食率与体质量抑制率整体高于90%,且对化蛹及羽化有一定的影响。【结论】光叶铁仔提取物对小菜蛾幼虫具有较强的杀虫活性,其活性成分及作用机理值得深入研究。  相似文献

【目的】探究天敌昆虫异色瓢虫Harmonia axyridis对草地贪夜蛾Spodoptera frugiperda的捕食潜力。【方法】在室内条件下评价异色瓢虫成虫和4龄幼虫对草地贪夜蛾1~2龄幼虫的捕食能力,观察并总结异色瓢虫的捕食行为特点。【结果】通过观察,发现异色瓢虫捕食草地贪夜蛾幼虫主要经过静止、搜寻、试探、取食和爬行5种行为,其中异色瓢虫成虫捕食3龄草地贪夜蛾幼虫时出现3对足抱握猎物的现象。异色瓢虫成虫和4龄幼虫对草地贪夜蛾1~2龄幼虫的捕食功能反应符合HollingⅡ模型。异色瓢虫成虫和4龄幼虫对草地贪夜蛾1龄幼虫的日最大捕食量分别为196.08和161.29头,瞬时攻击率分别为1.390 6和1.451 4,处理时间分别为0.005 1和0.006 2 d;异色瓢虫成虫和4龄幼虫对草地贪夜蛾2龄幼虫的日最大捕食量分别为65.36和41.84头,瞬时攻击率分别为1.145 7和1.271 0,处理时间分别为0.015 3和0.023 9 d,即异色瓢虫成虫对草地贪夜蛾幼虫的捕食能力强于4龄幼虫。【结论】异色瓢虫对草地贪夜蛾有一定的潜在控害能力,可用于防控草地贪夜蛾。  相似文献

The isolates of Phytophthora infestans on tomato in the Guangxi Zhuang Autonomous Region, China, were determined for the sensitivities to metalaxyl, cymoxanil and dimethomorph to give the basic information for integrating disease management. Sensitivities were tested by measuring the radial growth on agar medium amended with fungicide, compared with the floating-leaf-disk method. 239 isolates were collected from eight tomato growing areas during 2000-2006. The testing results indicated that the frequencies of sensitive, intermediate, and resistant isolates to metalaxyl were 42.26, 35.98, and 23.53%, respectively. Variations in sensitivities amongst isolates from different areas or different years were very high for metalaxyl. All isolates from Tianlin and Wuxuan were sensitive to metalaxyl, but the metalaxyl-resistant isolates predominated in Tianyang, with the frequency of 51.35%. The EC50 values of certain isolates from Tianyang were higher than 500 μg·mL^-1 and their resistance levels were over 100 000 folds. Cymoxanil has been used for nearly 10 years in Guangxi, and dimethomorph has been used for 5-6 years. However, there was no decrease in sensitivity of P. infestans populations and the sensitivities of the pathogen were nearly normally distributed. Hence, their mean ECs0 value [cymoxanil (0.1647 ±0.0255) μg·mL^-1, dimethomorph (0.0970 ± 0.0052) μg·mL^-1 could be used as the baseline sensitivities for monitoring the field resistance development. The comparison with the floating-leaf-disk method indicates that both the techniques provided equivalent results. These studies suggested that metalaxyl can be continuously applied in Tianlin, Wuxuan, and Nanning due to the resistant isolates that have not been found, while for those areas with resistant isolate, the use of metalaxyl should be reduced or alternated, and cymoxanil or dimethomorph was recommended for controlling late blight disease of tomato.  相似文献

小菜蛾(Plutella xylostella)是危害十字花科植物的重要害虫之一,防治方法主要是应用化学农药防治,使得农药残留、抗药性及再猖獗问题日益严重。为明确不同绿色防治技术对小菜蛾的防治效果,采用田间试验方法,在甘蓝田进行不同类型的诱捕器和诱芯比较、7种生物农药筛选及释放螟黄赤眼蜂防治小菜蛾的3种绿色防控技术试验。结果表明:不同诱捕器和诱芯诱蛾效果差异显著,天然橡胶塞诱芯诱蛾效果明显优于毛细管型诱芯,船型诱捕器配绿色天然橡胶塞型诱芯或红色塑料毛细管型诱芯为最佳组合。生物药剂中乙基多杀菌素防治小菜蛾的速效性最好,药后1 d防效达到78.93%;小菜蛾颗粒体病毒在药后7 d防效达到最高,达到86.68%,但持效性较短。田间释放螟黄赤眼蜂对小菜蛾卵寄生率均达到65%以上,对小菜蛾卵的寄生效果良好。这3种技术对小菜蛾的绿色防控具有一定的指导和参考价值,可在田间示范和推广。  相似文献

In this study, the oxidative damage of α-terthienyl (α-T) to the Spodoptera litura (SL) cell and its mechanism were investigated. MTT was used to compare the toxicity of α-T and rotenone to the SL cell. The output of malondialdehyde and relative content of glutathione were determined with 2-thiobarbituric acid (TBA) and 5, 5'-dithio-bis (2-nitrobenzoic acid) (DTNB), respectively. Transmission electron microscope (TEM) was employed to observe the influence of α-T on the membrane and organelle of the SL cell. The result showed that the IC50 value of α-T to the SL cell was 0.21 μg mL^-1, whereas the corresponding dose of rotenone was 12.25 μg mL^-1. The output of MDA had the same changing tendency with the concentration of α-T, whereas the content of GSH had the negative correlation with it. According to TEM, the cell membrane and karyotheca swelled and couldn't retain integrity, the intracellular substances leaked out, unidentified granules appeared in the SL cell. The mitochondria expanded, and the membrane and subcellular organelle were damaged severely. In this study, it was found that after oxidative damage induced by α-T, the output of MDA increased notably, whereas the relative content of GSH decreased. This indicated that the antioxidant ability of cell weakened. The result of TEM implied that the SL cell suffered from oxidative damage under the appointed dose.  相似文献

通过室内外系统试验探讨了体积分数1%苦皮藤乳油对小菜蛾(Plutella xylostella L.)种群的控制作用,并采用生命表技术进行评价。结果表明,其控制作用主要表现在对成虫产卵的忌避作用、对幼虫的拒食作用以及对生长发育的影响和减少幼虫危害力等方面。体积分数1%苦皮藤乳油10.00mL/L处理对成虫的忌避率为73.2%,对幼虫的选择性和非选择性拒食率分别为94.7%和93.5%,幼虫总取食量是对照的40.1%,使小菜蛾种群趋势指数(I值)由对照的21.954降为0.6684,干扰作用控制指数(IIPC)为0.0304。采用状态空间分析法就苦皮藤乳油施用后小菜蛾种群数量动态的模拟分析亦表明,其显著的控制作用表现在种群发展初期对成虫产卵的忌避作用。  相似文献

从烟田烟青虫(Helicoverpa assulta)自然感病虫体上新分离鉴定一株球孢白僵菌Bb062,将其与已有的不同来源地和寄主的10株白僵菌(Beauveria spp.)一起,分别测定其对烟青虫3龄幼虫致病力以及烟青虫3龄幼虫感染高毒力白僵菌后体内保护酶活性的变化。结果表明,在供试的11个白僵菌菌株中,Bb062对烟青虫3龄幼虫毒力最高,10 d累计校正死亡率达91.07%,LT50(致死中时)为4.67 d,且与其他菌株差异显著。以孢子1.0×106、1.0×107和1.0×108个·m L-1悬浮液处理时,烟青虫3龄幼虫累计死亡率随菌液浓度的提高和接菌后时间的延长而增大,LC50(致死中浓度)为孢子1.82×107个·m L-1。烟青虫3龄幼虫感染球孢白僵菌后72 h,其体内超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性均表现出先急剧升高,再急剧下降的变化过程。在所测试的菌株中,Bb062菌株对烟青虫毒力最高,且能够显著抑制其体内保护酶活性,因此,该菌株具有作为烟青虫生防菌的应用潜力。  相似文献

为明确芹菜茎叶提取物对菜粉蝶拒食作用和产卵行为的影响,采用水蒸气蒸馏法从芹菜中提取其挥发性次生物质,以菜粉蝶成幼虫为供试昆虫,测定芹菜提取物对幼虫拒食作用和对成虫产卵行为的影响。结果表明,芹菜茎叶提取物对幼虫具有显著的拒食作用,对成虫具有一定的产卵忌避作用。当提取物质量浓度为1 000g/L时,处理24、48和72h对幼虫的选择性拒食率分别为62.5%、77.7%和84.3%,非选择性拒食率分别为65.4%、76.5%和82.5%,对成虫的选择性忌避率分别为86.8%、75.2%和58.5%,非选择性忌避率分别为72.6%、64.4%和43.4%。  相似文献

Insecticidal activities of the petroleum ether-, chloroform-, ethyl acetate-, and water-soluble fractions of the methanolic extract of Ficus sarmentosa var. henryi were assayed against Musca domestica adults. The chloroform- and ethyl acetate-soluble fractions were the most active with 92.6 and 88.9% mortalities (24 h after treatment) respectively. Therefore, the two fractions were combined and four compounds, isolated from the fractions by activity-guided fractionation, were elucidated as 7-hydroxycoumarin, apigenin, eriodictyol, and quercetin by spectroscopic method and displayed excellent insecticidal activities against adults of M. domestica and 4th instar larva of Aedes albopictus. Among those, 7-hydroxycoumarin showed the strongest insecticidal activities with lethal concentrations (LC50) values of 72.13 μg g−1 sugar and 4.87 μg mL−1 (48 h after treatment) against the test insects respectively. The cytoxicities of these compounds on BTI-Tn-5Bl-4 cell were also investigated for the insecticidal mechanism and found that quercetin represented superior inhibitory activity with MTT assay and reactive oxygen species (ROS) against BTI-Tn-5Bl-4 cell, but slightly weaker than that of the positive control (azadirachtin) and significantly greater than the negative control (DMSO only). Meanwhile, eriodictyol demonstrated the strongest effect on the mitochondrial membrane potentials (MMP). In conclusion, based on their comparative toxicities to commercial insecticides and their cytotoxic effects, some of the compounds from the F. sarmentosa have potential as botanical insecticides.  相似文献

【目的】评价印楝素对草地贪夜蛾Spodoptera frugiperda的毒力与防效,为草地贪夜蛾的防治提供技术参考。【方法】采用浸叶法与喷雾法测定了印楝素对草地贪夜蛾的室内毒力和田间防治效果。【结果】在室内条件下,0.3%(φ)印楝素乳油处理草地贪夜蛾7 d后,2龄和3龄幼虫的LC50值分别为0.59和0.46 mg/L;40%(w)印楝素干粉处理草地贪夜蛾7 d后,2龄和3龄幼虫的LC_(50)值分别为0.93和0.79 mg/L。0.3%印楝素乳油处理草地贪夜蛾3 d后,2龄和3龄幼虫的AFC50值分别为0.30和0.12 mg/L;40%印楝素干粉处理草地贪夜蛾3 d后,2龄和3龄幼虫的AFC50值分别为0.53和0.30 mg/L。0.3%印楝素乳油稀释500倍喷雾处理后1、3和7 d对草地贪夜蛾的防效分别达到了24.83%、50.34%和75.50%。【结论】印楝素对草地贪夜蛾具有较好的毒杀活性与拒食活性,7 d后的防治效果较好;印楝素对草地贪夜蛾的防治具有广阔的应用前景。  相似文献

The paper was to find the bacteria to degrade aflatoxin B 1 (AFB 1) and realize the application of biological degradation on AFB 1. Using cumarin as the carbon source and energy on the first screening, then the ten strains which were first screened out were taken to degrade AFB 1 100 pg kg^-1. Strain NMO-3 was screened out of ten strains, the degradation ratio of AFB 1 reached 85.7%, which was more prominent than the others (P 〈 0.01). With the analysis of colony morphology, physiological and biochemistry experiments, and 16S rDNA gene sequence, the strain NMO-3 was finally identified as Stenotrophomonas sp. Using cumarin as the carbon source and energy could screen out the AFB 1 degradation strains. Acute toxicity tests show that the viable number of NMO-3 lower than 3.12 × 10^10 cfu mL-1 is safety. The crude enzyme was obtained by 65% ammonium sulfate fractionation, and it could degrade AFB1. It is the first report for the strain's detoxi- AFB1.  相似文献

【目的】黄野螟Heortia vitessoides是珍贵树种土沉香Aquilaria sinensis的重要食叶害虫,通过大面积林间调查土沉香受害情况,筛选可能存在的抗虫植株,为黄野螟的科学预防和土沉香抗虫品种的选育奠定基础。【方法】在黄野螟危害盛期,对野外土沉香林定期进行大面积调查,在严重受害的土沉香林中,观察不同受害水平土沉香的外观形态和叶片物理结构,同时采集具有不同抗虫性植株的叶片饲养黄野螟幼虫,观察初孵幼虫对不同抗性植株叶片的选择和拒食情况,取食不同抗性土沉香叶片后,测定黄野螟幼虫存活率、生长发育、化蛹和羽化的差异。【结果】在土沉香严重受害的林分中发现了2株未受害的土沉香植株,其表现出较好的抗虫性(抗1和抗2)。抗性植株(抗1和抗2)与感虫植株在叶片长度和厚度上差异显著(P0.05),而叶片长宽比无显著差异。在叶片物理结构上,抗2土沉香叶片的上表皮角质层厚度显著高于感虫叶片。抗2土沉香植株对幼虫取食抑制率高于抗1土沉香植株,两者均达44.81%以上。强迫取食抗性土沉香试验的幼虫存活率、成虫羽化率、蛹质量、成虫寿命均显著低于取食感虫土沉香叶片幼虫的相应指标,而取食抗性土沉香的幼虫、蛹发育历期均显著长于取食感虫土沉香叶片的害虫。【结论】叶片嫩绿的土沉香植株较易受黄野螟的为害,而叶片厚的或叶片颜色偏黄、墨绿的土沉香植株对黄野螟具有较强的抗性。抗性土沉香植株对黄野螟幼虫取食活性具有较强的抑制作用,对幼虫的发育有阻碍作用。  相似文献

为明确蛇莓[Duchesnea indica(Andr)Focke]中杀蚊幼活性成分,以淡色库蚊(Culex pipiens pallens)和白纹伊蚊(Aedes albopictus)3龄幼虫为试虫,采用活性跟踪法,对蛇莓乙醇提取物进行活性成分分离、鉴定和活性测定。从蛇莓乙醇提取物乙酸乙酯相中得到2个具有杀蚊幼活性的化合物,鉴定为齐墩果酸和芹菜素。浸渍法测定表明,乙酸乙酯萃取相对淡色库蚊和白纹伊蚊3龄幼虫LC50值分别为459.12和438.09mg/L,正丁醇相为471.41和430.81mg/L,显示较好的杀蚊幼活性,且乙酸乙酯相和正丁醇相差异性不显著;齐墩果酸对淡色库蚊和白纹伊蚊3龄幼虫24h处理LC50值分别为25.36和22.31mg/L,芹菜素对淡色库蚊和白纹伊蚊3龄幼虫24h处理LC50值分别为31.29和27.22mg/L。  相似文献

29株虫生真菌的鉴定及对烟粉虱的毒力   总被引:1,自引:0,他引:1  
【目的】对29株虫生真菌菌株进行分类学鉴定并筛选出对烟粉虱Bemisia tabaci具有高毒力的菌株。【方法】采用形态学及ITS区、TEF区、Bloc区基因序列相似性分析鉴定菌株,并采用浸渍法测定真菌对烟粉虱的毒力。【结果】29株虫生真菌有26株符合白僵菌Beauveria的特征,菌株SB003、SP665和SP670被鉴定属于棒束孢属Isaria;利用TEF区序列、Bloc区序列、TEF-Bloc联合序列构建的系统发育树,结果显示29株菌株共包括24株球孢白僵菌B. bassiana、2株假性球孢白僵菌B. pseudobassiana和3株环链棒束孢I. cateinannulata。烟粉虱2龄若虫的死亡率随孢子浓度的增加而增加,不同菌株致病力不同;用1×108 mL-1的孢子悬浮液处理烟粉虱2龄若虫7 d,致死率最高的菌株为SP433,其次为SB009,分别为87.37%和82.93%。【结论】本研究可为虫生真菌的分类提供理论基础,筛选出的高致病力菌株SP433和SB009可为烟粉虱的生物防治提供参考。  相似文献

为了寻找可产生鬼臼毒素类化合物的内生真菌,从砂地柏(Sabina vulgarisAnt.)不同组织中分离到126株内生真菌。以鬼臼毒素,脱氧鬼臼毒素和苦鬼臼毒素为标准品,采用HPLC法筛选得到一株可代谢产生重要药用和杀虫活性物质鬼臼毒素类似物的内生真菌SC13。对菌株SC13进行ITS-5.8S rDNA区域扩增,测序及遗传进化关系分析,初步推测该菌株属于链格孢属(Alternariasp.)真菌。采用小叶碟添加法测定该菌株发酵液萃取物对三龄初期粘虫的拒食活性,其48h拒食中浓为7.259 mg.mL-1,采用浸毒叶片法测定了三龄初期小菜蛾的毒杀活性,其48h致死中浓为6.136 mg.mL-1。  相似文献

研究不同质量浓度紫茎泽兰叶水提液对蛞蝓的毒杀作用及毒杀机理。胃毒活性测定结果表明,当紫茎泽兰叶水提液质量浓度≥0.09 g·mL-1时,其水提液对蛞蝓具有较好的毒杀作用,但作用缓慢,4 d后才能表现出明显的毒杀作用。Na+,K+ ATPase酶及乙酰胆碱酯酶活力测定结果表明,紫茎泽兰叶水提液对蛞蝓Na+,K+ ATPase酶活力具有明显的抑制作用,对乙酰胆碱酯酶活力也有一定的影响。可溶性糖和可溶性蛋白含量测定结果表明,紫茎泽兰叶水提液可降低蛞蝓体内可溶性糖质量分数,提高可溶性蛋白质量分数。说明紫茎泽兰叶水提液对蛞蝓具有明显的毒杀作用,其毒杀机理主要为抑制Na+,K+ ATPase酶活力。  相似文献

This study was designed to control plant fertility by cell lethal gene Barnase expressing at specific developmental stage and in specific tissue of male organ under the control of Cre/lox system, for heterosis breeding of chili pepper (Capsicum annuum L.). Chili pepper inbred lines (A, D, E, and I) were transformed with Cre gene and Barnase gene situated between loxp, separately, by means of Agrobacterium co-culture. In this study, we had established a high transformation system by extensive study of affecting factors including genotype, selection of marker, and lethal dose. Cotyledon with petiole from 9-11-day-old seeding was pre-cultured on media MR[MB(MS mineral+vitamine B5)+BA(6-Benzyladenine) 5.0 mg·L^-1 +IAA(indoleacetic acid) 1.0 mg·L^-1+GA3(gibberellic acid) 1.0mg·L^-1+sucrose 3%+agar 6.5g·L^-1] for 2d. The explants were infected by Agrobacterium tumefaciens when their OD600(optical density at 600 nm)reached 0.6-0.9. After co-cultured for 4-5 d on media MC [MB+BA5.0 mg·L^-1+IAA 1.0 mg·L^-1 +GA3 1.0 mg·L^-1+sucrose 3% +agar 6.5 g·L^-1+AS (acetosyringone) 200μmol·L^-1, these cotyledons with petiole were cultured on selective differentiation medium in the media MT[MB medium supplemented with BA [5.0 mg·L^-1+ IAA 1.0 mg·L^-1+ GA3 1.0 mg·L^-1+ AgNO3 5.0 mg·L^-1+ CW (coconut water) 5% + Km (kanamycin) 65 mg·L^-1+ Cb (carbenicillin) 500 mg·L^-1+ 3% sucrose + agar 6.5 g·L^-1].The Kmr (kanamycin resistant) bud rosettes were elongated on selective elongation medium and rooted on rooting medium. PCR and Southern blotting analysis of Kmr plantlet indicated that the foreign genes had been integrated into the genome of pepper. The transgenic plants with Cre gene developed well, blossomed out, and set fruit normally. The transgenic plants with Barnase gene grew well with normal appearance of flower, but they showed different fertility from complete sterility, partial sterility to complete fertility, and similar results were obtained from in vitro pollen germination experiments.  相似文献

通过对不同寄主或不同来源的11种29株拟青霉进行抗白色念珠菌和清除自由基的活性筛选,发现古尼拟青霉菌株RCEF0866的菌丝体乙酸乙酯提取物具有较强的抗真菌活性,在菌丝浓度为100.0 mg.mL-1时,其提取物的抑菌圈达到(18.1±1.7)mm;菌株RCEF0866和RCEF4118的菌丝体甲醇提取物还具有较强的清除自由基活性,在菌丝浓度为5.0 mg.mL-1时,其对0.4 mg.mL-1的DPPH(1,1 diphenyl-2-picryhydrazyl)自由基的清除率分别为82.75%和79.53%。对菌株RCEF0866菌丝体的乙酸乙酯提取物进行薄层抑菌试验发现,其抗真菌主要成分的Rf值为0.90,表明该抗真菌成分为低极性成分。二苯代苦味肼基自由基薄层试验结果表明,RCEF0866菌丝体的甲醇提取物中具清除自由基活性的成分的Rf值为0.80,表明该清除自由基成分也为低极性成分。  相似文献
