Twelve gilts, with an average initial BW of 38 kg, were fitted with a simple T-cannula at the distal ileum and two catheters in the external jugular veins. The animals were fed 700 g of one of the experimental diets, twice daily at 0800 and 2000. Diets 1 and 2 were cornstarch-based diets formulated to contain 16% CP from soybean meal and canola meal, respectively. Diets 3 and 4 contained 93.9% wheat and barley, respectively. Chromic oxide was included to determine nutrient digestibilities. Following recovery from surgery, 15N-leucine was infused continuously via a jugular catheter for 9 d at a rate of 40 mg.kg BW-1.d-1. Blood samples were taken at every feeding. Ileal digesta were collected for 24 h on d 7 and 9 of the infusion period. Assuming that the 15N-enrichment excess in the trichloroacetic acid-soluble fraction of the blood was similar to that in endogenous N, the contribution of endogenous to total N in ileal digesta was calculated. The amount of endogenous protein (N x 6.25) recovered in digesta collected from the distal ileum were 25.5, 30.5, 27.4 and 27.7 g/kg DM intake for the pigs fed diets 1, 2, 3 and 4, respectively. In the same order for the diets, the real ileal protein digestibilities were 97.5, 84.1, 99.0 and 94.2%, respectively. In addition, the real ileal amino acid digestibilities were calculated. The recoveries of endogenous protein in ileal digesta and the real ileal protein and amino acid digestibilities were higher than those determined by feeding protein-free diets for determining the recovery of endogenous protein and amino acids in ileal digesta.  相似文献

Three pigs of 34 kg live weight were fitted with a re-entrant cannula in the duodenum, and with two catheters placed in the jugular vein and carotid artery. They were fed 1.2 kg/d of wheat-dried-skimmed milk diet. Digesta from the proximal duodenal cannula were collected for 12 h on 2 consecutive days; 50% were reintroduced into the respective distal cannula and 50% were stored at -20 degrees C. Three days later 14C-leucine was infused into the jugular vein for 12 h, starting with the morning meal. During this period the digesta from the proximal cannula were collected and stored for analysis while the digesta collected previously were reintroduced into the distal cannula. Blood samples were taken from the carotid artery. The total flow of duodenal digesta in 12 h was 5760 +/- 530 g. On average 70 percent of the radioactivity in digesta was associated with the TCA-precipitable fraction. During hours 0-3 and 11-12 of infusion 60-70% and 96-98% of the radioactivity in the TCA precipitable fractions was in leucine. In the TCA soluble fraction only 30-40 per cent of the radioactivity was associated with leucine. At the plateau 2.1 and 3.3% of infused 14C-leucine and of radioactivity were recovered in the duodenal digesta. The calculated amount of endogenous protein passing the duodenum was 20.4 g/d/pig. The results are discussed in relation to previous studies on protein synthesis and secretion in which 14C- and 15N-amino acids were used.  相似文献

The objectives of the two studies were to determine whether the guanidination and isotope dilution methods applied both by labeling the animal (15N-infusion method) and by diet (15N-dilution method) give similar estimates of ileal endogenous lysine (EL) and endogenous nitrogen (EN) flows, respectively, to that of the enzyme-hydrolyzed casein (EHC) method in the growing pig and to determine whether the guandination and 15N-dilution methods give similar estimates of EL and EN flows, respectively, to that of the EHC method in the rat. For the first study, the test diet contained guanidinated and enzymatically hydrolyzed casein (molecular weight < 5,000 Da), which was also labeled with 15N. Rats (n = 30; mean BW 178 g) and pigs (n = 6; mean BW 19.2 kg) received a preliminary EHC-based diet for 7 d. The test diet was then given to the rats and pigs on d 8. Digesta were sampled from the terminal 20 cm of ileum of killed animals. The EL flows determined using the guanidination method were lower than those determined using the EHC method (means of 298 vs. 382, and 214 vs. 287 microg/g of DMI, in the rat and pig, respectively; P < 0.05 for the rat and P < 0.01 for the pig). The EN flows determined with the 15N-dilution method were lower than those determined using the EHC method (means of 1,034 vs. 1,942 and 1,011 vs. 1,543 microg/g of DMI, in the rat and pig, respectively, P < 0.001 for the rat and P < 0.05 for the pig). In the second study, pigs (n = 6; mean BW 27 kg) were continuously infused via the jugular vein with 15N-leucine for 11 d. The pigs received an EHC-based diet (molecular weight < 5,000 Da) during this 11-d period, after which digesta were sampled at the terminal ileum under anesthesia. The EN flow determined using the 15N-infusion method (mean of 1,971 microg/g DMI) was higher (P < 0.01) than that determined using the EHC method (mean of 1,233 microg/g DMI). The guanidination method gave a lower estimate of EL flow than did the EHC method in both the pig and rat. The 15N-dilution method also gave a lower estimate of EN flow than the EHC method in the pig and rat, and the 15N-infusion method gave a higher estimate of EN flow than the EHC method in the pig.  相似文献

The experiment was carried out on 3 pigs of 34 kg body weight equipped with re-entrant cannulas both in the duodenum and terminal ileum. Furthermore, catheters were inserted into the external jugular vein and into the carotid artery. During a preliminary period digesta from both cannulas were collected and stored. This digesta was than reintroduced during a 12 hours period of intravenous infusion of 14C-leucine while outflowing digesta from the proximal parts of the cannulas were collected, stored and analyzed for leucine content, 14C-radioactivity and specific radioactivity of leucine. This paper reports the net secretion of 14C-activity and of 14C-leucine by the small intestine and the recovery of both in the content of the digestive tract at the end of the infusion. It was concluded that endogenous leucine enters the lumen of the small intestine through the intestinal wall mainly in a protein bound form. Free leucine is secreted, however, only in small amounts. Metabolites of leucine were also secreted into or formed in the lumen of the small intestine. A proportion of endogenous leucine was reabsorbed during the passage of digesta along the small intestine.  相似文献

Six heifers with a live weight of 215, 227 and 238 kg (experiment 1) and 220, 227 and 233 kg, resp. (experiment 2), were supplied with ileocaecal re-entrance cannulae, jugular venous catheters and bladder catheters. The ration consisted of 4 kg maize silage and 4 kg wheat straw pellets per animal per day. Up to 3.5 kg of the straw pellets, consisting of 73% wheat straw, 10% barley, 12% molasses, NPN salts and a mineral mixture, were consumed per animal per day. In a preliminary period 50% of the digesta flow was collected over 12 h/d on 5 consecutive days and stored in a deep-freeze. During the main trial the re-entrance cannula was disrupted and the flowing digesta were quantitatively collected at the end of the ileum; previously collected digesta were supplemented with 15N urea and every hour over 24 h infused into the caecal part of the re-entrance cannula. Between the 24th and 30th hours the digesta were infused without 15N urea supplement. In trial 2 the digesta were also supplemented with partly hydrolysed straw meal between the 1st and 30th hours (approximately 10% straw meal DM related to digesta DM). There were no differences between trials 1 and 2 with regard to the increase of atom-% 15N excess (15N') in the plasma urea. The 15N labelling decrease of the plasma urea N shows that the half-life is 7.9 h in trial 1 and 7.0 h in trial 2. The NH3 nitrogen in faeces was distinctly higher labelled in trial 2 after the supplement of straw meal than in trial 1. The total N in faeces was also twice as highly labelled as in trial 1. Atom-% 15N' in urine was significantly higher in trial 2 than in trial 1 between the 6th and 16th hours after the beginning of 15N urea supplementation. In the decrease curve of atom-% 15N' (after the 26th hour of trial) the values in trial 1 were generally higher than in trial 2. The higher bacterial protein synthesis in the large intestine in trial 2 (after the supplement of partly hydrolysed straw meal) had the effect that 13.6% of the supplemented 15N' was excreted in faeces by the 30th hour of trial, in contrast to this only 4.7% in group 1. Up to the 4th day after the 15N urea infusion these values increased to 16.2 and 6.1%, resp., only.  相似文献

Sixteen male growing pigs of about 24 kg BW were fitted with both a duodenal re-entrant and a post-valve T-shaped cannula inserted in the caecum. The animals were divided into four groups. Each group received one of the following diets: corn starch-soybean protein isolate-based diet without (diet C) and with carboxymethylcellulose (diet CMC) or a rye-wheat-based diet without (diet RW) and with xylanase addition (diet RWX). The diets provided similar levels of apparent precaecal digestible crude protein (CP), lysine, methionine + cystine, threonine and tryptophan. Additionally, [15N]-yeast was given with the diets during the first 10 days of the experiment. For estimation of digesta viscosity, N-flow of dietary and endogenous origin, apparent precaecal digestibilities of dry matter (DM), CP, amino acids and non starch polysaccharides (NSP) (only in pigs fed diets RW and RWX), ileal and duodenal digesta were quantitatively collected on day 16 and 17, respectively. The endogenous N-proportion was measured by the ratio of 15N enrichment in the digesta and urine. The duodenal and ileal digesta supernatant viscosity increased as carboxymethylcellulose was included into the diet. Xylanase addition to the rye-wheat based diet reduced the viscosity in the ileal digesta. There were no differences in precaecal digestibilities of DM, CP and amino acids between diet C and CMC. The precaecal digestibilities of DM and soluble and insoluble NSP increased from 69.5% to 73.9%, from 1.3% to 47.9% and from 17.0% to 35.4%, respectively, as xylanase was added to the rye-wheat-based diet. The apparent precaecal digestibility of most essential amino acids increased by 2 to 5 percent units. The amounts of endogenous N at the duodenal level were estimated to be 158, 233, 313 and 276 mg per 12 h per kg0.75 BW of pigs fed diets C, CMC, RW and RWX, respectively. The corresponding values at the ileal level were 95, 107, 164 and 150 mg per 12 h per kg0.75 BW. For endogenous N amounts, significant differences were observed between diets C and CMC (duodenum) and also between semi-purified and cereal-based diets (duodenum and ileum). Methodological aspects for the estimation of endogenous N using the isotope dilution technique are discussed. Obviously, the digesta viscosity per se does not affect the nutrient absorption and endogenous N flow within the small intestine of pigs. Other properties of complex dietary fibre, digesta passage rate or bacterial activity probably contribute to the observed changes.  相似文献

60 Wistar rats (5 animals/group) received 12 different feedstuffs over a period of 7 days and were simultaneously labelled with 15N (orally by means of 15NH4Cl in the feed). In the subsequent 5 days faeces were collected in order to determine the apparent and true digestibility of crude protein. On the 13th experimental day the animals were killed 3 hours after the intake of half the daily ration and the atom-% 15N excess (15N') was determined in the TCA-soluble and TCA-precipitable fractions of the blood plasma and the digesta of the 2nd and 3rd thirds of the small intestine. Precaecal N-absorption was calculated with the help of the quotient [formula: see text] the blood plasma and the digesta The following values (in %) were registered in comparison to true N-digestibility (in the following in brackets): casein = 95.8 (99.2), whole egg = 92.1 (97.5), fish meal = 85.8 (93.4), dried skimmed milk = 98.4 (96.1), soybean meal = 79.6 (90.6), assay protein = 94.2 (98.9), wheat = 92.9 (90.7), barley = 84.3 (84.8), yeast, grown on molasses = 85.6 (87.2), yeast, grown on whey = 86.1 (88.5), biomass of liquid manure = 43.6 (68.9), activated sludge = 54.4 (64.1). One can conclude that the isotope dilution technique demonstrated here as an evaluation method is very well suited for the characterization of the N-digestibility of a feedstuff in the small intestine.  相似文献

Four 40 kg castrated male pigs fitted with simple 'T' cannulas in the terminal ileum were given 15N-labelled ammonium salts, added to a low protein diet, for 6 days. Excretion of 15N in urine and faeces was monitored daily throughout the labelling and subsequent experimental periods. During the experimental period the pigs were given a diet based on wheat and fish meal, supplemented with varying levels of partially hydrolysed straw meal to give crude fibre contents ranging from 40 to 132 g/kg. After adaptation to the particular levels of straw meal, faeces and ileal digesta were collected during successive 24 h periods. N digestibility values were determined by the chromic oxide ratio method. The retention of 15N labelled non-specific N was 0.46 of the dose given. The validity of using urine values as a measure of 15N abundance in endogenous N was demonstrated by the similarity of 15N abundance in urine immediately before slaughter at the end of the experiment and in the digestive secretory organs thereafter. The average amount of endogenous N passing the terminal ileum was 3.4 g/day or 0.30-0.50 of total ileal N flow. This was not affected by dietary fibre level. The proportion of faecal N which was of endogenous origin was similar to that in ileal digesta, suggesting similar utilization of endogenous and residual dietary N by hindgut bacteria. Half the endogenous N entering the large intestine was reabsorbed there. Increasing dietary crude fibre from 40 to 132 g/kg increased faecal endogenous N excretion from 1.3 to 2.0 g/animal and day.  相似文献

Barrows with an average initial weight of 55 kg were fitted with simple T-cannulas at the distal ileum. The animals were fed a protein-free diet that consisted of 79.7% cornstarch, 10% sucrose, 3% Alphafloc (a source of cellulose), 3% canola oil and a vitamin-mineral premix. The pigs were fed 700 g of diet twice each day, at 0800 and 2000. A balanced amino acid mixture or a saline solution was administered intravenously while the protein-free diet was fed. Ileal digesta were collected for 24 h following a 7-d adaptation period. The administration of amino acids reduced (P less than .05) the recovery of endogenous protein from 18.5 to 12.7 g per kg dry matter intake. For the amino acids, the reduction was only significant (P less than .05) for proline, from 3.6 to .6 g per kg dry matter intake. If the total endogenous protein losses are assumed to be constant and the differences in the amino acid composition of non-reabsorbed endogenous protein, as observed in this study, are used to calculate true ileal digestibilities, differences in the digestibilities of the indispensable amino acids are large (up to 7.4 percentage units for threonine). The amino acid composition of endogenous protein determined in pigs fed a protein-free diet and parenterally administered with amino acids should provide a better estimate for the calculation of true amino acid digestibilities when based on the determination of true protein digestibility by the 15N-isotope dilution technique.  相似文献

Three heifers with live weights of 255, 261 and 300 kg were supplied with ileo-caecal re-entrant cannulas, jugular vena catheters and bladder catheters. The ration consisted of 4 kg maize silage and 4 kg wheat straw pellets. In a previous period 50% of the digesta flow was collected over 12 h/d on 5 consecutive days and stored in a deep-freeze. During the main period the re-entrant cannulas were disrupted and the flowing digesta was quantitatively collected. Precollected digesta and pectin were infused into the distal part of cannula hourly for about 30 hours. During the first 24 hours the digesta was also supplemented with 15N-labelled urea. The amount of pectin corresponded to about 10% of digesta dry mater. An analysis of urine, faeces, digesta and blood plasma were carried out. The application of pectin increased the 15N-incorporation in the bacterial protein of faeces from 4.7% (without supplementation in an earlier experiment) to 10.5% of the introduced 15N. The ammonia-fraction of faeces was markedly higher than the bacterial fraction. The 15N-utilization of urea by the microbes of large intestine was lower in the actual trial evident than with supplementation of starch in the anterior experiment. During the pectin administration the amount of urine increased in comparison with earlier experiments and according to the literature to about the 4.5 fold. The amount of passage of 15N at the ileum cannula (recycled 15N) was 3.8% of the 15N intake. It is the same amount as in experiments in which starch was applied.  相似文献

Pigs received a diet based on wheat and dried skimmed milk in equal amounts at 12 h intervals. During the preliminary period the digesta flowing from both duodenal and ileal cannulas were collected over 12 h after feeding on two consecutive days and half of them were reintroduced into the gut and half were stored at -20 degrees C. During the experimental period 15N-urea was infused into the jugular vein for 12 hours starting with the morning meal. Total amount of urea infused was 5 g containing 1.22 g 15N-excess. The digesta from both proximal duodenal and ileal cannulas were collected and stored, while the digesta from the preliminary period were reintroduced into the respective distal cannulas. Blood samples were taken at different time of infusion. At the end of infusion period the animals were sacrificed and samples of the contents of the digestive tract and tissues were taken. Urea flux calculated according to atom-% 15N-excess of urea N in plasma was 1.23 to 2.37 g/kg body weight/day. In the duodenal digesta 94.5 +/- 0.2 and in ileal digesta 57.1 +/- 7.39 per cent of 15N were in the TCA soluble fraction. The total amount of 15N in the duodenal digesta was 1.7 to 6.3 times greater than in the ileal digesta. Only small amount of 15N was found in the caecum and almost none in the contents of colon and rectum. It is concluded that urea is secreted into all parts of the digestive tract, the main sites of urea secretion being pancreatic juice and/or bile as well as the small intestine. The total amount of urea secreted is assumed to be similar to the daily urea excretion.  相似文献

A study was conducted using the ileal digestibility technique with grower-finisher pigs to evaluate the effects of particle size reduction of soybean meal (SBM) on amino acid and energy digestibility. Soybean meal was processed through a hammer mill to achieve average particle sizes of 900, 600, 300, and 150 microm. The treatments included the use of two soybean meal sources and soy protein concentrate. One source of SBM was ground to four different particle sizes (i.e., 949, 600, 389, 185); a second source was a common SBM source used in other trials (i.e., 800 microm). The soy protein concentrate had an average particle size of 385 microm. A low-protein (5% casein) diet was fed to determine endogenous amino acid losses. This experiment was conducted in a 7 x 7 Latin Square design in two replicates using 14 crossbred barrows ([Landrace x Yorkshire] x Duroc) that averaged 28 kg BW and 60 d of age. Animals were surgically fitted with a T-cannula at the distal ileum. Treatment diets were fed in meal form, initially at 0.09 kg BW(0.75) and at graded increases at each subsequent period. Pigs within replicate were fed a constant quantity of their treatment diet for a 5-d adjustment period followed by a 2-d collection of ileal digesta samples. Apparent and true digestibility of amino acids was calculated by use of chromic oxide (0.5%) as an indigestible marker. Apparent digestibility of isoleucine, methionine, phenylalanine, and valine increased linearly (P < 0.05) as particle size decreased. True digestibility of isoleucine, methionine, phenylalanine, and valine increased linearly (P < 0.05) as particle size decreased. When the essential amino acids were averaged, apparent digestibility increased (P < 0.10) from 83.5% to 84.9% as particle size decreased, whereas, nonessential amino acid digestibility increased only slightly (P > 0.15). Essential amino acid true digestibility increased numerically from 91.0% to 92.4% as particle size decreased. Energy digestibility was not affected by particle size (P > 0.15). These results suggest that a reduction in particle size of soybean meal resulted in a small increase in the digestibility of its amino acids with the essential amino acids being affected more than the nonessential amino acids. The largest improvement in digestibility, however, was obtained when the particle size was reduced to 600 microm.  相似文献

Four gilts (average BW 80 kg) were used in the first experiment to study the effect of i.v. infusion of urea on urea kinetics by means of a radioisotope dilution technique. The pigs were fed twice daily 600 g of a cornstarch-based diet formulated to contain 16% CP by supplementation with isolated soy protein. Infusion of urea, compared with saline, increased (P < .05) plasma urea concentration, urea pool size, urea entry, urea excretion, and urea degradation rates; urea turnover rate and urea space were not affected (P > .05). Expressed as a percentage of the total entry rate, a lower (P < .05) percentage of urea was recycled in pigs infused with urea. The urea infused was almost completely excreted in urine, so there were no differences (P > .05) in N balance. In the second experiment, four gilts (average BW 40 kg), fitted with ileocecal reentrant cannulas, were used to determine whether the upper or the lower digestive tract represents the preferential site of urea secretion in pigs. Two pigs were fed twice daily 600 g of a cornstarch-based diet, formulated to contain 16% CP from soybean meal. The other two pigs were fed the same diet in which 15% cornstarch was replaced by beet pulp. After labeling the body urea pool of one pig on each treatment with [15N]urea, the reentrant cannulas were disconnected to prevent the flow of digesta from the small into the large intestine.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

An experiment was conducted to investigate the effects of BW, feed intake, and the physiological condition of the animal on the loss and amino acid composition of endogenous protein in swine. Ten growing barrows and five multiparous sows were equipped with a T-cannula in the distal ileum for digesta collection. A protein-free diet was fed to all animals. The barrows were given free access to the experimental diet. The sows also were allowed to consume the diet on an ad libitum basis, and digesta were collected during lactation and in the following gestation period. In addition, digesta from the gravid sows were collected after restricting the sows to 2 kg of feed per day. For each animal group, the endogenous losses of protein and amino acids were calculated in relation to DMI, and the amino acid composition of endogenous protein was calculated. The total endogenous gut protein loss at the distal ileum of growing pigs, lactating sows, and gestating sows, given free access to feed, was 12.4, 9.4, and 11.2 g/kg DMI, respectively. These values were not different (P > .10). However, when gestating sows were fed only 2 kg/d, 17.8 g of endogenous protein was lost per kilogram of DMI, which was higher (P < .05) than for any of the other groups. This difference was mainly caused by higher (P < .05) losses of glycine, proline, and serine. There were no differences (P > .05) in amino acid composition of endogenous protein between growing pigs, lactating sows, and gestating sows given free access to feed, but restricted-fed gestating sows had an amino acid composition of endogenous protein that was significantly different from that of the other groups. The results from the experiment showed that age, BW, and the physiological condition of the animal have little or no effect on the amount of endogenous protein and amino acids lost at the distal ileum of hogs if calculated in relation to DMI. Likewise, the amino acid composition was not affected by the BW or physiological condition of the animal. However, DMI had a significant effect on endogenous protein losses in sows as well as on amino acid composition of endogenous protein.  相似文献

The experiments were carried out on 3 bulls (body weight: 172, 229 and 193 kg), equipped with ileo-caecal cannulas and with catheters in the jugular veins on both sides. The offered pelleted ration consisted of straw 73%, molasses 12%, cereals 10%, ammonium hydrogen carbonate 3% and urea 2%. Feed intake amounted to about 3 kg per animal and day. During a preliminary period of 5 days 50% of ileal digesta were collected for 12 hours daily, deep-freezed and stored. In the main experiment 15N-urea was infused intravenously for 24 hours. During this period and during the following 6 hours ileal digesta were collected and replaced by precollected, unlabelled digesta. The urea metabolism was estimated by the 15N-labelling of blood urea, by the 15N-excretion via faeces and urea, by the appearance of 15N in ileal digesta and by the 15N-labelling of faecal NAN, NH3 and bacterial fraction. The time course of the 15N-labelling of plasma urea during infusion can be described by an exponential function. The urea flux rate was estimated from the calculated plateau value. The flux rate for the 3 animals was 28.8, 30.7 and 34.8 mumol per minute per kg0.75, respectively. During the infusion of 15N-urea 1.0-2.4% of the infused amount of 15N' appeared in ileal digesta, half of it in the TCA precipitable fraction. At the same time the 15N-labelling of faecal NH3 increased sharply, however, the 15N-labelling of the faecal bacterial fraction was smaller by one order of magnitude. Deficiency of fermentable substrates and problems of inhomogenity of the NH3 pool are supposed as reasons for this result. 30 to 50% of the urea flux entered the digestive tract, the direct entry of urea into the large intestine seems to be only very low.  相似文献

To compare the effects of time of daily protein supplementation on grazing behavior, forage intake, digesta kinetics, ruminal fermentation, and serum hormones and metabolites, 12 ruminally cannulated Holstein steers (449 and 378 kg average initial and final BW, respectively) were allotted to three groups. Treatments consisted of CON = no supplement, AM = cottonseed meal (.25% of BW) at 0600, and PM = cottonseed meal (.25% of BW) at 1200. Steers grazed a dormant (1.1% N) intermediate wheatgrass (Thinopyrum intermedium Host) pasture. Sampling trials occurred in December, January, and February. Supplementation altered (P = .01) time spent grazing; CON steers grazed approximately 1.5 h longer than supplemented steers. Supplemented steers lost less (P = .02) BW (-40 kg) than CON steers (-75 kg) did. Supplementation did not alter (P greater than .15) forage OM intake; however, total OM intake was greater (P = .01) for supplemented steers (22.3 g/kg of BW) than for CON (18.4 g/kg of BW) steers. Supplementation did not affect (P greater than .15) digesta kinetics. Extent of in situ NDF (96 h) and rate (%/h) of disappearance for supplemented steers was greater (P = .01) than for CON steers. Across all periods, ruminal NH3 N and total VFA concentrations were lower (P = .01) for CON steers than for supplemented steers. Serum insulin (ng/mL) concentration was lower (P = .03) and concentration of serum growth hormone (ng/mL) was higher (P = .02) for CON steers than for supplemented steers. Cottonseed meal supplementation enhanced utilization of intermediate wheatgrass; however, supplementation time had minimal effects on the variables measured.  相似文献

Male broilers were given a low protein diet (15.5% CP) spiked with [15N]H4HCO3 from day 12 to day 18 of age to label the endogenous N-constituents. Experimental diets were subsequently fed from day 19 to day 24 of age and consisted of a rye based diet (56% dietary inclusion) which contained either 10% soya oil (S) or 10% beef tallow (T), each of which was either unsupplemented (-) or supplemented (+) with a xylanase containing enzyme preparation (2700 IU/kg at pH 5.3). [15N]-atom percent excess (APE) of excreta, faeces and urine were monitored on a daily basis during both experimental periods. Furthermore, APE was measured in various tissues at the end of the experiment. The APE of urine on the last day of the experiment was between the APE of the pancreas and that of the jejunal tissue, an observation which supported the usefulness of using urinary APE as an indicator for the endogenous N-pool. Endogenous N-proportions were estimated by an isotope dilution technique at the end of the experiment by examination of the ratio of APE in faeces and urine. The endogenous N-proportion in the faeces was greatest in birds receiving the T(-) diet. The proportions were 0.321, 0.319, 0.451 and 0.289 in S(-), S(+), T(-) and T(+) fed groups, respectively. Xylanase addition reduced endogenous N-proportion, a factor which was used to correct apparent crude protein digestibility (85.6, 86.2, 84.3 and 88.5% in S(-), S(+), T(-) and T(+) fed birds, respectively) for endogenous losses resulting in almost equal true digestibilities of crude protein for all treatments (90.3, 90.6, 90.4 and 91.5%). The amounts of endogenous N in faces were estimated to be 87, 69, 244 and 81 mg per day per kg0.67 body weight in S(-), S(+), T(-) and T(+) fed birds, respectively. It was concluded that xylanase supplementation of a rye based broiler diet does not change endogenous N-secretions when the supplemental fat is soya oil. However, addition of tallow rather than soya oil increased these N-losses significantly, an effect which was reversed by xylanase addition.  相似文献

The aim of the study was to determine and compare the endogenous ileal excretions of nitrogen and amino acids under protein-free and peptide alimentation by the dog and rat. Two diets were prepared, one that was devoid of protein and the other containing 23% enzyme hydrolysed casein. Chromic oxide was included in the diets as an indigestible marker. A total of 10 mixed breed dogs were fed hourly either a protein-free or enzymatically hydrolysed casein diet for a total of 10 days. A faecal sample was obtained from each dog on day 9 while digesta was obtained from the terminal 20 cm of the ileum directly after euthanasia on day 10. A total of 12 8-week-old Sprague-Dawley rats received the same diets as the dogs. A faecal sample from each rat was obtained on day 7 while ileal digesta samples were obtained on day 8. The endogenous ileal excretions of most amino acids were greater in the dogs and rats that received the enzymatically hydrolysed casein diet compared with those receiving the protein free diet. Whereas the pattern of endogenous amino acid excretion was similar in the rats and dogs, the dogs excreted a significantly greater amount of nitrogen (1.91 vs. 2.27 and 1.63 vs. 4.12 g/kg dry matter intake for the protein-free and peptide alimentation method, respectively) and all amino acids except for glycine, isoleucine and leucine. Endogenous ileal amino acid excretions are higher in dogs compared to omnivorous animals such as rats and pigs but similar to the carnivorous cat.  相似文献

The digestibility of AA in dry extruded-expelled soybean meal (DESBM) and regular, solvent-extracted soybean meal (SBM) were determined in pigs and poultry. In the pig assay, 4 Cotswold barrows (average initial BW of 80.4 kg) fitted with a T-cannula at the distal ileum were allotted to 4 semipurified diets in a 4 x 4 Latin square design. Diet 1, a low protein diet (5% casein), was used to quantify endogenous CP and AA losses. Diets 2, 3, and 4 were formulated to contain 35% regular, solvent-extracted SBM; batch 1 of DESBM (DESBM-1); and batch 2 of DESBM (DESBM-2), respectively, as the sole source of protein. The DESBM samples were obtained from 2 different batches but were subjected to the same processing conditions. Chromic oxide (0.3%) was included as a digestibility marker in all diets. Compared with DESBM-1 and DESBM-2, apparent ileal digestibility of DM in SBM was greater (P < 0.05). Apparent and true ileal digestibilities of AA in SBM were greater (P < 0.05) compared with DESBM-2. In the poultry assay, 4 dietary treatments were each assigned to adult cecectomized roosters in a completely randomized design. Treatment 1 was a nonnitrogenous diet (NND; 90% sucrose and 10% vegetable oil) used to estimate endogenous N and AA losses. Treatments 2, 3, and 4 contained SBM, DESBM-1, and DESBM-2 as the only source of protein. Each of these diets was fed in 25-g quantities formulated to provide 5 g of CP from the respective soybean meal source. The SBM had greater (P < or = 0.05) true digestibility for isoleucine, leucine, cysteine, proline, serine, and tyrosine compared with DESBM-1. The results indicate that, relative to regular, solvent-extracted soybean meal, AA digestibilities of different batches of dry extruded-expelled soybean meal varied in pigs and poultry.  相似文献

1. Three experiments were designed to determine the response of broiler chickens to dietary isoleucine, and to quantify the antagonistic effects of excess leucine and valine on this response. 2. A dilution technique was used to measure the responses in growth rate and food intake to a range of diets differing in their isoleucine concentrations. A summit diet was formulated to contain isoleucine at 1.14 times the requirement and with leucine (1.76 times the requirement) and valine (1.87 times the requirement) at the minimum possible concentrations, given the ingredients available. A dilution mixture, devoid of protein, was formulated to correspond in all respects, other than in amino acid content, to the summit diet. These two basal diets were blended in different proportions to give a range of diets of decreasing isoleucine and protein content. 3. In experiment 1 the response was measured to isoleucine with leucine and valine remaining in the same proportion to isoleucine throughout the range of diets fed. In experiments 2 and 3, however, L-leucine and L-valine were added to the diets either singly or in combination to give 6 isoleucine concentrations and 3 ratios of each of leucine and valine to isoleucine. 4. Weight gain decreased as the isoleucine content of the diet was reduced, whereas food intake of broilers fed on the marginally deficient diets increased to a maximum and then decreased. FCE decreased curvilinearly as the isoleucine concentration in the food decreased, reflecting a concomitant change in the fat content of the broilers. 5. It is possible that the amount of dietary isoleucine assumed to be available to the broilers in these experiments was overestimated by hydrolysing the food samples for 72 h, and the doubt thus created makes an estimate of the efficiency of retention of isoleucine suspect. 6. Excess valine had no effect on the response to isoleucine, whereas an increase in the leucine to isoleucine ratio depressed food intake and hence weight gain, but only at the lowest concentrations of isoleucine. 7. If the food content of isoleucine is sufficient to meet the requirements of the broiler, relatively large excesses of leucine, of valine, or of both will not depress growth.  相似文献
