Ogura cytoplasmic male sterility (Ogu-CMS) is widely used in the production of commercial hybrids of Brassica oleracea. However, the widespread application of the Ogu-CMS system in B. oleracea has hindered the germplasm innovation of Ogu-CMS resources due to the lack of a natural restorer line. Previously, the Ogu-CMS fertility-restored interspecific hybrids between rapeseed 15Y403 (2n = 38, AACC) and Chinese kale JL1 (2n = 18, CC) have been successfully produced. However, these progenies, which still contained a large proportion of rapeseed genomic components, showed poor fertility and a low seed setting rate under natural pollination. To improve fertility and seed setting, a successive backcross with JL1 was performed to produce BC2 progenies. Screening with the Rfo-specific marker, five individuals harboring the Rfo gene were identified among 98 BC2 progenies. These five individuals underwent background marker screening and an evaluation of agronomic traits and fertility. One individual (code: 15Q23) was identified with higher pollen viability, better seed setting under natural pollination, and a closer genetic background to the parent Chinese kale JL1. Many morphological traits showed no significant differences (P < 0.05) between 15Q23 and the backcross parent JL1. However, the average seed setting of 15Q23 under natural pollination was 0.72 seeds per pod, which was 50 times higher than that of BC1 progenies, and the average pollen viability was 87.07%, which was significantly better than that of the F1 and BC1 plants (P < 0.01). The genetic background of 15Q23 was more closer to the parent JL1 than that of BC1 plants and another BC2 fertility-restored individual, with 82% of the polymorphic alleles being the same as those of the parent Chinese kale JL1. Thus, the individual 15Q23 could be used as a donor plant for further backcrosses. This study lays the foundation for the development of Ogu-CMS restorer material in B. oleracea.  相似文献

A new tomato rootstock seed parent which combines the characters green stem, potato leaf and long style in a Lycopersicon esculentum genotype has been produced. This will facilitate seed production of the interspecific F1 hybrid rootstock which is used for grafted crops, and also permit the positive identification of any off-types which arise from accidental self-pollination of the seed parent. The new seed parent has been named GCR 66. It is proposed that rootstocks derived from this type of seed parent shall be called Markerstocks. GCR 66 is very similar to the cultivar Ailsa Craig in its agronomic characters and will be useful for studies with the tomato when pollination needs to be accurately controlled.  相似文献

Summary A study was made over three seasons of seed yield characters in replicated genotypes of two maturity classes selected from within both Mbarara and Masaba Rhodes.Within maturity classes significant 2–4 fold ranges were observed in PGS (pure germinating seed) yield. The component most responsible for this increase was seed setting. Significant differences were also observed in seed retention, 1000-seed weight, head number and the number and length of racemes.Even though heading date, within a maturity class of a variety did not vary more than 10 days, differences were significant and the early-heading plants were the most productive in seed yield characters.Heritability estimates, based on individual plants, ranged from less than 0.10 for % PGS to over 0.30 for 1000-seed weight and raceme characters.  相似文献

The ongoing rise in temperatures caused by global climate change is a critical climatic risk factor for rice production, and enhancing rice heat tolerance is an area of particular research interest. A recombinant inbred line (RIL) mapping population was developed from heat sensitive, rice cultivar IAPAR-9 crossed with heat tolerant, Liaoyan241. RIL and parental lines were exposed to high temperature at the heating and flowering stage in experiments in 2014 and 2015. As indicators of heat tolerance, the seed setting rate under natural (NS) and heat stress (HTS) conditions were measured, and the reduction rate of seed set (RRS) was calculated. Quantitative trait loci (QTL) analysis revealed eleven heat tolerance QTLs located on chromosomes 1, 3, 4, 5, and 6. Single QTL contribution rates were 4.75–13.81% and effect values were ? 5.98 to 5.00. Four major QTLs (qNS1, qNS4, qNS6, and qRRS1) were stable detected in different environments in both years. Thirteen QTLs with epistatic interactions and nine QTLs with environmental interactions were also detected. Major QTLs were all involved in epistatic and environmental interactions. Three QTLs from the SSR marker interval RM471 to RM177 region of chromosome 4 (qNS4, qHTS4, and qRRS4) were all involved in epistatic and environmental interactions and contributed to phenotypic variation, indicating that this region constituted a major QTL hotspot. The major QTL for heat tolerance identified in this study will aid in breeding tolerant cultivars and facilitating investigation of the molecular underpinnings of heat tolerance in rice.  相似文献

Knowledge of the biological properties of pollination and fertilization are essential for breeding and the development of breeding concepts. In this investigation, panmictically cross-pollinated seeds of two selected combining partners of valerian (Valeriana officinalis L.) were produced and in the progeny, the outcrossing rates (OCR) were determined by using amplified fragment length polymorphism analysis (AFLP). The previous assumption that Valeriana officinalis L. shows a predominantly high outcrossing rate (OCR) was confirmed. The OCR ranged from 76.5 to 97.7%. Several mother plants showed an OCR of 100% in their progeny. Partially involved heterozygous DNA-fragments could have led to undetected outcrossings and to a lower OCR. The preferred outcrossing direction and the individual seed amount of the mother plants may influence the performance of a seed mixture, generated by both partners as mother plants.  相似文献

Summary Four apple (Malus X domestica) genotypes, Erovan, Golden Delicious, R1-49 and X6677, were pollinated with a marked pollen irradiated by -rays at doses ranging from 125 to 1000 Gy. Pollination with such irradiated pollen affected fruit set, seed number and seed contents, and induced the formation of parthenocarpic fruits and the development of parthenogenetic embryos. The immature embryos extracted from seeds. 2 and 3 months after pollination, were cultured in vitro and germinated after 2 months of cold treatment (3°C). Haploid plants were obtained in all 4 genotypes, after pollination with pollen irradiated at doses from 200 to 500 Gy. The optimum conditions for induction of apple haploids, by irradiated pollen approach, have been established.  相似文献

Broadening the genetic base of the C genome of Brassica napus canola by use of B. oleracea is important. In this study, the prospect of developing B. napus canola lines from B. napus?×?B. oleracea var. alboglabra, botrytis, italica and capitata crosses and the effect of backcrossing the F1’s to B. napus were investigated. The efficiency of the production of the F1’s varied depending on the B. oleracea variant used in the cross. Fertility of the F1 plants was low—produced, on average, about 0.7 F2 seeds per self-pollination and similar number of BC1 seeds on backcrossing to B. napus. The F3 population showed greater fertility than the BC1F2; however, this difference diminished with the advancement of generation. The advanced generation populations, whether derived from F2 or BC1, showed similar fertility and produced similar size silique with similar number of seeds per silique. Progeny of all F1’s and BC1’s stabilized into B. napus, although B. oleracea plant was expected, especially in the progeny of F1 (ACC) owing to elimination of the A chromosomes during meiosis. Segregation distortion for erucic acid alleles occurred in both F2 and BC1 resulting significantly fewer zero-erucic plants than expected; however, plants with?≤?15% erucic acid frequently yielded zero-erucic progeny. No consistent correlation between parent and progeny generation was found for seed glucosinolate content; however, selection for this trait was effective and B. napus canola lines were obtained from all crosses. Silique length showed positive correlation with seed set; the advanced generation populations, whether derived from F2 or BC1, were similar for these traits. SSR marker analysis showed that genetically diverse canola lines can be developed by using different variants of B. oleracea in B. napus?×?B. oleracea interspecific crosses.  相似文献

Pollen viability and stigma receptivity are prerequisites for successful pollination and seed set in flowering plants. In this study, the pollen viabilities and stigma receptivities of nine Lilium genotypes (six cultivars and three species native to China) were assayed by in vitro pollen germination and the benzidine-H2O2 method, respectively. Embryo sac development during anthesis was observed to further ensure the timing of controlled pollination. In addition, the relationship between stigma secretion and stigma receptivity was studied to estimate the pollination time based on phenotype. Anthers cracked on the day of flowering in all genotypes, but pollen germination during anthesis was not observed in Asiatic hybrids excepted for ‘Tiny pudhye’, which exhibited low pollen viability for a short period of time (from 0 to 1 day after anthesis). In the other genotypes, pollen germination rates were highest on anthesis (five of seven genotypes), 0–1 day after anthesis (L. sulphureum), or 0–2 days after anthesis (one Longiflorum hybrid), and then gradually decreased with days after anthesis. While, stigma receptivity first increased and then decreased during anthesis. For most genotypes, stigmas began to be receptive 1 day after anthesis, and all genotypes exhibited stigma receptivity at 2 days after anthesis. The durations of stigma receptivity and strongest stigma receptivity, were genotype dependent, and were 5–8 days and 1–4 days, respectively. Moreover, on the first flowering day, 6 of 7 genotypes had mature embryo sacs, and at the time at which stigmas began to be receptive, all tested genotypes had mature embryo sacs. Some Lilium genotypes showed stigma secretion, which can be a sign of stigma receptivity. Stigmas became receptive and reached highest receptivity within 1 day of the first appearance of secretion on the surface of the stigma and at peaking, respectively. The results of this study are valuable for the implementation of successful Lilium breeding programs.  相似文献

Summary Using the finger nails to remove most of the petals is a fast method of preparing apple flowers for hand pollination in the balloon stage. Such flowers still appeared quite attractive to insects; left to open pollination, the seed set per flower averaged two thirds of that of similarly treated, but hand-pollinated flowers. With the aid of marker pollen of scab or mildew resistant donors and the subsequent screening of the seedling progenies for resistance, it was shown that the stray pollen was responsible on average for one of every three seeds formed by unbagged depetalled and hand-pollinated flowers.  相似文献

Forsythia suspensa and F.Courtaneur’ were used as female parents to cross with Abeliophyllum distichum in 2011 and an intergeneric hybrid of F. suspensa × A. distichum was obtained, though with very low seed set. The morphological characteristics, flower fragrance and volatile organic compounds of flowers were analysed. The intergeneric hybrid had intermediate morphological characteristics of both parents and flower fragrance and was confirmed as a true intergeneric hybrid by amplified fragment length polymorphism (AFLP) markers. Compared with its mother parent (F. suspensa), flowers of the intergeneric hybrid are pale yellow with delicate fragrance. Volatile organic compounds of flowers were retrieved by purge-and-trap techniques, and determined by gas chromatography and mass spectrometry (GC–MS). The main volatile organic components of F. suspensa were isoprenoids, while the main volatile organic components of A. distichum and the hybrid of F. suspensa × A. distichum were aliphatics. To determine the time and the site of intergeneric hybridizing barriers occured, the pollen tubes’ behavior after pollination was observed under fluorescence microscopy. It was found that significant pre-fertilization incompatibility existed in intergeneric crossing combinations [F. ‘Courtaneur’ (Pin) × A. distichum (Thrum) and F. suspensa (Pin) × A. distichum (Thrum)], and only a few pollen tubes of A. distichum penetrated into the ovaries of Forsythia. In our research, an intergeneric hybrid between Forsythia and Abeliophyllum was obtained for the first time, which will provide a solid foundation for expanding the flower color range of Forsythia and breeding fragrant-flowered cultivars.  相似文献

The individual segregations of 14 seed protein loci named, SpA to SpM and Pha (phaseolin), were analyzed in a RIL population developed from the cross Xana × Cornell 49242. These seed protein loci were included in a genetic map previously developed in the same population. Protein loci, SpA, SpB, SpE, SpI, SpJ, and Pha, are organized in two different clusters, both located in linkage group (LG) 7; SpF, SpG, SpK, SpL, and SpM, form a single cluster in LG 4; SpC, is located in LG 3; and SpD, in LG 1. A close linkage was identified between the SpD seed protein locus, and the fin gene, controlling determinate growth habit. The usefulness of the SpD locus as a marker for the indirect selection of determinate growth habit and photoperiod insensitivity was checked in a F2 population derived from the cross G12587 (an indeterminate and photoperiod sensitive nuña bean) × Sanilac (determinate and photoperiod insensitive) and in a set of Mesoamerican and Andean genotypes. Results indicate that SpD protein locus was useful to detect individuals having determinate growth habit and photoperiod insensitivity in the cross G12587 × Salinac although some recombinants were found. However, the linkage between the SpD locus and the genes controlling growth habit and photoperiod sensitivity should be checked before using the SpD locus for the indirect selection of these traits in different backgrounds.  相似文献

To obtain crossing products, an efficient pollination and subsequent fertilization is essential. This efficient pollination is achieved by pollen germination and tube growth. Here, these pollen characteristics of 2 genetically differentiating cultivars of Anemone coronaria L. were investigated in vitro. For the essential components boron, calcium and an osmoticant, only calcium showed to be crucial for pollen germination. Boron concentrations influenced the pollen tube length with a concentration of 100 mg l?1 H3BO3 resulting in the longest pollen tubes. For the osmoticant sucrose, a concentration of 100 g l?1 caused a significant positive effect on both pollen germination and pollen tube length for the 2 cultivars. The cultivars reacted similarly with respect to the investigated compounds. Next to this, the pollen development was correlated with 8 different flower stages in A. coronaria. As a final point, the germination of an optimized pollen germination medium was compared with in vivo pollen germination in cross-pollinations within the same cultivar (identified by aniline blue staining). For ‘Mistral Wine’, pollen germination percentage was lower in vitro than in vivo, while ‘Wicabri Blue’ pollen showed no significant difference in germination rates in vivo and in vitro. To achieve fertilization a following requisite is that the stigma is receptive. To study this, the most receptive female flower stage of the 8 different flower stages for A. coronaria was characterized by aniline blue staining. A. coronaria clearly showed protogyny.  相似文献

Rice seed storability is an important characteristic of seed quality so that the cultivars with strong seed storability are expected in the production of hybrid seeds. Presently, little is known about the genetic and physiological mechanisms controlling rice seed storability. In this study, a double haploid population derived from the cross between a japonica cultivar CJ06 and an indica cultivar TN1 was used to identify the quantitative trait loci (QTLs) for seed germination percentage (GP) and fatty acid content (FA) during natural storage or artificial aging. A total of 19 QTLs, including ten QTLs for GP and nine QTLs for FA, were identified on nine chromosomes with the phenotypic variations ranged from 2.1 to 22.7%. Besides, six and three pairs of epistatic interactions were identified for GP and FA, respectively. Moreover, qGP-9, a QTL for germination percentage, was delimited in an interval of 92.8 kb between two STS markers P6 and P8, which contains 15 putative open reading frames. These results provide important information for understanding the genetic mechanisms on rice seed storability, and will be useful for breeding new rice varieties with high seed storability.  相似文献

Both low-temperature germinability (LTG) and cold tolerance at the seedling stage (CTS) are important traits for rice. In this study, a rice population of recombinant inbred lines (RILs), derived from the backcross population of a cross between Dongnong422 and Kongyu131, was developed to detect quantitative trait loci (QTL) affecting LTG and CTS by using seed of different storage times. Correlation analysis indicated that there was no significant relationship between LTG and CTS, suggesting that cold tolerance might be genetic differences for LTG and CTS. In total, Twelve and twenty-three major QTLs were detected for LTG and CTS, respectively, which could explain greater than 10% of the phenotypical variation. Eight (qCG12-1, qGI12-1, qGV9-1, qMLIT12-1, qPV6-1, qMDG12-1, qLDWcold10-1, qLFWcold10-1) significant QTLs were mapped for different storage time, it concluded that such QTLs were not affected by environment (storage time) and were closely related QTLs to cold tolerance. One or more QTLs were identified for each trait with some of these QTLs co-locating, qMLIT7-1, qCG7-1, and qGI7-1 for LTG, qLFWcold10-1, and qLDWcold10-1 for CTS with contributions over 15% were mapped common marker interval, respectively, co-location of QTLs for different traits can be an indication that a locus has pleiotropic effects on multiple traits due to a common mechanistic basis. Two lines, RIL128 and RIL73, might be valuable to improve the LTG and CTS through a combination of crosses. The identified QTLs might be applicable to improve the rice cold tolerance by the marker-assisted selection approach.  相似文献

Panicle exsertion is an important agronomic trait involved in pollination and hybrid rice seed production. The identification of genes controlling panicle exsertion will play a central role in hybrid rice breeding. C115, a chromosome segment substitution line carrying introgression segments of Nipponbare in genetic background of indica variety 9311, had significantly shortened panicle exsertion, uppermost internode length and plant height. Genetic analysis in F2 and F3 populations derived from a cross of C115 and 9311 indicated that the shortened panicle exsertion of C115 was recessive and controlled by a single Medelian factor, and the allele from Nipponbare decreased the phenotypic value. Primary mapping suggested qPE12, the Nipponbare introgression segment locating on the long arm of chromosome 12, was responsible for this phenotypic variation. Based on map-based cloning strategy, fine mapping was carried out with a total of 1130 recessive individuals selected from F2 and F3 populations, delimiting qPE12 to a 190-kb region. This result provides important information for isolation of a new gene controlling panicle exsertion in the future.  相似文献

The production of carrot seed sometimes presents the problem of the occurrence of seedless umbels. During an investigation into the cause of this phenomenon, a number of floral-biological data have been collected which are valuable both for the seed production and breeding of this crop.

    The distribution of male flowers and bisexual flowers. Both in the wild carrot and in 2 cultivated varieties the tendency to produce male flowers was encountered. In all forms tested the percentage of male flowers was practically negligible in the main umbel and was found to increase regularly in umbels of higher order. There were, however, clear differences in the mean percentage between the forms tested. Presumably the tendency to produce male flowers is determined genetically, but it may also be affected by the environment.  相似文献
