Out of six isolates of coccidia of the species Eimeria tenella, obtained in the years 1984 and 1985 on farms with a stationary occurrence of coccidiosis, five were found to have reduced sensitivity or resistance to monensin. Neither were these isolates sensitive to further monovalent ionophorous anticoccidials narasin and salinomycin. The bivalent ionophorous anticoccidial lasalocid controlled five isolates completely, but for one its effectiveness was reduced. An isolate obtained on a farm with long-term absence of coccidiosis was sensitive to all anticoccidial drugs.  相似文献

The efficacy of decoquinate against Eimeria infections in broiler chickens was evaluated using two drug sensitive laboratory strains of Eimeria tenella and 20 field isolates of Eimeria spp. collected from farms in China where various anticoccidials (including maduramicin) had been used. Decoquinate (20-40 ppm in feed) and maduramicin (5 ppm) were efficacious against E. tenella laboratory strains, but decoquinate more so than maduramicin. Body weight gains of E. tenella infected chickens were significantly improved, and caecal lesions were prevented, by feeding either decoquinate or maduramicin. Decoquinate also prevented oocyst production, but maduramicin did not. Most (18/20) Eimeria field isolates were resistant to maduramicin, judged by oocyst production; decoquinate at > or =20 ppm completely controlled all 20 field isolates. Decoquinate has potential value as a broiler anticoccidial in China and other countries where it has not been previously used.  相似文献

Eimeria parasites were isolated from Nanhai Guangdong province (southern China) and studied in chickens in wire cages to evaluate their drug resistance against commonly used ionophores: monensin (100 mg/kg of feed), lasolacid (90 mg/kg), salinomycin (60 mg/kg), maduramicin (5 mg/kg) and semduramicin (25 mg/kg). Chinese Yellow Broiler Chickens were infected with 40,000 crude sporulated Eimeria oocysts at 15 days of age and prophylactic medication commenced a day prior to infection. Drug resistance was assessed for each ionophore drug by calculating the anticoccidial index (ACI) and percentage optimum anticoccidial activity (POAA) based on relative weight gain, rate of oocyst production and lesion values. Results revealed that Nanhai Eimeria oocysts comprising of E. tenella, E. maxima and E. acervulina, were resistant to monensin, sensitive to both salinomycin and lasolacid and partially sensitive to maduramicin and semduramicin. By selection for early development of oocysts during passage through chickens, the prepatent time of E. tenella, E. maxima and E. acervulina were reduced by 49, 36 and 22 h, respectively. The precocious lines are less pathogenic than the parent strains from which they were selected and conferred a satisfactory protection for chickens against coccidiosis. These ionophore-tolerant precocious lines could have wider applications in the development of anticoccidial vaccines for sustainable control of coccidiosis.  相似文献

Coccidia were isolated from 90 broiler farms in 15 poultry-producing areas in Brazil and Argentina. Sixty isolates were tested for sensitivity to 7 anticoccidial drugs. The common species were: a) Eimeria tenella, 47 isolates; b) E. maxima, 49 isolates; c) E. acervulina, 44 isolates; d) E. mitis, 26 isolates; and e) E. brunetti, 12 isolates. Isolates were considered sensitive to drugs if intestinal lesion scores of medicated broilers were reduced by at least 50% compared with unmedicated infected broilers or if weight gain was at least 75% of that of uninfected birds in a 6-day laboratory test. According to lesion scores, there was evidence of resistance or seriously reduced sensitivity to monensin in 20 isolates, narasin in 29, salinomycin in 11, maduramicin in 1, clopidol in 36, amprolium in 40, and nicarbazin in 1. According to broiler weight gain, there was resistance to monensin in 36 isolates, narasin in 32, salinomycin in 28, maduramicin in 2, clopidol in 28, amprolium in 50, and nicarbazin in 4. These results suggested incomplete cross resistance of coccidia to polyether ionophorous drugs. The degree of resistance might be explained by previous patterns of use of these drugs.  相似文献

为检测巨型艾美耳球虫和柔嫩艾美耳球虫早熟株对常用抗球虫药的敏感性,选用地克珠利、氯苯胍、二硝托胺、癸氧喹酯、尼卡巴嗪、马杜米星、盐霉素和拉沙洛西等8种抗球虫药进行鸡体试验,通过抗球虫活性百分比(percent of optimum anticoccidial aetivity,POAA)、病变记分减少率(reduction of lesion scores,RLS)、相对卵囊产量(relative oocyst production,ROP)和抗球虫指数(anticoccidial index,ACI)4项指标进行综合评价。结果表明,巨型艾美耳球虫早熟株对8种药物敏感;柔嫩艾美耳球虫早熟株对盐霉素轻度敏感,对地克珠利、氯苯胍、二硝托胺、癸氧喹酯、尼卡巴嗪、马杜米星和拉沙洛西敏感。本研究结果可为对球虫早熟株疫苗的开发与应用提供实验依据。  相似文献

Coccidia were isolated from 30 broiler farms in major poultry-producing provinces of Canada; 21 mixed-species isolates were tested for sensitivity to anticoccidial drugs. Eimeria acervulina was identified in 20 samples, E. maxima in 15, and E. tenella in 10 of the mixed-species isolates. On the basis of weight gain after infection, 3 isolates were judged sensitive to amprolium, 5 to monensin, 9 to salinomycin, 14 to lasalocid, 19 to halofuginone, and 20 to nicarbazin. Similar trends in sensitivity/tolerance were identified from the percent reduction of intestinal lesion scores of medicated birds compared with scores of unmedicated, infected controls. These results were consistent with the previous pattern of use of anticoccidial drugs in commercial poultry in Canada.  相似文献

A battery trial was conducted to evaluate the drug sensitivity in Eimeria tenella field isolates against the commonly used anticoccidials salinomycin (60 ppm), maduramicin (5 ppm), and clopidol (125 ppm) in broiler chicks. These anticoccidials were mixed in feed at d 12 of age, and inoculation was given on d 14 of age. Drug sensitivity was determined by using the global index, which is composed of percentage weight gain, FCR, lesion score, oocyst index, and mortality percentage. In the present study, all the E. tenella isolates showed partial resistance against salinomycin, whereas varying degrees of sensitivity were observed against maduramicin and clopidol.  相似文献

为了测试柔嫩艾美耳球虫的4种抗药虫株(抗地克珠利、抗拉沙里菌素、抗盐霉素和抗克球粉虫株)对4种抗球虫药(地克珠利、拉沙里菌素、盐霉素和克球粉)的敏感性进行了本试验。结果表明:柔嫩艾美耳球虫地克珠利抗药虫株对拉沙里菌素、盐霉素和克球粉均表现敏感,拉沙里菌素抗药虫株、盐霉素抗药虫株和克球粉抗药虫株对地克珠利也表现为高度敏感,因此,柔嫩艾美耳球虫对地克珠利与拉沙里菌素、地克珠利与盐霉素、地克珠利与克球粉3组药物不存在交叉抗药性。  相似文献

The objective of this study was to identify and characterize species of Eimeria in broiler chickens using traditional morphological and pathological plus molecular (DNA amplification) diagnostic methodologies. Using a combination of those techniques it was possible to identify the presence of multiple circulating species in the flock as well as higher frequencies for some of them, especially Eimeria praecox and Eimeria maxima, which were identified in 100% of the flocks. The frequencies of the other species were Eimeria mitis and Eimeria necatrix (93.3%), Eimeria tenella (76,7%), Eimeria acervulina (56.7%) and Eimeria brunetti (16.7%). However using the lesion score, the most common species were E. maxima (46.7%), E. acervulina (30%), E. tenella (23.3%), and E. necatrix (10%). E. brunetti and E. praecox were not identified by using lesion score. DNA amplification had detection sensitivity for Eimeria species in the field samples of at least 20 oocysts. The implementation of DNA amplification as a routine diagnostic technique in aviaries can assist Eimeria population.  相似文献

The study was aimed to explore the sensitivity of Eimeria acervulina (E.acervulina) precocious strain to some common anticoccidials (diclazuril, toltrazuril, sulfamonomethoxine, dinitolmide, decoquinate, nicarbazin, maduramicin, salinomycin and oxytetracycline). The sensitivity of E.acervulina precocious strain to 9 kinds anticoccidials was assessed by using 4 indices: anticoccidial activity percentage (POAA), reduction of lesion scores (RLS), relative oocyst production (ROP) and anticoccidial index (ACI). The results of these sensitivity trials indicated that the four indices POAA, RLS, ROP and ACI of groups toltrazuril, sulfamonomethoxine, dinitolmide, decoquinate, nicarbazin, salinomycin and oxytetracycline were all negative; the ROP index of diclazuril group was positive, the other three indices were negative. The ROP and ACI indices of maduramicin group were positive, the other two indices were negative. The results showed that the Eimeria acervulina precocious strain was sensitive to diclazuril, toltrazuril, sulfamonomethoxine, dinitolmide, decoquinate, nicarbazin, salinomycin and oxytetracycline, it was partly sensitive to maduramicin.  相似文献

抗球虫药在鸡胚球虫感染中活性峰期测定方法的研究   总被引:2,自引:2,他引:0  
用鸡胚感染柔嫩艾美耳球虫子孢子作为抗球虫药活性峰期测定的模型。每一鸡胚在接种5000个子孢子后,分别于感染的当天、第2天、第3天和第4天注入测试药物,以不同注药时间对鸡胚卵囊所产生的抑制率的差异,判定药物的活性峰期。结果表明,氯甲吡啶酚和丁氧喹啉的活性峰期是球虫生活史的第1天,卵囊抑制率分别为100%和98%;莫能菌素、盐霉素、球R和加福的作用峰期是第2天,卵囊抑制率均为100%;氨丙啉的峰期是第3天,卵囊抑制率为100%;尼卡巴嗪的峰期是第4天,卵囊抑制率为100%;速丹则在球虫生活史的整个无性阶段均呈现活性作用。加福+氨丙体+乙氧乙胺苯甲酯、球R+氨丙啉+乙氧乙胺苯甲酯、加福+速丹、加福+尼卡巴嗪及加福+乙氧乙胺苯甲酯等合剂亦在球虫生活史的整个无性阶段发挥高峰作用,其印囊的抑制率均为100%;尼卡巴嗪+氨丙啉合剂则在球虫生活史的第2~4天发挥最大作用,卵囊抑制率为100%。作者认为,本方法具有操作简单,结果可信及成本低廉的特点,可作为抗球虫药的筛选方法。  相似文献

Pamaquine and primaquine, the well known antimalarial 8-aminoquinolines, have not been reported for their anticoccidial activity. A series of battery experiments was conducted to investigate their activity against a laboratory strain of Eimeria tenella, E. necatrix, E. acervulina, E. maxima, or E. brunetti and revealed that both drugs were effective against E. tenella and E. necatrix, but not against the other three species. Pamaquine suppressed the symptoms of E. tenella induced coccidiosis at concentrations above 125 ppm in feed and primaquine controlled the clinical signs as well at levels above 31.2 ppm. The activity against E. necatrix was observed with pamaquine at 250 ppm and with primaquine at levels above 125 ppm. Pamaquine showed a tendency apparently to reduce body weight gain at 125-500 ppm, whereas primaquine showed the same tendency at 500 ppm. In a concomitantly conducted experiment, this adverse effect of pamaquine was averted in its molecular complexes with benzophenone, nitropyrazole, dinitrobenzoic acids and quinoline, and in its salts of sulfate or zinc chloride, and yet these compounds retained the same anticoccidial activity as of pamaquine. This suggests that these compounds had the broadened safety margin. Judging from their susceptibility to these compounds. E. tenella and E. necatrix will have similar metabolic functions to those of blood cell parasitizing protozoa like plasmodia and prioplasma, which are easily suppressed by this class of compound.  相似文献

Drug-sensitivity of 99 isolates of coccidia from broiler farms   总被引:7,自引:0,他引:7  
Coccidia were isolated from 99 broiler farms in 12 broiler-producing states and tested for sensitivity to contemporary anticoccidial drugs. The isolates usually comprised two or more species, including 92 Eimeria acervulina, 47 E. maxima, 28 E. tenella, and 10 E. brunetti. Based on intestinal-lesion-score reduction of 30% or more, 38% of isolates were resistant to monensin (110 ppm), 29% to salinomycin (60 ppm), 20% to nicarbazin (125 ppm), and 46% to amprolium + ethopabate (125 + 4 ppm). Isolates with 50% or greater reduction of lesion scores were considered sensitive. Thirty-three percent were sensitive to monensin, 53% to salinomycin, 67% to nicarbazin, and 39% to amprolium + ethopabate. Other isolates had intermediate response to drugs and were considered to have "reduced sensitivity." There was some evidence for incomplete cross resistance to the polyether, ionophorous products.  相似文献

Twenty European Eimeria spp. field isolates were subjected to an anticoccidial sensitivity test (AST). The anticoccidial drugs tested were diclazuril (Clinacox) and monensin (Elancoban). The assay was performed in a battery cage trial. Infected medicated birds were compared with an unmedicated control group. Coccidial lesion scores and oocyst shedding were used as parameters. The results of the AST show that resistance is common amongst coccidiosis field isolates, especially Eimeria acervulina (68% and 53% resistance for diclazuril and monensin, respectively). Resistance is less frequent amongst Eimeria maxima (38% and 50% resistance for diclazuril and monensin, respectively) and Eimeria tenella isolates (23% and 38% resistance for diclazuril and monensin, respectively). A highly significant influence of the coccidiosis prevention program (live coccidiosis vaccination with Paracox-5 vs. anticoccidial drugs in feed) on the sensitivity patterns of Eimeria spp. field isolates for both diclazuril (P= 0.000) and monensin (P= 0.001) was found. Further, when looking at the single species and each anticoccidial drug level, significantly more sensitivity of E. acervulina for monensin (P= 0.018), E. maxima for diclazuril (P = 0.009), and E. tenella for diclazuril (P = 0.007) was found in isolates originating from vaccinated flocks. Moreover, for E. acervulina and diclazuril, E. maxima and monensin, and E. tenella and monensin a trend toward higher sensitivity of isolates for these products was found when live coccidiosis vaccination was applied. The present study shows that sensitivity for the anticoccidial drugs diclazuril and monensin is more frequent in Eimeria spp. field isolates originating from broiler farms where a coccidiosis vaccination policy is followed.  相似文献

To determine whether chemotherapeutic compounds available for use in domestic poultry are effective at controlling coccidiosis in northern bobwhites (Colinus virginianus), we tested 13 chemotherapeutic anticoccidials including amprolium (250 parts per million [ppm]), clopidol (125 ppm), diclazuril (1 ppm and 2 ppm), decoquinate (30 ppm), lasalocid (120 ppm), monensin (90 ppm), narasin/nicarbazin (36/36 ppm), robenidine (33 ppm), roxarsone (50 ppm), sulfadimethoxine/ ormetoprin (125/75 ppm), salinomycin (60 ppm), semduramicin (25 ppm), and zoalene (125 ppm and 150 ppm). Three tests were conducted using two replicates of 10 birds each: Infected, unmedicated controls and medicated birds were challenged with 1 x 10(6) oocysts of a field isolate consisting primarily of Eimeria lettyae. Subsequently, we tested clopidol, lasalocid, salinomycin, diclazuril (1 ppm), and monensin against mixed-species field isolates containing E. lettyae, E. dispersa, E. colini, or all. Weight gain, gross intestinal lesions, severity of diarrhea, and feed conversion ratio (FCR) 6 days postinfection were recorded. Lesion score, as previously reported, was unreliable as a measure of severity of infection in comparison with weight gain, fecal scores, and FCR. Excellent to good efficacy was found in clopidol, decoquinate, diclazuril (1 ppm and 2 ppm), and in lasalocid, narasin and nicarbazin, robenidine, sulfadimethoxine/ormetoprin, and zoalene (150 ppm). Marginal protection was found using monensin, salinomycin, semduramicin, or a roxarsone/semduramicin combination. Amprolium, roxarsone, and zoalene (125 ppm) were ineffective at controlling coccidia. Two of the six isolates tested against diclazuril 1 ppm and clopidol demonstrated a high degree of resistance, but none of the six isolates was resistant to lasalocid. Four of the eight isolates showed mild to moderate, and moderate to high, resistance against monensin and salinomycin, respectively. These findings indicate that several available compounds are effective at controlling coccidiosis in bobwhites.  相似文献

Four isolates of Eimeria tenella obtained from the field were partially resistant to monensin. This resistance was not lost after 10 passages in unmedicated chickens, indicating that it was stable. A previously described multiplex PCR was evaluated for the identification and prevalence of Eimeria species in market-age commercial chicken flocks in Ontario. The multiplex PCR based on species-specific RAPD-SCAR markers was able to distinguish six available laboratory strains of Eimeria species (E. tenella, E. maxima, E. necatrix, E. mitis, E. acervulina, and E. brunetti) and E. tenella, E. maxima and E. acervulina in unknown field samples, including multiple infections in single reactions. No backyard (0/77) and 20/360 market-age commercial chickens were oocyst-positive using standard fecal flotation methods. PCR identified E. tenella alone (9/360, 2.5%), E. maxima alone (5/360, 1.38%), E. maxima plus E. tenella (5/360, 1.38%) and E. acervulina alone (1/360, 0.27%) in market-age commercial broilers. This is probably the first time the multiplex PCR has been evaluated in poultry establishments in Canada and illustrates the value of the tool in coccidiosis epidemiology on commercial farms.  相似文献

The objective of this study was to confirm the presence of seven species of Eimeria involved in chicken coccidiosis in Australia by comparing internal transcribed spacer 1 (ITS-1) sequences, ITS-1 polymerase chain reaction (PCR) methods and to apply phylogenetic analysis to assess evolutionary relationships of Australian isolates. Twenty-two distinct ITS-1 regions of 15 Australian Eimeria isolates were sequenced, and analysed using maximum parsimony, distance and maximum likelihood methods. Poor bootstrap support, resulting from high ITS-1 sequence heterogeneity between all species groups, resulted in polychotomy of the Eimeria species in all three trees generated by these analyses. Percentage identity analyses revealed two distant ITS-1 lineages in both E. mitis and E. maxima at the same levels that separate the two species E. tenella and E. necatrix. One E. maxima lineage consisted of Australian isolates, the other American isolates, with one European sequence (originating from the same isolate) in each lineage. One Australian E. praecox sequence was only distantly related (33% variation) to three E. praecox sequences from Australian and European isolates. Short and long ITS-1 variants were isolated from both E. tenella (cloned line) and E. necatrix isolates with deletions (106 and 73 bp, respectively) in the short variants within the 3' region of the ITS-1 sequence. ITS-1 sequences of strains of both E. brunetti and E. acervulina species varied the least. Apart from E. maxima, all of the ITS-1 sequences of the six remaining individual species clustered to the exclusion of other species in all phylogenetic trees. Published ITS-1 tests for E. necatrix, E. acervulina, E. brunetti and E. tenella, combined with three new tests for E. mitis, E. praecox and Australian E. maxima amplified all respective Australian isolates specifically in a nested format using conserved ITS-1 PCR products as template to improve the sensitivity. All PCR tests were confirmed against a collection of 24 Australian chicken Eimeria isolates and contaminating species were detected in some instances. In conclusion, once the genetic variation between species and strains is determined, the ITS-1 is a good target for the development of species-specific assays, but the ITS-1 sequences alone do not seem suitable for the confirmation of phylogenetic inferences for these species. This study reports the first attempt at the analysis of the phylogeny and sequence comparison of the Eimeria species involved in chicken coccidiosis in Australia.  相似文献

The anticoccidial activity of maduramicin ammonium (5 mg/kg food) administered alone or with roxarsone (50 mg/kg food) and/or avoparcin (10 mg/kg food) was evaluated in battery-reared broilers artificially challenged with recent field culture mixtures containing Eimeria acervulina, E. maxima and E. tenella or E. mivati, E. necatrix and E. brunetti. 2. Maduramicin ammonium exhibited a high degree of anticoccidial activity and the addition of roxarsone and/or avoparcin in food at recommended concentrations did not adversely affect the activity.  相似文献

In this study, nine anticoccidial drugs commonly used in poultry were tested for efficacy for the prevention and treatment of Goussia carpelli (Apicomplexa) infection in common carp (Cyprinus carpio L.). To establish experimental infection with G. carpelli, paratenic host oligochaetes of the genera Tubifex and Limnodrilus were infected with oocysts, and laboratory-cultured parasite-free common carp fingerlings were infected by feeding to them oligochaetes containing sporozoites. The anticoccidial drugs (amprolium, narasin, maduramicin, salinomycin Na, lasalocid Na, diclazuril, robenidine HCl, monensin Na and toltrazuril), mixed in the food of the fish in a dose of 200 mg/kg, were fed for 12 days. Common carp fingerlings fed diclazuril, lasalocid, robenidine HCl or maduramicin and killed on day 14 after exposure were free from infection, while other groups treated with amprolium, toltrazuril, monensin Na, narasin or salinomycin Na harboured oocysts in the mucus and epithelium of the gut.  相似文献
