Phytophthora infestans was isolated from potato leaves and tubers collected from different parts of Finland in 1990–96 and Norway in 1993–96. Isolates were assessed for mating type, resistance to metalaxyl and virulence phenotype. In Finland 15% of 200 isolates tested and in Norway 25% of 642 tested were A2 mating type. Differences in the A1/A2 ratio were evident among regions and A2 was not found in northern areas. In Finland the frequency of metalaxyl-resistant isolates, among 1834 tested, decreased from 59% in 1990–95 to 2% in 1996. In Norway 59% of 491 isolates tested were resistant to metalaxyl in 1996. Among 269 Finnish and 105 Norwegian isolates, all known virulence genes were found in both countries and race 1.3.4.7.10.11 was the most common (resistance gene R9 was not included in the differential set). Oospores were observed in potato leaves from three locations in the southern part of Norway. The implications of the 'new' populations in the Nordic countries are discussed.  相似文献

Disease observations and amplified fragment length polymorphism (AFLP) markers were used to study recent developments in the Puccinia striiformis f.sp. tritici population in Denmark. The fungus appeared spontaneously at 10 locations in Denmark in 1997 after it was not observed under natural conditions in 1996. The pattern of disease development and prevailing winds suggested that the fungus reappeared by airborne spores from the south or west. In 1998, disease incidence was more evenly distributed throughout the country. Forty-eight single lesion isolates were collected from most crops where the disease was observed in these years; all except one from 1997 belonged to two pathotypes that were not previously detected in the country, and both possessed the newly discovered Yr17 virulence. The isolates were characterized with AFLP markers together with 28 isolates representing eight of 13 pathotypes observed prior to 1996. Initial screening of 240 Pst I/ Mse I AFLP primer combinations on four isolates showed that a primer combination, on average, revealed 0·4 polymorphisms between any isolate pair. A selection of 21 primer combinations resulted in 28 AFLP markers, which revealed 16 AFLP phenotypes among all 76 isolates. The two Yr17- virulent pathotypes consisted of three AFLP phenotypes, which were observed in both 1997 and 1998; the two most frequent AFLP phenotypes occurred at most sampling locations and often within the same crop. AFLP diversity was larger among samples collected prior to 1996, and also in this period most AFLP phenotypes were observed at different sampling locations. These results are consistent with the features of an entirely asexually reproducing pathogen dispersed by aerial spores across large areas.  相似文献

This study was conducted on potato late blight samples collected for monitoring early attacks in Finland from 1997 to 2000. Most of the 1726 Phytophthora infestans isolates were collected soon after the onset of the epidemics. Most of the isolates were tested for mating type as well as metalaxyl and propamocarb hydrochloride (HCl) sensitivity, while virulence on potato R-gene differentials and mitochondrial DNA (mtDNA) haplotype were determined for a subset of the isolates. In half of the fields from which more than one isolate was tested, both mating types were detected, indicating strong potential for sexual reproduction. The mating types coexisted more often in organic fields or gardens than in conventional fields. The proportion of A1 mating type decreased from approximately 80% (1997 and 1998) to 22% by the year 2000. Simultaneously, the proportion of isolates resistant to metalaxyl decreased from approximately 40% to 16%. Resistance to metalaxyl was confined to the IIa mtDNA haplotype and clearly associated with the A1 mating type, as resistance was 10 times more common among A1 isolates than among A2 isolates. Resistance to metalaxyl therefore probably derives from common descent from an isolate with the IIa haplotype. Most of the regional variation in metalaxyl sensitivity was also linked to mating type, as both metalaxyl resistance and the A1 mating type were most common in the north and south-west of Finland. In contrast to metalaxyl, only three propamocarb-HCl-insensitive isolates were found, but propamocarb-HCl sensitivity decreased significantly during the study.  相似文献

Barley leaf rust caused by Puccinia hordei is an important disease of barley in Ethiopia. In the 2003 and 2004 cropping seasons, surveys of P. hordei were conducted on fields in the main rainy, residual moisture and short rainy season-barley production systems. A total of 381 isolates were analysed on 12 barley differential hosts carrying different Rph resistance genes ( Rph1 – Rph12 ). Based on infection phenotypes on leaf rust ( Rph ) resistance genes, seven pathotypes were identified, namely ETPh7611, ETPh7631, ETPh6611, ETPh7651, ETPh7671, ETPh7653 and ETPh7633, with frequencies of 63·0, 21·5, 6·8, 2·9, 2·6, 2·1 and 1·2%, respectively. ETPh7611 and ETPh7631 were the most common pathotypes found in all the surveyed areas of the three production systems. ETPh7653 was found in the small rainy season production system only. ETPh7671, ETPh7653 and ETPh7633 were the most virulent, but the least frequent, pathotypes. All isolates were virulent on resistance genes Rph1 , Rph4 , Rph8 , Rph9 , Rph11 and Rph12 . Virulence against Rph3 and Rph7 was absent. Genes Rph2 , Rph6+2 , Rph5 and Rph10 were effective against 96·3, 88·9, 65 and 2·4% of the rust isolates tested, respectively.  相似文献

Ninety-five isolates ofPhytophthora infestans collected throughout Poland during 1985–1991 and characterized for multilocus genotypes based on mating type, allozymes and DNA fingerprint, were analyzed for specific virulence to differential potato cultivars carrying ten major resistance genes. The multilocus analysis led to three groupings. The first group contained 22 isolates of a clonal lineage (PO-1) that is postulated to have been present in Europe during most of the twentieth century, but PO-1 isolates were recovered in Poland only during 1985–1988. This group contained, on average, virulence to 5.5 specific resistance genes per isolate. The second group consisted of 30 isolates in a clonal lineage (PO-4) that had not been detected before 1988. PO-4 isolates had virulence to a mean of 6.5 resistance genes per isolate. The third group was composed of 43 isolates representing 38 multilocus genotypes also not detected before 1988. These diverse genotypes had virulence to an average of 6.7 specific resistance genes per isolate. More than half (53%) of the PO-4 isolates shared a single pathotype. The group of 43 isolates was dominated by two pathotypes: the most common one (47% of the isolates) was the same pathotype that dominated PO-4 isolates; the next most common one (21%) differed from the most common one by the absence of virulence to resistance gene R5. The recent immigrant isolates (not detected before 1988) generally had virulence to a greater number of specific resistance genes than did isolates in the previous population [detected before 1988 (PO-1)]. Recent immigrant populations were dominated by one or two pathotypes, so their pathotypic diversity values were somewhat less than that of the previous population.  相似文献

A collection of 101 isolates of Phytophthora infestans, obtained from seven sampling sites representing central, east and south-east Estonia during 2002 and 2003 were assessed for several phenotypic and genotypic markers. All 101 isolates were assessed for virulence and resistance to metalaxyl. Virulence to each of the 11 classic resistance genes was found among the tested isolates. The mean number of virulences per isolate was 6.3, with a very low frequency of virulence against resistance genes R5 (5%) and R9 (14%). The most common pathotypes were 1.3.4.7.8.10.11 and 1.3.4.7.10.11, representing altogether 12% of the studied strains. In terms of metalaxyl resistance, 30 resistant, 52 intermediate and 19 sensitive isolates were found. A subgroup of 50 isolates was assessed for mating type, allozymes [glucose-6-phosphate isomerase (Gpi) and peptidase (Pep)], DNA fingerprints with probe RG57 and mtDNA haplotype. Of this subset, 30 were A1 and 20 were A2. Collections from three of the seven fields contained both mating types. Allozyme analysis did not reveal any polymorphism. However, 19 diverse RG57 fingerprints were detected, and two mitochondrial DNA haplotypes, Ia and IIa, were detected. By combining the mating type, mtDNA haplotype and RG57 fingerprint data, 26 multilocus genotypes were identified, of which 18 were detected only once. Genotypic diversity measured by the normalised Shannon diversity index was high (0.76). The large number of multilocus genotypes and the presence of both mating types in some fields indicate that sexual reproduction may take place in Estonian populations of P. infestans.  相似文献

 2003~2005年,从云南省德宏州新兴冬作马铃薯主产区采集的马铃薯晚疫病病样中分离纯化到86株致病疫霉菌,对其代表性菌株的表型特征进行了较系统的研究。交配型实验结果表明:所测定的62个菌株都属于A1交配型,未发现A2交配型或自育型菌株。毒性基因鉴定、监测及生理小种组成分析显示:德宏州存在所有已知可鉴定的毒性基因,而且生理小种组成非常复杂;所测定的66个菌株可区分出36个生理小种,其中,出现频率最高的小种为1,2,3,4,5,6,7,8,9,10,11,占被测菌株的18.18%,在主产区潞西县和盈江县均有分布。对60个菌株的甲霜灵抗性测定结果表明:56.67%的菌株表现为敏感,25.00%的菌株中抗,18.33%的菌株对甲霜灵有抗性。本文对此实验研究结果作了初步的分析和讨论。  相似文献

A large collection of German rye leaf rust isolates was analysed to characterize the diversity, spatial variation and temporal dynamics of virulences. Virulence-avirulence phenotypes (=pathotypes) were determined on 23 host differentials. We found 93 pathotypes among 177 single-uredinial isolates in 2000, 201 pathotypes among 437 isolates in 2001, and 125 pathotypes among 213 isolates in 2002. In total, the 827 analyzed isolates represented 317 pathotypes. Frequency of virulences on the individual differentials varied from 2% to 97%. Eight of the differentials showed a high resistance level with virulence frequencies <10%. Virulence complexity of the isolates ranged from 3 to 21 with a mean of nine. The percentages of highly virulent isolates (>14 virulences) increased from 4 to 15% during the sampling period. A high level of virulence diversity was observed within and between individual sampling sites with Simpson indices around 0.9. Evenness indices ranged from 0.88 to 0.92. Four of the five most frequent pathotypes were found in each year but their frequency never exceeded 10%. Isolates with unusual virulence combinations could be clearly separated by principal component analysis. Location-specific pathotype frequencies were revealed in each year, but the frequency patterns varied across years. On four fields a considerable increase of highly virulent pathotypes occurred within 6 weeks during the epidemic. The high diversity of pathotypes as well as the fast accumulation of highly virulent pathotypes favour the adaptation of the pathogen to race-specific host resistances. More durable resistance might be achievable by combining new effective race-specific resistances with adult-plant and/or race-non-specific quantitative resistances.  相似文献

采用国际通用的小麦叶锈菌鉴别寄主和辅助鉴别寄主分析了来自1999年我国不同地区小麦叶锈菌的毒性基因,479个叶锈菌株共划分为162个毒性类型,其中23个为主要毒性类型.毒性类型中出现频率最高的为FHB、PHT、FHG、THT,它们对抗叶锈基因Lr2a、Lr2b、Lr3、Lr10、Lr14b、Lr16、 Lr26的平均毒性频率高于80%,而对Lr3ka、Lr25、Lr19、Lr24、Lr30、Lr15、Lr35的平均毒性频率低于30%;发现对Lr35有毒力的菌株,出现频率为1.04%;至今尚未发现对Lr38、Lr45抗性基因有毒力的菌株.研究同时发现,不同地区小麦叶锈菌的毒性类型不同,毒性频率存在一定的差异.Lr9、Lr15、Lr19、Lr24、Lr35、Lr38、Lr45为小麦抗叶锈育种可利用的有效抗病基因.  相似文献

Since its initial detection in Australia in 1979, wheat yellow (stripe) rust ( Puccinia striiformis f.sp. tritici ) has evolved in Australia and New Zealand into more than 20 pathotypes with assorted virulence characteristics. This evolution is believed to have occurred in a stepwise fashion from an original single pathotype, with no subsequent new introductions. A combination of random amplified polymorphic DNA (RAPDs) and amplified fragment length polymorphisms (AFLPs) was used to examine the level of molecular variation in Australian and New Zealand isolates, and to compare this with variation amongst other isolates of P. striiformis . Using 60 RAPD primers on seven Australian isolates representing seven different pathotypes collected between 1979 and 1991, more than 300 potentially polymorphic loci were analysed and no polymorphisms were detected. Using the same primers on two UK isolates, 3% of loci showed a polymorphism. A similar level of polymorphism was found between UK isolates using AFLP primers, and between 5 and 15% of fragments were polymorphic between an isolate from the UK, an isolate from Denmark, and one from Colombia. However, no AFLP polymorphisms were found amongst 14 Australian and New Zealand isolates tested, at over 100 potentially polymorphic loci. The lack of molecular variation in the Australian and New Zealand collection is consistent with the stepwise mutation theory of pathotype evolution from a single introduction.  相似文献

The genetic and virulence variability of 112 isolates of Phaeoisariopsis griseola , collected from various locations in Central America, were studied using seven random amplified polymorphic DNA (RAPD) primers and 12 common-bean differential genotypes. Broad molecular diversity ( H  = 0·92) among isolates was found using RAPD markers. Fifty pathotypes were identified on 12 differential bean genotypes, 29 of which were represented by only one isolate. Only 18 pathotypes were found in two or more countries. Pathotype 63-63 was the most virulent and caused leaf spots on all 12 common-bean differential genotypes. Comparison of virulence phenotypes and RAPD profiles to known Andean P. griseola isolates confirmed that all isolates belonged to the Mesoamerican group. Pairwise comparison between individual RAPD loci showed that the majority were in gametic phase linkage disequilibrium, revealing that P. griseola maintains a genetic structure that is consistent with asexual reproduction. The molecular and virulence diversities of P. griseola isolates from Central America imply that using single resistance genes to manage angular leaf spot is inadequate and stacking resistance genes may be necessary to manage the disease effectively.  相似文献

ABSTRACT The population genetic structure of Phytophthora infestans in Ecuador was assessed from 101 isolates collected from 1990 to 1992 and 111 isolates collected in 1993. All isolates were analyzed for mating type and allozyme genotype. Both samples were dominated (>95%) by a clonal lineage (EC-1) defined from neutral markers: 90/100 genotype for glucose-6-phosphate isomerase, 96/100 genotype for peptidase, A1 mating type, and a previously unreported nuclear DNA fingerprint. The remaining isolates belonged to the US-1 clonal lineage, which has a worldwide distribution. Isolates in the 1993 sample were analyzed for virulence and metalaxyl sensitivity. All representatives of EC-1 had complex patho-types, with three pathotypes representing >60% of the collection. There was variation for metalaxyl sensitivity. There was no evidence for geographical substructuring on the basis of neutral markers, but there was evidence for limited substructuring based on metalaxyl sensitivity and specific virulence. We hypothesize that EC-1 has been recently introduced to Ecuador.  相似文献

The pathogenic variability of the barley scald fungus, Rhynchosporium secalis , in central Norway was examined in 1994. The climate in this region is usually cold and wet during the growing season of spring barley. Leaf blotch is prevalent and causes significant yield losses. Forty-two isolates of the fungus, from naturally infected spring barley in four counties, were differentiated into 32 pathotypes by the standard differential set for R. secalis . All pathotypes were complex and had virulence for nine to 22 differentials. The cultivar Osiris was resistant to all isolates tested. The cultivars C.I.8162, Hudson, Atlas 46 and C.I.3515 were resistant to the majority of the isolates. Several differentials with various resistance genes were susceptible to up to 100% of the isolates. Isolates were derived from local cultivars with no known resistance genes, suggesting that R. secalis populations in central Norway are characterized by a high degree of seemingly unnecessary pathogenicity. Because of the great variability and complexity of the pathotypes, traditional breeding methods using single major genes are not likely to be effective in central Norway.  相似文献

In this study, the Estonian population of Phytophthora infestans was characterized with mating type, sensitivity to metalaxyl, virulence on 11 potato R-gene differentials and 12 SSR markers to show the outcome of potential sexual reproduction in the population. During the three years 2010–2012, 141 P. infestans isolates, collected from 23 potato fields, showed quite a high and stable frequency of the A2 mating type, 48% of the total population. In 87% of all sampled potato fields, both mating types were recorded, suggesting continuous sexual reproduction of P. infestans and possible oospore production. Metalaxyl-sensitive isolates prevailed in all three years (68 out of 99 isolates). Amongst the 95 isolates tested, 51 virulence races were found. The race structure was diverse, and most pathotypes were unique, appearing only once; the two most common pathotypes, 1.2.3.4.6.7.10.11 and 1.2.3.4.7.10.11, comprised 35% of the population. The P. infestans population was genetically highly diverse and most of the multilocus genotypes (MLGs) appeared only once. Furthermore, all of the MLGs appeared in only one of the three sampling years. Our results confirm that the high diversity in the Estonian P. infestans population is most likely the result of frequent sexual reproduction, which benefits the survival, adaptability and diversity of the pathogen in the climate of North-Eastern Europe.  相似文献

A 6·7-kb DNA fragment has been isolated from an R-type isolate of Pseudocercosporella herpotrichoides that exhibits specific hybridization to R-type isolates and not to W, C or S pathotypes or to P. anguioides. R-type isolates are polymorphic with respect to this fragment and three sizes were present (11·6, 6·7 and 4·5 kb) in Eco RI-digested DNA from isolates of a world-wide collection. Infection of rye seedlings was demonstrated by hybridization of this probe to DNA extracted from infected plants.  相似文献

Virulence analysis of 224 isolates of Blumeria graminis f. sp. hordei (barley powdery mildew) from South Africa was performed. The isolates were collected from eight fields and a greenhouse in 2004 and 2007. The isolates were tested for virulence on a set of 20 differential varieties. All isolates were virulent on the resistance genes Mla8 and Ml(Ch) and avirulent for the resistance genes Mla3, Mla6, Mla7, Mla9, Mla13, Mla23, Mlp1 and MlaN81. Virulence frequencies of field isolates for the resistance genes Mla12 + MlaEm2, Mlat, Mla22, Mlk1 and Mlh were 52.9–99.5 % and for Mla1 + MlaAl2, MlLa, Mlra, Mlg + MlCP and Ml(Ru2) were 0.5–23.5 %. In total, 46 pathotypes were detected in the field and seven other pathotypes in the greenhouse. Only nine pathotypes were found in both years, but they included 61.8 % of the isolates. The predominant pathotype represented 15.9 % of the isolates, and was the only one common to all three field populations. The average relative virulence complexity per field isolate increased from 0.405 in 2004 to 0.486 in 2007. Two powdery mildew metapopulations in geographically distant and separated areas (North West and Western Cape) were deduced. The South African population of Blumeria graminis f. sp. hordei had unique virulence frequencies and virulence associations when compared to populations from other parts of the world.  相似文献

The pathogenic variability of Aphanomyces euteiches on pea was investigated using a collection of 88 pea-infecting isolates from France and 21 isolates from Denmark, Sweden, Norway, USA, Canada and New Zealand. Aggressiveness and virulence were assessed by scoring the root symptoms on a differential set of six pea genotypes. Eleven virulence types were characterised. The virulence type I, previously described as virulent on the whole set, was predominant and included the most aggressive isolates of all geographical origins. The other types were much less prevalent, existing as one to five isolates. Three virulence types (III, IV and V) contained no French isolates. The type III, avirulent on MN313, was composed of American isolates only, and resembled the major group recently described in the USA. A wide range of aggressiveness was found within the virulence type I, and the French isolates appeared globally more aggressive than the foreign isolates. These findings indicate that isolates from the virulence type I should be used as references in breeding programs, and that pea lines PI180693 and 552 may be the most interesting resistance sources to date, despite their only partial resistance.  相似文献

Potato fields in Denmark, Finland, Norway and Sweden were sampled for single-lesion isolates of Phytophthora infestans . The aggressiveness of the isolates was determined on detached leaflets of potato cvs Bintje (susceptible) and Matilda (moderately resistant). The aggressiveness tests were carried out in the respective home countries of the isolates, with the exception of the Danish isolates. Fifteen Danish isolates were studied in each of the other three countries, including five isolates tested in all three laboratories. Results obtained from the Danish isolates revealed substantial differences between the test laboratories for infection efficiency, lesion growth rate and sporulation capacity on detached leaflets. When the laboratory effect was taken into account, the differences in aggressiveness between the countries were generally small or inconsistent between the test cultivars and epidemiologically insignificant. By contrast, variation among isolates within countries was substantial. The magnitude of the variation depended on country and cultivar. Maximal variation for the means of the isolates was between 89 and 185 h for latent period, between 100 and 1297 sporangia mm−2 for sporulation capacity and between nearly zero and 6 mm day−1 for lesion growth rate. Typically less than 1% of sporangia were able to cause infections, except in Norway. These extraordinarily low values may be an artefact of the testing method. High variation in results between the test laboratories emphasizes the need for caution when comparing results obtained by different research groups.  相似文献

ABSTRACT Severe Verticillium dahliae attacks have occurred in artichoke crops in the Comunidad Valenciana region of eastern-central Spain since the late 1990s. Knowledge of genetic and virulence diversity in the pathogen population is a key factor for the management of the disease through disease risk assessment as well as development and use of resistant cultivars. V. dahliae isolates from artichoke (109 isolates) and cotton (three isolates) in that region were characterized by vegetative compatibility grouping (VCG), and specific polymerase chain reaction assays using three sets of primer pairs that differentiate the cotton-defoliating (D) and -nondefoliating (ND) V. dahliae pathotypes. In all, 35 and 39 V. dahliae isolates representative of the identified VCGs and geographic origins were tested for virulence to artichoke cvs. Nun 6374 and Nun 9444, and cotton cv. Acala SJ-2, respectively. Four VCGs were identified among 107 artichoke isolates, and 2 isolates were heterokaryon self-incompatible: VCG1A (one isolate), VCG2A (31 isolates), VCG2B (72 isolates), and VCG4B (three isolates). The three cotton isolates were VCG1A. Isolates in VCG2B were distributed across the region and were the most prevalent isolates in the northern part. Conversely, 83.9% of isolates in VCG2A were recovered from the southern part of the region. Two subgroups of isolates were identified in VCG2B based on heterokaryon compatibility with either international or local tester isolates, which further showed diversity in the amplification of 334- and 824-bp DNA fragments which are markers of the D and ND pathotypes, respectively. Virulence of isolates to artichoke and cotton correlated with VCG but the pattern of correlation varied with the host. VCG1A isolates from artichoke and cotton induced defoliation in cotton but not in artichoke. Collectively, isolates of VCG2B and VCG4B were the most virulent and isolates of VCG1A or HSI were the least virulent to artichoke; but isolates of VCG1A were more virulent to cotton than those of any other VCG. Also, molecular subgrouping in VCG2B determined by amplification of the 334- and 824-bp markers correlated with virulence of isolates to the two hosts tested.  相似文献
