Torque teno viruses (TTVs) are vertebrate infecting, small viruses with circular single stranded DNA, classified in the Anelloviridae family. In pigs, two different TTV species have been described so far, Torque teno sus virus 1 (TTSuV1) and 2 (TTSuV2). TTSuVs have lately been linked to postweaning multisystemic wasting syndrome (PMWS). In the present study, TTSuV1 and TTSuV2 prevalence and DNA loads in longitudinally collected serum samples of healthy and PMWS affected pigs from Spanish conventional, multi-site farms were analyzed. Serum samples were taken at 1, 3, 7, 11 and around 15 weeks of age (age of PMWS outbreak) and viral DNA loads determined by quantitative PCR. For both TTSuV species, percentage of viremic pigs increased progressively over time, with the highest prevalence in animals of about 15 weeks of age. TTSuV1 and TTSuV2 viral DNA loads in healthy and TTSuV1 loads in PMWS affected animals increased until 11 weeks of age declining afterwards. On the contrary, TTSuV2 DNA loads in PMWS affected pigs increased throughout the sampling period. It seems that TTSuV species differ in the in vivo infection dynamics in PMWS affected animals.  相似文献

Anelloviruses are small, non-enveloped viruses with circular single stranded DNA, which infect a number of animal species as well as humans. In pigs, two distinct Torque teno sus virus (TTSuV) species have been described so far, being one of them linked to disease occurrence. Specifically, TTSuV2 loads in serum have been found increased in pigs suffering from postweaning multisystemic wasting syndrome (PMWS). Since this pathological condition is able to be controlled by means of porcine circovirus type 2 (PCV2) vaccination, it was hypothesized the possibility that such vaccination would have an impact on TTSuV2 prevalence and loads. A total of 150 pigs were divided in two study groups. Half of them received a PCV2 commercial vaccine, while the other half remained as non-vaccinated controls. PCV2 infection was monitored at 3-4, 8, 12, 16 and 21 weeks of age by means of an standard PCR, while TTSuV2 loads were determined at 8, 16 and 21 weeks of age by a quantitative PCR. No obvious PMWS clinical signs were observed among studied animals, although PCV2 infection was confirmed in both groups of pigs. Almost all pigs got TTSuV2 infection throughout the study period, independently of the PCV2 vaccination status of animals. Moreover, TTSuV2 load did not show significant differences between different pig groups at each sampling time, but mean viral load increased with age. Taking into account that previous results suggest that TTSuV2 load in serum is increased in the background of PMWS, the present study suggests that this is not the case in a PCV2 subclinical infection scenario. Therefore, vaccination of PCV2 subclinically infected pigs did not modify the outcome of TTSuV2 infection.  相似文献

We investigated the seroprevalence rate of Torque teno sus virus types 1 (TTSuV1) and 2 (TTSuV2) in the sera of 38 post weaning multisystemic wasting syndrome (PMWS)-suspected pigs and 43 porcine circovirus type 2 (PCV2)-vaccinated normal pigs on 3 commercial pig farms in southern Japan by using nested polymerase chain reaction. High seroprevalence rate of TTSuVs was observed in both PMWS-suspected pigs (100%) and PCV2-vaccinated normal pigs (90.7%). The seroprevalence rate of TTSuV2 was significantly higher in the PMWS-suspected pigs than in PCV2-vaccinated normal pigs (97.4% versus 81.4%, P<0.05), whereas no such difference was observed for TTSuV1 between the 2 groups of pigs. In both pig groups, the seroprevalence rate of TTSuV2 was significantly higher than that of TTSuV1 (P<0.01-0.05). These results show that TTSuVs are highly seroprevalent in both PMWS-suspected pigs and PCV2-vaccinated normal pigs with TTSuV2 being more seroprevalent in the former than in the latter.  相似文献

A fluorescent microbead-based immunoassay (FMIA) for simultaneous detection of IgG antibodies against Torque teno sus virus 1 (TTSuV1), TTSuV2, porcine reproductive and respiratory syndrome virus type 1 (PRRSV-1) and PRRSV-2 was developed. Serum samples were obtained over time from 20 pigs. Twelve of 20 were exposed to TTSuV2 on day 0, 20/20 were vaccinated with a PRRSV-2 vaccine on day 35, and 20/20 were exposed to PRRSV-2 on day 63. Anti-TTSuV antibodies were detected in 30% of the pigs on day 0, and 90% by day 35. All PRRSV-2 vaccinated pigs had detectable anti-PRRSV-2 IgG 21 days after vaccination. Field samples from 17 farms were also tested. The seroprevalence of both PRRSV and TTSuV increased with age. Comparison of the PRRSV-2 FMIA to an ELISA revealed good correlation in young pigs but a high rate of false positives in older pigs. Cross-reaction between PRRSV types was a problem.  相似文献

The role of swine torque teno sus viruses (TTSuVs) as co-factors in disease syndromes involving porcine circovirus strain 2 (PCV2) and porcine reproductive and respiratory disease syndrome virus (PRRSV) has been a debatable subject. In this study, the prevalence of TTSuVs in Iowa, the leading pork producing state in the U.S., was estimated by a duplex PCR. The PCR is capable of simultaneously detecting both teno sus viruses 1 and 2 (TTSuV1 and 2). Based on an analysis of 300 random samples representing six major geographical regions of the state, the overall prevalence rates for TTSuV1 and 2 were 47.34% and 24.67% respectively while the combined prevalence rate was 52.33%. The epidemiological association of TTSuV1 and 2 with the common etiological agents of the porcine respiratory disease complex (PRDC) namely porcine PRRSV, PCV2, Mycoplasma hyopneumoniae and swine influenza virus (SIV) was estimated in lung tissue derived from 45 pigs showing clinical signs of PRDC. Notably, 86.67% of the PRDC-suspect samples were positive for TTSuV1 in comparison to the baseline population prevalence rate of 47.34%. However, the prevalence of TTSuV2 (26.67%) was not significantly different. TTSuV1 was detected in 80.00%, 81.81%, 75.00% and 77.78% of the PRRSV, SIV, M. hyopneumoniae and PCV2 positive PRDC-suspect samples respectively. Our results indicate that TTSuV1 is strongly associated with clinical PRDC and support the hypothesis that TTSuVs might function as co-factors in PRDC. Further studies to define their possible role in the pathogenesis of swine respiratory diseases are warranted.  相似文献

A field-based case-control study was conducted to assess the strength of association of porcine circovirus type 2 (PCV2) and some major swine viruses with postweaning multisystemic wasting syndrome (PMWS). Cases were defined as individual pigs with a clinical history of progressive weight loss and histopathological lesions characteristic of PMWS. Controls were pigs without clinical signs and histopathological lesions typical of PMWS. A total of 31 cases and 56 controls was identified from diagnostic submissions. Serum and various tissues were collected from all animals and assayed for PCV, porcine reproductive and respiratory syndrome virus (PRRSV), porcine parvovirus, porcine enterovirus types 1-3, swine influenza virus, porcine respiratory coronavirus, transmissible gastroenteritis virus, porcine endogenous retrovirus, porcine lymphotropic herpesvirus type 1, and bovine viral diarrhea virus. The proportion of case and control pigs positive for each virus was determined and statistically compared for determining the strength of the association that each virus had with PMWS individually or in combinations. Porcine circovirus type 2 had the strongest association (OR = 9.3, P = 0.006) with PMWS among the viruses tested for. Risk for PWMS was much higher (OR = 31.2, P = 0.0009) if the animal was concurrently infected with PCV2 and PRRSV, suggesting that development of PMWS may be enhanced by cofactor(s). Because PCV2 was also found in 62.5% of the controls, PCV2 from 5 cases and 4 controls were selected and genetically compared. No significant genetic difference was observed between PCV2 from PMWS and control pigs.  相似文献

Torque teno viruses (TTVs), of the genus Anellovirus, are single-stranded circular DNA viruses that infect many vertebrate species. Although viruses of this type have quite a stable genome, they exhibit low nucleotide homology. Torque teno virus infection has not been consistently linked to specific diseases, although there is epidemiological evidence of an association with disease in humans. The recent recognition of naturally occurring TTV infection in swine and its epidemiological resemblance to human TTV raises the possibility of using the pig as a model to study human TTV infection. Such an approach will require the development of novel investigative tools to study the epidemiology, transmission, immune responses and potential pathogenesis of TTV infection. The present review summarises research on animal TTV infection, focussing in particular on TTV infection in the pig, and considers how a porcine experimental infection model might assist in the study of human TTV infection.  相似文献

为了解吉林地区猪细环病毒(TTSuVs)与II型猪圆环病毒(PCV2)混合感染情况,分析TTSuVs感染与仔猪多系统衰竭征(PMWS)的相关性。试验通过PCR技术对2019年收集到的吉林部分地区的10家规模化猪场130份血清样本进行检测;并应用本实验已建立的PCV2、TTSuVs实时荧光定量PCR检测方法,对PCV2阳性样品进行病毒载量的分析,区分PMWS病猪与PCV2亚临床感染猪,比较PMWS病猪与PCV2亚临床感染猪中TTSuVs的载量。结果显示:TTSuV1感染率为38.46%;TTSuV2感染率为57.69%;PCV2感染率为46.15%;TTSuV1与TTSuV2的混合感染率为31.54%;TTSuV1与PCV2的混合感染率为32.30%;TTSuV2与PCV2的混合感染率为44.62%;三种病毒的混合感染率为22.30%。且PMWS病猪血清中TTSuV2载量明显高于PCV2亚临床感染猪(P<0.01)。结果表明,TTSuV2载量与PMWS病的发生存在一定程度的相关性。  相似文献

Fourteen diseased pigs from four farms in which there had been an outbreak of salmonellosis were investigated. Granulomatous inflammation with depletion of lymphocytes was observed in the swollen lymph nodes in these pigs. Antigens to porcine circovirus type 2 (PCV2) were immunolabeled in the lesions along with detection of viral DNA as PCV2 by polymerase chain reaction (PCR). In addition, antigens to porcine reproductive respiratory syndrome virus (PRRSV) were immunodetected in the lungs and Salmonella Choleraesuis was isolated from the affected pigs. The nine salmonellosis affected pigs, five (55.6%) with salmonellosis and PMWS concurrently infected with PRRSV were much higher than those infected with salmonellosis and PMWS (22.2%) or with salmonellosis and PPPRV (22.2%).  相似文献

Postweaning multisystemic wasting syndrome (PMWS) is an emerging disease in swine. Recently, the disease has been reproduced with inocula containing a newly described porcine circovirus (PCV), designated PCV 2, and porcine parvovirus (PPV). In order to determine if these viruses interact in naturally acquired PMWS, affected tissues from field cases were examined by immunohistochemistry (IHC) and polymerase chain reaction (PCR) for PCV 2 and PPV, as well as by PCR for the other recognized porcine circovirus, PCV 1. Porcine circovirus 2 was detected by PCR or IHC in affected fixed or frozen tissues from 69 of 69 cases of PMWS collected over 3 years from 25 farms. Porcine parvovirus was detected in 12 of the same cases, and PCV 1 was detected in 9 of 69; however, an apparent decrease was found in the sensitivity of the PCRs used to detect the latter 2 viruses when fixed tissue from the same cases were compared with the use of frozen tissues. Porcine circovirus 2 was not detected by PCR in affected tissues from 16 age-matched pigs that had Streptococcus suis-associated disease. Electron microscopic examination of plasma pooled from 15 pigs with PMWS revealed the presence of PCV and PPV, whereas these viruses were not observed in pooled plasma from 5 age-matched clinically normal pigs. These results confirm and extend previous findings documenting a consistent association of PCV 2 with PMWS. As well, infection by PPV or PCV 1 or both may be an important cofactor in the pathogenesis of some, but apparently not all, cases of PMWS.  相似文献

Torque teno sus virus (TTSuV) is emergent in swine herds. Recent studies have shown an increased frequency of TTSuV2 in Porcine circovirus type 2 (PCV2)-associated diseases (PCVAD), which are endemic in many swine-producing countries, including Brazil. Coinfection with several other viral and bacterial agents results in an increased incidence of more severe PCVAD. Given the limited information on TTSuV and PCV2 coinfection, especially in Brazilian swine herds, this study made a preliminary estimation of the occurrence of coinfection in swine herds by testing samples from different categories. Between 2008 and 2009, 111 samples of feces and 23 serum samples from 5 swine herds were tested for PCV2 and TTSuVs and the results analyzed for associations between these agents. No significant differences in coinfection frequency were observed for PCV2 + TTSuV1 or for PCV2 + TTSuV2 between nursery piglets (P = 0.730), growing pigs (P = 0.331), or sows (P = 0.472). However, a significant difference was observed for PCV2 + TTSuV1 + TTSuV2 between nursery piglets and growing pigs (P = 0.004; Fisher’s exact test). Phylogenetic studies agreed with the grouping of TTSuV1 and TTSuV2 into 2 different clades, with no distinct pattern of clustering of these isolates with the animal categories.  相似文献

为分析我国猪细环病毒(TTSuV)和猪圆环病毒2型(PCV2)的共感染情况,利用所设计的TTSuV(TTSuV1、TTSuV2)和PCV2特异性引物对我国29个省市采集的猪群血清样品同时进行PCV2和TTSuV(TTSuV1、TTSuV2) PCR检测,分析混合感染情况.结果所检测的1898份样品中,TTSuV阳性为1103份(58%),PCV2阳性为435份(23%).阳性样品中呈混合感染的有275份(14%),其中TTSuV1和PCV2为249份(13%),TTSuV2和PCV2为200份(10%),均为阳性的有174份(9%).调查结果显示,我国猪群中TTSuV和PCV2混合感染现象较为普遍,对地区性分布特征和饲养模式等影响因素的分析表明TTSuV和PCV2混合感染情况存在地区性差异(P<0.01),但饲养模式并不是共感染的关键因素.  相似文献

Postweaning multisystemic wasting syndrome (PMWS) is a worldwide emerging disease of weaned piglets. The objective of this review is to summarize the current knowledge regarding PMWS, its causative agent, mode of transmission, diagnostic techniques to detect PCV-2, the possible control measures, and the association of PMWS and PCV-2 with porcine dermatitis and nephropathy syndrome (PDNS). The causative agent of PMWS is porcine circovirus type 2 (PCV-2), however, not all pigs infected with PCV-2 develop the syndrome. PCV-2 is consistently associated with PMWS and PMWS is considered not to occur without it. Both the syndrome and the virus are not regarded as new. Co-factors that could activate PCV-2 to cause PMWS are considered. This enigmatic nature of both the syndrome and the virus is triggering a concern towards uncertainties of the viral transmission, its introduction in to the herd, effective tools of diagnosis, and control strategies.  相似文献

The lymphoid, renal, pulmonary, and hepatic lesions of naturally occurring postweaning multisystemic wasting syndrome (PMWS) affected pigs have been studied by means of immunohistology. Ten conventionally reared pigs showing acute clinical signs of PMWS were selected from a farm on which animal were seronegative to porcine reproductive and respiratory virus and to Aujeszky's disease virus. All pigs were positive in tests for porcine circovirus type 2 by ISH and IHC. Monoclonal and polyclonal antibodies to CD3, CD79alpha, CD45RA (3C3/9), lysozyme, SLA-II-DQ (BL2H5), and MAC387 were used to characterise cells in PMWS lesions. The most relevant changes were reduction or loss of B and T lymphocytes, increased numbers of macrophages, and partial loss and redistribution of antigen presenting cells throughout lymphoid tissues compared to uninfected controls. The characteristics of lymphoid lesions in the present study strongly suggest an immunosuppressive effect of PMWS in affected pigs.  相似文献

Porcine circovirus type 2 (PCV2) is the essential infectious agent of postweaning multisystemic wasting syndrome (PMWS). Despite first sequencing studies did not find any association between PCV2 sequences and PMWS occurrence, recent works have suggested the opposite. In the present study, 87 open reading frame 2 (ORF2) sequences obtained from pigs with different clinical conditions and coming from farms with different PMWS status were analyzed. Results further confirmed the existence of two genogroups and the definition of two PCV2 genotypes (1 and 2) is proposed. All sequences included in genotype 1 came from pigs from PMWS affected farms, while all sequences obtained from non-PMWS affected farms corresponded to genotype 2. Moreover, infection of single pigs from PMWS affected farms harbouring both genotypes is described. Present results suggest that PCV2 genotype 1 may potentially be more pathogenic than PCV2 genotype 2.  相似文献

A retrospective study for the detection of porcine circovirus 2 (PCV-2) DNA was conducted by nested PCR method in 16 cases of swine post-weaning multisystemic wasting syndrome (PMWS) in Thailand. Histopathology showed characteristic lesions of PMWS and intracytoplasmic viral inclusion bodies in macrophages infiltrating in lymphoid tissues. PCV-2 DNA was detected from formalin-fixed and/or formalin-fixed paraffin-embedded tissues from all pigs with PMWS. The amplified products were digested with Hae III.  相似文献

This study was carried out to investigate the presence of Torque teno sus virus types 1 (TTSuV1) and 2 (TTSuV2) in a longitudinally (14 to 150 days of age) collected paired pooled sera (pSE) and peripheral blood mononuclear cells (pPBMCs) using nested polymerase chain reaction. The detection rate of TTSuV1 in pSE increased from 14 to 90 days of age, but a progressive decline was observed from 120 to 150 days of age, while in pPBMC, a high value was maintained till the end of growing-finishing period. On the contrary, except in PBMCs at 30 days of age, high detection rates of TTSuV2 were found in both pSE and pPBMCs in all sampling ages. The detection rate of TTSuVs between pSE and pPBMCs was positively correlated at all sampling ages except for TTSuV1 at 150 days of age. This is the first study showing the presence of TTSuVs in PBMCs from pigs and describing the in vivo infection dynamics of TTSuV in paired sera and PBMCs during the entire growing and finishing periods of pigs reared in conventional farms.  相似文献

The emergence of porcine circovirus 2b genotype (PCV-2b) in swine in Canada   总被引:6,自引:0,他引:6  
Since late 2004, the swine industry in the province of Quebec has experienced a significant increase in death rate related to postweaning multisystemic wasting syndrome (PMWS). To explain this phenomenon, 2 hypotheses were formulated: 1) the presence of a 2nd pathogen could be exacerbating the porcine circovirus 2 (PCV-2) infection, or 2) a new and more virulent PCV-2 strain could be infecting swine. In 2005, 13 PMWS cases were submitted to the Quebec provincial diagnostic laboratory and PCV-2 was the only virus that could be found consistently by PCR in all 13 samples. The PCR detection results obtained for other viruses revealed the following: 61.5% were positive for porcine reproductive and respiratory syndrome virus, 30.8% for swine influenza virus, 15.4% for porcine parvovirus, 69.2% for swine torque teno virus (swTTV), 38.5% for swine hepatitis E virus (swHEV) and 84.6% for Mycoplasma hyorhinis; transmissible gastroenteritis virus and porcine respiratory coronavirus (TGEV/PRCV) was not detected. Sequences of the entire genome revealed that these PCV-2 strains belonged to a genotype (named PCV-2b) that has never been reported in Canada. Further sequence analyses on 83 other Canadian PCV-2 positive cases submitted to the provincial diagnostic laboratory during years 2005 and 2006 showed that 79.5% of the viral sequences obtained clustered in the PCV-2b genotype. The appearance of the PCV-2b genotype in Canada may explain the death rate increase related to PMWS, but this relationship has to be confirmed.  相似文献
