The composition of seminal plasma and metabolism of sperm of the cyprinid fishAlburnus alburnus were investigated. Statistically significant correlations were found between motility parameters and seminal fluid osmolality, pH, Na+, K+ and protein levels (negative correlations: % immotile spermatozoa-Na+, K+; positive correlations: % motile spermatozoa-osmolality, pH, Na+, K+, protein; % linear motile spermatozoa-pH protein; swimming velocity of spermatozoa-pH, Na+, protein). Spermatozoan motility and ATP metabolism and glycolysis were correlated as indicated by measurement of ATPase, pyruvate kinase, adenylate kinase and lactate dehydrogenase activity. Determination of semen quality is necessary to understand the basic biochemical processes occurring during motility of sperm and during fertilization to evaluate the reproductive ability of different fish species and to create an optimal environment for storage of spermatozoa; in this regard less information is available for Acipenseridae compared with Cyprinidae and Salmonidae. The aim of the present study is to determine chemical composition and osmolality of seminal fluid and their relationship with sperm motility in Acipenser persicus. The results obtained show that sodium (Na+), chloride (Cl?) and potassium (K+) were predominant ions in the seminal plasma and the average of osmolality of seminal plasma was 82.56 mOsm kg?1. The higher chemical contents and osmolality compared with other sturgeon species reveal species‐specific characteristics and high secretory activity of spermatic duct in A. persicus. Significant positive correlations were observed between osmolality‐Cl?, Na+‐osmolality and Na+–Cl? (P<0.05, P<0.001 and P<0.05 respectively). But statistically significant correlation was not observed between seminal plasma parameters and sperm motility. Probably, the Na+ and Cl? are the main electrolytes playing a major role in maintaining the osmolality of the seminal plasma and the viability of the spermatozoa in vivo.  相似文献

Sperm motility, pH and osmolality of seminal plasma varied throughout the reproductive season spanning the period from June to September. Initially, sperm motility was low, peaked in July and August and then fell again towards the end of the spawning season. While the pH of seminal plasma increased from pH 7.4 to 7.9 during the period of spermiation, the average seasonal pH (7.78 ± 0.03) remained close to an experimentally determined optimum pH range for ocean pout sperm motility (pH 8–9). Likewise, although the values for seminal plasma osmolality fell during the reproductive season, from 416–339 mmol kg-1, the average osmolality value 356 ± 3 was within the optimum for sperm motility (300–400 mmol kg-1). In comparing fluctuations in sperm motility with the biochemical composition of ocean pout seminal plasma during the spawning season, this analysis showed that increased Mg++ levels were correlated with the summer period of maximum sperm motility. A seasonal decline in Na+ and Cl ion levels was reflected in lower seminal plasma osmolality values.  相似文献

Boleophthalmus boddaerti submerged in 10%, 50% and 80% seawater (sw) for 7 days, had whole body transepithelial potentials (TEP) of 3.3, 18.3 and 22.9 mV, respectively. Hypophysectomy significantly decreased the TEP ofB. boddaerti and reversed the polarity of the TEP of the fish exposed to 10% sw.Hypophysectomy also significantly decreased the branchial Na+-K+ activated adenosine triphosphatase (Na+,K+-ATPase) activity but increased the activity of branchial HCO3 -Cl stimulated adenosine triphosphatase (HCO3 ,Cl-ATPase) inB. boddaerti exposed to 10% sw. However, survival in 10% sw was not significantly impaired by hypophysectomy and no significant change in plasma osmolality and plasma Na+ and Cl concentrations was observed.Various doses of ovine-prolactin or salmon-prolactin were unable to restore the TEP of hypophysectomizedB. boddaerti in 10% sw to that of the sham-operated fish. However, cortisol increased TEP to a positive value in hypophysectomizedB. boddaerti, though it was still lower than the sham-operated control. Cortisol treatment also affected the plasma osmolality, plasma Na+ and Cl contents and branchial Na+,K+-ATPase and HCO3 ,Cl-ATPase activities. Overall, the hormonal control of osmoregulation inB. boddaerti appeared to differ from that of other teleosts.  相似文献

The mudskipperB. boddaerti, was able to survive in waters of intermediate salinities (4–27). Fish submerged in dechlorinated tap water suffered 60% mortality by the fifth day while 60% of those in 100% sea-water (sw) died after the third day of exposure. After being submerged in 50% or 80% sw for 7 days, the plasma osmolality, plasma Na+ and Cl concentrations and the branchial Na+ and K+ activated adenosine triphosphatase (Na+,K+-ATPase) activity were significantly higher than those of fish submerged in 10% sw for the same period. However, the activities of the branchial HCO3 and Cl stimulated adenosine triphosphatase (HCO3 ,Cl-ATPase) and carbonic anhydrase of the latter fish were significantly greater than those of the former. Such correlation suggests that Na+,K+-ATPase is important for hyperosmotic adaptation in this fish while HCO3 -Cl-ATPase and carbonic anhydrase may be involved in hypoosmotic survival.  相似文献

The high sperm density, together with the short spermatozoa swimming time, makes European eel sperm manipulation and assessment for quality difficult. Two diluting media (K15 and K30) previously designed for Japanese eel sperm were tested. After 24 h, European eel sperm showed significant reduction in the percentage of motile spermatozoa after activation and different motility parameters (VAP, angular velocity; VCL, curvilinear velocity; VSL, straight line velocity; BCF, beating cross frequency), concluding that these media are not suitable to preserve the sperm of this species. After a hormonal treatment to induce spermiation, sperm volume, density and motility were recorded at weekly samplings. The variation of the osmolality (325–330 mOsm kg−1), pH (8.4–8.6) and the ionic composition (concentration of Na+, K+, Mg2+ and Ca2+) of the seminal plasma were registered. Physio-chemical results were related with sperm quality throughout the treatment, to determine which must be the suitable characteristics of one extender for the sperm of this species, and to find the best conditions to obtain suitable cryopreservation media for European eel sperm. K+ concentration increased, while Ca2+ and Mg2+ concentrations showed a progressive reduction in correlation with the sperm quality improvement. Na+ showed a decreasing, but not significant tendency. P1 and P2 freezing media were designed considering the physio-chemical parameters as well as the ionic composition shown by the best quality sperm samples, and then compared with the previously described solutions, TNK and K30. Sperm quality was determined, checking the percentage of motile spermatozoa and motility parameters using computer-assisted sperm analysis (CASA) software. Samples were frozen after dilution (1:5, 1:20, 1:100) in different freezing media supplemented with 10% dimethyl sulfoxide (DMSO). After thawing, samples frozen with low dilution ratio (1:5) in TNK and P1 media showed higher, although not significant, spermatozoa survival (35.5 ± 14.5 and 36.6 ± 6.7%). The addition of l-α-phosphatidylcholine to the media seems to have a positive effect, as reported in the Japanese eel.  相似文献

The capacity of cortisol, ovine growth hormone (oGH), recombinant bovine insulin-like growth factor I (rbIGF-I) and 3,3,5-triiodo-l-thyronine (T3) to increase hypoosmoregulatory capacity in the euryhaline teleost Fundulus heteroclitus was examined. Fish acclimated to brackish water (BW, 10 ppt salinity) were injected with a single dose of hormone suspended in oil and transferred to seawater (SW, 35 ppt salinity) 10 days post-injection. Fish were sampled 24 h after transfer and plasma osmolality and gill Na+, K+-ATPase activity were examined. Transfer from BW to SW induced significantly increased plasma osmolality but not gill Na+, K+-ATPase activity. Cortisol (50 g g–1 body weight) improved the ability to maintain plasma osmolality and to increase gill Na+, K+-ATPase activity. oGH (5 g g–1 body weight) also increased hypoosmoregulatory ability and gill Na+, K+-ATPase activity. A cooperation between oGH and cortisol was observed in increasing hypoosmoregulatory ability but not in increasing gill Na+, K+-ATPase activity. rbIGF-I (0.5 g g–1 body weight) alone was without effect in increasing salinity tolerance or gill Na+, K+-ATPase activity. rbIGF-I and oGH showed a positive interaction in increasing salinity tolerance, but not gill Na+, K+-ATPase activity. Treatment with T3 (5 g g–1 body weight) alone did not increase salinity tolerance or gill Na+, K+-ATPase activity, and there was no consistent significant interaction between cortisol and T3 or between GH and T3. The results confirm the classical role of cortisol as a seawater-adapting hormone and indicate an interaction between cortisol and the GH/IGF-I axis during seawater acclimation of Fundulus heteroclitus.  相似文献

The osmoregulation capabilities of 7-month-old juvenile Chinese sturgeon (Acipenser sinensis Gray) (128.8 ± 15 g) transferred directly from fresh water (0‰, 46 mOsmol kg−1) to brackish water (10‰, 273 mOsmol kg−1) were studied over a 20-day period. Changes in serum osmolarity, chloride (Cl), sodium (Na+), potassium (K+) and calcium (Ca2+) ion concentrations, as well as gill and spiral valve Na+,K+-ATPase activities were measured at 3, 12, 24, 72, 216 and 480 h after transfer to BW. The serum osmolarity and ion concentrations (Na+, Cl and Ca2+) increased immediately after the transference to BW, reaching maximum at 24 h and returned to a new steady state at 216 h, while the FW control group maintained basal levels which showed lower (P < 0.05) than the BW group. Gill Na+,K+-ATPase activity of BW group exhibited an abrupt decrease in the first 3 h after transfer, but began to increase at 3 h, reaching a peak value at 24 h, and returned to a new steady state at 216 h. The differences between gill Na+,K+-ATPase activity of BW and FW fish were significant (P < 0.05) after 12 h. In contrast, Na+,K+-ATPase activity of the spiral valve showed transient increase after transference from FW to BW, and then decreased rapidly at 3 h, reaching the lowest at 24 h after transference. At 216 h after exposure to BW, Na+,K+-ATPase activities of the spiral valve increased slowly to the levels of FW control. The results of our study indicate the existence of hyposmoregulatory adaptive mechanisms in 7-month-old juvenile Chinese sturgeon which enable this fish to acclimate itself successfully to brackish water.  相似文献

Spermatozoa concentration, ionic composition, osmolality, glucose and total protein contents of seminal plasma and sperm motility were determined in Barbus sharpeyi (Cyprinidae, Teleosotei). Spermatozoa concentration ranged from 9.77 to 20.20 × 109 spermatozoa mL?1. Osmolality (mOsmol kg?1) and ionic contents (mM L?1) of the seminal plasma were 274.5±9.0, 70.0±3.4 Na+, 28.8±0.9 K+, 101.7±3.1 Cl?, 0.9±0.1 Mg2+ and 2.1±0.1 Ca2+ respectively. Total protein and glucose were 5.3±0.2 g L?1 and 76.7±4.3 mM L?1 respectively. Sperm motility was initiated in a hypo‐osmotic condition, composed of either an ionic (KCl or NaCl) or a non‐ionic (sucrose) activation medium. Duration of sperm motility was very short: <2 min after activation in distilled water. Percentage of motile spermatozoa was significantly higher in an activation medium containing NaCl compared with that of distilled water. An activating medium containing NaCl or KCl higher than 150 mM or sucrose higher than 275 mM totally inhibited the activation of sperm motility. Immediately after sperm activation, wave(s) propagated along the flagellum, but waves were restricted to the proximal part of the flagellum (close to the head) at 1 min post activation. Studied characteristics in the present study were compared with those of other cyprinids for understanding inter‐species differences.  相似文献

The exposure of freshly spawned, immotile carp sperm to hypoosmotic media triggers the initiation of calcium-dependent flagellar motility. Intracellular calcium concentration has been thought to be the critical component in motility initiation, possibly acting through a novel signalling pathway. The sensitivity of sperm cells to changes of osmolality of the environment raises the question whether a mechanoregulated osmosensitive calcium pathway is involved in the activation mechanism of carp sperm motility. The sperm cells are in a depolarized state in the seminal plasma (Ψ = –2.6 ± 3 mV) and they hyperpolarize upon hypoosmosis-induced activation of motility (Ψ = –29 ± 4 mV). The intracellular sodium [Na+]i, potassium [K+]i and calcium [Ca2+]i ion concentrations were determined in quiescent cells, and at 20, 60 and 300 s after activation. The [Na+]i and [K+]i of the quiescent cells were similar to the [Na+]e and [K+]e of the seminal plasma. Following hypoosmotic shock-induced motility, both [Na+]i and [K+]i decreased to one-fourth of the initial concentration. The [Ca2+]i doubled at initiation of the motility of the sperm cells and remained unchanged for 5 min. Bepridil (50–250 μM), a blocker of the Na+/Ca2+ exchanger, blocked carp sperm motility reversibly. Gadolinium, a blocker of stretch-activated channels (10–20 μM), inhibited sperm motility in a dose-dependent manner and its effect was reversible. Hypoosmotic shock fluidized the membrane and gadolinium treatment made it more rigid in both quiescent cells and hypotonic shock treated but immotile sperm cells. Based on these observations, it is suggested that, besides the well-known function of potassium and calcium channels, stretch-induced conformational changes of membrane proteins are also involved in the sperm activation mechanism of common carp.  相似文献

This study was carried out to evaluate milt quality in male Caspian brown trout (Salmo trutta caspius) over the course of the winter spawning season. Milt samples were collected biweekly during December and January. Chemical composition of seminal fluid, sperm production (milt volume, sperm density, spermatocrit,) and sperm motility characteristics (percentage and duration of motility) were measured. Milt volume, sperm density, osmolality, seminal minerals (Ca2+, Mg2+, K+, Na+, Cl?), and total protein gradually decreased over the spawning season. Glucose and triglyceride content of milt did not show significant changes over the spawning season. Milt pH and the percentage and duration of motility were comparatively stable, declining only at the end of the season. Significant positive correlations were found between sperm density and seminal minerals, total protein and spermatocrit; percentage of motile spermatozoa and seminal minerals, total protein; and duration of motility and K+, Cl?, total protein, and pH. Results show that season has a significant influence on milt quality in male Caspian brown trout, with the best milt being available at the beginning of spawning season.  相似文献

A study was conducted to examine the effect of dietary cation-anion difference (CAD, Na++K+-Cl, mEq kg –1) on acid-base balance, food consumption and growth of juvenile African catfish, Clarias gariepinus. Four dietary CAD levels (-100, 100, 500, 700 mEq kg–1) were established by altering levels of NaHCO3, CaCl2 and NH4Cl in the diets. The group fed the diet of excess anions (CAD: -100 mEq kg–1) had low food consumption and growth rate. Food consumption and growth increased linearly with an increase in dietary CAD within the range -100 to 700 meq. kg–1. Blood pH was not significantly influenced by dietary CAD. A quadratic relationship between Na+, Cl and CAD level in plasma and dietary CAD was found. Body composition of fish and nitrogen/energy utilisation efficiency differed between dietary CAD treatments. The results suggest that dietary CAD influence acid-base regulation and thus the energy costs for maintaining homeostasis. The increased food intake and growth performance of C. gariepinus at higher CAD levels may be related to a reduced maintenance costs for homeostasis.  相似文献

The motility and fertilizing ability of the Persian sturgeon, Acipenser persicus, spermatozoa were investigated. Optimum ionic content (Na+, K+, Ca2+ and Mg2+) and pH of activation solution as well as the optimum dilution rate were determined. The results show optimum motility characteristics of spermatozoa in buffered solutions containing 25, 0.2, 3 and 10 mM L?1 Na+, K+, Ca2+ and Mg2+, respectively, at dilution rate 1:50 and pH 8.0. To test the fertilizing ability of sperm, two buffered saline solutions were used as activation solution of sperm motility. The present study indicated (1) spermatozoa motility is one of key factors that influence on fertilizing ability of sperm, (2) a high fertilizing ability of sperm is obtained after dilution in saline solutions rather than in freshwater and (3) a maximum fertilization rate occurs in buffered saline solution containing 0.2 mM L?1 K+. There is also a good correlation between biochemical characteristics of seminal plasma and fertilizing ability of sperm.  相似文献

The effect of sulfide on K+ influx pathways was measured in red blood cells (RBCs) of sulfide-sensitive rainbow trout (Oncorhynchus mykiss) and sulfide-tolerant crucian carp (Carassius carassius). In trout RBCs, maximal inhibition of Na+, K+-ATPase was attained at 10 mol l–1 sulfide and amounted to 32% without being influenced by pH between 6.7 and 8.3. Ouabain-resistant K+ influx in the absence and presence of sulfide was insignificant at pH values between 6.7 and 7.7. At higher pH values ouabain-resistant K+ influx increased, but was inhibited to about 15% by 30 mol l–1 sulfide. In RBCs of crucian carp neither Na+, K+-ATPase nor ouabain-resistant K+ influx were affected by sulfide concentrations up to 850 mol l–1. Differences in sulfide-sensitivity of K+ influx between both species can be based upon different properties of the membrane transporter themselves. The reduced Na+, K+-ATPase activity in trout RBCs may also result from a slightly reduced (by 9%) ATP level after sulfide exposure. In addition, intracellular sulfide concentrations were higher in trout RBCs as compared to crucian carp. In trout, intracellular sulfide concentrations reached extracellular levels within 5 min of incubation whereas sulfide concentrations in crucian carp RBCs remained about 2-fold lower than extracellular concentrations. Although the physiological basis of sulfide-insensitive K+ influx in crucian carp RBCs is currently unknown it may contribute to the extremely high sulfide-tolerance of this species.  相似文献

The spermiation of tench males was stimulated with Supergestran containing mammalian LHRHa lecireline at the following doses: 5, 10, 20 and 40 g kg−1 b.w.; then with carp pituitary suspension (CPS) at a dose of 2 mg kg−1 b.w. and with a control of saline physiological solution. The following days, meaning 24, 48 and 72 h after injection, sperm was collected to evaluate volume and the number of sperm per male per kg body weight (B.W.) The percentage of motile sperm and velocity of spermatozoa were measured 48 h after hormonal injection, and 72 h after hormonal injection the sperm was evaluated for fertilization and hatching ability. All 42 males in experimental groups were diploid. Live weight did not differ significantly among experimental groups. The strongest stimulation of spermiation was achieved with LHRHa in dosage of 20 and 40 g kg−1 b.w. and CPS compared to males of the control group and lower dosage of LHRHa. Analysis of variance showed no significant influence of the treatment on the velocity and percentage of motile spermatozoa. The effect of different treatment on the fertilization capacity (the number of spermatozoa per egg was equilibrated) was significant. Significantly the highest quality of sperm collected 72 h after injection expressed by percentage of fertilization and hatching (62–65% fertilization and 61–64% hatching rates, respectively) was found for LHRHa in dosage of 20 and 40 g kg−1 b.w. Significantly the lowest parameters of fertilization and hatching were found for the control group, on the 12% level.  相似文献

Summer flounder (Paralichthys dentatus) that survived flounder infectious necrotizing enteritis (FINE) provided an unusual model for testing the role of the intestine in maintaining salt and water balance. The survivors lost 67% of the posterior-most intestine. The remaining stump of the anterior intestine healed and became a blind pouch. We sampled the fish three months after the epizootic ended, demonstrating that these fish survive with only a portion of their intestine. We hypothesized that salt and water balance would be disturbed in the survivors of FINE. However, plasma osmolality and concentrations of Na+, Cl, and K+ were the same in intact flounder and those with only an intestinal stump. No compensatory changes in gill Na+, K+-ATPase activity were detected. The fish with intestinal stumps continued to feed, but their body weight was only 41% that of intact fish. Summer flounder can maintain salt and water homeostasis, but fail to thrive, using only one-third of their intestine.  相似文献

Changes in semen quality and selected biochemical markers were analyzed during a week of spawning season of common carp Cyprinus carpio L. Semen was obtained twice, on May 30 and on June 7, and each time it was collected 24 h after hormonal stimulation using Ovopel [(d-Ala6, Pro9 NEt)-mGnRH + metoclopramide] in 1 pellet kg?1. The total volume of semen (ml), volume of semen per kg of body weight (ml kg?1 b.w.), sperm concentration (×109 ml?1), total number of sperm per kg of body weight (×109 kg?1 b.w.), pH of semen, pH of seminal plasma, seminal plasma osmotic pressure (mOsm kg?1) and the total protein content in seminal plasma (mg ml?1) were determined. A 10 mM Tris buffer containing 100 mM NaCl with 0.5 % BSA (pH 9.0, osmolality 200 mOsm kg?1) was used to activate sperm. The following computer-assisted sperm analysis (CASA) parameters were determined: percentage of motile sperm (MOT, %), progressively motile sperm (PRG, %), curvilinear velocity (VCL, μm s?1), straight-line velocity (VSL, μm s?1), movement linearity (LIN, %), wobbling index (WOB, %), amplitude of lateral head displacement (ALH, μm) and beat cross-frequency (BCF, Hz). The volume of semen per kg of BW, total number of sperm per kg of BW and semen pH were significantly lower at the second semen sampling compared to the first semen sampling. Volume of semen at the second sampling correlated positively with CASA parameters. A lack of differences among CASA parameters between both collection periods indicates good quality of carp sperm hormonally stimulated with Ovopel twice at a 1-week interval.  相似文献

The aim of this work was to determine the effects of supplemental dietary sodium chloride on salt water acclimation of tilapia Oreochromis niloticus. Fish were fed a basal diet supplemented with NaCl (8%) during three weeks in fresh water (FW) and then transferred to salt water (SW) at 15 and 20. Changes in plasma osmolality, chloride ion concentration (Cl), plasma level of cortisol and gill Na+, K+-ATPase activity were measured at 6, 12, 24, 48, 72 and 168 h after transfer to 15SW, while the higher strength SW group (20) was only monitored up to 24 h. Morphological changes in the gill mitochondria-rich (MR) cells were examined in relation to environmental salinity. The changes associated with dietary NaCl were sporadic and of small magnitude. The plasma osmolality and Cl increased immediately after transfer up to 12–24 h, but fish fed dietary salt (S) showed lower values than the control group (C). The S group showed higher plasma levels of cortisol than the control, which maintained its initial levels during the experiment. Gill Na+, K+-ATPase activity of the S group began to increase in the first hours after transfer, reaching maximum at 12 h and returned to basal level at 24 h, while the control group maintained basal levels. The differences between gill Na+, K+-ATPase activity of S and C fish were significant (p < 0.05) at 12 h. Transmission electron microscopy (TEM) revealed that MR cells in SW show more mitochondria and a more developed tubular system arising from the basolateral membrane. The MR cells of both groups frequently formed a multicellular complex in SW, consisting of a main MR and one or more accessory cells. Such complexes are rarely observed in FW. Some MR cells of fish fed supplemented dietary salt displayed convex apical membrane in FW.  相似文献

An experiment was conducted to compare the potency of different levels of luteinizing hormone-releasing hormone analogue (LHRHa) for increasing the sperm counts in sexually mature male Oreochromis niloticus (L.). One hundred males (106.49 ± 3.59g, 15.2 7 ± 0.20 cm) were randomly distributed in four treatment groups, each receiving intramuscular injections of: (i) control, 0.05 ml phosphate-buffered saline, (ii) 10 μg kg-1 LHRHa, (iii) 20 μ kg-1 LHRHa and (iv) 30 μ kg-1 LHRHa. Sperm counts were determined prior to injection (day 0) and for four consecutive days (days 1 to 4) thereafter. Results showed a significant increase in sperm counts (P < 0.01) 1 day after injection among males injected with 10 to 20 μ kg-1 LHRHa. The effect of injecting 30 μ kg-1 LHRHa is similar to that in the control group. Across all treatments, sperm counts declined with time over the 4-day sampling period.  相似文献

The fluid uptake rate of the posterior intestine of salmonids increases during the parr-smolt transformation. Intestinal fluid uptake in post-smolt Atlantic salmon was investigated after treatment with cortisol and growth hormone (GH), alone or together. Two replicate experiments were conducted in August 1991 and August 1992. Cortisol was emulsified in vegetable shortening and vegetable oil (1:1) and implanted into the peritoneal cavity. GH was administered as intraperitoneal injections in a saline vehicle on days 0 and 2. On days 5 and 6, plasma cortisol levels, gill Na+,K+-ATPase activity, andin vitro measurements of fluid transport rate (Jv) across the posterior intestine were measured. Implants of cortisol elevated the plasma cortisol levels within a physiological range, and resulted in elevated gill Na+,K+-ATPase activity, as expected. The fluid uptake rate across the posterior intestine was roughly doubled by cortisol treatment. GH treatment did not affect intestinal fluid transport, gill Na+,K+-ATPase activity, or plasma cortisol concentrations. The seawater-adapting increase in the rate of fluid uptake by the posterior intestine of smolting salmon is probably stimulated by elevated plasma cortisol concentrations.  相似文献
