Bacteria from genera Chlamydia (Ch.) and Chlamydophila (Chl.) are very pathogenic and may infect humans and animals. They also may cause latent infection, especially in animals. In this paper we discuss the non-specific and specific cellular and humoral immunity in farm animals, after infection or immunisation with Chlamydia sp. and Chlamydophila sp. bacteria. It has been shown, that the infection or immunisation with the microorganisms influenced the activity of polimorphonuclear cells (PMN) and mononuclear cells (MN) in the process of phagocytosis. It has also been shown that the bacteria influenced the amount and activity of lysozyme, activities of myeloperoxidase and lysosomal enzymes. Infection or immunisation with the microorganisms was demonstrated to affect numbers of lymphocytes T and B and those of their subpopulation as well as the activity of cytokines and levels of serum and secreted immunoglobulins. The changes were detected just a few hours after infection or immunisation and persisted for a few days to a few decades.  相似文献

Acute interdigital phlegmon (AIP) is a commonly occurring anaerobic bacterial infection in cattle. This study examined in vitro the interaction of bovine polymorphonuclear granulocytic neutrophils (PMN) from blood with bacterial species involved in AIP. Polymorphonuclear neutrophils were purified from whole bovine blood, exposed to one of the three putative etiologic agents of AIP and comparatively assessed for phagocytosis using light microscopy. Fusobacterium necrophorum and Prevotella intermedia were effectively phagocytosed by PMN, but Porphyromonas levii was phagocytosed significantly less effectively by PMN. The effect of high titre anti-P. levii bovine serum on antibody-mediated phagocytosis by PMN was also evaluated. High titre serum increased the efficiency of phagocytosis of P. levii by bovine PMN. This was independent of heat labile complement factors. Antibodies specific for P. levii were assessed for protease activity capable of cleaving bovine immunoglobulins (IgG, IgG1, IgG2, and IgM). Partially purified supernatant from broth cultures of P. levii were incubated with biotinylated immunoglobulins (Igs). Samples were taken from times 0 to 72 h and examined using SDS-PAGE followed by Western blot analysis. Streptavidin-alkaline phosphatase and NBT-BCIP were used to visualize the Igs for heavy and light chains as well as lower molecular weight fragments of these glycoproteins. Porphyromonas levii produced an immunoglobulin protease which readily cleaved bovine IgG into fragments, but did not act against IgM. Specifically, the enzyme may be a significant virulence factor as it may act to neutralize the antibodies demonstrated necessary for effective PMN-mediated phagocytosis.  相似文献

The effect of serum from horn cancer affected bullocks and cows on E-rosetting capacity of peripheral blood lymphocytes (PBL) from unaffected control animals was examined. The E-rosette counts were made using 2-aminoethyl isothiouronium bromide (AET) treated sheep red blood cells. A significant decrease in the percentage of EAET rosette forming cells was noticed when PBL were incubated with 50 per cent serum from animals affected with horn cancer. However, no such effect was noticed when PBL were treated with 50 per cent serum from unaffected control animals. A linear relationship was observed between percentage of EAET rosette forming cells of animals affected with horn cancer and E-rosette inhibitory activity of the corresponding serum on PBL from control animals.  相似文献

Recombinant major outer membrane proteins (rMOMP) of Chlamydophila (Ch.) abortus, Ch. pecorum, and Chlamydia (C.) suis were used as antigens to distinguish chlamydial species-specific antibodies in (i) immune sera from six rabbits and three pigs raised against native purified elementary bodies, (ii) serum samples from 25 sows vaccinated with Ch. abortus, and (iii) 40 serum samples from four heifers experimentally infected with Ch. abortus. All post-exposition sera contained chlamydial antibodies as confirmed by strong ELISA seroreactivities against the chlamydial LPS. For the rMOMP ELISA mean IgG antibody levels were at least 5.8-fold higher with the particular rMOMP homologous to the chlamydial species used for immunisation or infection than with heterologous rMOMPs (P <0.001). Preferential rMOMP ELISA reactivities of sera were confirmed by Western blotting. The results suggest that the entire chlamydial rMOMP could provide a species-specific serodiagnostic antigen.  相似文献

Examinations were performed of 31 fattened pigs suffering from papular dermatitis (PD) and 33 fattened pigs without dermal changes (C) coming from the D. large pig-fattening farm and the production breeding herd of pigs. The weight of the pigs was from 95 to 105 kg. Both farms are sited in a mining area (intensive lignite extraction). The PD pigs, compared with the controls (C), had less total protein, cholesterol and calcium in their blood serum and increased concentrations of thyroxine (T4), non-esterified fatty acids (NEFA), vitamins A and E, inorganic phosphorus and potassium, and the higher alanine aminotransferase (ALT) activity. The PD pigs also had an increased leucocyte count in blood. In their peripheral lymphocytes the PD pigs had a significantly higher number of aberrant cells, 3.9 times higher number of chromosome breaks, slightly increased rate of sister chromatid exchanges, but half the number of chromosome exchanges of the C pigs. No differences between PD pigs and the controls were recorded in the response of the T-lymphocytes of peripheral blood to non-specific phytohaemagglutinin stimulation; neither were there any substantial differences in the concentration of serum immunoglobulins of the IgG, IgM and IgA classes. The PD and C pigs of both herds (the D. and M. farms) had low concentrations of all three serum immunoglobulins. The higher occurrence of PD in the D. herd, compared with the M. farm, is associated with a greater reduction of serum immunoglobulins (IgM by 38%, IgA by 25%, IgG by 15%).  相似文献

Three adult horses were evaluated for signs of musculoskeletal pain, dullness, ataxia, and seizures. A diagnosis of bacterial meningitis was made on the basis of results of CSF analysis. Because primary bacterial meningitis is so rare in adult horses without any history of generalized sepsis or trauma, immune function testing was pursued. Flow cytometric phenotyping of peripheral blood lymphocytes was performed, and proliferation of peripheral blood lymphocytes in response to concanavalin A, phytohemagglutinin, pokeweed mitogen, and lipopolysaccharide was determined. Serum IgA, IgM, and IgG concentrations were measured by means of radial immunodiffusion, and serum concentrations of IgG isotypes were assessed with a capture antibody ELISA. Serum tetanus antibody concentrations were measured before and 1 month after tetanus toxoid administration. Phagocytosis and oxidative burst activity of isolated peripheral blood phagocytes were evaluated by means of simultaneous flow cytometric analysis. Persistent B-cell lymphopenia, hypogammaglobulinemia, and abnormal in vitro responses to mitogens were detected in all 3 horses, and a diagnosis of common variable immunodeficiency was made.  相似文献

Anti‐ferritin autoantibody is a ferritin‐binding protein commonly found in mammals; it is thought to form an immune complex with ferritin and thereby mediate the rapid clearance of circulating ferritin. The aim of this study is to determine concentrations of ferritin and anti‐ferritin autoantibodies (immunoglobulin (Ig)M, IgG and IgA) in serum and colostrum of Holstein (H) and Japanese Black (JB) cows within 24 h of normal calving. Blood and colostrum samples were collected from cows of various ages (2–11 years) and calving number (1–8 live births). Mean ferritin concentrations were higher in colostrum than in serum for both breeds, and higher colostrum ferritin concentrations were found in H than JB cows. IgA antibodies in serum and colostrum from both breeds had negligible ferritin‐binding activity. For both breeds, IgM and IgG antibodies had higher ferritin‐binding activity in colostrum than in serum. There was a significant correlation between IgM and IgG ferritin‐binding activities in serum and colostrum of H and JB cows. These results suggest that ferritin and IgM and IgG autoantibodies are actively transferred from the blood stream to the colostrum at prepartum or early lactation.  相似文献

Infectious diseases are common in foals aged 1-5 months. The objectives of this investigation were to evaluate immunologic parameters in foals from birth to weaning to establish reference values for the proportion of circulating lymphocytes that were helper (CD4+) or cytotoxic (CD8+) T cells, or B cells; to measure serum immunoglobulin (IgM and IgG) concentrations; and to compare these immunologic parameters to values in foals with naturally occurring Rhodococcus equi pneumonia and in adult horses. Peripheral blood lymphocyte subpopulations were determined by flow cytometric analysis, and serum IgG and IgM concentrations were determined by radial immunodiffusion. Flow cytometric analysis of lymphocyte subpopulations suggested age-related changes in the cell-mediated immune system in horses. Absolute circulating CD4+ and CD8+ T lymphocytes and B cells increased linearly up to 3 months of age. Circulating B cell concentrations from birth to 6 months of age were greater than values in adult horses and the lymphocyte differences among the age groups are mainly due to variation in B lymphocytes. Both absolute and proportional B cell concentrations were greater in foals with R equi pneumonia than in healthy foals at the same age. The increase in absolute cell counts of each subpopulation was dependent on the increase of absolute peripheral blood lymphocyte count. Serum IgG concentration increased linearly from 1 to 3 months of age, and serum IgM concentrations increased from 1 to 6 months of age. These data suggest age-dependent cell-mediated and humoral development in young foals.  相似文献

A 12-month-old Thoroughbred colt was presented for chronic recurrent respiratory disease characterised by persistent nasal discharge, intermittent pyrexia and coughing of several months' duration. These episodes were responsive to empirical antimicrobial therapy but returned once therapy was discontinued. History, physical examination and initial diagnostics were suggestive of bacterial pneumonia and possible underlying immunodeficiency. Serum immunoglobulin analysis and peripheral blood lymphocyte phenotyping revealed low serum IgG and IgM concentrations. In this 12-month-old colt, low serum IgG concentration was the most diagnostic parameter and supported a presumptive diagnosis of transient hypogammaglobulinaemia of the young. The colt responded favourably to repeated treatment with antimicrobials for bacterial pneumonia, and serial immunologic testing over the following months showed gradual increase in serum IgG and IgM concentrations, supporting the diagnosis of delayed development of humoral immunity in this patient.  相似文献

The transfer of immunoglobulin (Ig) isotypes (IgG1, IgG2, IgM), gamma-glutamyl transpeptidase (gamma-GT) and added D-xylose from colostrum to serum was investigated in newborn Holstein bull calves. Significant differences were observed in the time courses of the serum concentrations of these colostrum constituents following absorption from pooled colostrum. A computer model was devised to simulate the process of absorption of Ig isotypes, gamma-GT and D-xylose from colostrum in the newborn calf. A Fortran program was used to generate plots of the time course of the concentration of colostrum constituents in serum and other body fluids following a single feed of colostrum. These plots show how the changes in serum concentration of absorbed Ig isotypes, gamma-GT and D-xylose are affected by different rates of intestinal absorption, redistribution in body fluids and removal from plasma. A critical examination of data from the computer model and from the calf feeding experiments supports the view that the absorption of IgG1, IgG2 and IgM is not selective in the calf. The data were compared with earlier studies of the efficiency of the colostral transfer of Ig to the calf. In the present study the transfer efficiencies of IgG1, IgG2, IgM, gamma-GT and D-xylose were 46 per cent, 49 per cent, 47 per cent, 18 per cent and 21 per cent, respectively.  相似文献

Phagocytosis of Bacteroides nodosus by ovine peripheral blood leucocytes (PBL) was examined after organisms had been opsonized in sera from normal sheep, or from animals immune to, or infected with ovine footrot. Ingestion of bacteria, as assessed microscopically or by counting isotopically-labelled organisms spectrometrically was effected in suspensions by polymorphonuclear leucocytes (PMN). Opsonization of bacteria in immune serum, particularly its IgG2 isotype, enhanced the rate of phagocytosis by PMN compared with that promoted by normal serum or medium alone. Whereas IgG2 from immune serum also increased the rate of ingestion of B. nodosus by adherent PMN, IgM and IgG1 from immune serum also initiated phagocytosis of bacteria by adherent ovine monocytes. Leucocytes from normal, immune or infected sheep of different breeds ingested B. nodosus with equal facility.  相似文献

The relationship between Cowdria ruminantium and Chlamydia trachomatis was studied by immunofluorescence. A monoclonal antibody directed against the major outer membrane protein of C. trachomatis recognized rickettsial colonies of C. ruminantium in infected goat brain. No specific fluorescence was observed in non-infected brain. Two commercial Chlamydia-specific monoclonal antibodies as well as polyvalent anti-Chlamydia rabbit serum recognized C. trachomatis, but did not recognize Cowdria. Moreover, polyvalent Cowdria antiserum failed to recognize C. trachomatis cultivated in HeLa cells. It is concluded that Cowdria and Chlamydia are to a certain extent related, confirming similarities in ultrastructure and developmental cycle.  相似文献

Immunoglobulin G, IgM, and IgA concentrations were measured in serum collected from 36 Standardbred mares within 12 hours of foaling, in colostrum collected within 6 hours of foaling, and in serum collected from foals 24 to 48 hours after birth. In serum collected from mares after parturition, mean concentrations of IgG, IgM, and IgA were 2,463.9 +/- 1,337.3 mg/dl, 136.4 +/- 218 mg/dl, and 305.2 +/- 237.5 mg/dl, respectively. In serum from foals, mean concentrations of IgG, IgM, and IgA were 1,953.3 +/- 1,635 mg/dl, 33.8 +/- 30.4 mg/dl, and 58.4 +/- 42.2 mg/dl, respectively. In colostrum, mean concentrations of IgG, IgM, and IgA were 8,911.9 +/- 6,282.2 mg/dl, 957 +/- 1088.1 mg/dl, and 122.9 +/- 77.3 mg/dl, respectively. The IgG concentrations in foal serum were poorly correlated with IgG concentrations in colostrum (r = 0.462, P less than 0.01). Correlations of IgM or IgA concentrations in serum from foals with IgM or IgA concentrations in colostrum and correlations of IgG concentrations in serum from mares with those in colostrum were not significant (P less than 0.01). Of 36 foals, 1 (2.8%) had a serum IgG concentration less than 400 mg/dl. Of 36 foals monitored for 4 months, 6 developed infectious respiratory tract disease requiring antimicrobial therapy at ages varying from 55 to 113 days; these infections were probably not related to failure or partial failure of passive transfer of antibody.  相似文献

The phagocytosis and killing of Escherichia coli (strain P4) and Staphylococcus aureus (strain M60) by bovine polymorphonuclear lymphocytes (PMN) suspended in phosphate buffered saline, requires the presence of calcium ions and opsonins. The highest dilution of normal decomplemented adult sera in which the opsonins are still active is approximately 1/1000 for S aureus and 1/200 for E coli. In fetal and precolostral sera heat labile factors are also required for opsonising E coli but these are not required for S aureus. IgG1 and IgG2 from adult sera did not opsonise the bacteria even though receptors for IgG2 anti-erythrocyte antibodies have been reported on bovine and ovine PMN. A systematic separation of adult serum proteins was carried out by salt fractionation, anion exchange and gel filtration chromatography. The results suggest strongly that the opsonin in adult bovine sera is IgM.  相似文献

The prevalence of Coxiella burnetii and Chlamydia psittaci antibodies was investigated in 530 dog specimens divided into six groups, i. e. A = private watch dogs, B1 = service dogs from Bratislava, B2 = service dogs from other localities of Slovakia and Moravia, C = watch dogs from farms, I = household dogs, T = stray dogs. The dogs demonstrated the higher seropositivity to C. burnetii (11.7%) than to Ch. psittaci (5.5%). The highest percentage of antibodies to C. burnetii was found in stray dogs (23.7%), less prevalence of antibodies was observed in the animals in group C (13.6%), almost the same positivity was proved in the dogs of group B1 and B2 (10.5 and 10.6%). The highest positivity to Ch. psittaci was demonstrated in the dogs of group A (8.7%), less in group B2 (6.6%) and the least number in group B1 (1.9%). The stray dogs occupied the intermediate position in this data (Tab. I). Ninety four localities were tested, from which 38 were seropositive. Neither acute coxiellosis nor chlamydiosis were proved in any animals examined. Ninety per cent of dogs were found healthy, but 10% of dogs demonstrated hepatopathia and gastroenteritis. Two of them (category A and I) were seropositive to C. burnetii (titer 1:8 to 1:16) and one to Ch. psittaci (titer 1:16). Both C. burnetii and Ch. psittaci attack dogs parallely with the agents of other zoonoses, of which the most common is Toxoplasma gondii (Tab. II). Four gilts, sisters from one litter, aged 70 days and weighing 20-24 kg, were used for a trial. Two experimental gilts (P) were administered an experimental feed mixture containing phenylmercury chloride (40 mg/kg). Two control gilts (K) were fed the same mixture but without phenylmercury chloride. P gilts began to lag behind in their growth from day 60 of the experiment, they manifested nonphysiological postures (dog's sitting posture), paresis of hind limbs and uncoordinated movements. P gilts had cloudy, orange-brown urine from day 70 and from day 75 they began to suffer from diarrhoea. Mercury (Hg) contents in urine and blood serum of P gilts were irregularly variable: urine 0.58-2.15 mg/l, blood serum 0.02-0.37 mg/l. Hg content in excrements of P gilts fluctuated from 23 to 26 mg/kg. Vitamin A concentrations in blood serum and liver decreased in P gilts. Phenylmercury chloride feeding caused mutagenic changes in peripheral lymphocytes of P gilts (an increase in the number of aberrant cells from 2-3% to 8-9%) and reduced IgA, IgM and IgG immunoglobulin levels in blood serum. Pathological lesions were observed in the colon, kidneys and liver. None of the above-mentioned changes were observed in K gilts. Increased resistance to the negative effects of Hg was found in one experimental gilt. In comparison with K gilts, Hg concentrations in P gilts after 130 days of the experiment increased as follows: 427 times in kidneys, 333 times in liver, 106 times in guts, 71 times in pancreas, 53 times in ovaries, 50 times in muscles, 47 times in bristles and 16 times in the brain.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

In a pilot experiment, bullocks were given diet with a sodium monensinate supplement as the Rumensin preparation. The dose of the supplement was 125 mg per head/day and the medicated feed was given to the bullocks for 14 months. The daily weight gain of the tested bullocks increased by 3.85% and feed consumption decreased by 7.12%. The meat of the ten bullocks examined (m. longissimus dorsi) showed no statistically significant changes in the basic chemical composition as compared with the meat of ten control bullocks. In the tested group the content of hydroxyproline in meat decreased by 35.14% (from 0.12 to 0.08 g per 100 g of fresh tissue - p less than 0.05). The content of glycine decreased by 8.82% (from 3.63 to 3.31 g per 100 g of dry matter - P less than 0.01). The content of alanine decreased by 5.86% (from 5.12 to 4.82 g per 100 g of dry matter - P less than 0.05). As to essential amino acids, the content of valine in the meat of the tested bullocks increased by 4.76% (from 3.99 to 4.18 g per 100 g of dry matter - P less than 0.05). Sodium monensinate exerted a favourable influence on weight gains and nutrient conversion in the bullocks. It had no effect on the basic chemical composition and no marked influence on the content of amino acids, except hydroxyproline and, particularly, essential amino acids in the meat of the tested bullocks. A reduction in the content of hydroxyproline signalizes higher-value proteins.  相似文献

Colostral volume and IgG and IgM concentrations were determined in 6 multiparous mares at foaling and them every 2 hours from 16 to 20 hours after parturition. Serum IgG and IgM concentrations at foaling also were determined in each mare. The rate of mammary secretion was 292 +/- 26 ml/h (range, 202 to 389 ml/h), and the colostral volume was 5.1 +/- 0.5 L (range, 3.2 to 7.0 L). The colostral IgG and IgM contents were 440 +/- 106 g (range, 199 to 855 g) and 3.1 +/- 0.9 g (range, 0.7 g to 7.1 g), respectively. There was no significant correlation between serum and initial colostral IgG and IgM concentration or between serum and total colostral IgG or IgM values. The colostral IgG and IgM concentrations at foaling correlated well with the total colostral IgG and IgM contents, respectively. The initial 250 ml of colostrum contained 10 +/- 1.4% (range, 6.0 to 13.9%) and 6 +/- 1.0% (range, 2.4 to 8.5%) of the total IgG and IgM contents, respectively, and the initial 500 ml of colostrum contained 20 +/- 2.7% (range 12.0 to 27.1%) and 14 +/- 1.2% (8.2 to 17%) of the total colostral IgG and IgM contents, respectively.  相似文献

Absorption of bovine colostral immunoglobulins G and M in newborn foals   总被引:1,自引:0,他引:1  
The uptake of colostral IgG and IgM, their serum half-lives, and the rates of endogenous synthesis of IgG and IgM were evaluated in 6 newborn foals fed bovine colostrum (principals) and 6 foals allowed to suckle their dams (controls). The principal foals were fed 400 ml of bovine colostrum (IgG, 10,000 mg/dl and IgM, 200 mg/dl) at 2-hour intervals, from 2 to 20 hours after foaling (total dose, 4 L). Serum IgG and IgM concentrations were determined by single radial immunodiffusion from birth to 98 days of age. At foaling, principal foals had no detectable serum equine IgG, but 1 control foal had serum equine IgG of 185 mg/dl. After ingestion of colostrum, there was no significant difference in the maximal serum bovine IgG concentration (range, 1,350 to 3,300 mg/dl) in the principal foals, and maximal serum equine IgG concentration in the control foals (range, 500 to 6,000 mg/dl). The calculated biological bovine and equine IgG half-life in the principal and control groups was 9.4 and 26 days, respectively. Endogenous IgG synthesis was first detected in 1 principal foal at 3 days of age, but was detected first between 28 and 42 days in the other principal foals. Starting on day 56 there was no significant difference in serum equine IgG concentration between groups. At foaling, foals in both groups had low equine IgM concentrations. Immunoglobulin concentrations (IgG, IgM, and IgA) in bovine serum, follicular fluid, and uterine and vaginal secretions were determined. The specificities of IgG, IgM, and IgA for virus-neutralizing antibody against bovine viral diarrhea (BVD) and infectious bovine rhinotracheitis (IBR) viruses were also examined. High concentrations of IgG were present in both serum and follicular fluid. The IgG, IgM, and IgA concentrations were low in uterine and vaginal secretions. There was more IgG in the uterus during estrus than at any other time. Virus-neutralizing antibodies against BVD and IBR in serum of cows were mainly the IgG class. There was positive correlation between serum and follicular fluid virus-neutralizing antibody titers fro BVD and IBR. These antibodies may provide some protection for recently ovulated ova.  相似文献
