尾型同源盒转录因子(Cdx2)能促进早期胚胎滋养外胚层(TE)的发育,并在胚胎发育体节的延伸中发挥着重要的作用;非典型性蛋白激酶(aPKC)则控制着细胞对称性和胚胎的极性,以维持哺乳动物神经细胞的功能和干细胞的分化,同时aPKC还参与调节外胚层细胞的发育,此外,Cdx2的表达水平也影响着aPKC在胚胎卵裂球顶端的定位.论文对Cdx2和aPKC在胚胎发育和极性建立方面的作用机制进行综述.  相似文献

囊胚形成过程中,胚胎卵裂球分化为滋养外胚层和内细胞团细胞。囊胚孵出后,滋养外胚层又分化为极滋养层和壁滋养层。滋养外胚层的分化对哺乳动物早期胚胎发育至关重要。研究表明,影响滋养外胚层发育的关键基因主要为Tead4、Cdx2和Eomes,敲除或突变这些基因将导致滋养外胚层细胞分化错误,而滋养外胚层的完整性、渗透性和液体转运功能受损也将导致囊胚形成失败。本文主要综述了哺乳动物滋养外胚层的发育、滋养外胚层发育需要的关键基因、调控滋养外胚层完整性和功能相关的蛋白,以及这些蛋白与滋养外胚层发育关键基因之间可能存在的关系,为进一步研究哺乳动物早期胚胎滋养外胚层的调控机制提供参考。  相似文献

采集屠宰母牛卵巢和输卵管中的卵母细胞,进行体外受精,受精卵继续在体外培养。将发育3d的牛胚胎于42℃下分别培养0.5、2.0和4.0h,与对照组(39℃)牛胚胎相比,接触最高热应激(42℃,4h)处理的胚胎,约509/6的胚胎发育速度明显下降。而处理组牛胚胎发育到第9d的囊胚数、细胞数及内细胞团与滋养外胚层比率与对照组差异不显著(P〉0.05)。表明,温度升高对牛囊胚期胚胎的发育没有明显损害。对9d的囊胚进行基因性别分析发现,正常体外培养的雄胚发育快于雌胚,而在发育第3d受42℃热应激后,到囊胚期生存的胚胎性别比率发生改变,雌胚比率高于雄胚,表明雌性胚胎比雄性胚胎具有更强的抗热应激能力。  相似文献

UBE2C( Ubiquitin- conjugating enzyme 2C )属于E2泛素偶联酶家族成员,较为明确的作用是调控有丝分裂检验点以及控制细胞周期进程,但其在小鼠胚胎中的作用尚不明确。本文首先探讨了小鼠多种组织以及胚胎不同发育时期UBE2C表达图谱,其次在小鼠受精卵中通过显微注射过表达UBE2C分析其对小鼠早期胚胎发育的影响。结果表明:过表达UBE2C后导致胚胎2-细胞发育阻滞,证明UBE2C在胚胎发育过程中具有重要功能,其作用可能是通过降解卵母细胞储存的母源蛋白,从而确保小鼠早期胚胎正常发育进程。  相似文献

以Whiten氏液为基本培养液,研究了卵丘细胞、谷氨酰胺及不同浓度的葡萄糖对HCG注射后17~18h、25~27h和36~38h昆明小鼠胚胎体外发育的影响。结果表明,不论葡萄糖或谷氨酰胺存在与否,卵丘细胞与受精卵的共同培养对胚胎克服体外发育阻断无明显效果。葡萄糖能抑制1-细胞胚胎发育,促进4-细胞以后的胚胎发育。谷氨酰胺是小鼠早期胚胎发育的良好能源物质,可使96%(308/321)的体外发育到2-细胞的胚胎克服阻断而继续发育到4-细胞。谷氨酰胺与葡萄糖联合使用,则可使小鼠受精卵的囊胚发育率达57.5%(229/398)。HCG注射后36~38h的小鼠胚胎能利用葡萄糖。  相似文献

用光镜、电镜及特殊染色方法,对胚兔15-25日龄的胚胎发育阶段牙板(dental laminae,DL)进行形态学观察,表明胚兔在发育过程中DL来源于外胚层衍化的上皮,在可能发生牙齿部位,外胚、间充质这些细胞的相互作用(胚胎诱导),为DL发生创造条件,使口腔黏膜上皮向间充质组织增生的DL形成牙胚;在牙胚形成的过程中,DL的牙源性上皮细胞存在着增生与调亡,以调控牙胚发育生理性的稳定及牙齿的数目、形态的异常,进一步表明胚兔DL在发育过程中存在着形成牙齿遗传的潜能及细胞凋亡现象,为研究人类DL形成、分化、结构及颌骨内牙源性肿瘤和牙畸形提供动物模型。  相似文献

猪胚胎的附植是通过广泛的细胞运动和重塑发生。胚胎的成功附植主要取决于胚胎延伸和滋养外胚层与子宫内膜上皮紧密连接的成功,这种紧密连接保证了孕体摄取其存活所必需的子宫腺上皮分泌物质。这些过程需要子宫和胚胎来源的细胞因子完成子宫内膜和滋养外胚层之间的细胞重塑,营造胚胎附植时的促炎微环境,维持附植阶段免疫系统的平衡,协调子宫和胚胎之间的相互作用等。本文主要综述了猪胚胎附植时期,由子宫和胚胎来源的细胞因子包括白细胞介素-1β(IL-1β)、干扰素(IFN)、成纤维细胞生长因子(FGF)、肿瘤坏死因子-α(TNF-α)、转化生长因子-β(TGF-β)发挥的作用及其作用机理,以期为胚胎附植及发育调控相关研究提供参考。  相似文献

卵母细胞胞质中存在着大量的母源性信息,正常受精卵的发育启动和早期卵裂主要由这些母源性信息所控制。为了评估卵胞质含量对早期胚胎发育的影响,以假显微注射兔原核胚为对照,采用显微注射技术将兔原核期胚胎去除或增加一定量的细胞质:试验一,增加5%和20%的细胞质;试验二,移入5%小鼠MⅡ期卵母细胞胞质。胚胎经显微操作后进行R1/R2序贯培养。结果显示,(1)增量5%组胚胎的2细胞胚(72.00%)、8细胞胚(60.00%)、桑椹胚(58.00%)和囊胚(54.00%)的发育率均显著(P〈0.05)高于增量20%组胚胎(分别为47.62%、35.71%、33.33%和30.95%),但与对照组之间无显著差异(P〉0.05);囊胚细胞数在2个增量组和对照组3者之间也无显著差异(P〉0.05)。(2)将5%小鼠MⅡ期卵胞质移入兔原核胚后,各阶段胚胎发育率和囊胚细胞数与增量5%组、对照组相比均无显著差异;早期胚胎经PCR检测,在2细胞胚(5/5)、8细胞胚(5/5)和桑椹胚(5/5)均全部检测到了供体小鼠mtDNA的D—loop区3’端片段,而在囊胚却仅有1枚(1/5)能够检测到。结果表明,增加少量细胞质不会对兔原核胚的体外发育造成显著影响,但胚胎发育率随着细胞质体积的增加有下降趋势,而异种卵胞质对兔原核胚的发育没有明显影响,在囊胚期虽仍能检测到异种卵胞质中mtDNA的存在,但异种mtDNA会随胚胎发育而逐渐减少。  相似文献

滋养层干细胞(TSc)是形成胎盘组织细胞的前体细胞,与胚胎着床和胎盘形成密切相关。体外分离滋养层干细胞为研究滋养层的发育与功能提供了研究工具和基础。滋养层干细胞已经成功从小鼠附植前囊胚中获得,在猪上也有相关报道,但并没有关于6 d囊胚中分离到猪滋养层干细胞(pTSc)的报道。本试验通过对6 d孤雌囊胚透明带进行划口处理, 饲养层和基础培养液中附加碱性成纤维生长因子(bFGF)和人白血病抑制因子(hLIF)分离猪滋养层干细胞样细胞。同时,通过采用形态学和基因表达分析的方法对获得的猪滋养层干细胞样细胞进行了初步鉴定。结果显示,本试验分离得到的细胞呈现上皮样细胞形态,细胞间结合紧密,边缘光滑,核质比较大,RT-PCR结果显示表达滋养层干细胞标记基因Cdx2,体现了滋养层干细胞特征。结果表明,本试验成功在6 d孤雌囊胚分离得到猪滋养层干细胞样细胞,为后续研究猪滋养层发育提供了试验基础。  相似文献

In the present study, 1‐cell stage mouse embryos were cultured with or without glucose, and their development to the blastocyst stage was compared. Embryos cultured in a glucose‐free medium had a higher percentage of development to the 8‐cell stage, and they had higher developmental speeds compared with those cultured in a glucose‐containing medium. The percentages of embryos that developed to the early blastocyst stage, blastocyst stage and expanded blastocyst stage were much lower than those developed in a glucose‐containing medium. This suggests that the culture of 1‐cell stage embryos in a glucose‐containing medium inhibits development at the 8‐cell stage, and that glucose is necessary for blastocyst formation. Previous reports indicate that, from the 1‐cell stage to the 4‐cell stage, glucose inhibits embryo development. In the present study, exposure of early embryos to a glucose‐free medium improved subsequent embryo development, and there was no difference in the percentage of development to the stages ranging from the 8‐cell stage to the expanded blastocyst stage between embryos cultured in a glucose‐free medium from the 1‐cell stage to the 2‐cell, 4‐cell and 8‐cell stage. This indicates that embryo development is improved when a 1‐cell stage embryo is exposed to a glucose‐free medium before and during the 2‐cell stage, and glucose only has an inhibitory effect on embryo development during conversion from the 1‐cell stage to the 2‐cell stage.  相似文献

为了分析小鼠早期胚胎在体外发育阻滞的原因 ,建立早期胚胎体外培养系统 ,应用几种不同的培养系统对昆明小鼠单细胞胚胎在体外培养了 96~ 12 0 h。结果表明 ,小鼠单细胞胚胎在 M1 6 和 BWW培养液中卵裂均受到阻滞 ,且两者之间差异不显著 (P>0 .0 5 ) ;CZB和 HTF培养液能有效地克服小鼠胚胎的 2 -细胞发育阻滞 ,与 M1 6 和 BWW相比 ,2 -细胞卵裂率和囊胚率均存在显著性差异 (P<0 .0 1) ;在 M1 6 培养液中添加牛磺酸对早期胚胎发育有促进作用 (P<0 .0 1) ;小鼠早期胚胎在 CZB和牛输卵管上皮细胞 (COEC)共培养体系中的卵裂率较对照组明显提高 (P<0 .0 5 )  相似文献

鸡胚因发育过程清楚,长久以来作为基础和应用科学研究领域重要的实验模型,尤其在鸡胚发育早期绒毛尿囊膜阶段,因其血管丰富,是天然的免疫缺陷宿主,是病理学、药理学和肿瘤学等研究领域的理想实验模型。作者简述了发育早期的鸡胚组织结构,并介绍了鸡胚绒毛尿囊膜在肿瘤研究、血管生成、器官移植、烧伤等疾病机理研究中的应用,以及在鸡胚病理模型基础上进行的抗肿瘤药物筛选的应用。重点介绍了鸡胚及禽类细胞系在病毒繁殖和疫苗生产、治疗性蛋白和单抗生产方面的应用研究进展。多种人源病毒、禽源病毒、支原体等可在鸡胚及禽类细胞上增殖,并用于疫苗生产。作者对常用的禽类纤维原细胞和多能干细胞的发展和特点进行了阐述,并总结了商业化的禽类细胞系来源以及部分易感病毒。鸡胚表达系统能够在目的蛋白特定位点产生人源化糖基,减少目的蛋白对人的过敏反应,且禽蛋廉价易得,可作为生产人用单克隆抗体和治疗性蛋白的合适供体。作者介绍了鸡胚及禽类细胞系在生物医药领域应用的最新进展,并对鸡胚作为动物模型在未来的应用进行了展望。  相似文献

Chicken embryo has long been an important experimental model in basic and applied science because of its clear development process,especially in the early development of chicken embryo chorioallantoic membrane stage,due to its abundant blood vessels,it is a natural immunodeficiency host and can be used as an ideal experimental model for pathology,pharmacology and oncology research.The authors briefly described the tissue structure of early stage of chicken embryo,the application of chick embryo chorioallantoic membrane model in tumor,angiogenesis,organ transplantation,burn and other diseases,and the application of anti-cancer drug screening based on the pathological model of chicken embryo.The advances in the application of chicken embryo and avian cell lines in virus reproduction,vaccine production,therapeutic protein production and monoclonal antibody production were reviewed.A variety of human viruses,avian viruses and mycoplasma can proliferate in chicken embryos and avian cell lines and be used in vaccine production.In this paper,the development and characteristics of commonly used avian fibroblasts and pluripotent stem cells were described,and the source of commercial avian cell lines and some susceptible viruses were summarized.Chicken embryo expression system can produce human glycosylates at specific sites of target proteins,reduce the allergic reaction of the target protein to human,and poultry eggs are cheap and easily available,so it can be used as a suitable donor for the production of human monoclonal antibodies and therapeutic proteins.In this paper,the recent progress in the application of chicken embryos and avian cell lines in the field of biomedicine was introduced,and the future application of chicken embryos as animal models was forecasted.  相似文献

试验就胚胎的发育阶段、胚胎保存时间及移植胚胎数等因素对胚胎移植效果的影响情况进行了研究。结果表明,胚胎发育日龄对受体妊娠率没有显著影响(P>0.05),但移植8~16细胞的胚胎的妊娠率略高于2~4细胞(58.49%vs 61.47%)。人员不是影响受体妊娠率的主要因素。胚胎在体外保存0~120min对妊娠率没有显著影响,但以在体外保存0~30 min内移植的妊娠率最高(65.61%)。黄体与移植胚胎的相对位置对受体的妊娠率没有显著影响(P>0.05),但移植双胚的受体的妊娠率较移植单胚高(61.76%vs 45.17%)。同时,试验统计了经胚胎移植产生的羔羊和自然繁殖羔羊之间的差异,发现经胚胎移植产生的波尔山羊母羔的初生重极显著地高于自然繁殖的母羔(P<0.01),但公羔间的初生重以及公、母羔间的体高、体长、胸围、死亡率和性别比例与自然繁殖羔羊差异不显著(P>0.05)。  相似文献

为了提高延边黄牛体细胞克隆重组胚的发育率,研究了在体外培养液中添加不同浓度的甘氨酸对重组胚发育的影响。分别在体外培养液中添加0、7、14、21mmol/L的甘氨酸,结果表明,在体外培养液中添加甘氨酸能够显著提高延边黄牛体细胞克隆重组胚的发育率,且以7mmol/L浓度的甘氨酸囊胚的发育率最高。  相似文献

细胞外囊泡(extracellular vesicles,EVs)作为一种细胞交流媒介,能够通过其携带的miRNA、蛋白质等分子作用于靶细胞/靶组织,影响靶细胞/靶组织的功能和表型。最近研究表明,来自于胚胎、子宫内膜与子宫微环境等的细胞外囊泡参与了哺乳动物胚胎附植过程,通过影响子宫内膜的容受性和滋养层细胞的黏附能力,进而保证了胚胎成功附植和胎儿生长发育。本文主要阐述了细胞外囊泡的发生、生物学功能及其在哺乳动物胚胎附植过程中的作用。  相似文献

Embryonic stem (ES) cells are expanded versions of the inner cell mass cells that compose the early mammalian blastocyst. Components derived from ES cells may contain various bioactive materials (BM) helpful for early preimplantation embryo growth. In this study, we examined the effect of human ES cell derived BM (hES-BM) on in vitro culture of bovine embryos. When bovine parthenogenetic day 2 embryos were cultured in 10% hES-BM, a significantly higher embryo development rate (44.3%) and increased cell numbers were observed relative to control medium containing 3 mg/ml BSA (19.5%; P<0.01). Among the various concentrations (5, 10 and 15%) and days of treatment (2 or 4 days) tested, 10% hES-BM treatment for 4 days provided the best culture environment to support the growth of bovine embryos in vitro (P<0.05). Little difference was observed between 10% hES-BM and 10% FBS treatment in the examined parthenogenetic or in vitro fertilized embryos, although the hES-BM group developed at a slightly better rate. However, the ICM cell numbers were significantly higher in the hES-BM group in irrespective of embryo origin (P<0.05). In addition, the relative levels of pluripotency (Oct4, × 1.8 fold; Nanog. × 3.3 fold), embryogenesis (Stat3, × 2.8 fold; FGF4, × 18.8 fold; E-cad, × 2.0 fold) and growth (Glut5, × 2.6 fold) genes were significantly higher in the 10% hES-BM group than in the 10% FBS group (P<0.05), while the levels of other genes (Bax, Bcl2, MnSOD and Connexin43) were not different. This is the first report examining the positive effects of hES-BM on bovine embryo development in vitro. Based on our results, we conclude that hES-BM can be used as a new protein supplement for bovine preimplantation embryo development.  相似文献
