Morphological changes in gill mitochondria-rich (MR) cells were examined in cultured Japanese eel acclimated to deionized freshwater (DFW), freshwater (FW), 30%-diluted seawater (DSW), and seawater (SW). The gill Na+/K+-ATPase activity was higher in SW-acclimated eel than in those acclimated to DFW, FW, and DSW. Immunocytochemical observations revealed that MR cells in the gill filaments were most developed in SW, whereas MR cells in the lamellae were preferentially observed in DFW, suggesting that filament and lamellar MR cells are responsible for ion secretion and absorption, respectively. In scanning electron microscopic observations, the apical membrane of lamellar MR cells appeared as a flat or slightly projecting disk with a mesh-like structure on its surface. In contrast, the apical membrane of filament MR cells showed a slightly concave surface. Whole-mount immunocytochemistry revealed that most MR cells showed cystic fibrosis transmembrane conductance regulator immunoreaction in their apical region in fish in DSW and SW, but not in those in DFW and FW, indicating that MR cells developed in DSW and SW function as an ion-secreting site. In addition to MR cells, distinct Na+/K+-ATPase immunoreaction was observed in the outermost layer of gill epithelia, suggesting that pavement cells are an additional site of ion uptake in the gills.  相似文献

Branchial activities of Na+,K+-ATPase (ouabain sensitive), Mg2+ ATPase (ouabain insensitive) and kinetic analysis of high and low affinity Ca2+ ATPase were measured inAnguilla anguilla that had been acclimated to demineralized water (DW, Ca < 10 M), freshwater (FW, Ca = 2 mM), and Low calcium freshwater (L-Ca, Ca = 0.9 mM). Na+,K+-ATPase activity decreased while ouabain insensitive activity increased when ambient Ca2+ decreased. Two kinetic forms of Ca2+ ATPase could be resolved in each environmental condition. The stimulation coefficients of both sites or enzymes were not affected by ambient Ca2+ concentrations. The maximal velocity of both the high and the low affinity Ca2+ ATPase was increased when external Ca2+ was decreased during acclimation. The low affinity Ca2+ ATPase and the Mg2+ stimulated enzyme could be a non specific enzyme accepting either Ca2+ or Mg2+. Results are compared with previous results in the literature and in relation to the branchial morphology and ionic exchanges in fish.  相似文献

We explored osmoregulatory ability and mechanisms of ion and water regulation in Japanese eel leptocephali. Tissue osmolality of leptocephali ranged from 360 to 540 mOsm/kg·H2O. Immunocytochemical observations revealed that Na+/K+-ATPase-immunoreactive mitochondrion-rich (MR) cells were distributed over the entire body surface of leptocephali. Using a fluorescent sodium indicator and the chloride test, we localized Na+ and Cl? secreting sites at the apical region of cutaneous MR cells. To further examine drinking behavior and water absorption in the intestine, leptocephali were exposed to seawater containing dextran labeled with Alexa Fluor. To calculate relative water absorption, fluorescent intensity was measured along the digestive tract. Whereas water was hardly absorbed in the stomach and intestine, water absorption predominantly took place in the rectum. Our findings indicate that Japanese eel exert hyposmoregulatory ability as early as during leptocephalus stages, secreting Na+ and Cl? through cutaneous MR cells and primarily absorbing water from ingested seawater in the rectum.  相似文献

An 8‐week feeding experiment was conducted to determine the dietary magnesium (Mg) requirement and physiological responses of Litopenaeus vannamei in low salinity water of 2 g L?1. Casein–gelatin‐based diets supplemented with seven levels of Mg (0, 0.4, 0.8, 1.6, 3.2, 6.4 and 8.0 g kg?1) were fed to juvenile shrimp. Prior to the experiment, the postlarvae were gradually acclimated to the low salinity media and fed with a basal diet (0.5 g Mg kg?1) for 2 weeks. After 8 weeks of feeding, survival ranged from 80.11% to 85.65% with no significant difference among the treatments. Hepatopancreas Mg2+‐ATPase and Na+/K+‐ATPase activities and muscle content of lipid and protein were not significantly affected by graded levels of Mg. The weight gain and mineral (calcium, potassium, sodium and total phosphorus) content of different tissues were significantly affected by dietary Mg levels, while there were no significant differences in ash and zinc content in tissues. The Mg content in tissues except hepatopancreas was maintained relatively constant regardless of dietary treatments. The dietary Mg requirement for optimal growth was 2.60–3.46 g Mg kg?1 by using the polynomial regression analysis based on growth.  相似文献

We evaluated the performance of whiteleg shrimp Penaeus vannamei (Boone, 1931) in response to different stocking densities and acclimation periods. Shrimp postlarvae were acclimated from seawater (30 g L?1) to low‐salinity well water (<1.0 g L?1) at a constant hourly reduction rate of 40, 60, 80 and 100 h. After acclimation to low‐salinity well water, postlarvae from each acclimation time treatment were stocked in three replicate tanks at densities of 50, 100, 150 or 200 shrimps m?2 for 12 weeks of growth. Salinity averaged <1.0 g L?1 for each growth study. The different treatments resulted in significant differences in both the final body weight and the survival rate (SR). Shrimp acclimated for 100 h showed substantially improved survival (83%) relative to shrimp acclimated for shorter periods. Shrimp yields for all cultured periods ranged from 0.32 kg m?2 in tanks stocked at 50 m?2 to 1.14 kg m?2 in tanks stocked at 200 m?2. We conclude that whiteleg shrimp can be successfully grown in low‐salinity well water, and that the growth, production output and SRs are significantly higher when shrimp are acclimated for longer periods.  相似文献

Gibel carp (Carassius auratus gibelio) of mean initial weight 3.1 g were fed one of seven casein‐dextrin‐based diets containing graded levels of magnesium (Mg) (39, 120, 220, 380, 700, 1600 and 2900 mg kg?1) for 3 months with the waterborne Mg concentration of 10.6–12.7 mg L?1. Magnesium sulphate was used as the supplementation Mg source in the diets. The experiment was carried out in a flow‐through system. Growth, survival rate, Na+/K+‐ATPase, Mg2+‐ATPase and tissue mineral contents were measured to investigate the effect of dietary magnesium in gibel carp. At the end of the experiment, the hepatopancreas of fish were collected for enzyme determination. The hepatopancreas, vertebrae and whole body were collected for tissue magnesium content analysis. After 3 months, dietary magnesium supplementation did not improve the growth performance, including feed intake, weight gain and feed conversion efficiency of juvenile gibel carp. On the contrary, negative impacts on survival, reduced growth performance and dramatically decreased Na+/K+‐ATPase, Mg2+‐ATPase and superoxide dismutase activities were observed in gibel carp fed a high Mg diet of 2900 mg kg?1. Although serum and hepatopancreas Mg and Ca contents were not affected by dietary Mg supplementation, vertebrae and whole‐body Mg contents increased significantly with the increasing dietary Mg concentrations. Based on the relationship between whole‐body Mg retention and dietary Mg concentration, a suitable dietary Mg level of 745 mg kg?1 could be estimated for gibel carp. It could be concluded that dietary Mg supplementation did not improve the growth performance, but could increase vertebrae Mg contents of gibel carp. Considering the adverse effects, a dietary Mg concentration of above 2900 mg kg?1 is not recommended and it should be careful to supplement magnesium in practical diets for gibel carp as most feed ingredients contain high magnesium concentrations.  相似文献

Sakhalin taimen Hucho perryi populations have decreased drastically to near extinction. It is urgent to establish an effective conservation strategy based on an understanding of the characteristics of migration and habitat use of this species. We examined the migration history of anadromous Sakhalin taimen captured off the Sarufutsu coast, northern Hokkaido, Japan, using otolith Sr:Ca ratios and also examined the relationship between their otolith Sr:Ca ratios during freshwater and seawater residence in a rearing experiment. Otolith Sr:Ca ratios of some fish from the Sarufutsu coast showed freshwater levels (0.5–4.0 × 10−3) near the core, which thereafter increased to brackish water levels (4.0–6.0 × 10−3), and then to seawater levels (6.0–10.0 × 10−3) in the outermost regions. Those findings indicate that specimens from the Sarufutsu coast migrated to the brackish water region or the sea and spent most of their lives there. The anadromous migration pattern including the timing of downstream migration seems to be flexible among individuals in the species. They migrate between freshwater and seawater or brackish water several times during their lives, showing extensive habitat use. It is essential to secure the continuity among the freshwater, brackish water, and seawater areas for their effective conservation.  相似文献

The high sperm density, together with the short spermatozoa swimming time, makes European eel sperm manipulation and assessment for quality difficult. Two diluting media (K15 and K30) previously designed for Japanese eel sperm were tested. After 24 h, European eel sperm showed significant reduction in the percentage of motile spermatozoa after activation and different motility parameters (VAP, angular velocity; VCL, curvilinear velocity; VSL, straight line velocity; BCF, beating cross frequency), concluding that these media are not suitable to preserve the sperm of this species. After a hormonal treatment to induce spermiation, sperm volume, density and motility were recorded at weekly samplings. The variation of the osmolality (325–330 mOsm kg−1), pH (8.4–8.6) and the ionic composition (concentration of Na+, K+, Mg2+ and Ca2+) of the seminal plasma were registered. Physio-chemical results were related with sperm quality throughout the treatment, to determine which must be the suitable characteristics of one extender for the sperm of this species, and to find the best conditions to obtain suitable cryopreservation media for European eel sperm. K+ concentration increased, while Ca2+ and Mg2+ concentrations showed a progressive reduction in correlation with the sperm quality improvement. Na+ showed a decreasing, but not significant tendency. P1 and P2 freezing media were designed considering the physio-chemical parameters as well as the ionic composition shown by the best quality sperm samples, and then compared with the previously described solutions, TNK and K30. Sperm quality was determined, checking the percentage of motile spermatozoa and motility parameters using computer-assisted sperm analysis (CASA) software. Samples were frozen after dilution (1:5, 1:20, 1:100) in different freezing media supplemented with 10% dimethyl sulfoxide (DMSO). After thawing, samples frozen with low dilution ratio (1:5) in TNK and P1 media showed higher, although not significant, spermatozoa survival (35.5 ± 14.5 and 36.6 ± 6.7%). The addition of l-α-phosphatidylcholine to the media seems to have a positive effect, as reported in the Japanese eel.  相似文献

Otolith microchemical analyses of the strontium (Sr) and calcium (Ca) concentrations in the eels Anguilla japonica and A. anguilla caught in Tokyo Bay were undertaken to reconstruct the eels’ migratory histories. A. japonica in the yellow stage (immature stage) were caught in a bay without any adjacent rivers or streams. A. anguilla was in the silver stage (early maturing stage), and the eel was confirmed to have just begun spawning migration to the open ocean from Tokyo Bay based on the otolith Sr:Ca ratios, which showed a typical catadromous life history with low Sr:Ca ratio values throughout the eel’s life after recruitment. The mean Sr:Ca ratios in A. japonica from the elver mark to the otolith edge indicated the eels belonged to several general categories of migratory histories, including sea eels (average Sr:Ca ratio ≥6.0 × 10−3) and estuarine eels (average Sr:Ca ratio 2.5 to 6.0 × 10−3) based on the criteria reported previously in A. japonica. All eels had a certain freshwater life period, although the period was highly variable among fish. These results indicate that A. japonica has a flexible pattern of migration, with the ability to adapt to various habitats and salinities.  相似文献

A feeding experiment was conducted to examine the efficacy of a vitamin C (L‐ascorbic acid, AsA) derivative, l ‐ascorbyl‐2‐monophosphate Na/Ca (AMP‐Na/Ca) by comparing to l ‐ascorbyl‐2‐monophosphate‐Mg (AMP‐Mg) in terms of growth, AsA content in tissues, and hematology in yellowtail Seriola quinqueradiata juvenile (1.10 ± 0.01 g). Furthermore, a stress resistance test for the species fed AMP‐Na/Ca was also conducted. In experiment 1, 11 test pellet diets with different levels of AMP‐Na/Ca (0, 12, 43, 88, 440 and 881 mg AsA kg?1diet) and AMP‐Mg (0, 16, 52, 106, 595 and 1164 mg AsA kg?1 diet) were formulated and fed to the yellowtail three times per day for 50 days. In both the vitamin C sources, the survival rates of the fish fed diets without supplemental AsA were lower than 50% at day 20, and more than 50% mortality occurred in fish that fed the diets containing 12 or 16 mg AsA kg?1 after day 30. However, no significant differences were detected in survival and growth among fish that fed the diets containing more than 43 mg AsA kg?1. Liver and brain AsA concentrations generally increased with increasing dietary AsA level in both sources. In experiment 2, test diets were formulated to contain 43, 88, 440 and 881 mg AsA kg?1 using AMP‐Na/Ca, and after 60 days of feeding, yellowtail juveniles were subjected to low salinity and air exposure stress. The fish that received diets with 440 mg AsA kg?1 showed significantly higher tolerance to low salinity stress and higher survival rate in air exposure stress than those of other groups. The present study demonstrated that yellowtail juveniles could utilize AMP‐Na/Ca as an AsA source like AMP‐Mg, and that supplementation of 43–52 mg AsA kg?1 diet was optimum for normal growth. However, this study showed that dietary inclusion of 440 mg AsA kg?1 would be necessary to improve stress resistance of this species.  相似文献

There is expanding interest in the culture of the Australian shortfin eel Anguilla australis Richardson; however, there is a lack of fundamental biology and husbandry information necessary to further develop an industry within Australia. The present study was undertaken to gain a preliminary understanding of basic husbandry requirements for rearing of juvenile A. australis (glass eels and elvers) in tanks and earthen ponds. Newly caught glass eels were successfully acclimated to culture conditions. During tank culture trials, specific growth rates (SGR) and survival rates ranged from ?2.1 to 2.8% day?1 and 52% to 100% respectively. Glass eels weaned onto a commercial eel diet exhibited a significantly greater SGR and survival rate than those weaned onto a commercial trout diet. Glass eels weaned onto an eel diet over a 15‐day period grew slightly faster than eels weaned over a 5‐day period, but survival rates were not significantly different for each treatment. SGRs (up to 2.8% day?1) were significantly higher for glass eels fed at 9 and 12% day?1 than at 6% day?1. Stocking densities between 2.5 kg m?3 and 30 kg m?3 did not influence either SGR or survival rates. SGRs were significantly higher for glass eels cultured at 25 °C than at lower temperatures. During pond culture trials, SGRs and survival rates ranged from 1.36 to 1.65% day?1 and 39% to 77% respectively. The SGR and survival rates of juvenile eels stocked into ponds receiving supplementary feeding with a commercial eel diet were not significantly different to those of eels stocked into ponds that did not receive supplementary feeding.  相似文献

Viable chloride and pavement cells were isolated from the gill epithelium of Japanese eel, Anguilla japonica, by a 3-step percoll gradient centrifugation at low speed. Viability of the isolated cells were tested by the trypan blue exclusion test, rhodamine-123, or pre-labelling the cells with fluo-3 Ca2+ dye and examined by laser confocal microscopy. Isolated chloride cells responded to ionomycin with a rapid increase in Ca2+ fluorescence, which was abolished by chelating external Ca2+ with EGTA. Peptide hormones, including arginine vasotocin, isotocin, insulin-like growth factor I and II, and urotensin I increased Ca2+ entry, urotensin II had no effect, and eel corpuscles of Stannius extract reduced residual Ca2+ fluorescence. Isolated chloride cells and pavement cells from eels were analyzed for their enzymatic activities involved in intermediary and nitrogen metabolism. Chloride cells had high levels of glutaminase I, glutamate dehydrogenase, glutamate oxalacetate transaminase, HCO3-ATPase and carbamoyl phosphate synthetase II. Pavement cells had highly active glucose-6-phosphate dehydrogenase and AMP deaminase. Both had high levels of lactate dehydrogenase compared with other tissues.  相似文献

In order to determine the salinity effect on otolith strontium (Sr):calcium (Ca) ratios, the Sr and Ca contents of otoliths of Sakhalin taimen, Hucho perryi, reared for 60 days in four salinities (freshwater, one-third seawater, two-thirds seawater, and full seawater) were examined. The Ca and Sr contents and the resultant Sr:Ca ratios in the rearing water increased linearly with salinity. Otolith Sr:Ca ratios were also found to be positively correlated with salinity. However, the Sr:Ca did not correspond to temperature fluctuations during the experiment. These results indicate that Sr:Ca ratios in otoliths can be used to reconstruct the migratory history of the fish by differentiating when the fish migrated between freshwater and seawater.  相似文献

Osmoregulation plays an important role in the migration process of catadromous fish. The osmoregulatory mechanisms of tropical marbled eel (Anguilla marmorata), a typical catadromous fish, did not gain sufficient attention, especially at the molecular level. In order to enrich the protein database of A. marmorata, a proteomic analysis has been carried out by iTRAQ technique. Among 1937 identified proteins in gill of marbled eel, the expression of 1560 proteins (80 %) was quantified. Compared with the protein expression level in the gill of marbled eel in freshwater (salinity of 0 ‰), 336 proteins were up-regulated and 67 proteins were down-regulated in seawater (salinity of 25 ‰); 33 proteins were up-regulated and 32 proteins were down-regulated in brackish water (salinity of 10 ‰). These up-regulated proteins including Na+/K+-ATPase, V-type proton ATPase, sodium–potassium–chloride co-transporter and heat shock protein 90 were enriched in many KEGG-annotated pathways, which are related to different functions of the gill. The up-regulated oxidative phosphorylation and seleno-compound metabolism pathways involve the synthesis and consumption of ATP, which represents extra energy consumption. Another identified pathway is the ribosome pathway in which a large number of up-regulated proteins are involved. It is also more notable that tight junction and cardiac muscle contraction pathways may have correlation with ion transport in gill cells. This is the first report describing the proteome of A. marmorata for acclimating to the change of salinity. These results provide a functional database for migratory fish and point out some possible new interactions on osmoregulation in A. marmorata.  相似文献

Sperm density, mineral and organic composition of the seminal plasma and their physiological relationship were investigated in the Caspian brown trout (Salmo trutta caspius). To establish a rapid and accurate method for assessment of sperm density, three different techniques were used: sperm counting, spectrophotometry (at 480 nm) and determination of the spermatocrit. The seminal plasma contained 159.26±8.84 mM sodium (Na), 33.72±2.01 mM potassium (K), 133.04±5.96 mM chlorine (Cl), 1.68±0.2 mM calcium (Ca) and 0.988±0.13 mM magnesium (Mg). The following organic components were found: total protein 0.75±0.14 mg 100 mL−1, cholesterol 2.86±0.58 mg L−1 and glucose 3.81±1.04 mM L−1. The mean sperm density was estimated to be 3.3 × 109 spermatozoa mL−1. The spermatocrit (%) ranged from 25 to 52 in sperm samples. Highly significant linear relationships were found between sperm density and spermatocrit (R2=0.703, P<0.001) and sperm density and optical density (R2=0.909, P<0.001), indicating that optical density can be used as a quick and accurate method of estimating sperm density. Significant relationships were also found between sperm density and Ca, Mg and total protein of seminal plasma. A significant correlation was also observed between the Ca and Mg concentrations (R2=0.774, P<0.01). The following correlations were observed between mineral and organic components: total protein and Ca (R2=0.462, P<0.05), total protein and Mg (R2=0.518, P<0.05) and glucose and Cl (R2=0.374, P<0.05). These parameters should be considered when developing procedures for either artificial fertilization or for cryopreservation of sperm.  相似文献

Marbled eels, Anguilla marmorata (Quoy & Gaimard), cultured in Taiwan exhibited haemorrhage and mortality in January 2012. The severely diseased eels bled from the gills and showed congestion of the central venous sinus of the gill filaments and haemorrhage throughout the body similar to viral endothelial cell necrosis of eel. In this study, a novel polyomavirus (AmPyV) was isolated from the diseased eels using the AMPF cell line established from the pectoral fin of healthy marbled eels. AmPyV was found to encode a long T‐antigen orthologous gene. Phylogenetic analysis showed that AmPyV was closely related to Japanese eel endothelial cell‐infecting virus. PCR assays revealed AmPyV infection throughout the systemic organs. AmPyV proliferated in the AMPF, EK‐1 and EO‐2 cells at temperatures 25–30 °C, and the progeny virus yields were 107.0, 107.4 and 107.7 TCID50 mL?1, respectively. The purified virions were icosahedral particles, 70–80 nm in diameter. No clinical signs or mortality was observed among the eels injected with the virus; however, the virus was reisolated from the brain, eyes, kidneys, fins and gills of infected eels 2 month after injection. Our results suggest that AmPyV exhibits a latent infection. Pathogen of the disease needs to study further.  相似文献

The involvement of the freshwater fish gill chloride cells (CCs) in trans-branchial calcium uptake (JinCa2+) was investigated. This was accomplished by assessing the interspecific relationships between the apical surface area of CCs exposed to the external environment and JinCa2+. Three species of freshwater teleosts, the rainbow trout (Oncorhynchus mykiss), the American eel (Anguilla rostrata) and the brown bullhead catfish (Ictalurus nebulosus), were used. Chronic (ten-day) treatment with cortisol in each species was used as a tool to evoke variations in both JinCa2+ and gill CC morphology in order to assess intraspecific relationships between CC surface area and JinCa2+. The results of quantitative morphometry, based on analysis of scanning electron micrographs, demonstrated that catfish possessed the lowest fractional area of exposed CC (CCFA) on the gill filament epithelium (12,744 ± 2248 m2/mm2) and was followed, in increasing order, by American eel (21,355 ± 981 m2/mm2) and rainbow trout (149,928 ± 26,545 m2/mm2). With the exception of catfish, chronic treatment with cortisol caused significant increases in CCFA owing to proliferation of CCs and/or enlargement of individual CCs (eel only). The rates of JinCa2+ closely reflected the CC fractional area in each species. The results of correlation analysis revealed significant correlations between CC fractional area and JinCa2+ in trout and eel. Owing to the absence of an effect of cortisol treatment, there was no significant correlation in catfish because of insufficient variation in CC fractional area in this species. CC fractional area was significantly correlated with JinCa2+ among the three species examined. These results suggest that CC is involved in calcium uptake in freshwater teleosts and that both intra- and interspecific differences in the rates of calcium uptake can be accounted for by variability in the surface area of exposed CCs on the gill epithelia.  相似文献

Growth hormone (GH) effectively promotes seawater (SW) adaptation in salmonids, but little is known of its effect in tilapias. Experiments were performed to investigate the effects of recombinant eel GH (reGH) on osmoregulatory actions and ultrastructural features of gill chloride cells in juvenile tilapia, Oreochromis niloticus. Tilapia showed a markedly improved SW survival, when directly transferred from freshwater (FW) to 62.5% SW 24h after a single reGH injection (0.25 or 2.5 µg g-1) or 3 reGH injections (0.25 µg g-1 every other day). Plasma Na+ and Mg2+ levels were significantly reduced by reGH (0.25 and 2.5 µg g-1) compared with saline injections; Ca2+ concentrations were reduced significantly by high dose of reGH (2.5 µg g-1) after SW transfer. However, fish failed to survive more than 24h when directly transferred to 70% SW, although the fish treated with reGH could survive longer than the controls. When examined by electron microscopy, the chloride cells were identified as mitochondrion-rich and an extensive tubular system was induced by GH treatment. The results of the present study suggest that, similar to its effect on salmonids, GH also exerts acute osmoregulatory actions and enhances SW adaptation in juvenile tilapia. A quantitative analysis of the essential mineral content (mg kg?1 dry weight) was carried out in 31 samples, including molluscs, crustaceans, fish and meals in an attempt to identify those most suitable for formulating cephalopod diets. The mineral ratios (MR: content in the test sample/content in whole Octopus vulgaris) were used as index of nutritional quality. Both crustaceans and oysters presented an optimal profile that covered the macro‐ and microelements composition of O. vulgaris. These samples differed from the rest by their higher Ca, Mg, B, Cu and Zn contents based on a principal component analysis. Fish were deficient in macroelements, such as Na (MR: 70–420 g kg?1) and Mg (MR: 220–690 g kg?1), but would be good source of K, Ca and P. Most fish were also deficient in Fe, Zn and Cu, although the copper content would be the most affected (MR: 3–130 g kg?1). Fish and krill meals showed a high content of Ca and P, although both would be deficient in Na (MR: 440–470 g kg?1) and Cu (130–540 g kg?1), along with K, Fe and Zn in krill and Mg and B in fish. Among the plant meals, sunflower and soybean were the most appropriate, presenting higher total content of minerals and MRs above 1000 g kg?1 for all minerals, except Na, Cu and Zn.  相似文献
