Published research has indicated that ethyl 2-methylpropanoate, ethyl 2-methybutanaote, ethyl 3-methylbutanoate, hexanoic acid, butanoic acid, and 3-methylbutanoic acid are responsible for fruity fermented (FF) off-flavor; however, these compounds were identified in samples that were artificially created by curing immature peanuts at a constant high temperature. The objective of this study was to characterize the volatile compounds contributing to naturally occurring FF off-flavor. Volatile compounds of naturally occurring FF and no-FF samples were characterized using solvent-assisted flavor evaporation (SAFE), solid phase microextraction (SPME), gas chromatography-olfactometry (GC-O), and gas chromatography-mass spectrometry (GC-MS). Aroma extract dilution analysis (AEDA) identified 12 potent aroma active compounds, none of which were the previously identified esters, with no consistent differences among the aroma active compounds in no-FF and FF samples. Hexanoic acid alone was identified in the naturally occurring FF sample using the SAFE GC-MS methodology, whereas two of the three previously identified esters were identified in natural and artificially created samples. The same two esters were confirmed by SPME GC-MS in natural and artificially created samples. This study demonstrated the need for caution in the direct application of data from artificially created samples until those compounds are verified in natural samples. However, these results suggest that a laboratory method using SPME-GC techniques could be developed and correlated on an ester concentration versus FF intensity basis to provide an alternative to sensory analysis for detection of FF off-flavor in peanut lots.  相似文献

A dynamic solid-phase microextraction (SPME) method to sample fresh headspace volatile compounds released during the grinding of roasted coffee beans was described and the analytical results using gas chromatography/mass spectrometry (GC/MS) and GC/olfactometry (GC/O) were compared to those of the conventional static SPME sampling methods using ground coffee. Volatile compounds released during the grinding of roasted coffee beans (150 g) were obtained by exposing the SPME fiber (poly(dimethylsiloxane)/divinylbenzene, PDMS/ DVB) for 8 min to nitrogen gas (600 mL/min) discharged from a glass vessel in which the electronic coffee grinder was enclosed. Identification and characterization of volatile compounds thus obtained were achieved by GC/MS and GC/O. Peak areas of 47 typical coffee volatile compounds, separated on total ion chromatogram (TIC), obtained by the dynamic SPME method, showed coefficients of variation less than 5% (n = 3) and the gas chromatographic profile of volatile compounds thus obtained was similar to that of the solvent extract of ground coffee, except for highly volatile compounds such as 4-hydroxy-2,5-dimethyl-3(2H)-furanone and 4-ethenyl-2-methoxyphenol. Also, SPME dilution analysis of volatile compounds released during the grinding of roasted coffee beans showed linear plots of peak area versus exposed fiber length (R (2) > 0.89). Compared with those of the headspace volatile compounds of ground coffee using GC/MS and GC/O, the volatile compounds generated during the grinding of roasted coffee beans were rich in nutty- and smoke-roast aromas.  相似文献

固相微萃取技术是一项新型的提取技术,它具有很多的优点:需样量少、简单、快捷、以及无需溶剂等。为了解苦菜中的挥发性成分的抗便秘作用,用固相微萃取-气质联用方法对挥发成分进行了定性分析及定量测定。该法采用顶空固相微萃取技术,用65μm聚乙二醇-二乙烯基苯萃取纤维头取样,在25℃下萃取30min。通过实验,得出以下结论:本实验通过65μmCW-DVB固相微萃取GC-MS方法鉴定了苦菜挥发性成分中的41种化合物,主要包括戊酸乙酯,顺-3-己烯基异戊酸,苯乙烯,3-甲基丁酸乙酯,3-甲基丁酸乙酯等,经文献检索尚未发现这些物质有“缓解便秘”的报道;对苦菜的营养成分分析发现:其粗纤维含量较高;苦菜具有很高的营养价值,适合进行综合开发  相似文献

Volatiles from stored Kuerle fragrant pears (Pyrus serotina Reld) were studied using high-resolution gas chromatography and the solid-phase microextraction (SPME) method of gas chromatography/mass spectrometry (GC/MS). The dominant components were hexanal, ethyl hexanoate, ethyl butanoate, ethyl acetate, hexyl acetate, ethanol, alpha-farnesene, butyl acetate, and ethyl (E,Z)-2,4-decadienoate. By using GC-olfactometry, it demonstrated that the volatile compounds from SPME were responsible for the aroma of the Kuerle fragrant pear. The levels of sugars, organic acids, and phenolic acids in Kuerle fragrant pears were investigated using high-performance liquid chromatography (HPLC). Fructose was the dominant sugar, followed by glucose and sucrose. With increasing storage time, sucrose levels decreased; however, changes in fructose and glucose levels were not remarkable. There was a slight decrease in flesh firmness during storage. The general soluble solids concentration (SSC) declined slightly after 5 months storage. Some aroma-related volatile components increased during storage, while others decreased, especially the esters. The organic acids and phenolic acids also changed. The flavor of the Kuerle fragrant pears was affected by the change of volatile compounds and changes in chemical and physical properties.  相似文献

A biocatalyst was prepared by immobilization of Saccharomyces cerevisiae strain AXAZ-1 on delignified cellulosic material (DCM). Repeated batch fermentations were conducted using these biocatalysts and free cells, separately, at temperatures of 20, 15, and 10 degrees C. Solid phase microextraction (SPME) was used in monitoring the formation of volatile alcohols, acetate esters, and ethyl esters of fatty acids. The kinetics of volatile production were similar for free and immobilized cells. In all cases immobilized cells showed a better rate of volatile production, which was directly connected to sugar consumption. The main difference observed was in propanol production, which increased with temperature decrease for the immobilized cells, whereas it remained constant for the free ones. In the case of immobilized cells significant amounts of esters were also produced. It is well-known that esters contribute to the fruity aroma of wine. It was also established that SPME is a very sensitive, accurate, and reliable technique and can be used without any reservation in the characterization of volatile constituents of wine.  相似文献

Many volatile alcohol and ester metabolites of the lesser grain borer (LGB, Rhyzopertha dominica) cultured on wheat grain were identified. Volatiles from infested samples at 80 degrees C were collected on Tenax absorbent, thermally desorbed, and analyzed by gas chromatography (GC) using infrared (IR) and mass (MS) detectors for component identification. A solid-phase microextraction (SPME) technique was used to analyze selected samples with a GC-MS system set up for obtaining chemical ionization mass spectra. SPME was also used in a synthesis process required to identify ester metabolites. Predominant compounds in LGB-infested grains were 2-pentanol and its esters of 2-methyl-2-pentenoic (A) and 2,4-dimethyl-2-pentenoic (B) acids, which are known aggregation pheromones, dominicalures 1 and 2. 2-Pentanol esters of saturated A, beta-keto- and beta-hydroxy derivatives of A and B, homologues of A and B, and acid moieties lacking the 2-methyl substitution were found. Other straight- and branched-chain secondary alcohols and their esters were also observed. Reexamination of GC-MS-IR data acquired in previous investigations of LGB cultured on sorghum grain and commercial samples in a grain odor study showed the presence of many LGB metabolites in addition to the known dominicalures.  相似文献

In this study, the optimization and comparison of an internally cooled fiber [cold fiber with polydimethylsiloxane (PDMS) loading] and several commercial solid-phase microextraction (SPME) fibers for the extraction of volatile compounds from tropical fruits were performed. Automated headspace solid-phase microextraction (HS-SPME) using commercial fibers and an internally cooled SPME fiber device coupled to gas chromatography-mass spectrometry (GC-MS) was used to identify the volatile compounds of five tropical fruits. Pulps of yellow passion fruit (Passiflora edulis), cashew (Anacardium occidentale), tamarind (Tamarindus indica L.), acerola (Malphigia glabra L.), and guava (Psidium guajava L.) were sampled. The extraction conditions were optimized using two experimental designs (full factorial design and Doehlert matrix) to analyze the main and secondary effects. The volatile compounds tentatively identified included alcohols, esters, carbonyl compounds, and terpernes. It was found that the cold fiber was the most appropriate fiber for the purpose of extracting volatile compounds from the five fruit pulps studied.  相似文献

Changes in the volatility of selected flavor compounds in the presence of nonvolatile food matrix components were studied using headspace solid-phase microextraction (HS-SPME) combined with GC-MS quantification. Time-dependent adsorption profiles to the SPME fiber and the partition coefficients between different phases were obtained for several individual volatiles, showing that HS-SPME analysis with a short sampling time can be used to determine the "true" headspace concentration at equilibrium between the headspace and a sample matrix. Equilibrium dialysis followed by HS-SPME/GC-MS was carried out to confirm the ability of HS-SPME extraction for monitoring the free volatile compounds in the presence of proteins. In particular, a short sampling time (1 min) avoided additional extraction of volatiles bound to the protein. Interactions between several selected flavor compounds and nonvolatile food matrix components [beta-lactoglobulin or (+)-catechin] were also studied by means of HS-SPME/GC-MS analysis. The volatility of ethyl hexanoate, heptanone, and hexanal was significantly decreased by the addition of beta-lactoglobulin compared to that of isoamyl acetate. Catechin decreased the volatility of ethyl hexanoate and hexanal by 10-20% and increased that of 2-heptanone by approximately 15%. This study indicates that HS-SPME can be a useful tool for the study of the interactions between volatile compounds and nonvolatile matrix components provided the kinetic and thermodynamic behavior of the volatiles in relation to the fiber chosen for the studies is carefully considered.  相似文献

Volatile esters contribute important floral and fruity sensory properties to wine. Numerous factors influence the biosynthesis and hydrolysis of esters throughout yeast fermentation; however, methods to monitor the dynamic changes in ester production that occur during winemaking processes are limited. In this study, we showed that solid phase microextraction (SPME), a rapid, solventless sampling procedure, combined with GC/MS analysis is a useful method for the nearly continuous analysis of volatile compounds such as esters that are produced during fermentation. Accuracy, precision, and limits of quantification were comparable to those of other sample preparation methods such as liquid-liquid extraction. Using GC/MS-SPME to monitor fatty acid ethyl esters and acetate esters, we obtained detailed information on the production patterns of ester formation during fermentation. This method now enables the monitoring of volatiles during fermentation and can provide greater insight into yeast metabolism and flavor formation.  相似文献

Solid-phase microextraction (SPME) with a polydimethylsiloxane fiber coupled with gas chromatography-mass spectrometry (GC-MS) was applied to the study of variability in volatiles released by 13 apple varieties. The relative amounts of 40 esters and alpha-farnesene were determined. Principal component analyses of these results clustered the apples into three groups according to skin color: red, green, and red-green. Total ester contents were highest with the red cluster apples, and the green cluster apples had the highest alpha-farnesene levels. This technology was also applied to the monitoring of changes in volatiles for apples removed from controlled-atmosphere storage with subsequent storage at 4 degrees C and room temperature. Total ester contents increased 25-fold, with the greater increases coming at room temperature, whereas alpha-farnesene levels increased only 5-fold. For apples stored at room temperature, after 11 days, the amount of increase was inversely proportional to the size of the ester: levels of smallest esters (molecular weight 116) increased 12.5-fold, and the largest esters (molecular weight 228) increased approximately 1.3-fold.  相似文献

The aroma compounds of young and aged Chinese "Yanghe Daqu" liquor samples were extracted by solid phase microextraction (SPME) and analyzed by gas chromatography (GC)-olfactometry dilution analysis. The original liquor samples were diluted with deionized water to give a final alcohol content of 14% (v/v). The samples were stepwise diluted (1:1) with 14% (by volume) ethanol-water solution and then extracted by headspace SPME. The samples were preequilibrated at 50 degrees C for 15 min and extracted with stirring at the same temperature for 30 min prior to injection into GC. The aroma compounds were identified by both GC-MS and GC-olfactometry using DB-Wax and DB-5 columns. The results suggested that esters were the major contributors to Yanghe Daqu liquor aroma. Ethyl hexanoate, ethyl butanoate, and ethyl pentanoate had very high flavor dilution values in both young and aged liquors (FD > 8192). Methyl hexanoate, ethyl heptanoate, ethyl benzoate, and butyl hexanoate could also be very important because of their high flavor dilution values (FD > or = 256). Moreover, two acetals, 1,1-diethoxyethane and 1,1-diethoxy-3-methylbutane, also were shown high flavor dilution values in Yanghe Daqu liquors (FD > or = 256). Other aroma compounds having moderate flavor dilution values included acetaldehyde, 3-methylbutanol, and 2-pentanol (FD > or = 32). Comparing young and aged liquors, the aroma profiles were similar, but the aroma compounds in the aged sample had higher flavor dilution values than in the young ones.  相似文献

White wines experimentally produced from the white grape varieties Loureira, Dona Branca, and Treixadura have been analyzed over four consecutive harvests. The contents of monoterpenes, volatile phenols, alcohols, sulfur components, acetates, fatty acids, and ethyl esters were determined by gas chromatography (FID) and gas chromatography-mass spectrometry. The mean values from four vintages confirmed that these wines have characteristic profiles. Loureira wines are characterized by a high content of free terpenes, 1-hexanol, ethyl acetate, and fatty acids. Dona Branca wines present the highest concentrations of methanol and sulfur constituents, and the lowest concentrations of higher alcohols, acetates, diethyl succinate, and ethyl esters. The levels of monoterpenes in Treixadura wines are very low, but they have the highest concentrations of volatile phenols, principally due to the vanillin, diethyl succinate, ethyl lactate, and ethyl esters. These results were confirmed by principal component and linear discriminant analysis, which show a clear differentiation among these wines as a function of the varietal origin.  相似文献

Substrates controlling the formation of branched chain volatile esters in ripening bananas were investigated by the application of alcohol and amino acid precursors to whole fruit and tissue samples. The resulting changes in the profile of the volatile esters were determined using SPME and GC. These changes revealed the selectivity characteristics of the esterification enzyme AAT, the availability of acyl CoA's for ester formation, and the role of substrate supply on volatile production. The results obtained suggest that substrate supply is a major determinant of the quantitative and qualitative composition of the resulting aroma profile.  相似文献

The development and application of a solid-phase microextraction (SPME) method in the analysis of vanilla extracts and vanilla flavorings was studied. The SPME method was developed to be used in conjunction with gas chromatography mass spectrometry (GC-MS). The optimized SPME sampling parameters for the determination of the volatile components included a poly(acrylate) fiber, a 40-min sampling time at room temperature, and a 2-min desorption time. The reproducibility of the method was good, with a percent relative standard deviation between 2.5 and 6.4% for the target compounds. The data suggest that the origin of natural extracts can be readily determined from the GC profile and that differences exist between nature-identical and synthetic flavorings and the natural extracts. The method also has potential for identifying the type of vanilla extract/flavoring used to flavor food.  相似文献

The aroma-active compounds in two apple ciders were identified using gas chromatography-olfactometry (GC-O) and GC-mass spectrometry (MS) techniques. The volatile compounds were extracted using solvent-assisted flavor evaporation (SAFE) and headspace solid-phase microextraction (HS-SPME). On the basis of odor intensity, the most important aroma compounds in the two apple cider samples were 2-phenylethanol, butanoic acid, octanoic acid, 2-methylbutanoic acid, 2-phenylethyl acetate, ethyl 2-methylbutanoate, ethyl butanoate, ethyl hexanoate, 4-ethylguaiacol, eugenol, and 4-vinylphenol. Sulfur-containing compounds, terpene derivatives, and lactones were also detected in ciders. Although most of the aroma compounds were common in both ciders, the aroma intensities were different. Comparison of extraction techniques showed that the SAFE technique had a higher recovery for acids and hydroxy-containing compounds, whereas the HS-SPME technique had a higher recovery for esters and highly volatile compounds.  相似文献

Solid phase microextraction (SPME) is an ideal sample preparation technique because of its speed and solvent-free features. Sampling by SPME is selective and only the dissolved concentration is measured, which allows measurement of the bioavailable fraction of a contaminant in aqueous media. One potential application of SPME is for analysis of enantiomers of chiral contaminants in environmental samples. In this study, a method was developed for determining enantiomers of (Z)-cis-bifenthrin and cis-permethrin in water using coupled SPME and enantioselective gas chromatography (GC). Following SPME sampling, enantiomers of (Z)-cis-bifenthrin and cis-permethrin were separated at the baseline on a beta-cyclodextrin-based enantioselective column, and analyte enrichment onto the SPME fiber was not enantioselective. The GC response increased as sampling time was increased from 0 to 240 min, and as sampling temperature was increased from 20 to 40 degrees C. Organic solvents such as methanol, acetone, and acetonitrile enhanced, while soil extracts slightly decreased, the GC response. The integrated SPME-enantioselective GC method was used to analyze surface runoff samples. The analysis showed preferential degradation of the 1S-3S enantiomer over the 1R-3R enantiomer for both (Z)-cis-bifenthrin and cis-permethrin. The concentrations detected by SPME-GC were substantially smaller than those determined following solvent extraction, suggesting that SPME-enantioselective GC analysis selectively measured the dissolved fraction.  相似文献

The flavor volatiles in three Japanese rice cultivars, Nihonbare, Koshihikari, and Akitakomachi, during cooking were directly extracted by using a modified headspace solid‐phase microextraction (SPME) method and analyzed by gas chromatography‐mass spectrometry (GC‐MS). A total of 46 components were identified, including aldehydes, ketones, alcohols, and heterocyclic compounds, as well as fatty acids and esters, phenolic compounds, hydrocarbons, etc. The amount of key odorant compounds increased with cooking, while the amount of low‐boiling volatiles decreased. The similarities and differences of the three rice cultivars were determined through a comparison of their volatile components. Nihonbare was characterized by a higher amount of indole but an absence of the chemical class of fatty acid esters. In contrast, both Koshihikari and Akitakomachi had a higher amount of 4‐vinylphenol and an abundance of those esters. Koshihikari and Akitakomachi were quite similar in regard to those flavor volatiles. Furthermore, the observations in the research may suggest that the volatile components at cooking stage (I) were the representatives of the flavor volatiles of uncooked rice, while the volatile constituents at cooking stage (IV) were the representatives of the flavor volatiles of cooked rice.  相似文献

利用高效液相色谱法测定贮藏期间五九香梨的糖和有机酸的组成及变化,采用固相微萃取方式(SPME)进行萃取,并结合气相色谱和质谱(GC/MS)分析检测梨果实中的挥发性香气成分。得出以下主要结论:果糖是五九香梨的主要糖类,其次是葡萄糖和蔗糖。随着贮藏期的延长,果糖先有增加的趋势,在贮藏的第三个月内,果糖含量处于最高,而蔗糖和葡萄糖减少趋势不明显。五九香梨果实中的主要有机酸是苹果酸和柠檬酸,整个贮藏期间,柠檬酸和酒石酸含量处于上升趋势,苹果酸和奎宁酸含量在第一、二月贮藏期间增加,而后开始下降;莽草酸含量在贮藏期第一到第三阶段上升,而后略下降;琥珀酸含量在第一到第二阶段变化不明显,接着有上升的趋势,在第三阶段含量达到最高,第四和第五阶段略低于第三阶段;乙酸含量变化不明显。梨果实的硬度略有下降,贮藏的前3个月,可溶性固形物含量有上升的趋势,随后开始缓慢下降。五九香梨的主要香气成分化合物有乙酸己酯 (49.35%),乙酸丁酯 (19.56%),己酸乙酯 (5.16%),丁酸乙酯 (4.92%),乙酸乙酯 (1.08%),(E,Z)-2,4-癸二烯酸乙酯 (0.84%)等。在整个贮藏期间,部分香气成分随着贮藏期的延长而略有增加,尤其是酯类化合物。结果显示:果实硬度、可溶性固形物、糖、有机酸和香气的组成与变化对五九香梨果实的品质都有一定的影响。  相似文献

An apparatus that allows extraction and enrichment of flavor volatiles of rice during cooking using solid-phase microextraction (SPME) was designed and tested in the Japanese rice cultivar Akitakomachi. Because it is a solvent-free technique, no solvent contaminants were extracted during sampling. This technique enables one to place the fiber of SPME in the effluent of the flavor volatiles of the rice during cooking and it can also be used for simultaneous extraction of those volatiles in the course of the cooking process. Therefore, variations in the composition and amount of volatile compounds of rice during cooking can be determined. The overall flavor volatiles of rice during cooking have been analyzed by using this SPME method. Compounds that have been previously highlighted as flavor volatile markers, such as volatiles from oxidative degradation of lipids, products from thermal decomposition, and fatty acids existing in rice, were extracted directly by SPME in conjunction with this apparatus.  相似文献
