The effect of different conditions of pea germination on dietary fiber (DF) composition was studied. Insoluble dietary fiber (IDF) and soluble dietary fiber (SDF) were subjected to acid hydrolysis, and the resultant neutral sugars, uronic acids, and Klason lignin were quantified. Germinated peas exhibited significantly higher contents of total dietary fiber (TDF) than the raw sample, due to the increases of both DF fractions. Under darkness conditions, germination exhibited the highest contents of IDF and SDF. Decreasing IDF/SDF ratios showed that the carbohydrate changes did not take place to the same extent during germination, the SDF fraction being the most affected. The detailed chemical composition of fiber fractions reveals increases of cellulose in the IDF of germinated samples, whereas SDF exhibits a decrease of pectic polysaccharides and also increases of polysaccharides rich in glucose and mannose. The DF results were corroborated by a comparative examination of the cell wall carbohydrate composition.  相似文献

Onion tissues of three varieties were evaluated for dietary fiber (DF) composition. Insoluble (IDF) and soluble (SDF) dietary fibers were subjected to acid hydrolysis, and the resultant neutral sugars, uronic acids, and Klason lignin were quantified. Brown skin exhibited the highest total dietary fiber (TDF) content (65.8%) on a dry matter basis, followed by top (48.5%) and bottom (38.6%), IDF being the main fraction found. The SDF:IDF ratio decreased from inner to outer tissues. Brown skin and outer leaves byproducts appear to be the most suitable sources of DF that might be used in food product supplementation. The chemical composition reveals that cellulose and pectic polysaccharides were the main components of onion DF in all tissues, although differences between them were noticed. An increase in the uronic acids/neutral sugars ratio from inner to outer tissues was found, suggesting that the galactan side chain shows a DF solubilization role.  相似文献

A collaborative study was conducted to determine the insoluble dietary fiber (IDF), soluble dietary fiber (SDF), and total dietary fiber (TDF) content of food and food products by using a combination of enzymatic and gravimetric procedures. The method was basically the same as that developed for TDF only, which was adopted official final action by AOAC, except for changing the concentration of buffer and base and substituting hydrochloric acid for phosphoric acid. These changes were made to improve the robustness of the method. Duplicate blind samples of soy isolate, white wheat flour, rye bread, potatoes, rice, corn bran, oats, Fabulous Fiber, wheat bran, and a high fiber cereal were analyzed by 13 collaborators. Dietary fiber values (IDF, SDF, and TDF) were calculated as the weight of residue minus the weight of protein and ash. The coefficients of variation (CVs) of both the independent TDF determination and the sum of IDF and SDF were better than 15 and 18%, respectively, with the exception of rice and soy isolate. These 2 foods, however, contained only about 1% TDF. The CVs of the IDF were equally good, except for Fabulous Fiber, for which filtration problems occurred. The CVs for the SDF were somewhat high, but these products had very low SDF content. There was excellent agreement between the TDF determined independently and the TDF determined by summing the IDF and SDF. The method for separate determination of IDF and SDF requires further study. The modifications (changes in concentration of buffer and base and the use of hydrochloric acid instead of phosphoric acid) to the official final action method for TDF have been adopted.  相似文献

The effect of fermentation on antinutritional factors and also on total dietary fiber (TDF), insoluble (IDF) and soluble (SDF) dietary fiber fractions was studied in beans (Phaseolus vulgaris L.). The processes studied were two types of fermentation (lactic acid and natural), and a portion of the obtained flours were processed by autoclaving. The dietary fiber (DF) content and its components were determined using the enzymatic-gravimetric and enzymatic-chemical methods. The TDF content ranged from 24.5% dry matter (DM) in the raw to 25.2% DM in the processed beans. All the processing treatments significantly decreased the SDF content, and irrelevant changes were noticed in the IDF content of processed beans. Cellulose content of all samples was reduced by the processing treatments. Correspondingly, higher amounts of resistant starch was observed in the processed beans, except in the lactic acid fermented ones. However, the levels of pectic polysaccharides and Klason lignin were higher in the samples fermented by Lactobacillus plantarum. The action of microorganisms was determinant for the different degradation of the bean cell wall, disrupting the protein-carbohydrate integration, thus reducing the solubility of DF.  相似文献

Dietary fiber (DF) is one of the main dietary factors contributing to consumers' well-being. In this work the possibility of using the roasted coffee silverskin (CS), a byproduct of roasted coffee beans, as a DF-rich ingredient has been evaluated. The results of our investigation showed that this material has 60% total DF, with a relevant component (14%) of soluble DF. Although a small amount of free phenol compounds is present in CS, it has a marked antioxidative activity, which can be attributed to the huge amount of Maillard reaction products, the melanoidins. Static batch culture fermentation experiments showed that CS induces preferential growth of bifidobacteria rather than clostridia and Bacteroides spp. CS can be proposed as a new potential functional ingredient in consideration of the high content of soluble DF, the marked antioxidant activity, and the potential prebiotic activity.  相似文献

The enzymatic hydrolysis of brewers' spent grain (BSG) has been investigated through treatment with commercial carbohydrases and proteases. Resultant residues were then chemically fractionated and delignified. Enzymatic treatments released 25-30% of the BSG mass and yielded precursors suitable for subsequent conversion to potentially value-added products. Controlled chemical fractionation selectively solubilized arabinoxylan but with no differences apparent due to prior enzyme treatment. The loss of non-polysaccharide components during alkali treatment suggests the presence of a high proportion of alkali-soluble lignin. Further delignification of the alkali-insoluble residues and further chemical fractionation released the remaining hemicellulose, to yield a residue which was >90% cellulose. Further knowledge of the properties and interaction between BSG polymers will facilitate an improved enzyme-assisted total deconstruction of BSG and hence the exploitation of its biomass.  相似文献

以大豆渣为原料,采用微生物发酵、微射流均质机高压均质处理和高温蒸煮的方法来提高大豆膳食纤维中可溶性成分含量,研究不同发酵时间、不同处理压力和蒸煮温度以及蒸煮时间对提高豆渣可溶性膳食纤维(SDF)含量的影响。结果表明:利用发酵法可提高可溶性膳食纤维的含量达15%之多,并且在发酵的前6天,SDF含量增长很快,6 d后SDF的含量变化较少;而高压均质处理法提高可溶性膳食纤维含量的幅度在10%~28%之间,并随着处理压力的升高而增大;高温蒸煮法能提高可溶性膳食纤维的含量20%,121℃,20 min为较理想的蒸煮条件。  相似文献

A new soluble cocoa fiber product (SCFP), obtained after enzymatic treatment of cocoa husks, was characterized and its potential health effects studied in an animal model of dietary-induced hypercholesterolemia. The SCFP was rich in soluble dietary fiber (DF) and antioxidant polyphenols. Consumption of a cholesterol-rich diet containing the SCFP as a source of DF resulted in lower food intake and body weight gain in comparison with control groups consuming cholesterol-free or cholesterol-rich diets with cellulose as DF. The cholesterol-rich diet caused remarkable hypercholesterolemia. However, the SCFP diminished the negative impact of the cholesterol-rich diet, buffering the decrease of high density lipoprotein-cholesterol, and the increase of total and low density lipoprotein-cholesterol levels, and lipid peroxidation (malondialdehyde levels) induced by the fatty diet. The SCFP also decreased triglyceride levels to values lower than those in the group fed the cholesterol-free diet. These results put forward the potential application of the SCFP as a dietary supplement or functional food ingredient.  相似文献

Contradictory results have been reported in studies associating dietary fiber (DF) intake and the incidence of colorectal cancer. Most studies focused mainly on the amount of total DF, but DF is a heterogeneous complex and its components are generally ignored. The aim of this work was to compare the amount and composition of DF intake in Murcia (Spain) and Copenhagen (Denmark). Total dietary fiber intake was a 31% higher in Murcia than in Copenhagen, the ratio of insoluble DF/soluble DF being similar in the two diets. DF in Murcia contains major amounts of nonstarch polysaccharides and polyphenolic compounds, mainly derived from the high consumption of fruits and vegetables (6.7 servings). Resistant starch intake was of the same order in the two regions while resistant protein intake was higher in Murcia. The quantitative and qualitative differences in DF intake may contribute to understand the differences in colorectal cancer incidence in Mediterranean and Scandinavian areas.  相似文献

The proximate composition of sclerotia of Pleurotus tuber-regium, Polyporus rhinoceros, and Wolfiporia cocos, together with the yield, purity, monosaccharide profile, and microstructure of their insoluble dietary fiber (IDF) and soluble dietary fiber (SDF) fractions prepared from AOAC enzymatic-gravimetric methods were investigated and compared. All three sclerotia were typical carbohydrate rich sclerotia [ranging from 90.5 to 98.1% dry matter (DM)] with an exceptionally low amount of crude lipid content (ranging from 0.02 to 0.14% DM). Besides, all three sclerotia possessed substantial amounts of IDF (ranging from 77.4 to 94.6% DM) with notably high levels of nonstarch polysaccharides (NSP) (89.9-92.2% DM) in which glucose was the predominant sugar residue (90.6-97.2% of NSP DM). On the contrary, both the yield (only ranging from 1.45 to 2.50% DM) and the amount of NSP (ranging from 22.4 to 29.6% DM) of the three sclerotial SDF fractions were very low. Scanning electron micrographs showed fragments of interwoven hyphae and insoluble materials in the three sclerotial IDF fractions, but only an amorphous structure of soluble materials was observed in the SDF fractions. The potential use of these fiber preparations was discussed.  相似文献

Metabolite production and antioxidant released during colonic fermentation of naturally occurring dietary fiber (DF) from two European diets (Mediterranean and Scandinavian) were determined. With this aim, DF and associated components were isolated from both whole diets, as well as from cereals and fruits and vegetables comprising the diets. DF was used as substrate for colonic fermentation in a dynamic in vitro model of the colon, samples were collected, and fermentation metabolites were analyzed. Statistical differences between samples were observed in the concentrations of short-chain fatty acids and ammonia and in the ratio acetate/propionate/butyrate. Whole grain cereal DF generated a larger amount of propionate than refined flour cereal DF. Fruit and vegetable DF generated higher amounts of butyrate than cereal DF. Most antioxidant compounds were released from DF during in vitro colonic fermentation. It is concluded that different sources of DF may play a specific role in health maintenance mediated by metabolites produced during colonic fermentation.  相似文献

The effects of soaking, cooking, and industrial dehydration treatments on soluble carbohydrates, including raffinose family oligosaccharides (RFOs), and also on total dietary fiber (TDF), insoluble dietary fiber (IDF), and soluble (SDF) dietary fiber fractions were studied in legumes (lentil and chickpea). Ciceritol and stachyose were the main alpha-galactosides for chickpea and lentil, respectively. The processing involved a drastic reduction of soluble carbohydrates of these legumes, 85% in the case of lentil and 57% in the case of chickpea. The processed legume flours presented low residual levels of alpha-galactosides, which are advisable for people with digestive problems. Processing of legumes involved changes in dietary fiber fractions. A general increase of IDF (27-36%) due to the increase of glucose and Klason lignin was observed. However, a different behavior of SDF was exhibited during thermal dehydration, this fraction increasing in the case of chickpea (32%) and decreasing in the case of lentil (27%). This is probably caused by the different structures and compositions of the cell wall networks of the legumes.  相似文献

Dietary fiber (DF) intakes in Western countries only accounts for about one-third of the substrates required for colonic bacterial cell turnover. There is a general trend among nutritionists to extend the DF concept to include all food constituents reaching the colon. In this line, a method to quantify the major nondigestible components in plant foods, namely, the indigestible fraction (IF), is presented. Analytical conditions for IF determination are close to physiological. Samples, analyzed as eaten, were successively incubated with pepsin and alpha-amylase; after centrifugation and dialysis, insoluble and soluble IFs were obtained. IF values include DF, resistant starch, resistant protein, and other associated compounds. IF contents determined in common foods (cereals, legumes, vegetables, and fruits) were higher than DF contents. Calculated IF intakes were close to the estimated amount of substrates reaching the colon. IF data could be more useful than DF data from a nutritional point of view; therefore, IF is proposed as an alternative to DF for food labeling and food composition tables.  相似文献

肥料减量深施对土壤N2O排放和冬油菜产量的影响   总被引:4,自引:3,他引:1  
为揭示肥料深施条件下减量施肥对土壤N2O排放及作物产量的影响,提出有效的减氮减排及增产增效措施,该研究以冬油菜为对象,设置肥料深施条件下当地推荐缓释肥量(750 kg/hm2,N-P2O5-K2O:25-7-8)的100%(DF100)、80%(DF80)和60%(DF60)3个施肥水平,以地表撒施当地推荐缓释肥量(BF100)和不施肥(F0)为对照,共5个处理;采用静态箱-气相色谱法对N2O排放进行原位监测,分析不同施肥处理对土壤N2O排放、土壤充水孔隙率(Water-Filled Pore Space, WFPS)、土壤温度及冬油菜产量的影响。结果表明:较地表撒施相比,肥料深施土壤N2O排放量增加了13.3%,但不显著(P>0.05);冬油菜产量显著增加了20.1%,肥料偏生产力(Partial Factor Productivity,PFP)和农学利用率(Agronomic Efficiency,AE)分别显著提高了20.1%和31.9%(P<0.05)。减量施肥显著减小了土壤N2O排放、冬油菜产量及肥料利用率(P<0.05),DF100处理较DF80、DF60和F0处理土壤N2O排放量分别增加了22.7%、42.5%和153.7%;DF100处理冬油菜产量分别是DF80、DF60和F0处理的1.30、2.24和3.24倍;DF100处理较DF80和DF60处理PFP分别增加了3.8%和34.5%且AE分别增加了19.7%和201.3%。综合考虑产量和温室效应,在深施当地推荐缓释肥施用量的基础上能够适当减量施肥,但需高于600 kg/hm2。该研究为冬油菜区N2O减排及油菜机械化直播种植合理施肥提供参考。  相似文献

为明确玉米籽粒营养成分的分布差异及不同部位富集特征,应用快速缓苏、微量着水半湿法分层破胚剥皮技术,结合靶向代谢组学方法,对郑单958玉米不同部位的营养成分及基础代谢物质进行分析与比较。结果表明玉米籽粒不同部位的淀粉、脂肪、矿物元素和膳食纤维等营养物质含量存在显著差异(P<0.05)。该研究中的玉米内皮层可能主要由种皮、糊粉层及部分外胚乳构成,该部位营养成分的种类及含量均较为丰富,其中水溶性膳食纤维含量显著高于其他部位(P<0.05),可作为玉米水溶性膳食纤维的提取分离来源。K、P和Mg元素是玉米中含量最高的矿物元素,主要存在于胚芽中,Fe、Zn、Mn和Cu元素在胚芽和玉米皮层中均有较多分布,精制加工会导致这些矿物元素的损失。玉米胚芽中水解氨基酸种类较其他部位丰富且含量较高(P<0.05),甜味氨基酸占总游离氨基酸含量的24.49%,高于玉米皮层部位、显著高于胚乳部位。研究结果为玉米营养健康食品的创制、玉米精深加工及相关专用装备的研发提供参考。  相似文献

Brewer's spent grain (BSG) is an abundant, protein-rich coproduct from the beer industry. There is a growing interest in increasing and diversifying the exploitation of BSG and related coproducts for economic and environmental reasons. In this paper, we report on a study of the solubilization of proteinaceous material from BSG using several commercial peptidase preparations. Our data show that Alcalase is the most effective peptidase for solubilization of BSG proteins, with an ability to release up to 77% of total protein. The peptides produced by Alcalase had lower average molecular weight than peptides produced by the less effective enzymes. Processes that combined peptidase treatment with carbohydrate-degrading enzyme preparations such as Depol740 increased the solubilization of dry matter (from 30 to 43% under optimal conditions). However, such additional treatment had little effect on the solubilization of protein. The choice of enzyme dosage depends on the desired hydrolysis time and was assessed through several experiments. Protein solubilization was consistently better at pH 8.0 as compared to pH 6.8. Maximum protein solubilization at pH 8.0 within 4 h required the use of 10-20 microL Alcalase per g of dry matter. However, a considerable degree of solubilization (64%) and hydrolysates with high protein content could be obtained using doses down to only 1.2 microL. Amino acid composition analyses showed that Alcalase treatment solubilizes proline and glutamine (constituents of barley hordein) slightly more efficiently than the other amino acids in BSG.  相似文献

限水条件下,等量氮不同施用方式对小麦旗叶光合和衰老特性的调控效应不同;不同抗旱性品种对施氮方式的响应也有所差异。与拔节初期1次施氮处理(SF)相比,拔节初期、挑旗期2次施氮处理(DF)增加了供试品种旗叶中后期的叶绿素含量(Chl)、可溶蛋白含量(SP)、RuBPCase活性、气孔导度(Gs)和光合速率(Pn),延长了叶绿素含量缓降期(RSP)、光合速率高值持续期(PAD),提高了叶源量(LSC);使旗叶生长中后期的超氧化物歧化酶(SOD)活性和过氧化物酶(POD)活性增加,细胞的活性氧(O2-)含量和丙二醛(MDA)含量降低。与抗旱品种石麦12相比,DF处理对抗旱性中等的品种石新733的旗叶光合生理参数、光合功能高值持续期具有更大调控效应。DF处理使供试品种的千粒重、产量和水分利用效率增加,其中石麦12后两指标增幅达显著水平。表明在春季适宜施氮量范围内,在肥力中等偏下地力条件下,生育中后期(挑旗期)追施适量氮素,具有提高叶片的细胞保护酶活性、维持细胞的较强活性氧清除能力,改善限水条件下小麦旗叶光合碳同化能力、延缓旗叶衰老、促进子粒灌浆和提高水分利用效率的作用。  相似文献

This research aims at developing a biorefinery platform to convert lignocellulosic corn fiber into fermentable sugars at a moderate temperature (37 °C) with minimal use of chemicals. White-rot (Phanerochaete chrysosporium), brown-rot (Gloeophyllum trabeum), and soft-rot (Trichoderma reesei) fungi were used for in situ enzyme production to hydrolyze cellulosic and hemicellulosic components of corn fiber into fermentable sugars. Solid-substrate fermentation of corn fiber by either white- or brown-rot fungi followed by simultaneous saccharification and fermentation (SSF) with coculture of Saccharomyces cerevisiae has shown a possibility of enhancing wood rot saccharification of corn fiber for ethanol fermentation. The laboratory-scale fungal saccharification and fermentation process incorporated in situ cellulolytic enzyme induction, which enhanced overall enzymatic hydrolysis of hemi/cellulose components of corn fiber into simple sugars (mono-, di-, and trisaccharides). The yeast fermentation of the hydrolyzate yielded 7.8, 8.6, and 4.9 g ethanol per 100 g corn fiber when saccharified with the white-, brown-, and soft-rot fungi, respectively. The highest ethanol yield (8.6 g ethanol per 100 g initial corn fiber) is equivalent to 35% of the theoretical ethanol yield from starch and cellulose in corn fiber. This research has significant commercial potential to increase net ethanol production per bushel of corn through the utilization of corn fiber. There is also a great research opportunity to evaluate the remaining biomass residue (enriched with fungal protein) as animal feed.  相似文献

This study explored the potential of soluble dietary fiber (SDF) from agrowastes, okara (soybean solid waste), oil palm trunk (OPT), and oil palm frond (OPF) obtained via alkali treatment, in the nanoencapsulation of Lactobacillus acidophilus . SDF solutions were amended with 8% poly(vinyl alcohol) to produce nanofibers using electrospinning technology. The spinning solution made from okara had a higher pH value at 5.39 ± 0.01 and a higher viscosity at 578.00 ± 11.02 mPa·s (P < 0.05), which resulted in finer fibers. FTIR spectra of nanofibers showed the presence of hemicellulose material in the SDF. Thermal behavior of nanofibers suggested possible thermal protection of probiotics in heat-processed foods. L. acidophilus was incorporated into the spinning solution to produce nanofiber-encapsulated probiotic, measuring 229-703 nm, visible under fluorescence microscopy. Viability studies showed good bacterial survivability of 78.6-90% under electrospinning conditions and retained viability at refrigeration temperature during the 21 day storage study.  相似文献

Coffee beans are rich in nondigestible polysaccharides (dietary fiber), which may partially pass into brewed coffee; however, to the authors' knowledge, there is not enough literature on dietary fiber in brewed coffee. A specific method to determine dietary fiber in beverages (enzymatic treatment plus dialysis) was applied to the coffees brewed by the most common methods (espresso, filter, soluble); results showed that brewed coffee contained a significantly higher amount of soluble dietary fiber (0.47-0.75 g/100 mL of coffee) than other common beverages. Coffee dietary fiber contains a large amount of associated antioxidant phenolics (8.7-10.5 mg/100 mL of brewed coffee).  相似文献
