AIM: To investigate the effect of miR-496 over-expression on the growth and metastasis of colon cancer cells and its molecular mechanism.METHODS: The proteins interacting with miR-496 were screened by bioinformatic method. The levels of miR-496, CTNNB1 mRNA and β-catenin protein in colon cancer cell lines, HT29, HCT116 and SW480, and normal colonic epithelial cell line NCM460 were detected by real-time PCR and Western blot. HT29, HCT116 and SW480 cells were transfected with miR-496 mimics using Lipofectamine 2000 and named as HT29-miR-496 mimics, HCT116-miR-496 mimics and SW480-miR-496 mimics cells, respectively, and the cells transfected with the scramble served as negative control. The cell viability, lactate dehydrogenase (LDH) leakage, and colony formation and metastatic abilities were determined by MTT assay, LDH assay, colony formation assay and Transwell method, respectively. The promoter activity of miR-496 was measured using luciferase reporter gene assay. The protein levels of β-catenin, eukaryotic translation initiation factor 4E-binding protein 1 (4E-BP1), p-4E-BP1, low-density lipoprotein receptor-related protein 6(LRP6), p-LRP6, MMP-7, MMP-9, MMP-13 and TIMP-2 were monitored by Western blot.RESULTS: Endogenous miR-406 interacted with β-catenin was found in the colon cancer cells. Low miR-496 expression in the HT29, HCT116 and SW480 cells and high miR-496 expression in NCM460 cells were detected. In contrast, high β-catenin expression was found in the HT29, HCT116 and SW480 cells and low β-catenin expression was observed in the NCM460 cells. Compared with control group, the cell viability, colony formation rate and the number of metastatic cells remarkably decreased in the HT29-miR-496 mimics, HCT116-miR-496 mimics and SW480-miR-496 mimic cells (P<0.05). The promoter activity of miR-496 was significantly increased in colon cancer cells transfected with miR-496 mimics, and was 1.75, 2.04 and 1.61 times as high as control group. miR-496 over-expression inhibited β-catenin levels, and p-4E-BP1 and p-LRP6 protein levels were also reduced. siRNA- or over-expressed miR-496-mediated β-catenin down-regulation inhibited MMP-7 and MMP-9 expression, but promoted TIMP-2 expression.CONCLUSION: The expression level of miR-496 in the colon cancer cells is low, but in the normal colonic epithelial cells is high. miR-496 over-expression inhibits the protein levels of MMP-7 and MMP-9, and promotes the protein expression of TIMP-2 via inhibiting Wnt/β-catenin pathway, thus suppressing malignant phenotype in the colon cancer cells.  相似文献

AIM: To investigate the function of microRNA-125a-5p (miR-125a-5p) on epithelial-mesenchymal transition (EMT) of breast cancer cells via GSK-3β/Snail signaling pathway.METHODS: The expression of miR-125a-5p in normal breast epithelial cells and breast cancer cells, as well as the transfection efficiency of miR-125a-5p plasmid in MDA-MB-231 cells was detected by RT-qPCR. The chemotaxis ability and invasion ability were detected by chemotaxis assay and Transwell invasion assay. The changes of EMT-related markers, the protein level of phosphorylated glycogen synthase kinase-3β (p-GSK-3β) and the nuclear translocation of Snail were determined by Western blot. RESULTS: The expression of miR-125a-5p in the breast cancer cells was significantly lower than that in the normal breast epithelial cells. The expression of miR-125a-5p was significantly higher in MDA-MB-231/miR-125a-5p cells than that in MDA-MB-231/NC cells. The ability of epithelial growth factor (EGF) at 10 μg/L to induce chemotaxis of MDA-MB-231 cells was the strongest. Compared with MDA-MB-231/NC group, stimulation of EGF decreased the invasion ability of MDA-MB-231/miR-125a-5p cells, and resulted in the increase in E-cadherin expression, while significantly decreased the protein levels of vimentin and p-GSK-3β. Meanwhile, the nuclear localization of Snail was significantly inhibited. The invasion capacity of MDA-MB-231/miR-125a-5p+GAB2 cells was significantly enhanced compared with MDA-MB-231/miR-125a-5p+Con cells, the expression of E-cadherin was decreased, and the protein levels of vimentin and p-GSK-3β were significantly increased, while the nuclear localization of Snail was promoted. CONCLUSION: miR-125a-5p suppresses EMT via GSK-3β/Snail signaling pathway, thus inhibiting the invasion ability of breast cancer cells.  相似文献

AIM:To study the effects of microRNA-105(miR-105) on the cell proliferation, migration and invasion abilities of non-small-cell lung cancer (NSCLC) H460 cells, and further to explore its mechanism. METHODS:The expression of miR-105 and kinesin family member C1 (KIFC1) mRNA in the NSCLC tissues and adjacent tissues and cells was detected by RT-qPCR. The protein expression of KIFC1 in the NSCLC tissues, adjacent normal tissues and cells was determined by Western blot. The H460 cells were divided into miR-105 group (transfection with miR-105 mimics), miR-negative control (NC) group (transfection with miR-NC), inhibitor-NC group (transfection with NC of inhibitor), inhibitor-miR-105 group (transfection with miR-105 inhibitor), si-NC group (transfection with NC siRNA), si-KIFC1 group (transfection with KIFC1 siRNA), miR-105+vector group (miR-105 mimics and pcDNA 3.1 co-transfection) and miR-105+KIFC1 group (miR-105 mimics and pcDNA 3.1-KIFC1 co-transfection). The cell proliferation was measured by MTT assay and colony formation assay. The migration and invasion abilities were detected by Transwell methods. The relative luciferase acitivity was evaluated by double luciferase reporter assay. RESULTS:Compared with the adjacent tissues, the expression of miR-105 was significantly decreased and the expression of KIFC1 was significantly increased in NSCLC tissues (P<0.05). Compared with human normal embryonic lung fibroblasts MRC-5, the expression of miR-105 in the H460 cells was significantly decreased, and the expression of KIFC1 was significantly increased (P<0.05). miR-105 inhibited the relative luciferase activity of H460 cells with wild-type KIFC1 and negatively regulated the protein expression of KIFC1. Over-expression of miR-105 and knockdown of KIFC1 expression significantly inhibited the proliferation, migration and invasion abilities of H460 cells. Over-expression of KIFC1 reversed the inhibitory effect of miR-105 on the cell proliferation, migration and invasion abilities of H460 cells. CONCLUSION:miR-105 inhibits the proliferation, migration and invasion abilities of NSCLC cells. The mechanism may be related to targeting and negatively regulating expression of KIFC1.  相似文献

AIM: To study the effect of miR-18a on angiogenesis of human aortic artery endothelial cells. METHODS: After 10 nmol/L miR-18a mimics or 20 nmol/L inhibitor was transfected into human aortic endothelial cells, the expression level of miR-18a was determined by qRT-PCR, and the capacities of endothelial cell angiogenesis, such as migration, adhesion and tube formation, were observed. RESULTS: Forty-eight hours after transfection with miR-18a mimics, the expression of miR-18a was as 608 folds as the control (P<0.05), but decreased to 31% of the control level in miR-18a inhibitor transfection group (P<0.05). Compared with control group, the numbers of endothelial cells, which migrated to the lower Transwell chamber and formed capillary-like tubes, declined by 60% and 52%, respectively, in miR-18a mimics group (P<0.01), and they increased by 100% and 84%, respectively, in miR-18a inhibitor transfection group (P<0.05, P<0.01). In addition, the inhibitor treatment group displayed more potent adhesion capacity, 43% higher than that in control group (P<0.05). CONCLUSION: miR-18a is involved in angiogenesis of human arterial endothelial cells, and might be a potential molecular therapeutic target of vascular diseases.  相似文献

AIM: To investigate the effect of macrophage peroxisome proliferator-activated receptor α (PPARα) activation on macrophage inflammation-induced activation and migration of cardiac fibroblasts. METHODS: Mouse bone marrow-derived macrophages were treated with vehicle, PPARα agonist WY14643 (10 μmol/L), angiotensin Ⅱ (Ang II; 1 μmol/L) or Ang II+WY14643 for 24 h, and the supernatants were collected as conditioned medium (CM) to stimulate cardiac fibroblasts for additional 24 h. The mRNA levels of PPARα, interleukin-6 (IL-6), IL-1β and tumor necrosis factor-α (TNF-α) in the macrophages as well as fibrotic markers collagen type Ⅰ alpha 2 chain (Col1a2), collagen alpha 1 chain (Col3a1) and actin alpha 2 (Acta2) in the cardiac fibroblasts were detected by RT-qPCR. The protein levels of IL-6 and IL-1β in the macrophages as well as collagen I, collagen III and α-smooth muscle actin (α-SMA; encoded by Acta2 gene) in the cardiac fibroblasts were determined by Western blot. Wound-healing assay was applied to eva-luate the migration ability of cardiac fibroblasts. RESULTS: Ang II significantly increased the mRNA levels of pro-inflammatory factors, such as IL-6, IL-1α and TNF-α, but decreased the mRNA level of PPARα in the macrophages. Administration of PPARα agonist WY14643 dramatically decreased Ang II-induced mRNA levels of IL-6, IL-1β and TNF-α in the macrophages, and significantly decreased Ang II-induced protein expression of IL-6 and pro-IL-1β in the macrophages. The CM from Ang II-treated macrophages significantly up-regulated the mRNA levels of Col1a2, Col3a1 and Acta2 in the cardiac fibroblasts, which were inhibited by the CM from WY14643-treated macrophages. The same results were observed in the protein levels of collagen I, collagen III and α-SMA in the cardiac fibroblasts. Moreover, the CM from Ang II-treated macrophages significantly promoted cardiac fibroblast migration, whereas the CM from WY14643-treated macrophages markedly inhibited macrophage inflammation-induced cardiac fibroblast migration. CONCLUSION: WY14643-activated PPARα inhibits activation and migration of cardiac fibroblasts by attenuating Ang II-induced macrophage inflammatory response.  相似文献

AIM:To explore the effects of miR-126 on the expression of vascular endothelial growth factor (VEGF) in vascular endothelial cell line EA.hy926. METHODS:EA.hy926 cells were cultured in vitro and transfected with miR-126 mimics or miR-126 inhibitor by cation-mediated transfection method. The total RNA was extracted from the culture cells 36 h after transfection of miR-126 mimics or miR-126 inhibitor. The expression of VEGF at mRNA and protein levels was detected by real-time PCR and Western blotting. RESULTS:Thirty-six hours after transfection of miR-126 inhibitor at concentration of 50 nmol/L, the expression of VEGF at mRNA and protein levels increased significantly as compared with the negative control (P<001). However, transfection of miR-126 mimics at concentration of 50 nmol/L for 36 h significantly decreased the expression of VEGF at mRNA and protein levels as compared with the negative control. CONCLUSION:miR-126 inhibits the expression of VEGF. VEGF may be one of the target genes regulated by miR-126.  相似文献

AIM: To explore the effect of microRNA-221 (miR-221) on the proliferation of lung cancer A549 cells, and to investigate its mechanism. METHODS: The A549 cells were transfected with miR-221 mimics by Lipofectamine 2000. The expression of miR-221 was detected by RT-qPCR. The expression of PTEN at mRNA and protein le-vels was detected by RT-qPCR and Western blot, respectively. The cell proliferation was examined by CCK-8 assay and colony formation assay. The 3'-UTR of PTEN was cloned into luciferase reporter vector and its enzymatic activity was detected to verify whether miR-221 targeted to PTEN. RESULTS: The expression level of miR-221 in the A549 cells was significantly increased after transfection with miR-221 mimics as compared with negative control group and blank group (P<0.01). The mRNA and protein levels of PTEN were significantly down-regulated compared with control group and blank group (P<0.05). In addition, miR-221 over-expression significantly promoted the proliferation of A549 cells (P<0.05). Moreover, miR-221 inhibited the enzymatic activity of luciferase reporter vector of PTEN. CONCLUSION: Over-expression of miR-221 significantly promotes the proliferation ability of human lung cancer A549 cells by down-regulation of PTEN.  相似文献

AIM:To investigate the expression level of microRNA-130b (miR-130b) and the molecular me-chanisms of miR-130b in temozolomide (TMZ)-resistant glioma. METHODS:The relative levels of miR-130b in 3 glioma cell lines (U251, SHG-44 and U87) were assessed by RT-qPCR. The half maximal inhibitory concentration (IC50) of TMZ for the glioma cell lines was analyzed. To establish the TMZ-resistant glioma cell line, U251 cells were exposed to gradually increasing concentrations of TMZ. The IC50 and resistance index (RF) were calculated with GraphPad Prism software. miR-130b-overexpressing U251/TR cells and miR-130b-knockdown U251 cells were established by transient transfection with miR-130b mimics and miR-130b inhibitor, respectively. The viability of the glioma cells was measured by CCK-8 assay. The apoptosis of glioma cells was analyzed by Annexin V/PI apoptosis assay. Bioinformatics software was used to predict the potential target gene of miR-130b, and such prediction was validated by luciferase reporter assay. Electrophoretic mobility shift assay was performed to detect the DNA binding ability of NF-κB. Western blot was used to determine the protein levels of tumor necrosis factor-α (TNF-α), Bcl-2, X-linked inhibitor of apoptosis protein (XIAP) and survivin in the glioma cells. RESULTS:The IC50 values of TMZ for the giloma cell lines U251, SHG-44 and U87 were 54.8, 94.8 and 149.6 μmol/L, respectively. U251/TR cells were approximately 8.1 times resistant to TMZ as compared with its parental cells. Up-regulation of miR-130b significantly reduced the resistance of U251/TR cells to TMZ. On the contrary, down-regulation of miR-130b dramatically increased the tolerance of U251 cells to TMZ. The overexpression of miR-130b promoted apoptosis induced by TMZ in the U251/TR cells. However, the knockdown of miR-130b expression decreased the percentage of apoptotic cells in the U251 cells induced by TMZ (P<0.05). Luciferase reporter assay confirmed that TNF-α was a direct target gene of miR-130b. Knockdown of miR-130b in the U251 cells significantly promoted, while overexpression of miR-130b in the U251/TR cells reduced the DNA binding ability of NF-κB as well as the levels of TNF-α, Bcl-2, XIAP and survivin. Furthermore, NF-κB inhibitor Bay 11-7082 enhanced TMZ-induced apoptosis in the U251/TR cells. CONCLUSION:The expression of miR-130b is significantly decreased in TMZ-resistant glioma cells. miR-130b inhibits resistance of glioma to TMZ by targeting TNF-α/NF-κB pathway.  相似文献

AIM: To observe the effect of microRNA-16 (miR-16) on the megakaryocytic differentiation of K562 cells, and to explore the potential mechanism.METHODS: miR-16 was over-expressed or silenced by transfection with miR-16 mimics or inhibitor in K562 cells. The level of miR-16 was detected by real-time PCR. The expression of CD41, CD42b and CD61, as megakaryocytic differentiation markers, was detected by flow cytometry. The effect of miR-16 on the expression of myeloblastosis oncogene (MYB) was measured by Western blotting, and flow cytometry was performed to confirm whether the effect of miR-16 on expression of CD41, CD42b and CD61 was mediated by MYB.RESULTS: Transfection with miR-16 mimics dramatically elevated the level of miR-16 and the expression of CD41, CD42b and CD61 in the K562 cells. Transfection with miR-16 inhibitor decreased the level of miR-16 and the expression of CD41, CD42b and CD61 in the K562 cells (P<0.05). The expression of MYB was regulated by miR-16, and MYB silencing reversed the regulation of CD41, CD42b and CD61 induced by miR-16.CONCLUSION: miR-16 regulates the megakaryocytic differentiation of K562 cells by targeting MYB.  相似文献

AIM: To explore the effect of microRNA-146a (miR-146a) on apoptosis of human gastric cancer SGC-7901 cells and the underluing mechanism. METHODS: miR-146a mimic (up-regulated miR-146a expression) and miR-146a inhibitor (down-regulated miR-146a expression) were transfected into the SGC-7901 cells by liposome method. At the same time, miRNA nonsense sequence transfection group as the negative control group (NC group) was set up. RT-qPCR was used to evaluate the levels of miR-146a in the SGC-7901 cells after transfection. The effects of miR-146a on the cell apoptosis and growth were assessed by flow cytometry analysis and CCK-8 assay, respectively. The effect of over-expression or knockdown of miR-146a on transforming growth factor-β-activated kinase 1 (TAK1)/nuclear factor-kappa B (NF-κB) signaling was evaluated by RT-qPCR and Western blot. RESULTS: miR-146a modulated apoptosis of SGC-7901 cells. Over-expression of miR-146a significantly increased apoptosis, whereas knockdown of miR-146a inhibited the apoptosis of SGC-7901 cells. The expression of TAK1 at mRNA and protein levels was significantly decreased when miR-146a mimic was transfected into the SGC-7901 cells (P<0.05). On the contrast, the expression of TAK1 at mRNA and protein were significantly higher in miR-146a inhibitor transfection group than that in NC group (P<0.05), suggesting that miR-146a negatively regulated TAK1 expression. Moreover, knockdown of TAK1 enhanced the apoptosis of SGC-7901 cells (P<0.01), while over-expression of TAK1 inhibited the apoptosis of SGC-7901 cells(P<0.01). Additionally, both over-expression of miR-146a and knockdown of TAK1 led to a prominent increase in the expression of NF-κB inhibitor protein alpha (IκBα) and a significat decrease in B cell lymphoma-2 (Bcl-2) level in the SGC-7901 cells. CONCLUSION: miR-146a significantly promotes apoptosis of SGC-7901 cells by inhibition of NF-κB pathway via targeting TAK1.  相似文献

AIM: To investigate the effect of microRNA-708-5p(miR-708-5p) on the migration of human mesenchymal stem cells(hMSCs). METHODS: The expression of miR-708-5p was determined by miRNA arrays and real-time PCR. By transfection of miR-708-5p mimic or inhibitor, the up-regulation or down-regulation of miR-708-5p expression in hMSCs was evaluated. The cell scratch and Transwell tests were used to detect the migration capability of hMSCs. The effects of transmembrane protein 88(TMEM88), a miR-708-5p target gene, on β-catenin expression and migration of hMSCs were detected. RESULTS: The expression of miR-708-5p was down-regulated in the old hMSCs compared with the young hMSCs. Up-regulation of miR-708-5p resulted in increasing migration of hMSCs. Conversely, down-regulation of miR-708-5p resulted in decreasing cell migration. The expression of TMEM88 was up-regulated in the old hMSCs compared with the young hMSCs, while the expression of β-catenin was down-regulated. Directly repression of TMEM88 expression increased the β-catenin expression and migration of hMSCs. The regulation of miR-708-5p on hMSCs was attenuated by inhibiting the expression of miR-708-5p and TMEM88 together. CONCLUSION: miR-708-5p increases β-catenin expression and Wnt/β-catenin activity by repressing TMEM88, thus enhancing the migration of hMSCs.  相似文献

AIM: To investigate the effect of microRNA-375 (miR-375) on the viability, cell cycle and apoptosis of HCT116 cells.METHODS: The expression of miR-375 in different colorectal cancer cell lines was detected by real-time PCR. The miR-375 mimics was transfected into HCT116 cells by LipofectamineTM 2000. The mRNA expression of miR-375 and AEG-1 was detected by real-time PCR. The HCT116 cell viability was detected by MTT assay. The changes of apoptosis and cell cycle distribution were analyzed by flow cytometry.RESULTS: Real-time PCR showed that miR-375 expression was the lowest in HCT116 among 4 colorectal cancer cell lines. The expression level of miR-375 significantly increased in miR-375 mimics group compared with that in the negative control group. The high expression level of miR-375 significantly inhibited the mRNA expression of AEG-1. After transfection with miR-375 mimics, the cell viability was inhibited, the apoptotic rate was increased, the proportion of G1-stage cells was increased, and the proportion of S-stage cells was decreased.CONCLUSION: miR-375 inhibits the viability, mediates the cell cycle arrest and promotes the apoptosis of colon cancer HCT116 cells. miR-375 may act as a tumor suppressor in colorectal cancer by inhibiting AEG-1.  相似文献

AIM: To explore the effects of microRNA-129-3p (miR-129-3p) on the viability and migration of NIH3T3 cells during transforming growth factor-β (TGF-β)-induced transformation into myofibroblasts and the underlying molecular mechanisms. METHODS: RT-qPCR was used to examine the relative expression of miR-129-3p in renal cell carcinoma (RCC)-adjacent tissues and fibrotic renal tissue. NIH3T3 cells were stimulated with TGF-β to transform into myofibroblasts, and miR-129-3p expression level was detected. After transfection with miR-129-3p mimics for 48 h in vitro, the cell viability was measured by MTT assay, the protein expression level of Ki-67 was determined by Western blot, and the cell migration was observed by wound healing assay. The direct target of miR-129-3p was predicted by online database TargetScan and confirmed by dual-luciferase reporter assay. The expression level of target protein was further confirmed by Western blot. RESULTS: Compared with the RCC-adjacent tissues, the expression of miR-129-3p was down-regulated in fibrotic renal tissue (P<0.01). In TGF-β-induced NIH3T3 cell transformation into myofibroblasts, the expression of miR-129-3p was also decreased (P<0.01). Transfection with miR-129-3p mimics followed by TGF-β stimulation in the NIH3T3 cells inhibited the viability, Ki-67 expression and migration. TargetScan analysis showed miR-129-3p had binding sites in the 3'-UTR of Smad3, which was confirmed by dual-luciferase reporter assay. The results of Western blot further confirmed that miR-129-3p affected the expression of Smad3. CONCLUSION: miR-129-3p inhibits the viability and migration ability of NIH3T3 cells during TGF-β-induced transformation into myofibroblasts by directly targeting Smad3.  相似文献

AIM: To investigate the changes of peroxisome proliferator-activated receptors (PPAR)α/peroxisome proliferator activated receptor coactivator 1 alpha (PGC-1α) in doxorubicin (DOX) induced dilated cardiomyopathy (DCM) and its effect on the energy metabolism and myocardial function in mice. METHODS: Forty mice were randomly divided into 4 groups: control group, DOX group, PPARα inhibitor group and PPARα agonist group. The DCM model was established by injection of DOX. The protein levels of PPARα/PGC-1α were detected. The PPARα inhibitor and PPARα agonist were used 2 weeks beforeinjection of DOX. The contents of adenine acid and phosphocreatine (Pcr) in the mitochondria were measured by high-performance liquid chromatography (HPLC). The ANT activity was analyzed by the atractyloside-inhibitor stop technique. The changes of the echocardiography and hemodynamics were also observed. RESULTS: DOX induced DCM model was successfully established. The protein levels of PPARα and PGC-1α in control group were significantly higher than those in DOX group (P<0.05). Both of the high-energy phosphate contents and the transport activity of ANT were decreased in DOX group (P<0.05), and the hemodynamic parameters were disordered (P<0.01). Compared with DOX group, PPARα inhibitor pre-treatment significantly reduced the PPARα/PGC-1α expression. Meanwhile, high-energy phosphate contents in the mitochondria and the ANT transport activity of the mitochondria decreased, as well as the left ventricular function (P<0.05). On the other hand, PPARα agonist significantly increased the expression of PPARα and PGC-1α, and improved the transport activity of ANT. In addition, the hemodynamic parameters were ameliorated, but the high-energy phosphate contents of the mitochondria did not significantly change. CONCLUSION: PPARα/PGC-1α plays an important role in the regulation of ANT transport activity in dilated cardiomyopathy induced by DOX, and the activation of PPARα/PGC-1α has protective effects on the DCM induced by DOX.  相似文献

AIM: To investigate the effect of microRNA-17 (miR-17) on the senescence of vascular smooth muscle cells (VSMCs) and the underlying mechanism.METHODS: The medial layer of the thoracic aorta was collected from the SD rats and isolated for primary culture. VSMCs were identified by immunofluorescence staining. The VSMCs were collected at the 4th~6th generations, and then the miR-17 mimics and miR-17 inhibitor were transfected into the VSMCs by liposome method. After 24 h, the cell senescence was induced by D-galactose. The VSMCs were divided into the following 6 groups:aging induction+miR-17 mimics (A-miR-17) group, aging induction+miR-17 inhibitor (A-anti-miR-17) group, A-control group, normal (N)+miR-17-mimics (N-miR-17) group, N-anti-miR-17 group, and N-control group. On day 3 after the addition of D-galactose, the senescence of VSMCs was observed with β-galactosidase staining. The expression of miR-17, p16 and p21 was detected by RT-qPCR and immunohistochemistry. RESULTS: miR-17 expression in the VSMCs was significantly lower in A-control group than that in N-control group (P<0.01). Compared with A-control group, the expression of miR-17 in the VSMCs was significantly increased in A-miR-17 group (P<0.01), while that was significantly decreased in A-anti-miR-17 group (P<0.01). The number of β-galactosidase positive staining cells in A-anti-miR-17 group was significantly higher than that in A-miR-17 group (P<0.01). The expression of p21 at mRNA and protein levels in the VSMCs was significantly lower in A-miR-17 group than that in A-control group (P<0.01), and the expressions of p21 at mRNA and protein levels was significantly higher in A-anti-miR-17 group than that in A-miR-17 group (P<0.01). CONCLUSION: miR-17 inhibits rat VSMCs senescence induced by D-galactose, the underlying mechanism is associated with the inhibition of p21 expression.  相似文献

AIM: To investigate the role of endoplasmic reticulum (ES) stress in cardiomyocyte apoptosis induced by β1-adrenoceptor autoantibody (β1-AA). METHODS: The rat model of active immunization with the second extracellular loop of β1-adrenoceptor was established, and SA-ELISA was applied to detect the level of β1-AA in serum of actively immunized rats. The apoptosis of cardiomyocytes was detected by TUNEL staining, and the protein expression levels of glucose-regulated protein 78 (GRP78), C/EBP homologous protein (CHOP) and caspase-12 in rat heart tissues were determined by Western blot and immunohistochemistry. After purified β1-AA obtained by affinity chromatography was used to treat H9c2 myocardial cells, the cell viability was measured by CCK-8 assay and the apoptosis was analyzed by flow cytometry with Annexin V-FITC/PI double staining. The H9c2 cells were treated with ER stress inhibitor 4-phenoxybutyric acid (4-PBA) before interfered with β1-AA, and the changes of cell viability and apoptosis were determined by CCK-8 assay and flow cytometry. RESULTS: Compared with vehicle group, the level of β1-AA in the serum of rats was significantly increased after active immunization for 2 weeks and further rised in 8 weeks, and increased apoptosis was observed in cardiomyocytes after active immunization for 2 weeks, lasting till 8 weeks. Compared with vehicle group, the protein expression of GRP78, CHOP and caspase-12 increased after active immunization for 4 weeks and 8 weeks. Continuous reduction of cell viability and increased apoptosis of H9c2 cells were induced by β1-AA. ER stress inhibitor 4-PBA pretreatment in H9c2 cells reversed the increased apoptosis and decreased cell viability induced by β1-AA, indicating that suppression of ER stress effectively reduced cardiomyocyte apoptosis. CONCLUSION: β1-AA induces increased apoptosis in cardiomyocytes by activating ER stress.  相似文献
