Summary

The acid‐base status of 58 calves (all in normal anterior presentation) was determined from venous blood samples before the onset of traction and immediately after vaginal delivery.

Calves were assigned to one of three groups according to their blood pH value: group 1 ‐ normal, pH >7.2; group 2 ‐ acidotic, pH 7.2 to 7.0; group 3 ‐ severely acidotic, pH < 7.0. Before the onset of traction (i.e. during the 30 minutes following the appearance and rupture of the membranes), 43 calves (74.1%) had normal acid‐base values, 14 (24.1%) had slight acidosis, and only 1 (1.7%) had severe acidosis. At birth the three groups of calves showed the following distribution: 23 (39.7%) were normal, 29 (50%) had slight acidosis, and 6 (10.3%) had severe acidosis.

Seven calves (12. 1%) died during birth or within 48 hours after birth; 2 were normal shortly before birth, 4 were acidotic shortly before birth, and 1 was severely acidotic even before the onset of traction. Traction was significantly longer for cows that delivered severely acidotic calves compared to cows in the other two groups.  相似文献

The adaptation of newborn calves to extra-uterine life was evaluated by measuring arterial blood gases, acid-base values, blood ions and lung mechanical function parameters in normal and acidotic calves during the first 24 h. Twenty-seven Holstein Friesian newborn calves were divided into two groups according to their immediate post partum arterial blood pH values (Group A blood pH > or = 7.2: normal group; Group B blood pH between 7.2 and 7.0: acidotic group). Pulmonary function parameters were measured and arterial blood samples were analysed for blood gases, acid-base variables and ion content immediately post partum (within 2 min) and then 6 and 24 h after calving.Lung resistance and maximal difference between pressure maximum and pressure minimum (max delta Ppl) decreased, while dynamic lung compliance increased significantly in both groups. Immediately post partum the lung resistance and max delta Ppl were significantly higher in the acidotic group than in the normal group. The arterial blood pH progressively compensated with time in both groups during the first 24 h and there was no difference in arterial blood pH values between the two groups 6 h after birth. These results showed that the compensation of acidosis was associated with the improvement in lung mechanics and these changes occurred mainly during the first 6 h of life. Moderate to pronounced acidosis did not affect the pulmonary adaptation negatively, although some respiratory mechanics parameters (max delta Ppl), blood pH and Ca(2+)ion concentrations remained significantly different between the normal and acidotic groups at 24 h. This might be the result of overcompensation of acidosis and the interdependence between blood pH and Ca(2+)concentrations.  相似文献

Blood pH, total plasma carotene, plasma vitamin A fractions and some plasma proteins were determined in newborn calves (n = 38) and their dams. Calves were assigned to one of two groups according to their blood pH immediately after birth: group 1 (normal) with pH above 7.2 (n = 27) and group 2 (slightly acidotic) with pH 7.0 to 7.2 (n = 11). The difference between the neonatal blood pH values of the two groups immediately post partum disappeared 24 h after birth. The blood pH of the dams was normal. The extremely low plasma carotene concentration (group 1: 32.7 micrograms/l; group 2:20.5 micrograms/l) and the low plasma globulin fractions (group 1:19.6 g/l; group 2:19.2 g/l) immediately after birth were elevated by the intake of colostrum. There was no significant change in plasma vitamin A fractions (retinol, retinyl ester) during the early postnatal period (3 days post partum) in newborn calves. The acid-base balance of newborn calves does not have any influence on the physiological alterations of plasma vitamin A, carotene and some plasma proteins during the first three days of life.  相似文献

OBJECTIVE: To determine concentrations of electrolytes, total bilirubin, urea, creatinine, and hemoglobin; activities of some enzymes; and Hct and number of leukocytes and erythrocytes of newborn calves in relation to the degree of acidosis and treatment with a hypertonic sodium bicarbonate (NaHCO(3)) solution. ANIMALS: 20 acidotic newborn calves with a blood pH < 7.2 and 22 newborn control calves with a blood pH > or = 7.2. PROCEDURES: Approximately 10 minutes after birth, acidotic calves were treated by IV administration of 5% NaHCO(3) solution. The amount of hypertonic solution infused was dependent on the severity of the acidosis. RESULTS: Treatment resulted in a significant increase in the mean +/- SEM base excess from -8.4 +/- 1.2 mmol/L immediately after birth to 0.3 +/- 1.1 mmol/L 120 minutes later. During the same period, sodium concentration significantly increased from 145.3 +/- 0.8 mmol/L to 147.8 +/- 0.7 mmol/L. Mean chloride concentration before NaHCO(3) administration was significantly lower in the acidotic calves (99.6 +/- 1.1 mmol/L) than in the control calves (104.1 +/- 0.9 mmol/L). Calcium concentration in acidotic calves decreased significantly from before to after treatment. Concentrations of potassium, magnesium, and inorganic phosphorus were not affected by treatment. CONCLUSIONS AND CLINICAL RELEVANCE: Administration of hypertonic NaHCO(3) solution to acidotic neonatal calves did not have any adverse effects on plasma concentrations of several commonly measured electrolytes or enzyme activities. The treatment volume used was smaller, compared with that for an isotonic solution, which makes it more practical for use in field settings.  相似文献

Under adverse obstetrical conditions, appropriate supervision and assistance during the immediate neonatal period are of the utmost importance, especially for weak calves. The aim of this study was to establish the effects of dystocia and oxytocin infusion on neonatal vitality, acid-base balance, and electrolyte and haematological homeostasis of dairy calves. Data were collected for 30 Holstein calves which were allocated to three groups: normal calving (n = 10); dystocia with mild to severe obstetric assistance (n = 10); and uterine inertia treated with oxytocin (n = 10). All 30 calves exhibited normothermia at birth, but had a significant decrease in body temperature after 60 min. Dystocic calves had lower Apgar scores than calves in the other two groups, and had respiratory and metabolic acidosis. Calves from normal calvings had normal blood pH, but base excess below the reference range. The mean partial pressure (Pa) of oxygen of calves whose dam had been treated with oxytocin was lower than that of calves from normal calvings. In all experimental groups, there was improvement in metabolic status in the first 60 min postpartum as PaCO2 values significantly decreased. All calves had normonatraemia, normokalaemia and normochloridaemia during the study period, but calves born to dams treated with oxytocin had a higher sodium concentration than those in the two other groups.  相似文献

Evaluation of acid-base disturbances in calf diarrhoea   总被引:1,自引:0,他引:1  
The severity of acid-base disturbances in diarrhoeic calves was investigated and a simple, inexpensive method of monitoring them was evaluated. The Harleco apparatus measures the 'total carbon dioxide' in a blood sample, mostly generated from the bicarbonate present, and any abnormalities are mainly due to metabolic acidosis or alkalosis. Its performance was tested against a standard blood gas analyser by comparing the results obtained by both methods with nearly 2000 blood samples from healthy or diarrhoeic calves. After technical modifications, the technique gave excellent precision and accuracy for the clinical evaluation of acid-base balance, using venous whole blood. The samples were very stable, especially at 0 degrees C, but also at room temperature. The normal range (mean +/- 1.96 sd) for total carbon dioxide in whole blood from calves was 21 to 28 mmol/litre. For samples corresponding to mild, moderate or severe acidosis, 79 per cent were correctly classified by the Harleco apparatus and only 0.1 per cent were beyond the adjacent degree of severity. After four days of diarrhoea, the calves which later died had twice the deficit in plasma bicarbonate of those which survived. As death approached, the deficit was almost three times that in surviving calves and the blood pH shortly before death was as low as 6.79 +/- 0.08. The Harleco apparatus was less successful with alkalotic samples, but metabolic alkalosis is less common and usually less severe.  相似文献

The effect of postnatal acid-base status on the absorption of colostral immunoglobulins by calves was examined in 2 field studies. In study 1, blood pH at 2 and 4 hours after birth was related to serum IgG1 concentration 12 hours after colostrum feeding (P less than 0.05). Decreased IgG1 absorption from colostrum was associated with respiratory, rather than metabolic, acidosis, because blood PCO2 at 2 and 4 hours after birth was negatively related to IgG1 absorption (P less than 0.05), whereas serum bicarbonate concentration was not significantly related to IgG1 absorption. Acidosis was frequently observed in the 30 calves of study 1. At birth, all calves had venous PCO2 value greater than or equal to 60 mm of Hg, 20 of the calves had blood pH less than 7.20, and 8 of the calves had blood bicarbonate concentration less than 24 mEq/L. Blood pH values were considerably improved by 4 hours after birth; only 7 calves had blood pH values less than 7.20. Calves lacking risk factors for acidosis were examined in study 2, and blood pH values at 4 hours after birth ranged from 7.25 to 7.39. Blood pH was unrelated to IgG1 absorption in the calves of study 2. However, blood PCO2 was again found to be negatively related to colostral IgG1 absorption (P less than 0.005). Results indicate that postnatal respiratory acidosis in calves can adversely affect colostral immunoglobulin absorption, despite adequate colostrum intake early in the absorptive period.  相似文献

In depressed calves (modified APGAR score 4-6) there is at birth an evident combined respiratory-metabolic acidosis (pH = 7.082 +/- 0.175; pCO2 = 73.3 +/- 26.8 mm Hg; BE = -10.6 +/- 7.2 mmol/l). The metabolic adaptation is completed after 6 hours, the respiratory acidosis is present up to 24 hours after delivery. In comparison to normal calves there are significant deviations in pH-values, base excess standard bicarbonate and actual bicarbonate during the whole investigation time. The carbon dioxide tensions of the depressed calves are at birth similar to those of normal calves, but in the following hours they are significantly higher. A definite relationship can be demonstrated between the 1 minute APGAR score and pH-value, base excess, standard bicarbonate and actual bicarbonate. Oxygen tension, oxygen saturation and carbon dioxide do not correlate with the clinical condition.  相似文献

After examination of the clinical state of 128 new-born calves, blood was collected from their vena jugularis for the determination of blood actual pH value, concentration of lactic acid, pCO2, base excess, buffer base and standard acid bicarbonate. The course and difficulty of parturition exerted a significant influence on the vitality of the calves and on the studied parameters of acid-base state. In the normally born calves, compared with those after dystocia, the following values were obtained: pH 7.20 +/- 0.03 : 7.11 +/- 0.07, pCO2 = 8.4 +/- 0.9 : 10.0 +/- 1.1 kPa, base excess -2.30 +/- 2.10 : 5.80 +/- 4.60 mmol/l, buffer base 43.0 +/- 2.4 : 39.5 +/- 6.5 mmol/l, standard acid bicarbonate 22.3 +/- 1.8 : 19.6 +/- 4.1 mmol/l and lactic acid concentration 5.6 +/- 2.0 : 10.7 +/- 5.1 mmol/l. The differences were statistically significant (P less than 0.05) and statistically highly significant (P less than 0.01). The continual study of the blood actual pH value and lactic acid concentration in the calves in the first 24 hours of life showed that with the same trend of changes in calves after dystocia the initial values were less favourable and that their normalization lasted longer. Attention is drawn to the importance of dystocia for the rise of respiratory metabolic acidosis and its effect on the vitality of newborn calves, and/or on their survival. The discussion deals with the importance of immunoglobulin levels in calves in the first days after birth for their further development. The determination of antibody content in colostral serum from the first milking in 33 and 29 cows on two farms showed great drawbacks in quality. A satisfactory level of IgG was found only in 36.36% and 58.62% of the cows, and a satisfactory level of IgM only in 12.12% and 24.13% of the studied cows. The determination of immunoglobulin content in their calves two to three days from birth (33 + 33 animals) showed normoglobulinemia only in 24.24% and 15.15% of cases. In 33 and 29 cows on two farms the colostrum serum from the first milking had an average content of immunoglobulins of class G amounting to 27.99 +/- 20.25 mg/ml and 36.95 +/- 21.62 mg/ml, and class M amounting to 3.64 +/- 1.25 and 2.04 +/- 1.42 mg/ml. Three days from birth, their calves had an IgG content of 4.25 +/- 2.57 mg/ml and 3.99 +/- 1.86 mg/ml and an IgM content of 0.30 +/- 0.20 and 6.38 +/- 0.25 mg/ml.  相似文献

Thirty-six dehydrated diarrheic neonatal calves were used to study the effects of various alkalinizing compounds on acid-base status, the changes in central venous pressure (CVP) in response to rapid IV infusion of large volumes of fluid, and the correlation of acid-base (base deficit) status, using a depression scoring system with physical determinants related to cardiovascular and neurologic function. Calves were allotted randomly to 4 groups (9 calves/group). Over a 4-hour period, each calf was given two 3.6-L volumes (the first 3.6 L given in the first hour) of a polyionic fluid alone (control group) or were given the polyionic fluid with sodium bicarbonate, sodium L-lactate, or sodium acetate added (50 mmol/L). Acid-base status, hematologic examination, and biochemical evaluations were made immediately before infusion of each fluid (at entry) and after 3.6, 4.8, and 7.2 L of fluid had been given. Compared with control values, bicarbonate, lactate, and acetate had significantly greater alkalinizing effects on pH (P less than 0.01) and base deficit (P less than 0.01) after 3.6, 4.8, and 7.2 L of fluid were given. Bicarbonate had the most rapid alkalinizing effect and induced greater changes in base deficit (P less than 0.01) than did acetate or lactate at each of the 3 administered fluid volumes evaluated. Acetate and lactate had similar alkalinizing effects on blood. Rehydration alone did not improve acid-base status. The CVP was elevated in 10 (28%) of the 36 calves after 1 hour of fluid (3.6 L) administration, but significant differences in body weight, PCV, and clinical condition or depression score at entry were not found between calves with elevated CVP and those with normal CVP.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

We investigated the diet content and properties of blood and milk in 11 pairs of Japanese Black cows and their calves for the one month following delivery. Four pairs (group A) had been no cases of white diarrhea during the year prior to this study, and 7 pairs (group B) had been a high occurrence of white diarrhea in calves during the same period. Properties of dams' diet in groups A and B before the onset of white diarrhea in calves were as follows: TDN fullness rate 98 +/- 2% vs. 110 +/- 5%, DCP fullness rate 151 +/- 2% vs. 200 +/- 33%, and starch content 5 +/- 2% vs. 14 +/- 3%. Blood and milk samples were collected from cows weekly and at the onset of white diarrhea in calves. No calves in group A had white diarrhea, while 5 out of 7 calves in group B had the diarrhea in this study. All cows were fed mixed hay, but 3 out of 5 cows whose calves had white diarrhea had been additionally given 3.0-3.5 kg of corn-silage a day from 4-5 days before onset. In all cows in the white diarrhea development group, the fat content of milk increased by 2.94 +/- 1.82% at the day of onset in comparison before. Serum TG and BUN, respectively, increased by 3.8 +/- 1.3 mg/dl and 3.7 +/- 2.0 mg/dl. Feeding cows a low-starch diet and additional corn-silage may induce a transient increase in blood TG, BUN and milk fat, which may contribute to the occurrence of white diarrhea in calves.  相似文献

OBJECTIVES: To determine whether high PaCO2 reduced apparent efficiency of IgG absorption (AEA) in calves and whether assisted ventilation of calves with high PaCO2 increased AEA. ANIMALS: 48 Holstein calves. PROCEDURES: Arterial and venous blood samples were collected 1, 13, and 25 hours after birth; an additional venous sample was collected at 37 hours after birth. Arterial samples were analyzed for PaCO2, PaO2, pH, and bicarbonate and base excess concentrations; venous samples were analyzed for plasma IgG concentrations. On the basis of 1-hour PaCO2, calves were assigned to nonrespiratory acidosis (PaCO2 < 50 mm Hg; n = 19) or respiratory acidosis (PaCO2 > or = 50 mm Hg; 29) groups. Calves in the respiratory acidosis group were assigned randomly to receive no further treatment (n = 17) or to be given 5 minutes of assisted ventilation (12). All calves received between 1.8 and 2 L of colostrum 2, 14, 26, and 38 hours after birth. Plasma volume and AEA were determined 25 hours after birth. RESULTS: 1-hour PaCO2 had no effect on AEA or on plasma IgG concentrations determined 13, 25, or 37 hours after birth. Artificial ventilation had no effect on plasma IgG concentration or AEA. CONCLUSIONS AND CLINICAL RELEVANCE: Lack of effect of 1-hour PaCO2 on AEA and IgG concentration indicated that calves compensated for moderate acid-base imbalances associated with birth. Calves born with high PaCO2 achieved adequate plasma IgG concentrations if fed an adequate amount of high-quality colostrum early in life. The effect of artificial ventilation on PaCO2 was temporary and did not increase AEA.  相似文献

Acid-base balance was evaluated in calves with experimentally induced viral diarrhea. When blood pH decreased to less than 7.200, calves were assigned to treatment groups and fed milk replacer, electrolyte solution without bicarbonate, or electrolyte solution containing bicarbonate. Calves in the electrolyte treatment groups had lower mortality (P less than 0.05), were better hydrated (P less than 0.05), and were less acidotic (P less than 0.05) than calves fed milk replacer. Bicarbonate-containing electrolyte solution restored acid-base balance (P less than 0.05) and corrected depression better (P less than 0.05) than electrolyte solution that did not contain bicarbonate. Both electrolyte solutions were equally good at correcting dehydration.  相似文献

The respiratory component PvCO2 of acid-base-status was observed in n = 36 calves (age: x +/- s = 8.7 +/- 5.0 d) with neonatal diarrhea and an acidosis (venous blood-pH: < 7.30; x +/- s = 7.08 +/- 0.15). In n = 10 (28%) calves with a severe metabolic acidosis (pH: x +/- s = 7.03 +/- 0.12; BE: x +/- s = -22.1 +/- 5.3 mmol/l) the PvCO2 was decreased < 5.3 kPa (x +/- s = 4.5 +/- 0.5 kPa) and showed a distinct respiratory compensation. A PvCO2 between 5.3-6.7 kPa (x +/- s = 6.0 +/- 0.4 kPa) was observed in n = 16 (44%) acidotic calves (pH: x +/- s = 7.11 +/- 0.13; BE: x +/- s = -15.2 +/- 7.4 mmol/l). These n = 26 (72%) calves showed a simple metabolic acidosis which is well known for calves with neonatal diarrhea. The remaining n = 10 (28%) calves showed an increase of the PvCO2 > 6.7 kPa (x +/- s = 8.0 +/- 1.5 kPa). These animals had a mixed respiratory-metabolic acidosis (pH: x +/- s = 7.08 +/- 0.20; BE: x +/- s = -13.9 +/- 10.3 mmol/l), as the decrease of the pH could not be determined by the decreased metabolic component HCO3- of acid-base-status alone. Calves which died during hospitalization and calves with a PvCO2 > 6.7 kPa tended to be younger and showed partially significant lower values for the parameters of oxygen-supply PvO2 and SvO2. Lactate was significantly higher in dying calves but not in calves with a mixed acidosis which on the other hand were more dehydrated. The functional capacity of respiratory compensation of acidotic disorders in the calves studied promised to be almost the same as in dog and man. One reason for the failure of respiratory compensation in some calves could be a more severe hypovolemia. With the use of "venous hypoxemia" (decrease PvO2 and decrease SvO2) the detection of tissue hypoxia was easier than with lactate concentration.  相似文献

The aim of this study was to evaluate the efficiency of vaccination of young calves and to see whether maternal antibodies may influence the immunological response in calves. For this project 20 matched-pairs of cows and their offspring were selected. Of each pair, one cow received a placebo 8 and 4 weeks before term (group A) and the other was vaccinated against Feline Leucose Virus, FeLV, with Leucogen? (group B). All calves received colostrum from their respective mother shortly after birth and all calves were vaccinated with Leucogen? 10 days after birth. Blood samples from the cows and calves were taken during the whole study period (till four weeks after calf vaccination). An ELISA test was done in the lab to define the FeLV antibody concentration. 30 % of the vaccinated cows showed a seroconversion, 13 out of 20 vaccinated cows passed the antibodies onto their calves. 11 calves of group B did not convert in comparation of only 4 of group A. All seroconverted calves had low antibody concentration before their vaccination. Calves of group B with a low passive antibody level at the beginning showed a higher seroconversion as compared to calves with higher antibody concentrations of the same group. Two thirds of the calves without maternal antibodies reacted adequately to the vaccination. Therefore, an early vaccination of calves can be recommended.  相似文献

The aim of the study was to compare the acid-base (AB-) findings in cattle obtained parallelly by the equilibration method after Astrup and the partial method (titration determination of total carbon dioxide in blood plasma) and at the same time to determine pH, net acid-base exudate in urine and the specific weight of urine. Altogether we made 65 parallel examinations in three groups of cows according to phases of reproduction. In 70.8% of cases correlation between the two results was very good or good, however under the condition that in many cases it was necessary to know also blood pH for the employment of the partial method. Without the knowledge of the blood pH good correlation would be obtained only in 33.8% of all cases. The partial method does not provide satisfactory results mainly in diagnosing respiratory disturbances and in determining compensatory degree of acid-base disturbances. We analysed two alternative values for the top, reference limit of vol. % CO2 in plasma: 60.0 or 55.0. The results imply that from the aspect of correct partial method interpretation it is more accurate to use the value 60.0. A high interpretation correlation between pH and net acid-base urine exudate was also confirmed and that independent urine pH values may be used in a complex acid-base examination to obtain acid-base findings. The representation of the basic types of acid-base disturbances indicates that metabolical acidoses are most frequent in highly pregnant cows (36.9%) mostly without proved compensation. The largest proportion of normal acid-base findings (43.4%) was determined in cows after calving in which also the compensatory mechanisms were most often activated. In connection with these findings the possibilities of impaired health of calves are discussed.  相似文献

The effect of the competitive relationship between potassium and hydrogen in the excretory mechanism of the kidneys on metabolism was studied in calves. The intraruminal exposure of calves to 5.6 meq potassium chloride (KCl) per kg per day has, in itself, only a slight acidotic effect ascribable to a slight reduction of the excretion of acid by the kidney. The same dose of ammonium chloride (NH4Cl) has a strong acidotic effect combined with a negative influence on weight gain. Both basic types of exposure increase the concentration of inorganic phosphorus and reduce the concentration of chlorine in the blood serum. The addition of 2.8 meq of potassium chloride per kg per day to the basic dose of ammonium chloride increases metabolic acidosis, worsens the health condition (being, at times, hazardeous to life) and leads to weight losses. The same amount of ammonium chloride added to the basic dose of ammonium chloride has a relatively small acidotic action on the blood, evoking only a slight and temporary potassium retention in the serum. The negative action on the weight gains and clinical symptoms are generally weaker than in the other group. The results indicate that the acidotic environment creates in ruminants a very dangerous situation if the organism is excessively exposed to the effect of potassium at the same time.  相似文献

Carbicarb (an equimolar mixture of sodium bicarbonate and sodium carbonate) was compared with sodium bicarbonate alone for the treatment of acidosis in newborn calves: 25 of 49 calves with a blood pH at birth of less than 7-2 and a base deficit of less than -3 mmol/litre were treated intravenously with sodium bicarbonate and 24 were treated with carbicarb. The doses were calculated on the basis of the base deficit in a blood sample taken 10 minutes after birth, and further blood samples were taken immediately after the treatment and 30 and 60 minutes after the treatment for the determination of acid-base status, blood gases and haematological and biochemical variables. Both treatments resulted in a significant increase in blood pH, but there was no difference between them. The mean (sd) blood pH before treatment was 7.09 (0.02) and after treatment it was 7.28 (0.01). There was no increase in the partial pressure of carbon dioxide after treatment with either sodium bicarbonate or carbicarb. Both treatments were associated with an increase in sodium concentration and decreases in the total erythrocyte count, haematocrit and haemoglobin concentration.  相似文献

Thirty-seven of 53 diarrhoeic calves hospitalised for intravenous fluid therapy were classified as very severely acidotic (total carbon dioxide less than 8 mmol/litre) by using a Harleco apparatus. All the calves were given intravenously 10 to 20 litres of electrolyte solution which contained 144 mmol/litre sodium, 4 mmol/litre potassium, 113 mmol/litre chloride and 35 mmol/litre bicarbonate, and in addition the 37 very severely acidotic calves received 400 ml of 1M sodium bicarbonate in the first 5 litres of fluid administered. Sixteen of the 37 very severely acidotic calves had a distended right flank, suggesting the presence of a dilated fluid-filled viscus. Neither their history nor other clinical signs were useful predictors of the distension. The distended calves had significantly higher plasma concentrations of sodium and chloride, and significantly lower plasma creatinine concentrations than the calves which were not distended. Treatment was successful in all the 21 non-distended calves but four of the distended calves died despite treatment. The resolution of the distension in the successfully treated calves, coincided with a significant increase in plasma bicarbonate concentration and the passage of large amounts of malodorous mucoid faeces.  相似文献

OBJECTIVE: To compare plasma disposition of alkaloids after lupine challenge in cattle that had given birth to calves with lupine-induced arthrogryposis and cattle that had given birth to clinically normal calves and determine whether the difference in outcome was associated with differences in plasma disposition of anagyrine. ANIMALS: 6 cows that had given birth to calves with arthrogryposis and 6 cows that had given birth to clinically normal calves after being similarly exposed to lupine during pregnancy. PROCEDURES: Dried lupine (2 g/kg) was administered via gavage. Blood samples were collected before and at various time points for 48 hours after lupine administration. Anagyrine, 5,6-dehydrolupanine, and lupanine concentrations in plasma were measured by use of gas chromatography. Plasma alkaloid concentration versus time curves were generated for each alkaloid, and pharmacokinetic parameters were determined for each cow. RESULTS: No significant differences in area under the plasma concentration versus time curve, maximum plasma concentration, time to reach maximum plasma concentration, and mean residence time for the 3 alkaloids were found between groups. CONCLUSIONS AND CLINICAL RELEVANCE: Because no differences were found in plasma disposition of anagyrine following lupine challenge between cattle that had given birth to calves with arthrogryposis and those that had not, our findings do not support the hypothesis that between-cow differences in plasma disposition of anagyrine account for within-herd differences in risk for lupine-induced arthrogryposis.  相似文献
