Leptospirosis is a zoonotic infection caused by pathogenic members of the genus Leptospira spp. Knowledge of the prevalent serovars and their maintenance hosts is essential to understand the disease. The aim of this study was to evaluate the ability of serology by the microscopic agglutination test (MAT) to predict the serogroups compared with results of identification of leptospires in São Paulo, Brazil. MAT correctly assigned the serogroup of the infecting isolate in 49/52 cases (94.23%). The serogroup Icterohaemorrhagiae was the predominant serogroup (88.46%). This study showed the usefulness of the MAT to correctly identify the infecting serogroup with a good overall agreement between the serologically-identified infecting serogroup and by identification of the isolate and can be used in epidemiological surveys in São Paulo. However, it should be complemented by the identification of Leptospira isolates.  相似文献

During an investigation of pig abortions and stillbirths in Northern Ireland, leptospires were isolated from 55 of the 78 litters examined. Strains belonging to four serogroups (Australis, Icterohaemorrhagiae, Hebdomadis and Autumnalis) were recovered but leptospires of the Australis serogroup accounted for 91 per cent of the isolates. Leptospirosis in horses is often associated with reproductive disorders. In the southern states of Brazil, horses are used for various jobs and cultural practices; nevertheless, serological surveillance for Leptospira is rare. Therefore, the objective of this study was to determine the seroprevalence of Leptospira spp. in horses in southern Brazil, as well as to identify the risk factors for infection and its impacts on reproduction. We performed microscopic agglutination tests for 12 serovars that corresponding 9 serogroup (Sejroe, Icterohaemorrhagiae, Australis, Pyrogenes, Pomona, Canicola, Grippotyphosa, Tarassovi and Ballum) in 595 samples from 60 herds. A brief history was obtained to analyze risk factors for reproductive disorders. A total of 45.9% of the tested horses were seropositive, of which the most frequent serogroups were Icterohaemorrhagiae (Icterohaemorrhagiae and Copenhageni serovars) and Ballum (Ballum serovar). Simple infections were found in 45.4% of seropositive animals, while mixed infections occurred in 54.6% of horses. There was a correlation between seropositivity and age and sex, that is, seropositivity was more frequent in animals over 6 years old and in females. There was no correlation between seropositivity and reproductive disorders. We conclude that there is a high seroprevalence of Leptospira spp. in southern Brazil with predominance of Icterohaemorrhagiae serogroup, mainly in older animals. Location, breeds, contact with dogs or other domestic animals are not risk factors, whereas gender is a risk factor. Reproductive disorders are not due to leptospirosis in the study region.  相似文献

Leptospirosis is a re-emerging bacterial zoonosis. North-Central Italy is characterized by a geographic area that promote Leptospira circulation. Data on sero-epidemiological survey carried out from 2002 to 2016 in North-Central Italy were reported and discussed. Overall, 709 out of the 8488 (8.35%) tested sera were positive for Leptospira at the cut-off titer (1:100) and 218 (2.57%) at higher titer (≥1:400). The highest percentages of positivity was recorded for coypus (22.86%), swine (19.74%) and bovine (13.03%). Pomona and Australis resulted the serogroup more often detected, followed by Sejroe and Icterohaemorrhagiae; while, a low number of positive sera was detected for serogroups Ballum, Canicola and Tarassovi. Percentage of positive sera for each year slightly decreased from 2002 to 2008 and rose from 2009. High percentages of positive reactions were recorded in 2014 (17.23%), 2015 (19.61%) and 2016 (38.05%). In conclusion, the results of this investigation reported an increase of leptospirosis in North-Central Italy. Furthermore, several animals resulted infected as accidental hosts by unusual Leptospira serovars. These data could suggest a change in host range for some serovars, that may promote the adaptation to new hosts.  相似文献

Four hundred and eighty bovine kidneys were collected from an abattoir near Harare between December 1987 and November 1988, and leptospires were recovered from 50 (10.4 per cent) of them. The isolates were identified to serogroup level by the microscopic agglutination test; 32 belonged to serogroup Sejroe, seven were Pyogenes, four Hebdomadis, two Tarassovi, and one isolate each belonged to serogroups Australis, Bataviae, Grippotyphosa, Icterohaemorrhagiae and Pomona.  相似文献

A study was conducted to determine the seroprevalence of leptospirosis and infecting serovars across livestock (cattle, sheep, goats, and pigs) in Trinidad using the microscopic agglutination test with an international panel of 23 serovars. Of a total of 590 cattle tested, 21.5% were seropositive with agglutinations to 13 of the 23 antigens used in the panel. Icterohaemorrhagiae (9.3%), Sejroe (4.1%), Ballum (4.1%), and Autumnalis (1.9%) were the predominant serogroups detected in the cattle sampled (n = 590). Of 222 sheep tested, 5.0% were seropositive with agglutinations to five serovars belonging to two serogroups. These serogroups were Autumnalis at 2.7%, and Icterohaemorrhagiae at 2.3% of all sheep tested (n = 222). Of a total of 180 goats tested, 3.3% were seropositive, all agglutinating to the Icterohaemorrhagiae serogroup, 1.7% to serovar Copenhageni, 1.1% to serovar Mankarso, and 0.6% to serovar Icterohaemorrhagiae. Among pigs (n = 200), 5.0% were seropositive for five serovars belonging to three serogroups. These serogroups were Icterohaemorrhagiae at 2.5%, Australis at 2%, and Ballum at 0.5%. Overall, age and sex of animals were not significantly associated with leptospirosis with the exception of cattle where age was a significant factor for seropositivity. It was concluded that for livestock, leptospirosis may be an important zoonotic and economic disease, particularly in the case of cattle. It is imperative that the impact of leptospirosis on abortion, stillbirths, and decreased milk production in livestock in the country be assessed.  相似文献

Leptospirosis is a worldwide zoonosis which can affect many species. Control programs need accurate diagnosis to be successful, and currently, diagnosis relies on serology. It presents three main issues: the sampling, the antigen panel, and the cutoff point. Herein, we propose a systematic review on leptospirosis among dogs, pigs, and horses in Latin America in order to improve the understanding of the seroepidemiology of leptospirosis in these species in the region as well as the temporal development of the research on this topic and, consequently, improve the chances of success on control programs. Internet databases were consulted over 2015. Inclusion criteria included serosurvey using MAT; a relevant number of animals; the presence in the antigen panel of at least one representative of serogroups Icterohaemorrhagiae and Canicola for dogs, Icterohaemorrhagiae, Australis, and Pomona for pigs, and Icterohaemorrhagiae and Australis for horses; and a cutoff point of ≥100. Overall, 240 papers were studied, of which 87 referred to dogs, 66 to pigs, 39 to horses, and 48 to more than one of the studied species. In relation to those that met all the inclusion criteria, it was 45 (66.2%) in dogs, 23 (41.8%) in pigs, and 23 (63.9%) in horses. Leptospirosis is widespread in Latin America. Predominant serogroups are Canicola to dogs and Icterohaemorrhagiae to pigs and horses. Therefore, research on animal leptospirosis should be encouraged in Latin America, in order to reach a greater standardization in studies and then achieve better results on control programs.  相似文献

Cattle are commonly infected with pathogenic leptospires, and similarly to rodents, they excrete the bacteria in their urine and can transmit the pathogen from animal to animal or animal to human. Thus, surveillance and monitoring systems for detection of new Leptospira serovars are important for the control of leptospirosis. Here, we report the isolation of a spirochete from a stillborn bovine foetus and its characterization by immunological and molecular techniques. A variable number tandem repeat profile using seven discriminatory primers identified the spirochete as belonging to species Leptospira interrogans serogroup Australis serovar Muenchen. A phenotypic analysis using monoclonal antibodies (mAbs) against leptospiral membrane-associated proteins confirmed the expression of important virulence and pathogenicity factors (LipL32 and LigBrep). Out of 120 reference sera tested, 22 positive (36.66%) and 9 negative (15%) also reacted with the new isolate. Furthermore, the serovar Muenchen isolate was virulent in hamster model. The animal inoculated developed acute lethal infection characterized by hepatic, pulmonary and renal lesions. Local isolates exhibited unique characteristics that differed from those of reference strains; therefore, isolation of leptospires is useful in the surveillance of local pathogenic serovars. In conclusion, the data obtained from this study can contribute to the epidemiological understanding and control of leptospirosis in southern Brazil.  相似文献

Leptospirosis is a serious bacterial disease that affects both humans and animals. A wide range of symptoms have been described in humans; the disease in dogs is commonly associated with kidney and/or liver disease. In Malaysia, information about the common serovars infecting dogs is limited. Therefore, we investigated the occurrences of leptospirosis in 124 pet dogs diagnosed with kidney and/or liver disease. Blood, urine, abdominal effusion, and/or kidney and liver were collected from the dogs. Based on microscopic agglutination testing, 53 of 124 (42.7%) dogs were seropositive for leptospiral exposure. Sera were frequently positive to serovars Bataviae (n = 12), Javanica (n = 10), and Icterohaemorrhagiae (n = 10). Direct detection using PCR showed that 42 of 124 (33.9%) of the whole blood and 36 of 113 (31.9%) urine samples were positive for pathogenic Leptospira spp. By PCR, 2 of 23 (9.1%) kidney and 2 of 23 (9.1%) liver were positive for pathogenic Leptospira spp. Abdominal effusion from 4 dogs were PCR-positive for pathogenic Leptospira spp. The species detected were L. interrogans, L. borgpetersenii, L. kirschneri, and L. kmetyi by partial 16S rRNA sequencing. We further identified and characterized 11 Leptospira spp. isolates from 8 dogs as serovars Bataviae, Javanica, and Australis. The mortality rate of the Leptospira-infected dogs was high (18 of 53; 34%).  相似文献

Eighteen isolates from the Australis serogroup from free-living and domestic animals were identified using the cross agglutination absorption test. Serovar muenchen was found only in England and Wales in wood mice, short tailed and bank voles, a grey squirrel and a pig. Serovar bratislava was found in hedgehogs in England, Wales and Northern Ireland and also in a brown rat from Northern Ireland. Serovar bratislava was isolated from sheep in both England and Northern Ireland and from horses in Northern Ireland. The distribution of these serovars in relation to possible maintenance hosts is discussed.  相似文献

Leptospirosis is a systemic disease affecting humans and animals, and pigs are generally considered the reservoir host species for the serovars Pomona, Bratislava and Tarrasovi. Endemic infections in swine herds generally remain subclinical, as do the vast majority of leptospire infections. However, when a susceptible breeding herd is infected for the first time or its immunity is compromised, considerable losses can occur due to abortion, stillbirths, weakly piglets or infertility. Infections in pigs caused by other serovars tend to occur only incidentally, vary regionally, and depend on other reservoir hosts, primarily rodents. Leptospires persist in porcine kidneys, and the Bratislava serovar, in the genital tract; it is excreted in urine and genital fluids. Leptospirosis is transmitted by direct or indirect contact with an infected animal. Fundamental research on porcine leptospirosis was conducted in the 1970s and 1980s. However, despite subsequent application of the most recent molecular biological methods, the pathogenesis of porcine leptospirosis is still largely unknown, and research results from the last 25 years on its incidence are very heterogeneous, due not only to regional differences but also to differences in the evaluation of diagnostic and population studies. Serological testing of pigs showed serovar prevalences ranging between as much as 16.3% (Pomona) and generally no more than 2.9% (Tarassovi), whereas antibodies against Bratislava were found in as many as 41.8% of pigs tested during the last 20 years, as in previous studies, indicating that this remains the most prevalent serovar.  相似文献

Leptospirosis, a disease more common in the tropics, can cause a life-threatening multisystemic syndrome in humans and animals. Immunity, whether natural or vaccine-induced, is serogroup-specific with the infecting serovars varying according to geographical locality. In South Africa, in spite of the fact that the bacterin vaccine for some Leptospira serovars is often used, there is no recent information on the incidence of canine leptospirosis as well as the infecting serovar/s. The aim of this study, which was undertaken on sera collected in 2008 and 2009 from both strays and owned dogs predominantly in the coastal regions of South Africa, was to determine the presence of leptospiral antibodies to 15 serovars known to infect dogs. Of the 530 samples tested, 25 tested positive to 7 different serovars with the microscopic agglutination test (MAT). Nine of the 25 samples tested positive to more than one serovar. The 2 serovars most frequently represented were Canicola, which reacted to 17 sera, and Pyrogenes, which reacted to 10 sera. Currently the only vaccines available in South Africa in different combinations contain serovars Canicola, Icterohaemorrhagiae, Pomona and Grippotyphosa. The results showed that the use of vaccines containing serovar Canicola is still justifiable in certain regions of the country. However, the presence of antibodies to serovar Pyrogenes in several dogs, pending a broader investigation, indicates that this serovar should also be included in the range of Leptospira vaccines for use in South Africa.  相似文献

Wildlife plays a pivot role in the epidemiology of leptospirosis and rodents have a reservoir function for several Leptospira serogroups. The crested porcupine is the largest rodent of the Italian fauna and shares the same environment with red foxes, badgers, coypus and wild boars that are known to be infected by Leptospira. Between 2018 and 2019 the seroprevalence of Leptospira in crested porcupine was investigated for the first time. Overall 7 out of 14 sera (50 %) were found positive to Leptospira. Icterohaemorrhagiae resulted as the most detected serogroup (57 %) followed by Pomona, Australis and Sejroe. The highest titer (1:1600) was detected for the serogroup Australis. These results indicate that porcupines could be infected by several serogroup of Leptospira and the role of reservoir or accidental host need to be addressed. Further investigations are necessary in order to clarify the leptospirosis – epidemiology – wildlife framework in light of its potential zoonotic source.  相似文献

The prevalence of Leptospirosis in goat herds of the State of Minas Gerais has seldom been studied. The present research had as its objectives: (1) investigate the seroprevalence of Leptospirosis in the county of Uberlandia, MG, (2) verify the Leptospirosis serovars, and (3) identify the risk factors associated with infection on the farms examined. Serum samples from 230 animals in 11 properties were tested using the microscopic agglutination test. An epidemiological examination furnished data for analysis regarding the risk factors. The prevalence of Leptospirosis was found to be 31.3% with variation from 1:100 to 1:800. The most frequent serovars were: Autumnalis (30.30%), Tarassovi (19.20%), Pyrogenes (13.13%), and Icterohaemorrhagiae (11.11%). The ages and races of the animals were among the risk factors found to be significantly correlated (P < 0.05) with infection. At the farm level, the intensity of production, use of salaried workers, and association of other animals were all found to be related with the frequency of Leptospirosis. The results demonstrated that inadequate management was a factor which favored the occurrence of infection in the region of the study.  相似文献

Clinical and serological evidence has indicated that human leptospirosis in Fiji is an important disease, and the prevalence of antibody is exceptionally high. A serological survey of the rural population showed that only 12% of the people studied did not have complement fixing (CF) leptospiral antibody. As the origin of this infection could not be explained by the known distribution of leptospiral infection in domestic and wild animals, a serological survey using the complement fixation test (CFT) was undertaken as the first stage of an epidemiological investigation into human and animal leptospirosis. Sera from domestic and wild animals were tested for CF antibody to 12 leptospiral serovars, namely: pomona, copenhageni, grippotyphosa, hardjo, ballum, tarassovi, canicola, australis, bratislava, autumnalis, pyrogenes and bataviae. Antibody was detected in 27.5% of 480 cattle, 17.1% of 70 sheep, 10.3% of 252 goats, 10.0% of 480 pigs, 57.0% of 100 dogs, 55.8% of 34 rats (Rattus rattus, R. frugivorus, R. exulans and R. norvegicus), 53.1% of 32 mongooses (Herpestes auropunctatus) and 40.0% of 10 mice (species unknown.) Cross-reactivity precluded the identification of infecting serogroups with the exception of pomona in pigs and icterohaemorrhagiae, ballum and australis in dogs. Infection of dogs with a serovar of the australis serogroup may explain the predominance of serological reactions to bratislava in man. The survey revealed a significant level of leptospiral antibody in the animal populations of Fiji and indicated that cattle, dogs, rats, mongooses and mice are probably the most important maintenance hosts. Consequently, further investigation will concentrate on the attempted isolation of leptospires from these species.  相似文献

Three pregnant gilts were experimentally infected with leptospires of the serovar mozdok, isolated from an aborted pig fetus from a Portuguese pig farm with abortion problems. All the gilts aborted dead or dying piglets on days 105 or 106 of pregnancy. Serovar mozdok was isolated from 12 of the 22 piglets in the three litters. Histological examination of the livers and kidneys of the gilts at the end of the experiment revealed evidence of disease, and leptospires were isolated from their kidneys. Their serological responses up to 42 days after inoculation were monitored by means of a microscopic agglutination test, using 21 antigens from 18 serogroups. Cross reactions to heterologous antigens belonging to the Grippotyphosa, Australis, Icterohaemorrhagiae and Cynopteri groups were observed in all of them.  相似文献

Swine are one of significant reservoirs and sources of Leptospira infections for man. Serological screenings help to effectively control the epidemiological situation in swine herds and to prevent transmission of Leptospira from animals to man. The purpose of this study was to investigate, by the use of serological methods, the prevalence of infections caused by selected Leptospira serogroups in swine population in Poland. A total of 7112 swine serum samples were examined. The samples were collected from January to October 2008 and came from 280 counties situated in all 16 provinces of Poland. All sera were examined preliminary by enzyme-linked immunosorbent assay (ELISA) using heat-stable antigenic preparation. The samples positive or doubtful in ELISA were investigated by microscopic agglutination test (MAT) with use of serovars Icterohaemorrhagiae, Pomona, Canicola, Sejroe, Tarassovi and Of the collected sera examined by ELISA 73 (1.02%) samples were positive, 85 (1.20%)--doubtful and 6954--negative. Among ELISA-positive and doubtful sera 64 samples (coming from 14 provinces) were recognized in MAT as positive. Among MAT positive samples 42.19% of sera demonstrated titres with serovar Pomona, 32.81%--with Sejroe, 14.06%--with Icterohaemorrhagiae, 6.25%--with Tarassovi, 3.13%--with Grippotyphosa and 1.56% with Canicola.  相似文献

Serum samples from 77 animals belonging to 38 species and 19 families in Rio de Janeiro Zoo, Brazil were tested for antibodies against serovars of Leptospira interrogans by microscopic agglutination test. Antibodies were detected in serum samples of 37.7% of all animals belonging to 10 families. Seropositivity was more common in the Carnivora Canidae (7/9), Procyonidae (5/9) and the Edentata Myrmecophagidae (5/9). Icterohaemorrhagiae was the most common serogroup. Leptospirosis is a zoonosis of worldwide distribution affecting domestic animals, wildlife and man. The bacterial disease is caused by pathogenic Leptospira spp., which are transmitted from reservoir hosts to accidental hosts. Horses are accidental hosts and can become susceptible to leptospiral infections. Widespread exposure to leptospires exists and is significantly more common than clinical disease. Leptospirosis can have different clinical manifestations including abortion, still birth, systemic disease with hepatic or renal dysfunction, and equine recurrent uveitis (ERU). ERU is the most frequently encountered clinical manifestation and this article will focus on the review of leptospira‐associated ERU. Equine recurrent uveitis is the most common cause of vision impairment and blindness in horses. The pathogenesis of leptospira‐associated ERU involves direct bacterial effects and immune‐mediated responses. Clinical signs vary between the acute and chronic phases of the disease and progress over time. The diagnosis of leptospira‐associated ERU can be difficult and usually requires a combination of diagnostic tests. Medical and surgical treatments have been described with varying outcomes. The prognosis for sight is usually poor, although core vitrectomy may improve the outcome. Avoidance of leptospiral exposure of horses is the only reliable prevention of leptospira‐associated disease.  相似文献

Various tissues, nasal swabs, urine and blood samples were collected from 376 feral swine at two federally inspected abattoirs in Texas during six separate sampling periods in 2015. Samples were tested for Brucella spp. by culture and serology. Brucella spp. were cultured from 13.0% of feral swine, and antibodies were detected in 9.8%. Only 32.7% of culture‐positive feral swine were also antibody positive, and 43.2% of antibody‐positive feral swine were culture positive. Approximately, the same number of males (14.0%) and females (12.1%) were culture positive, and slightly more males (10.5%) than females (8.7%) were antibody positive. Our results indicate that serology likely underestimates the prevalence of feral swine infected, and that those who come in contact with feral swine should be aware of the symptoms of infection with Brucella spp. to ensure prompt treatment.  相似文献
