Objective To measure changes in the ocular and orbital blood flow velocities by color Doppler imaging (CDI) in beagles with primary open angle glaucoma as the disease progressed from early to advanced stages. Methods CDI measurements were performed periodically on 13 glaucomatous Beagles during the nontreated mild, moderate and advanced stages of POAG over the course of 4 years. CDI was performed with the dogs lightly anesthetized (butorphanol 0.1 mg/kg IV, acepromazine maleate 0.02 mg/kg IV, and atropine sulfate 0.05 mg/kg) while the CD transducer was placed directly on the cornea anesthetized with 0.5% tetracaine hydrochloride. Intraocular pressure (IOP) by pneumatonography or TonoPen XL, heart rate and mean arterial blood pressure were measured at the beginning, middle and end of each study. The ophthalmic vessels examined included: external ophthalmic arteries and veins, long and short posterior ciliary arteries, anterior ciliary arteries and veins, primary retinal arteries, and vortex veins. Recordings of each vessel included peak systolic velocity (PSV), end diastolic velocity (EDV) and time averaged velocity (TAV), and when possible the resistive index (RI) and pulsatility index (PI) were computed. Results CDI abnormalities were present before intraocular pressure exceeded the normal range. As the animals aged, and the glaucoma progressed with higher levels of IOP, significant changes occurred in nearly all vessels, and generally included a major increase in RI (P < 0.001) and an increase in the PI (P < 0.001). Mean arterial blood pressure (105 ± 18 mmHg) and heart rate (118 ± 33/min) remained reasonably constant. The IOP gradually increased as the disease progressed (early and normotensive: 19.4 ± 3.9 mmHg; moderate: 29.7 ± 2 mmHg; and advanced: 44.5 ± 6 mmHg). The ocular veins seemed most influenced early on in the disease. Late in the disease, ocular venous blood flow could not be consistently demonstrated. An increase in the PI of ocular veins occurred in the moderately and severely affected glaucomatous Beagles. As the IOP increased, there were trends of increasing resistive index and pulsatility index in most arteries, and periods of marked decreased velocities of the vortex and external ophthalmic veins in severe cases. Conclusion CDI measurements in Beagles with primary open angle glaucoma during the course of 4 years indicate easily measurable and repeatable progressive blood flow abnormalities before the elevation of IOP and, thereafter, with gradually increased levels of IOP.  相似文献

Testicular tissue maintenance is performed by the testicular artery, and the hemodynamics of this artery can be evaluated using Doppler ultrasonography. The aim of this study was to characterize the peak systolic velocity (PSV), end diastolic velocity (EDV), pulsatility index (PI) and resistivity index (RI) of five regions of the testicular artery in dogs, including two proposed regions and three that have been previously described. Twenty‐two dogs were used, and the PSV, EDV, PI and RI of the testicular artery were measured in five regions: proximal, medial and distal supratesticular; marginal; and intratesticular. The median values for PSV (cm/s), EDV (cm/s), PI and RI in the five regions were as follows: proximal supratesticular (23.1, 3.7, 2.1 and 0.8); medial supratesticular (17.2, 4.5, 1.5, and 0.7); distal supratesticular (12.2, 5.7, 0.8, and 0.5); marginal (11.3, 6.5, 0.5, and 0.4); and intratesticular (5.7, 3.5, 0.5, and 0.4). There was a difference between the PSV of the medial and distal supratesticular regions. There were differences in the PSV, EDV, PI and RI among the distal supratesticular, marginal and intratesticular regions. Measurements of PSV, EDV, PI and RI of the testicular artery in dogs at the proposed regions showed different results due to the hemodynamic and morphological differences of the artery during its course in the spermatic cord and to the testicles. It is necessary to identify the region in testicular artery Doppler velocimetric evaluations of dogs, given that there is a difference according to the region measured.  相似文献

Background

Canine atopic dermatitis can be a result of exposure to aeroallergens or trophallergens. Hemodynamic alterations occur in dogs with food hypersensitivity.

Hypothesis/Objectives

To evaluate if hemodynamic alterations occur in dogs with NFICAD with lowered resistance to diastolic flow at fasting, after feeding, or both.

Animals

Ten healthy dogs and 22 dogs with NFICAD were included from the hospital population.

Methods

Blinded prospective study. Peak systolic velocity (PSV), end diastolic velocity (EDV), mean velocity (MV), pulsatility index (PI), resistive index (RI) and PSV/EDV ratio were measured at fasting for both arteries (cranial mesenteric artery [CMA], celiac artery [CA]) and at 40 minutes after feeding in CMA and at 60 minutes in CA. The results were analyzed statistically with a mixed model.

Results

There was no difference detected between groups of dogs for any variable except EDV during fasting (= .01).

Conclusions and Clinical Importance

There is no decrease in resistance in NFICAD to diastolic flow. This observation could be explained by the absence intestinal inflammation in NFICAD.  相似文献

The accessory sex glands play a major role in the production of seminal plasma, and testicular artery blood flow seems to strongly influence testicular function. However, very little ultrasound imaging of these organs has been undertaken in donkeys. The present work reports the results of such examinations in five jackasses along the year. The accessory glands were inspected by B-mode ultrasound while the testicular artery blood flow was assessed by colour pulsed-wave Doppler ultrasound. The testicular artery was examined at pampiniform plexus (PPT), supratesticular area (ST) and capsular artery (CA). Values were recorded for the total arterial blood flow (TABF), peak systolic velocity (PSV), end-diastolic velocity (EDV), resistive index (RI), pulsatility index (PI) and time average maximum velocity (TAMV). Semen was obtained and assessed for sperm concentration, viability, abnormalities and motility using a CASA system. The bulbourethral glands, prostate and ductus deferens ampullae were relatively larger than in the stallion. Bulbourethral glands and ampullae sizes were inversely correlated with sperm motility. An reduction in blood flow between the level the PPP and the CA was observed, helping to reduce testis temperature and oxygen pressure. Blood flow at the CA showed the strongest correlation with semen production. PI and RI were positively correlated with the CASA motility variable STR (p = .02, p = .06) and sperm viability (p = .01), while sperm concentration (p = .05) correlated inversely with PSV, EDV, TAMV and TABF. EDV also correlated negatively with the CASA variables VSL, LIN, STR and VAP (p ≤ .05). PI and RI were also negatively correlated with testis length (p = .0093, p = −.0438).  相似文献

Objective To evaluate the effect of central corneal thickness (CCT) on the measurement of intraocular pressure (IOP) with the rebound (TonoVet®) and applanation (TonoPen XL®) tonometers in beagle dogs. Animal studied Both eyes of 60 clinically normal dogs were used. Procedures The IOP was measured by the TonoVet®, followed by the TonoPen XL® in half of the dogs, while the other half was measured in the reverse order. All CCT measurements were performed 10 min after the use of the second tonometer. Results The mean IOP value measured by the TonoVet® (16.9 ± 3.7 mmHg) was significantly higher than the TonoPen XL® (11.6 ± 2.7 mmHg; P < 0.001). The IOP values obtained by both tonometers were correlated in the regression analysis (γ2 = 0.4393, P < 0.001). Bland–Altman analysis showed that the lower and upper limits of agreement between the two devices were ?0.1 and +10.8 mmHg, respectively. The mean CCT was 549.7 ± 51.0 μm. There was a correlation between the IOP values obtained by the two tonometers and CCT readings in the regression analysis (TonoVet® : P = 0.002, TonoPen XL® : P = 0.035). The regression equation demonstrated that for every 100 μm increase in CCT, there was an elevation of 1 and 2 mmHg in IOP measured by the TonoPen XL® and TonoVet®, respectively. Conclusions The IOP obtained by the TonoVet® and TonoPen XL® would be affected by variations in the CCT. Therefore, the CCT should be considered when interpreting IOP values measured by tonometers in dogs.  相似文献

To investigate the ovarian and uterine blood flow responses, hemodynamic, circulating ovarian hormones and nitric oxide (NO) after end of treatment by Folltropin. Holstein Friesian (12) cows previously synchronized with CIDR underwent Doppler ultrasound after administrating of FSH daily for 4 days in eight injections started on day 10 of the second ovulation (day −5). Oestradiol (E2), progesterone (P4) and nitric oxide (NOMs) were measured. During the follicular phase, follicle area and antrum area of the second cycle reached maximum value on the day of ovulation compared with that in the first cycle, while during the luteal phase, both showed a pattern of increase and decrease. The luteal area and total coloured area increased till day 10 in the first and second cycle. The first cycle ipsilateral ovarian artery (Ov.A) had higher pulsatility (PI) (p = .001), resistance (RI) (p = .001), peak velocity (PSV) (p = .009) and lower end-diastolic velocity (EDV) (p = .003) compared with the second cycle. The increased ipsilateral Ov.A PSV (p = .009) was accompanied by lower EDV. The first cycle ipsilateral middle uterine artery (MUA) had higher PI (p = .001) and RI (p = .001), with lower PSV (p = .001) and EDV (p = .001). It was concluded that blood flow of ovarian and middle uterine arteries changed after the end of superstimulation as the increased ipsilateral Ov. A and MUA PSVs accompanied by lower EDV and both Doppler indices that reflect the amount of ovarian and uterine blood flow waveform.  相似文献

The haemodynamics of the canine left renal artery (LRA) and interlobar artery (ILA) were evaluated in eleven fasted, healthy, conscious beagles with severe acute (haematocrit [Hct] 16%), moderate chronic (Hct 26%) and mild chronic (Hct 34%) normovolaemic anaemia using Doppler ultrasound. Heart rate, peak systolic velocity (PSV), end diastolic velocity (EDV), time-averaged mean velocity (TAVmean), pulsatility index (PI) and resistive index (RI) were recorded. Doppler values in the dogs following the induction of anaemia states were compared with corresponding values in the same dogs prior to the induction of anaemia. Left renal artery mean PSV, mean PI and mean RI were significantly higher and the mean EDV was significantly lower in severe acute anaemia. No significant change was seen in mean values of the same parameters in moderate or mild chronic anaemia. There was no significant change in TAVmean of the LRA or mean PI and mean RI of the ILA in any grade of anaemia. Acute, severe normovolaemic anaemia significantly altered LRA Doppler parameters in resting dogs without influencing those of the ILA. Moderate or mild chronic anaemia had no effect on any renal Doppler parameter.  相似文献

This study was performed to characterize the normal blood flow of the canine testis and to measure the peak systolic velocity (PSV), end-diastolic velocity (EDV), resistive index (RI) and pulsatility index (PI) of testicular arteries weekly during a period of 6 months in five healthy Beagle dogs and to evaluate whether there were any change along this time. The ultrasonographic exams were made with an 11 MHz linear transducer. The vessels of the testes were subdivided into three categories: supratesticular arteries, marginal artery and intratesticular vessels. At the supratesticular arteries, two measurements were recorded at the cranial and the looping parts. No significant differences in any of the parameters studied were observed for the 6 months that the study was performed. The cranial part of the supratesticular artery showed a flow pattern of high-resistive vessel, whereas in the looping part of the supratesticular artery, marginal and intratesticular arteries, the flow showed a low-resistance pattern. PSV, RI and PI values were higher at the cranial part of the supratesticular artery, followed by the looping part of the supratesticular artery, marginal and intratesticular vessels. EDV measurements were higher in the looping part of the supratesticular artery.  相似文献

This study was carried out with the objectives to test the differences in the haemodynamic characteristics of the prostatic artery in normal and benign prostatic hyperplasia (BPH)–affected dogs using Doppler ultrasonography. In sixteen male German shepherd dogs, prostatic volume was detected and prostatic biopsy was performed. The prostatic artery blood flow parameters determined were as follows: peak systolic velocity (PSV), end diastolic velocity (EDV), Resistive Index (RI) and Pulsatility Index. The power Doppler mode was used for colour flow mapping of the prostatic artery. In PW mode at marginal locations, the waveforms recorded showed a continuous pattern typical of the high‐resistance vessels, while in subcapsular locations, the waveforms recorded were continuous characteristic of the low‐resistance vessel. Peak systolic velocity and EDV in both locations were significantly higher in BPH group (p < 0.001) than normal group (p < 0.05). Pixel number in BPH group in both locations was significantly higher (p < 0.05) compared to the normal group. This study shows that Doppler ultrasonography represents a valid and non‐invasive method for the characterization of the blood flow in the prostatic artery in dogs affected with BPH. Moreover, statistically significant differences of blood flow velocities in prostatic artery in normal and BPH‐affected dogs were detected. The RI was not able to differentiate normal dogs from dogs affected by BPH and therefore is not a parameter usable for diagnostic purposes, while Power Doppler could represent an additional diagnostic tool.  相似文献

The objective of this study was to compare the sedative and peripheral hemodynamic effects of acepromazine (ACP) and promethazine (PTZ) in the standing healthy horse. Nine healthy Warmblood horses randomly received either intravenous ACP at 0.1 mg/kg or PTZ at 0.1, 0.2, and 0.3 mg/kg. A sedation score based on clinical examination was recorded, and systolic arterial blood pressure was noninvasively evaluated using a Doppler flow detector at the tail, just before and every 15 minutes until 60 minutes after drug injection. Hemodynamics of the median artery of the left forelimb was studied using Doppler ultrasonography just before and 45 minutes after injection of the drug, which allowed calculation of surface (SURF), diameter (DIAM), and circumference (CIRC) of the vessel and peak systolic velocity (PSV), end diastolic velocity (EDV), mean velocity (MV), volumetric flow (VF), and resistivity index (RI) of the blood flow. Regardless of the dose used, PTZ had lesser sedative and hypotensive effects than ACP at 0.1 mg/kg and did not induce significant variations in SURF, DIAM, CIRC, PSV, EDV, MV, VF, and RI of the studied standing horses. Conversely, the vasodilatory properties of ACP were illustrated by a significant increase in SURF, DIAM, CIRC, PSV, EDV, MV, and VF and a significant reduction of the RI. Unlike ACP, PTZ did not induce alterations on the morphology of the Doppler waveform. PTZ appears to have less sedative and peripheral vasodilator effects than ACP, thus it could be safer than ACP in patients suffering from hypotension.  相似文献

Purpose (1) To determine the effect of the l ‐type calcium channel blocker amlodipine on color Doppler ultrasound‐determined vascular resistance and blood flow velocities in the distal retrobulbar arteries of dogs; (2) to determine any effect of blood pressure and PCO2 rate on such color Doppler‐determined circulatory measurements. Methods Color Doppler imaging measurements of the short posterior ciliary artery, long posterior ciliary artery, and ophthalmic artery of normal eyes of 10 dogs were obtained under isofluorane anesthesia before and 1 week after oral amlodipine administration. Mean systemic arterial blood pressure and PCO2 were monitored. Results The mean resistive index decreased significantly in the short posterior ciliary artery (P = 0.0347), in the long posterior ciliary artery (P = 0.0092), and ophthalmic artery (P = 0.0004) following systemic amlodipine administration. The end diastolic velocity increased significantly in the long posterior ciliary artery (P = 0.0368) and ophthalmic artery (P < 0.0001). The peak systolic velocity increased significantly in the ophthalmic artery (P = 0.0256). Mean systemic arterial blood pressure was significantly negatively associated with resistive index (P < 0.0001) and significantly associated with the log of the end diastolic velocity (P < 0.0001). Conclusions Systemically administered amlodipine increases color Doppler imaging‐determined blood flow velocity and decreases vascular resistive index in the ophthalmic artery, short posterior ciliary artery and long posterior ciliary artery of normal dogs. Changes in systemic arterial blood pressure can significantly affect the measurement of color Doppler imaging parameters.  相似文献

Objective  To validate intraocular pressure (IOP) readings obtained in cats with the TonoVet® tonometer. Animals studied  IOP readings obtained with the TonoVet® were compared to IOP readings determined by manometry and by the Tono‐Pen XL? in 1 normal cat and two glaucomatous cats. TonoVet® and Tono‐Pen XL? readings were also compared in a further six normal and nine glaucomatous cats. Procedures  The anterior chambers of both eyes of three anesthetized cats were cannulated and IOP was varied manometrically, first increasing from 5 to 70 mmHg in 5 mmHg increments, then decreasing from 70 to 10 mmHg in 10 mmHg decrements. At each point, two observers obtained three readings each from both eyes, with both the TonoVet® and Tono‐Pen XL?. IOP was measured weekly for 8 weeks with both tonometers in six normal and nine glaucomatous unsedated cats. Data were analyzed by linear regression. Comparisons between tonometers and observers were made by paired student t‐test. Results  The TonoVet® was significantly more accurate than the Tono‐Pen XL? (P = 0.001), correlating much more strongly with manometric IOP. In the clinical setting, the Tono‐Pen XL? underestimated IOP when compared with the TonoVet®. Conclusions  Both the TonoVet® and Tono‐Pen XL? provide reproducible IOP measurements in cats; however, the TonoVet® provides readings much closer to the true IOP than the Tono‐Pen XL?. The TonoVet® is superior in accuracy to the Tono‐Pen XL? for the detection of ocular hypertension and/or glaucoma in cats in a clinical setting.  相似文献

Objective To compare intraocular pressure (IOP) measurements made on healthy adult rabbits without the effect of tranquilizers using the new applanation tonometer, Tono‐Pen Avia®, and the rebound tonometer Tonovet®. Methods Intraocular pressure was measured throughout the day (6:00, 9:00, 12:00, 15:00, and 18:00 h) in 38 adult New Zealand White rabbits (76 eyes). The animals were 20 males and 18 females, with a mean weight of 3.5 kg and an average age of 6 months. A complete ocular exam (including Schirmer tear test, fluorescein staining, slit‐lamp biomicroscopy, and direct ophthalmoscopy) was performed on all animals at the beginning of the trial. Rebound tonometry was performed, and after 10 min, anesthetic drops were instilled and applanation tonometry was carried out. IOP values obtained using the two techniques were analyzed statistically. Results The mean IOP was 9.51 ± 2.62 mmHg with Tonovet®, and 15.44 ± 2.16 mmHg with the Tono‐Pen Avia®. Significant differences between measurements with the two tonometers were observed (P < 0.001). The linear regression equation describing the relationship between the two tonometers was y = 0.4923x + 10.754 (y = Tonovet® and x = Tono‐Pen Avia®). High IOPs were recorded in the early measurements (6:00), but the average IOPs from both devices were statistically similar throughout the day (P = 0.086). The correlation coefficient was r2 = 0.357. No significant difference in IOP regarding gender was observed. Conclusion The Tono‐Pen Avia® recorded higher levels of IOP compared with the Tonovet®. Early in the day, the IOP of rabbits was higher than later in the day, regardless of the tonometer used.  相似文献

The purpose of this study was to evaluate the effects of medetomidine administration on the Doppler variables of abdominal arteries. The study population consisted of 20 healthy dogs. The haemodynamic effects of the medetomidine were defined using Doppler variables of the abdominal aorta, renal arteries, cranial mesenteric artery and celiac artery. The dogs were monitored continuously and different measurements were performed before medetomidine injection, at 10, 40 and 80 minutes after medetomidine medication and after atipamezole administration. Changes in the characteristic Doppler spectra of different vessels were more marked in the abdominal aorta, in which a greater reverse flow was found. There was a significant decrease in peak systolic velocity (PSV), end diastolic velocity (EDV), and mean velocity (MV) at ten minutes in every vessel studied and this effect persisted until atipamezole administration. Pulsatility index (PI) increased significantly in the abdominal aorta at ten minutes and persisted during the study until atipamezole administration. Resistive index (RI) did not vary significantly in any vessel. A significant decrease was found in flow volume of the abdominal aorta, the cranial mesenteric artery and the celiac artery at ten minutes, persisting until atipamezole administration. We conclude that medetomidine can be a good sedative in aiding sonographic evaluation of RI in all the abdominal vessels studied. On the other hand, the changes in other Doppler variables suggest that medetomidine administration causes significant hemodynamic differences between sedated and non-sedated dogs.  相似文献

Insonation of Circle of Willis by transcranial Doppler duplex color sonography is described in 30 healthy dogs with 15 weighing <33 lb and 15 weighing >33 lb. Imaging was via a temporal window to explore the rostral, middle, and caudal cerebral arteries on both the left and right-hand sides; and through an suboccipital window to study the basilar artery. Normal mean values of the peak systolic velocity (PSV), end diastolic velocity, mean velocity, resistance index (RI), and pulsatility index (PI) were characterized and compared with those obtained in previous studies. There was significant differences in the PSV, RI, and PI in the rostral cerebral artery between dogs weighing <33 vs. >33 lb. Mean PSV was higher in weighing over 33 lb, whereas the mean resistive index and mean PI were lower in these dogs.  相似文献

Objectives To establish normal reference ranges of ocular parameters including phenol read thread, palpebral fissure length, horizontal and vertical corneal diameter, upright and hanging intraocular pressure (IOP) and to report ophthalmic examination findings of the anterior segment and lens, in a population of captive fruit bats. Animals studied Eyes of 30 bats of three species were included in this study: 10 (5 males, 5 females) Malayan Flying Foxes (Pteropus vampyrus), 10 (5 males, 5 females) Little Golden‐mantled Flying Foxes (Pteropus pumilus), and 10 (4 males, 6 females) Island Flying Foxes (Pteropus hypomelanus). Results The most common ophthalmic examination findings included iris‐iris persistent pupillary membranes (83%), nuclear sclerosis (56.7%), prominent arterial circle (40%), iridal hyperpigmented foci (30%), pupillary margin cysts (27%), and third eyelid defects (20%). The mean, among all species for: phenol red thread was 20.23 ± 1.28 mm/15 s both eyes (OU); palpebral fissure length was 13.34 ± 0.33 mm for OU; for horizontal corneal diameter was 10.72 ± 0.32 mm for OU; for vertical corneal diameter was 9.90 ± 0.30 mm for OU; for the hanging intraocular pressures was 19.38 ± 0.77 mmHg for OU; for upright IOP was 13.95 ± 0.60 mmHg for OU. Measurements for the individual species groups and eyes were also calculated. Conclusions Results revealed the IOP of bats in a hanging position were significantly higher than the IOP of bats in an upright position. The size of the bat, between the species, affected palpebral fissure length, horizontal corneal diameter, and vertical corneal diameter. Information about the ocular structures and normal ophthalmic parameters for the Pteropus species is crucial for species protection because of dependence on vision for survival.  相似文献

Objective The present study was undertaken to establish reference values for Schirmer tear test (STT) and intraocular pressure (IOP) in the long‐eared hedgehog (Hemiechinus auritus). Animals Fourteen healthy long‐eared hedgehogs (H. auritus) of either sex were studied. Procedures The hedgehogs were individually immobilized with an intramuscular injection of combined Ketamine (20 mg/kg) and Diazepam (0.5 mg/kg), and each animal underwent ophthalmic examinations including: STT, tonometry, biomicroscopy, and indirect ophthalmoscopy. Results No significant effects of animal gender, weight, side (right vs. left eye) were found in this study. Mean (SD) STT values for all eyes (n = 28) were 1.7 ± 1.2 mm/1 min with a range of 0–4 mm/1 min. Mean STT in male animals was 2.2 ± 1.2. Mean STT in female Hedgehogs was 1.3 ± 1.1. Mean (SD) IOP values by applanation tonometry were 20.1 ± 4.0 mmHg (range 11.5–26.5 mmHg). Mean (SD) IOP values by applanation tonometry were 18.2 ± 4.0 and 22.0 ± 3.2 mmHg for males and females, respectively. Conclusions This study reports STT and IOP findings in long‐eared hedgehogs (H. auritus).  相似文献

Objective

To determine the effect of a bimatoprost sustained‐release intracameral implant (Bimatoprost SR) on episcleral venous pressure (EVP) in normal dogs.

Methods

Normotensive beagle dogs were randomized to receive Bimatoprost SR 30 μg (n = 7) or sham injection (needle insertion only, n = 7) in one eye on day 1. EVP was measured with an episcleral venomanometer through day 65. Episcleral aqueous outflow vessels were identified using fluorescence imaging following intracameral injection of indocyanine green in one additional animal. A separate cohort of dogs that had been trained for conscious intraocular pressure (IOP) measurements received Bimatoprost SR 30 μg (n = 8) in one eye; IOP was evaluated through day 66.

Results

Baseline mean EVP was 10.0 mmHg in the Bimatoprost SR group and 10.4 mmHg in the sham group. Eyes treated with Bimatoprost SR exhibited a transient increase in mean EVP that peaked at day 8, followed by a decrease to levels below baseline. From day 29 to day 65, the change in mean EVP from baseline ranged from ?2.4 to ?3.9 mmHg (P < 0.05 vs. sham). Baseline mean IOP in eyes treated with Bimatoprost SR was 14.9 mmHg, and a steady IOP reduction was maintained through day 66. Bimatoprost SR‐treated eyes exhibited a selective, sustained dilation of aqueous outflow vessels that was not observed in sham‐treated eyes.

Conclusions

In normal dogs, Bimatoprost SR was associated with a transient increase in EVP followed by a sustained decrease. Changes in EVP were accompanied by a sustained dilation of aqueous outflow vessels.

Reasons for performing study: Only few drugs with limited efficacy are available for topical treatment of equine glaucoma. Objective: To evaluate the effect of topical administration of 1% brinzolamide on intraocular pressure (IOP) in clinically normal horses. Methods: Healthy mature horses (n = 20) with normal ocular findings, were studied. The IOP was measured 5 times daily (07.00, 11.00, 15.00, 19.00 and 23.00 h) over 10 days. On Days 1 and 2, baseline values were established. On Days 3–5 one eye of each horse was treated with one drop of 1% brinzolamide every 24 h immediately following the 07.00 h measurement. On Days 6–8 the same eye was treated with 1% brinzolamide every 12 h (07.00 and 19.00 h). Measurements on Days 9 and 10 documented the return of IOP to baseline values. Statistical analysis of the data was performed. Results: In the treated eye a significant decrease in IOP compared to baseline values was noted during both the 24 and 12 h dosing periods (P<0.001). During the once‐daily treatment protocol an IOP reduction of 3.1 ±1.3 mmHg (14%) from baseline was recorded. During the twice‐daily protocol a total IOP reduction of 5.0 ± 1.5 mmHg (21%) was achieved. Conclusion: Intraocular pressure was significantly decreased by 1% brinzolamide in a once‐daily and a twice‐daily treatment protocol in normotensive eyes. These findings suggest that brinzolamide might also be effective in horses with an elevated IOP. Potential relevance: This drug may be useful for treatment of equine glaucoma.  相似文献

Objective To evaluate effects of Coherin? on intraocular pressure (IOP), pupil size (PS), and heart rate (HR) in glaucomatous Beagles in single‐dose studies in a pilot study. Materials and methods Intraocular pressure, PS, and HR were measured in eight glaucomatous Beagles. One randomly chosen eye received single 50 μL doses of differing concentrations of Coherin? (treated eye) or vehicle (placebo‐treated eye), and the fellow eye served as the untreated control. After the first measurements, a single dose of either Coherin? or sterile water vehicle was instilled in the drug and placebo eyes, respectively. Results The mean ± SEM diurnal changes in IOP after 0.005%, 0.01%, 0.2%, 0.284%, 1%, 2%, and 4% topical Coherin? once daily were 7.6 ± 3.2 mmHg, 15.5 ± 5.3 mmHg, 11.2 ± 4.4 mmHg, 11.8 ± 4.4 mmHg, 19.1 ± 3.8 mmHg, 5.0 ± 1.8 mmHg, and 8.8 ± 2.8 mmHg, respectively. The declines in IOP were significantly different (P < 0.05) from the untreated control eyes with the 0.2% and 0.284% Coherin?‐treated eyes and suggestive for 1% Coherin? concentrations. No signs of irritation, significant PS, and HR changes were detected in the Coherin?‐treated eyes. Conclusion Of seven different concentrations, 2% and 0.248% Coherin? produced significant declines in IOP in the glaucomatous beagle in single‐dose studies when compared to both untreated control and placebo‐treated eyes. One percent Coherin? solution produced significant IOP decreases compared with the placebo‐treated eye but not the untreated control eyes. No local ocular irritation, PS and HR changes were observed in Coherin?‐treated eyes. This pilot study suggests that topical Coherin? has potential as an ocular hypotensive agent.  相似文献
