The present investigation was conducted to elucidate the influence of an opioid peptide, leucine-enkephalin (L-ENK), on the reproductive axis of the tilapia Oreochromis mossambicus. In the first experiment, administration (i.p.) of 25, 100, and 300 μg L-ENK to the stripped female tilapia, for a period of 22 days, resulted in a significantly higher number of stage I follicles compared to those of initial controls and experimental controls, whereas the mean number of stage II and III follicles and serum levels of E2 did not significantly differ among different experimental groups. A significant increase in the number of stage V (fully ripened) follicles was concomitant with significant reduction in the follicular diameter in 25 or 100 μg L-ENK-treated fish compared to those of experimental controls. However, significant reduction in the mean number and diameter of these follicles was observed in 300 μg L-ENK-treated fish compared to those of experimental controls and 25 or 100 μg L-ENK-treated fish. In the second experiment, the stimulatory effect of 25 μg L-ENK on the ovary was abolished in combination with gonadotropin-releasing hormone antagonist (GnRH-A). In conclusion, these results suggest that L-ENK exerts stimulatory as well as inhibitory effects on the ovary in a dose-dependent manner, and that these effects are possibly mediated through the GnRH, for the first time in fish.  相似文献

Bioaccumulation of mercury and histomorphological changes in the olfactory epithelium of Labeo rohita were investigated after exposing the fish to two sublethal concentrations of HgCl2 (66 and 132 μg/L) for 15 and 30 days. Mercury deposition increased in the tissue significantly (p < 0.05) with dose- and duration-dependent manner. Severe damage to the olfactory epithelium was evident. When fish exposed to 66 μg/L for 15 days, the histology of olfactory epithelium exhibited that mucous cell proliferation was upregulated and cell size was significantly increased from the control. Similar trends were found in 30 days exposure in both treated groups. Histology showed that mercury induced degeneration of columnar sensory cells, supporting cells and ciliated non-sensory cells and induced basal cell proliferation. Basal cell hyperplasia led to form intraepithelial proliferative lesion, thickening of epithelium, basal lamina disruption and cyst formation. Scanning electron microscopy revealed that mercury exposure at 66 μg/L caused clumping and loss of cilia, erosion in microridges on the supporting cells and proliferation of mucous cell opening. Complete degeneration of ciliated cells and cyst formation was observed in the fish when exposed to 132 μg/L HgCl2. This result suggests that prolonged exposure to mercury might cause irreversible damage to the olfactory epithelium and impair the olfactory function of fish.  相似文献

The effect of serotonin after intracerebroventricular (ICV) injections in dosage of 0.3 and 0.15 μg/g of the body weight on the thermoregulatory behavior in juvenile carp Cyprinus carpio L. and goldfish Carassius auratus (L.) has been studied in two experiments of 10- to 11-day duration. It has been found that ICV injection of serotonin in dosage of 0.3 μg/g caused the initial decrease in preferred temperatures (PT) (2–3 days) followed by their further increase (4–8 days after the administration) in carp. ICV injection of serotonin in dosage of 0.15 μg/g caused more durable initial decrease in PT (2–8 days) followed by the increase of once (9–11 days of observation) in goldfish. The values of the ultimate preferred temperature in fish of the experimental group exceeded that in control specimens by 4.1 °C at the first experiment and by 3.1 °C in the second experiment. The data for the first time demonstrated long-term effect of a single ICV injection of serotonin on the thermoregulatory behavior in two closely related species of teleosts.  相似文献

In the current study, laboratory evaluations were made to assess the immunomodulatory effect of cypermethrin on fingerlings of common carp (Cyprinus carpio L.). Results showed that 96-h LC50 of cypermethrin in common carp was estimated at 0.85 μg/L. Fish were exposed for 21 days to cypermethrin at three sub-lethal concentrations of 0.042, 0.085, and 0.17 μg/L that represented 5, 10, and 20%, respectively, of the 96-h LC50 of the pesticide for this fish species. Blood samples were taken after 7, 14, and 21 days of exposure. Immunological indices and resistance against bacterial infection were determined. Compared to the control group, the fish exposed to cypermethrin showed a significant increase in neutrophil ratio but exhibited a significant decrease in leukocyte number and lymphocyte ratio in treatments exposed to 0.17 and/or 0.085 μg/L after 21 days of exposure (p < 0.05). Serum protein level was significantly decreased in group exposed to 0.17 μg/L on day 14 and also in groups exposed to 0.085 and 0.17 μg/L on day 21 (p < 0.05). Immunoglobulin value was significantly reduced in groups exposed to 0.085 and 0.17 μg/L after 21 days of exposure (p < 0.05). Serum lysozyme activity and phagocytic activity were significantly decreased following exposure to 0.17 μg/L determined on days 14 and 21, post-exposure (p < 0.05). Mortality rate following the challenge with Aeromonas hydrophila significantly increased in fish exposed to 0.17 μg/L of cypermethrin. Overall, the present results indicate severe immunotoxicological effects of cypermethrin in common carp. Therefore, the use of cypermethrin in the proximities of common carp farms should be carefully considered.  相似文献

The size of a fish is an important factor in its physiology, and metal uptake is affected by animal physiology. In this study, small and large tilapias (Oreochromis niloticus) differing approximately twofold in length and fivefold in weight were compared for their antioxidant response. Both groups were exposed to Cu or Cr (1.0 μg/mL) in a freshwater (?80 mg CaCO3/L, conductivity 1.77 mS/cm) using 2 exposure protocols (20 μM for 48 h and 10 μM for 6 days). Following the exposures, the antioxidant enzyme activities (superoxide dismutase, SOD; catalase, CAT; glutathione peroxidase, GPX; glutathione reductase, GR and glutathione S-transferase, GST) and glutathione (GSH) levels were measured in the liver of fish. Results showed that small fish was affected from exposure conditions much more than large ones as their antioxidant parameters significantly decreased even in controls. Metal exposures of small fish caused significant increases in SOD and CAT activity in acute Cu or Cr exposures. Subchronic Cr exposure of small fish also caused significant increases in CAT, GPx and GST activities, while there was no significant change in Cu-exposed ones. Large fish, however, showed different antioxidant responses as their levels mostly decreased. This study demonstrated that the response of antioxidant system in the liver of tilapia varied in relation to fish sizes and emphasized using different size groups in environmental monitoring and also in evaluation of fish biomarkers.  相似文献

Farmed fish are exposed to risks from feed-borne chemical contamination, such as leucomalachite green (LMG) in fish meal. Consequently, the use of malachite green is prohibited in aquacultural practice. An improved understanding of the toxicity of dietary LMG provided to farmed fish is needed in order to manage risk. Oreochromis niloticus specimens were fed experimental diets containing 0, 100, 500, and 2500 μg/kg LMG for 28 days. On sampling days 7, 14, and 28, fish in the exposed groups had detectable levels of LMG. Accumulation levels were approximately 12.2 % (in the liver) and 6.2 % (in the muscle) of the LMG concentration in the feed (104, 510, and 2200 μg/kg). Hematocrit and mean corpuscular hemoglobin concentrations in the 500 and 2500 μg/kg groups were significantly elevated at 7 and 14 days. Hemoglobin in the group that received the highest dose was significantly higher than that in the control group. Significant increases in the activities of alanine aminotransferase and alkaline phosphatase were also detected in the group receiving the highest dose. Total cholesterol concentrations in all of the exposed groups were significantly higher than those in the control group. These observations of toxicity were dose dependent. Histological changes in gills and livers were observed in LMG-exposed fish.  相似文献

The present study was conducted to evaluate the effects of calcium (Ca) and copper (Cu) exposure on growth performance, metal element composition, enzymatic activities and histology in Synechogobius hasta and tested the hypothesis that Ca could protect against Cu-induced toxicity in the fish species. Three hundred sixty S. hasta (initial mean weight 22.6 ± 0.2 g, mean ± SEM) were stocked in 18 circular fiberglass tanks (water volume: 300 l), 9 of which were pre-exposed to Ca at a rate of 600 mg Ca/l for 10 days and then exposed to concentrations of 0, 77 and 154 μg Cu/l for 30 days. Another 9 tanks were cultured in natural seawater (no extra Ca addition) for 10 days and then exposed to concentrations of 0, 77 and 154 μg Cu/l for 30 days. Both Ca exposure and then waterborne Cu exposure influenced the accumulation of metal elements (Cu, iron, zinc and manganese) in several tissues (muscle, intestine, gill, liver and spleen), changed hepatic lipogenic metabolism and induced histological alterations in liver, gill and spleen. In general, Ca exposure seemed to mitigate the severity of Cu-induced mortality and histopathological injuries, indicating that Ca exposure had the capacity to decrease Cu toxicity in S. hasta. To our knowledge, it is the first study involved in the effects of Ca and Cu exposure on hepatic lipogenic metabolism and metal element compositions in a wide range of tissues and organs in fish, which provides new insight into the protective effect of Ca on toxic metal elements in fish.  相似文献

The need to expand aquaculture production has led to other fish to be considered as potential species for culture, such as the sub-Antarctic notothenioid Eleginops maclovinus (Valenciennes, 1830). The aim of this study was to determine the cumulative effect of density and pathogen infection by protein extract of Piscirickettsia salmonis on skeletal muscle metabolism. In a first experiment, specimens were submitted to three different stocking densities: (1) 3.1 kg m?3, (2) 15 kg m?3 and (3) 60 kg m?3, for a period of 10 days. In a second experiment, metabolic changes caused by an infection of P. salmonis protein extract (a single injection of 0.5 μL P. salmonis protein extract g body weight?1 was inoculated in the fish) and its combined effect with stocking density was assessed during a period of 10 days. This study concludes that stress caused by high stocking density led to the reorganization of some metabolic routes to fulfill skeletal muscle energy needs. Furthermore, infection response by pathogen P. salmonis differed when stocking density increased, suggesting an increase of energy needs with density in skeletal muscle of infected fish.  相似文献

We studied accumulation and biochemical effects of microcystin-LR (MCLR) in Odontesthes hatcheri after dietary administration of the cyanobacteria Microcystis aeruginosa (1.3 μg MCLR/g body mass, incorporated in standard fish food). After 12 h, MCLR content in liver did not differ between fish fed with crushed or intact cells, demonstrating O. hatcheri’s capacity to digest cyanobacteria and absorb MCLR. In the second experiment, fish received toxic cells, non-toxic cells, or control food; MCLR accumulation was monitored for 48 h. Protein phosphatase 1 (PP1), catalase (CAT), glutathione-S-transferase (GST) activities, and lipid peroxidation (as MDA) were measured in liver and intestine. Methanol-extractable MCLR was determined by PP1 inhibition assay (PPIA); extractable and protein-bound MCLR were measured by Lemieux oxidation-gas chromatography/mass spectrometry (GC/MS). MCLR accumulated rapidly up to 22.9 and 9.4 μg MCLR/g in intestine and liver, respectively, followed by a decreasing tendency. Protein-bound MCLR represented 66 to ca. 100 % of total MCLR in both tissues. PP1 activity remained unchanged in intestine but was increased in liver of MCLR treated fish.CAT and GST activities and MDA content were significantly increased by MCLR only in liver. We conclude that O. hatcheri is able to digest cyanobacteria, accumulating MCLR mostly bound to proteins. Our data suggest that this freshwater fish can be adversely affected by cyanobacterial blooms. However, the rapid decrease of the detectable MCLR in both tissues could imply that sublethal toxin accumulation is rapidly reversed.  相似文献

Triplicate groups of one hundred Tra catfish (8 g?±?0.2) were fed seven test diets containing increasing levels of AFB1 (0, 50, 100, 250, 500, and 1000 μg AFB1 kg?1). Additionally Mycofix® Secure was added at 1.5% to one diet containing 500 μg AFB1 kg?1. Results showed that Tra catfish are sensitive to AFB1. Reduction in weight gain (P?<?0.05) was observed for fish fed 50 μg AFB1 kg?1 and declined further with increasing levels of AFB1 in the diets. Fish fed diets contaminated with 500 and 1000 μg AFB1 kg?1 showed increased (P?>?0.05) hepatosomatic index (HIS), while an increase in adipose somatic index (ASI) was observed in fish fed 50 μg AFB1 kg?1 and above when compared to the control and Mycofix® diets. After 12 weeks, blood serum analysis revealed higher alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels in fish fed the 50, 100, and 250 μg AFB1 kg?1 suggesting occurrence of liver damage. Disease resistance of fish exposed to Edwardsiella ictaluri was also compromised by the presence of AFB1 in the feed and was directly related to the contamination level. Seven days after Edwardsiella ictaluri exposure, survival rates were 50, 41.7, 31.7, and 8.3% for fish fed control, 50, 100, and 250 μg AFB1 kg?1, respectively. This trial shows that AFB1 at a level of 50 μg AFB1 kg?1 and above can affect fish performance and disease resistance. Application of an effective mycotoxin management in the feed seems to be useful to prevent the negative effects of AFB1.  相似文献

In vitro accumulation of tetrodotoxin (TTX) and paralytic shellfish toxin (PST) in tiger puffer fish Takifugu rubripes was investigated using liver tissue slices. When T. rubripes liver slices were incubated with Leibovitz’s L-15 medium containing 0.13 mM TTX at 20 °C in air with saturated humidity, they accumulated 21.5 ± 7.3 μg TTX g?1 liver after the incubation for 12 h and increased to 55.3 ± 8.2 μg TTX g?1 liver at 48 h. In the incubation of T. rubripes liver slices with 0.13 mM PST-containing medium, PST was detected 6.3 ± 0.9 μg g?1 liver at 12 h and reached a plateau thereafter. These results reveal the difference between TTX and PST in accumulation in T. rubripes liver tissue slices. To examine the variation in PST accumulation among fish species, the liver tissue slices from tiger puffer fish T. rubripes, parrot-bass Oplegnathus fasciatus and green ling Hexagrammos otakii were incubated at a concentration of 0.027 mM PST. The toxin contents of 3.0 μg g?1 liver were observed at 8 h regardless of fish species but were not increased subsequently, showing no variety among these three species as to accumulation patterns of PST. It is noted that the tiger puffer fish T. rubripes liver specifically accumulate TTX in preference to PST.  相似文献

Sesamin (S) is a known lipid modulator and has been shown to increase the conversion of α-linolenic acid (ALA) to docosahexaenoic acid in rainbow trout (Oncorhynchus mykiss) fed vegetable oil mixtures including linseed oil. In this study, we evaluated the effects of S supplementation in linseed oil-based diets, content of α- and γ-tocopherols, fatty acid (FA) composition, as well as the gene expression of lipid-related genes. Fish with an average weight of 36.5 g were fed different combinations of commercial linseed oil (LO), purified linseed oil triacylglycerols (TAG) with polar fraction removed and a mixed linseed-sunflower oil (6:4 v/v) (MO). S was added at 0.58 g 100?1g feed and fed to the fish for a period of 58 days. Expression of PPARα was downregulated in white muscle of fish fed S containing diets (P < 0.05). The expression of PPARβ1A was not affected by S supplementation except where TAG oil was used. The expression of PPARβ1A declined significantly in TAG + S fed group (P < 0.05), which indicates that some minor compounds in linseed oil might suppress the effect of S on the expression of PPARβ1A. The expression of PPARγ(long) declined in LO + S and MO + S fed group (P < 0.05). The β-oxidation-related genes CPT1 and ACO were upregulated by vegetable oils compared to fish oil. S decreased percentage of ALA in white muscle of fish fed LO + S (P < 0.05). The increased desaturation index and the decreased ALA levels suggest that S may increase the biosynthesis of highly unsaturated FA in rainbow trout.  相似文献

The aim of the study was to investigate the effects of the diets that contain safflower oil and fish oil as lipid sources, on growth, feed conversion and body composition in turbot (Psetta maxima). Two iso-nitrogenous and iso-lipidic diets (55 % protein and 14 % lipid) were prepared that include 100 % fish oil (FO group) and 100 % safflower oil (SFO group) for turbots with average weight of 62.21 ± 1.28 g, and fish were fed with these diets for 104 days. At the end of the experiment, the weight gain, specific growth rate and feed conversion ratio were the highest in SFO group than in FO group (p < 0.05). Fatty acid composition of fish body reflected the fatty acid composition determined in the experimental diets. The amounts of palmitic acid (PA; C16:0), oleic acid (OA; C18:1n-9), linoleic acid (LA; C18:2n-6) and docosahexaenoic acid (DHA; C22:6n-3) were dominant fatty acids in fish body. It was confirmed that the usage of safflower oil instead of fish oil in turbot feed did not generate any negative effects on growth, feed conversion and the values regarding the growth performance.  相似文献

The aim of the study was to assess the effects of a cyanobacterial extract containing microcystins (MCs) on selected hematological and biochemical parameters in common carp (Cyprinus carpio L.), as well as to determine the accumulation of toxins in fish tissues. The fish were immersed for 5 days in water containing toxins at a final concentration of 12 μg/L of microcystin LR equivalent. Microcystin LR residues were detected in fish liver, reaching 207, 238 and 260 ng/g f.w. of the tissues taken 24 h, 72 h and 5 days after the end of intoxication, respectively. The most substantial changes were found in fish plasma, including increases in creatine kinase, lactate dehydrogenase, ammonia, glucose, aspartate aminotransferase and alanine aminotransferase levels. A decline of about 50% in lysozyme activity was observed by the end of the experimental period. Moreover, a marked increase in ceruloplasmin activity was detected 24 h after the end of intoxication with a subsequent decrease in its activity after 72 h and 5 days. This study concludes that not only consumption of food containing toxins but also MCs dissolved in water may pose a threat to fish health. Additionally, detected changes in lysozyme and ceruloplasmin activity may have distinct effects in fish resistance against pathogens or oxidative stress, which should be taken into account in the future studies.  相似文献

Little is known about the effects of brominated flame retardants in teleosts and some of the information currently available is inconsistent. This study examined effects of dietary exposure to 2,2′,4,4′-tetrabromodiphenyl ether (BDE-47) on thyroid condition, body mass and size, and gonadal development of zebrafish. Pubertal, 49-day-old (posthatch) fish were fed diets without BDE-47 (control) or with 1, 5 or 25 μg/g BDE-47/diet. Treatments were conducted in triplicate 30-L tanks each containing 50 zebrafish, and 15 fish per treatment (5 per tank) were sampled at days 40, 80 and 120 of exposure. Measurements were taken of body mass, standard length, head depth and head length. Sex (at 40–120 days of exposure), germ cell stage (at 40 days) and thyroid condition (at 120 days; follicular cell height, colloid depletion, angiogenesis) were histologically determined. Whole-body BDE-47 levels at study completion were within the high end of levels reported in environmentally exposed (wild) fishes. Analysis of variance was used to determine differences among treatments at each sampling time. No effects were observed on thyroid condition or germ cell stage in either sex. Reduced head length was observed in females exposed to BDE-47 at 80 days but not at 40 or 120 days. In males, no apparent effects of BDE-47 were observed at 40 and 80 days, but fish exposed to 25 μg/g had lower body mass at 120 days compared to control fish. These observations suggest that BDE-47 at environmentally relevant whole-body concentrations does not affect thyroid condition or pubertal development of zebrafish but does affect growth during the juvenile-to-adult transition, especially in males.  相似文献

The present study investigates the effects of different doses (0, 40, 60, 100 μg/L) of 4-nonylphenol (NP) on testis histology and sperms motility of mature Oreochromis niloticus. A tendency for a dose-dependent reduction in the gonado-somatic index (GSI) in the NP-exposed groups was observed. Histological examination revealed that high doses of NP (60 and 100 μg/L) cause hyperplasia of interlobular connective tissue components coincide with decrease in the size of seminiferous lobules and amount of lumen spermatozoa in comparison with the control and low-dose (40 μg/L)-exposed groups. In addition, within the seminiferous lobules, rupture of spermatogenic cysts, vacuolation and necrosis of primary spermatocytes due to damage of sertoli cells were clearly observed and lead to decrease in advanced spermatogenic cells. The highest dose caused significant decrease in the GSI associated with appearance of testis–ova (intersex) state. The percentage of abnormal sperms increased with increasing the dose of NP reaching to 96 % for those subjected to 100 μg NP/L, while it was only 11 % for the control group specimens. The study indicates that NP has estrogenic potency induced marked alteration in the sexual development including inhibition in testicular growth and deformation in the sperms.  相似文献

Eight isonitrogenous (approximately 35 % crude protein) and isocaloric (17.58 kJ g?1) diets incorporating raw and fermented sesame oilseed meal replacing other feed ingredients including fishmeal at 10, 20, 30 and 40 % levels by weight into a fishmeal-based reference diet (RD) were fed to rohu, Labeo rohita, fingerlings (mean initial weight 3.19 ± 0.09 g) for 80 days. Two phytase-producing bacterial strains (LF1 and LH1 of Bacillus licheniformis) isolated from the foregut and hindgut regions of adult L. rohita were used for fermentation of oilseed meal for 15 days at 37 ± 2 °C. Fermentation of sesame seed meal was effective in significantly reducing the crude fibre content and anti-nutritional factors such as tannins and phytic acid and enhancing available free amino acids, free fatty acids and mineral concentration. In terms of growth, feed conversion ratio and protein efficiency ratio, 30 % fermented oilseed meal incorporated diet resulted in a significantly (P < 0.05) better performance of rohu fingerlings. In general, growth and feed utilization efficiencies of diets containing fermented oilseed meal were superior to diets containing raw meal. The apparent digestibility of protein, lipid, ash and minerals (phosphorus, calcium, manganese, copper and iron) was significantly (P < 0.05) higher in fish fed diet containing 30 % fermented oilseed meal in comparison with those fed RD. The maximum deposition of protein in the carcass was recorded in fish fed the diet containing 30 % fermented seed meal.  相似文献

It was previously reported that the oral administration of lactoferrin (LF) provides antimicrobial activity in animals against bacterial and viral infections and is involved in immunomodulatory properties. In this report, a hybrid strain of Oreochromis nilotica (male) × O. mossambicus (female) was fed homemade diets with or without supplementation with 10, 50, 100, and 150 mg bovine LF g?1 feed for 60 days. After dietary treatment, the experimental fish were infected with Streptococcus agalactiae. LF supplementation resulted in a significantly higher survival rate and suppression of bacterial growth at 24–96 h in the liver, spleen, kidneys, eyes, and gills. A reduction in peroxidase activity was followed by a similar reduction in the peroxidase content of leukocytes at 24 h as analyzed by spectrophotometry. Respiratory burst (RB) activity was detected regardless of the time at which LF was administered to fish in relation to the bacterial infection. A beneficial effect of LF on RBs was also detected before bacterial challenge (with 100 mg LF/g) and 24 h after bacterial challenge with 150 mg/g. The results obtained validate LF’s beneficial effects on RBs by phagocytes in tilapia, which was only shown at 48 h after supplementation with 100 mg bovine LF g?1 feed after the bacterial infection. Moreover, after oral administration of LF, it was detected in the mucosa of the small intestines of tilapia. In conclusion, we demonstrate a reduction in fish mortality after the oral administration of LF, and we examined its immunomodulatory properties in tilapia.  相似文献

Vitamin E, a potent antioxidant consisting of four isomers each (α, β, γ, δ) of tocopherol (T) and tocotrienol (T3), is found naturally in plant oils at different concentrations. In this study, four semi-purified isonitrogenous and isolipidic (10 %) diets containing canola oil, cold-pressed soybean oil, wheat germ oil, or palm fatty acid distillates (PFAD) as the sole vitamin E source were fed to triplicate groups of red hybrid tilapia (Oreochromis sp.) fingerlings (14.82 ± 0.05 g) for 45 days. Vitamin E concentrations and composition were measured in the muscle, liver, skin, and adipose tissue. Deposition of α-T (53.4–93.1 % of total vitamin E) predominated over deposition of other isomers, except in the liver of fish fed the SBO diet, where α-T and γ-T deposition was in the ratio 40:60. T3 deposition (2.6–29.4 %) was only detected in tissues of fish fed the PFAD diet; adipose tissue was the major storage depot. Fish fed the SBO diet contained significantly more (P < 0.05) muscle thiobarbituric acid-reactive substances. Muscle fatty acid composition reflected dietary fatty acid profile. This is the first study to compare the deposition in fish tissues of the naturally occurring vitamin E isomers present in plant oils. The type and concentration of endogenous vitamin E and the fatty acid composition of plant oils can affect the oxidative stability of tilapia tissues.  相似文献
