[目的]证实从蜡梅花基因组提取DNA等同于从叶基因组提取DNA。[方法]采用CTAB法,对保康野生蜡梅花与叶基因组进行遗传多样性研究。[结果]在用CTAB法提取DNA时,65℃水浴振荡并二次抽提后花基因组DNA在浓度和OD260/OD280上都与叶基因组DNA达到相近的水平。[结论]从性状表现更显著的蜡梅花基因组提取DNA进行遗传性分析能达到叶基因组的水平。 相似文献

在分析现有提取微生物基因组DNA的原理和方法基础上,对SDS-NaCl法进行了改良,以商品化微生物基因组提取试剂盒提取的基因组结果为对照,利用改良的SDS-NaCl法,分别提取大肠杆菌(Escherichia coli)、枯草芽孢杆菌(Bacillus subtilis)和金黄色葡萄球菌(Staphylococcus aureus)的基因组DNA.结果表明,改良的SDS-NaCl法提取的大肠杆菌、枯草芽孢杆菌和金黄色葡萄球菌基因组DNA的OD260/OD280分别为1.88、1.89、1.81,浓度分别为61.67、64.32、53.22μg/mL;而商品化试剂盒提取大肠杆菌、枯草芽孢杆菌和金黄色葡萄球菌基因组DNA OD260/OD280分别为1.85、1.87、1.83,浓度分别为64.07、58.43、52.34μg/mL.结果证明改良SDS-NaCl法提取的革兰氏阳性和阴性细菌的基因组DNA可用于后续试验如全基因组测序和PCR等,同时可快速获得大量的高质量微生物基因组. 相似文献

[目的]探讨四倍体菘蓝组培苗高质量DNA的提取方法。[方法]分别采用CTAB法1、CTAB法2、SDS法1、SDS法2和碱裂解法对四倍体菘蓝组培苗DNA进行提取试验,通过紫外分光光度法和琼脂糖凝胶电泳,比较了5种DNA提取方法对四倍体菘蓝组培苗的提取效果。[结果]CTAB法2提取DNA的OD260/OD230值为2.311,OD260/OD280值为1.774,DNA纯度最大,浓度最大,提取率最高,PCR扩增条带清晰,重复性好,所得DNA的质量最好。[结论]CTAB法2是提取四倍体菘蓝组培苗基因组DNA最有效的方法。 相似文献

川续断叶片中很有较多的蛋白质、多糖、酚类以及其他一些次生代谢物,采用传统的CTAB法很难得到高质量的DNA。试验以川续断叶片为材料,提出一种改良的CTAB法提取叶片基因组DNA,对提取的基因组DNA进行琼脂糖电泳检测及紫外分光光度计分析。利用中药材条形码ITS2序列的引物对提取的基因组DNA进行PCR验证。结果表明,改良的CTAB法得到的DNA质量较好,其OD260/OD280的值介于1.8~2.0之间,ITS2序列的引物PCR扩增成功率达到100%。改良的CTAB法为较为理想的川续断基因组DNA提取方法,获得的总DNA能够满足下游的PCR试验要求。 相似文献

4种经优化的DNA提取方法即CTAB区室法、SDS区室法、CTAB常规法、SDS常规法提取猕猴桃嫩叶基因组DNA,效果均好。经综合对比分析,本实验认为CTAB常规法是适于AFLP分析最佳的提取方法,因其步骤简单、快捷,得到的基因组DNA降解少、杂质含量少,能获得高质量的DNA模板。通过对所得到的基因组DNA模板进行AFLP分析,得到了清晰的DNA指纹图谱。 相似文献

[目的]探索一种用于RAPD分析的羌活基因组DNA提取方法。[方法]分别采取CTAB法S、DS法和改良的CTAB法提取羌活基因组DNA,通过紫外分光光度法、琼脂糖凝胶电泳和RAPD分析对提取的DNA进行检测。[结果]3种方法均能有效地从羌活中获得较高产量的DNA,以改良的CTAB法所得的DNA纯度最高,此法提取的羌活基因组DNAOD260/OD280为1.8~2.0,DNA干重得率约为0.235μg/mg。[结论]改良的CTAB法更适于羌活基因组DNA的提取,可以完全满足RAPD扩增的需要。 相似文献

[Objective] The study aimed to introduea a rapid and effective method that is suitable for extracting genomic DNA from animal and plant. [Method] The genomic DNAs were extracted from tender leaves of 24 peanut cuhivars and from the liver, lung and kidney of white mouse through the spe-cifically modified CTAB method. The DNAs were run on agarose gel, next detected by DNA/Protein analyzer. Finally PCR amplification was conducted to detect the quality of DNAs extracted using the modified CTAB method. [Result] The clear and orderly bands were observed in gel detection, and the val-ues of OD260/OD280 for DNAs extracted via modified CTAB method were between 1.77 - I. 83. [目的]比较雷公藤叶片中基因组DNA的提取方法。[方法]采用常规CTAB法、改良CTAB法、常规SDS法、高盐低pH值法和一步法分别提取雷公藤叶片基因组DNA,利用琼脂糖凝胶电泳、紫外分光光度法测定样品OD260与OD280的比值以及RAPD扩增结果,判断所得DNA样品的纯度,并根据OD260值计算不同提取方法的DNA得率。[结果]改良CTAB法提取效果最佳,所提DNA的平均得率为219.3μg/g,OD260/OD280比值在1.869~1.903,用于RAPD-PCR均有较好的扩增结果。[结论]改良CTAB法是雷公藤叶片基因组DNA提取的最佳方法。 相似文献
