The immunohistolocalization of carbonic anhydrase isozymes (CA-I, II, III) in canine salivary glands was studied using antiserum against CA-I, II, III. In parotid glands, immunostaining intensely localized cytosolic CA-II antiserum throughout the cytoplasm of acinar secretory cells and ductal epithelial cells, especially in the striated duct region. CA-III reactivity in the glands was only seen selectively at the intercalated ductal cells. In contrast, no immunoreaction localized CA-I in the gland. In the submandibular and sublingual glands, CA-I, II, and III were all observed in the ductal segments of the glands, whereas serous demilune appeared devoid of all three cytosolic CA isozymes. In contrast, in zygomatic glands (i.e. dorsal buccal glands) all CA isozymes were observed in both serous demilune and ductal segments. In all of the salivary glands examined, no mucous acinar cells were found to be reactive for any CA.  相似文献

The glycoconjugate content of major horse salivary glands was investigated by means of horseradish peroxidase-conjugated lectins. Qualitative differences were observed in the terminal sugar residues of secretory glycoproteins and glycoconjugates linked to the apical surface of excretory duct epithelial cells. Mucous acinar cells in mandibular and sublingual glands contained oligosaccharides with D-galactose, α- and β-N-acetylgalactosamine, N-acetylglucosamine and fucose residues, whereas mandibular, sublingual and parotid serous cells contained only oligosaccharides with terminal α- and β-N-acetylgalactosamine residues. The apical portion of striated and interlobular duct lining cells of mandibular and sublingual glands stained for α- and β-N-acetylgalactosamine and for N-acetylglucosamine. In parotid gland the cytoplasm of intercalated duct cells and the apical surface of striated duct epithelial cells stained for α-N-acetylgalactosamine.  相似文献

采用免疫组织化学方法对不同日龄的长爪沙鼠颌下腺IgA的定位分布进行了研究。结果表明,长爪沙鼠的颌下腺由导管部和分泌部构成,分泌部主要由浆液腺构成,导管部包括闰管、纹管、颗粒曲管和小叶间导管等。DAB显色结果表明,IgA阳性细胞主要分布于浆液性腺泡、闰管、纹管、颗粒曲管和小叶间导管,并可分布于腺泡和腺管间结缔组织,IgA阳性产物的分布具有不均一性,无明显随年龄变化的规律性。阳性产物分布于胞质中,胞核呈阴性,对照组阴性。提示从浆细胞产生或循环而来的IgA先经结缔组织进入颌下腺组织,进而定位分布于浆液腺泡和各级导管,导管部有较多的IgA分布。  相似文献

The morphology of the pancreatic duct system did not receive much attention as compared to the microanatomy of the exocrine and endocrine pancreas. The histological peculiarities of the excretory duct system are of major importance especially in laboratory animals like guinea pigs. The paper describes the histological peculiarities of the major interlobular and extrapancreatic ducts in guinea pigs. The pancreatic tissue samples were collected from five guinea pigs. For histological investigation, several pancreatic fragments underwent fixation in 10% buffered formalin and were later processed by the standard paraffin technique. Subsequentially, tissue sections were stained by Goldner's trichrome staining. The mucous substances were assessed by Alcian blue and Periodic acid–Schiff staining methods. The interlobular and main pancreatic ducts in the guinea pig present a simple columnar epithelium surrounded by a thick layer of dense connective tissue. The aforementioned epithelium of the main pancreatic ducts includes principal cells, goblet cells and basal cells. Additionally, the ductal epithelium presents occasional unicellular multiloculated intraepithelial mucous glands and prominent extraepithelial glands. The latter adopts a simple or compound tubular feature. The mucus elaborated by the three glandular types is mostly neutral in goblet cells, predominantly acidic in extraepithelial ductal glands, and a similar amount of acidic and neutral mucin in intraepithelial glands. In conclusion, the epithelium-associated mucous glands in the interlobular and main pancreatic ducts in the guinea pig are restricted not only to goblet cells. A substantial mucous discharge probably with a protecting role against irritative pancreatic juice derives from the main ductal intraepithelial and extraepithelial glands.  相似文献

The present paper described the immunohistochemical distributions of carbonic anhydrase (CA) isozymes. CA-I, CA-II and CA-III, in the epithelium lining the rat gastrointestinal tract, with rabbit antibodies to equine CA-I, CA-II and CA-III. Prior to the immunohistochemical examinations, the crossreactivities of these antibodies to the rat-antigens were confirmed in this study. In the stomach, surface epithelial cells and parietal cells of the glandular region showed an immunoreactivity only to CA-II. In the large intestine, each immunoreactivity to CA isozyme (CA-I, CA-II and CA-III) was localized in the upper portion of intestinal glands, and decreased toward the distal digestive tract, but absent in the small intestine. The present histological findings suggested that the CA isozymes might play a role in the ion-transportation during the water absorption in the rat large intestine.  相似文献

A study was made of the Harderian and lachrymal glands of fowls, pheasants, turkeys and pigeons. Accumulation of heterophils and plasma cells commenced soon after hatching, was well advanced by four weeks of age and was always greater in the Harderian than in the lachrymal glands. Plasmablasts entered via interlobular capillaries, maturing as they migrated to and congregated round the lobular collecting ducts. Their secreted immunoglobulin was readily demonstrated in the duct lumen. Russell body containing cells of diverse morphology were a consistent feature of infiltrated Harderian glands. Though scant in the glands proper, lymphoid tissue was abundant in the walls of their draining ducts.  相似文献

The immunolocalization of the efferent duct and the epididymis in canine was firstly examined using an the immunohistochemical method with the canine carbonic anhydrase (CA) -I, CA-II and CA-III antisera. The efferent duct was immunonegative for all present canine CA antisera. However, some slender shaped epithelial cells in the head and body segments of the epididymal duct were intensely reacted to the CA-II antiserum. These results suggested that the CA-II might be controlled in the luminal environment in the head and body segments of the canine epididymis by the proton and bicarbonate balance for the maintenance of the spermatozoal stability and movement.  相似文献

Research on the disease called shimao zheng, which had resulted in widespread fleece-eating and shedding by sheep and goats in the Haizi area of Akesa County of Gansu Province of China, was carried out by both laboratory studies and pathological observations. There was a marked deficiency of sulfur in the wool and tissues of the affected animals. The pathological changes were mainly loss of fleece, exposure of the skin, keratinization of the epidermal cells, narrowing and reduced numbers of the fleece follicles, and fewer sebaceous glands and sweat glands. Histologically, there was atrophy of the striated and cardiac muscles and of the epithelium of the convoluted tubules of the kidney, together with a glomerulonephritis. The disease was considered to be a local nutritional and metabolic disorder, probably mainly caused by sulfur deficiency.  相似文献

The purpose of this study was to determine the expression and distribution of band 3 in the collecting duct and connecting tubules of the kidney of the marmoset monkey (Callithrix jacchus), and to establish whether band 3 is expressed in type A intercalated cells. The intracellular localization of band 3 in the different populations of intercalated cells was determined by double-labeling immunohistochemistry. Immunohistochemical microscopy demonstrated that band 3 is located in the basolateral plasma membranes of all type A intercalated cells in the connecting tubule (CNT), cortical collecting duct (CCD), and outer medullary collecting duct (OMCD) of the marmoset. However, type B intercalated cells and non-A/non-B intercalated cells did not show band 3 labeling. Electron microscopy of the CNT, CCD and OMCD confirmed the light microscopic observation of the basolateral plasma membrane staining for band 3 in a subpopulation of interacted cells. Basolateral staining was seen on the plasma membrane and small coated vesicles in the perinuclear structure, some of which were located in the Golgi region. In addition, there was no labeling of band 3 in the mitochondria of the CNT, CCD and in OMCD cells. The intensity of the immunostaining of the basolateral membrane was less in the CNT than in the CCD and OMCD. In contrast, band 3 immunoreactivity was greater in the intracellular vesicles of the CNT. From these results, we suggest that the basolateral Cl-/HCO3- exchanger in the monkey kidney is in a more active state in the collecting duct than in the CNT.  相似文献

The intrarenal distribution of renin in the cat kidney was evaluated in a semi-quantitative immunocytochemical study using a cross-reacting antiserum directed against purified renin. In addition, the morphology of the juxtaglomerular apparatus was studied electron-microscopically. The majority of renin in the cat kidney was located in the distal portion of the afferent arteriole. The values for the juxtaglomerular index and the renin-positive portion of the afferent arteriole were 30.6 +/- 3.2% and 49.9 +/- 8.3 microns, respectively. In addition, scattered renin-positive cells were found far from the vascular pole and even in the interlobular artery. Also, some vasa efferentia contained renin-positive cells. In 70% of the cats studied, renin-positive cells were located in the glomerular mesangium. The majority of the renin-positive mesangial cells were located in the vicinity of the glomerular stalk, but in some cases the centrilobular mesangium also contained renin-positive cells.  相似文献

The location of carbonic anhydrase III (CA-III) in frozen sections of biopsies of Thoroughbred horse skeletal muscle was studied. Fibre types were determined by ATP-ase and succinate dehydrogenase staining. CA-III isozyme was detected using a peroxidase conjugated anti-CA-III antibody. CA-III was found to be localised in slow twitch oxidative fibres (ST), but was also present in fast twitch oxidative (FTH) fibres in small amounts. Fast twitch glycolytic (FT) fibres were stained lightly compared with control sections. The concentrations of CA-III in muscle and liver were 70 micrograms/mg protein and 4 micrograms/mg protein, respectively. CA-I and CA-II were not found in muscle extracts by the double immunodiffusion method.  相似文献

Parotid glands from adult dogs were stained with a battery of seven horseradish peroxidase-conjugated lectins (PNA, UEA, LTA, DBA, SBA, WGA and ConA). In some cases (PNA and DBA) neuraminidase digestion was followed by lectin staining. Acinar cells contained conspicuous quantities of oligosaccharides with terminal sialic acid radicals. Galactosil-(13)N-acetylgalactosamine was the most abundant penultimate sugar linked toN-acetylneuraminic acid. Sialylated components having the terminal dimer sialic acid-N-acetylgalactosamine were found in the acinar cells. Secretory cells presented a heterogeneous distribution of glycoconjugates with terminal fucose and -N-acetylgalactosamine. Fucose,N-acetylglucosamine and -N-acetylgalactosamine were present on the apical cytoplasm and surface of the striated and interlobular duct cells. This glycosidic composition was unaffected by extensive selective breeding. In the present work, gustatory glands (von Ebner's glands) of the horse tongue were examined by means of five peroxidase-conjugated lectins (PNA, DBA, SBA, UEA I, WGA), with and without prior sialidase digestion, in order to investigate the presence and distribution of carbohydrate residues in secretory cells and duct cells. The most intense staining of secretory cells was observed with PNA after pre-treatment with neuraminidase. This indicates that the terminal trisaccharide sequence sialic acid- (α2→3, 6) galactosyl (β1→3) N-acetylgalactosamine is the most frequent oligosaccharide chain present in glycoproteins secreted by horse gustatory glands. Secretory cells also contained oligosaccharides with terminal α-N-acetylgalactosamine and N-acetylglucosamine, whereas fucose was found in only a few glandular cells. The apical cytoplasm of duct lining cells reacted with all the lectins except WGA.  相似文献

Nasolabial glands are serous glands forming a thick subcutaneous layer in the bovine muzzle. In order to identify the different epithelial cell types, both immunofluorescent and immunoperoxidase techniques were employed on frozen and fixed sections using monoclonal antibodies to cytoskeletal proteins and S-100. Actin was also detected with phalloidin. The results show that four cell types can be identified on the ground of the different composition of the cytoskeletal filaments: acinar, basket, luminal duct and basal duct cells. Acinar, luminal duct cells and basal duct cells express different patterns of cytokeratins, as shown by the 12 anti-cytokeratin monoclonal antibodies used, and both basket cells and the basal cells of intercalated ducts are also reactive to phalloidin and anti-x-smooth muscle actin monoclonal antibody. The presence of actin supports the conclusion that basal duct cells are also contractile elements, i. e. myoepithelial cells. Vimentin, glial fibrillary acidic protein (GFAP), and S-100, molecules considered to be markers of myoepithelial cells by many AA., were not found. The intermediate filaments of the duct epithelium appear more complex and heterogeneous in comparison with those present in the acinar cells.  相似文献

The study investigated seasonal changes in the morphology of the seminal vesicles of Eidolon helvum in a typical African tropical environment. Seminal vesicles of forty-eight bats were examined during the early rainy, late rainy and peak dry seasons, using gross anatomical, histological and ultrastructural techniques. The interlobular connective tissue septa from the external capsule demarcated the glandular parenchyma into numerous lobules of tubulo-alveolar glands. A simple cuboidal epithelium of mono- or bi-nucleated cuboidal cells lined the glandular mucosa during the rainy season. This was replaced by a bistratified cuboidal epithelium, whose upper layer of cells showed evidence of degeneration, during the dry season. The PAS-positive secretory products appeared to be numerous during the rainy season, but few and clumped together during the dry season. The mean gross weight, interlobular septal thickness, acini diameter and epithelial height of the glands varied significantly with season. Ultrastructural features of the secretory cells during the rainy season showed well-developed cytoplasmic organelles, numerous electron lucid secretory vesicles and electron-dense granules. Secretory epithelial cells of the seminal vesicles are very active during the rainy season, while regeneration of degenerated epithelium in preparation for the next reproductive cycle occurs during the dry season.  相似文献

The physiology of the muscle systems of the human lower urinary tract is still not known in detail. To study the functional basics of this complex organ system, experiments are often performed in animal models including rhesus monkeys. To apply the results of animal model studies to the humans, a clear knowledge of the comparative anatomy of both species is necessary. However, detailed comparative studies of the lower urinary tract of the rhesus monkey and the humans are lacking. Accordingly, a detailed study on the sphincteric musculature of the lower urinary tract of the rhesus monkey was performed in order to demonstrate anatomical correspondences and differences between both species. The lower urinary tract anatomy was investigated in 18 male and female rhesus monkeys (Macaca mulatta) by serial sections. Immunohistochemical staining methods were used to differentiate striated and smooth musculature. Three-dimensional reconstructions were performed in order to demonstrate the topographical anatomy of the different muscle systems. In both man and male rhesus monkeys, a urethral sphincter muscle exists independent of the pelvic floor musculature, with a smooth and a striated muscular part. A urinary diaphragm (diaphragma urogenitale) does neither exist in the rhesus monkey nor in the human. In contrast to women, a striated muscle encircles the urethra and vagina together in the female rhesus monkey. A vesical sphincter muscle, found in the human bladder outlet, does not exist in the rhesus monkey.  相似文献

Sheep lacrimal glands are mixed glands, consisting of tubulo-acinar units succeeded by ducts of simple morphology. The secretory portions consist of three cell types: mucous, seromucous and serous, which may be intermingled in the same acinus or may form acini wholly made of only serous or mucous cells. Mucous cells show a rough endoplasmic reticulum that is reduced to a few cisternae located near the cell base and among the interstices of the secretory droplets. Mucous granules appear uniformly electron-lucent. Serous cells display a typical structure; serous granules can be uniformly electron-dense or composed of dense inclusions dispersed in an electron-lucent matrix. The seromucous granules have a bizonal substructure: a dense core is embedded in a highter matrix. Secretory acini are succeeded by intercalated ducts; the epithelium of these ducts gradually increases in height to form a kind of excretory duct, without the intervention of striated ducts.  相似文献

The poll glands of the camel are tubuloalveolar glands. They consist of lobules separated by interlobular connective tissue. Adrenergic axons and blood vessels including fenestrated capillaries are present in the intralobular connective tissue in close proximity to the secretory cells. The prominent features of the secretory cell cytoplasm are many mitochondria, smooth endoplasmic reticulum, and many vesicles in various secretory stages.
It is concluded that the secretory cells have apocrine as well as merocrine modes of secretion. The glands may have the ability to uptake hormones and concentrate them. Additional work is needed before considering these glands as steroid-secreting glands.  相似文献
