以长叶点地梅(Androsace longifolia)无菌实生苗为试验材料,研究植物生长调节剂、大量元素、琼脂、蔗糖、暗处理等因子对长叶点地梅愈伤组织诱导的影响。结果表明,2,4-D更适合长叶点地梅叶片愈伤的诱导,诱导愈伤组织最适宜的培养基是MS基本培养基附加1.0 mg·L-1 2,4-D+0.2 mg·L-1 6-BA+3%蔗糖+0.8%琼脂,诱导率为95.0%;暗处理不利于长叶点地梅叶片愈伤的诱导。  相似文献

本文分别以大蕉、香蕉、粉蕉的未成熟雄花为材料,取其幼嫩花序和花序轴薄片作外植体,研究了不同浓度的2,4-D、6-BA、KT、IAA、NAA对其胚性愈伤组织的诱导和增殖的影响及诱导过程的一系列反应。结果表明,前胚性性愈伤的诱导难易程度为:大蕉难于香蕉,粉蕉最难,胚性愈伤组织相似。诱导前胚性愈伤组织适宜的培养基配方,大蕉为:MS 6-BA4.0 2,4-D1.;香蕉为:MS BA6.0 2,4-D2.0;粉蕉的配方和香蕉一样。诱导胚性愈伤组织适宜的培养基配方MA IAA1.0 2,4-D4.0 NAA1.0。增殖适宜的培养基,大蕉为:MS 6-BA3.0 2,4-D0.5。香蕉为:MA IAA1.0 2,4-D4.0 NAA1.0粉蕉的配方和香蕉一样。  相似文献

以文冠果成熟种子的子叶为外植体,优化培养基配方,诱导收集胚性愈伤组织,建立了高效的同步胚液体培养扩增体系,同步胚转移到固体培养基上,实现了植株的高效再生,为文冠果转基因平台的建立打下了基础.结果表明,文冠果子叶外植体胚性愈伤组织诱导优化培养基为附加2,4-D 2.0 mg·L-1、NAA 1.0 ·L-1、6-BA 1.0 mg·L-1和3%的蔗糖的MS固体培养基,可产生大量胚性愈伤组织;胚性细胞系经继代和初步扩增后,在附加2,4-D 0.5 mg·L-1、NAA 0.5 mg·L-1、6-BA 0.5 mg·L-1和2%的蔗糖的B5液体培养基中增殖产生大量的同步球形胚;球形胚转入NAA 0.25 mg·L-1、6-BA 0.5 mg·L-1的B5固体培养基上,经心形胚、鱼雷形胚和子叶形胚,再生成植株.  相似文献

以佛手的叶片、茎段为材料,建立了其组织培养系统.研究表明,B5比MS更适合作为诱导佛手愈伤组织的基础培养基,诱导愈伤组织效率较高的培养基配方为B5+2.0 mg·L-1 2,4-D+0.2 mg·L-1ZT和B5+1.5 mg·L-1 2,4-D+1.0 mg·L-1 NAA+0.4 mg·L-1 6-BA.在叶片对卡那霉素敏感性的试验中发现,叶片外植体预培养2 d后进行筛选的效果较好,卡那霉素的适宜筛选浓度为25 rng·L-1.  相似文献

【目的】越南紫薯〔Ipomoea batatas(L.)Lam〕品质优良,薯块均匀,商品性好,但产量偏低,产量性状亟待遗传改良,体细胞胚胎发生是快速繁殖和保持种性稳定的基础,对紫薯的遗传改良有重要意义。采用激素处理越南小紫薯外植体,研究越南紫薯的体细胞胚胎再生途径,为通过体细胞无性系变异筛选紫薯高产优质新材料提供新方法。【方法】以市场销售的越南紫薯为材料,以叶片和茎段作外植体,探讨植物生长调节物质种类(6-BA、NAA、2,4-D、ABA)及配比对越南紫薯培养的影响,建立越南紫薯体细胞胚胎发生及再生植株体系。【结果】叶片和茎段诱导愈伤组织培养基相同,难易程度一致,诱导配方为 MS+6-BA 1.0 mg/L + NAA 0.2 mg/L;在普通愈伤组织诱导形成胚性愈伤组织过程中,2,4-D 起重要作用,其他激素起辅助作用,较好的诱导配方为 MS+6-BA 0.1 mg/L + 2,4-D 2.0 mg/L;在胚性愈伤组织诱导产生体细胞胚过程中,ABA 起着重要作用,并进一步发育和分化的适宜的培养基为 MS+6-BA 0.1 mg/L + NAA 0.2 mg/L + ABA 0.5 mg/L。【结论】叶片和茎段愈伤组织诱导最佳培养基为 MS+6-BA 1.0 mg/L+0.2 mg/L NAA,愈伤组织转化为胚性愈伤最佳培养基为 MS+6-BA 0.1 mg/L +2,4-D 2.0 mg/L;胚性愈伤形成体细胞胚胎最佳培养基为 MS+6-BA 0.1 mg/L + NAA 0.2 mg/L + ABA 0.5 mg/L。  相似文献

以小菊七月红、金莲、七月蜂黄、矮红、盏黄5个品种的叶片为外植体,用MS+2.0 mg@L-1 2,4-D+0.2 mg@L-1 6-BA诱导愈伤组织培养基和MS+2.0 mg@L-1 6-BA+0.1 mg@L-1 NAA分化培养基,选出分化能力较强的品种七月红.再以七月红的叶片上部、叶片下部、叶柄、茎段为外植体,在不同培养基(B5、MS)上诱导胚性愈伤组织,发现茎段最易诱导出胚性愈伤组织,培养基以B5添加0.5 mg@L-1 2,4-D+0.2 mg@L-1 6-BA,MS添加1.0 mg@L-1 2,4-D+0.2 mg@L-1 6-BA效果较好;在添加2.0 mg@L-1 6-BA和0.1 mg@L-1 NAA的分化培养基(B5、MS)上,茎段再生能力较理想.  相似文献

为了获得优质的愈伤组织,以火棘的离体叶片为外植体,MS为基本培养基,研究2,4-D和6-BA以不同浓度组合,叶片的暗处理以及不同部位的叶片对愈伤组织诱导的影响。结果表明:培养基MS+2.0mg·L-12,4-D+2.5mg·L-1 6-BA对愈伤组织的诱导最好;暗处理10d,然后转入光照下培养可提高叶片愈伤组织的诱导效果;较上部叶片愈伤组织诱导率可达96.7%。  相似文献

以半夏叶片为外植体,研究了生长调节物质6-BA(6-苄氨基腺嘌呤)、2,4-D(2,4-二氯苯氧乙酸)、α-NAA(α-萘乙酸)、IBA(吲哚丁酸)对半夏人工种胚诱导和增殖的影响。结果表明,6-BA和2,4-D有利于半夏叶片愈伤组织的诱导和增殖,适宜培养基为MS+6-BA 1.0 mg/L+2,4-D 2.0 mg/L;6-BA、IBA和NAA对半夏人工种胚的诱导和增殖效果较好,适宜培养基为MS+6-BA 1.0 mg/L+IBA 2.0 mg/L或MS+6-BA1.0 mg/L+NAA 2.0 mg/L。结果为对半夏人工种胚诱导及工厂化提供了试验依据。  相似文献

The physical and chemical properties of the hydrated alpha-Al(2)O(3) (0001) surface are important for understanding the reactivity of natural and synthetic aluminum-containing oxides. The structure of this surface was determined in the presence of water vapor at 300 kelvin by crystal truncation rod diffraction at a third-generation synchrotron x-ray source. The fully hydrated surface is oxygen terminated, with a 53% contracted double Al layer directly below. The structure is an intermediate between alpha-Al(2)O(3) and gamma-Al(OH)(3), a fully hydroxylated form of alumina. A semiordered oxygen layer about 2.3 angstroms above the terminal oxygen layer is interpreted as adsorbed water. The clean alpha-Al(2)O(3) (0001) surface, in contrast, is Al terminated and significantly relaxed relative to the bulk structure. These differences explain the different reactivities of the clean and hydroxylated surfaces.  相似文献

 采用新方法精制脱落酸(ABA)异构体试样,改变以往靠光学离析法分离racemic型(SR)-(±)-ABA,提高了(S)-(+)-ABA与(R)-(-)-ABA两镜像体纯度。抑制生长试验和残留量分析结果表明:天然型(S)-(+)-ABA活性,显着高于非天然型(R)-(-)-ABA或(SR)-(±)-ABA活性。抑制莴苣种子发芽50%的活性强度(S)-(+)-ABA约是(R)-(-)-ABA的5倍,(SR)-(±)-ABA介于二者之间。抑制萝卜下胚轴生长试验,最显着有效期2~6天,(6天后差异渐小,8日甚微),生理作用期约为一周,(S)-(+)-ABA活性是(R)-(-)-ABA的3.6倍。差异原因可能是两异构体分子立体结构不同所致。  相似文献

2006年,于山西的侯马、长治、交城、祁县、太原,在Steiner诱捕器内加T,Cue,Me诱剂诱集各种实蝇,监测山西境内实蝇的发生动态。结果表明,三点棍腹实蝇(Dacus(Callantra)trimacula(wang))和具条实蝇(Bactrocera(Zeugodacus)scutelltat(Hendel))是山西的优势实蝇种群,各诱集点两种实蝇种群动态基本一致,每年的6月底7月初由南到北相继发生,高峰期出现在8月中下旬至9月上中旬,种群消退结束于9月底到10月上旬。两种实蝇一年发生一代,种群羽化整齐。  相似文献

Optical spectra of high-transition-temperature superconductors in the mid-infrared display a gap of in-plane conductivity whose role for superconductivity remains unresolved. Femtosecond measurements of the mid-infrared reflectivity of YBa(2)Cu(3)O(7-delta) after nonequilibrium optical excitation are used to demonstrate the ultrafast fill-in of this gap and reveal two gap constituents: a picosecond recovery of the superconducting condensate in underdoped and optimally doped material and, in underdoped YBa(2)Cu(3)O(7-delta), an additional subpicosecond component related to pseudogap correlations. The temperature-dependent amplitudes of both contributions correlate with the antiferromagnetic 41-millielectronvolt peak in neutron scattering, supporting the coupling between charges and spin excitations.  相似文献

A comprehensive inelastic neutron scattering study of magnetic excitations in the near optimally doped high-temperature superconductor YBa(2)Cu(3)O(6.85) is presented. The spin correlations in the normal state are commensurate with the crystal lattice, and the intensity is peaked around the wave vector characterizing the antiferromagnetic state of the insulating precursor, YBa(2)Cu(3)O(6). Profound modifications of the spin excitation spectrum appear abruptly below the superconducting transition temperature T(c), where a commensurate resonant mode and a set of weaker incommensurate peaks develop. The data are consistent with models that are based on an underlying two-dimensional Fermi surface, predicting a continuous, downward dispersion relation connecting the resonant mode and the incommensurate excitations. The magnetic incommensurability in the YBa(2)Cu(3)O(6+)(x) system is thus not simply related to that of another high-temperature superconductor, La(2-)(x)Sr(x)CuO(4), where incommensurate peaks persist well above T(c). The temperature-dependent incommensurability is difficult to reconcile with interpretations based on charge stripe formation in YBa(2)Cu(3)O(6+x) near optimum doping.  相似文献

本文对我国北方杨树蛀干性害虫杨大透翅蛾(Aegerla apiformls clerck)进行了研究。除对形态特征,生活史,生活习性记述外,着重该虫的发生量与营林措施及不同杨树品种的关系,以及一系列的防治试验。在此基础上提出成虫期采用活雌虫性诱;卵期树干涂白;幼虫期使用熏蒸毒签等经济有效的防治手段。同时营造乔灌混交林,选用抗虫性较强的毛白杨等13个品种,可有效地控制该虫的发生量。  相似文献

本文对齐口裂腹鱼和重口裂腹鱼消化道的大体形态及显微结构进行了观察和比较,并探讨了结构与食性的关系。两种鱼消化管分为口腔、咽、食道、肠等四个部分;消化道管壁除口腔、咽外均由粘膜、粘膜下层、肌层和浆膜构成;食道上皮均为复层扁平上皮;肠上皮主要由柱状细胞和杯状细胞组成,其间有嗜酸性颗粒细胞;杯状细胞的数量在前、中肠少,后肠增多;两种鱼在口腔、咽形态组织结构,肠长度和盘曲程度方面差异较大,与其食性密切相关。  相似文献

We report that the doping and temperature dependence of photoemission spectra near the Brillouin zone boundary of Bi(2)Sr(2)CaCu(2)O(8+delta)exhibit unexpected sensitivity to the superfluid density. In the superconducting state, the photoemission peak intensity as a function of doping scales with the superfluid density and the condensation energy. As a function of temperature, the peak intensity shows an abrupt behavior near the superconducting phase transition temperature where phase coherence sets in, rather than near the temperature where the gap opens. This anomalous manifestation of collective effects in single-particle spectroscopy raises important questions concerning the mechanism of high-temperature superconductivity.  相似文献
