Tomato plants, susceptible toFusarium oxysporum f. sp.lycopersici, were inoculated by immersing the roots in a conidial suspension ofF. oxysporum f. sp.lycopersici race 1,F. oxysporum f. sp.dianthi race 2 or a mixture of both fungi. Plants inoculated withF. oxysporum f. sp.lycopersici showed disease symptoms after 2 weeks, whereas plants inoculated withF. oxysporum f. sp.dianthi or a mixture of both fungi remained symptomless for over 7 weeks, the duration of the experiment. In another experiment root systems of plants were split and each half was separately inoculated. One half was firstly inoculated withF. oxysporum f. sp.dianthi or treated with water, followed after a week by a second inoculation of the other half withF. oxysporum f. sp.lycopersici or by a water treatment. The disease symptoms in the half firstly inoculated withF. oxysporum f. sp.dianthi were significantly delayed, compared to plants of which that half had been treated with water. BecauseF. oxysporum f. sp.dianthi reduced disease symptoms caused byF. oxysporum f. sp.lycopersici without any direct interaction with this pathogen, it is concluded thatF. oxysporum f. sp.dianthi is able to induce resistance againstF. oxysporum f. sp.lycopersici in tomato plants.  相似文献

Twenty isolates of fluorescent pseudomonads were evaluated for their ability to control damping-off in tomato (Lycopersicon esculentum) and hot pepper (Capsicum annuum). These isolates were characterized as Pseudomonas fluorescens and Pseudomonas putida. Two isolates, PFATR and KKM 1 belonged to P. putida and the remaining 18 isolates belonged to P. fluorescens. Among these isolates, P. fluorescens isolate Pf1 showed the maximum inhibition of mycelial growth of Pythium aphanidermatum and increased plant growth promotion in tomato and hot pepper. P. fluorescens isolate Pf1 was effective in reducing the damping-off incidence in tomato and hot pepper in greenhouse and field conditions. Isolate Pf1 was further tested for its ability to induce production of defense-related enzymes and chemicals in plants. Earlier and increased activities of phenylalanine ammonia lyase (PAL), peroxidase (PO) and polyphenol oxidase (PPO) were observed in P. fluorescens Pf1 pretreated tomato and hot pepper plants challenged with Pythium aphanidermatum. Moreover, higher accumulation of phenolics was noticed in plants pretreated with P. fluorescens isolate Pf1 challenged with Pythium aphanidermatum. Thus, the present study shows that in addition to direct antagonism and plant growth-promotion, induction of defense-related enzymes involved in the phenyl propanoid pathway collectively contributed to enhance resistance against invasion of Pythium in tomato and hot pepper.  相似文献

Filtrates from shake-cultures of Fusarium oxysporum f. sp. lycopersici race 1, concentrated to 20% of the original volume, caused cell death in tomato leaf protoplasts from near-isogenic lines corresponding to the compatible cultivar/race reactions of whole plants. Maximum activity was found in late log phase cultures on Czapek-Dox supplemented with 2% casamino acids. Selective toxicity was associated only with the protein fraction of the culture filtrate. LD50 values for susceptible Ace and Moneycross to F. oxysporum f. sp. lycopersici race 1 culture filtrates were 1·92 and 0·36 μg protein ml−1. Corresponding values for cvs Royal Ace and MM161, each containing the I-gene conferring resistance to race 1, were >350. Culture filtrates from F. oxysporum f. sp. lycopersici race 2 gave LD50 values of 2·34 and 2·08 μg protein ml−1 on cvs Ace and Royal Ace, both susceptible to race 2. The LD50 of cv. Ace to a non-pathogenic isolate of F. xysporum f. sp. lycopersici was > 350. Culture filtrates from non-host formae of F. oxysporum were 9–149-fold less toxic on cv. Ace. Protoplasts from Pisum sativum, Lactuca sativa, Zea mays, Gossypium barbadense and Solanum melongena, all non-hosts of F. oxysporum f. sp. lycopersici, were 6–175 times less sensitive to F. oxysporum f. sp. lycopersici filtrates than susceptible tomato. The putative toxins lycomarasmin and fusaric acid showed no differential toxicity to I+ and I tomato protoplasts. The results are discussed in the wider context of host-pathogen interaction in which specificity is considered as the recognition of susceptibility by a proteinaceous toxic metabolite of the pathogen. This hypothesis is further extended to include the specificity of F. oxysporum formae and races.  相似文献

Simultaneous inoculation with races 1 and 2 of the vascular wilt pathogenFusarium oxysporumf.sp.lycopersiciprovided a high level of protection against race 2 in three tomato cultivars carrying resistance geneI, which confers resistance to race 1 but not race 2. However, simultaneous inoculation did not provide any protection in cultivars lacking this gene. Protection resulted in reduction and delay of wilt symptoms. Similarly, avirulent races ofF. oxysporumf.sp.melonisprotected muskmelon plants against virulent races of the sameforma specialis.A ratio 10:1 between spore concentrations of inducer and challenger organism gave the highest cross protection, but ratio 0.1:1 still provided significant disease reduction. Cross protection was also obtained when inoculation with the inducer organism was performed 6 or 12 h before inoculation with the challenger organism. Autoclaved spores of the inducer did not have any protective effect, indicating that living propagules were required to initiate protection. The results suggest the presence of a gene-for-gene interaction betweenF. oxysporumf.sp.lycopersici-tomato andF. oxysporumf.sp.melonis-muskmelon, in which cross protection against a virulent race is mediated by recognition of a specific elicitor from the avirulent race by the plant resistance gene product and by subsequent induction of the plant defense reaction.  相似文献

Forms ofFusarium oxysporum specific on hosts other than tomato induce in this plant greater initial increases of the phenols content than the pathogenic f. sp.lycopersici. Mixed inoculations of f. sp.lycopersici and f. sp.dianthi are on the contrary no more effective in inducing the phenol accumulation 24 h after the infection than f. sp.lycopersici alone. Activities of polygalacturonase, pectin lyase, and cellulase increased in susceptible, catechol-treated, and resistant tomato plants, after inoculation withFusarium oxysporum f.lycopersici (Sacc.) Snyder and Hansen race 2. The catechol-treated and the resistant plants remained symptomless, while susceptible plants developed symptoms of disease. It is therefore suggested that increased activity of cell-wall-degrading enzymes in inoculated plants does not necessarily cause the development of disease symptoms.  相似文献

Fusarium oxysporum f. sp. lycopersici (FOL) races and F. oxysporum f. sp. radicis lycopersici (FORL), the causal agents of root rot and crown rot diseases, respectively, cause serious economic losses in tomato greenhouses where production is intensive in the West Mediterranean region of Turkey. The isolates were collected from West Mediterranean region of Turkey and were characterized by specific primers based on three races (r1, r2, r3), besides pathogenicity tests in in vivo conditions Additionally, a scheme was developed using newly tested ISSR and SRAP markers to a genotyping database and to determine the possible origin of these pathogens. The present study provided new information on these pathogens based on their races and their dominant existence in this region that has not been reported before. Genetic diversity detected in the same races of the pathogen may be associated with difficulties in controlling the pathogen and a possible resistance formation effort exerted by the pathogen to chemicals used in plant protection in tomato greenhouses. Molecular analyses indicated genetic diversity in pathogen isolates identified as r3, r2 and FORL, which may be associated with abiotic stress to which the pathogens were exposed.  相似文献

Fusarium crown and root rot caused by Fusarium oxysporum f. sp. radicis-lycopersici (FORL) is a serious soilborne disease reducing tomato yields in Florida, a leading state in fresh market tomato production in the United States. One hundred and twenty five isolates of FORL were collected from the three main tomato-growing counties in Florida between 2006 and 2008. Vegetative compatibility groups (VCGs) and 10 microsatellite loci were used to infer the population structure of FORL. Up to 69.8 % of the isolates could be assigned to one of three VCGs, 0094, 0098 or 0099, with frequencies of 38.6, 24.4, and 6.8 % respectively. A medium level of population differentiation (Φst?=?0.159) was detected among the three counties, and three optimal clusters (populations) were supported by discriminant analysis of principal components. In addition, each population had some individuals that likely migrated from other populations. Migration probably played an important role in shaping the population structure of FORL since repeated VCGs and multilocus genotypes were observed in the three counties. Considerable migrants (> 1.33 migrants per generation) were also detected between the three counties, resulting in an increase in the effective population size and genetic diversity of F. oxysporum f. sp. radicis-lycopersici. Although migration is an important evolutionary force, mutation and parasexual recombination could not be completely ruled out as contributing causes to the genetic diversity of FORL. Management strategies that limit the movement of F. oxysporum f. sp. radicis-lycopersici are necessary to reduce the evolutionary potential of the pathogen.  相似文献

The suppression of fusarium wilt of tomato achieved by Penicillium oxalicum (PO) applied one or several times (up to four) was assessed during three glasshouse experiments. The first application of PO (106 conidia g-1 substrate) to the growing substrate (peat and vermiculite, 1 : 1, v : v) was performed prior to its infestation with Fusarium oxysporum f. sp. lycopersici (FOL) (104–106 chlamydospores g-1). Repeated applications of PO prolonged the duration of control of fusarium wilt especially when disease incidence was high. The timing of repeated applications of PO did not affect the efficacy of the control. Disease reduction was not associated with a decrease in density of FOL in the rhizosphere, irrespective of the number of applications of PO. Density of PO in the tomato rhizosphere was higher when repeated applications were made. No relationship was observed between reduction of disease and high densities of PO. Reasons for a longer disease reduction in tomato plants following several applications with PO are discussed.  相似文献

We selected a reduced-pathogenicity mutant of Fusarium oxysporum f. sp. lycopersici, a tomato wilt pathogen, from the transformants generated by restriction enzyme-mediated integration (REMI) transformation. The gene tagged with the plasmid in the mutant was predicted to encode a protein of 321 amino acids and was designated FPD1. Homology search showed its partial similarity to a chloride conductance regulatory protein of Xenopus, suggesting that FPD1 is a transmembrane protein. Although the function of FPD1 has not been identified, it does participate in the pathogenicity of F. oxysporum f. sp. lycopersici because FPD1-deficient mutants reproduced the reduced pathogenicity on tomato.The nucleotide sequence data reported are available in the DDBJ/EMBL/GenBank databases under the accession number AB110097  相似文献

Arbuscular mycorrhiza (AM) colonization led to a decrease in the severity of fusarium wilt disease caused by Fusarium oxysporum f. sp. lycopersici in tomato plants. The involvement of two plant defense hormones, namely methyl jasmonate (MeJA) and salicylic acid (SA), in the expression of mycorrhiza induced resistance (MIR) against this vascular pathogen was studied in the AM colonized and non-colonized (controls) plants. Activity of lipoxygenase (LOX), which plays a role in jasmonic acid (JA) biosynthesis, as well as levels of methyl jasmonate (MeJA) increased in AM colonized plants as compared to controls, but did not show any further changes in response to F. oxysporum inoculation. On the other hand, activity of phenylalanine ammonia lyase (PAL), which is an enzyme from salicylic acid (SA) biosynthetic pathway, as well as SA levels, increased in both controls and AM colonized plants in response to application of F. oxysporum spores. Hence the JA and not the SA signalling pathway appeared to play a role in the expression of MIR against this vascular pathogen. The resistance observed in AM colonized plants was completely compromised when plants were treated with the JA biosynthesis inhibitor salicylhydroxamic acid (SHAM). This confirmed that the AM-induced increase in JA levels was involved in the expression of resistance toward F. oxysporum. The SA response gene pathogenesis-related 1 (PR1) showed an increased expression in response to F. oxysporum infection in SHAM treated AM colonized plants as compared to plants that were not treated with this JA inhibitor. This suggested the possibility that JA inhibited SA responses, at least in the roots. AM colonization therefore appeared to prime plants for improved tolerance against the vascular pathogen F. oxysporum, which was mediated through the JA signalling pathway.  相似文献

Plant growth promoting Bacillus subtilis MSS9 and Bacillus licheniformis MSS14 were isolated from the tomato rhizosphere. These isolates were capable of inhibiting the fungal pathogen, Fusarium oxysporum f. sp. lycopersici causing fusarium wilt in tomato, tested by dual culture method and by mycolytic enzyme production. The isolates have the capacity to form biofilm on the microtitre plate. Scanning electron microscopy revealed good colonization capacity of Bacillus licheniformis MSS14 on tomato plant root as compared to Bacillus subtilis MSS9, pot experiments were also analyzed to study the effects of both rhizobacterial cultures on pathogen development and plant growth. It was observed that MSS14 reduces the incidence of Fusarium oxysporum f. sp. lycopersici in tomato and there was significant increase in vegetative parameters like root length, shoot length, plant wet weight, dry weight and chlorophyll content after which indicates that the root colonization property of the culture MSS14 helps in enhancing the biocontrol capacity against pathogen than that of MSS9.  相似文献

The response of microconidia from pathogenic and non-pathogenic Fusarium oxysporum to root exudates from tomato plants inoculated with different pathogenic and non-pathogenic F. oxysporum strains was studied. Root exudates from non-inoculated tomatoes highly stimulated the microconidial germination of the two tomato pathogens, F. oxysporum f.sp. lycopersici strain Fol 007 and F. oxysporum f.sp. radicis-lycopersici strain Forl 101587. In root exudates from tomato plants challenged with the pathogen Fol 007 the microconidial germination of Fol 007 was increased, whereas in root exudates from plants challenged with Forl 101587 the microconidial germination of Fol 007 was reduced. Root exudates of tomato plants challenged with the non-pathogenic unspecific F. oxysporum strain Fo 135 and the biocontrol strain Fo 47 clearly reduced microconidial germination of the pathogenic strain Forl 101587. Moreover, the microconidial germination rate of the biocontrol strain Fo 47 was increased in the presence of root exudates of tomato plants challenged with the tomato wilt pathogen Fol 007. These results indicate that pathogenic and non-pathogenic F. oxysporum strains alter the root exudation of tomato plants differently and consequently the fungal propagation of pathogenic and non-pathogenic F. oxysporum strains in the rhizosphere is affected differently.  相似文献

Bacillus amyloliquefaciens IUMC7 isolated from mushroom compost inhibited growth of Fusarium oxysporum f. sp. lycopersici (FOL) on culture plates, and a culture supernatant of IUMC7 inhibited in vitro germ tube elongation of FOL. When compared with control soils, mushroom compost inoculated with IUMC7 significantly reduced disease severity caused by FOL in tomato plants. PCR tests for expression of PR genes indicated that IUMC7 did not induce resistance in tomato plants. These results suggested that the suppression of disease was mainly caused by antimicrobial compounds produced by IUMC7.  相似文献

Biological soil disinfestation (BSD), or reductive soil disinfestation, achieved by amendment with organic materials such as wheat bran followed by flooding and covering the soil surface, has been used to control some soilborne diseases including Fusarium wilt and bacterial wilt of tomato. During a BSD treatment, accumulation of acetic acid and/or butyric acid was detected with high-performance liquid chromatography. Survival of Fusarium oxysporum f. sp. lycopersici or Ralstonia solanacearum was suppressed by these organic acids. Amendment of these organic acids into soil suppressed the survival of R. solanacearum at lower concentrations than the maximum detected in BSD treatment, indicating that production of these organic acids is one of the mechanisms of control. However, F. oxysporum f. sp. lycopersici in soil survived with the maximum concentrations of these organic acids achieved by BSD; thus, involvement of factors other than organic acids may be involved.  相似文献

The lipids metabolism of tomato and bean plants during biological control of wilt pathogens (Fusarium oxysporum f.sp.lycopersici andF. oxysporum f.sp.phaseoli, respectively) byBacillus subtilis was investigated. The interaction of wilt pathogens with both tomato and bean caused an imbalance and drastic reduction in total lipids, triacylglycerol, sterol and all phospholipd fractions except phosphatidic acid. The application of a formulated biocontrol agent,B. subtilis, eliminated the detrimental effect of both wilt pathogens and consequently prevented catabolism of lipid fractions in both tomato and bean. Three strains of Fusarium oxysporum were tested for their antagonism to Fusarium oxysporum f.sp. dianthi, two from alluvial soils of the Durance valley (Provence, FR) known to be suppressive to F.o. f. sp. melonis (strains 47 and 20 of Alabouvette) and a third from carnation growing medium, in which it spontaneously appeared (TF3). Strains 47 and 20 were introduced into perlite before planting carnations and found to have variable efficacy against fusarium wilt, according to the formulation of the inoculum. With a talc preparation, these strains gave only 15% witted plants, compared with 49% in the control. With a bran-perlite inoculum, only strain TF3 gave better results than the control. Repeated introduction of TF3 through the season increased the incidence of disease, whereas a single treatment before planting in perlite was effective whatever the formulation (humid bran-perlite, bran-perlite powder, talc, liquid). However, the antagonistic activity of TF3 was not sufficient to control the disease in a naturally infested soil.  相似文献

Pseudomonas fluorescens strains CHA0 and Pf1 were investigated for their biocontrol efficacy against Banana bunchy top virus (BBTV) in banana (Musa spp.) alone and in combination with chitin under glasshouse and field conditions. Bioformulation of P. fluorescens strain CHA0 with chitin was effective in reducing the banana bunchy top disease (BBTD) incidence in banana under glasshouse and field conditions. In addition to disease control, the bioformulation increased the economic yield significantly compared to the untreated control. Increased accumulation of oxidative enzymes, peroxidase (PO), polyphenol oxidase (PPO), phenylalanine ammonia lyase (PAL), pathogenesis-related (PR) proteins, chitinase, β-1,3-glucanase and phenolics were observed in CHA0 bioformulation amended with chitin-treated plants challenged with BBTV under glasshouse conditions. Indirect ELISA indicated the reduction in viral antigen concentration in P. fluorescens strain CHA0 with chitin-treated banana plants corresponding to reduced disease ratings. The present study revealed that induction of defence enzymes by P. fluorescens with chitin amendment reduced the BBTD incidence and increased bunch yield in banana.  相似文献

The behaviour of Fusarium oxysporum f.sp. lycopersici (Fol) and the effectiveness of the microbial control agent Trichoderma asperellum strain T34 were examined in hydroponically grown tomato plants under five ammonium/nitrate ratios. The results showed that disease severity was reduced by the action of T34 under increasing concentrations of ammonia. Furthermore, rhizosphere F. oxysporum populations decreased with T34 application. The presence of T34 augmented leaf nitrogen concentration in treatments infested with Fol. In addition, T34 application reduced iron concentration in tomato leaves at high ammonium/nitrate ratios and reduced the severity of Fusarium wilt at high iron and nitrogen leaf concentrations.  相似文献
