Localization of the sheep FcRn in the mammary gland   总被引:5,自引:0,他引:5  
Among the multiple functions, which have been identified for the neonatal Fc receptor (FcRn), we study its role in the IgG transport in the mammary gland during the colostrum formation. For this reason, we have obtained several mammary gland biopsies from a pregnant sheep around parturition. The presence of the FcRn heavy chain mRNA was detected exclusively in the acinar and ductal epithelial cell by in situ hybridization (ISH). We detected strong signal in samples harvested 24 and 10 days prepartum; however, in samples we collected postpartum was barely detectable. Immunohistochemistry confirmed our ISH data. The cytoplasm of the epithelial cells of the acini and ducts in the mammary gland biopsies stained homogeneously before parturition, although a remarkable difference was observed in the pattern after lambing. The signal indicated uneven distribution of the FcRn alpha chain in the epithelial cells 1 and 5 days postpartum, since the apical sides of the epithelial cells were highlighted. The presence of the FcRn in the acinar and ductal epithelial cells and the obvious change of its distribution before and after parturition suggest that FcRn plays an important role in the IgG transport during colostrum formation. FcRn expression was also found in the lamb duodenal crypt epithelial cells, which have been previously demonstrated to secrete IgG1 in newborn ruminants, suggesting secretory role of the FcRn in ruminant epithelial cells.  相似文献

Many of the receptors for the Fc domain of immunoglobulins in cattle, sheep, pigs and horses have been cloned and characterized recently. This review summarises recent developments and relates them to the current understanding of the primary structure, cellular specificity and binding properties of Fc receptors (FcRs). Although there is an obvious overall similarity to their human and mouse counterparts, some Fc receptors in domestic animals are unusual, perhaps most notably the bovine Fcgamma2R, which although related to other mammalian FcgammaRs, belongs to a novel gene family and the porcine FcgammaRIIIA, which associates with a molecule that contains significant homology to the cathelin family of antimicrobial proteins. Accumulating data suggest the possibility of a different role for the FcRn in ruminants, which may secrete IgG onto the mucosal surfaces, rather than absorbing it, as was suggested by mouse studies. These differences may be linked to the diversity of immunoglobulin classes in different mammalian species, and may contribute to different immune functions. The observations made so far emphasize the importance of elucidating and analyzing the roles of these molecules within the immune system of each host animal, rather than inferring roles from conclusions made in human and mouse studies. A better understanding of Fc receptor expression on immune effector cells should help in developing new immunization protocols, while knowledge of the Fc receptors involved in immunoglobulin transport, especially in the mammary gland, may help to develop new products which could be used not only for veterinary purposes but perhaps also for human therapy.  相似文献

Hepatocyte growth factor/scatter factor (HGF/SF) is a pleiotropic cytokine that plays a crucial role in the embryonic and postnatal development of various organs including the mammary gland. We cloned bovine HGF and its c-Met receptor cDNAs, and examined their expression during mammary gland development in dairy cows. The 2.5-kbp HGF cDNA clone contained a 2190 bp open reading frame coding a 730 amino acid protein, while the 4.8-kbp c-Met cDNA clone contained a 4152 bp open reading frame coding a 1384 amino acid protein. The bovine HGF and c-Met sequences exhibited more than 87% identity with those of other mammals. RT-PCR analysis revealed ubiquitous expression of both HGF and c-Met mRNAs in various bovine tissues tested. HGF mRNA was detected only in the inactive stage of bovine mammary gland development and not in the developing, lactating, and involuting stages, while c-Met mRNA was detected in the inactive and involuting stages. Immunohistochemical analysis demonstrated that the c-Met protein was found on mammary epithelial cells in the inactive, developing, and involuting stages, and on myoepithelial cells in all stages. These results suggest pivotal roles of HGF and c-Met in the development of bovine mammary gland.  相似文献

Marsupial young are born in an under-developed state without mature immune responses. Prior to the maturation of an immune system, marsupial young are heavily reliant upon immune factors secreted in the milk to defend them against potential microbial pathogens in the environment. In this study, we identified and characterized the immunoglobulin heavy chain constant regions, light chains, polymeric Ig receptor (pIgR), J chain, neonatal Fc receptor (alpha chain) (FcRn) and the chemokine CCL28 from the model marsupial species, the tammar wallaby (Macropus eugenii). Low levels of conservation were seen in motifs in C and Cγ associated with receptor binding and or transcytosis, and this may have potential implications for functionality. We evaluated the expression of immunoglobulin genes in the tammar mammary gland throughout lactation and found that two periods of increased expression of immunoglobulin genes occur. These two periods coincide with the birth of the young, and with its first emergence from the pouch. This increased expression may represent a strategy for maternal immunological protection of the pouch young.  相似文献

In neonatal calves, maternal immunoglobulin (Ig) is transferred into respiratory secretion which contributes to protection against pathogens. The early predominance of IgG1 in respiratory tract secretions is progressively reduced in favor of IgA by age but in the lower, bronchoalveolar system secreted IgG remains the dominant secreted Ig even in adulthood. The trans-epithelial transport of secretory IgA into mucosal secretions is carried out by the polymeric Ig receptor. However, the mechanism by which IgG crosses epithelial cells to provide defense on mucosal surfaces is still unknown. In order to investigate the possibility that the neonatal Fc receptor, FcRn is involved in this transport we have first analyzed the localization of this receptor in the upper and lower respiratory tracts. Consistent with the in situ hybridization data, immunohistochemistry showed undetectable expression in the tracheal epithelial cells, relatively weak expression in epithelial cells of the bronchi, apparent staining those lining the bronchioli and randomly scattered signal over the alveolar tissue. The bovine FcRn may thus play a role in IgG transport across mucosal epithelial barriers as a trafficking receptor and ensure IgG predominance in the lower respiratory tract.  相似文献

Our previous report demonstrated that high concentration of taurine is present in rat milk for the first few days of lactation and plays an important role in the body growth of rat pups. In the present study, gene expression of rate-limiting enzyme for taurine biosynthesis, cysteine sulfinate decarboxylase (CSD) were examined in rat mammary gland. By RT-PCR, CSD mRNA was found to be expressed in rat mammary gland like that in the liver. The expression level of CSD mRNA in the mammary gland was higher in the earlier lactational stage (days 1 and 6 of lactation) than that in the later lactational stage (day 14). CSD mRNA expression in the mammary gland of non-pregnant rats was only a trace level. By in situ hybridization analysis, CSD mRNA was demonstrated in the epithelial cells of the mammary gland. These results suggest that high concentrations of taurine in the milk are at least partially resulted from de novo synthesis of taurine in mammary gland epithelial cells and that the expression pattern of CSD mRNA may be responsible for the changes in taurine levels in the milk during a lactational period.  相似文献

To examine the PTH/PTHrP receptor in the mammary gland, molecular cloning of bovine PTH/PTHrP receptor and measurement of its mRNA expression were carried out in cows during the periparturient period. The PTH/PTHrP receptor gene was partially cloned, and expression of bovine PTH/PTHrP receptor mRNA was detected in various tissues of the cow. In the mammary gland, PTH/PTHrP receptor mRNA expression was constantly low during the periparturient period, whereas PTHrP mRNA expression dramatically increased after parturition. This suggested that expression of PTH/PTHrP receptor mRNA in the mammary gland is not affected by lactation in cows.  相似文献

Changes in mare mammary secretion composition were characterized prior to and after foaling. Eighteen mares were used for collection of mammary secretions from 23 days prepartum through 44 days postpartum. Concentrations of lactose, total protein, IgG and lysozyme (activity assay) were determined. Considerable variabilty was observed among mares. Mean lactose concentrations by mare were lower (P<.05) in the prepartum period compared with the postpartum period. Mean total protein and immunoglobulin G concentrations by mare were higher (P<.05) in the prepartum period compared with the postpartum period. Mean lysozyme activities by mare tended to be lower (P<.08) during the prepartum period compared with the postpartum period. Mammary secretion protein profiles, characterized by SDS-PAGE, were consistent with changes in concentrations of specific proteins measured in the secretions. Dynamic changes occurring in mammary secretions during the transition from prepartum accumulation of colostrum to postpartum production of milk include factors like IgG and lysozyme which have protective roles in the neonatal foal.  相似文献

Recently, the type II sodium-dependent phosphate cotransporter NaPi IIb, which mediates the intestinal phosphate transport, was detected in the apical membranes of caprine mammary gland epithelial cells. Regulatory influences of developmental stages, dietary phosphorus (P) supply and hormones like calcitriol are well described for the intestinal NaPi IIb. Therefore, it was the aim of this study to examine the influence of involution and dietary P restriction on the expression of mammary gland NaPi IIb and of vitamin D receptor (VDR). During involution both, NaPi IIb and VDR, showed an initial increase of expression. This resulted in a delayed response on protein level. Dietary P restriction resulted in a decrease of mRNA expression which was not reflected on protein level. Influenced expression pattern, at least on mRNA level, indicate that mammary gland NaPi IIb is a regulated phosphate transporter which might have an important role especially during involution. Coexpression pattern with VDR provides an indication that calcitriol could be the modulator of these adaptive responses to involution and dietary P restriction. Therefore, a physiological meaning of NaPi IIb in mammary gland epithelia during processes of cell regeneration has to be considered.  相似文献

No knowledge regarding the peripartum changes in mammary secretions in the jenny are presently available in literature. In the mare, instead, several studies report the role of these changes as indicators of foetal readiness for birth and impending parturition. This experiment was designed to determine calcium, sodium, potassium concentrations, and the value of sodium/potassium ratio in mammary fluids during prepartum in the jenny. Samples were daily collected by hand milking, after mammary gland size increased noticeably, from 17 Martina Franca jennies. Prepartum mammary secretions were analysed every other day between day 10 and day 2 antepartum, and then once a day from the day before to the day of parturition. Calcium concentration showed a significant increase between day 10 and day 6 antepartum and then between day 6 and days 4 and 2. Afterwards, another statistical significant increase was observed at parturition. Sodium concentration significantly decreased from day 10 to day 2 prepartum. Potassium concentration significantly increased between day 10 and day 8 before parturition, then showed a further increase at day 4, followed by none significant changes until foaling. All jennies showed a reversal in sodium/potassium ratio between 2 days antepartum and the day before. In conclusion, the evaluation of mammary fluid calcium concentrations and the reversal of sodium/potassium ratio could be used as good indicators of foetal maturity in the jenny. As far as the prediction of parturition is concerned, the reversal of sodium/potassium ratio is the best parameter, since it was detected 48–24 h before parturition in all considered animals.  相似文献

Kinetic properties and substrate specificity of the lysine transport system in porcine mammary gland were studied using mammary tissue explants from nine lactating sows. Sodium dependence of lysine uptake was determined by replacing sodium in the medium with choline. Kinetic parameters of lysine uptake were determined using lysine concentrations from 5 microM to 5.12 mM. Competition of lysine uptake by other amino acids was determined using the cationic amino acids, arginine and ornithine, and using other essential amino acids. Transport of lysine was time-dependent and was unaffected by replacing sodium with choline. Lysine uptake occurred by a transport mechanism with a Km of approximately 1.4 mM and a Vmax of 7.9 mmol x kg cell water(-1) x 30 min(-1). Lysine uptake was inhibited by arginine and ornithine and by high concentrations of L-alanine, L-methionine, L-leucine, cycloleucine, and D-lysine, but not by 2-(methylamino)-isobutyric acid. This transport mechanism is the primary system responsible for uptake of cationic amino acids in lactating sow mammary tissue. The relatively high Km, compared with physiological blood concentrations of lysine, indicates that the kinetic properties of the lysine transport system should not be limiting to milk protein synthesis. Transmembrane transport of lysine by lactating sow mammary tissue should be a direct function of plasma concentrations. However, interactions of other amino acids with the uptake system may affect lysine uptake.  相似文献
