Occurrence of a variant of cucumber green mottle mosaic virus in ISRAEL   总被引:1,自引:0,他引:1  
A virus was isolated from naturally infected melons and squirting cucumber(Ecballium elaterium (L.) Rich) plants collected in the northern part of Israel. Some data are presented on the biological, serological and physical properties of the virus, which was identified as a variant of cucumber green mottle mosaic virus.  相似文献

番茄斑驳花叶病毒(tomato mottle mosaic virus, ToMMV)是2013年发现的烟草花叶病毒属一个新种,目前在多国(地区)有发生。本文采用小RNA深度测序及RT-PCR检测方法在广东省广州市南沙区辣椒疑似病样中检测到ToMMV,命名为番茄斑驳花叶病毒广东分离物(ToMMV-GD-2020)。采用RT-PCR分段扩增获得了ToMMV-GD-2020的基因组全序列,该分离物基因组全长6 399 nt,包含4个开放阅读框,分别编码4个蛋白。序列相似性分析表明,ToMMV-GD-2020与已登录GenBank的14个ToMMV分离物基因组序列相似性分别为99.0%~99.7%,其中与中国辽宁分离物ToMMV-LN(GenBank登录号:MN853592)的相似性最高(99.7%),与危害我国番茄、同属烟草花叶病毒属的番茄花叶病毒(tomato mosaic virus, ToMV)、番茄褐色皱果病毒(tomato brown rugose fruit virus, ToBRFV)代表分离物的相似性分别为84.6%和81.0%。系统进化分析表明,ToMMV-GD-2020...  相似文献

A mosaic and stunt disease of chilli pepper (Capsicum frutescens) in Ivory Coast was found to be caused by pepper veinal mottle virus, a member of the potato virus Y group, recently described from chilli pepper in Ghana. The Ivory Coast isolate was first transmitted by aphids toPhysalis floridana and subsequently maintained by mechanical inoculation inP. floridana andNicotiana megalosiphon. The host range includes mainly Solanaceae, butSolanum species were not or very little susceptible.Aphis gossypii, A. spiraecola andToxoptera citricidus transmitted the virus in a non-persistent manner. No dodder transmission was found. Serological relationship with onion yellow dwarf virus and Columbian Datura virus was established.Samenvatting Een zeer algemeen voorkomende virusziekte van spaanse pepers (Capsicum frutescens) in Ivoorkust bleek veroorzaakt te worden door het pepper veinal mottle virus (PVMV) door Brunt en Kenten (1971 en 1972) uit Ghana beschreven. Het virus maakt deel uit van de aardappelvirus-Y-groep.Hoewel ook de waardplantenreeks van de in Ivoorkust voorkomende stam van het virus (PVMV-CI) voornamelijk Solanaceeën omvat, bleek het geslachtSolanum echter in hoge mate onvatbaar.Physalis floridana enNicotiana megalosiphon zijn goede diagnostische soorten. PVMV-CI kan op non-persistente wijze overgebracht worden doorAphis gossypii, A. spiraecola enToxoptera citricidus. De laatste twee zijn nog niet eerder als vectoren vermeld. Overdracht door middel van warkruid,Cuscuta subinclusa, bleek niet mogelijk. Het virus werd gezuiverd uitN. megalosiphon, waarbij opbrengsten van 5–20 mg virus per kg vers blad verkregen werden. 以来自甘肃的南瓜果实样品中提取的总RNA为模板进行RT-PCR扩增,序列测定和分析结果表明:扩增片段包含CGMMV的外壳蛋白基因和3’非编码区,证明南瓜果实中存在黄瓜绿斑驳花叶病毒,将其印基因序列与来自国内不同地区的其他CGMMV分离物进行序列比对,发现国内各分离物间的同源率高,有很近的亲缘关系。  相似文献

A viral disease of broad bean ( Vicia faba ) was observed in Jiangsu, Zhejiang and Sichuan Province, the People's Republic of China. Its typical symptom on infected leaves was a white mottle, and, rarely, red necrotic lesions on several broad bean cultivars. A non-aphid-transmissible strain of turnip mosaic virus (TuMV) was identified as the causal agent of the disease on the basis of biological properties, particle morphology, seed and aphid transmission and serological tests. The virus could not be transmitted by aphids in the non-persistent manner.  相似文献

香石竹斑驳病毒的鉴定和RT-PCR检测   总被引:7,自引:0,他引:7  
从表现为叶斑驳、花碎色症状的香石竹病株上获得一病毒分离物 ,电镜负染观察到直径为28~33nm的球状粒子。病毒提取液经紫外光测定呈典型核蛋白吸收曲线 ,OD260/OD280=1.70;血清学反应与CarMV抗血清出现明显的沉淀线。通过以上实验结果 ,确定该病毒分离物为香石竹斑驳病毒(carnation mottle virus ,CarMV)。根据该病毒的RNA序列设计引物 ,对病健材料进行了RT-PCR检测 ,结果从感病材料中扩增出大约600bp的特异片段 ,而健康植物无此扩增带。将PCR产物连接 pGEM-T-easy载体 ,转化大肠杆菌JM109,得到了含目的片段的重组子 ,经双脱氧序列分析 ,与Guilley报道的序列对应部分的核苷酸序列基本一致 (其同源性达96% ) ,最低检出病毒核酸含量为5ng ,表明应用RT-PCR检测香石竹斑驳病毒是可行的  相似文献

黄瓜绿斑驳花叶病毒(Cucumber green mottle mosaic virus,CGMMV)主要危害葫芦科作物,已被世界上许多国家和地区列为检疫性有害生物。CGMMV目前在我国23个省、市、区已有报道发生和危害,严重影响葫芦科作物的生产;近年来该病害在国内外呈现迅猛扩展的趋势并对生产造成危害。本文综述了防治该病害的种子处理、化学及生物防治、嫁接以及转基因等分子生物学方法;分析了CGMMV与寄主黄瓜互作研究的最新进展,对小分子RNA参与调控寄主对CGMMV病毒的防控策略提出了展望,并概述了下一代测序技术、基因编辑技术在植物新病毒的检测、鉴定以及培育抗病新品种等方面的应用。  相似文献

玉米褪绿斑驳病毒实时荧光RT-PCR检测方法研究   总被引:2,自引:0,他引:2  
玉米褪绿斑驳病毒(Maize chlorotic mottle virus,MCMV)是我国对外公布的检疫性有害生物。本研究根据该病毒外壳蛋白基因的保守序列,设计得到特异性引物及Taqman荧光探针,建立了MCMV的实时荧光RT-PCR方法,并对其灵敏度与特异性进行了研究。该方法针对2个不同来源的毒株均能得到典型扩增曲线,而没有从小麦线条花叶病毒、玉米粗缩病毒和玉米矮花叶病毒的RNA得到扩增曲线,表明引物与荧光探针具有良好的特异性。针对玉米褪绿斑驳病毒RNA不同稀释度样品,实时荧光RT-PCR检测低限达到10-5稀释度,检测灵敏度要比普通RT-PCR高出100倍。因此,本研究建立的MCMV实时荧光方法具有特异性强、灵敏度高和快速有效的优点。  相似文献

通过病毒dsRNA分离技术、非序列依赖性PCR扩增(sequence-independent amplification,SIA)技术和RTPCR等手段对采自山西太谷的西瓜病样进行了检测与鉴定,确定其为黄瓜绿斑驳花叶病毒(Cucumber green mottle mosaic virus,CGMMV)所侵染。为明确CGMMV西瓜分离物(CGMMV-SXTG)的分类地位,本研究进一步克隆了CGMMV-SXTG的外壳蛋白基因(coat protein,CP)全序列并进行序列分析,系统进化树分析显示该分离物与亚洲分离物亲缘关系较近,而与欧洲分离物亲缘关系较远,表明该病毒的不同分离物在分组上可能与地域有一定的关系;接着对不同分离物的CP基因核苷酸序列和氨基酸序列进行同源性比对分析,结果表明该分离物与中国山东分离物(TANG)同源性最高,分别达到99.8%和99.6%。  相似文献

Momordica charantia L. plants systemically infected with Cucumber mosaic virus (CMV) were found in Oita Prefecture. The virus isolated from the host plant was characterized by biological, serological, and molecular biological methods. The purified virus was used to mechanically inoculate the host and produced green mottle, green mosaic, and/or chlorotic spots in the noninoculated upper leaves of the host. The virus was identified as an isolate of CMV containing genomic RNA3 derived from subgroup IA by several lines of evidence based on electron microscopy, serological detection, host range, symptoms, and the entire nucleotide sequence of RNA3.  相似文献

ABSTRACT The occurrence of more than one virus species in a single plant is not uncommon in cultivated and native plant species. A mixed virus infection may lead to greater disease severity than individual viral components and this is sometimes referred to as a synergistic disease. Although, in some cases, synergism has been demonstrated for various plant growth parameters such as plant height, weight, and yield, proof of synergy typically has not been demonstrated for symptom severity when the mixed virus infection was not lethal. We demonstrated synergy in bell pepper plants co-infected with Cucumber mosaic virus (CMV) and Pepper mottle virus (PepMoV) relative to each virus alone for stem height (two of three trials) and aboveground fresh weight (one of three trials) using factorial analysis and Abbott's equation for synergy. This approach allowed affirmation of the type of response (i.e., synergistic rather than antagonistic) and statistical proof of synergy. A detailed evaluation of symptom severity for each viral treatment revealed three phases associated with host plant developmental stages. A numerical symptom severity rating scale was developed and used in each of two equations to demonstrate statistical proof for synergy based on symptom severity for co-infected plants. Virus accumulation in noninoculated leaves was determined by enzyme-linked immunosorbent assay. In singly infected plants, CMV titers declined in mildly symptomatic leaves representing later stages of plant development, but titers increased in similar leaves of co-infected plants. In contrast, PepMoV titers did not differ in singly or co-infected plants.  相似文献

从广西南宁市郊温室大棚中的葫芦[Lagenaria siceraria(Molina)Stand.]上采集到一个表现脉绿、花叶症状的病毒样品,ELISA检测表明,该样品与黄瓜绿斑驳花叶病毒(Cucumber green mottle mosaic virus,CGMMV)有密切的血清学关系,利用RT-PCR方法从样品中扩增获得约500bp的DNA片段,序列分析表明,该片段是CGMMV的外壳蛋白基因,暂将该病毒分离物定名为GX-BG。外壳蛋白基因核苷酸序列系统进化树分析表明,已报道的CGMMV主要分为3大群体,GX-BG与中国辽宁分离物(CGMMV-LN)分别属于不同的群体。  相似文献

应用MNP-RT-PCR方法检测黄瓜绿斑驳花叶病毒   总被引:4,自引:0,他引:4  
 A novel RT-PCR method integrated with Magnetic Nano Particles (MNP), MNP-RT-PCR, was set up for detection of Cucumber green mottle mosaic virus (CGMMV). After the virus particles in crude sap were concentrated by MNP, viral RNAs were released and were detected by RT-PCR. CGMMV could be detected in as less as 10 ng watermelon leaf materials. Compared with normal RT-PCR, the method decreased the inhibitors of plant material and steps for extracting RNA, and also increased the sensitivity of RT-PCR detection in less time. The method is simple and suitable for quick detection of plant virus in a large number of samples.  相似文献

在昆明地区栽培的香石竹上发现了香石竹斑驳病毒,人工接种可局部侵染苋色藜、墙生藜、千日红及番杏,可系统侵染昆诺阿藜和美国石竹,分别造成褪绿斑、斑驳或坏死斑。经二轮PEG(分子量6000)沉淀,病毒提纯量约220mg/kg鲜组织。电镜观察病毒颗粒直径为28nm,等轴对称。提纯物紫外扫描呈典型病毒核蛋白吸收峰,A260/A280值为1.56。提纯病毒经4次免疫家兔,所得抗血清经试管沉淀测定效价为1∶4096,琼脂糖双扩散效价为1∶1024。  相似文献

1985—1987年研究表明北京地区花生上流行的病害主要有黄瓜花叶病毒CA株系(CMV-CA)引起的黄花叶病害和花生轻斑驳病毒(PMMV)引起的轻斑驳病害。血清鉴定451份花生病害样品,黄花叶病害样品受CMV-CA侵染占95.7%,轻斑驳病害样品受PMMV侵染占91.3%,中后期病害样品受CMV-CA和PMMV复合感染较普遍。CMV种传率2%左右,种传病苗在花生出苗后开始出现,6月病害进入高峰。三年CMV-CA流行程度明显不同,主要取决于苗期蚜虫发生早晚、发生量及苗期降雨量。PMMV种传率低于1%,病苗出现稍晚,但病害扩散快,6月上旬进入高峰,7月上、中旬发病率达100%。三年流行程度差异不明显。  相似文献

Twenty-four isolates of Chilli veinal mottle virus (ChiVMV) from China, India, Indonesia, Taiwan and Thailand were analysed to determine their genetic relatedness. Pathogenicity of virus isolates was confirmed by induction of systemic mosaic and/or necrotic ringspot symptoms on Capsicum annuum after mechanical inoculation. The 3' terminal sequences of the viral genomic RNA were determined. The coat protein (CP) coding regions ranged from 858 to 864 nucleotides and the 3' untranslated regions (3'UTR) from 275 to 289 nucleotides in length. All isolates had the inverted repeat sequence GUGGNNNCCAC in the 3'UTR. The DAG motif, conserved in aphid-transmitted potyviruses, was observed in all isolates. All 24 isolates were considered as belonging to ChiVMV because of their high CP amino acid and nucleotide identity (more than 94·8 and 89·5%, respectively) with the reported ChiVMV isolates including the pepper vein banding virus (PVBV), the chilli vein-banding mottle virus (CVbMV) and the CVbMV Chiengmai isolate (CVbMV-CM1). Based on phylogenetic analysis, ChiVMV isolates including all 24 isolates tested, PVBV, CVbMV and CVbMV-CM1 can be classified into three groups. In addition, a conserved region of 204 amino acids with more than 90·2% identity was identified in the C terminal of the CP gene of ChiVMV and Pepper veinal mottle virus (PVMV), and may explain the serological cross reaction between these two viruses. The conserved region may also provide useful information for developing transgenic resistance to both ChiVMV and PVMV.  相似文献

2016年9月贵州省贵阳市发生严重的辣椒病毒病,症状复杂,主要表现为植株矮化,叶片黄化、花叶、皱缩、畸形以及枯死斑,果实有坏死斑等,根据症状难以判断病毒种类。本文采用小RNA深度测序技术对田间自然发病的2株辣椒标样进行了毒源鉴定,发现样品1由蚕豆萎蔫病毒2号(Broad bean wilt virus2,BBWV2)、辣椒脉斑驳病毒(Chilli veinal mottle virus,ChiVMV)、黄瓜花叶病毒(Cucumber mosaic virus,CMV)和辣椒内源RNA病毒(Bell pepper endornavirus,BPEV)4种病原复合侵染;样品2中除鉴定到上述4种病毒外,还检测到马铃薯Y病毒(Potato virusY,PVY)。进一步通过反转录PCR(RT-PCR)对深度测序结果进行了验证,证明其准确可靠。其中4个辣椒标样中均有的辣椒内源RNA病毒(BPEV)为贵州省首次报道。多种病毒复合侵染是辣椒产量和品质的重要限制因素之一,是辣椒生产的主要威胁。  相似文献
