Fusarium head blight of cereals has, in recent years, become one of the most important pre-harvest diseases worldwide. This paper examines the in vitro efficacy of fungicides to control Fusarium species in cereals and the efficacy in the field on both Fusarium infection of ripening ears as well as their impact on mycotoxin production. Field studies suggest that fungicides such as tebuconazole and metconazole give good control of both Fusarium infection of ears and control of deoxynivalenol (DON) production. However, azoxystrobin and related fungicides are less effective, and grain from treated crops has sometimes been found to have increased concentrations of DON and nivalenol. Studies of isolates of Fusarium culmorum from different parts of Europe showed that complex interactions occur between environmental factors, fungicide type and isolate in relation to growth inhibition and DON production. These studies confirmed the ineffectiveness of azoxystrobin and suggest that environmental stress factors, particularly water availability and temperature, and low fungicide doses may stimulate mycotoxin production by Fusaria in vitro and in wheat grain.  相似文献

Fusarium head blight of wheat is caused by a disease complex comprised of toxigenic pathogens, predominantly Fusarium spp., and a non-toxigenic pathogen Microdochium nivale, which causes symptoms visually indistinguishable from Fusarium and is often included as a causal agent of Fusarium head blight. Four field trials are reported here, including both naturally and artificially inoculated trials in which the effect of fungicide treatments were noted on colonisation by Fusarium and Microdochium, and on the production of deoxynivalenol (DON) mycotoxin. The pathogen populations were analysed with quantitative PCR and samples were tested for the presence of the mycotoxin DON. Application of fungicides to reduce Fusarium head blight gave a differential control of these fungi. Tebuconazole selectively controlled F. culmorum and F. avenaceum and reduced levels of DON, but showed little control of M. nivale. Application of azoxystrobin, however, selectively controlled M. nivale and allowed greater colonisation by toxigenic Fusarium species. This treatment also lead to increased levels of DON detected. nobreak Azoxystrobin application two days post-inoculation increased the production of DON mycotoxin per unit of pathogen in an artificially inoculated field trial. This result indicates the potential risk of increased DON contamination of grain following treatment with azoxystrobin to control head blight in susceptible wheat cultivars. This is the first study to show differential fungicidal control of mixed natural pathogen populations and artificial inoculations in field trials.  相似文献

Glasshouse studies were undertaken to determine if fungicides used for the control of Fusarium head blight (FHB) result in elevated concentrations of the trichothecene mycotoxin, deoxynivalenol (DON) in harvested wheat grain. Metconazole and azoxystrobin, at double, full, half or quarter the manufacturer's recommended dose rate, were applied to ears of wheat (cv. Cadenza), artificially inoculated with conidia of either Fusarium culmorum or F. graminearum. Metconazole demonstrated high activity against both pathogens, reducing significantly the severity of FHB and the DON concentrations at each of the four dose rates tested when compared to untreated controls. Applications of azoxystrobin significantly reduced FHB and DON compared to unsprayed controls. However, their effectiveness was significantly less than that of metconazole and no dose rate response was observed. Quantification of the amount of trichothecene-producing Fusarium present in harvested grain was determined using a competitive PCR assay based on primers derived from the trichodiene synthase gene (Tri5). Simple linear regression analyses revealed strong relationships between the amount of trichothecene-producing Fusarium present in grain and the DON concentrations (r 2=0.72–0.97). It is concluded that fungicides, applied for the control of FHB, affect DON concentrations indirectly by influencing the amount of trichothecene-producing Fusarium species present in wheat grain. There was no evidence that fungicide applications directly increase the concentration of DON in grain.  相似文献

恶苗病是水稻生产上较为严重的种传真菌病害,咪唑类广谱内吸性杀菌剂咪鲜胺是目前防治该病害的主要药剂。以对咪鲜胺抗性及敏感的田间水稻恶苗病菌为试材,研究了其适合度及对几种常用杀菌剂的交互抗性。结果显示:抗性菌株的抗药性可稳定遗传,其温度敏感性与敏感菌株无明显差异,部分抗性菌株在菌丝生长速率、产孢量、孢子萌发率和致病力方面显著高于田间敏感菌株;咪鲜胺与三唑类及2-氰基丙烯酸酯类杀菌剂之间均无交互抗性。研究表明,对咪鲜胺产生抗性的水稻恶苗病菌具有较强的适合度,在田间自然条件下有可能形成优势群体,因此需合理轮换使用不同作用机制的杀菌剂,以延缓其抗药性的发展。  相似文献

Fusarium head blight (FHB), a devastating disease that affects wheat, is caused by a complex of Fusarium species. The overall impact of Fusarium spp. in wheat production arises through the combination of FHB and mycotoxin infection of the grain harvested from infected wheat spikes. Spike infection occurs during opening of flowers and is favoured by high humidity or wet weather accompanied with warm temperatures. Available possibilities for controlling FHB include the use of cultural practices, fungicides and biological approaches. Three cultural practices are expected to be of prime importance in controlling FHB and the production of mycotoxins: soil preparation method (deep tillage), the choice of the preceding crop in the rotation and the selection of appropriate cultivar.  相似文献

Determination of the Fusarium protein equivalent (FPE) levels in kernels for better characterisation of genotypes showing Fusarium head blight (FHB) resistance, and better detection of susceptibility to kernel infection among genotypes with slight symptom expression was carried out. Twelve wheat cultivars and eight hexaploid winter wheat lines derived from a cross of Triticum aestivum with related species T. macha, T. polonicum, and T. dicoccoides were evaluated for levels of spike and kernel infection, the content of the mycotoxin deoxynivalenol (DON) and FPE in kernels after artificial inoculation with the fungus Fusarium culmorum in the field in 2006–2007. The ELISA immunochemical method was employed for the quantitative analyses of DON and FPE. Three wheat lines had a significantly low infection of spikes and kernels compared to cvs Sumai 3 and Nobeoka Bozu, indicating the presence of specific resistance mechanisms to FHB. The significantly low AUDPC (area under the disease progress curve) and the high level of FPE and DON content in kernels indicated a lack of resistance in one wheat line (crossed with T. polonicum). The results showed highly significant correlations (P < 0.01) between FPE and DON content and between FPE and AUDPC. In addition, correlations between FPE and reductions in yield components were also highly significant. Quantification of Fusarium spp. in wheat kernels can be helpful for evaluating wheat genotypes for their levels of resistance to FHB.  相似文献

Details of our long-term research programme concerning the epidemiology of Fusarium spp. and mycotoxin production are summarized. Evaluation of the occurrence of Fusarium spp., mainly on winter wheat (Triticum aestivum), was carried out by investigating Fusarium infection and mycotoxin contamination. Two to 15% of grains were infested during 1995–1998 at three climatologically differing localities of the Rhineland, Germany. Disease progress was accelerated by rainfall during the flowering season. The species most frequently isolated were Fusarium avenaceum, F. poae, F. culmorum and F. graminearum. The mean deoxynivalenol (DON) content varied from 19gkg–1 (1995) to 310gkg–1 (1998) and was not always correlated with disease severity. Organic farming systems showed lower rates of infection with ear blight and lower mycotoxin contamination than conventional farming systems.  相似文献

Fungicide resistance in plant pathogens is often caused by a single point mutation in a gene encoding fungicide target proteins. Such is the case for resistance to MBI-D (inhibitors of scytalone dehydratase in melanin biosynthesis) fungicides in rice blast fungus (Magnaporthe oryzae), which is caused by a mutation in the scytalone dehydratase gene that results in a replacement of valine with methionine at codon 75 of the fungicide target protein. PCR-Luminex, a novel system developed for high-throughput analysis of single nucleotide polymorphisms (SNPs) was successfully introduced to diagnose MBI-D resistance using specific oligonucleotide probes coupled with fluorescent beads. The PCR-Luminex system was further tested for its potential in identifying species causing Fusarium head blight on wheat. Four major pathogens, Fusarium graminearum (=F. asiaticum), F. culmorum, F. avenaceum, and Microdochium nivale, known to cause the disease, were tested, and the species were identified using the PCR-Luminex method. So far, this report is the first on the application of the DNA-based PCR-Luminex system in the area of crop protection and/or agricultural sciences.  相似文献

BACKGROUND: It is possible that a single nucleotide polymorphism (SNP) (G143A mutation) in the cytochrome b gene could confer resistance to quinone outside inhibiting (QoI) fungicides (strobilurins) in rice blast fungus because this mutation caused a high level of resistance to fungicides such as azoxystrobin in Pyricularia grisea Sacc. and other fungal plant pathogens. The aim of this study was to survey Magnaporthe oryzae B Couch sp. nov. isolates in Japan for resistance to QoIs, and to try to develop molecular detection methods for QoI resistance. RESULTS: A survey on the QoI resistance among M. oryzae isolates from rice was conducted in Japan. A total of 813 single‐spore isolates of M. oryzae were tested for their sensitivity to azoxystrobin using a mycelial growth test on PDA. QoI fungicide resistance was not found among these isolates. The introduction of G143A mutation into a plasmid containing the cytochrome b gene sequence of rice blast fungus was achieved by site‐directed mutagenesis. Molecular diagnostic methods were developed for identifying QoI resistance in rice blast fungus using the plasmid construct. CONCLUSION: As the management of rice blast disease is often dependent on chemicals, the rational design of control programmes requires a proper understanding of the fungicide resistance phenomenon in field populations of the pathogen. Mutation of the cytochrome b gene of rice blast fungus would be specifically detected from diseased leaves and seeds using the molecular methods developed in this study. 为评价牛至精油及其2种主要成分香芹酚和百里酚在植物真菌病害防治方面的应用前景,采用菌丝生长速率法和孢子萌发法测定3种物质对16种植物病原真菌菌丝生长和孢子萌发的抑制活性,并采用黄瓜子叶法测定其对黄瓜灰霉病的防治效果。结果表明,牛至精油在500 mg/L浓度处理下可完全抑制番茄早疫病菌Alternaria solani、黄瓜灰霉病菌Botrytis cinerea和小麦根腐病菌Bipolaris sorokiniana等12种植物病原真菌的菌丝生长,对棉花黄萎病菌Verticillium dahliae的菌丝生长抑制率也达到97.66%。牛至精油对这13种病原真菌的EC50在83.09~236.58 mg/L之间,香芹酚和百里酚对这13种病原真菌的毒力均高于牛至精油,其中香芹酚对黄瓜灰霉病菌、油菜菌核病菌Sclerotinia sclerotiorum、小麦全蚀病菌Gaeumannomyces graminis、棉花黄萎病菌和小麦茎基腐病菌Fusarium pseudograminearum菌丝生长的毒力较强,EC50分别为9.09、22.45、24.28、28.40和28.80 mg/L;百里酚对黄瓜灰霉病菌和小麦全蚀病菌菌丝生长的毒力较强,EC50分别为21.32 mg/L和27.08 mg/L。香芹酚和百里酚对棉花黄萎病菌、玉米大斑病菌Setosphaeria turcica、苦瓜枯萎病菌Fusarium oxysporum f.sp.momordicae、黄瓜灰霉病菌和黄瓜炭疽病菌Colletotrichum orbiculare的孢子萌发亦有较强毒力,其EC50为3.78~289.07 mg/L。1 000 mg/L牛至精油对黄瓜灰霉病的预防效果与400 mg/L嘧霉胺悬浮剂的预防效果无显著差异;500 mg/L百里酚对黄瓜灰霉病的预防效果和治疗效果均与400 mg/L嘧霉胺悬浮剂的效果相当,而香芹酚的治疗效果显著优于400 mg/L嘧霉胺悬浮剂。表明牛至精油及其主要成分香芹酚和百里酚的抑菌谱广且活性强,具有作为植物源杀菌剂的开发应用潜力。  相似文献

Saprophytic Fusarium strains, isolated from Fusarium-suppressive soils, have been tested in pot and glasshouse experiments in order to reduce soil colonization by Fusarium oxysporum f.sp. dianthi. The highest amount of antagonist inoculum tested (200 g m-2) showed the best results in controlling carnation wilt. The antagonistic Fusarium strains showed good efficacy when applied by dipping rooted cuttings in conidial suspensions of the antagonists or by mixing inoculated wheat kernels or perlite + wheat bran with the soil. The effectiveness of the antagonistic Fusarium strains in reducing incidence of F.o. f.sp. dianthi was improved by combining chemical (benomyl, 20 g m-2) with biological measures.  相似文献

The broad-spectrum fungicide CGA 219417 inhibits mycelial growth of Botrytis cinerea Pers ex Fr. Pseudocercosporella herpotrichoides (Fron.) Deighton and Helminthosporium oryzae B. de Haan on defined media lacking amino acids. The growth inhibition of B. cinerea is reversed by the addition of a mixture of 19 amino acids at a concentration of 100 μM each or by the addition of methionine or homocysteine in concentrations of 100 μM or 1 mM, respectively. In the case of B. cinerea, the reversal of growth inhibition by methionine could also be shown for pyrimethanil and mepanipyrim. These findings suggest that the pyrimidinamine fungicides inhibit the biosynthesis of methionine in phytopathogenic fungi.  相似文献

Strobilurins, broad-spectrum fungicides used to control various diseases, may also inhibit the growth of nontarget fungi in the natural environment. Acibenzolar-S-methyl (ASM) is a plant activator; however, the effect of ASM on the growth of microflora on the leaf surface is not clear yet. ASM, strobilurins and other fungicides were sprayed on Japanese pear and cucumber plants in the field. Variations in the morphological characteristics and rDNA-ITS nucleotide sequence of isolated phylloplane fungi were evaluated. The isolation frequency of Alternaria spp., Aureobasidium spp., Cladosporium spp. and Myrothecium spp. on Japanese pear leaves treated with kresoxim-methyl and polycarbamate was lower than on nontreated pear leaves. Moreover, the frequency of Alternaria spp., Cladosporium spp., Epicoccum spp. and Fusarium spp. on leaves of two cucumber cultivars treated with azoxystrobin, triflumizole and mancozeb was lower than the frequency of fungal isolation on nontreated cucumber leaves. The effect of ASM on fungal isolation frequency was not significantly different from that on nontreated pear or cucumber leaves. The fungal genera observed and isolation frequency on the leaves treated with polycarbamate, azoxystrobin, triflumizole and mancozeb tended to be reduced compared to the no-spray treatment.  相似文献

BACKGROUND: Phoma stem canker, caused by the coexisting related fungal pathogens Leptosphaeria maculans (Des.) Ces. & de Not and L. biglobosa Shoemaker & H Brun, is a major disease of winter oilseed rape in the UK. Annually, over 90% of UK crops receive at least one foliar application of fungicide, but little is known about the sensitivity of the more damaging L. maculans and the less damaging L. biglobosa to these fungicides. The effects of flusilazole, tebuconazole and Methyl Benzimidazole Carbamate (MBC) fungicides (benomyl and carbendazim) on the germination of ascospores, conidia and germ tube growth of both species were examined. Isolates collected from different oilseed rape crops in England and Wales were assessed for their mycelial growth on fungicide‐amended medium, and ED50 values were calculated. RESULTS: Leptosphaeria maculans and L. biglobosa differed in their sensitivity to fungicides. Conidial germination of L. maculans was more sensitive to these fungicides than that of L. biglobosa. Isolates of L. maculans had smaller ED50 values for mycelial growth for all fungicides tested than isolates of L. biglobosa. CONCLUSION: These results suggest that fungicide applications might affect the structure of L. maculans/L. biglobosa populations in UK winter oilseed rape crops. The spatial pattern of Fusarium‐infected kernels and their mycotoxin contamination was studied in four wheat fields in Germany using geo‐referenced sampling grids (12–15 × 20–30 m, 28–30 samples per field) at harvest. For each sample, frequency of Fusarium‐infected kernels and spectrum of species were assessed microbiologically; mycotoxin contents were determined by HPLC‐MS/MS analysis. Spatial variability of pathogens and mycotoxins was analysed using various parameters including Spatial Analysis by Distance IndicEs (sadie® ). Microdochium majus, the most frequent head blight pathogen in 1998, was less frequent in 1999 and could not be detected in kernels from two fields in 2004. Fusarium avenaceum, F. graminearum and F. poae were the most frequent Fusarium species, with 7–8 species per field. The frequency of Fusarium‐infected kernels was 3–15% and the incidence of species showed considerable within‐field variability. Spatial patterns varied among Fusarium species as well as from field to field. Although pathogens and mycotoxin were often distributed randomly in the field, F. avenaceum, F. graminearum, F. poae, F. sporotrichioides, F. tricinctum and the mycotoxin moniliformin had an aggregated pattern in at least one field. Patterns are discussed in relation to spread of Fusarium species depending on inoculum sources, spore type, kind of dispersal, availability of susceptible host tissue and micro‐climate. Sampling of wheat fields for representative assessment of mycotoxins is complicated by random patterns of Fusarium‐infected kernels, especially where the frequency of infection is small.  相似文献

Roots of seedlings of wheat and barley affected by bare patch disease at a field site in Western Australia were assessed for root damage and plated to isolate fungi. The patches were variable in shape and size and had the most severely affected plants in the centre. Of the 165 isolates ofRhizoctonia spp. obtained, 90% were multinucleate and 10% binucleate, the former being predominant in the plants at the centre of the patch. The relative frequency of binucleate isolates increased with proximity to the periphery. The increase in activity of avirulent binucleate isolates towards the periphery of the patch may be related to the sharp and abrupt edging of the patch. A variety of other species of fungi such asFusarium spp.,Mortierella spp.,Bipolaris sorokiniana, Pythium sp. andTrichoderma sp. were encountered within the patches. The multinucleate isolates belonging to anastomosis groups (Ag) 2–1, 2–2 and 8 (Thanatephorus cucumeris) were most pathogenic to wheat. The binucleate isolates of Ag C, D, E, and K (Ceratobasidium sp.) were less pathogenic. It is suggested that the bare patch disease is caused by a complex of root rot fungi composed of one or more anastomosis groups ofRhizoctonia spp. and other associated fungi.Samenvatting Van kiemplanten van tarwe en gerst, afkomstig van een met kale-plekkenziekte besmet perceel in West Australië werd de mate van wortelbeschadiging bepaald en werden schimmels uit de wortels geïsoleerd. High year-to-year variability in the incidence of Fusarium spp. and mycotoxin contamination was observed in a two-year survey investigating the impact of maize ear rot in 84 field samples from Germany. Fusarium verticillioides, F. graminearum, and F. proliferatum were the predominant species infecting maize kernels in 2006, whereas in 2007 the most frequently isolated species were F. graminearum, F. cerealis and F. subglutinans. Fourteen Fusarium-related mycotoxins were detected as contaminants of maize kernels analyzed by a multi-mycotoxin determination method. In 2006, a growth season characterized by high temperature and low rainfall during anthesis and early grain filling, 75% of the maize samples were contaminated with deoxynivalenol, 34% with fumonisins and 27% with zearalenone. In 2007, characterized by moderate temperatures and frequent rainfall during the entire growth season, none of the 40 maize samples had quantifiable levels of fumonisins while deoxynivalenol and zearalenone were detected in 90% and 93% of the fields, respectively. In addition, 3-acetyldeoxynivalenol, 15-acetyldeoxnivalenol, moniliformin, beauvericin, nivalenol and enniatin B were detected as common contaminants produced in both growing seasons. The results demonstrate a significant mycotoxin contamination associated with maize ear rots in Germany and indicate, with regard to anticipated climate change, that fumonisins-producing species already present in German maize production may become more important.  相似文献

Combined analyses of the natural occurrence of fusarium head blight (FHB), mycotoxins and mycotoxin‐producing isolates of Fusarium spp. in fields of wheat revealed FHB epidemics in 12 of 14 regions in Hubei in 2009. Mycotoxin contamination ranged from 0·59 to 15·28 μg g?1 in grains. Of the causal agents associated with symptoms of FHB, 84% were Fusarium asiaticum and 9·5% were Fusarium graminearum, while the remaining 6·5% were other Fusarium species. Genetic chemotyping demonstrated that F. asiaticum comprised deoxynivalenol (DON), 3‐acetyldeoxynivalenol (3‐AcDON), 15‐acetyldeoxynivalenol (15‐AcDON) and nivalenol (NIV) producers, whereas F. graminearum only included DON and 15‐AcDON producers. Compared with the chemotype patterns in 1999, there appeared to be a modest shift towards 3‐AcDON chemotypes in field populations during the following decade. However, isolates genetically chemotyped as 3‐AcDON were present in all regions, whereas the chemical 3‐AcDON was only detected in three of the 14 regions where 3‐AcDON accounted for 15–20% of the DON and acetylated forms. NIV mycotoxins were detected in seven regions, six of which also yielded NIV chemotypes. The number of genetic 3‐AcDON producers was positively correlated with amounts of total mycotoxins (DON, NIV and acetylated forms) or DON in wheat grains. Chemical analyses of wheat grains and rice cultures inoculated with different isolates from the fields confirmed their genetic chemotypes and revealed a preferential biosynthesis of 3‐AcDON and 4‐AcNIV in rice. These findings suggest the importance of chemotyping coupled with species identification for improved prediction of mycotoxin contamination in wheat.  相似文献

A ring spot disease of Aloe vera was found on leaves of potted seedlings of Aloe vera in Hachijojima and Chichijima Islands, Tokyo. From tissue of ring spot lesions, a fungus producing Fusarium-type conidia was consistently isolated. After 1 month, reddish perithecia of nectriaceous fungus had formed on the colonies of this isolate on PDA. These nectriaceous and Fusarium fungi were identified as Haematonectria haematococca and Fusarium sp., respectively. From a single ascospore isolation, the former was confirmed to be the teleomorph of the Fusarium sp. Typical ring spot lesions were reproduced by artificial inoculations using single ascospore and single conidium isolates. Inoculations of five species of genus Aloe revealed that they were highly susceptible except for A. arborescens. This is the first report of a disease on Aloe caused by H. haematococca (anamorph: Fusarium sp.) in Japan, and it was named aloe ring spot.  相似文献

In 1993 we observed the sensitivity of wheat powdery mildew populations (Erysiphe graminis DC f.sp. tritici Marchal) from the Czech Republic, Austria, Hungary and Slovakia to the fungicides triadimenol, tebuconazole, propiconazole, flutriafol and fenpropimorph. The highest resistance value was shown to triadimenol, which attained a mean resistance factor (MRF) of 29 (expressing how many times the population is more resistant than are standard sensitive isolates) in the mildew population from the Czech Republic. The mildew populations from eastern Slovakia and eastern Hungary, populations geographically isolated from the other populations, showed very high sensitivity to all fungicides tested. There was most sensitivity to fenpropimorph (smallest MRF values) compared with the other fungicides. Cross-resistance was established among all triazoles used, but not between triazoles and fenpropimorph. Sensitivity of wheat powdery mildew populations from Central Europe to these fungicides is considered adequate, and the development of resistance has shown a decreasing tendency in recent years.  相似文献
