Culliton discusses what she terms the "unprecedented" public scrutiny to which the technology of gene therapy and its potential application in humans have been subjected. She summarizes the federal government's interest in this area, ranging from the Office of Technology Assessment's report, Human Gene Therapy, to the creation of the Working Group on Human Gene Therapy. The latter, a subcommittee of the National Institutes of Health's Recombinant DNA Advisory Committee, released a list of "points to consider" by researchers planning to design and submit protocols to NIH for gene therapy research in humans. Culliton notes that some of the questions that the working group asks potential researchers "go beyond the usual" to address controversial areas of gene therapy such as primate studies, public health considerations, public relations, patents, and eugenics. 相似文献

Over 400 academic and corporate leaders attended a gathering, "Partners in the Research Enterprise: A National Conference on University-Corporate Relations in Science and Technology," held 14-16 Dec 1982 at the University of Pennsylvania. Problems explored included the protection of academic freedom and values in collaborative research, the sums of money involved, faculty roles, and patents. Representative Albert Gore, previewing his House subcommittee's report, called for a national guideline-writing conference. The Working Group on Human Gene Therapy, a subcommittee of the NIH Recombinant DNA Advisory Committee (RAC), has revised its draft guidelines, published in the 22 January 1985 Federal Register, for researchers submitting protocols for human gene therapy experiments. The major revisions in the "Points to Consider" are the elimination of a required response in the protocol to complex social and ethical questions and a greater flexibility in requirements for animal testing prior to human experimentation. Other modifications include provisions for public review of protocols, a requirement of patient agreement to long term follow-up and autopsy, and the limiting of review to only somatic cell therapy for the present. The stages of protocol review will involve local ethics and biosafety committees, then the Working Group, the full RAC, and finally the director of NIH. 相似文献

The National Institutes of Health has recommended that John Darsee be denied NIH funding for ten years and that Eugene Braunwald's cardiac research laboratory at Harvard, where Darsee worked, be subject to an on-site review in one year. These recommendations follow the report of an NIH-appointed panel which investigated data falsification by Darsee and supervisory procedures at the laboratory. We investigated the association between a U.S. National Institutes of Health (NIH) R01 applicant's self-identified race or ethnicity and the probability of receiving an award by using data from the NIH IMPAC II grant database, the Thomson Reuters Web of Science, and other sources. Although proposals with strong priority scores were equally likely to be funded regardless of race, we find that Asians are 4 percentage points and black or African-American applicants are 13 percentage points less likely to receive NIH investigator-initiated research funding compared with whites. After controlling for the applicant's educational background, country of origin, training, previous research awards, publication record, and employer characteristics, we find that black applicants remain 10 percentage points less likely than whites to be awarded NIH research funding. Our results suggest some leverage points for policy intervention. 相似文献

The laboratory of which the National Institutes of Health (NIH) is a lineal descendant was founded in 1887. During discussions of our plans for a year-long centennial observance with members of our House and Senate appropriations subcommittees, congressional members urged us to set two specific objectives: making NIH better known to the American people, and presenting the attractions of the many roles in health-related research to young people who have not yet formulated career plans. When I was invited to prepare an article for Science dealing with my personal experiences as director of NIH since April 1982, it seemed an opportunity to address the same objectives for the scientific community, for in my view there is much misinformation and far too much pessimism throughout the country about the state of biomedical research and its support. 相似文献

The CML-specific P210 bcr/abl protein, unlike v-abl, does not transform NIH/3T3 fibroblasts 总被引：32，自引：0，他引：32
The v-abl oncogene of the Abelson murine leukemia virus (A-MuLV) is known to efficiently transform NIH/3T3 fibroblasts in vitro and to cause an acute lymphosarcoma in susceptible murine hosts. The role of its relative, the bcr/abl gene product, in the etiology of human chronic myelogenous leukemia (CML) remains speculative. To assess the transforming properties of the bcr/abl gene product, complementary DNA clones encoding the CML-specific P210 bcr/abl protein were expressed in NIH/3T3 fibroblasts. In contrast to the v-abl oncogene product P160, the P210 bcr/abl gene product did not transform NIH/3T3 cells. Cell lines were isolated that expressed high levels of the P210 bcr/abl protein but were morphologically normal. During the course of these experiments, a transforming recombinant of bcr/abl was isolated which fuses gag determinants derived from helper virus to the NH2-terminus of the bcr/abl protein. This suggests that a property of viral gag sequences, probably myristylation-dependent membrane localization, must be provided to bcr/abl for it to transform fibroblasts. 相似文献

Scientists may also work with embryonic stem cells, but may obtain them only from private sources and must ensure that derivation meets certain ethical conditions. 用高抗枯萎病亚麻品种“晋亚 7号”与高感枯萎病品种“晋亚 1号”配制杂交组合 ,接种鉴定其正反交F1代以及F2 代分离群体的枯萎病发生情况 ,结果表明 ,晋亚 7号对枯萎病的抗性属于细胞核遗传 ,受 2个显性基因控制。用 4 8个EcoRI/MseI引物组合对“晋亚 7号”、“晋亚 1号”两个亲本及其F2 代抗病和感病基因池进行AFLP分析 ,共扩增出约 330 0条可分辨的带 ,其中 3条为稳定的差异。用“晋亚 7号”和“晋亚 1号”杂交产生的F2 代分离群体对 3个特异条带与目的基因的遗传连锁性进行分析 ,发现特异条带AG/CAG与暂定名为FuJ7(t)的抗枯萎病基因紧密连锁 ,二者之间的遗传距离为 5 .2cM。将AG/CAG片段回收、克隆和测序 ,成功地将其转化为SCAR标记 ,可以更加方便地用于对FuJ7(t)基因的分子检测和标记辅助选择。 相似文献

[目的/意义]文章基于全球“海洋水产饲料”领域的专利数据,通过对比分析世界各区域在该领域的技术发展特征,揭示中国在“海洋水产饲料”领域实施技术创新面临的国际形势和存在的问题。[方法/过程]采用专利文献计量法分析了国际“海洋水产饲料”领域的专利发展态势、布局趋势、专利竞争力、专利权人特征、专利技术发展特征等。[结果/结论]国际“海洋水产饲料”的专利技术正在向新兴市场转移,精细化、突破性的技术研发是未来发展的方向。近10年中国大陆的专利技术呈现出强劲的增长势头,但中国专利布局及市场流入在国际上的发展状况并不乐观,而且存在专利的规模增长与质量提高不协调、专利维护及合作力度偏低、专利市场应用度不足等问题。未来应从深化政策引导支持、跟踪国际技术及市场动向、加强“产学研”合作与专利质量提升等方面出发,不断推动“海洋水产饲料”技术的高质量发展与应用。 相似文献

The epidermal growth factor (EGF) receptor gene EGFR has been placed in a retrovirus vector to examine the growth properties of cells that experimentally overproduce a full-length EGF receptor. NIH 3T3 cells transfected with the viral DNA or infected with the corresponding rescued retrovirus developed a fully transformed phenotype in vitro that required both functional EGFR expression and the presence of EGF in the growth medium. Cells expressing 4 x 10(5) EGF receptors formed tumors in nude mice, while control cells did not. Therefore, the EGFR retrovirus, which had a titer on NIH 3T3 cells that was greater than 10(7) focus-forming units per milliliter, can efficiently transfer and express this gene, and increased numbers of EGF receptors can contribute to the transformed phenotype. 相似文献

根据牛病毒性腹泻病毒(BVDV)5′-UTR保守序列建立的特异性荧光RT-PCR,对采自福州市周边屠宰场的359份屠宰猪血清进行BVDV感染的病原学检测。结果待检的359份血清样品BVDV阳性52份,阳性率为14.5%(52/359)。对其中13份BVDV阳性样品5′端UTR片段进行序列测定分析,发现所测13份BVDV阳性样品5′端UTR片段均与VEDEVAC进化谱系较为密切,均为基因Ⅰ型。以上结果表明以牛源BVDV建立的特异性荧光RT-PCR方法适用于猪源BVDV感染的监测,并揭示福州市屠宰猪存在BVDV感染。 相似文献
