A 12‐week feeding trial was conducted using Nile tilapia, Oreochromis niloticus (L.) to evaluate the interactive effects of fishmeal replacement and salinity on growth, feed utilization efficiencies and relative expression of growth related genes. Two iso‐nitrogenous and iso‐energetic diets were prepared (32% protein). The control diet included 15% fishmeal (FM diet) and fishmeal component in non‐fishmeal diet (NFM) was eliminated by a mixture of poultry by‐product meal, high protein distillers dried grains and distillers dried grains with soluble. The NFM diet was supplemented with DL‐methionine and L‐lysine. Duplicated group of fish with initial mean weight of 6 g, reared in four salinity levels (0, 4, 8 and 12 g/L) were fed one of the two diets twice a day to near satiety. At the end of the experiment, growth, feed utilization efficiency and expression of growth related genes were compared. The specific growth rate (SGR), mean feed intake (MFI) and feed conversion ratio (FCR) were not affected by the diets while salinity effects were significant. The fish in the 4 g/L salinity showed the highest SGR and MFI while fish in the 0 g/L treatment showed the lowest FCR. Relative expression of hepatic IGF‐I and IGF‐II was regulated by salinity but not by the diet. Expression of growth hormone receptor gene was not affected by either diet or salinity. The present findings provide evidence for the possibility of total fishmeal replacement in saline waters (0–12 g/L) without compromising growth, feed utilization and body composition of Nile tilapia.  相似文献

We assessed effects of feed restriction and inclusion of 200 g kg?1 extracted soybean meal in the diet on gene expression in Atlantic salmon using a cDNA microarray (SFA2.0) and real‐time qPCR. The trial lasted for 54 days. Restricted feeding and soybean inclusion reduced the thermal growth coefficient by respectively 51% and 22% compared with fish fed with the fishmeal‐based control diet to satiation. Soybean decreased distal intestinal expression of lysosomal (cathepsins C, D, L, Y and Z) and extracellular proteases while genes involved in responses to cellular stress were up‐regulated. Expression changes of immune genes suggested both pro‐ and anti‐inflammatory regulation. The hepatic responses to soybean and restricted feeding were highly similar, which could be because of negative effects of soybean meal on digestion and nutrient absorption. We observed up‐regulation of ribosomal proteins and down‐regulation of genes involved in lipid and steroid metabolism. Of note, growth reduction in both study groups was associated with coordinated down‐regulation of genes involved in oxidative and cellular stress responses, metabolism of xenobiotics and protein degradation. High expression of stress‐related genes in salmon fed with the control diet suggests that maximum growth rates can be associated with health problems.  相似文献

Fishmeal could only be replaced by plant proteins at limited levels in aquafeeds, especially for carnivorous fish. In this study, an experiment was designed to evaluate the possibility of improving the utilization of plant proteins by maggot meal supplementation in turbot diet. Five diets were formulated: a reference diet (FM) containing 63% fishmeal and four experimental diets (35(0%), 35(3%), 40(0%), 40(3%)) in which fishmeal was substituted at different levels by plant proteins with 0–3% maggot meal. Turbot (4.90 ± 0.03 g) was fed with these diets for 9 weeks. Fishmeal was successfully replaced by plant proteins in turbot diet without growth reduction at 35% but not 40%. However, maggot supplementation (3%) at 40% plant protein replacement level achieved comparable growth performance with that of fishmeal. Maggot meal supplementation improved apparent digestibility coefficients, plasma hydroxyproline levels, intestine trypsin activities and activated target of rapamycin (TOR) signalling, all of which were decreased or down‐regulated after high plant protein replacement. Therefore, this study demonstrated maggot meal, a potential valuable protein source for turbot.  相似文献

Previously, we reported that methionine intake determined the taurine concentration in the liver of on‐growing Atlantic salmon fed plant protein diets. Further, the methionine intake and/or the increased taurine concentration following increased methionine intake affected the liver lipid metabolism. The following study therefore aimed to test whether taurine affected the growth or the type of growth when added in high plant protein diets naturally low in taurine but equal and adequate in dietary methionine. Juvenile Atlantic salmon [initial body weight (BW) of 2 g] were fed plant protein diets (16.5% fishmeal), which were supplemented with taurine or not for a period of 56 days. As a control for growth and normal metabolism, a fishmeal‐based commercial diet (68% fishmeal) was used. Supplementation with taurine to high plant protein diets had a slightly negative effect on weight gain, but the final body weight was not different. Interestingly, the pool of free amino acids in the liver and muscle was significantly higher in fish fed the supplemented diet as compared with fish fed the plant protein diet without taurine supplementation. Liver polyamine concentration was higher in fish fed the supplemented diet than in fish fed the similar diet without supplementation. Additionally, juvenile salmon fed the plant‐based diet supplemented with taurine had a lower body lipid‐to‐protein ratio due to a reduced whole‐body lipid content, while the whole‐body protein content was similar between treatments. Our study thus indicates that the addition of a low concentration of taurine to high plant protein diets interacts with lipid metabolism and storage, concomitantly affecting the general metabolism as the concentrations of the free amino acids and polyamines in the liver were significantly higher. The possible reasons for these changes are discussed.  相似文献

ABSTRACT:   Expressed sequence tag (EST) analysis is an efficient tool for gene discovery and for profiling gene expression. A cDNA library developed from messenger RNA of the Japanese flounder, Paralichthys olivaceus spleen was constructed by directional cloning, in order to isolate functional genes involved in immunity in fish. A total of 1010 ESTs from the library were sequenced and compared with sequences in the GenBank database. Of the 1010 ESTs, 618 ESTs (61%) were identified as being homologous with known genes from many organisms by BLAST searches, whereas 392 (39%) appeared to be unknown and are likely to represent newly described genes. Of the identified genes, 105 (17%) encoded proteins associated with cell/organism defence and homeostasis. Of these 105 genes, 21 were identified for the first time in Japanese flounder. These included macrophage inflammatory protein (MIP)-3 α, granulin-A, novel immune-type receptor (NITR), interferon regulatory factor (IRF)-10, presenilin-like protein-2, and antizyme inhibitor. A comparison of ESTs derived from spleen, kidney, liver and leukocytes suggested that expression of immune-related genes in different tissues, organs, or cells are different.  相似文献

Fishmeal replacement with a mixture of plant protein (PP) raw materials (soya, wheat gluten meal, corn gluten meal and rapeseed meal) in diets for 300‐950 g turbot was tested. Eight different diets with fishmeal protein stepwise varying from 53.7% of crude protein (CP) to 93% of CP of the total protein in the diet were tested. The fish was weighed at monthly intervals for following of weight development and calculation of specific growth rates, daily feed intake and feed conversion ratio. At the end of the experiment, fish was sampled for sensory evaluation. Average final weight was 950 g and did not vary between the experimental groups. There were no effects of dietary treatment on specific growth rates, daily feed intake, feed conversion ratio or sensory attributes measured. The least‐cost diet (with 53.7% fishmeal protein) is about 12% lower in raw material cost (based on material price of diet components) than the all fishmeal diet. The results therefore indicate that the raw material cost in feed for on‐growing turbot can be reduced considerably without any negative effect on growth and feed utilization.  相似文献

The inclusion of phytogenics in fish feed is a promising strategy to compensate for the negative performance effects of replacing fishmeal (FM) with vegetable sources. The present work assessed the interactive effects of different dietary FM levels (22.5 and 10% of formulation) and the supplementation of a commercial blend of anise, citrus, and oregano essential oils (Digestarom PEP M.G.E 150) on European sea bass, Dicentrarchus labrax growth performance, nutrient utilization, gut morphology, antioxidant status, and immunological response over a 60‐day growth trial. Results showed decreased growth and protein efficiency ratio and increased feed conversion ratio with a low dietary FM level. In contrast, supplementation of the phytogenic product demonstrated improved performance and nutrient utilization together with increased protein and energy retention. Supplementation with the plant essential oils fully compensated for the negative intestinal changes observed in sea bass fed a low‐FM diet but showed little improvement in fish immunological response, except for the 30% increase in lysozyme activity observed in fish fed the low FM‐supplemented diet compared to those fed the standard high‐FM diet. Overall, this study supports the use of this phytogenic product in low‐FM diets as a possible tool to decrease feed costs associated with FM without compromising fish performance, nutrient utilization, and health.  相似文献

Two feeding experiments were carried out to evaluate the utilization of fermented soybean meal (FSBM) in diets for Pacific white shrimp (Litopenaeus vannamei). The digestibility experiment was assessed with four diets: a reference diet and three ingredient test diets containing FSBM, local and Chilean fishmeal. The growth experiment was conducted including four isonitrogenous and isocaloric diets formulated to contain the graded levels of dietary fishmeal (0 g/kg, 150 g/kg, 300 g/kg and 450 g/kg) substituted by FSBM. The results indicated that ADCs of crude protein in FSBM were high (888.4 g/kg) and equal in local fishmeal but little lower than Chilean fishmeal while without any significant differences observed in ADCs of crude lipid among the different test ingredients. After 75 days of growth trial, survival rates and feed conversion ratio were not significantly different among the experimental treatments. However, shrimp fed the diets with increasing inclusions of FSBM had a tendency to reduce weight gain and specific growth rate. Based on the correlation between weight gain and substituted fishmeal level analysed by broken‐line regression, the optimum level of fishmeal replaced by FSBM in diet was 253.6 g/kg without adverse effects on growth and feed utilization of Pacific white shrimp.  相似文献

The utilization of plant‐based diets in fishes could be affected by their genetic backgrounds. To examine this, three strains of one‐year‐old gibel carp, Dongting (strain DT: 30.42 ± 0.05 g), CAS III (strain A: 43.34 ± 0.08 g) and CAS Ⅴ (strain F: 61.78 ± 0.10 g), were fed fishmeal (FM), soybean meal (SBM) and rapeseed meal (RM) diets for 8 weeks. The present results showed that growth performances, feed utilization, body composition, postprandial kinetics of plasma metabolites and gene expression of lipid metabolism markers in the three strains were affected by diets and strains (p < 0.05). Strain A had higher SGR than strain DT and F (A > F > DT) with the three diets, whereas strain F had higher FE than the other two strains with FM and RM diets (p < 0.05). Compared to FM diet, plant protein‐based diets resulted in poor growth, feed use and nutrient retention in all the three strains, and different strains showed differing changes in fatty acid synthesis and oxidation to different dietary proteins. Overall, strain A showed better growth performances than the other two strains irrespective of diet, and all the three strains could utilize RM diet better than SBM diet.  相似文献

This study examined the genetic variability and genotype × diet interactions during early growth (initial mean body weight 1.2 g) among seven heterozygous clones of rainbow trout, Oncorhynchus mykiss. The clones were hand-fed a diet containing either fishmeal or plant proteins during a 49-day trial divided into two periods (P1, 26 days, and P2, 23 days). Weight, variation of weight within clone, feed intake, feed efficiency and mortality were calculated for both periods.There was a highly significant effect of diet and of clone for all traits at both periods, except for feed efficiency and mortality at P1. Highly significant interactions between diet and clone were also recorded for all these traits, except for mortality at P1. The occurrence of genotype × diet interactions when feeding juvenile rainbow trout with an all plant-protein diet indicates that a highly performing genotype on a fishmeal diet may perform poorly when fed a plant-protein diet. Interactions were found for the two major determinants of growth, i.e. feed intake and feed efficiency, showing that the dietary response differs according to the genotype. Monitoring of the within-clone variability of weight showed that a plant-based diet is likely to enhance the overall phenotypic variance in a population, whatever its initial genetic variability.  相似文献

Rainbow trout (23.1 ± 0.4 g) were fed either a fishmeal‐ or plant‐based diet supplemented with various levels of zinc (0, 15, 30, 60 or 120 mg kg?1) for 12 weeks. Trout fed the fishmeal diet had significantly higher weight gain than with the plant‐based diet. Zinc supplementation in the fishmeal diet had no effect on growth performance, suggesting that additional dietary supplementation of zinc is not required. However, in trout fed the plant‐based diet, growth increased significantly up to 30 mg kg?1 zinc after which growth was not affected. Trout fed the plant‐based diet containing no zinc exhibited severe growth retardation, and in fish fed the 0 and 15 mg kg?1 zinc diets, cataracts were present. Use of broken‐line quadratic modelling suggests that dietary supplementation of zinc needed to prevent deficiency and promote adequate growth in rainbow trout fed the plant‐based diet in this study was 30.1 mg kg?1 (80 mg kg?1 total dietary zinc). This is higher than the NRC (2011, Nutrient Requirements of Fish and Shrimp) dietary recommended level of 15 mg kg?1 for rainbow trout. Following the NRC recommendation could lead to zinc deficiency in rainbow trout fed a plant‐based diet.  相似文献

植物蛋白替代鱼粉一直是水产研究的热点,但植物蛋白往往不能完全替代鱼粉,其潜在的原因值得进一步探索。本试验配制了3组等氮等能饲料,分别为鱼粉组(FM)、混合植物蛋白组(豆粕,菜粕,玉米蛋白粉)(PPB)和混合植物蛋白添加氨基酸组(PPB/Aa),对日本沼虾(0.123±0.03 g)进行8周的营养试验。结果发现:PPB和PPB/Aa组日本沼虾增重率显著低于FM组,而饲料系数显著高于FM组(P<0.05),各组之间成活率和肌肉必需氨基酸组成无显著差异(P>0.05);FM组肝胰腺丙二醛(MDA)含量、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性显著低于PPB组和PPB/Aa组(P<0.05),各组谷草转氨酶(AST)和谷丙转氨酶(ALT)无显著差异(P>0.05),PPB与PPB/Aa组血清碱性磷酸酶(AKP)、酸性磷酸酶(ACP)和ALT的活性均显著高于FM组(P<0.05);与FM组相比,PPB与PPB/Aa组显著下调雷帕霉素靶蛋白(TOR)、核糖体蛋白S6激酶(S6K)及神经肽(NPY)基因的表达(P<0.05),但各组瘦素(Leptin)基因的表达无显著差异(P>0.05)。综上,鱼粉与植物蛋白对日本沼虾肌肉氨基酸组成没有影响,植物蛋白降低了日本沼虾生长并引起氧化应激,且添加氨基酸不能缓解这种负面影响,TOR信号通路与食欲调节相关基因表达的抑制可能是植物蛋白抑制日本沼虾生长的原因之一。  相似文献

为了探讨鱼蛋白水解物对黄颡鱼生产性能的影响,以日本鳀粉为对照,以实用型黄颡鱼饲料配方模式为基础开展实验:1以30.5%鱼粉为对照(FM),在相同配方模式下,以6%鱼蛋白水解物(MPH6)替代20%的鱼粉;2以30.5%鱼粉为对照(FM),在无鱼粉日粮中分别添加3%(FPH3)、6%(FPH6)、12%(FPH12)鱼蛋白水解物;共设计5组等氮等能实验日粮,在池塘网箱中养殖黄颡鱼[初始体质量(30.08±0.35)g]60 d。结果显示:与FM相比,FPH12在SGR、FCR、PRR和FRR方面均无显著差异,而MPH6、FPH3、FPH6组SGR降低了15.45%~24.39%,FCR升高了32.14%~42.86%,MPH6、FPH6差异显著,在PRR和FRR方面,MPH6、FPH3、FPH6组PRR降低了21.11%~27.78%,MPH6组FRR降低了41.51%;全鱼水分、粗蛋白、粗脂肪和灰分各组间差异不显著,FPH3、FPH6、FPH12肌肉多种游离氨基酸水平显著高于FM,其中Thr、Val、His与其在日粮中的水平显著相关;FPH6组HSI显著低于FM,鱼蛋白水解物对CP、VSI、肠体比的影响不显著;血清AST、ALT、HDL、LDL、TP、CHOL、TG无显著差异,FPH3组ALB显著低于FM。研究表明:黄颡鱼日粮中,12%鱼蛋白水解物(干物质)与30.5%鱼粉在生长速度、饲料效率、血清生理指标等方面具有一定的等效性;过高的植物蛋白日粮影响了黄颡鱼的生产性能;饲喂鱼蛋白水解物日粮使黄颡鱼肌肉游离氨基酸的含量升高,特别是呈味氨基酸的含量增加。  相似文献

Our recent results in hybrid striped bass (HSB) concluded that ideal protein theory accurately predicts first‐limiting amino acids in commercial diet formulations if accurate amino acid availability data are used and that appropriate levels of supplemental lysine are needed to improve fish performance from fishmeal‐free diets. Our goal in this study was to elucidate how dietary lysine supplementation of a commercial fishmeal‐free diet influences the expression of two genes, myostatin and myogenin, controlling myogenesis in differentially growing groups of HSB. Real‐time RT‐PCR results in HSB suggest that the levels of lysine added to the diet (17.8, 35.1, 51.0 g kg?1 of diet) has an impact on myogenin relative to the basal unsupplemented diet, but no effect on myostatin. Moreover, our data further suggest that the amount of dietary lysine supplementation influenced the ratio of myostatin/myogenin expression in HSB and that this pattern mimicked that of most of the growth, composition of growth and nutrient retention data from our previous study and may therefore be a useful marker for selecting fish for improved growth performance.  相似文献

表达序列标签是一种快捷、高效的揭示基因组信息的方法。本研究以蓝塘猪与大白猪差异表达的5条EST在各组织中的表达情况为目的,采用大白猪、蓝塘猪为实验材料,从成年种猪的10个组织中提取总RNA,运用RT-PCR方法分析,结果显示5条EST在猪的多数组织中均有表达;同源性比对后发现,在5条差异显示cDNA中,EST1、EST2与已知序列无同源性;电子表达谱分析表明EST1、EST2、EST4及EST5在骨骼肌、肝脏组织中有表达。这为进一步研究该EST的功能奠定基础。  相似文献

Using rainbow trout, Oncorhynchus mykiss (Walbaum), of approximately 50 g, a 63 day feeding trial was undertaken to evaluate the efficacy of individual and multiple amino acid supplements in diets where soyabean meal was used as the principle protein source. Iso-nitrogenous diets (45% crude protein) were formulated where the control diet (F) contained fishmeal as the reference protein and a solvent extracted soyabean meal (S) replaced approximately 66% of this protein source. The soyabean containing diets were then supplemented with crystalline amino acids thus; methionine only, dual supplemented with two methionine and lysine levels and finally, a supplement comprising methionine, lysine, tryptophan, threonine, arginine and histidine. The results showed that soyabean meal (SBM) was inferior to the reference protein when SBM was used to replace 66% of the fishmeal and that no significant restoration in growth, feed efficiency and apparent net protein utilization was obtained by either methionine only or dual methionine and lysine supplementation. However, by comparison with the fish fed the unsupplemented, single and dual supplemented diets, multiple amino acid incorporation was associated with significantly improved percentage weight gain, specific growth rate and marginal improvements in apparent net protein utilization. However, performance was not equal to that of the fish fed the fishmeal based control diet. The results are discussed with respect to the level of each of the essential amino acids (EAA) as a proportion of the total EAAs (A/E index) of test diets by reference to the whole body tissue amino acid profile of rainbow trout.  相似文献

The effect of the substitution of fishmeal with tuna by‐product meal (TBM) in the diet of Korean rockfish on growth, body composition, plasma chemistry and amino acid profiles was determined. Nine experimental diets were prepared. The control (Con) diet consisted of 550 g kg?1 fishmeal. The 10, 20, 30, 40, 60, 80 and 100 % fishmeal diets were substituted with tuna by‐product meal (TBM), referred to as the TBM10, TBM20, TBM30, TBM40, TBM60, TBM80 and TBM100 diets, respectively. Finally, the fishmeal and soybean meal was completely replaced with TBM in the diet TBM100‐S. There was no significant difference in weight gain and SGR of the fish that were fed the TBM40 and Con diets. The feed conversion ratio (FCR) of the fish that were fed the Con, TBM10, TBM20 and TBM30 diets was lower than that of the fish that were fed all of the other diets. The protein efficiency ratio (PER) of fish that were fed the TBM10, TBM20 and TBM300 diets was higher than that of fish that were fed the TBM40, TBM60, TBM80, TBM100 and TBM100‐S diets. It can be concluded that the substitution of fishmeal with up to 40 and 30 % TBM in the diet of juvenile Korean rockfish could be made without exerting a detrimental effect on their growth (SGR) and feed utilization (FCR and PER), respectively.  相似文献
