Pathological changes in respiratory tracts were studied in 30 calves following exposure to aerosols of Pasteurella multocida or to bovine herpesvirus 1 and P. multocida. Two groups of five calves were exposed to aerosols of one of two types of P. multocida only, which produced lobar pneumonia in one calf of each group. Another five groups of four calves were exposed to aerosols of bovine herpesvirus 1 and four to seven days later to one of the two types or one sub-type of P. multocida. Extensive necropurulent lesions were produced throughout the respiratory tract with each type of P. multocida in all four calves in three groups but none in the remaining two groups. The pathological changes differed from those produced following similar exposures to bovine herpesvirus 1 and P. haemolytica, in that the exudate in air passages and alveoli was more purulent and streaming (oat) cells and large mononuclear eosinophilic granulocytes were absent. This is the first report of experimental respiratory disease in cattle as a result of aerosol exposure to P. multocida alone or in combination with bovine herpesvirus 1.  相似文献

The development of pathological lesions in the small intestine of neonatal calves is described. Seven newborn calves were challenged orally with a known enteropathogenic strain of E coli 0101k?(A) and killed at varying times after inoculation. Adhesion of bacteria to the mucosa of the small intestine was observed in all calves. A few organisms were seen in the distal small intestine at three hours after inoculation and thereafter adhesion progressed anteriorly along the intestine in calves killed from six to 36 hours. In these calves pathological changes occurred between six and 12 hours after inoculation. Villi were stunted and thickened and the epithelial surface was irregular. A further calf, anaesthetised from five-and-a-half to 10 hours after inoculation and repeatedly sampled from the distal small intestine, developed similar lesions abruptly at nine hours after inoculation. Villus and crypt lengths in the challenged calves were compared with those in three normal uninoculated control calves.  相似文献

Thirteen healthy neonatal Holstein calves were cold stressed twice by hosing with cold water for 20 minutes, 12 hours between hosings. Measurements of the pattern of ventilation [tidal volume (VT), respiratory frequency (f), minute ventilation (VMIN), and functional residual capacity (FRC)], gas exchange properties of the lungs [alveolar ventilation (VA), oxygen uptake (VO2), CO2 production (VCO2), dead space ventilation (VD), dead space/tidal volume ratio (VD/VT), arterial oxygen tension (PaO2), arterial CO2 tension (PaCO2) and alveolar-arterial oxygen difference (AaDO2)] and of the mechanical properties of the pulmonary system [dynamic compliance (Cdyn), pulmonary resistance (RL), and total respiratory system resistance (RRS)] were taken. Calves responded to chilling by increasing VO2 and VCO2 necessitating an increase in VA. This was accomplished by increasing VT with reciprocal decreases in f so that VMIN remained constant. There was no change in Cdyn, RL, or AaDO2. Seven of these 13 calves were then exposed to intratracheal inoculation of 2 X 10(9) organisms of Pasteurella haemolytica, the remaining calves serving as controls. Within 1 hour, calves exposed to P haemolytica had increased VMIN, f, VD/VT, and VD. There was a decrease in PaO2 associated with increased AaDO2, but no change in PaCO2, Cdyn or RL. By 3 hours after inoculation, there were pronounced changes in PaO2 and AaDO2, and Cdyn was reduced below base-line values. By 12 hours after inoculation, calves infected with P haemolytica had increased RL and RRS and PaCO2, in addition to the previously mentioned changes. Data from Pasteurella-exposed calves indicate that gas exchange impairment and peripheral lung injury occur rapidly and that increases in airway resistance develop relatively late in the disease.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Thirteen field cases of chronic respiratory disease in crossbred calves aged between 1 and 8 months were subjected to a standardized examination in which clinical criteria were scored and weighted to allow quantification. Predicted values for pulmonary function variables were generated using regression equations previously established in normal calves which related measured pulmonary function variables to thoracic perimeter. Subsequent pulmonary function testing of each calf was undertaken and values obtained compared with predicted values and correlated with clinical scores. Diagnosis was confirmed at post-mortem examination. Calves with chronic respiratory disease had significantly reduced inspiratory and expiratory times, tidal volume, dynamic compliance and PaO2, significantly increased respiratory frequency, airway resistance and PaCO2 and more negative transpulmonary pressure values when compared to predicted pulmonary function values for the same calves. PaO2 and PaCO2 were the only variables correlated with clinical scores, suggesting their value in the assessment of severe cases of calf respiratory disease.  相似文献

[目的]为了掌握宁夏固原地区示范村杨坪村和马沟村的肉犊牛呼吸道疾病的流行情况,[方法]2019年-2021年对2村犊牛呼吸道疾病发病情况进行统计并采用分子生物学技术对采集的犊牛呼吸道鼻拭子样本进行6种病原的鉴定。[结果] 2019年-2021年杨坪村犊牛呼吸系统疾病发病率呈下降趋势,但不同年份间差异不显著(p>0.05);马沟村犊牛呼吸系统疾病发病率呈下降趋势,2021年与2019年相比差异显著(p<0.05)。2019年-2021年,病毒性呼吸道疾病呈下降趋势,而牛支原体和多杀性巴氏杆菌由于没有有效的疫苗预防而呈现较高的阳性率。[结论]本研究揭示了牛支原体和多杀性巴氏杆菌为目前犊牛主要的呼吸道致病病原,做好牛支原体病和多杀性巴氏杆菌病的预防、早期诊断和治疗可有效降低犊牛呼吸道疾病的发病率。  相似文献

Twelve Pasteurella-free Holstein-Friesian calves were used in a study to test the efficacy of a live streptomycin-dependent Pasteurella multocida A:3 and streptomycin-dependent Pasteurella haemolytica A1 vaccine. The calves were inoculated intramuscularly twice at 14-day intervals with either the streptomycin-dependent vaccine, containing 1 X 10(6) colony forming units/mL P. multocida and 4 X 10(8) colony forming units/mL P. haemolytica, commercial bacterin, or phosphate buffered saline. Two weeks following the second vaccination, all calves were challenged by intranasal inoculation of 10(8) TCID50/4.0 mL infectious bovine rhinotracheitis virus followed three days later by intratracheal injection with 2.3 X 10(7) colony forming units/mL of a 16 hour culture of P. multocida A:3 and 2.6 X 10(8) colony forming units/mL of an 8 hour culture of P. haemolytica A1. Seven days after challenge with Pasteurella, calves were killed for collection of tissues at necropsy. Each calf was given a score based on macroscopic and microscopic lesions. The scores for the calves receiving live vaccines were significantly lower (p less than 0.025) than those for the controls. Also, the calves receiving live vaccines had a significant (p less than 0.05) increase in the level of serum antibody to P. haemolytica. The results of this preliminary study showed that the streptomycin-dependent vaccine offered better protection than the commercial bacterin against a virulent homologous challenge.  相似文献

Seventeen Holstein-Friesian calves weighing an average of 139.8 +/- 13.5 (mean +/- standard deviation) kg were used in a study to determine the efficacy of a live vaccine containing of Pasteurella multocida A:3 and Pasteurella haemolytica A:1. Eleven calves received the vaccine by intramuscular injection in the right shoulder, whereas six calves received vaccine diluent and served as non-vaccinated controls. Fourteen days following vaccination (Day 15) all calves were inoculated deep intranasally with 3.6 X 10(7) TCID50 bovine herpes virus-1. On Day 16, calves were stressed by transports, and on Day 17 calves were challenged intratracheally with P. multocida A:3. On Day 22 calves were euthanized and necropsied, and tissues were collected for pathological and microbiological evaluations. Scores were assigned to each calf based on the severity of observed clinical signs. Macroscopic lung lesions were expressed as percentage of tissue involved relative to the total lung tissue of a calf. Plasma fibrinogen concentration, rectal temperature, serum antibody level, microscopic appearance of lung, and microbiologic results were also recorded for analyses. The control calves had significantly higher clinical-sign scores (P less than 0.05) and more severe gross lesions (P less than 0.05) than the vaccinated calves. Although the vaccinated calves had a slight increase of immunoglobulins M and G classes, the differences were not statistically significant (P greater than 0.05, P greater than 0.05). The results of the study indicate that the live Pasteurella vaccine is effective against experimental P. multocida infection in calves.  相似文献

OBJECTIVE: To examine effects of danofloxacin and tilmicosin on continuously recorded body temperature in beef calves with pneumonia experimentally induced by inoculation of Mannheimia haemolytica. ANIMALS: 41 Angus-cross heifers (body weight, 160 to 220 kg) without a recent history of respiratory tract disease or antimicrobial treatment, all from a single ranch. PROCEDURE: Radiotransmitters were implanted intravaginally in each calf. Pneumonia was induced intrabronchially by use of logarithmic-phase cultures of M. haemolytica. At 21 hours after inoculation, calves were treated with saline (0.9% NaCl) solution, danofloxacin, or tilmicosin. Body temperature was monitored from 66 hours before inoculation until 72 hours after treatment. Area under the curve (AUC) of the temperature-time plot and mean temperature were calculated for 3-hour intervals and compared among treatment groups. RESULTS: The AUCs for 3-hour intervals did not differ significantly among treatment groups for any of the time periods. Analysis of the mean temperature for 3-hour intervals revealed significantly higher temperatures at most time periods for saline-treated calves, compared with temperatures for antimicrobial-treated calves; however, we did not detect significant differences between the danofloxacin- and tilmicosin-treated calves. The circadian rhythm of temperatures before exposure was detected again approximately 48 hours after bacterial inoculation. CONCLUSIONS AND CLINICAL RELEVANCE: Danofloxacin and tilmicosin did not differ in their effect on mean body temperature for 3-hour intervals but significantly decreased body temperature, compared with body temperature in saline-treated calves. Normal daily variation in body temperature must be considered in the face of respiratory tract disease during clinical evaluation of feedlot cattle.  相似文献

The study aimed to establish an experimental model to investigate the pathogenesis of lung infection by Pasteurella multocida, an important cause of bovine respiratory disease. An experimental model is required to assist the development of an effective vaccine. Sixteen 8-week-old calves were challenged intratracheally with 10(9) or 10(10) colony forming units of P. multocida in either 60 or 300 ml saline in a 2 x 2 factorial experiment. All animals became dull within 2-6h post-infection (p.i.) and two calves were killed humanely because of suspected endotoxic shock. Remaining animals showed increased respiratory rates by 15-20 h p.i. and, at 23 h p.i., calves given the high dose, high volume challenge showed higher (P < 0.05) rectal temperatures. From 24 to 36 h p.i., clinical signs decreased in a majority of animals. Plasma haptoglobin concentrations increased (P < 0.05) in calves given the high volume challenge irrespective of the number of bacteria. At post-mortem examination (4d p.i.), lung lesions, mainly in the apical lobes, were found in all calves. Histopathological examination showed areas of purulent pneumonia with a tendency to abscessation and inflamed interlobular septa characterised by accumulation of neutrophils and oedema. The clinical and pathological responses described were typical of bovine pneumonic pasteurellosis.  相似文献

Clinical changes and acute phase responses, including tumour necrosis factor-alpha (tnfalpha), in six buffalo calves were examined following intravenous inoculation of a bolus of endotoxin (1 microg kg(-1) bodyweight in 10 ml of phosphate-buffered saline [ pbs ]) extracted from Pasteurella multocida serotype B:2, the bacterium responsible for haemorrhagic septicaemia (hs) in Asia. Endotoxin injection caused a rapid onset of clinical signs characterised by dullness, sternal recumbency, elevated rectal temperatures, excessive salivation and dyspnoea that lasted for up to 12 hours post-inoculation (p.i.). Serum concentrations of tnfalpha rose within 1 hour p.i. to reach peak values ranging between 8 and 140 ng ml(-1) at 1-2 hours p.i. and then declined rapidly to baseline levels 3-5 hours p.i. Endotoxin injection induced other acute phase changes, including a rapid leucopenia and reductions in the serum concentrations of iron and zinc and a delayed but prolonged increase in haptoglobin from 12 hours p.i. that reached a plateau from about 60 hours p.i. Three control calves injected with 10 ml pbs showed no clinical or blood compositional changes. By reproducing key signs of hs the work confirms a pivotal role of endotoxin in the pathogenesis of hs and emphasises the exquisite sensitivity of the buffalo to P multocida endotoxin.  相似文献

Collection of exhaled condensate (freezing of expired air under conditions of spontaneous breathing) is a non-invasive method permitting the collection of material originating from the lung and the lower respiratory tract so that it can be used for diagnostic examinations. In order to be able to evaluate the diagnostic evidence of exhaled condensate samples in cases of respiratory disease of the calf, leukotriene B4 (LTB4) in bovine exhaled condensate was determined. The influence of the breathing pattern and body temperature on the quantity of condensate to be collected was tested in a total of 49 exhaled condensate samples. It became obvious that the exhaled condensate quantity obtained per time unit is dependent on the ventilation volume per time unit (minute volume). In exhaled condensate samples from 35 clinically healthy calves, LTB4 concentrations of less than 250 pg/mL exhaled condensate were detected. A total of 14 exhaled condensate samples from 7 calves was analyzed before and after experimental respiratory infection with Pasteurella multocida D. In parallel to the analysis of LTB4 in exhaled condensate, the lung function of the calves was examined by means of impulse oscilloresistometry. The increase of LTB4 in the exhaled condensate post infection correlated significantly (p < or = 0.05) with decreases of respiratory reactance. The determination of LTB4 concentrations in exhaled condensate seems to be suitable, in principle, for the detection of inflammations in the respiratory system of the calf. Further studies are needed for the evaluation of the diagnostic validity of the method.  相似文献

In 5 Friesian calves given 3-methylindole (3-MI) (100 mg/kg once a week for 8 weeks, except calf 4, given a 50 mg/kg dose on weeks 3 to 8), pulmonary function (PF) values and arterial blood gas tensions (PaO2 and PaCO2) were measured 24 hours after dosing was done and were correlated with clinical, biochemical, and pathologic changes. Three of the calves (No. 1, 2, and 3) showed acute respiratory distress syndrome 24 hours after the first 3-MI treatment, with a large increase in respiratory frequency, minute viscous work, and PaCO2 and a large decrease in tidal volume, dynamic lung compliance, and PaCO2. They died 36, 38, and 84 hours after dosing. Pulmonary function changes were compatible with the severe pulmonary edema and alveolar damage observed at necropsy. The 2 other calves, after they were given the 1st dose, showed only subacute respiratory distress syndrome with less severe changes in PF values recorded at 24 hours. Furthermore, they became progressively more tolerant to the 2nd, 3rd, and 4th weekly treatments, and showed base-line PF values after the 5th weekly treatment. Pathologic changes were not observed in lung biopsy material from these 2 animals at 2 and at 12 weeks after the 8th (or last) 3-MI treatment.  相似文献

Inoculation of calves with potassium thiocyanate (KSCN) extract of Pasteurella multocida type A in saline-tris buffer or in Freund's incomplete adjuvant or modified Freund's incomplete adjuvant resulted in the elicitation of agglutinating, hemagglutinating, bactericidal and homocytotropic antibodies. The antibody response was significantly (P = 0.05) higher in calves inoculated with the KSCN extract in either adjuvant than in those inoculated with the extract in saline-tris buffer. Hemagglutination also was observed if the KSCN extract of P hemolytica was used for sensitizing the tanned sheep red blood cells. Further, calf anti-P multocida extract antisera also was bactericidal to P hemolytica. The KSCN extract of P multocida was found to be nontoxic to calves at 2 doses tested, as judged by an evaluation of total and differential leukocyte counts, body temperature, and pulse rates at various intervals after inoculation.  相似文献

Respiratory syncytial virus (RSV) infection causes severe lower respiratory tract disease in infants and calves. Neonatal respiratory tract infection in children often produces persistent changes in lung function. The specific objective of this study was to determine whether neonatal calves have transient or persistent alterations in pulmonary function and airway reactivity following RSV infection. Six 2- to 3-day-old Holstein bull calves were inoculated with 10 ml of bovine respiratory syncytial virus (BRSV) inoculum (10(2.7) to 10(3.8) cell culture infective doses/ml) intranasally and 10 ml of BRSV inoculum (10(4.8) to 10(5.9) cell culture infective doses/ml) intratracheally for 4 consecutive days, and 5 other calves were sham-inoculated. Prior to inoculation (day 0) and on days 4, 14, and 30 after the last inoculation, body weight (kg), dynamic compliance (Cdyn), pulmonary resistance (RL), and 2 indices of airway reactivity (effective dose [ED] 65Cdyn and ED200RL) were measured. Control calves gained weight progressively throughout the study, whereas RSV-inoculated calves failed to gain weight for 14 days, but equaled control calf weight by 30 days after inoculation. The Cdyn of control calves increased significantly by 30 days, but did not in the RSV-infected calves. Pulmonary resistance was increased significantly at 4, 14, and 30 days, but was unaffected by sham inoculation. The ED65Cdyn and ED200RL indicated an age-dependent increase in reactivity to histamine and an increase in responsiveness in the infected group beginning at 14 days and persisting until the end of the study.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

为了克隆从患肺炎犊牛和健康犊牛上分离的多杀性巴氏杆菌血红素结合受体(heme acquisition system receptor,HasR)基因并进行序列分析,分别以致犊牛肺炎和健康犊牛携带的多杀性巴氏杆菌DNA为模板,通过PCR反应扩增出血红素结合受体基因的全部序列,PCR产物纯化后克隆到pMD19-T载体上,经菌液PCR和酶切鉴定后进行序列测定。各菌株HasR基因序列比对结果发现,Pm-BS-a、Pm142-x-4、Pm142-x-3、Pm149-x、Pm-BS-d的序列之间亲源关系较近,而Pm149-xby与参考菌株Pm70的同源性较高。健康牛源的多杀性巴氏杆菌与致犊牛肺炎多杀性巴氏杆菌之间HasR基因的同源性非常高,说明致犊牛肺炎多杀性巴氏杆菌是一种条件性致病菌,其引起的犊牛肺炎可能属内源性感染。  相似文献

Sixty-four, 10-week-old turkeys were inoculated with a highly virulent field isolate (86-1913) of Pasteurella multocida serotype A:3,4 by an oculo-nasal-oral route. Inoculated turkeys were examined at 4, 8, 16, 20, and 24 hours post-inoculation for bacteremia and histologic lesions. Bacteremia was detected in one of six turkeys 8 hours after inoculation and in four of six turkey poults at 16 hours post-inoculation. Pasteurella multocida was isolated from the spleens of two turkeys at 8 hours and from the spleens of all six poults 16 hours after inoculation. Peak concentrations of P. multocida reached 10(9) colony forming units per ml of blood. At 4 to 8 hours post-inoculation, isolate 86-1913 produced a fibrinopurulent bronchopneumonia followed by severe pulmonary necrosis, pleuritis, vasculitis; and, at 16 to 24 hours post-inoculation numerous extracellular bacteria were observed. Hepatic lesions included focal heterophil aggregates 8 hours after inoculation; these progressed to hepatic necrosis. Numerous extracellular bacteria within sinusoids were present 16 to 24 hours after inoculation. At 16 to 24 hours post-inoculation, there was degeneration of periarteriolar reticular cells in the spleen; these cells progressed to coalescing coagulative splenic necrosis with extracellular bacterial colonies. Eighty-four calves with respiratory disease from 18 herds in different parts of Finland were chosen for a study evaluating the capacity of different respiratory pathogens to cause changes in different acute phase protein concentrations, white blood cell (WBC) count and clinical signs. The selected acute phase proteins were fibrinogen, haptoglobin, serum amyloid-A, lipopolysaccharide binding protein and alpha1-acid glycoprotein. From each calf, a paired blood sample was obtained for serological studies of bovine parainfluenza virus-3, bovine respiratory syncytial virus, bovine coronavirus, bovine adenovirus-3 and bovine adenovirus-7. Tracheobronchial lavage was performed to detect bacteria and mycoplasma. Isolation of Pasteurella multocida was associated with increased concentrations of all tested acute phase proteins. For other pathogens, no significant relationships were observed. No association was present between viral or bacterial findings and WBC count.  相似文献

Twenty veal calves were fed milk substitute containing 19 mg Fe/kg; ten calves were supplemented with iron in accordance with their requirements and the other 10 were unsupplemented controls. Serum iron (SI), hemoglobin (Hb) and packed cell volume (PCV) were determined at the start of the experiment and then every third week. From 3 weeks after the start to the end of the experiment there were significant differences between SI mean values between treated and untreated calves. In the treated group the Hb mean values were unchanged during the experiment while the untreated group had a significant decrease. The mean PCV values were also decreased. The daily gain was 1.36 g higher per calf in the treated group. The feed consumption per kg gain was 18 g lower and the carcass weight was 4.2 kg higher per calf in the treated group. The points for color and structure of carcass were 2.5 units higher per calf in the untreated group. The incidence of enteritis was less in the treated calves than in the untreated ones.  相似文献

Tests were performed on 130 one-month-old calves. In 16 animals, treatment with a suspension of Trichophyton verrucosum culture at the dosage of 5 mil. conidia for an area of 10 x 10 of the clipped skin caused clinical trichophytic changes. The length of the incubation time was shorter in the calves scarified before inoculation of the infecting culture (10 to 13 days), as compared with the group of calves without scarification of the skin (11 to 19 days). Only 3 in 6 calves, infected by the same dose into non clipped skin, did contract trichophytosis, manifesting themselves as separate mycotic deposits, which appeared 26 to 35 days after inoculation. Trichophytic changes lasted longest in the group with scarification of the infected area. The ID50 in T. verrucosum and T. equinum cultures was about 1500 conidia per one calf in the case of the method of infecting into clipped scarified skin (area 100 sq cm). A 10 times lower dose (150 conidia per one calf) of the T. verrucosum suspension did not induce visible mycotic changes, but the T. equinum inoculation produced infection in one of 10 calves.  相似文献

The prevalence of respiratory pathogens in diseased veal calves was determined in 24 respiratory disease outbreaks in 15 herds in Belgium. Bacteria were cultured from nasopharyngeal swabs and seroconversion against viruses and Mycoplasma bovis was determined on paired sera. At the individual calf level, Mycoplasma species, Mannheimia haemolytica and Pasteurella multocida, were isolated from 70.5 per cent, 21.5 per cent and 26.0 per cent of swabs, respectively. At the herd level, the presence of M bovis could be confirmed in 84.6 per cent of the herds examined. Seroconversion against bovine viral diarrhoea virus (BVDV) was present in 71.4 per cent of herds, parainfluenzavirus type 3 in 53.3 per cent, bovine respiratory syncytial virus in 40.0 per cent, bovine adenovirus type 3 in 46.7 per cent, bovine coronavirus in 30.0 per cent, and bovine herpesvirus type 1 in 26.7 per cent. At postmortem examination, Mycoplasma species could be cultured from 61.9 per cent of pneumonic lungs (n=21). Sixty per cent of calves tested were positive for BVDV (n=20), and 20.0 per cent were positive for bovine respiratory syncytial virus (n=16).  相似文献
