From June 2006 to January 2007 passive samplers (solid phase adsorbing toxin tracking, SPATT) were tested as a monitoring tool with weekly monitoring of phytoplankton and toxin content (liquid chromatography–mass spectrometry, LC-MS) in picked cells of Dinophysis and plankton concentrates. Successive blooms of Dinophysis acuminata, D. acuta and D. caudata in 2006 caused a long mussel harvesting closure (4.5 months) in the Galician Rías (NW Spain) and a record (up to 9246 ng·g resin-week−1) accumulation of toxins in SPATT discs. Best fit of a toxin accumulation model was between toxin accumulation in SPATT and the product of cell densities by a constant value, for each species of Dinophysis, of toxin content (average) in picked cells. Detection of Dinophysis populations provided earlier warning of oncoming diarrhetic shellfish poisoning (DSP) outbreaks than the SPATT, which at times overestimated the expected toxin levels in shellfish because: (i) SPATT accumulated toxins did not include biotransformation and depuration loss terms and (ii) accumulation of toxins not available to mussels continued for weeks after Dinophysis cells were undetectable and mussels were toxin-free. SPATT may be a valuable environmental monitoring and research tool for toxin dynamics, in particular in areas with no aquaculture, but does not provide a practical gain for early warning of DSP outbreaks.  相似文献

Vegetative and reproductive growth information of lesquerella (Physaria fendleri), a new oilseed crop targeted for bio-products, is important to understand especially in the early commercialization stage of this new crop. The objective of this study was to determine the effect of fall, winter, and spring planting dates over three years on the ontogeny of the crop including biomass, floral buds, flowers, and siliques. Fall plantings always produced more than the other plantings due to the extended season. Winter and spring plantings had less biomass and produced fewer buds, flowers, and siliques. The compensation of lower crop management costs and a shorter growing season could make winter a viable option for planting in the southwest. Spring planting could become viable if seed shatter due to summer rains could be reduced. The information will help decide growing regions suitable for crop production and determine what reproductive stages could be manipulated to improve seed yields.  相似文献

Traditionally, harmful algal bloom studies have primarily focused on quantifying toxin levels contained within the phytoplankton cells of interest. In the case of paralytic shellfish poisoning toxins (PSTs), intracellular toxin levels and the effects of dietary consumption of toxic cells by planktivores have been well documented. However, little information is available regarding the levels of extracellular PSTs that may leak or be released into seawater from toxic cells during blooms. In order to fully evaluate the risks of harmful algal bloom toxins in the marine food web, it is necessary to understand all potential routes of exposure. In the present study, extracellular and intracellular PST levels were measured in field seawater samples (collected weekly from June to October 2004–2007) and in Alexandrium spp. culture samples isolated from Sequim Bay, Washington. Measurable levels of intra- and extra-cellular toxins were detected in both field and culture samples via receptor binding assay (RBA) and an enzyme-linked immunosorbent assay (ELISA). Characterization of the PST toxin profile in the Sequim Bay isolates by pre-column oxidation and HPLC-fluorescence detection revealed that gonyautoxin 1 and 4 made up 65 ± 9.7 % of the total PSTs present. Collectively, these data confirm that extracellular PSTs are present during blooms of Alexandrium spp. in the Sequim Bay region.  相似文献

This study investigates the occurrence of diarrhetic shellfish toxins (DSTs) and their producing phytoplankton species in southern Brazil, as well as the potential for toxin accumulation in co-occurring mussels (Perna perna) and octopuses (Octopus vulgaris). During the spring in 2012 and 2013, cells of Dinophysis acuminata complex were always present, sometimes at relatively high abundances (max. 1143 cells L−1), likely the main source of okadaic acid (OA) in the plankton (max. 34 ng L−1). Dinophysis caudata occurred at lower cell densities in 2013 when the lipophilic toxins pectenotoxin-2 (PTX-2) and PTX-2 seco acid were detected in plankton and mussel samples. Here, we report for the first time the accumulation of DSTs in octopuses, probably linked to the consumption of contaminated bivalves. Perna perna mussels were consistently contaminated with different DSTs (max. 42 µg kg−1), and all octopuses analyzed (n = 5) accumulated OA in different organs/tissues: digestive glands (DGs) > arms > gills > kidneys > stomach + intestine. Additionally, similar concentrations of 7-O-palmytoyl OA and 7-O-palmytoly dinophysistoxin-1 (DTX-1) were frequently detected in the hepatopancreas of P. perna and DGs of O. vulgaris. Therefore, octopuses can be considered a potential vector of DSTs to both humans and top predators such as marine mammals.  相似文献

The illness of three people in 2011 after their ingestion of mussels collected from Sequim Bay State Park, Washington State, USA, demonstrated the need to monitor diarrhetic shellfish toxins (DSTs) in Washington State for the protection of human health. Following these cases of diarrhetic shellfish poisoning, monitoring for DSTs in Washington State became formalized in 2012, guided by routine monitoring of Dinophysis species by the SoundToxins program in Puget Sound and the Olympic Region Harmful Algal Bloom (ORHAB) partnership on the outer Washington State coast. Here we show that the DSTs at concentrations above the guidance level of 16 μg okadaic acid (OA) + dinophysistoxins (DTXs)/100 g shellfish tissue were widespread in sentinel mussels throughout Puget Sound in summer 2012 and included harvest closures of California mussel, varnish clam, manila clam and Pacific oyster. Concentrations of toxins in Pacific oyster and manila clam were often at least half those measured in blue mussels at the same site. The primary toxin isomer in shellfish and plankton samples was dinophysistoxin-1 (DTX-1) with D. acuminata as the primary Dinophysis species. Other lipophilic toxins in shellfish were pectenotoxin-2 (PTX-2) and yessotoxin (YTX) with azaspiracid-2 (AZA-2) also measured in phytoplankton samples. Okadaic acid, azaspiracid-1 (AZA-1) and azaspiracid-3 (AZA-3) were all below the levels of detection by liquid chromatography tandem mass spectrometry (LC-MS/MS). A shellfish closure at Ruby Beach, Washington, was the first ever noted on the Washington State Pacific coast due to DSTs. The greater than average Fraser River flow during the summers of 2011 and 2012 may have provided an environment conducive to dinoflagellates and played a role in the prevalence of toxigenic Dinophysis in Puget Sound.  相似文献

Diaphorina citri Kuwayama (Hemiptera: Psyllidae), is a global pest of citrus and vector of Candidatus Liberibacter, a bacteria that causes huanglongbing or greening, a devastating disease of citrus. Mature citrus trees are dormant in winter and produce most new shoots in spring, followed by sporadic canopy growth in summer and fall. Young shoots are required for oviposition and nymphal development, but adults can survive and overwinter on hardened leaves. Surviving adults reproduce in spring shoots and their progeny are probably responsible for a large portion of disease spread as they disperse to search for food. Therefore, foliar sprays of broad-spectrum insecticides applied to mature trees in winter were evaluated in a commercial citrus orchard as tactic to reduce pest populations and insecticide use in spring and summer when beneficial insects are most active. A single spray of chlorpyrifos (2.8 kg a.i. ha−1) in January 2007 reduced adult psyllids an average of 10-fold over six months compared to untreated trees. The following year, differences with the untreated control averaged 15-fold for over five months following a single spray of chlorpyrifos, fenpropathrin (0.34 kg a.i. ha−1), or oxamyl (1.12 kg a.i. ha−1) applied in January. Spiders, lacewings and ladybeetles were equally abundant during the growing season in both treated and untreated trees both years (P = 0.05). Thus foliar sprays of broad-spectrum insecticides before spring growth suppressed D. citri for five to six months, with no detectible impact on key natural enemies. This tactic has been widely adopted to control the psyllid in Florida, in some cases area-wide. Additional sprays during the growing season should be based on scouting and targeted at adults before anticipated new flush.  相似文献

Murraya koenigii (L.) Spreng. (Rutaceae) is characterized by a large chemical intraspecific variability among the land races. This fact makes it difficult to detect real changes occurring in their essential oil composition during annual cycle. Based on this, variations of essential oil yield and composition in two chemotypes (‘A’ and ‘B’) of M. koenigii were assessed in spring, summer, rainy, autumn and winter seasons under foot hill conditions of northern India. The essential oil yield ranged from 0.15% to 0.18% in chemotype ‘A’, while it varied from 0.12% to 0.14% in chemotype ‘B’. Essential oils of both chemotypes from different seasons were analysed by gas chromatography (GC-FID) and gas chromatography-mass spectrometry (GC-MS). A total of fifty-eight constituents representing 93.7-98.8% of chemotype ‘A’ and fifty-six constituents forming 96.1-98.7% of the total composition of chemotype ‘B’ were identified. Chemotype ‘A’ was characterized by higher percentages of α-pinene (34.6-41.9%), sabinene (26.1-36.1%), (E)-caryophyllene (2.4-5.4%) and terpinen-4-ol (1.5-5.3%), whereas chemotype ‘B’ was dominated by higher amount of α-pinene (52.7-65.3%), β-pinene (10.7-12.9%), (E)-caryophyllene (3.1-10.3%) and limonene (5.1-7.8%). Comparative results showed considerable variations in the essential oil composition of both chemotypes due to season of collection. Present study concluded that the M. koenigii leaves of desired quality may be obtained by selecting suitable chemotype and season.  相似文献

Six new tetraprenylated alkaloids, designated as malonganenones L–Q (1–6), were isolated from the gorgonian Echinogorgia pseudossapo, collected in Daya Bay of Guangdong Province, China. The structures of 1–6 featuring a methyl group at N-3 and a tetraprenyl chain at N-7 in the hypoxanthine core were established by extensive spectroscopic analyses. Compounds 1–6 were tested for their inhibitory activity against the phosphodiesterases (PDEs)-4D, 5A, and 9A, and compounds 1 and 6 exhibited moderate inhibitory activity against PDE4D with IC50 values of 8.5 and 20.3 µM, respectively.  相似文献

The agronomic value of Syn 1 tall fescue, a synthetic variety derived from North African parent material, was determined in three experiments. Herbage yield, sward purity and digestibility were measured under cutting treatments for incremental sward growth during autumn/winter, and for sward regrowths during autumn/winter, spring and a complete season. Similar data were obtained for sward regrowths during autumn/winter and spring under defoliation by grazing. Syn 1 was compared with several of the north-temperate region tall fescues, e.g. S170, Rozelle and Manade, also with S143 cocksfoot Syn 1 showed yield superiority in autumn/early winter when there were comparable populations of tall fescue. Under cutting management, Syn 1 swards thinned out rapidly because of lack of winter-hardiness and yields were depressed in the second season; the effects were worst under the winter foggage-type management. Under grazing, with probably less severe defoliation, Syn 1 swards maintained yields for an additional season, but thereafter failed to persist Syn 1 swards had low spring- and annual-yields. The yield advantage of Syn 1 in autumn/early winter was not sufficient to be of great agricultural value and with its shortcomings in winter-hardiness and annual yield, it was considered unsuitable for West of Scotiand conditions. The main value of plant material such as Syn 1 may lie in ccmferring autumn/winter growth potential to winter-hardy north-temperate tall fescues in a breeding programme.  相似文献

Two different isolation techniques, microwave-assisted hydrodistillation (MAHD) and conventional hydrodistillation (HD), were used to extract the volatile constituents from the leaves and stems of Schefflera heptaphylla (L.) Frodin harvested during four different seasons, which were subsequently analyzed via gas chromatography with flame ionization detection and gas chromatography with mass spectrometry. MAHD is more advantageous than HD in terms of energy savings and extraction time (60 min vs. 180 min for MAHD vs. HD). Large variations in the composition among the different oils were observed. The seasonal changes in the percentages of the main constituents of the oils were irregular; this affected the quantity and composition of the oil. Oil yield was affected by the method of extraction and seasonal changes. It ranged from 0.11% to 0.27%, with the maximum amount of oil extracted from the leaves using MAHD in winter and the minimum from the stem oil extraction using HD in spring. The essential oil content varied significantly with the season. Sesquiterpene hydrocarbons constituted the major portion of the essential oils. Sesquiterpene hydrocarbons were highest in the leaf oil extraction by HD during winter (88.6%) and lowest in the stem oil extraction by HD during spring (56.3%). Microwave irradiation did not adversely affect the composition of the essential oils. MAHD, indeed, is a modern, green, and fast technology.  相似文献

The frequent occurrence of Moorea producens (formerly Lyngbya majuscula) blooms has been associated with adverse effects on human health. Hoiamide A is a structurally unique cyclic depsipeptide isolated from an assemblage of the marine cyanobacteria M. producens and Phormidium gracile. We examined the influence of hoiamide A on neurite outgrowth in neocortical neurons and found that it suppressed neurite outgrowth with an IC50 value of 4.89 nM. Further study demonstrated that hoiamide A stimulated lactic acid dehydrogenase (LDH) efflux, nuclear condensation and caspase-3 activity with EC50 values of 3.66, 2.55 and 4.33 nM, respectively. These data indicated that hoiamide A triggered a unique neuronal death profile that involves both necrotic and apoptotic mechanisms. The similar potencies and similar time-response relationships between LDH efflux and caspase-3 activation/nuclear condensation suggested that both necrosis and apoptosis may derive from interaction with a common molecular target. The broad-spectrum caspase inhibitor, Z-VAD-FMK completely inhibited hoiamide A-induced neurotoxicity. Additionally, hoiamide A stimulated JNK phosphorylation, and a JNK inhibitor attenuated hoiamide A-induced neurotoxicity. Collectively, these data demonstrate that hoiamide A-induced neuronal death requires both JNK and caspase signaling pathways. The potent neurotoxicity and unique neuronal cell death profile of hoiamide A represents a novel neurotoxic chemotype from marine cyanobacteria.  相似文献

Kernel hardness is mainly conditioned by allelic variations of Pina-D1 and Pinb-D1 genes located on the short arm of chromosome 5D. In this work, the Ecotilling approach was optimized to investigate Pina and Pinb alleles in the micro-core collections of Chinese wheat germplasm, and three Pina and eight Pinb alleles were found. Generally, more Pinb alleles were detected in the accessions coming from the regions that grow winter or a mixture of spring and winter wheats. This was particularly evident for the Southwestern winter wheat, Xinjiang winter–spring wheat and Yellow and Huai River Valley winter wheat regions. A novel variant (designated as Pinb-D1x) was discovered in one of the accessions from the Xinjiang winter–spring wheat region. Compared to wild type (WT) allele Pinb-D1a, two nucleotide substitutions occurred in the coding region of Pinb-D1x, one (at nucleotide position 257) resulting in the replacement of a WT cysteine residue by tyrosine and the other (at nucleotide position 382) creating a premature stop codon. The implications of our data to understanding the diversity of Pina and Pinb alleles in wheat and to future molecular breeding of wheat kernel hardness are discussed.  相似文献

The blood coagulation cascade involves the human coagulation factors thrombin and an activated factor VII (fVIIa). Thrombin and fVIIa are vitamin-K-dependent clotting factors associated with bleeding, bleeding complications and disorders. Thrombin and fVIIa cause excessive bleeding when treated with vitamin-K antagonists. In this research, we explored different strains of toxic Microcystis aeruginosa and cyanobacteria blooms for the probable fVIIa-soluble Tissue Factor (fVIIa-sTF) inhibitors. The algal cells were subjected to acidification, and reverse phase (ODS) chromatography-solid phase extraction eluted by water to 100% MeOH with 20%-MeOH increments except for M. aeruginosa NIES-89, from the National Institute for Environmental Studies (NIES), which was eluted with 5%-MeOH increments as an isolation procedure to separate aeruginosins 89A and B from co-eluting microcystins. The 40%–80% MeOH fractions of the cyanobacterial extract are active against fVIIa-sTF. The fVIIa-sTF active fractions from cultured cyanobacteria and cyanobacteria blooms were subjected to liquid chromatography-mass spectrometry (LC-MS). The 60% MeOH fraction of M. aeruginosa K139 exhibited an m/z 603 [M + H]+ attributed to aeruginosin K139, and the 40% MeOH fraction of M. aeruginosa NIES-89 displayed ions with m/z 617 [M − SO3 + H]+ and m/z [M + H]+ 717, which attributed to aeruginosin 89. Aeruginosins 102A/B and 298A/B were also observed from other toxic strains of M. aeruginosa with positive fVIIa-sTF inhibitory activity. The active fractions contained cyanobacterial peptides of the aeruginosin class as fVIIa-sTF inhibitors detected by LC-MS.  相似文献

The invasive mollusk Crepidula fornicata, occurring in large amounts in bays along the French Northeastern Atlantic coasts, may have huge environmental effects in highly productive ecosystems where shellfish are exploited. The present study aims at determining the potential economic value of this marine species in terms of exploitable substances with high added value. Lipid content and phospholipid (PL) composition of this mollusk collected on the Bourgneuf Bay were studied through four seasons. Winter specimens contained the highest lipid levels (5.3% dry weight), including 69% of PLs. Phosphatidylcholine (PC) was the major PL class all year, accounting for 63.9% to 88.9% of total PLs. Consequently, the winter specimens were then investigated for PL fatty acids (FAs), and free sterols. Dimethylacetals (DMAs) were present (10.7% of PL FA + DMA mixture) revealing the occurrence of plasmalogens. More than forty FAs were identified, including 20:5n-3 (9.4%) and 22:6n-3 (7.3%) acids. Fourteen free sterols were present, including cholesterol at 31.3% of the sterol mixture and about 40% of phytosterols. These data on lipids of C. fornicata demonstrate their positive attributes for human nutrition and health. The PL mixture, rich in PC and polyunsaturated FAs, offers an interesting alternative source of high value-added marine lecithin.  相似文献

Marine pelagic diazotrophic cyanobacteria of the genus Trichodesmium (Oscillatoriales) are widespread throughout the tropics and subtropics, and are particularly common in the waters of New Caledonia. Blooms of Trichodesmium are suspected to be a potential source of toxins in the ciguatera food chain and were previously reported to contain several types of paralyzing toxins. The toxicity of water-soluble extracts of Trichodesmium spp. were analyzed by mouse bioassay and Neuroblastoma assay and their toxic compounds characterized using liquid chromatography coupled with tandem mass spectrometry techniques. Here, we report the first identification of palytoxin and one of its derivatives, 42-hydroxy-palytoxin, in field samples of Trichodesmium collected in the New Caledonian lagoon. The possible role played by Trichodesmium blooms in the development of clupeotoxism, this human intoxication following the ingestion of plankton-eating fish and classically associated with Ostreopsis blooms, is also discussed.  相似文献

Potato psyllids became a topic of interest at the end of the 2011 growing season in the Pacific Northwest when they were first documented to transmit ‘Candidatus Liberibacter solanacearum,’ the putative pathogen causing zebra chip disease in the region. Zebra Chip is characterized by above and below ground symptoms that cause significant yield loss. For many decades it has been unclear whether the potato psyllid migrates to states in the Pacific Northwest from large, well-documented infestation areas in California or Texas. Thus, we conducted a study in the spring of 2012 in the lower Columbia Basin (north east Oregon and south east Washington state), and in the fall, winter, and spring of 2012 in southwestern Idaho to determine if psyllids could be found overwintering. Potato psyllid was observed in November 2011 in southwestern Idaho and March 2012 in the lower Columbia Basin living on Solanum dulcamara L. (i.e., bittersweet nightshade), a perennial weed of wet habitats and riverbanks. Potato psyllids were collected and brought to the laboratory. Emerging adults from both locations (i.e., Columbia Basin and Idaho) were offered potato plants and they completed a second generation in approximately 15 days under similar environmental conditions. While overwinter survival of the potato psyllid in the Pacific Northwest is concerning for the potato industry, weather trends in the region were mild during the winter of 2012. It is unknown whether a colder or longer winter season would affect the overwintering biology we have observed. Observations through several coming winters are warranted.  相似文献

In this study, we characterized the genetic structure of Myzus persicae (Sulzer) (Hemiptera: Aphididae) populations in China using microsatellites. We expected that these data will reveal the genetic relationships among various populations of M. persicae and will be of value in the development of better methods for pest control. Four hundred sixty individuals from 23 areas over 13 provinces were collected in the early spring of 2010, all from their primary host, Prunus persicae. The markers analyzed were highly polymorphic, as demonstrated by the expected heterozygosity value (He = 0.861) and the Polymorphism Information Content (PIC = 0.847), which indicated that M. persicae maintains a high level of genetic diversity. Analysis of molecular variance revealed an intermediate level of population differentiation among M. persicae populations (FST = 0.1215). Geographic isolation existed among these populations, and, consequently, the genetic structure of the populations was split into a southern group and a northern group divided by the Yangtse River.  相似文献

Putative precursors in pseudopterosin biosynthesis, the hydrocarbons isoelisabethatriene (10) and erogorgiaene (11), have been identified from an extract of Pseudopterogorgia elisabethae collected in the Florida Keys. Biosynthetic experiments designed to test the utilization of these compounds in pseudopterosin production revealed that erogorgiaene is transformed to pseudopterosins A–D. Together with our previous data, it is now apparent that early steps in pseudopterosin biosynthesis involve the cyclization of geranylgeranyl diphosphate to elisabethatriene followed by the dehydrogenation and aromatization to erogorgiaene.  相似文献

Evaluating decision rules for dryland rotation crop selection   总被引:1,自引:0,他引:1  
No-till dryland winter wheat (Triticum aestivum L.)-fallow systems in the central Great Plains have more water available for crop production than the traditional conventionally tilled winter wheat-fallow systems because of greater precipitation storage efficiency. That additional water is used most efficiently when a crop is present to transpire the water, and crop yields respond positively to increases in available soil water. The objective of this study was to evaluate yield, water use efficiency (WUE), precipitation use efficiency (PUE), and net returns of cropping systems where crop choice was based on established crop responses to water use while incorporating a grass/broadleaf rotation. Available soil water at planting was measured at several decision points each year and combined with three levels of expected growing season precipitation (70, 100, 130% of average) to provide input data for water use/yield production functions for seven grain crops and three forage crops. The predicted yields from those production functions were compared against established yield thresholds for each crop, and crops were retained for further consideration if the threshold yield was exceeded. Crop choice was then narrowed by following a rule which rotated summer crops (crops planted in the spring with most of their growth occurring during summer months) with winter crops (crops planted in the fall with most of their growth occurring during the next spring) and also rotating grasses with broadleaf crops. Yields, WUE, PUE, value-basis precipitation use efficiency ($PUE), gross receipts, and net returns from the four opportunity cropping (OC) selection schemes were compared with the same quantities from four set rotations [wheat-fallow (conventional till), (WF (CT)); wheat-fallow (no-till), (WF (NT)); wheat–corn (Zea mays L.)-fallow (no-till), (WCF); wheat–millet (Panicum miliaceum L.) (no-till), (WM)]. Water use efficiency was greater for three of the OC selection schemes than for any of the four set rotations. Precipitation was used more efficiently using two of the OC selection schemes than using any of the four set rotations. Of the four OC cropping decision methods, net returns were greatest for the method that assumed average growing season precipitation and allowed selection from all possible crop choices. The net returns from this system were not different from net returns from WF (CT) and WF (NT). Cropping frequency can be effectively increased in dryland cropping systems by use of crop selection rules based on water use/yield production functions, measured available soil water, and expected precipitation.  相似文献

Five insecticide application regimes were evaluated for two years to determine their influence on the development of esfenvalerate resistance by the Colorado potato beetle (CPB),Leptinotarsa decemlineata (Say). These included sequential applications of esfenvalerate; two alternated uses of esfenvalerate, azinphosmethyl, endosulfan, oxamyl, andBacillus thuringiensis or cryolite; an IPM program; and one early season esfenvalerate application. Both CPB adults and first instars were tested for esfenvalerate resistance before and after insecticide applications. The LC50’s in the early 1992 season were correlated with values in the late 1991 season. However, over the winter, esfenvalerate resistance of CPB adults decreased, but was relatively stable for first instars. Esfenvalerate LC50’s late in the season were generally higher than values early in the season. Increases in esfenvalerate LC50’s were correlated with the number of esfenvalerate applications made in these fields during the growing season, with the greatest increase found in the sequential esfenvalerate treatment.  相似文献
