丁吡吗啉是一种结构新颖的杀菌剂,化学名(E)-3-(2-氯吡啶-4-基)-3-(4-叔丁基苯基)-丙烯酰吗啉。室内生物测定结果表明,在离体条件下,丁吡吗啉对致病疫霉Phytophthora infestans、辣椒疫霉Phytophthora capsici、立枯丝核菌Rhizoctonia solani、古巴假霜霉Pseudoperonospora cubensis等重要植物病原菌均有很好的抑制活性,其对致病疫霉、辣椒疫霉、立枯丝核菌菌丝生长的抑制中浓度(EC50)分别为1.38±0.06、0.72±0.05和4.44±0.03 μg/mL,对古巴假霜霉菌孢子囊萌发的抑制中浓度为5.33±0.05 μg/mL;但其对尖镰孢萎蔫专化型菌Fusarium oxysporum f.sp.vasinfectum、轮纹大茎点菌Macrophthoma kawatsukai、玉蜀黍赤霉Gibeberlla zeae的菌丝生长无抑制效果。用400 μg/mL的丁吡吗啉药液喷雾处理番茄幼苗,分别在施药后1、3和8 d接种致病疫霉的游动孢子,其对番茄晚疫病的防效分别为94.2%、90.8%和78.1%,说明丁吡吗啉在防治番茄晚疫病中具有较好的保护作用和一定的持效期。  相似文献

Three diagrammatic grading keys were designed for the assessment of the severity of late blight (caused by Phytophthora infestans ) in tomato leaves. Simplified and broad keys considered, respectively, six (3, 12, 22, 40, 60 and 77%) and eight (3, 6, 12, 22, 40, 60, 77 and 90%) levels of disease severity, whilst a modified key based on a previous proposal for potato late blight considered six levels (1, 5, 10, 16, 32 and 50%). The keys were validated by 24 evaluators who assessed digital images of tomato leaves exhibiting different areas with lesions. Evaluator errors were compared using a mixed model in which evaluators were considered as random effects and the keys and evaluations as fixed effects. The accuracy and precision of the evaluators were compared by simple linear regression between the estimated and actual values of disease severity. The repeatability of evaluators was assessed using Pearson's correlation coefficient. There was significant ( P  ≤   0·001) variability amongst the errors made by evaluators, although the precision of each of the three keys was high with a coefficient of determination (R2) of 0·96, 0·93 and 0·83 for the simplified, broad and modified key, respectively. Repeatability of estimations amongst the evaluators was adequate (correlation coefficients of 0·91, 0·91 and 0·90 for the three keys, respectively). The simplified and broad keys resulted in higher precision and accuracy for the estimation of severity than did the modified key. Since the simplified key considers a smaller number of disease severity levels, its use is recommended in the assessment of late blight in tomato leaves.  相似文献

Phomalactone, 5,6‐dihydro‐5‐hydroxy‐6‐prop‐2‐enyl‐2H‐pyran‐2‐one, produced by the fungus Nigrospora sphaerica, was tested in vitro against nine plant pathogenic fungi, and specifically inhibited the mycelial growth of Phytophthora infestans, with an MIC value of 2.5 mg litre?1. Its inhibitory activities against sporangium and zoospore germination of P infestans were similar to those against Phytophthora capsici. 采用菌丝生长速率法,测定了雷帕霉素对番茄灰霉病菌、油菜菌核病菌、水稻纹枯病菌和棉花枯萎病菌的抑菌活性,比较了嘧菌酯、丙烷脒及雷帕霉素对番茄灰霉病菌的抑菌效果,并通过电子显微镜观察了雷帕霉素对番茄灰霉病菌菌丝生长的影响。结果表明:雷帕霉素对供试4种植物病原真菌菌丝均表现出了极强的抑制活性,其中对油菜菌核病菌的抑制作用最强,EC50值为2.23×10-4μg/mL,对番茄灰霉病菌、水稻纹枯病菌和棉花枯萎病菌的EC50值分别为1.32×10-3、4.05×10-3及3.82×10-3μg/mL;雷帕霉素对番茄灰霉病菌菌丝的抑制活性显著高于嘧菌酯(EC50值为3.24μg/mL)和丙烷脒(EC50值为3.81μg/mL)。电镜观察发现,经雷帕霉素处理后,番茄灰霉病菌菌丝表现出提前衰老等症状。研究结果可为深入探讨雷帕霉素对植物病原真菌的作用机制奠定基础。  相似文献

Effects of inoculum concentration, wetness duration and plant age on the development of tomato early blight were evaluated in relation to host susceptibility under controlled environmental conditions. The main effect of early blight was premature defoliation, which was linearly related to the percentage of leaf area showing symptoms. As ln(inoculum concentration, conidia mL−1) increased from 6·2 to 11·5, the percentages of leaf area affected and of defoliation increased linearly. Four h of leaf wetness after inoculation were sufficient to initiate the disease on plants of hybrid Skala RZ but not on those of cv. Rio Rojo, for which at least 6 h leaf wetness were needed. As wetness duration increased up to 24 h, there was an increase in the percentage leaf area showing symptoms and in the percentage of defoliation, but thereafter there was no significant increase in either parameter. Tomato plants were susceptible to Alternaria solani at all growth stages, but susceptibility increased as plants matured. There were no significant differences in susceptibility between tomato cultivars and hybrids.  相似文献

A survey was made on the occurrence of soilborne Phytophthora species in 35 oak stands on a range of geologically different sites in Bavaria. The most widespread species were P. quercina , P. cambivora and P. citricola . Seven other Phytophthora species were isolated infrequently. The fine root systems of 106 healthy and 111 declining mature trees of Quercus robur and Q. petraea were intensively investigated. The results indicate that, depending on the site conditions, at least two different complex diseases are referred to under the name 'oak decline'. On sites with a mean soil pH (CaCl2)  3·5 and sandy-loamy to clayey soil texture Phytophthora spp. were commonly isolated from rhizosphere soil, and highly significant correlations existed between crown transparency and various root parameters. Oaks with P. quercina or other Phytophthora spp. in their rhizosphere had markedly higher levels of fine root damage than oaks without Phytophthora spp., and were subject to a relative risk of severe crown symptoms of 2·1 and 2·8, respectively. In contrast, in stands with sandy to sandy-loamy soils and a mean soil pH  3·9, Phytophthora spp. were not found. In these stands, correlations between crown transparency and various root parameters were either less significant or not significant. It is concluded that Phytophthora species are strongly involved in oak decline on sandy-loamy to clayey sites with a mean soil-pH (CaCl2)  3·5.  相似文献

 枯草芽胞杆菌NCD-2菌株及其无细胞发酵液对多种植物病原真菌具有较强的拮抗活性。为了明确NCD-2菌株抑菌作用机理,本研究利用快速蛋白液相色谱技术(FPLC)结合抑菌活性测定,对NCD-2菌株的脂肽类抑菌物质进行分离、纯化,经质谱鉴定,该抑菌物质为芬荠素(fengycin)。从NCD-2菌株中克隆出fengycin合成酶基因fenC及其上游启动子序列,通过同源重组技术对fenC基因进行了内缺失突变,同NCD-2野生型菌株相比,fenC基因缺失突变子丧失了fengycin的合成能力,同时该突变子显著降低了对番茄灰霉病菌的拮抗活性。进一步在离体叶片上比较了NCD-2野生型菌株和突变子之间脂肽提取物和菌体细胞对番茄灰霉病的保护作用,结果发现,突变子不论是脂肽提取物还是菌体细胞显著降低了对番茄灰霉病的防治作用。  相似文献

The population of Phytophthora infestans in Brazil consists of two clonal lineages, US-1 associated with tomatoes and BR-1 associated with potatoes. To assess whether host specificity in these lineages resulted from differences in aggressiveness to potato and tomato, six aggressiveness-related epidemiological components – infection frequency (IF), incubation period (IP), latent period (LP), lesion area (LA), lesion expansion rate (LER) and sporulation at several lesion ages (SSLA) – were measured on detached leaflets of late blight-susceptible potato and tomato plants. Infection frequency of US-1 was similar on potato and tomato leaflets, but IF of BR-1 was somewhat reduced on tomato. Incubation period was longer on both hosts with US-1, although this apparent lineage affect was not significant. Overall there was no host effect on IP. On potato, BR-1 had a shorter LP (110·3 h) and a larger LA (6·5 cm2) than US-1 (LP = 162·0 h; LA = 2·8 cm2). The highest LER resulted when isolates of BR-1 (0·121 cm2 h−1) and US-1 (0·053 cm2 h−1) were inoculated on potato and tomato leaflets, respectively. The highest values of the area under the sporulation capacity curve (AUSC) were obtained for isolates of US-1 inoculated on tomato leaflets (6146) and for isolates of BR-1 on potato leaflets (3775). In general, higher values of LA, LER, SSLA and AUSC, and shorter values of LP were measured when isolates of a clonal lineage were inoculated on their original host than with the opposite combinations. There is evidence that there are quantitative differences in aggressiveness components between isolates of US-1 and BR-1 clonal lineages that probably contribute to host specificity of P. infestans populations in Brazil.  相似文献

Chemical and biological agents were evaluated for their ability to suppress root rot, caused by Phytophthora cactorum , in field-grown radiata pine seedlings in New Zealand. Trials were conducted over two seasons in an area of a forest nursery with a natural infestation of P. cactorum , and a history of root rot. In each season, symptoms of root rot developed during April, one month after root pruning, when seedlings were approximately six months old. In trial one, root rot incidence by mid July 2007 was 9·1% in untreated plots and 8·4% in plots that had been treated with metalaxyl-M/mancozeb (14 kg ha−1) at seedling emergence. Disease incidence was lowest (2·1%) in plots that received seven monthly applications of phosphorous acid (6·5 L ha−1). Other treatments, including seed coating with thiram or Trichoderma spp., and foliar applications of methyl jasmonate, did not control disease. In trial two, effects of treatment timing relative to root pruning were investigated. By late June 2008, three months after root pruning, root rot incidence was 22·2% in the untreated plots. Phosphorous acid was the most effective treatment and almost completely suppressed disease (0·1% incidence) when applied fortnightly from February until May (seven applications). Metalaxyl-M/mancozeb (15 kg ha−1) was not effective (21·4% incidence) when applied five months before root pruning. However, disease incidence was reduced when the chemical was applied one week after root pruning (14·9% incidence) and greater control was achieved (8·2% incidence) when the application rate was increased to 50 kg ha−1.  相似文献

The height of barley stunted by Rhizoctonia solani anastomosis group 8 was significantly increased by up to 72·8% after incubation for 8 days at 20°C in seedling tray tests following application of the mycoparasite Verticillium biguttatum. The pathogen and mycoparasite were applied at the rate of 1% Perlite maizemeal inoculum (w/w potting mixture) resulting in propagule densities of approximately 24·0 and 6·6 × 105 colony-forming units (cfu) per g potting mixture, respectively. Sieving (2 mm) R. solani inoculum prior to dilution in potting mixture increased the recovery of propagules from 1·2 × 2·1 × 103 cfu per g inoculum compared with recovery when inoculum was sieved after dilution. Applications of a V. biguttatum isolate from the UK (vbl) and a Dutch isolate (M73) reduced stunting to a similar extent but did not stimulate the growth of healthy plants. The height of stunted plants was significantly increased after application of V. biguttatum inoculum after 6 days if inoculated trays were preincubated for 1 day prior to planting but a similar increase was only detected after 7 days if seeds were planted immediately. The number of stunted plants which emerged after 4 days was significantly increased by treatment with V. biguttatum but preincubation had no additional effect. These results suggested that control of R. solani was effected both before and after the initiation of disease.  相似文献

In vitro inhibition of soil microorganisms by 2-phenylethyl isothiocyanate   总被引:8,自引:0,他引:8  
Isothiocyanates (ITCs) are produced by the enzymatic hydrolysis of glucosinolates in cruciferous plants and can kill fungi, oomycetes and bacteria. The effect of 2-phenylethyl ITC (2-PE ITC), the main ITC liberated from the roots of canola, was tested in vitro on a range of fungi, oomycetes and bacteria. Bacteria were generally more tolerant than the eukaryotic pathogens to 2-PE ITC, although both groups showed considerable variability in response (ED90 ranging from 0·005 to 1·5 m m for eukaryotes, and from 0·33 to greater than 3·34 m m for complete inhibition of bacterial growth). While intraspecies variability was low, interspecies variability was high within some genera. Amongst the eukaryotes, Trichoderma spp. were the most tolerant to 2-PE ITC, while other genera, including Aphanomyces , Gaeumannomyces , Phytophthora and Thielaviopsis , were very sensitive. A range of responses was exhibited by Pythium spp. and the different anastomosis groups of Rhizoctonia solani . A simple laboratory screening may be an effective way to identify candidate soilborne pathogens for control in rotation cropping systems which include a Brassica crop phase.  相似文献

The role of salicylic acid (SA) was investigated in basal defence and induced resistance to powdery mildew ( Oidium neolycopersici ) and grey mould ( Botrytis cinerea ) in tomato ( Lycopersicon esculentum ) and tobacco ( Nicotiana tabacum ). A comparison of NahG transgenic tomato and tobacco (unable to accumulate SA) to their respective wild types revealed that in tomato, SA was not involved in basal defence against O. neolycopersici but NahG tobacco showed an enhanced susceptibility to O. neolycopersici infection, the effect becoming more obvious as the plants grew older. In contrast, SA played no role in the basal defence of tobacco against B. cinerea , but seemed to contribute to basal defence of tomato against B. cinerea. Activation of the SA-dependent defence pathway via benzothiadiazole (BTH) resulted in induced resistance against O. neolycopersici in tobacco but not in tomato. Microscopic analysis revealed that BTH treatment could prevent penetration of the Oidium germ tube through tobacco leaves, whereas penetration was successful on tomato leaves irrespective of BTH treatment. In contrast, soil or leaf treatment with BTH induced resistance against B. cinerea in tomato but not in tobacco. It is concluded that the SA-dependent defence pathway is effective against different pathogens in tomato and tobacco.  相似文献

Stem canker and black scurf are diseases of potato caused by the fungus Rhizoctonia solani . Spatiotemporal experimentation and empirical modelling were applied for the first time to investigate the effect of antagonistic Trichoderma harzianum on the dynamics of soilborne R. solani on individual potato plants. Trichoderma harzianum reduced the severity of symptoms, expressed as 'rhizoctonia stem lesion index' (RSI), during the first 7 days post-inoculation when the inoculum of R. solani was placed at certain distances (30–60 mm) from the host. For example, with inoculum at 40 mm from the host, RSI was 6 and 40 with and without T. harzianum , respectively. At later observation times, the antagonistic effect was overcome. Trichoderma harzianum reduced the severity of black scurf on progeny tubers. Furthermore, the mean number of progeny tubers per potato plant was reduced by the biocontrol treatment (means of 6·5 ± 1·1 and 9·9 ± 2·7 tubers per plant with and without T. harzianum , respectively), as was the proportion of small (0·1–20·0 g) tubers (48% and 66% with and without T. harzianum , respectively). Using tomato seedlings, the plant defence activator acibenzolar-S-methyl (benzo-[1,2,3]-thiadiazole-7-carbothioic acid-S-methyl ester, ASM; Bion 50 WG) was assayed for its ability to induce resistance against Clavibacter michiganensis ssp. michiganensis , the causal agent of bacterial canker of tomato. In ASM-pretreated plants, reduction in disease severity (up to 76·3%) was correlated with lower bacterial growth (up to 68·2% lower) during the time course of infection. To understand the possible mechanism of action of ASM, alterations in the activities of peroxidase (POX) and chitinase were assessed as markers of resistance. Sensitivity of Botrytis cinerea from vegetable greenhouses to boscalid   总被引:4,自引:1,他引:4  
Between 2004 and 2006, 228 isolates of Botrytis cinerea from two regions in China were characterized for baseline sensitivity to boscalid, a new active ingredient that interferes with succinate ubiquinone reductase in the electron transport chain. The isolates showed similar sensitivity in different years and regions. Baseline sensitivities were distributed as unimodal curves with mean EC50 values of 1·07 (± 0·11) and 0·42 (± 0·05) mg L−1 for inhibition of mycelial growth and conidial germination, respectively. Laboratory studies were conducted to evaluate the risk of development of resistance to boscalid. Boscalid-resistant mutants were obtained by UV-treatment at lower frequencies and with smaller resistance factors than pyrimethanil-resistant mutants. All boscalid-resistant mutants were also significantly more sensitive to Qo inhibitors than their wild-type parents and showed reduced sporulation in vitro and pathogenicity on aubergine leaves. The results suggested that the risk of resistance developing for boscalid was lower than for pyrimethanil. However, as B. cinerea is a high-risk pathogen, appropriate precautions against resistance development should be taken. Synergism between the activity of boscalid and that of kresoxim-methyl was observed.  相似文献

The effects of osmotic water potential (Ψs) on mycelial growth and perithecial production of Monosporascus cannonballus , the cause of root rot and vine decline of melons, were examined at 25°C on potato dextrose agar (PDA) amended with KCl, NaCl or sucrose. Patterns of the growth responses of four isolates to decreasing Ψs were similar for each of the osmotica. Compared with growth on nonamended PDA (−0·3 MPa), growth of all isolates increased as Ψs was reduced to −0·8 MPa. Maximum growth occurred at Ψs values of −0·6 to −0·8 MPa. Growth was not reduced below that on nonamended PDA until Ψs was reduced to −1·8 MPa, and a 50% reduction in growth did not occur until Ψs was reduced to < −2·5 MPa. Reproduction was much more sensitive to reduced Ψs than was mycelial growth, and perithecia were produced only at Ψs ≥ −0·7 or −0·8 MPa on PDA amended with KCl or NaCl, respectively. Three isolates produced perithecia on PDA amended with sucrose only at Ψs ≥ −0·6 MPa, but the fourth isolate produced perithecia at ≥ −1·9 MPa. Colonization of the xylem early in disease development may provide an essential source of water for subsequent reproduction in the root cortex during plant senescence. Postharvest cultivation to expose and desiccate roots may prevent reproduction even when temperatures lethal to hyphae are not attained.  相似文献

Pink rot of potato, most commonly caused by Phytophthora erythroseptica , is a major field and post-harvest problem in southern Idaho, USA, particularly since 1998 when isolates resistant to the phenylamide fungicide metalaxyl-M (mefenoxam) were detected. Isolates of P. erythroseptica were collected from infected tubers in 2001 and 2002 from six Idaho counties and tested for resistance to metalaxyl-M on amended agar. Metalaxyl-M resistant (MR) and metalaxyl-M-sensitive (MS) isolates were identified in six counties; 160 isolates were highly resistant, seven moderately resistant and 57 sensitive to metalaxyl-M with mean EC50 values of 182, 23 and 0·5 mg L−1 ai metalaxyl-M, respectively. Mycelial growth rates and oospore production in agar were assessed for 20 MS and 20 MR isolates at 10, 15, 20, 25 and 30°C. Growth rates of MR isolates were between 2·5 and 3·1 times greater ( P  < 0·05) than those of MS isolates at 10, 15, 20 and 25°C, and oospore production was between 6·8 and 20·5 times greater ( P  < 0·0001) for MR than for MS isolates at the same temperatures. Colony growth in V8 broth at 18°C was greater for MR than MS isolates ( P  < 0·0032). However, zoospore production at 18°C was greater for MS than for MR isolates ( P  < 0·0109), and zoospore production m m −1 of colony circumference was also greater for MS than for MR isolates, 14 191 and 9959, respectively ( P  = 0·0109). Sexual reproduction of MR isolates in nature may be greater than MS isolates, but MS isolates may be more asexually fit based on the fitness parameters studied.  相似文献

Antifungal activity of oligochitosan against nine phytopathogens was investigated in vitro. Oligochitosan was more effective than chitosan in inhibiting mycelial growth of Phytophthora capsici and its inhibition on different stages in life cycle of P. capsici was observed. Rupture of released zoospores induced by oligochitosan was reduced by addition of 100 mM glucose. The effects of oligochitosan on mycelial growth and zoospore release, but not zoospore rupture, were reduced largely when pH value was above 7. The ultrastructural study showed that oligochitosan caused distortion and disruption of most vacuoles, thickening of plasmalemma, and appearance of unique tubular materials. Plasmalemmasomes in hyphal tip cells were not found in the presence of oligochitosan. These results suggest polycationic nature of oligochitosan contributes only partly to its antifungal activity and multiple modes of action of oligochitosan exist including the disruption of endomembrane system.  相似文献

Spray mixtures consisting of the plant activator BABA (DL-3-aminobutyric acid) and the protectant fungicide mancozeb were significantly more effective than BABA or mancozeb alone in controlling late blight (Phytophthora infestans) in potato and tomato and downy mildew (Pseudoperonospora cubensis) in cucumber. A mixture composed of 5 parts BABA and 1 part mancozeb (w/w, a.i.) exhibited a higher synergy factor than the 1+1 or the 1+5 (BABA + mancozeb) mixtures. No synergistic interaction was detected between BABA plus mancozeb in controlling sporangial or cystospore germination, nor mycelial growth ofP. infestans in vitro. 为明确单端孢菌素对主要作物病原菌的抑菌活性和田间防效,采用孢子萌发抑制测定法、菌丝生长速率法和田间药效常规试验方法,测定了单端孢菌素在不同质量浓度下对作物病原菌的室内毒力及田间药效。结果表明:单端孢菌素对供试16个病原菌的MIC为1.9~15.5mg/L,对辣椒炭疽病菌、番茄早疫病菌、水稻恶苗病菌、水稻稻瘟病菌、番茄灰霉病菌、玉米小斑病菌和马铃薯晚疫病菌等7个病原菌的EC50分别为5.93、7.90、9.27、10.78、13.34、14.95和16.50mg/L,500mg/L质量浓度对番茄灰霉病、番茄早疫病、辣椒炭疽病、马铃薯晚疫病、玉米小斑病和水稻稻瘟病的防治效果分别为78.98%、81.29%、85.28%、72.42%、78.99%和72.74%。单端孢菌素具有较强的抑菌活性和较广的抑菌谱,对几种主要作物真菌和卵菌病害具有较好的田间防治效果,应用前景广。  相似文献
