Pathogenesis of caseous lymphadenitis in sheep and goats   总被引:7,自引:0,他引:7  
The progress of caseous lymphadenitis in sheep and goats is described as involving primary wound infection, lymphatic and haematogenous dissemination, and secondary infection of lymph nodes and various visceral organs. This is followed by elimination or containment of infection, the latter being represented by the characteristic caseous lesions. Resistance to infection is complex involving components of both the specific and non-specific host response. The role of these in pathogenesis is discussed. Bacterial factors including the phospholipase exotoxin and cytotoxic lipid contribute to pathogenesis at a local level with little likelihood of systemic effect in the natural disease. In a disease such as caseous lymphadenitis, lesion site, size or number do not indicate severity of disease but rather, reflect the ability of the host to mount a cellular response to infection.  相似文献

羊口疮是一种人兽共患传染病,主要由羊口疮病毒(Orf virus,OrfV)感染所致,目前尚无特效药物可治疗。囊膜蛋白是OrfV的主要抗原性蛋白,其机理主要是为病毒侵入宿主细胞创造一系列条件,同时降低宿主免疫力,增强了病毒的毒力。当病毒侵入宿主体内后,宿主会产生抗病毒免疫应答来清除病毒粒子,包括特异性体液免疫和非特异性细胞免疫应答,但主要以非特异性的细胞免疫应答为主。宿主对病毒会产生抗病毒免疫应答,同时病毒也会对宿主的免疫应答形成一种免疫逃避机制,通过该机制来躲避宿主免疫细胞对病毒粒子的捕获和清除,为病毒粒子在宿主体内的增殖、成熟及增强病毒毒力创造了各种条件。作者归纳总结了近年来羊口疮病毒与宿主互作的研究,阐述了OrfV囊膜蛋白的生物学功能、宿主的抗病毒免疫应答、病毒在宿主体内的免疫逃避机制和OrfV的其他一些毒力因子的致病作用,以期为羊口疮病毒的致病机制及疫苗防制研究提供参考。  相似文献

The acute phase proteins (APP) form part of a non-specific host response to inflammation. They may be induced by a range of different causes, including infection, inflammation, cancer and trauma. As they form part of the earliest response to such insults, they have potential for early identification of disease. In people, APP levels have been shown to correlate both with the extent of disease and also the prognosis in several forms of neoplasia, including prostate, oesophageal and colorectal cancer. As such, they can be used as prognostic and monitoring tools. To date, similar studies in veterinary patients have been limited, largely retrospective in nature and many are non-specific for tumour type. The purpose of this study was to evaluate a panel of four APPs in dogs with naturally occurring mast cell tumours (MCTs) and sarcomas to identify in the first instance whether increased levels of individual APPs, or identifiable combinations of APPs, was linked with the presence of disease. In the patients with MCTs, C-reactive protein (CRP) and α-1 acid glycoprotein levels increased, with a concurrent drop in serum amyloid A levels. In the sarcoma patients, CRP, α-1 acid glycoprotein and haptoglobin were increased. These findings suggest that specific solid tumour types in dogs may be associated with specific changes in APP profiles.  相似文献

In order to monitor the progress of New Zealand's hydatids eradication campaign, a specific, serological, diagnostic test is required to identify infected sheep. An indirect haemagglutination test, using pyruvic aldehyde-stabilized sheep erythrocytes as the antigen carrier, was developed for the serodiagnosis of larval cestode infections of sheep. Using cyst fluid from Echinococcus granulosus, Taenia hydatigena and T. ovis as the antigens in this test, it was shown that the larval cestode species, responsible for an infection in sheep harbouring a single specific infection, could be identified by the higher titre given with the homologous antigen, in comparison to that given with the heterologous antigens. Sera of sheep infected with two or more species were also tested by this method, and the only specific infections to be diagnosed by differential titres were those due to the presence of live E. granulosus cysts. These antigens cannot be used for the diagnosis of specific larval cestode infections in the field because of the cross-reactivity between cyst fluids. However, the test did show that infection with larval cestodes could be diagnosed on a non-specific basis.  相似文献

Staphylococcus aureus is the most important and prevalent contagious mammary pathogen; it causes clinical and subclinical intramammary infection with serious economic loss and herd management problems in dairy cows. In vitro studies have shown that Staphylococcus aureus adheres to mammary epithelial cells and extracellular matrix components and invades into mammary epithelial as well as other mammary cells. Staphylococcus aureus strains from intramammary infection produce several cell surface-associated and extracellular secretory products. The exact pathogenic roles of most of the products and their effects on adhesion and invasion are not well evaluated. It is also known that mammary epithelial cell-associated molecules and extracellular matrix components interact with S. aureus during the pathogenesis of mastitis, but their roles on adhesion and invasion have not been characterized. The adhesion of S. aureus to epithelial cells may involve non-specific physicochemical interactions and/or specific interactions between bacterial cell-associated ligands and host cell surface receptors. In vitro adhesion depends on the S. aureus strain, the growth phase of the bacteria, the growth medium and the origin of the epithelial cells. Adhesion is hypothesized to be a prerequisite and crucial early step for mammary gland infection. Staphylococcus aureus invades mammary epithelial cells. It also invades other cells such as endothelial cells and fibroblasts. Bacteria are found enclosed in membrane bound vacuoles in the cytoplasm of mammary epithelial cells. Recent observations indicate that S. aureus escapes from the phagosome into the cytoplasm and induces apoptosis. The invasion into mammary epithelial cells may occur through an endocytic process that requires involvement of elements of the cytoskeleton or by direct binding of bacteria to epithelial cells through a process mediated by specific receptors that needs de novo protein synthesis by both cells. Thus, the recurrent subclinical infection may result from this intracellular existence of bacteria that are protected from host defenses and effects of antibiotics. This review emphasizes on recent findings on S. aureus adhesion to mammary epithelial cells and extracellular matrix components and invasion into mammary epithelial cells.  相似文献

Recent studies have shown that fish are able to mount protective immune responses against various parasites. One of the best characterized parasite-host system in this context is the ciliate Ichthyophthirius multifiliis (Ich) parasitizing a range of freshwater fishes. Both specific and non-specific host defence mechanisms are responsible for the protection of fish against challenge infections with this ciliate. The specific humoral components comprise at least specific antibodies. The non-specific humoral elements included are the alternative complement pathway and probably lectins. Cellular factors involved in the specific response are B-cells and putative T-cells. The non-specific effector cells recognized are various leukocytes. In addition, goblet-cells and mast cells (EGC-cells) may have a function. The NCC-cell (suggested analogue to NK-cells in mammals) seems to play a role in the non-specific response. This well documented protective response in freshwater fishes against Ich has urged the development of anti-parasitic vaccines. Indeed, such products based on formalin killed parasites have been developed and found to offer the vaccinated host a satisfactory protection. However, the collection of parasites for vaccine production is extremely laborious. It involves keeping infected fish due to the fact that in vitro propagation of the parasite is still insufficiently developed. Gaining knowledge of amino acid sequences and its encoding DNA-sequences for the protective antigens (i-antigens) in the parasite was a major breakthrough. That achievement made it possible to produce a recombinant protein in E. coli and preliminary results indicated a certain protection of fish vaccinated with this product. Recent work has shown that the free-living and easily cultivated ciliate Tetrahymena can be transformed and express the i-antigen. This path seems to be promising for future development of vaccines against Ich. A novel approach in fish is the development of DNA-vaccines. Successful DNA-vaccination trials have been conducted in fish against viral infections and the technology also makes it possible to develop a DNA-vaccine against Ich. Other approaches to immuno-protection against Ich have been the use of heterologous vaccines. Thus, both bath and injection vaccination using live or killed (un-transformed) Tetrahymena has been reported to offer treated fish a certain level of protection. Such protection could be explained by non-specific reactions and the efficacy and duration of this vaccination type should be further evaluated.  相似文献

The purpose of the present study was to investigate whether dietary antibiotic-induced changes in the fish intestinal microbiota altered host physiological responses to the infection with Aeromonas hydrophila in hybrid tilapia (Oreochromis niloticus ♀ × O. aureus ♂). After an 8-week induction period with an antibiotic-supplemented or antibiotic-non-supplemented diet, 160 hybrid tilapias in 16 tanks were each injected with phosphate buffered saline (PBS) or A. hydrophila at a dose of one-half of the LD(50). Then, all of the diets were changed to a prebiotic-supplemented one for the sequential 8-week response period. Parameters including production, gut microbial diversity and count, and non-specific immunity were determined at the end of the response period. Our results showed that A. hydrophila infection had no effects on the growth and diet conversion of tilapia, but it caused the decrease of the gut bacterial count, the number of visual bands, and the Shannon diversity and equitability indexes of gut bacteria in antibiotic-non-supplemented fish based on PCR-DGGE fingerprints. Infection with A. hydrophila reduced the gut bacterial evenness (lower Shannon equitability index), and slightly improved the gut bacterial richness (more visual bands) in antibiotic-supplemented tilapia. In addition, A. hydrophila infection affected non-specific immunity such as serum lysozyme activity and serum alternative complement pathway (C3 and C4) activities regardless of hybrid tilapia fed antibiotic-supplemented diets. These changes varied based on the intestinal microbial status of the fish before infection with A. hydrophila.  相似文献

Summary Acute febrile diseases are characterized by specific and non-specific symptoms. The non-specific responses are presented under the headings: fever, inflammation and pain, experimental models for investigating febrile reactions, haematologic changes, blood biochemical changes, cardiovascular effects, changes in gastric function, and the effects of fever upon pharmacokinetics of drugs. It was the purpose of this review to describe present concepts of thermoregulation and fever, the associated reactions produced by bacterial pyrogens and the mechanisms of these reactions. The available data illustrate, that many questions have not yet been clearly answered. However, the entire field of research involving endogenous substances, such as interleukin-1, is now moving ahead with great speed. Furthermore, there is some evidence which suggests that fever and the associated lower plasma zinc and iron levels act together as a co-ordinated non-specific host defence mechanism. Since experimental fever has a distinct effect upon the pharmacokinetics of drugs, more attention should be given to this aspect.  相似文献

The host cell-mediated immune response was examined following experimentally-induced infection of rats with Dermatophilus congolensis, the causal agent of the skin disease dermatophilosis. Mononuclear cells (MC) isolated from Wistar rats 10 days following the induction of a third infection underwent a strong and specific proliferative response, as assessed by a [3H]thymidine incorporation assay, when cultured with various concentrations of inactivated D. congolensis cocci. Using specific monoclonal antibodies in an indirect fluorescent antibody test, this in vitro response was found to be characterised by a large expansion of the W3/25 (T-helper phenotype) population to form 56% of the total. Finally, the primed and stimulated MC were assessed for their ability to produce factors capable of inhibiting macrophage migration. The culture supernatants of D. congolensis-stimulated MC from infected rats caused significant migration inhibition of normal rat peritoneal exudate cells, whilst the supernatants of similarly-stimulated MC from naive rats failed to cause significant inhibition. The results show that a MC subpopulation becomes primed following experimentally-induced infection with D. congolensis and becomes activated after subsequent, in vitro, exposure.  相似文献

A rich body of work has reported levels of infection with Toxocara species in definitive hosts, and the frequency of eggs in the environment, in many different regions and situations. These have greatly increased our understanding of the relationship between egg excretion from companion and wild animals and the risk of human infection by inadvertent ingestion of eggs from soil and other environmental reservoirs. Nevertheless, it is difficult to compare studies directly because of vagaries in sampling and laboratory methods, a preponderance of prevalence rather than abundance data, and a lack of studies that systematically sample different sympatric definitive host populations. Such comparisons could be instructive, for example to determine the relative contributions of different definitive host populations and categories to environmental contamination in specified areas, and hence guide priorities for control. In this article we use estimates of host density and infection levels in the city of Bristol, UK, as a case study to evaluate the relative contribution of sympatric cats, dogs and foxes to overall environmental contamination with eggs. Results suggest that dogs, especially those less than 12 weeks of age, dominate total egg output, but that this is modified by degree of access to public areas and removal of faeces, such that foxes could take over as the primary source of eggs. Results and conclusions are likely to differ among specific locations. The general aim is to show how an improved quantitative framework for epidemiological studies of Toxocara spp. egg contamination can help to advance understanding and the effectiveness of control strategies in future.  相似文献

The protozoan parasite Toxoplasma gondii is the causative agent of the zoonosis toxoplasmosis. In sheep and goats, it is one of the most prevalent causes of infectious abortion. Also in pregnant women, a primary infection can result in miscarriage. Humans acquire the infection either by ingestion of oocysts excreted by cats, the definitive host of the parasite, or by eating raw or undercooked meat from latently infected animals (Dubey & Beattie 1988). In Sweden, toxoplasmosis is a notifiable disease, and cases of clinical disease in humans as well as animals must be reported. In both veterinary and human medicine serological assays based on detecting the humoral antibody response of the host against the parasite are used as diagnostic tools. So far, solid phase assays, such as the indirect fluorescent antibody test (IFAT) and the enzyme-linked immunosorbent assay (ELISA), have been widely used to diagnose T. gondii infection in many species including cats, pigs and sheep (Dubey & Beattie 1988). However, both IFAT and ELISA require appropriate anti-species specific immunoglobulins (Ig) that must be carefully evaluated for each species prior to use. This makes these assays complicated and time consuming. Consequently, alternative, simpler methods that do not require specific antisera would be of great value. The direct agglutination test (DA), which is based on the principle that formalin-treated organisms agglutinate in the presence of specific IgG antibodies, is such an assay (Fulton & Turk 1959). The DA-test is widely used in human medicine as a screening test for T gondii infection but it has not yet been thoroughly evaluated for use in veterinary medicine (Uggla & Buxton 1990).  相似文献

The purpose of the study was to determine the influence of the Bioimmuno preparation administered in feed and/or immunisation with the Respisure One vaccine on the development of selected indices of non-specific and specific humoral immune response against Mycoplasma hyopneumoniae (Mhp) infections in pigs. The study was performed on 28 piglets at the age of 4 weeks, divided into four equal groups. The biopreparations were administered according to the following pattern: group I--Bioimmuno (IFI Olsztyn, Poland) with feedstuff at amount of 1 kg/50 kg of feed for 48 h before vaccination with Respisure One (Pfizer) on day 28 of life; group II--Bioimmuno only (1 kg/50 kg feedstuff) for 48 h before vaccination with Respisure One of groups I and III; group III--Respisure One only on day 28 of life (2 ml/animal i.m.) and group C (control)--PBS (2 ml/animal i.m.) simultaneously with vaccination of groups I and III. On days 0, 3, 7, 14 and 21 after immunomodulation and/or immunisation, the serum level of gamma-globulins, the activity of lysozyme (LSM) as well as the serum levels of cytokines: interferon gamma (IFNgamma), interleukin 1 beta (IL-1beta) and interleukin 6 (IL-6) were determined, as indices of non-specific immune response against Mhp infections in pigs. The study has revealed that in piglets after weaning the application of the Bioimmuno and/or Respisure One biopreparations improves the non-specific immunity parameters stimulating an increase in serum levels of gamma-globulins, lysozyme and cytokines (IFNgamma, IL-1beta, IL-6), while late appearing seroconversion confirms a minor role of specific humoral immunity in the protection against Mhp infection.  相似文献

It is the purpose of this review to describe present concepts of fever and the associated reactions produced by bacterial pyrogens and the mechanisms of those reactions. The available data indicate that interleukin-1 is a key mediator of host responses to bacterial infections. However, many questions have not yet been clearly answered. In particular, more detailed studies of the processes within the CNS responsible for anorexia are still required.  相似文献

Serum antibody responses in sheep naturally or experimentally infected with Echinococcus granulosus and/or other larval cestodes were examined using an enzyme-linked immunosorbent assay (ELISA) with various antigens prepared from sheep hydatid cyst fluid ( SHCF ). Serum donors included: sheep experimentally infected with E. granulosus and their age-matched non-infected controls; sheep experimentally infected with other helminth parasites; sheep naturally infected with E. granulosus both from Tasmania and the Australian mainland; sheep from Tasmania naturally infected with larval cestodes other than E. granulosus; and naturally reared sheep completely free from infection with larval cestodes. Attempts were made to eliminate serological reactions which were not specific for E. granulosus by using a series of antibody affinity chromatography steps to deplete crude SHCF antigen; these included adsorption with a monoclonal antibody, 3EgH 29-2, removal of host IgG using rabbit anti-sheep IgG antibody, and removal of antigens which bound non-specifically to normal sheep immunoglobulin. The final affinity-depleted antigen product was designated AD SHCF . Specific serological reactivity in infected sheep was very low. Affinity depletion of SHCF using 3EgH 29-2 did not appear to increase the specificity of serological diagnosis of E. granulosus infection when experimentally infected sheep were compared with their non-infected controls provided the latter were age-matched with experimental animals. The other affinity adsorption steps significantly reduced non-specific background binding to antigen by normal sheep serum. Despite this reduction in background in the ELISA, only low levels of antibody could be detected in naturally-infected sheep.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Chronic subclinical mastitis represents a vicious cycle of constant tissue irritation by bacteria and inflammatory response of the host. Antibiotics have proven relatively inefficient in eliminating chronic mastitic infections. The use of anti-inflammatory agents in mastitis is controversial. Theoretically, the inflammation should not be suppressed as this is a host defence mechanism of the body. However, because it is uncertain whether the infection or chronic inflammation causes more harm to the tissue, there have been efforts to use anti-inflammatory agents such as glucocorticoids and antipyretic analgetics. The problem is that there is insufficient evidence as to which inflammatory mediators (histamine, kinins, prostaglandins, leukotrienes, PAF, interleukin-1 etc.) alone or in combination are important in mastitis.  相似文献

Sporozoites of Cryptosporidium parvum were incubated in 1:10 dilutions of immune or non-immune, heat-inactivated lamb serum specimens or serum fractions. The infectivity of treated sporozoites was assessed by inoculating them, per rectum, into five-day-old rats followed by histological examination of their intestines at either three or five days after infection. The infectivity of sporozoites treated with heat-inactivated whole sera was greatly reduced. This neutralisation had both specific and non-specific components. The former was associated with the IgG fraction of hyperimmune serum raised against sporozoites and the latter with a heat-stable, non-dialysable component present in both IgG-depleted hyperimmune serum and uninfected, gnotobiotic serum.  相似文献

Feline leukemia virus is a naturally occurring, contagiously transmitted and oncogenic immunosuppressive retrovirus of cats. The effects of FeLV are paradoxical, causing cytoproliferative and cytosuppressive disease (eg, lymphoma and myeloproliferative disorders vs immunodeficiency and myelosuppressive disorders). In the first few weeks after virus exposure, interactions between FeLV and hemolymphatic system cells determine whether the virus or the cat will dominate in the host/virus relationship--persistent viremia and progressive infection or self limiting, regressive infection will develop. The outcome of these early host/virus interactions is revealed in the diagnostic assays for FeLV antigenemia and viremia. The latter, in turn, predict the outcome of FeLV infection in cats. Known host resistance factors include age and immune system functional status. Known virus virulence factors are magnitude of exposure and virus genotype. Molecular analysis of FeLV strains indicated that natural virus isolates exist as mixtures of closely related virus genotypes and that minor genetic variations among FeLV strains can impart major differences in pathogenicity. The genetic coding regions responsible for cell targeting and specific disease inducing capacity (eg, thymic lymphoma, acute immunosuppression, or aplastic anemia) have been mapped to the virus surface glycoprotein and/or long terminal repeat regions for several FeLV strains. Infection by specific FeLV strains leads to either malignant transformation or cytopathic deletion of specific lymphocyte and hemopoietic cell population, changes that prefigure the onset of clinical illness. Another notable feature of the biology of FeLV is that many cats are able to effectively contain and terminate viral replication, an important example of host immunologic control of a retrovirus infection and a process that can be selectively enhanced by vaccination. Thus, FeLV infection serves as a natural model of the multifaceted pathogenesis of retroviruses and as a paradigm for immunoprophylaxis against an immunosuppressive leukemogenic retrovirus.  相似文献

Bacteria from genera Chlamydia (Ch.) and Chlamydophila (Chl.) are very pathogenic and may infect humans and animals. They also may cause latent infection, especially in animals. In this paper we discuss the non-specific and specific cellular and humoral immunity in farm animals, after infection or immunisation with Chlamydia sp. and Chlamydophila sp. bacteria. It has been shown, that the infection or immunisation with the microorganisms influenced the activity of polimorphonuclear cells (PMN) and mononuclear cells (MN) in the process of phagocytosis. It has also been shown that the bacteria influenced the amount and activity of lysozyme, activities of myeloperoxidase and lysosomal enzymes. Infection or immunisation with the microorganisms was demonstrated to affect numbers of lymphocytes T and B and those of their subpopulation as well as the activity of cytokines and levels of serum and secreted immunoglobulins. The changes were detected just a few hours after infection or immunisation and persisted for a few days to a few decades.  相似文献

Enteric redmouth disease (ERM) is a serious septicemic bacterial disease of salmonid fish species. It is caused by Yersinia ruckeri, a Gram-negative rod-shaped enterobacterium. It has a wide host range, broad geographical distribution, and causes significant economic losses in the fish aquaculture industry. The disease gets its name from the subcutaneous hemorrhages, it can cause at the corners of the mouth and in gums and tongue. Other clinical signs include exophthalmia, darkening of the skin, splenomegaly and inflammation of the lower intestine with accumulation of thick yellow fluid. The bacterium enters the fish via the secondary gill lamellae and from there it spreads to the blood and internal organs. Y. ruckeri can be detected by conventional biochemical, serological and molecular methods. Its genome is 3.7 Mb with 3406–3530 coding sequences. Several important virulence factors of Y. ruckeri have been discovered, including haemolyin YhlA and metalloprotease Yrp1. Both non-specific and specific immune responses of fish during the course of Y. ruckeri infection have been well characterized. Several methods of vaccination have been developed for controlling both biotype 1 and biotype 2 Y. ruckeri strains in fish. This review summarizes the current state of knowledge regarding enteric redmouth disease and Y. ruckeri: diagnosis, genome, virulence factors, interaction with the host immune responses, and the development of vaccines against this pathogen.  相似文献
