The detection, by means of faecal egg count reduction tests and larval cultures, of two cases of multiple anthelmintic resistance in goats and two in sheep, is reported. The former cases appeared to involve resistance to all three broad-spectrum drench groups (benzimidazole, levamisole, ivermectin) by mainly Ostertugia spp., the latter resistance to two of them (benzimidazole, levamisole) bv Trichostrondus spp. (New Zealand Veterinary Journal 38, 114–117, 1990)  相似文献

The Himalaya region of Nepal provides a habitat for the endangered snow leopard (Panthera uncia) and its principal prey species, the blue sheep (Pseudois nayaur). The aim of this study was to describe the habitat, the distribution and the population structure of blue sheep, and to estimate their contribution to the carrying capacity of snow leopard in the upper Mustang region of Nepal. Blue sheep were recorded at altitudes from 3209–5498 m on slopes with gradients of 16–60° and aspects of 40°NE to 140°SE. A total of 939 blue sheep were counted in the upper Mustang region, and 98 were counted in the Yak Kharka region of Manang district; however, upper Mustang had the lowest population density of blue sheep recorded within their distribution range in Nepal (0.86 blue sheep/km2). The results of the study show that a higher density of blue sheep is associated with greater plant species diversity. The most important species present in the blue sheep habitat were Kobresia pygmaea, Artemesia spp., Lonicera spp., Lancea tibetica, Poa spp., Astragalus spp. and Ephedra gerardiana. It is estimated that the existing blue sheep population biomass of approximately 38 925 kg in the upper Mustang region could support approximately 19 snow leopards (1.6 snow leopards/100 km2).  相似文献

Clinical parasitism associated with multigeneric resistance to benzimidazole anthelmintics was investigated in four sheep flocks during February and March 1989. The genera considered resistant were Haemonchus and Nematodirus spp. on all four farms and Trichostrongylus and Ostertagia spp. on three of the four farms. It is suggested that when investigating cases of suspected drench resistance, postdrenching worm counts are useful to help identify the resistant worm species present.  相似文献

A study was made to compare the numbers of infective ovine nematode parasite larvae on an open pastured block and an adjacent block forested with Pinus radiate at 200 stems per hectare. Each block consisted of two paddocks which had similar nematode parasite egg contamination. Infective larvae of six genera were recovered over a 12 month period viz., Trichostrongylus spp., Ostertagia spp., Cooperia spp., Nematodirus spp., Chabertia/Oesophagostomum spp. and Haemonchus sp. A significantly higher recovery of Trichostrongylus spp. (p<0.01) and Ostertagia spp. (p<0.05) from the pine forested block was probably due to the more favourable climatic environment under the pines.

The lower mean body weights of sheep grazing the pine block was considered to be due to the greater exposure to infective larvae and marginal undernutrition.  相似文献

The prevalence and level of thiabendazole resistance of sheep nematodes in the North Island of New Zealand was investigated by means of an in vitro egg-hatch technique; samples from sheep on 52 properties in six country areas were examined. Resistance of Haemonchus contortus and Trichostrongylus spp. was identified on the basis of egg hatch in 0.1 ppm thiabendazole. Evidence of resistance was obtained on 11 (21.2%) of the properties. LD 50's and resistance ratios were calculated for the resistant nematode populations found.  相似文献

A recent national survey on anthelmintic resistance in cattle and sheep in New Zealand indicated that the magnitude of the problem has increased from very low levels only a few years ago to disturbingly high levels now. There is a particular problem with multiple resistance to all three action families of anthelmintic currently available in Ostertagia (= Teladorsagia) spp in sheep, and to both macrocyclic lactones (ML) and benzimidazoles in Cooperia spp in cattle. The prevalence and extent of resistance indicate that all cattle farmers and most sheep farmers should now be using a combination anthelmintic on most occasions just to achieve effective control of all parasites. Despite this, the presence of resistant parasites has generally not been appreciated by the majority of affected farmers, possibly because most have not formally tested to determine the resistance status of nematodes on their farms.

Anthelmintics will remain the cornerstone of gastrointestinal nematode control in sheep and cattle for the foreseeable future but to ensure their continued effectiveness farmers need to be constantly aware of the need to maintain adequate reservoirs of unselected nematodes, i.e. worms in refugia, to minimise the expansion of the resistant population. High-risk practices in relation to selection of resistance need to be identified and avoided or at least their use limited. These include: treating adult animals where there is no identified need, moving newly- treated animals onto ‘clean’ pasture, and failing to effectively quarantine-drench bought-in animals. None of these are new concepts but many have not been adopted or practised. In particular, sheep farmers should endeavour to avoid treating ewes pre-lambing with long-acting anthelmintics. Farmers needs to negotiate a balance between achieving good parasite control and the sustainability of their control options.  相似文献

This study describes the immunohistochemical and histochemical phenotypes of inflammatory cells in sheep lungs infected with lungworms. A total of 20 naturally infected sheep lungs were used. Protostrongylus spp., Muellerius capillaris, Neostrongylus linearis, and Cystocaulus ocreatus were the chief organisms determined from such lesions, which were of a chronic nature. All the lungs had many developmental stages of the parasites and a similar inflammatory response, which included numerous mast cells, eosinophils, T cells, B cells, dendritic cells, and macrophages. In the bronchial and interstitial tissues, the inflammatory cells were dominated by MHCII, CD1, CD4, CD5, CD14, CD21, IgM, and CD172a positive cells, whereas CD2 and WC1 positive cells were detected less. The data provided additional evidence that subsets of inflammatory cells were included within ovine lungs infected with lungworms; however, understanding the entire immune-response process and development of resistance to lungworms in sheep remain to be clearly elucidated.  相似文献

Abstract

AIM: To evaluate resistance to anthelmintics containing abamectin, levamisole, and oxfendazole (AB-LEV-OX), derquantal and abamectin (DEQ-AB), moxidectin, and monepantel in naturally acquired gastrointestinal nematodes present on a sheep farm.

METHODS: Faecal nematode egg count reduction tests (FECRT) were carried out on lambs that were approximately 7 months-old and infected with naturally acquired nematodes. Lambs were randomly allocated to one of five groups (n=15 per group): treatment with 2?mg/kg derquantel and 0.2?mg/kg abamectin; 0.2?mg/kg abamectin, 8?mg/kg levamisole HCl and 4.5?mg/kg oxfendazole; 2.5?mg/kg monepantel; 0.2?mg/kg moxidectin, or no treatment. Post-treatment samples were collected 12 days later. Abomasa and small intestines were collected from two slaughtered lambs from each of the DEQ-AB, AB-LEV-OX, moxidectin and control groups 15 days after treatment, for nematode counting.

RESULTS: The FECRT demonstrated that efficacy was 90.3 (95% CI=84.2–94.1)% for AB-LEV-OX, 54.5 (95% CI=28.4–71.1)% for moxidectin, 99.2 (95% CI=97.4–99.8)% for DEQ-AB and 100% for monepantel, across all genera. For Trichostrongylus spp. efficacy was 85.5% for AB-LEV-OX and 46.7% for moxidectin. Haemonchus spp. were fully susceptible to all treatments. Post-treatment nematode counts indicated that the resistant Trichostrongylus spp. were from the small intestine.

CONCLUSIONS: Anthelmintic resistance to both AB-LEV-OX and moxidectin was present in the Trichostrongylus genus on a commercial sheep farm. Monepantel and DEQ-AB were both effective against Trichostrongylus spp. based on FECRT results.

CLINICAL RELEVANCE: This finding of resistance to an AB-LEV-OX triple-combination anthelmintic in the Trichostrongylus genus in sheep in New Zealand further limits anthelmintic treatment options available, and calls into question whether this combination is suitable for use as a quarantine treatment.  相似文献

SUMMARY Three years after the start of an experiment to assess the merits of thiabendazole (TBZ) treatment of trichostrongylid parasites in weaner sheep, field isolates of Ostertagia spp and Trichostrongylus spp were made from weaner sheep treated under one of three treatment schemes. Treatment frequencies were “nil”, “planned” (5 or 6/year) and “regular” (every 3 weeks). In addition an isolate was taken from a group of “tracer” sheep drenched with TBZ every 10 days. Resistance to TBZ was assessed using an in vitro egg hatch assay, pre- and post-treatment faecal egg counts and a controlled anthelmintic efficiency test. Pre- and post-treatment egg counts revealed the presence of TBZ-resistance in field isolates of mixed species. Egg hatch assays indicated a level of resistance for Ostertagia spp which was proportional to the frequency of TBZ treatment. The “planned”, “regular” and “tracer” strains of Ostertagia spp had resistance ratios for eggs of 4, 13 and 15 respectively when compared to the “nil” strain. In the anthelmintic efficiency assay treatment with 44 mg kg-1 and 88 mg kg-1 of TBZ removed 82 and 96% respectively of the total Ostertagia burden (adults and larvae) from the “nil” strain and 30 and 75% respectively from the “planned” strain. The same dose rates against the “regular” and “tracer” strains and additional rates of 132 or 176 mg kg-1 against the “tracer” strain failed to reduce the Ostertagia burden significantly. Intestinal Trichostrongylus spp from all isolates were fully susceptible to TBZ at 44 mg kg-1. Levamisole at 7.0 mg kg-1 was highly effective (99% reduction) against the “tracer” strain of Ostertagia.  相似文献

Benzimidazole resistance involving several gastrointestinal nematode genera on a single sheep farm in New Zealand is reported for the first time. A controlled slaughter trial showed that at the recommended dose rate of 12.5 mg/kg, mebendazole had efficacies of 0, 60, 66, 90, 54 and 38% against Haemonchus contortus, Ostertagia spp., Nematodirus spp., intestinal Trichostrongylus spp., Strongyloides spp. and Oesophagostomum venulosum, respectively. The relevance of such multigeneric resistance to possible future options for controlling anthelmintic resistant sheep trichostrongylids is briefly discussed.  相似文献

AIM: To determine associations between resistance of Ostertagia (= Teladorsagia) spp to macrocyclic lactone (ML) anthelmintics and history of use of anthelmintics, by type, on commercial sheep farms in temperate regions of southern South Australia and Victoria, Australia.

METHODS: Faecal egg count reduction tests (FECRTs) were conducted during a 2.5-year period (from August 2001 to January 2004) and records of the type of anthelmintic used in the 5 years preceding the FECRTs were collected from commercial sheep farms (n=103) in southern South Australia and Victoria, and data analysed retrospectively. ML resistance was defined as <95% reduction of Ostertagia spp 10–14 days after treatment with ivermectin (IVM), orally, at half the manufacturer's recommended dose rate. Use of anthelmintics in the preceding 5 and 10 years on each property was classified according to the nett number of years each of the following classes of drug had been used: IVM oral liquid (IVO), IVM controlled-release capsules (CRCs), abamectin (ABA), moxidectin (MOX) or a non-ML an- thelmintic. The prevalence of ML resistance, by property, was analysed for associations with prior use of anthelmintics.

RESULTS: Resistance by Ostertagia spp to ML anthelmintics was evident on 51/103 (49.5%) properties. The prevalence of resistance was lowest (23%) on properties on which MOX had not been used, and was significantly higher (64–77%) on properties on which MOX had been used for ≥2 of the preceding 5 years (p<0.001). In contrast, the prevalence of resistance was highest (70–74%) on the properties on which IVM, or IVM and/ or ABA, had not been used in the previous 5 years (on which the use of MOX was predominant), and was markedly lower (20– 42%) on properties that had used IVM or IVM and/or ABA for at least one of the preceding 5 years. Prevalence of resistance was higher for properties on which the only ML anthelmintic used was MOX (19/29=66%) than for those on which the only ML used was IVO (2/19=11%; p<0.001). Properties on which the only ML used was MOX were 2.72 times more likely to have resistance than properties on which the only ML used was IVO (95% confidence interval (CI) = 1.01–5.08).

CONCLUSION: Use of MOX for ≥2 of the preceding 5 years was associated with a higher prevalence of resistance to ML by Ostertagia spp on sheep farms in south eastern Australia than the use of IVO.  相似文献

AIM: To determine the aetiolog y of a recurring and severe form of infectious keratoconjunctivitis (IKC) in sheep.

METHODS: Five sheep flocks that had experienced a severe form of IKC were examined. Clinical history, conjunctival swabs and blood samples were collected from affected animals. Culture for bacteria, and also specifically for Mycoplasma and Chlamydophila spp, and detection of Mycoplasma conjunctivae DNA by polymerase chain reaction (PCR) were attempted. Serum samples were tested for antibodies to M. agalactiae, M. capricolum, M. conjunctivae and Chlamydophila spp.

RESULTS: Mycoplasma conjunctivae DNA was detected using PCR in 3/5 flocks, and in all flocks antibodies to M. conjunctivae were detected in sera. A pure growth of Branhamella ovis was cultured from conjunctival swabs from a small proportion of sheep in two flocks. No other pathogens were detected.

CONCLUSIONS: This investigation demonstrated that M. conjunctivae was a primary pathogen causing severe IKC in sheep, and is the first report of detection of this organism in sheep in New Zealand. Introduction of clinically normal carrier sheep appeared to have caused the outbreaks.  相似文献

The growing problem of antimicrobial resistance affects veterinarians on a daily basis. Antimicrobial stewardship and responsible prescribing are essential for a future with effective antimicrobials, as it is unlikely that new antimicrobials will become available for use in horses in the near future. Extended-spectrum β-lactamase (ESBL) producing Enterobacteriaceae, methicillin-resistant Staphylococcus aureus (MRSA) and Salmonella spp. are pathogens of significant concern but there are also other opportunistic pathogens such as Pseudomonas spp., α-haemolytic Streptococcus spp., Enterococcus spp. and Acinetobacter spp. which, due to their high intrinsic resistance, have limited treatment options in adult horses. It is essential that highest priority critically important antimicrobials such as ceftiofur, enrofloxacin, rifampicin and polymyxin B are used prudently in horses and ideally based on culture and antimicrobial susceptibility testing (AST). For example, the use of polymyxin B at a low anti-endotoxic dose rather than at a higher antimicrobial dose in horses for the treatment of systemic inflammatory response syndrome is a potential driver for resistance to colistin (polymyxin E), an antimicrobial used as a last resort in the treatment of multidrug resistant (MDR) Enterobacteriaceae infections in humans. Serum procalcitonin levels are used in humans to distinguish noninfectious inflammatory conditions from inflammation caused by bacteria and other infectious agents and are also used to guide cessation of antimicrobial treatment. Although no such studies have been performed in horses, this or other markers may prove to be helpful in guiding antimicrobial treatment decisions in the future. Optimising sampling techniques and good communication with the microbiology laboratory are essential for generating the accurate culture and AST results that underpin appropriate antimicrobial use. Additionally, there is clearly a need for national and international harmonisation of laboratory methods in order to improve the reliability and consistency of results reported by different laboratories.  相似文献

Evidence of Anaplasma spp. in goats and sheep in Cyprus has been demonstrated by previous research. Herein, further research was performed for the identification of the exact Anaplasma spp. resulting in the identification of Anaplasma ovis strains in all samples examined. We used a bioinformatics as well as a molecular approach (study of groEl and mps4 genes) in order to verify the validity of the results. The present study was carried out to assess the prevalence of motile Aeromonas spp. in the faeces of clinically healthy sheep, cattle and horses and evaluate their susceptibility to some anti-microbial agents. Rectal swabs from 120 sheep, 85 cattle and 20 horses were examined for Aeromonas species using alkaline peptone water (pH 8.4) as the enrichment medium and Aeromonas Selective Agar containing 5 mg/l ampicillin as the isolation medium. Identification and antibiotic resistance of motile Aeromonas strains was performed using Gram Negative Enteric ID panel. Motile aeromonads were isolated from 12 (10%) sheep, 7 (8.2%) cattle and 1 (5%) horse. Of these 20 aeromonad isolates, 13 were A. caviae, 6 were A.sobria and 1 was A. hydrophila. Aeromonas species in the faeces of livestock might pose a public health problem for humans who are in direct contact with contaminated animals. However, further studies should be performed on aeromonads relating to their transmission between animals and humans.  相似文献

Samples were collected from clinically infected carthorses in Awassa. Fungus species affecting the carthorses were identified. Eight genera of fungal groups were isolated from swabs and skin scrapes taken from symptomatic horses. These included Aspergillus spp., Histoplasma spp., Penicillinum spp., Microsporum spp., Trichophyton spp., yeast cells of Candida spp., Cryptococcus spp. and Geotrichum spp. The most frequent isolates were from the genera Aspergillus (48 %), Penicillium (39.2 %) and Trichophyton (31.6 %). Clinical findings are reported, the economic, zoonotic and pathogenetic importance of fungi causing dermatomycoses is discussed, and further studies are recommended.  相似文献

An analysis was undertaken of 177 veterinary diagnostic case submissions to two North Island and two South Island animal health laboratories for faecal egg count reduction testing in sheep during 1993 to provide some comparative data on the frequency of occurrence of anthelmintic resistance. The results suggest that resistance to anthelmintics in sheep nematodes may be more common in the South Island than the North. In both instances this mainly involved resistance to a single drench type, the benzimidazoles. The data indicated a frequency of occurrence of resistance to benzimidazole anthelmintics of 61 and 72%, to levamisole-type anthelmintics of 29 and 29% and to benzimidazole-levamisole combinations of 11 and 22% for submissions to North and South Island laboratories, respectively. In submissions from the South Island, resistance to benzimidazoles involved infections of Nematodirus spp. alone much more frequently than those from the North.  相似文献

This study is conducted to determine the occurrence and antimicrobial resistance of Arcobacter spp. isolated from clinically healthy food animals. A total of 308 samples from cattle (200) and sheep (108) were collected from Shiraz slaughterhouse, southern Iran to investigate the presence of the important Arcobacter spp. using cultivation and Polymerase Chain Reaction (PCR) methods. Antimicrobial susceptibility of Arcobacter isolates was determined for 18 antibiotics using disk diffusion method. Among 308 samples, 27 (8.7%) and 44 (14.28%) were positive for the presence of Arcobacter species with cultivation and PCR procedures, respectively. The predominant species was A. butzleri in both cattle (58.33%) and sheep (55%). In addition, concurrent incidence of the species was observed in 25% of the positive samples. All Arcobacter isolates were resistant to rifampicin, vancomycin, ceftriaxone, trimethoprim and cephalothin. The isolates showed high susceptibility to tetracycline, oxytetracycline, erythromycin, ciprofloxacin, kanamycin, amikacin, gentamicin and enrofloxacin. No significant difference among cattle and sheep isolates in resistance pattern was observed. The results indicate that cattle and sheep are significant intestinal carriers for Arcobacter spp. Moreover, tetracycline and aminoglycosides showed great effects on Arcobacter species in antibiogram test and can be used for treatment of human Arcobacter infections.  相似文献

Rodents are sources of many zoonotic pathogens that are of public health concern. This study investigated bacterial pathogens and assessed their antimicrobial resistance (AMR) patterns in commensal rodents in Qatar. A total of 148 rodents were captured between August 2019 and February 2020, and blood, ectoparasites, and visceral samples were collected. Gram-negative bacteria were isolated from the intestines, and blood plasma samples were used to detect antibodies against Brucella spp., Chlamydophila abortus, and Coxiella burnetii. PCR assays were performed to detect C. burnetii, Leptospira spp., Rickettsia spp., and Yersinia pestis in rodent tissues and ectoparasite samples. Antimicrobial resistance by the isolated intestinal bacteria was performed using an automated VITEK analyzer. A total of 13 bacterial species were isolated from the intestine samples, namely Acinetobacter baumannii, Aeromonas salmonicida, Citrobacter freundii, Citrobacter koseri, Enterobacter aerogenes, Enterobacter cloacae, Escherichia coli, Hafnia alvei, Klebsiella pneumoniae, Providencia stuartii, Proteus mirabilis, Pseudomonas aeruginosa, and Salmonella enterica. The majority of them were E. coli (54.63%), followed by P. mirabilis (17.59%) and K. pneumoniae (8.33%). Most of the pathogens were isolated from rodents obtained from livestock farms (50.46%), followed by agricultural farms (26.61%) and other sources (22.94%). No antibodies (0/148) were detected against Brucella spp., C. abortus, or C. burnetii. In addition, 31.58% (6/19) of the flea pools and one (1/1) mite pool was positive for Rickettsia spp., and no sample was positive for C. burnetii, Leptospira spp., and Y. pestis by PCR. A total of 43 (38%) bacterial isolates were identified as multidrug resistant (MDR), whereas A. salmonicida (n?=?1) did not show resistance to any tested antimicrobials. Over 50% of bacterial MDR isolates were resistant to ampicillin, cefalotin, doxycycline, nitrofurantoin, and tetracycline. The presence of MDR pathogens was not correlated with rodent species or the location of rodent trapping. Seven (11.86%) E. coli and 2 (22.2%) K. pneumoniae were extended-spectrum beta-lactamases (ESBL) producers. These findings suggest that rodents can be a source of opportunistic bacteria for human and animal transmission in Qatar. Further studies are needed for the molecular characterization of the identified bacteria in this study.

AIM: To determine the distribution of the three common Trichostrongylus spp. infecting sheep and their resistance status on farms throughout New Zealand, using PCR.

METHODS: Cultures were prepared from faecal samples from 70 farms while conducting faecal egg count reduction tests (FECRT) in lambs between 2010 and 2012. Trichostrongylus-type infective stage larvae (L3) were recovered from cultures, derived from untreated control and albendazole-, levamisole- and ivermectin-treated groups of lambs on each of the farms involved, and these were identified to species using PCR analysis of the second internal transcribed spacer region of ribosomal DNA. The species composition of the larvae present in cultures from the untreated control groups was examined across all farms to assess any potential differences in geographical distribution. In addition, the species composition of larvae cultured from the untreated and anthelmintic-treated lamb groups were compared to determine which species exhibited resistance to each of the anthelmintics used in the FECRT.

RESULTS: Of 67 farms with Trichostrongylus spp. present, 42 (63%) cultures from the untreated control groups contained all three Trichostrongylus spp. and no significant geographical patterns in their distribution were detected. Seven samples contained only one species. Irrespective of the anthelmintic efficacy levels, Trichostrongylus colubriformis dominated cultures prepared from lambs following treatment with albendazole (99.1 (95%CI?=?97?100)% of larvae) or levamisole (81.6 (95%CI?=?75.3?87.9)% of larvae), indicating the presence of widespread resistance in this species. In cultures prepared from levamisole-treated lambs, small numbers of T. axei larvae were also frequently present (5.4 (95% CI?=?1.3?12.4)% of larvae). Resistance to ivermectin was not found in any of the three Trichostrongylus spp. after PCR identification. Although larvae were identified, based on length, as being Trichostrongylus spp., for 24 of the 48 samples cultured following treatment with ivermectin, 100% of the larvae present were identified as Teladorsagia circumcincta.

CONCLUSIONS: As in previous surveys, all three Trichostrongylus spp. were common throughout New Zealand and no geographical patterns were detected in the current study. On all farms where resistance to albendazole and/or levamisole was indicated (i.e. efficacy <95%), the species identified as being resistant was T. colubriformis. Even where efficacies were >95%, T. colubriformis still tended to dominate in post-treatment cultures. While this could reflect a lower susceptibility of T. colubriformis to these anthelmintics, it seems more likely to indicate the presence of resistant genotypes in these populations. Similarly, T. axei also tended to be present after treatment with levamisole, which likely reflects a known lower susceptibility of this species to these anthelmintics.  相似文献
