A prospective study of vesicular stomatitis was conducted in two bovine herds in southeastern Mexico. In July 1987, an initial serological screening showed that 64% and 87% of the 654 cattle tested negatively to vesicular stomatitis New Jersey and Indiana antibodies, respectively, using the plaque-reduction serum neutralization test. Most seropositive animals were at least 24 months of age. Based on the initial serological screening, cohorts of seronegative and seropositive cattle were monitered (January–December 1988) for the prevalence of vesicular stomatitis virus (VSV) antibodies using an enzyme-linked immunosorbent assay (ELISA). The serological results, using ELISA, indicated that no VSV activity occured in the two study herds. The seronegative cohort of cattle did not yield a positive seroconversion pattern to either VSV Indiana or New Jersey. The seropositive cohort showed a variable antibody response pattern against the VSV. There were no clinical cases of vesicular stomatitis (VS) in the two herds. The data from the national surveillance program for vesicular diseases suggested that 1988 was a year of low VSV infection incidence in southeastern Mexico.  相似文献

An ELISA was compared with the plaque-reduction serum neutralization (PRSN) test, for detection of vesicular stomatitis virus (VSV) antibodies in cattle in a vesicular stomatitis enzootic region of Mexico. A total of 325 bovine serum samples were screened for VSV antibodies. The PRSN test was performed, using Vero cells. The ELISA contained gradient-purified VSV Indiana (Lab strain) and VSV New Jersey (Hazelhurst) as the antigens. Regression analysis and weighted kappa statistic were used to estimate measures of agreement between the 2 assays for detection of VSV antibodies. The ELISA method proved useful for serodiagnosis of vesicular stomatitis. The ELISA and PRSN test results were highly correlated for detection of VSV antibodies.  相似文献

A liquid-phase enzyme-linked immunosorbent assay (ELISA) was developed for the detection of vesicular stomatitis virus (VSV) types New Jersey (VSV-NJ) and Indiana subtype Indiana I (VSV-IND1) antibodies in the sera of naturally and experimentally infected cattle, horses and swine. Four different VSV preparations were compared for use as antigen in the ELISA: virus used in neutralization tests, complement-fixation antigen, immunodiffusion ager gel antigen and viral glycoprotein. Comparative antibody titration results of virus neutralization (VN) and ELISA showed no statistically significant difference between serum titers obtained with the four antigens to VSV-NJ (P=0.21) and VSV-IND1 (P=0.14). The viral glycoprotein antigen was incorporated in the ELISA system because it is non-infectious and induces neutralizing antibodies. The reliability and variability of the ELISA was determined by testing 516 bovine, equine and swine sera which originated from areas free of vesicular stomatitis, and by testing 186 sera from areas where outbreaks occur. ELISA and VN results were correlated (P < 0.001 for both serotypes), and no statiscafically difference was found between replications of the tests. The ELISA allows the testing of a larger number of sera in a shorter time than is possible with the VN test and it can be used in diagnostic laboratories in VSV-free areas for the support of epidemiological surveillance programs.  相似文献

An indirect enzyme-linked immunosorbent assay (ELISA) was developed to detect bovine antibody to vesicular stomatitis virus (VSV). Serum samples from cows experimentally infected with the New Jersey serotype of VSV (VSV-NJ) were assayed by the ELISA and serum-neutralization (SN) assay. The ELISA was as sensitive as the SN assay in detecting bovine antibody to VSV. The correlation between SN titers and ELISA values at absorbance at 405 nm was statistically significant. The ELISA was not specific for VSV-NJ, however, and could detect serum samples positive to the Indiana serotype of VSV that had SN titers of greater than or equal to 480. Nonspecific reactions were due to cross-reactive group-specific viral proteins that are shared by both serotypes. The cross-reactivity allows the use of a single rapid test in identifying both serotypes of VSV from the other exotic vesicular diseases, especially foot-and-mouth disease. The ELISA titers of serum samples positive for VSV-NJ were comparable with the corresponding SN titers of each sample. The sensitivity, rapidity, and ease of the ELISA system and the use of a single test in identifying both serotypes of VSV from the other exotic vesicular diseases make this ELISA suitable as a rapid diagnostic assay for VS.  相似文献

为建立方便快捷的水泡性口炎病毒(VSV)检测方法,本研究以抗VSV单克隆抗体(MAb)为捕获抗体,兔抗VSV多克隆抗体为检测抗体,建立VSV双抗体夹心ELISA检测方法。结果显示,该方法的最佳工作条件为:抗VSV MAb 1A2的包被浓度为3.09μg/mL,兔抗VSV多克隆抗体和酶标抗体的工作浓度分别为5.16μg/mL和1∶5 000,以OD450nm≥0.231作为阳性判定标准。该ELISA方法对猪水泡病病毒、猪水疱疹病毒及羊传染性脓疱病毒等均无交叉反应;敏感度可达3.125μg/mL(101TCID50);其重复性变异系数小于10%。采用建立的ELISA方法与RT-PCR方法同时检测187份临床样品,符合率达到97.9%,具有良好的相关性。本实验建立的VSV双抗体夹心ELISA检测方法具有特异性好、敏感性高、成本低及方便快捷等优点,可以用于VSV的快速检测。  相似文献

The pathogenesis of vesicular stomatitis virus (VSV) infection has not been investigated previously in native New World rodents that may have a role in the epidemiology of the disease. In the present study, 45 juvenile and 80 adult deer mice (Peromyscus maniculatus) were inoculated intranasally with VSV New Jersey serotype (VSV-NJ) and examined sequentially over a 7-day period. Virus was detected by means of immunohistochemistry and in situ hybridization in all tissues containing histologic lesions. Viral antigen and mRNA were observed initially in olfactory epithelium neurons, followed by olfactory bulbs and more caudal olfactory pathways in the brain. Virus also was detected throughout the ventricular system in the brain and central canal of the spinal cord. These results support both viral retrograde transneuronal transport and viral spread within the ventricular system. Other tissues containing viral antigen included airway epithelium and macrophages in the lungs, cardiac myocytes, and macrophages in cervical lymph nodes. In a second experiment, 15 adult, 20 juvenile, and 16 nestling deer mice were inoculated intradermally with VSV-NJ. Adults were refractory to infection by this route; however, nestlings and juveniles developed disseminated central nervous system infections. Viral antigen also was detected in cardiac myocytes and lymph node macrophages in these animals. Viremia was detected by virus isolation in 35/72 (49%) intranasally inoculated juvenile and adult mice and in 17/36 (47%) intradermally inoculated nestlings and juveniles from day 1 to day 3 postinoculation. The documentation of viremia in these animals suggests that they may have a role in the epidemiology of vector-borne vesicular stomatitis.  相似文献

The occurrence of vesicular stomatitis (VS) was investigated in El Salvador through monthly visits to 12 sentinel cattle operations located in four different departments. Management, environmental, and spatial data were collected. Heifers were enrolled on the operations and were examined and bled monthly for 3 years. Two competitive ELISAs were used to detect antibodies on each sample for each serotype of VS virus (VSV). On 8 of the 12 operations, small terrestrial rodents were trapped, blood samples collected, and antibodies to both VS serotypes evaluated using a serum-neutralization test for each virus serotype.

Similar to other studies of VS in Central America, the seroprevalence of the New Jersey serotype was higher than the seroprevalence to the Indiana serotype. An outbreak of VS appeared to occur in the Department of Sonsonate in the summer of 1999. We confirmed that VS is endemic in the four departments investigated in El Salvador.  相似文献

将水泡性口炎病毒(VSV)经差速离心和蔗糖密度梯度离心法进行纯化后,以纯化的VSV作为免疫原免疫8~10周龄雌性BALB/c小鼠,取其脾细胞与SP2/0骨髓瘤细胞进行融合,经间接ELISA方法筛选能稳定分泌抗VSV单克隆抗体的杂交瘤细胞株,并对制备出的抗VSV单抗的特异性、抗体亚类等生物学特性进行鉴定。结果显示,试验成功筛选出2株能稳定分泌抗VSV单克隆抗体的杂交瘤细胞株,分别命名为1A2、4C3。ELISA鉴定结果表明,2株单抗均能特异性地与VSV结合,而与口蹄疫病毒(FMDV)、猪水泡病病毒(SVDV)均不发生交叉反应;诱生小鼠腹水产生的抗体效价可达1∶25 600~1∶51 200。杂交瘤细胞染色体核型鉴定结果显示,杂交瘤细胞染色体数为95~105,均高于2个亲本细胞的染色体数目,说明这2株细胞是两者的杂交产物。抗体亚类鉴定结果显示,所获得2株单抗1A2、4C3均为IgG1。Western blot分析结果表明,1A2可识别VSV G蛋白。2株抗VSV单克隆抗体的成功制备将为VSV快速检测方法的建立以及检测试剂的研制等奠定基础。  相似文献

OBJECTIVE: To determine whether pigs can be infected with strains of vesicular stomatitis virus New Jersey (VSV-NJ) and vesicular stomatitis virus Indiana (VSV-I) isolated during recent vesicular stomatitis outbreaks that primarily involved horses in the western United States and determine the potential for these viruses to be transmitted by contact. ANIMALS: 128 pigs. PROCEDURE: Pigs were challenged with VSV-NJ or VSV-I from the 1995 and 1997 outbreaks of vesicular stomatitis in the western United States, respectively, or with VSV-NJ (OS) associated with vesicular stomatitis in feral pigs on Ossabaw Island, Ga. Pigs (3/group) were inoculated with each virus via 3 routes and evaluated for viral shedding, seroconversion, and the development of vesicular lesions. In another experiment, the potential for contact transmission of each virus from experimentally infected to na?ve pigs was evaluated. RESULTS: Infection of pigs was achieved for all 3 viruses as determined by virus isolation and detection of seroconversion. In inoculated pigs, all 3 viruses were isolated from multiple swab samples at concentrations sufficient to infect other pigs. However, compared with results obtained with the 2 VSV-NJ strains, viral titers associated with VSV-I were low and the duration of virus shedding was reduced. Results from the contact transmission trials were consistent with these results; virus transmission was detected most frequently with the VSV-NJ strains. CONCLUSIONS AND CLINICAL RELEVANCE: Pigs can be infected with VSV-NJ and VSV-I. Differences in the extent of viral shedding and potential for contact transmission were apparent between serotypes but not between the VSV-NJ strains investigated.  相似文献

Physical trauma, dietary factors, certain toxins, immune mediated disorders, and vesicular stomatitis virus (VSV) infection are known causes of stomatitis in horses. There is evidence that some outbreaks of equine stomatitis are caused by as yet unidentified infectious agents. It remains to be determined whether stomatitis is an emerging equine infectious disease, or if the increase in reported cases is simply the result of greater public awareness as a consequence of widespread outbreaks of VSV in the southwestern United States in recent years. Focused laboratory and epidemiological studies are necessary to more adequately define non-VS related infectious and noninfectious causes of equine stomatitis.  相似文献

An indirect sandwich enzyme-linked immunosorbent assay (ELISA) has been used for vesicular stomatitis virus (VSV) typing using sets of monovalent and polyvalent rabbit/guinea pig antisera for identification of VSV types New Jersey (VNJ) and Indiana (VIND). The VIND polyvalent antiserum (VIND-P) detects any strain of the 3 subtypes of the VIND type (VIND-1, VIND-2, and VIND-3) with the same strong reactivity. It is also possible to subtype the VIND strains using VIND-P rabbit antiserum as capture antibody and monovalent VIND-1, VIND-2, or VIND-3 guinea pig antisera as detector. The ELISA proposed has about 10 times more sensitivity and provides 10% more positive results than does the complement fixation 50% (CF50) test when epithelial samples are tested.  相似文献

水疱性口炎(VS)是由水疱性口炎病毒(VSV)引起的人畜共患的重大动物疫病.VS病毒生态学复杂,易感动物较多,传播媒介种类广,病毒可在一定区域内长期存在.VSV主要呈嗜上皮性,大量流涎是家畜感染最重要的临床症状,其特征为口腔黏膜、乳房皮肤及蹄冠部皮肤出现水泡及糜烂,人感染后出现类似流感的症状.由于其临床症状与口蹄疫不易区别,发病时容易引起国际贸易恐慌,因此,对VS诊断防治的研究有着重要的社会经济和公共卫生意义.文章从分子生物学特征、流行病学、诊断和防控4个方面对水疱性口炎的研究进行了综述.  相似文献

水疱性口炎分为印第安那型 ( Indiana)和新泽西型 ( New Jersey)。这两种血清型病毒的快速和可靠的鉴别对该病的诊断、检疫、分子流行病学调查和监测至关重要。文章按照 VSV核蛋白基因序列 ,设计了一对两型通用引物和两型各自特异性探针。研究建立了 VSV实时荧光定量 PCR检测方法 ,对 VSV细胞培养物、人工感染实验动物组织、血清样品 ,以及系列稀释的不同 TCID50 样品、其它相关或相似病毒进行鉴定 ,同时与常规 PCR、病毒分离试验作了比较。Taq Man○R RT-PCR的特异性和敏感性相当于或优于对照方法。重复性和稳定性试验证实 ,该方法可靠。每个试验中设立阳性、阴性对照和标准稀释度对照 ,使试验结果可对病毒 RNA作准确定量 ,并可在 4h内获得结果。研究结果表明 ,Taq Man○R RT-PCR方法是一种特异性强、敏感性高、快速安全的定量检测方法。因此 ,可以作为 VSV的快速检测和定型。  相似文献

Contact transmission of vesicular stomatitis virus New Jersey in pigs   总被引:4,自引:0,他引:4  
OBJECTIVE: To determine how viral shedding and development or lack of clinical disease relate to contact transmission of vesicular stomatitis virus New Jersey (VSV-NJ) in pigs and determine whether pigs infected by contact could infect other pigs by contact. ANIMALS: 63 pigs. PROCEDURE: Serologically naive pigs were housed in direct contact with pigs that were experimentally inoculated with VSV-NJ via ID inoculation of the apex of the snout, application to a scarified area of the oral mucosa, application to intact oral mucosa, or ID inoculation of the ear. In a second experiment, pigs infected with VSV-NJ by contact were moved and housed with additional naive pigs. Pigs were monitored and sampled daily for clinical disease and virus isolation and were serologically tested before and after infection or contact. RESULTS: Contact transmission developed only when vesicular lesions were evident. Transmission developed rapidly; contact pigs shed virus as early as 1 day after contact. In pens in which contact transmission was detected, 2 of 3 or 3 of 3 contact pigs were infected. CONCLUSIONS AND CLINICAL RELEVANCE: Transmission was lesion-dependent; however, vesicular lesions often were subtle with few or no clinical signs of infection. Contact transmission was efficient, with resulting infections ranging from subclinical (detected only by seroconversion) to clinical (development of vesicular lesions). Long-term maintenance of VSV-NJ via contact transmission alone appears unlikely. Pigs represent an efficient large-animal system for further study of VSV-NJ pathogenesis and transmission.  相似文献

OBJECTIVE: To identify management factors affecting the risk of animals developing vesicular stomatitis (VS). DESIGN: Case-control study. ANIMALS: Horses, cattle, and sheep with suspected vesicular stomatitis on 395 premises in Colorado, New Mexico, Utah, and Arizona. PROCEDURE: Data were collected during the VS outbreak of 1997. Diagnosticians interviewed livestock owners and completed a supplemental questionnaire. Cases were defined as those premises that had a completed questionnaire and had > or = 1 animal positive for VS. Control premises were all premises investigated that had a completed questionnaire and on which the animals had been tested but VS was not detected. RESULTS: Animals that had access to a shelter or barn had a reduced risk of developing VS (OR, 0.6; 95% confidence interval [CI], 0.35 to 0.99). This effect was more pronounced for equine premises (OR, 0.5; 95% CI, 0.3 to 0.9). Conversely, during an adjusted analysis on equine premises, risk of developing disease was increased slightly where animals had access to pasture (OR, 2.01; 95% CI, 1.1 to 3.7). On all premises where owners reported insect populations were greater than normal, odds of developing disease were significantly increased (OR, 2.5; 95% CI, 1.47 to 4.47). Premises with animals housed < 0.25 miles from running water were more than twice as likely to have clinical signs of VS (OR, 2.6; 95% CI, 1.32 to 5.0). CONCLUSIONS AND CLINICAL RELEVANCE: These results support reports of others that suggest biting insects are a vector in VS virus transmission. Management practices to reduce exposure to biting insects might reduce the risk of VS.  相似文献

Definitive diagnosis of vesicular or vesicular-like lesions in livestock animals presents challenges both for veterinary clinicians and diagnostic laboratories. It is often impossible to diagnose the causative disease agent on a clinical basis alone and difficult to collect ample vesicular epithelium samples. Due to restrictions of time and sample size, once laboratory tests have ruled out foot-and-mouth disease, vesicular stomatitis and swine vesicular disease a definitive diagnosis may remain elusive. With the ability to test a small quantity of sample for a large number of pathogens simultaneously, DNA microarrays represent a potential solution to this problem. This study describes the application of a long oligonucleotide microarray assay to the identification of viruses known to cause vesicular or vesicular-like lesions in livestock animals. Eighteen virus isolates from cell culture were successfully identified to genus level, including representatives of each foot-and-mouth disease virus serotype, two species of vesicular stomatitis virus (VSV), swine vesicular disease virus, vesicular exanthema of swine virus (VESV), bovine herpesvirus 1, orf virus, pseudocowpox virus, bluetongue virus serotype 1 and bovine viral diarrhoea virus 1. VSV and VESV were also identified in vesicular epithelium samples, with varying levels of sensitivity. The results indicate that with further development this microarray assay could be a valuable tool for the diagnosis of vesicular and vesicular-like diseases.  相似文献

BackgroundRabies is a viral disease that causes severe neurological manifestations both in humans and various mammals. Although inactivated and/or attenuated vaccines have been developed and widely used around the world, there are still concerns with regard to their safety, efficacy, and costs.ObjectiveAs demand has grown for a new rabies vaccine, we have developed a new vesicular stomatitis viruses (VSVs) based rabies vaccine that replaces glycoproteins with rabies virus (RABV) glycoprotein (GP), or so-called VSV/RABV-GP.MethodsVSV/RABV-GP production was measured by sandwich ELISA. The generation of VSV/RABV-GP was evaluated with GP-specific antibodies and reduced transduction with GP-specific neutralizing antibodies. Virus entry was quantified by measuring the luciferase levels at 18-h post-transduction. BALB/c mice (three groups of six mice each) were intraperitoneally immunized with PBS, RABA, or VSV/RABV-GP at 0 and 14 days. At 28 days post-immunization serology was performed. Statistical significance was calculated using the Holm–Sidak multiple Student’s t test.ResultsMice immunized with VSV/RABV-GP produced IgM and IgG antibodies, whereas IgM titers were significantly higher in mice immunized with VSV/RABV-GP compared to inactivated RABV. The secretion profiles of IgG1 and IgG2a production suggested that VSV/RAVB-GP induces the T helper cell type-2 immune bias. In addition, the average (±SD; n = 3) serum neutralization titers of the inactivated RABV and VSV/RABV-GP groups were 241 ± 40 and 103 ± 54 IU/mL, respectivelyConclusionOur results confirm that VSV/RABV-GP could be a new potential vaccination platform for RABV.  相似文献
